Introducción: La infección del tracto urinario (ITU) es de las más frecuentes en los ancianos institucionalizados. Es necesario conocer las resistencias de algunos antibióticos a los gérmenes frecuentes, y el efecto del tratamiento fisioterapéutico. Objetivos: Conocer la prevalencia de ITU en ancianos, determinar los agentes etiológicos implicados, analizar las resistencias a los antibióticos utilizados en su tratamiento, y establecer relaciones entre dichas infecciones urinarias con el tratamiento fisioterapéutico. Material y Métodos: Estudio descriptivo, de corte transversal, de ITU en un centro geriátrico de 122 residentes. Se ha realizado cultivo y antibiograma de 56 pacientes con sospecha de ITU. Se ha estudiado la asociación de microorganismo aislado en relación al sexo, actividad física, rehabilitación y resistencias antibióticas. Resultados: Los datos de prevalencia son de un 45,90% (el I.C. al 95% es de 37,06%-54,74%) de infecciones urinarias clínicas y de un 37,70% (29,10-46,30) de infecciones urinarias confirmadas analíticamente. La inmovilidad es un factor de riesgo de infección urinaria, mientras que la rehabilitación es un factor protector, aunque estas medidas no alcanzan el valor de significación estadística en este estudio. Se observa mayor frecuencia de ITU en hombres que en mujeres pero estos valores tampoco son estadísticamente significativos. Escherichia coli es el principal microorganismo aislado. Conclusiones: Las características del paciente geriátrico institucionalizado podrían explicar algunos de los resultados hallados. Otros estudios determinan que la incidencia de ITU es mayor en mujeres que en hombres, siendo el principal microorganismo encontrado el E. coli en mujeres. Diversas publicaciones confirman que la fisioterapia geriátrica apoya a fortalecer los músculos y mejorar la calidad de vida. With the objective of evaluating the epidemic situation of the breathing processes in egg-laying hens, a retrospective epidemiological study was carried out during the period between January, of 2004 to December, 2008, in two commercial enterprises. From the reception of the information seated in the official registrations of the provincial Veterinary Medicine Institute, the relative mortality per group diseases, the prevalence of focal period and the positive focal incidence were calculated by WinEpi TASAS version 2.0 program. In order to know the temporary behavior of the health events referred, a analysis of chronological series was carried out in STATGRAPHICS PLUS 5.1, determining tendency, seasonal variation and ciclicidad. Kruskal Wallis and U the Mann-Whitney (Minitab 14 statistical package) test were applied in order to compare the relative mortality between months and season of the year. It was demonstrated that the health events more prevalent during the period analyzed were breathing and digestive diseases and they constitute the main causes of deaths in the category analyzed. A similar dynamic of the positive focal incidence in both breathing events; could be verified however, several behaviors were appreciated regarding levels of infection reached during the period. A marked seasonal and recurrent variation was observed in the rainy season in both breathing processes in egg-laying hens in the years analyzed. [ABSTRACT FROM AUTHOR]

Magnetic CoNi@Au core–shell nanorods have been electrochemically synthesized, characterized and functionalized to test their inherent cytotoxicity in order to assess their potential use for biomedical applications. The initially electrodeposited CoNi nanorods have been covered with a gold layer by means of galvanic displacement to minimize the nanowires toxicity and their aggregation, and favour the functionalization. The presence of a gold layer on the nanorod surface slightly modifies the magnetic behaviour of the as-deposited nanorods, maintaining their soft-magnetic behaviour and high magnetization of saturation. The complete covering of the nanorods with the gold shell favours a good functionalization with a layer of (11-Mercaptoundecyl)hexa(ethylene glycol) molecules, in order to create a hydrophilic coating to avoid the aggregation of nanorods, keeping them in suspension and give them stability in biological media. The presence of the organic layer incorporated was detected by means of electrochemical probe experiments. A cytotoxicity test of functionalized core–shell nanorods, carried out with adherent HeLa cells, showed that cell viability was higher than 80% for amounts of nanorods up to 10 μg mL − 1 . These results make functionalized nanorods promising vehicles for targeted drug delivery in medicine, which gives a complementary property to the magnetic nanoparticles. [ABSTRACT FROM AUTHOR]

Meeting the contemporary demand for the development of functional, biocompatible, and environment friendly self-assembled structures using efficient, cost-effective, and energy-saving methods, the field of colloids has witnessed a surge in interest. Research into cationic and anionic (catanionic) surfactant combinations has gained momentum due to their distinct advantages and synergistic properties in this context. Catanionic self-assemblies have emerged as promising contenders for addressing these requirements. Catanionic self-assemblies possess high stability, adjustable surface charge, and low critical aggregation concentration. This comprehensive review article distinguishes between cationic/anionic non-equimolar and equimolar ratio mixing formation of high-salt catanionic self-assemblies known as catanionic mixture and salt-free counterparts, termed ion-pair amphiphiles, respectively. It explores diverse synthesis techniques, emphasizing the roles of solvents, salts, and pH conditions and covers both experimental and theoretical aspects of state-of-the-art catanionic self-assemblies. Additionally, the review investigates the development of multi-responsive catanionic self-assemblies using light, pH, temperature, and redox, responsive cationic/anionic amphiphiles. It provides an in-depth exploration of potential synergistic interactions and properties, underscoring their practical importance in a wide range of industrial applications. The review explores challenges like precipitation, stability and identifies knowledge gaps, creating opportunities in the dynamic catanionic self-assembly field. Singlet oxygen (SO) reacts with organic molecules, for example in degrading environmental contaminants and causing toxicity to cells in photodynamic therapy (PDT). The relevance of SO in the environmental and biomedical domains has fuelled research towards improved methodologies for its efficient generation. In this paper, we report the use of a bis-imidazolium-based amphiphile that forms supramolecular gels in water-ethanol mixtures encapsulating SO generating chromophores. The gels comprise twisted fibres that incorporate one of four different porphyrins: 5,10,15,20-tetrakis(N-methyl-4-pyridinium)porphyrin tetraiodide (TPPP), 5,10,15,20-tetrakis(4-carboxyphenyl)porphyrin (TCPP), 5,10,15,20-tetrakis(3-hydroxyphenyl)porphyrin (TPP-3OH) and 5,10,15,20-tetrakis(4-hydroxyphenyl)porphyrin (TPP-4OH). Rheological measurements confirmed viscoelastic properties characteristic of these types of supramolecular gels, suggesting their potential as effective local PDT delivery systems. Enhanced SO generation within the hydrogel matrix compared to the solution of the chromophores was observed in suspension in a spectrophotometer using uric acid as the molecular probe. The SO generation was also shown through antimicrobial PDT (aPDT) studies. The TPPP-containing gel showed the highest reduction in the colony forming unit (CFU) count, which is 94% against E. coli and 100% against S. aureus. These results indicate that the porphyrin gels based on a gemini amphiphile with a high level of SO production are of significant interest and have a lot of potential use in aPDT., (Copyright © 2025 The Author(s). Glutathione (GSH) plays a vital role in the regulation of intracellular functions which alterations in physiological glutathione levels are associated to various diseases. Molecular bioimaging is a sensitive method for GSH detection, but challenges persist in the development of fluorescent probes, mainly concerning long-term tracking of intracellular GSH concentration because of aggregation of molecular probes and their washout in cells. Engineered nanomaterials have shown great promise for increasing the disease diagnosis accuracy. Microchips generated by advanced microfabrication techniques can be applied in designing biomedical devices due to control over size, shape, and bioactive coatings utilization. In the current work, the synthesis and characterization of two GSH probes, Bdpy1 and Bdpy2, is reported, each offering irreversible and reversible GSH reactions, respectively. These GSH probes are immobilized on silicon oxide microchips (SOμC), micro-fabricated using photolithographic techniques, to give SOμC-Bdpy1 and SOμC-Bdpy2. Both functionalized microchips exhibited sensitivity to GSH, and, notably, the reversible SOμC-Bdpy2 showed less time dependency, making it more suitable for long-term intracellular GSH sensing. In vitro experiments in HeLa cells reveal both SOμC-Bdpy1 and SOμC-Bdpy2 were internalized in living cells, showing SOμC-Bdpy2 more reliable results (due to its less time dependency) for quantifying intracellular GSH. Remarkably, the intracellular GSH measurement was monitored by SOμC-Bdpy2 for 48 h, indicating the functionalized microchips capability to detect GSH amount in different time intervals. This study introduces a promising approach for long term quantification of intracellular GSH, overcoming the limitation of fluorescent probes and offering valuable insights into microchip-based sensing methodologies.

Drug delivery advances rely on using nano- and microsized carriers to transfer therapeutic molecules, although challenges persist in increasing the availability of new and even approved pharmaceutical products. Particle shape, a critical determinant in how these carriers distribute within the body after administration, raises opportunities of using, for instance, micrometer-sized nonspherical particles for vascular targeting and thereby creating new prospects for precise drug delivery to specific targeted areas. The versatility of polycrystalline silicon microfabrication allows for significant variation in the size and shape of microchips, and so, in the current work, photolithography was employed to create differently shaped polysilicon microchips, including cuboids, cubes, bars, and cylinders, to explore the influence of particle shape on cellular interactions. These microchips with different shapes and lateral dimensions, accounting for surface areas in the range of ca. 15 to 120 μm 2 and corresponding total volumes of 0.4 to 27 μm 3 , serve as ideal models for investigating their interactions with macrophages with diameters of ca. 20 μm. Side-scattering imaging flow cytometry was employed for studying the interaction of label-free prepared microchips with RAW 264.7 macrophages. Using a dose of 3 microchips per cell, results show that cuboids exhibit the highest cellular association (ca. 25%) and uptake (ca. 20%), suggesting their potential as efficient carriers for targeted drug delivery to macrophages. Conversely, similarly sized cylinders and bar-shaped microchips exhibit lower uptakes of about 8% and about 6%, respectively, indicating potential benefits in evading macrophage recognition. On average, 1-1.5 microchips were internalized, and ca. 1 microchip was surface-bound per cell, with cuboids showing the higher values overall. Macrophages respond to microchips by increasing their metabolic activity and releasing low levels of intracellular enzymes, indicating reduced toxicity. Interestingly, increasing the particle dose enhances macrophage metabolic activity without significantly affecting enzyme release.

Glutathione (GSH) is one of the most common thiol-containing molecules discovered in biological systems, and it plays an important role in many cellular functions, where changes in physiological glutathione levels contribute to the progress of a variety of diseases. Molecular imaging employing fluorescent probes is thought to be a sensitive technique for online fluorescence detection of GSH. Although various molecular probes for (intracellular) GSH sensing have been reported, some aspects remain unanswered, such as quantitative intracellular analysis, dynamic monitoring, and compatibility with biological environment. Some of these drawbacks can be overcome by sensors based on nanostructured materials, that have attracted considerable attention owing to their exceptional properties, including a large surface area, heightened electro-catalytic activity, and robust mechanical resilience, for which they have become integral components in the development of highly sensitive chemo- and biosensors. Additionally, engineered nanomaterials have demonstrated significant promise in enhancing the precision of disease diagnosis and refining treatment specificity. The aim of this review is to investigate recent advancements in fabricated nanomaterials tailored for detecting GSH. Specifically, it examines various material categories, encompassing carbon, polymeric, quantum dots (QDs), covalent organic frameworks (COFs), metal-organic frameworks (MOFs), metal-based, and silicon-based nanomaterials, applied in the fabrication of chemo- and biosensors. The fabrication of nano-biosensors, mechanisms, and methodologies employed for GSH detection utilizing these fabricated nanomaterials will also be elucidated. Remarkably, there is a noticeable absence of existing reviews specifically dedicated to the nanomaterials for GSH detection since they are not comprehensive in the case of nano-fabrication, mechanisms and methodologies of detection, as well as applications in various biological environments. This research gap presents an opportune moment to thoroughly assess the potential of nanomaterial-based approaches in advancing GSH detection methodologies.

Nano- and micro-carriers of therapeutic molecules offer numerous advantages for drug delivery, and the shape of these particles plays a vital role in their biodistribution and their interaction with cells. However, analysing how microparticles are taken up by cells presents methodological challenges. Qualitative methods like microscopy provide detailed imaging but are time-consuming, whereas quantitative methods such as flow cytometry enable high-throughput analysis but struggle to differentiate between internalised and surface-bound particles. Instead, imaging flow cytometry combines the best of both worlds, offering high-resolution imaging with the efficiency of flow cytometry, allowing for quantitative analysis at the single-cell level. This study focuses on fluorescently labelled silicon oxide microchips of various morphologies but related surface areas and volumes: rectangular cuboids and apex-truncated square pyramid microchips fabricated using photolithography techniques, offering a reliable basis for comparison with the more commonly studied spherical particles. Imaging flow cytometry was utilised to evaluate the effect of particle shape on cellular uptake using RAW 264.7 cells and revealed phagocytosis of particles with all shapes. Increasing the particle dose enhanced the uptake, while macrophage stimulation had minimal effect. Using a ratio particle:cell of 10:1 cuboids and spheres showed an uptake rate of approximately 50%, in terms of the percentage of cells with internalised particles, and the average number of particles taken up per cell ranging from about 1-1.5 particle/cell for all the different shapes. Quantum biological tunnelling for electron transfer is involved in controlling essential functions for life such as cellular respiration and homoeostasis. Understanding and controlling the quantum effects in biology has the potential to modulate biological functions. Here we merge wireless nano-electrochemical tools with cancer cells for control over electron transfer to trigger cancer cell death. Gold bipolar nanoelectrodes functionalized with redox-active cytochrome c and a redox mediator zinc porphyrin are developed as electric-field-stimulating bio-actuators, termed bio-nanoantennae. We show that a remote electrical input regulates electron transport between these redox molecules, which results in quantum biological tunnelling for electron transfer to trigger apoptosis in patient-derived cancer cells in a selective manner. Transcriptomics data show that the electric-field-induced bio-nanoantenna targets the cancer cells in a unique manner, representing electrically induced control of molecular signalling. Optical tweezers (OT) have become an essential technique in several fields of physics, chemistry, and biology as precise micromanipulation tools and microscopic force transducers. Quantitative measurements require the accurate calibration of the trap stiffness of the optical trap and the diffusion constant of the optically trapped particle. This is typically done by statistical estimators constructed from the position signal of the particle, which is recorded by a digital camera or a quadrant photodiode. The finite integration time and sampling frequency of the detector need to be properly taken into account. Here, we present a general approach based on the joint probability density function of the sampled trajectory that corrects exactly the biases due to the detector's finite integration time and limited sampling frequency, providing theoretical formulas for the most widely employed calibration methods: equipartition, mean squared displacement, autocorrelation, power spectral density, and force reconstruction via maximum-likelihood-estimator analysis (FORMA). Multifunctional gold nanoparticles (AuNPs) are of great interest, owing to their vast potential for use in many areas including sensing, imaging, delivery, and medicine. A key factor in determining the biological activity of multifunctional AuNPs is the quantification of surface conjugated molecules. There has been a lack of accurate methods to determine this for multifunctionalized AuNPs. We address this limitation by using a new method based on the deconvolution and Levenberg-Marquardt algorithm fitting of UV-visible absorption spectrum to calculate the precise concentration and number of cytochrome C (Cyt C ) and zinc porphyrin (Zn Porph) bound to each multifunctional AuNP. Dynamic light scattering (DLS) and zeta potential measurements were used to confirm the functionalization of AuNPs with Cyt C and Zn Porph. Transmission electron microscopy (TEM) was used in conjunction with UV-visible absorption spectroscopy and DLS to identify the AuNP size and confirm that no aggregation had taken place after functionalization. Despite the overlapping absorption bands of Cyt C and Zn Porph, this method was able to reveal a precise concentration and number of Cyt C and Zn Porph molecules attached per AuNP. Furthermore, using this method, we were able to identify unconjugated molecules, suggesting the need for further purification of the sample. This guide provides a simple and effective method to quickly quantify molecules bound to AuNPs, giving users valuable information, especially for applications in drug delivery and biosensors.

Photodynamic therapy (PDT) is facing challenges such as poor solubility, precise delivery, self-aggregation, and photobleaching of photosensitizers with cancer cells due to their less tendency to accumulate in tumor tissues. To address these challenges, we have explored a Rose Bengal (RB)-loaded metallocatanionic vesicles (MCVs) nanosystem for the phototoxicity of cancer cells. Different sets of MCVs were prepared by two different cationic single-chain metallosurfactants, i.e. , hexadecylpyridinium trichlorocuprate (CuCPC I) and hexadecylpyridinium trichloroferrate (FeCPC I) in combination with anionic double-chain sodium bis(2-ethylhexyl)sulfosuccinate (AOT) surfactant in phosphate buffer saline of pH 7.4. The RB-loaded CuCPC I:AOT and FeCPC I:AOT vesicles enhanced the maximum singlet oxygen ( 1 O 2 ) generation by 1-fold and 3-fold, respectively, compared to pure RB. Upon irradiation with a 532 nm laser for 10 min, these RB-loaded CuCPC I:AOT and FeCPC I:AOT MCVs significantly decreased the metabolic activity of U-251 cells by 70% and 85% at MCVs concentration of 0.75 μM, respectively. Furthermore, RB-loaded MCVs showed the highest intracellular 1 O 2 -mediated membrane damage and cell-killing effect as confirmed by singlet oxygen sensor green and differential nuclear staining assay, which is attributed to the cellular uptake profile of different RB-loaded MCVs fractions. Caspase 3/7 assay confirmed the apoptotic pathway of cell death by activating caspase. Therefore, the photoactivation of RB-loaded MCVs led to a significant reduction in the viability of U-251 cells (maximum 85%), which resulted in cell death. Our study demonstrated the advantage of using these dual-charge and biocompatible metallocatanionic vesicles as a promising delivery system of photodynamic therapy that can enhance 1 O 2 generation from PS and can be further utilized in photomedicine.

Curcumin shows anti-inflammatory activity, and it has been widely investigated for neurodegenerative diseases, adjuvant treatment in AIDS and antitumor activity against different tumors, among other activities. The goal of this work was to evaluate the capacity of curcumin and its derivatives (bisdemethoxycurcumin and bisdemethylcurcumin) in preventing the irritant effects of topically applied xylol and to assess the intrinsic capacity of curcuminoids in permeating human skin by ex vivo permeation tests. Its secondary goal was to validate an HPLC method to simultaneously determine the curcuminoids in the samples from the ex vivo permeation studies and drug extraction from the skin. Curcuminoid quantification was performed using an RP-C18 column, at isocratic conditions of elution and a detection wavelength of 265 nm. The method was specific with a suitable peak resolution, as well as linear, precise, and accurate in the range of 0.195-3.125 μg/mL for the three curcuminoids. Bisdemethylcurcumin showed the greatest permeation through the human skin, and it was the curcuminoid that was most retained within the human skin. The anti-inflammatory activity of the curcuminoids was evaluated in vivo using a xylol-induced inflammation model in rats. Histological studies were performed to observe any changes in morphology at the microscopic level, and these three curcuminoids were found to be respectful within the skin structure. These results show that these three curcuminoids are suitable for anti-inflammatory formulations for dermal applications, and they can be properly quantified using HPLC-UV.

The mixing of [V 10 O 28 ] 6- decavanadate anions with a dicationic gemini surfactant (gem) leads to the spontaneous self-assembly of surfactant-templated nanostructured arrays of decavanadate clusters. Calcination of the material under air yields highly crystalline, sponge-like V 2 O 5 (gem-V 2 O 5 ). In contrast, calcination of the amorphous tetrabutylammonium decavanadate allows isolation of a more agglomerated V 2 O 5 consisting of very small crystallites (TBA-V 2 O 5 ). Electrochemical analysis of the materials' performance as lithium-ion intercalation electrodes highlights the role of morphology in cathode performance. The large crystallites and long-range microstructure of the gem-V 2 O 5 cathode deliver higher initial capacity and superior capacity retention than TBA-V 2 O 5 . The smaller crystallite size and higher surface area of TBA-V 2 O 5 allow faster lithium insertion and superior rate performance to gem-V 2 O 5 ., (© 2023 The Authors. Photodynamic therapy (PDT) is an innovative technique for cancer treatment with minimal side effects, based on the use of a photosensitizer, oxygen, and light. Photosensitizers (PSs) have several limitations, that may limit their clinical use, like poor solubilization, self-aggregation, and lack of specific targeting, which can be addressed with the use of nanomaterials. Herein, a unique type of catansomes (CaSs) was prepared using a gemini imidazolium-based surfactant (1,3-bis[(3-octadecyl-1-imidazolio)methyl]benzene dibromide (GBIB) and a double chain surfactant, diaoctyl sodium sulfosuccinate or Aerosol OT (AOT). The formation of CaS GBIB/AOT was optimized in various ethanol/water (E/W) solvent ratios by employing a facile, quick, and most reliable solution-solution mixing method. The CaS was characterized by dynamic light scattering (DLS) and field emission gun scanning electron microscopy (FEG-SEM) techniques. The experimental results reveal that stable CaSs with a spherical shape were obtained at lower concentration (100 μM). Rose Bengal (RB), a PS of the xanthene family, was incorporated into these prepared CaSs, as proven by fluorescence spectroscopy, UV-visible absorption spectroscopy, and confocal laser scanning microscopy. Singlet oxygen ( 1 O 2 ) generation studies revealed the relevant role of the E/W solvent ratio as there was a 4-fold boost in the 1 O 2 production for GBIB/AOT in E/W = 50:50 and around 3-fold in E/W = 30:70. Also, the GBIB-rich 80:20 fraction was more efficient in increasing the 1 O 2 generation as compared to the AOT rich fraction (20:80). Further, their phototoxicity was tested in a water-rich solvent ratio (E/W = 30:70) against MCF-7 cells. Upon irradiation with a 532 nm laser (50 mW) for 5 min, RB@GBIB/AOT(20:80) fraction caused 50% decrease in the metabolic activity of MCF-7 cells, and RB@GBIB/AOT(80:20) fraction produced a maximum 85% decrease in cell viability. Furthermore, the enhancement in intracellular 1 O 2 generation by RB@GBIB/AOT, as compared to pure RB, was confirmed with singlet oxygen sensor green (SOSG). This new type of CaS based on gemini surfactants exhibiting a large amount of 1 O 2 generation, holds great interest for several applications, such as use in photomedicine in future.

The use of micrometric-sized vehicles could greatly improve selectivity of cytotoxic compounds as their lack of self-diffusion could maximize their retention in tissues. We have used polysilicon microparticles ( SiμP ) to conjugate bipyridinium-based compounds, able to induce cytotoxicity under regular intracellular conditions. Homogeneous functionalization in suspension was achieved, where the open-chain structure exhibits a more dense packing than cyclic analogues. The microparticles internalized induce high cytotoxicity per particle in cancerous HeLa cells, and the less densely packed functionalization using cyclophanes promotes higher cytotoxicity per bipy than with open-chain analogues. The self-renewing ability of the particles and their proximity to cell membranes may account for increased lipid peroxidation, achieving toxicity at much lower concentrations than that in solution and in less time, inducing highly efficient cytotoxicity in cancerous cells.
