3. Ergostane triterpenoids from cultures of basidiomycete Favolaschia minutissima.

Abstract

Five undescribed ergostane triterpenoids, favolons D–G (1 – 4) and favolanone A (5), were isolated together with three known compounds, favolon (6), favolon C (7), and laschiatrion (8), from cultures of basidiomycete Favolaschia minutissima TBRC-BCC 19434. The undescribed compounds were evaluated for antimalarial, antitubercular, and antibacterial activities. Favolanone A (5) exhibited an IC 50 value of 13 μM in the antimalarial activity assay against Plasmodium falciparum K1 (multidrug-resistant strain). However, it also showed moderate cytotoxicity to Vero cells (IC 50 22 μM). In the present study, it was newly found that favolon (6) exhibits activity against the plant pathogenic fungus Alternaria brassicicola (MIC 3.13 μg/ml). 19. Maleic anhydride and chromone derivatives from the endophytic fungus BCC 54265 (Botryosphaeriaceae).

Abstract

A maleic anhydride derivative, botryoanhydride (1), and a chromone derivative, botryochromone (2), together with three known chromones, eugenitin (3), 6-hydroxymethyleugenin (4) and 6-methoxymethyleugenin (5), were isolated from cultures of the endophytic fungus BCC 54265 of the family Botryosphaeriaceae. Abstract

A new seco-tremulane, 11,12-epoxy-5,6-secotremula-1,6(13)-dien-5,12-olide ( 1), was isolated together with the known compounds, conocenolides A ( 2) and B ( 3), tremulenediol A ( 4), tremulenolide A ( 5), and two lanostane triterpenoids, trametenolic acid B ( 6), and pinicolic acid A ( 7), from cultures of the basidiomycete Flavodon flavus BCC 17421. Interconversion of conocenolides A/B was demonstrated. Compound 1 exhibited weak cytotoxic activities, whereas tremulenediol A showed antiplasmodial activity ( IC50 8.6 μg/ml). Two new hirsutane sesquiterpenes, marasmiellins A (1) and B (2), were isolated from cultures of the basidiomycete Marasmiellussp. BCC 22389. The structures were elucidated on the basis of NMR spectroscopic and mass spectrometry data. The absolute configuration of marasmiellin B was determined by application of the modified Mosher’s method.

Biological activities of the new compounds were evaluated.

Abstract

Paecilodepsipeptide A (1), a new cyclohexadepsipeptide possessing three d-amino acid residues, together with its linear analogues paecilodepsipeptides B (2) and C (3), was isolated from the insect pathogenic fungus Paecilomyces cinnamomeusBCC 9616. Structures of these compounds were elucidated primarily by NMR and mass spectroscopic analyses. The absolute configurations of the amino acid and hydroxy acid residues of 1were addressed by HPLC analysis of its acid hydrolyzate using a chiral column and Marfey's method. Paecilodepsipeptide A (1) showed activity against the malarial parasite Plasmodium falciparumK1 with an IC50value of 4.9 M. This compound also showed cytotoxicity to two cancer cell lines, KB (IC505.9 M) and BC (IC506.6 M); however, it was inactive against noncancerous Vero cells up to 67 M (50 g/mL).

Two new γ-lactones, eutypellins A (1) and B (2), and two ent-eudesmane sesquiterpenes, ent-4(15)-eudesmen-11-ol-1-one (3) and ent-4(15)-eudesmen-1α,11-diol (4), together with three known pimarane diterpenes, diaporthein B, scopararane A, and libertellenone C, were isolated from the endophytic fungus Eutypellasp. BCC 13199. The structures of these compounds were elucidated by interpretation of spectroscopic data. The absolute configuration of 4was confirmed by application of the modified Mosher’s method. Eutypellin A (1) and sesquiterpene 3exhibited weak cytotoxic activities.

Two new prenylhydroquinone-derived compounds, lentinospirol (1)and 1-(2,5-dihydroxyphenyl)-4-hydroxy-3-methyl-1-butanone (2), were isolated from cultures of the basidiomycete Lentinus similisBCC 52578, together with the known compounds panepoxydone (3), panepoxydione (4), isopanepoxydone (5), 2,2-dimethyl-6-hydroxy-2H-chromene (6), and (3R,4S)-3,4-dihydroxy-2,2-dimethyl-6-methoxychroman (7). Compounds 3and 4exhibited cytotoxicity against all cell-lines tested, while the other compounds were inactive.
