Abstract

The increased environmental presence of micro-/nanoplastics (MNPLs) and the potential health risks associated with their exposure classify them as environmental pollutants with special environmental and health concerns. Consequently, there is an urgent need to investigate the potential risks associated with secondary MNPLs. In this context, using “true-to-life” MNPLs, resulting from the laboratory degradation of plastic goods, may be a sound approach. These non-commercial secondary MNPLs must be labeled to track their presence/journeys inside cells or organisms. Because the cell internalization of MNPLs is commonly analyzed using fluorescence techniques, the use of fluorescent dyes may be a sound method to label them. Five different compounds comprising two chemical dyes (Nile Red and Rhodamine-B), one optical brightener (Opticol), and two industrial dyes (Amarillo Luminoso and iDye PolyPink) were tested to determine their potential for such applications. Using commercial standards of polystyrene nanoplastics (PSNPLs) with an average size of 170 nm, different characteristics of the selected dyes such as the absence of impact on cell viability, specificity for plastic staining, no leaching, and lack of interference with other fluorochromes were analyzed. Based on the overall data obtained in the wide battery of assays performed, iDye PolyPink exhibited the most advantages, with respect to the other compounds, and was selected to effectively label “true-to-life” MNPLs. These advantages were confirmed using a proposed protocol, and labeling titanium-doped PETNPLs (obtained from the degradation of milk PET plastic bottles), as an example of “true-to-life” secondary NPLs. These results confirmed the usefulness of iDye PolyPink for labeling MNPLs and detecting cell internalization, A. Abstract

Javier Carvajal fue uno de los arquitectos más relevantes dentro del cambio que experimentó la arquitectura española de mediados del siglo XX. Este trabajo aborda su interés por la integración de las artes, en particular del diseño y la arquitectura en las tiendas que creó para Loewe. El contexto histórico en el que trabajó no sólo mostraba un cambio en el modo de ver la arquitectura, sino también en la variación de los hábitos de consumo de un país en evolución. Su colaboración con Loewe supuso la modernización de la marca y en particular de sus tiendas, donde el arquitecto consiguió crear un espacio entorno al ritual de la compra de lujo, y entender que la envolvente y el mobiliario iban a definir el concepto de la nueva tienda contemporánea. En el estudio que presentamos aquí se analizan cinco de las tiendas que Carvajal hizo para Loewe entre los años 1959 y 1964 -Madrid, Bilbao, Mallorca, Barcelona y Valencia- aplicando estas cuestiones y otras. Pero sobre todo, se estudia la relación del mobiliario singular de las tiendas, partiendo de los planos originales, en cuanto a su influencia en las fases del protocolo de compra. En definitiva, se trata de un trabajo que aborda el trabajo de Javier Carvajal para definir un modelo de tienda para Loewe a través un mobiliario acorde al carácter de la empresa., Javier Carvajal was one of the most relevant architects in the change that the spanisg architecture experienced in the mid-20th century. Throught this study we will see his interest in the integration os the arts, particularly in the design and the architecture into the commercial architecture that created for Loewe. The historical context in which he worked, not only showed a change in the way of seeing architecture, but also in the variation of consumer habits in development country. His collaboration with Loewe meant the modernization with the brand and aprticullary with his stores, where the architect managed to create a space around the ritual of luxury shopping, and with it, understanding that the envelope and furniture were going to define the concept of the new contemporany store. In this case study, we analyze five stores that Carvajal design for Loewe between 1959 and 1964 -Madrid, Bilbao, Mallorca, Barcelona y Valencia- applying this and other points on them. Above all, we study the relationship between the store´s furniture, starting from the original plans, in terms of its importance to influence and guide the phases of the purchase prorocol. Abstract

The alkaline comet assay, or single cell gel electrophoresis, is one of the most popular methods for assessing DNA damage in human population. One of the open issues concerning this assay is the identification of those factors that can explain the large inter-individual and inter-laboratory variation. International collaborative initiatives such as the hCOMET project a COST Action launched in 2016 represent a valuable tool to meet this challenge. The aims of hCOMET were to establish reference values for the level of DNA damage in humans, to investigate the effect of host factors, lifestyle and exposure to genotoxic agents, and to compare different sources of assay variability. A database of 19,320 subjects was generated, pooling data from 105 studies run by 44 laboratories in 26 countries between 1999 and 2019. A mixed random effect log-linear model, in parallel with a classic meta-analysis, was applied to take into account the extensive heterogeneity of data, due to descriptor, specimen and protocol variability. As a result of this analysis interquartile intervals of DNA strand breaks (which includes alkali-labile sites) were reported for tail intensity, tail length, and tail moment (comet assay descriptors). A small variation by age was reported in some datasets, suggesting higher DNA damage in oldest age-classes, while no effect could be shown for sex or smoking habit, although the lack of data on heavy smokers has still to be considered. Finally, highly significant differences in DNA damage were found for most exposures investigated in specific studies. In conclusion, these data, which confirm that DNA damage measured by the comet assay is an excellent biomarker of exposure in several conditions, may contribute to improving the quality of study design and to the standardization of results of the comet assay in human populations.(c) 2021 The Authors.
