Abstract

A truly organocatalytic approach to the Darzens reaction affording α,β-epoxy carbonyl compounds in good yields was developed taking advantage of the high basic strength and low nucleophilicity of cyclopropenimine superbases. The catalytic active free base can easily be generated in situ from its hydrochloride salt and maintained in the active deprotonated form by performing the reactions in a heterogeneous reaction system in the presence of excess potassium carbonate as a sacrificial base. Abstract

The divergent supramolecular behavior of a series of tripeptide stereoisomers was elucidated through spectroscopic, microscopic, crystallographic, and computational techniques. Only two epimers were able to effectively self-organize into amphipathic structures, leading to supramolecular hydrogels or crystals, respectively. Despite the similarity between the two peptides’ turn conformations, stereoconfiguration led to different abilities to engage in intramolecular hydrogen bonding. Self-assembly further shifted the pKavalue of the C-terminal side chain. As a result, across the pH range 4–6, only one epimer predominated sufficiently as a zwitterion to reach the critical molar fraction, allowing gelation. By contrast, the differing pKavalues and higher dipole moment of the other epimer favored crystallization. The four stereoisomers were further tested for gold nanoparticle (AuNP) formation, with the supramolecular hydrogel being the key to control and stabilize AuNPs, yielding a nanocomposite that catalyzed the photodegradation of a dye. Importantly, the AuNP formation occurred without the use of reductants other than the peptide, and the redox chemistry was investigated by LC–MS, NMR, and infrared scattering-type near field optical microscopy (IR s-SNOM). This study provides important insights for the rational design of simple peptides as minimalistic and green building blocks for functional nanocomposites.

Abstract

The interaction of para-substituted benzyl hydroxyalkyl nitroxide with the monolayer of water-soluble protected gold cluster made by a short alkyl chain and a triethylene glycol monomethyl ether unit is studied by using EPR spectroscopy. The inclusion of nitroxide probes in the more hydrophobic environment of the monolayer gave rise to a reduction of the value of both nitrogen and Beta-proton hyperfine splittings.

A demonstration that the core size of metallic nanoparticles passes on to the protective organic monolayer that has various water soluble properties could be measured with the help of ESR spectroscopy is presented. Results confirmed that ESR spectroscopy is an effective tool to check the properties of nanoparticles that binds them.

Abstract

One of the tremendous areas of growth on the research side involves using nanomaterials both as imaging agents and as drug delivery vehicles. Nuclear magnetic resonance imaging (MRI) is a particular attractive imagin modality: it provides high-resolution, detailed structural images, three-dimensional spatial reconstruction, and no ionizing radiation.[1] Moreover, MRI is superior to optical bio-imaging in living systems for the visualization of deep tissues. 1H and 19F are the most sensitive nucleai for MRI, with the 1H signal suited for collecting information about the body for diagnostic purposes due to its high sensitivity. However, 1H MRI often suffers from a low contrast-to-noise ratio because of the large background signals from water protons. Therefore, for specific imaging both in vivo and ex vivo, there is still an obvious need to develop new methodologies. In particular, 19F holds great promise as an alternative nuclide for MRI as it is highly sensitive in nuclear magnetic resonance (NMR) spectroscopy (83% relative to 1H) and has 100% natural abundance. A more important advantage of 19F is that in animal bodies essentially no endogenous 19F is detectable by MRI, which therefore eliminates interference from background signals. Moreover, unlike other prototypical metal-based contrast agents, 19F MRI also enables image quantification.[2] As 19F imaging agents, a variety of fluorinated small molecule compounds have been proposed, perfluoro-15-crown-5-ether (PFCE) or perfluorobenzene (PFB) being the most commonly used perfluorocarbons (PFCs) in 19F-MRI research. These PFC molecules have poor solubility in water and, as such, are typically emulsified into lipid micro/NP formulations for biological applications. Moreover, such lipid-based formulations sometimes have drawbacks, which limit imaging capabilities to the vasculature with intravenous administration. In the last years we have reported examples of water soluble gold nanoparticles (NPs) protected by homoligand and heteroligand fluorinated-/hydrogenated ligands.[3] We will reported in this communication the design, synthesis, and characterization of gold nanoparticles (NPs) protected by fluorinated ligands which are endowed with 19F NMR properties suitable for applications in diagnostic imaging. Considering that gold NPs are excellent scaffold for medical uses and that they can be easily modified introducing pro(drugs) and specific recognition elements, our fluorinated NPs may behave both as imaging and therapeutic agents.

Abstract

The detection of hepatocellular carcinoma (HCC) may be improved by analyzing the novel class of serumbiomarkers constituted by IgM immune complexes of well-known cancer antigens such as alpha-fetoprotein (AFP-IgM). To evaluate the relevance of the natural immune response to AFP for HCC diagnosis, we assessed the levels of IgM anti-AFP antibodies in serum samples of patients with HCC and with cirrhosis (CR) using a dedicated IgM anti-AFP assay. The assay result was compared to the diagnostic accuracy of the AFP-IgM test.
