Abstract Background The congenital ventricular outflow tract malformations (CVOTMs) is a major congenital heart diseases (CHDs) subtype, and its pathogenesis is complex and unclear. Lipid metabolic plays a crucial role in embryonic cardiovascular development. However, due to the limited types of detectable metabolites in previous studies, findings on lipid metabolic and CHDs are still inconsistent, and the possible mechanism of CHDs remains unclear. Methods The nest case-control study obtained subjects from the multicenter China Teratology Birth Cohort (CTBC), and maternal serum from the pregnant women enrolled during the first trimester was utilized. The subjects were divided into a discovery set and a validation set. The metabolomics of CVOTMs and normal fetuses were analyzed by targeted lipid metabolomics. Differential comparison, random forest and lasso regression were used to screen metabolic biomarkers. Results The lipid metabolites were distributed differentially between the cases and controls. Feature extraction and classification is a difficult area in motor imagery electroencephalogram (EEG) signal processing. In order to improve the classification accuracy of EEG signals, both a feature extraction method based on the combination of LMD-CSP and a classification algorithm based on the fusion of PSO-SVM are proposed. Firstly, the extended informax ICA algorithm is used to denoise the signal and reduce the influence of noise on the signal. Then, the pre-processed EEG signals are decomposed into multiple Product Function (PF) components by Local Mean Decomposition (LMD), and the most discriminative PF component is selected. Next, feature extraction is carried out from the selected PF components using the Common Space Pattern (CSP). Finally, the obtained features are input into a Support Vector Machine (SVM) classifier improved by Particle Swarm Optimization (PSO) for classification recognition. The experimental results show that compared with the traditional CSP-SVM method, CapsNet method, WPT-CSP+CNN method, FDCSP-SVM method, and EMD-CNN method, the classification accuracy of the proposed method is increased by 26.94 %, 14.9 %, 13.34 %, 8.34 % and 4.04 %, respectively. This further proves the superiority of the proposed method for EEG signal processing technology.

Abstract Whether the anesthesia technique, inhalational general anesthesia (IGA) or propofol-based anesthesia (PBA), influences the long-term survival of non-metastatic breast cancer (eBC) remain unclear and controversial. We carried out a literature search on 16thJuly, 2022 for studies comparing IGA and PBA in eBC undergoing standard surgery, according to PRISMA 2020. The major endpoint in our study was overall survival (OS). Seventeen studies including four randomized clinical trials and thirteen retrospective cohort studies were included in the meta-analysis. Ten studies provided data for crude OS in unweighted eBC patients (imbalance in baseline characteristics). The summarized estimate HRs of the PBA group versus the IGA group (ten studies, N = 127,774, IGA group: 92,592, PBA group: 35,182.) was 0.83 (95%CI: 0.78–0.89). Compared with IGA, PBA was associated with both better 1-year OS (two studies, N = 104,083, IGA group: 84,074, PBA group: 20,009. Pooled HR = 0.80, 0.73–0.89) and 5-year OS (six studies, N = 121,580, IGA group: 89,472, PBA group: 32,108. HR = 0.80, 0.74–0.87). Ten studies applied PSM method to balance the baseline characteristics. In these weighted patients, PBA still showed a better OS (ten studies, N = 105,459, IGA group: 79,095, PBA group: 26,364. HR = 0.93, 0.87–1.00), a better 1-year OS (two studies, N = 83,007, IGA group: 67,609, PBA group: 15,398. HR = 0.88, 0.78–0.98) and a trend towards a better 5-year OS (nine studies, N = 121,580, IGA group: 76,797, PBA group: 24,066. HR = 0.95, 0.88–1.03). Loco-regional recurrence-free survival (LRRFS) was also better in PBA group (HR = 0.73, 0.61–0.86). The present study is the first comprehensive meta-analysis to demonstrate that propofol-based anesthesia could significantly improve OS and LRRFS in non-metastatic breast cancer patients, compared with inhalational anesthesia.

Insufficient therapeutic strategies for acute kidney injury (AKI) necessitate precision therapy targeting its pathogenesis. This study reveals the new mechanism of the marine-derived anti-AKI agent, piericidin glycoside S14, targeting peroxiredoxin 1 (PRDX1). By binding to Cys83 of PRDX1 and augmenting its peroxidase activity, S14 alleviates kidney injury efficiently in Prdx1-overexpression (Prdx1-OE) mice. Besides, S14 also increases PRDX1 nuclear translocation and directly activates the Nrf2/HO-1/NQO1 pathway to inhibit ROS production. Due to the limited druggability of S14 with low bioavailability (2.6%) and poor renal distribution, a pH-sensitive kidney-targeting dodecanamine-chitosan nanoparticle system is constructed to load S14 for precise treatment of AKI. l-Serine conjugation to chitosan imparts specificity to kidney injury molecule-1 (Kim-1)-overexpressed cells. The developed S14-nanodrug exhibits higher therapeutic efficiency by improving the in vivo behavior of S14 significantly. By encapsulation with micelles, the AUC0‒t, half-life time, and renal distribution of S14 increase 2.5-, 1.8-, and 3.1-fold, respectively. The main factors contributing to the improved druggability of S14 nanodrugs include the lower metabolic elimination rate and UDP-glycosyltransferase (UGT)-mediated biotransformation. In summary, this study identifies a new therapeutic target for the marine-derived anti-AKI agent while enhancing its ADME properties and druggability through nanotechnology, thereby driving advancements in marine drug development for AKI.

In the era of Education 4.0, the development of smart schools represents a significant step forward. Learning spaces are critical to the success of smart school construction in primary and secondary schools, due to their significant impact on school-age children. This study focuses on identifying key spatial factors that influence students’ psychological preferences and learning utility in smart learning spaces. To achieve this, a visual stated preference method was employed to design a questionnaire, which was used to survey primary and secondary school students in Harbin, China. The resulting data was analyzed using a discrete choice model, which enabled the measurement of the utility levels of different learning space factors. The results show that students’ learning space preferences are significantly influenced by a variety of spatial factors, with the combination mode of classrooms and outdoors having the highest level of influence, followed by a completely open interface, U-shape seating layout, zoned color tone, and the combination mode of classrooms and corridors; the students surveyed show a significant negative preference for heteromorphic spaces compared to rectangular spaces. The discussion recommends the design of smart learning spaces around key spatial factors to enhance the utility of student learning.

Abstract Drug exposure during pregnancy lacks global fetal safety data. The maternal drug exposure birth cohort (DEBC) study, a prospective longitudinal investigation, aims to explore the correlation of maternal drug exposure during pregnancy with pregnancy outcomes, and establish a human biospecimen biobank. Here we describe the process of establishing DEBC and show that the drug exposure rate in the first trimester of pregnant women in DEBC (n = 112,986) is 30.70%. Among the drugs used, dydrogesterone and progesterone have the highest exposure rates, which are 11.97% and 10.82%, respectively. The overall incidence of adverse pregnancy outcomes is 13.49%. Dydrogesterone exposure during the first trimester is correlated with higher incidences of stillbirth, preterm birth, low birth weight, and birth defects, along with a lower incidence of miscarriage/abortion. Due to the limitations of this cohort study, causative conclusions cannot be drawn. Further follow-up and in-depth data analysis are planned for future studies.

Abstract N-Hydroxyapiosporamide (N-hydap), a marine product derived from a sponge-associated fungus, has shown promising inhibitory effects on small cell lung cancer (SCLC). However, there is limited understanding of its metabolic pathways and characteristics. This study explored the in vitro metabolic profiles of N-hydap in human recombinant cytochrome P450s (CYPs) and UDP-glucuronosyltransferases (UGTs), as well as human/rat/mice microsomes, and also the pharmacokinetic properties by HPLC–MS/MS. Additionally, the cocktail probe method was used to investigate the potential to create drug-drug interactions (DDIs). N-Hydap was metabolically unstable in various microsomes after 1 h, with about 50% and 70% of it being eliminated by CYPs and UGTs, respectively. UGT1A3 was the main enzyme involved in glucuronidation (over 80%), making glucuronide the primary metabolite. With a favorable bioavailability of 24.0%, N-hydap exhibited a higher distribution in the lungs (26.26%), accounting for its efficacy against SCLC. Administering N-hydap to mice at normal doses via gavage did not result in significant toxicity. Furthermore, N-hydap was found to affect the catalytic activity of drug metabolic enzymes (DMEs), particularly increasing the activity of UGT1A3, suggesting potential for DDIs. Understanding the metabolic pathways and properties of N-hydap should improve our knowledge of its drug efficacy, toxicity, and potential for DDIs. Graphical Abstract

Environment coatings are frequently considered, to improve the corrosion resistance of silicon carbide (SiC) in pressurized water reactor (PWR) and boiling water reactor (BWR) surroundings. Chromium (Cr) is one of the main candidate materials for coatings on zirconium-based alloys for accident-tolerant fuel (AFT) cladding. In this work, the magnetron sputtered Cr coatings on SiC substrates were irradiated by the 500 keV He+ ions at room temperature. Moreover, the microstructural characterization was conducted on the irradiated specimens by using scanning electron microscope (SEM), and transmission electron microscope (TEM). The scratch test showed that the coating-substrate adhesion decreased from 41 N to 28 N (∼31.7%) before and after the irradiation, maybe due to that the irradiation produced tensile strain in the coating and mesophase along the coating-substrate interface. It revealed that irradiation accelerated the migration and aggregation of C elements on the surface of the coating. After the 800 °C annealing for 20 min, massive Cr atoms diffused into the SiC matrix and reacted with Cr to form a chromium silicide interlayer. Some cracks were observed at the interface, being attributed to the Kirkendall effect. Therefore, it is very necessary to avoid the formation of chromium silicide interlayer and decrease swelling mismatch between the metal coatings and the SiC substrate for the actual application in nuclear systems.

Glaucoma is a common and complex neurodegenerative disease characterized by progressive loss of retinal ganglion cells (RGCs) and axons. Currently, there is no effective method to address the cause of RGCs degeneration. However, studies on neuroprotective strategies for optic neuropathy have increased in recent years. Cell replacement and neuroprotection are major strategies for treating glaucoma and optic neuropathy. Regenerative medicine research into the repair of optic nerve damage using stem cells has received considerable attention. Stem cells possess the potential for multidirectional differentiation abilities and are capable of producing RGC-friendly microenvironments through paracrine effects. This article reviews a thorough researches of recent advances and approaches in stem cell repair of optic nerve injury, raising the controversies and unresolved issues surrounding the future of stem cells.

Abstract Background Synonymous variants are non‐pathogenic due to non‐substitution of amino acids. However, synonymous exonic terminal nucleotide substitutions may affect splicing. Splicing variants are easily analyzed at RNA level for genes expressed in blood cells. Minigene analysis provides another method for splicing variant analysis of genes that are poorly or not expressed in peripheral blood. Methods Whole exome sequencing was performed to screen for potential pathogenic mutations in the proband, which were validated within the family by Sanger sequencing. The pathogenicity of the synonymous mutation was analyzed using the minigene technology. Results The proband harbored the compound heterogeneous variants c. [291G >A; 572‐50C >T] and c.681 + 1G >T in F7, of which the synonymous variant c.291G >A was located at the terminal position of exon 3. Minigene analysis revealed exon3 skipping due to this mutation, which may have subsequently affected protein sequence, structure, and function. Conclusion Our finding confirmed the pathogenicity of c.291G >A, thus extending the pathogenic mutation spectrum of F7, and providing insights for effective reproductive counseling.

Precise prediction of irrigation volumes is crucial in modern agriculture. This study proposes an optimized long short-term memory (LSTM) model-based irrigation prediction method that combines bidirectional LSTM networks. The model provides farmers with more precise irrigation management decisions, facilitating optimal utilization of water resources and effective crop production management. This proposed model aims to fully exploit spatio-temporal features and sequence dependencies to enhance prediction accuracy and reliability. We aim to fully leverage crop irrigation volumes’ spatio-temporal features and sequence dependencies to improve prediction accuracy and reliability. First, this study adopts a bidirectional LSTM (BiLSTM) model to simulate the temporal features of irrigation volumes and learn the sequential dependencies of crop growth data from historical records. Then, this study passes the irrigation volume data through a convolutional neural network (CNN) model to extract spatial features and capture correlations among various features such as temperature, precipitation, and wind speed. Our prediction performance significantly improved after incorporating an attention mechanism that involves weighting features and enhancing focus on crucial aspects. The proposed BiLSTM-CNN-Attention approach is used to predict irrigation volume for spring corn in significant irrigation areas in Jilin Province, China. The results demonstrate that the proposed method surpasses recurrent neural network (RNN), CNN, LSTM, BiLSTM, and BiLSTM-CNN methods in terms of mean square error (MSE), root mean square error (RMSE), mean absolute error (MAE) (0.000004, 0.005968, 0.004599), and R2 (0.9749), making a superior solution for predicting the volume of crop irrigation.

Abstract Background The majority of congenital heart diseases (CHDs) are thought to result from the interactions of genetics and the environment factors. This study aimed to assess the association of maternal non-occupational phthalates exposure, metabolic gene polymorphisms and their interactions with risk of CHDs in offspring. Methods A multicenter case-control study of 245 mothers with CHDs infants and 268 control mothers of health infant was conducted from six hospitals. Maternal urinary concentrations of eight phthalate metabolites were measured by ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UHPLC-MS/MS). Twenty single nucleotide polymorphisms (SNPs) in cytochrome P450 family 2 subfamily C member 9 (CYP2C9) and 19 (CYP2C19), uridine diphosphate (UDP) glucuronosyl transferase family 1 member A7 (UGT1A7), family 2 member B7 (UGT2B7) and B15(UGT2B15) genes were genotyped. The multivariate logistic regressions were used to estimate the association between maternal phthalates exposure or gene polymorphisms and risk of CHDs. Generalized multifactor dimensionality reduction (GMDR) was used to analyze the gene–gene and gene–phthalates exposure interactions. Results There was no significant difference in phthalate metabolites concentrations between the cases and controls. No significant positive associations were observed between maternal exposure to phthalates and CHDs. The SNPs of UGT1A7 gene at rs4124874 (under three models, log-additive: aOR = 1.74, 95% CI:1.28–2.37; dominant: aOR = 1.86, 95% CI:1.25–2.78; recessive: aOR = 2.50, 95% CI: 1.26–4.94) and rs887829 (under the recessive model: aOR = 13.66, 95% CI: 1.54–121) were significantly associated with an increased risk of CHDs. Furthermore, the associations between rs4124874 (under log-additive and dominant models) of UGT1A7 were statistically significant after the false discovery rate correction. No significant gene-gene or gene-phthalate metabolites interactions were observed. Conclusions The polymorphisms of maternal UGT1A7 gene at rs4124874 and rs887829 were significantly associated with an increased risk of CHDs. More large-scale studies or prospective study designs are needed to confirm or refute our findings in the future.

Sheet music recognition is a vital technology aimed at converting printed or handwritten musical scores into digital or machine-readable formats. The significance of this technology lies in making music compositions more accessible for editing, performance, learning, and sharing, thereby fostering music education, composition, and culture. It also provides a powerful tool for music analysis, research, and preservation. Our aim is to investigate a sheet music recognition method that offers a simple workflow, high recognition accuracy, and fast model convergence. Specifically, the proposed Deep Multilevel Cascade Residual Recurrent (MCRR) framework for sheet music recognition consists of the following components. Firstly, we introduce additive Gaussian white noise, additive Perlin noise, and elastic deformations such as rotation and stretching to simulate real-world noise in the sheet music images, thereby augmenting the dataset, enhancing model robustness, and mitigating overfitting. Secondly, in the feature extraction phase, we employ a residual Convolutional Neural Network (ConvNet) to address the issue of model degradation and use the multilevel cascade fusion technique to obtain comprehensive feature information, improving the model’s feature extraction capability and reducing recognition errors. For note recognition, we use a variant of RNN (Recurrent Neural Network) called SRU (Simple Recurrent Unit), which transforms most computations into parallel processing, speeding up model convergence. Finally, we combine the Connectionist Temporal Classification (CTC) loss function with SRU to eliminate the requirement for strict alignment between data and labels, enabling note classification and recognition. Extensive ablation experiments and comparative analyses, including visual analysis, intuitive illustrations, and quantitative assessments, confirm the effectiveness of the proposed method, demonstrating its superiority over various state-of-the-art methods. The proposed method achieved promising results in both the PrIMus and Camera-PrIMuS datasets. Specifically, in the PrIMus dataset, the method obtained an SeER (Symbol Error Rate) of 1.4571% and a SyER (System Error Rate) of 0.3234%. Notably, it demonstrated high accuracy in pitch, type, and note recognition, scoring approximately 97% in pitch and type accuracy and around 94% in note accuracy. The training time per epoch was relatively low, recorded at 0.56 seconds. In the case of the Camera-PrIMuS dataset, the method achieved slightly lower but still competitive results. It exhibited an SeER of 5.1488% and a SyER of 1.0612%, with pitch and type accuracies around 90%, and note accuracy at approximately 88%. The training time per epoch was slightly higher at 1.93 seconds Furthermore, we compare our method with existing commercial software, namely Capella-scan, PhotoScore, and SmartScore. Among these, Capella-scan delivers the best performance but exhibits lower robustness compared to the proposed method.

Abstract Emotional and mood disturbances are common in people with dementia. Non-pharmacological interventions are beneficial for managing these disturbances. However, effectively applying these interventions, particularly in the person-centred approach, is a complex and knowledge-intensive task. Healthcare professionals need the assistance of tools to obtain all relevant information that is often buried in a vast amount of clinical data to form a holistic understanding of the person for successfully applying non-pharmacological interventions. A machine-readable knowledge model, e.g., ontology, can codify the research evidence to underpin these tools. For the first time, this study aims to develop an ontology entitled Dementia-Related Emotional And Mood Disturbance Non-Pharmacological Treatment Ontology (DREAMDNPTO). DREAMDNPTO consists of 1258 unique classes (concepts) and 70 object properties that represent relationships between these classes. It meets the requirements and quality standards for biomedical ontology. As DREAMDNPTO provides a computerisable semantic representation of knowledge specific to non-pharmacological treatment for emotional and mood disturbances in dementia, it will facilitate the application of machine learning to this particular and important health domain of emotional and mood disturbance management for people with dementia.

Abstract Background Pancreatic neuroendocrine neoplasms (pNENs) are relatively rare. Hypoxia and lipid metabolism-related gene acetyl-CoA synthetase 2 (ACSS2) is involved in tumor progression, but its role in pNENs is not revealed. This study showed that hypoxia can upregulate ACSS2, which plays an important role in the occurrence and development of pNENs through lipid metabolism reprogramming. However, the precise role and mechanisms of ACSS2 in pNENs remain unknown. Methods mRNA and protein levels of ACSS2 and 3-hydroxy-3-methylglutaryl-CoA synthase1 (HMGCS1) were detected using quantitative real-time PCR (qRT-PCR) and Western blotting (WB). The effects of ACSS2 and HMGCS1 on cell proliferation were examined using CCK-8, colony formation assay and EdU assay, and their effects on cell migration and invasion were examined using transwell assay. The interaction between ACSS2 and HMGCS1 was verified by Co-immunoprecipitation (Co-IP) experiments, and the functions of ACSS2 and HMGCS1 in vivo were determined by nude mouse xenografts. Results We demonstrated that hypoxia can upregulate ACSS2 while hypoxia also promoted the progression of pNENs. ACSS2 was significantly upregulated in pNENs, and overexpression of ACSS2 promoted the progression of pNENs and knockdown of ACSS2 and ACSS2 inhibitor (ACSS2i) treatment inhibited the progression of pNENs. ACSS2 regulated lipid reprogramming and the PI3K/AKT/mTOR pathway in pNENs, and ACSS2 regulated lipid metabolism reprogramming through the PI3K/AKT/mTOR pathway. Co-IP experiments indicated that HMGCS1 interacted with ACSS2 in pNENs. Overexpression of HMGCS1 can reverse the enhanced lipid metabolism reprogramming and tumor-promoting effects of knockdown of ACSS2. Moreover, overexpression of HMGCS1 reversed the inhibitory effect of knockdown of ACSS2 on the PI3K/AKT/mTOR pathway. Conclusion Our study revealed that hypoxia can upregulate the lipid metabolism-related gene ACSS2, which plays a tumorigenic effect by regulating lipid metabolism through activating the PI3K/AKT/mTOR pathway. In addition, HMGCS1 can reverse the oncogenic effects of ACSS2, providing a new option for therapeutic strategy.

Abstract Background Patients with hematological malignancies received multiple hypodermic injections of recombinant human granulocyte colony-stimulating factor. Procedural pain is one of the most common iatrogenic causes of pain in patients with hematological malignancies. It is also identified as the most commonly occurring problem in clinical care in the Department of Hematology and Oncology at Shenzhen University General Hospital. However, providing immediate relief from pain induced by hypodermic injection of recombinant human granulocyte colony-stimulating factor remains a major challenge. This trial aims to evaluate the safety and analgesic efficacy of a fixed nitrous oxide/oxygen mixture for patients with hematological malignancies and experiencing procedural pain caused by hypodermic injection of recombinant human granulocyte colony-stimulating factor in the department. Methods The nitrous oxide/oxygen study is a single-center, randomized, double-blind, placebo-controlled trial involving patients with hematological malignancies who require hypodermic injections of recombinant human granulocyte colony-stimulating factor for treatment. This trial was conducted in the Hematology and Oncology Department of Shenzhen University General Hospital. A total of 54 eligible patients were randomly allocated to either the fixed nitrous oxide/oxygen mixture group (n = 36) or the oxygen group (n = 18). Neither the investigators nor the patients known about the randomization list and the nature of the gas mixture in each cylinder. Outcomes were monitored at the baseline (T0), immediately after hypodermic injection of recombinant human granulocyte colony-stimulating factor (T1), and 5 min after hypodermic injection of recombinant human granulocyte colony-stimulating factor (T2) for each group. The primary outcome measure was the score in the numerical rating scale corresponding to the highest level of pain experienced during hypodermic injection of recombinant human granulocyte colony-stimulating factor. Secondary outcomes included the fear of pain, anxiety score, four physiological parameters, adverse effects, total time of gas administration, satisfaction from both patients and nurses, and the acceptance of the patients. Discussion This study focused on the safety and analgesic efficacy during hypodermic injection of recombinant human granulocyte colony-stimulating factor procedure. Data on the feasibility and safety of nitrous oxide/oxygen therapy was provided if proven beneficial to patients with hematological malignancies during hypodermic injection of recombinant human granulocyte colony-stimulating factor and widely administered to patients with procedural pain in the department. AbstractChrysoglossum ornatum Blume, the type species of Chrysoglossum Blume, belongs to the tribe Collabieae of the subfamily Epidendroideae of Orchidaceae. In this study, we sequenced, assembled, and analyzed the complete chloroplast genome of C. ornatum. The result showed that the complete chloroplast genome of C. ornatum was 158,175 bp in size, consisting of a large single-copy (LSC) region of 87,235 bp, a small single-copy (SSC) region of 18,384 bp, and a pair of inverted repeats (IRs) of 26,278 bp. The chloroplast genome encoded 113 unique genes, comprising 80 protein-coding genes, 29 tRNA genes, and four rRNA genes. Phylogenetic analysis inferred from the complete chloroplast genome indicated that Chrysoglossum was closely related to Collabium Blume. This study provides genomic resources helpful for further phylogenetic and biodiversity research on Chrysoglossum.

Alkaline hydrogen evolution reaction (HER) is suppressed by the water dissociation, leading to more sluggish kinetics than acidic HER. Developing multifunction catalysts via constructing heterogeneous interfaces is a feasible tactic to accelerate the alkaline HER. Herein, NiO coupled with Ni and MoxN (NMN) nanorods were prepared via a hydro-thermal synthesis combined with a thermal decomposition under ammonia atmosphere. The low crystalline NMN nanorods are rich in heterointerfaces, and have sufficient high active sites for HER. The synergistic effect between NiO and Ni-MoxN promotes the water dissociation the hydrogen adsorption, and the charge transfer, contributing to excellent alkaline HER activity. The overpotential on NMN is only 36 and 150 mV for the current density of 10 and 300 mA cm−2, respectively, and the Tafel slope is 48 mV/dec, demonstrating a superior performance for alkaline HER, which is even comparable to the commercial electrocatalysts.

The temporal component of episodic memory has been recognized as a sensitive behavioral marker in early stage of Alzheimer's disease (AD) patients. However, parallel studies in AD animals are currently lacking, and the underlying neural circuit mechanisms remain poorly understood. Using a novel AppNL-G-F knock-in (APP-KI) rat model, the developmental changes of temporal order memory (TOM) and the relationship with medial prefrontal cortex and perirhinal cortex (mPFC-PRH) circuit were determined through in vivo electrophysiology and microimaging technique. We observed a deficit in TOM performance during the object temporal order memory task (OTOMT) in APP-KI rats at 6 month old, which was not evident at 3 or 4 months of age. Alongside behavioral changes, we identified a gradually extensive and aggravated regional activation and functional alterations in the mPFC and PRH during the performance of OTOMT, which occurred prior to the onset of TOM deficits. Moreover, coherence analysis showed that the functional connectivity between the mPFC and PRH could predict the extent of future behavioral performance. Further analysis revealed that the aberrant mPFC-PRH interaction mainly attributed to the progressive deterioration of synaptic transmission, information flow and network coordination from mPFC to PRH, suggesting the mPFC dysfunction maybe the key area of origin underlying the early changes of TOM. These findings identify a pivotal role of the mPFC-PRH circuit in mediating the TOM deficits in the early stage of AD, which holds promising clinical translational value and offers potential early biological markers for predicting AD memory progression.

Corneal transplantation is a common treatment for corneal diseases. Secondary glaucoma after corneal transplantation is the second leading cause of failure of keratoplasty. This article reviews the mechanism and treatment of secondary glaucoma after corneal transplantation.

Background: This study aimed to investigate the effects of the combination of Epimedii Folium (EF) and Ligustri Lucidi Fructus (LLF) on regulating apoptosis and autophagy in senile osteoporosis (SOP) rats. Methods: Firstly, we identified the components in the decoction and drug-containing serum of EL (EF&LLF) by Ultra performance liquid chromatography-quadrupole-time of flight-mass spectrometry (UPLC-Q-TOF-MS). Secondly, SOP rats were treated with EF, LLF, EL and caltrate to evaluate the advantages of EL. Finally, H2O2-, chloroquine-, and MHY1485-induced osteoblasts were treated with different doses of EL to reveal the molecular mechanism of EL. We detected bone microstructure, oxidative stress levels, ALP activity and the expressions of Bax, Bcl-2, caspase3, P53, Beclin-1, p-PI3K, PI3K, p-Akt, Akt, p-mTOR, mTOR, and LC3 in vivo and in vitro. Results: 36 compounds in EL decoction and 23 in EL-containing serum were identified, including flavonoids, iridoid terpenoids, phenylethanoid glycosides, polyols and triterpenoids. EL could inhibit apoptosis activity and increase ALP activity. In SOP rats and chloroquine-inhibited osteoblasts, EL could improve bone tissue microstructure and osteoblasts functions by upregulating Bcl-2, Beclin1, and LC3-II/LC3-I, while downregulating p53 in all treatment groups. In H2O2-induced osteoblasts, EL could upregulate the protein and mRNA expressions of Bcl-2 while downregulate LC3-II/LC3-I, p53 and Beclin1. Besides, EL was able to down-regulate PI3K/AKT/mTOR pathway which activated in SOP rats and MHY1485-induced osteoblasts. Conclusions: These findings demonstrate that EL with bone protective effects on SOP rats by regulating autophagy and apoptosis via PI3K/Akt/mTOR signaling pathway, which might be an alternative medicine for the treatment of SOP.

Hepatosteatosis is characterized by abnormal accumulation of triglycerides (TG), leading to prolonged and chronic inflammatory infiltration. To date, there is still a lack of effective and economical therapies for hepatosteatosis. Oridonin (ORI) is a major bioactive component extracted from the traditional Chinese medicinal herb Rabdosia rubescens. In this paper, we showed that ORI exerted significant protective effects against hepatic steatosis, inflammation and fibrosis, which was dependent on LXRα signaling. It is reported that LXRα regulated lipid homeostasis between triglyceride (TG) and phosphatidylethanolamine (PE) by promoting ATGL and EPT1 expression. Therefore, we implemented the lipidomic strategy and luciferase reporter assay to verify that ORI contributed to the homeostasis of lipids via the regulation of the ATGL gene associated with TG hydrolysis and the EPT1 gene related to PE synthesis in a LXRα-dependent manner, and the results showed the TG reduction and PE elevation. In detail, hepatic TG overload and lipotoxicity were reversed after ORI treatment by modulating the ATGL and EPT1 genes, respectively. Taken together, the data provide mechanistic insights to explain the bioactivity of ORI in attenuating TG accumulation and cytotoxicity and introduce exciting opportunities for developing novel natural activators of the LXRα-ATGL/EPT1 axis for pharmacologically treating hepatosteatosis and metabolic disorders.

Abstract The process of post-transcriptional regulation has been recognized to be significantly impacted by the presence of N6-methyladenosine (m6A) modification. As an m6A demethylase, ALKBH5 has been shown to contribute to the progression of different cancers by increasing expression of several oncogenes. Hence, a better understanding of the key targets of ALKBH5 in cancer cells could potentially lead to the development of new therapeutic targets. However, the specific role of ALKBH5 in pancreatic neuroendocrine neoplasms (pNENs) remains largely unknown. Here, we demonstrated that ALKBH5 was up-regulated in pNENs and played a critical role in tumor growth and lipid metabolism. Mechanistically, ALKBH5 over-expression was found to increase the expression of FABP5 in an m6A-IGF2BP2 dependent manner, leading to disorders in lipid metabolism. Additionally, ALKBH5 was found to activate PI3K/Akt/mTOR signaling pathway, resulting in enhanced lipid metabolism and proliferation abilities. In conclusion, our study uncovers the ALKBH5/IGF2BP2/FABP5/mTOR axis as a mechanism for aberrant m6A modification in lipid metabolism and highlights a new molecular basis for the development of therapeutic strategies for pNENs treatment. WO3 and silicone rubber (SR)-based gasochromic composites were fabricated to detect hydrogen leaks at room temperature. WO3 rod-like nanostructures were uniformly distributed in the SR matrix, with a particle size of 60–100 nm. The hydrogen permeability of these composites reached 1.77 cm3·cm/cm2·s·cmHg. At a 10% hydrogen concentration, the visible light reflectance of the composite decreased 49% during about 40 s, with a color change rate of 6.4% s−1. Moreover, the composite detected hydrogen concentrations as low as 0.1%. And a color scale was obtained for easily assessing hydrogen concentrations in the environment based on the color of composites. Finally, the composite materials as disposable sensors underwent testing at several Sinopec hydrogen refueling stations.

We developed a novel site-specific bimodal MRI/fluorescence nanoparticle contrast agent targeting gastrin-releasing peptide receptors (GRPrs), which are overexpressed in aggressive prostate cancers. Biocompatible ultra-small superparamagnetic iron oxide (USPIO) nanoparticles were synthesized using glucose and casein coatings, followed by conjugation with a Cy7.5-K-8AOC-BBN [7-14] peptide conjugate. The resulting USPIO(Cy7.5)-BBN nanoparticles were purified by 100 kDa membrane dialysis and fully characterized using transmission electron microscopy (TEM), dynamic light scattering (DLS), Fourier transform infrared (FTIR) spectroscopy, and magnetic resonance imaging (MRI) relaxivity, as well as evaluated for in vitro and in vivo binding specificity and imaging efficacy in PC-3 prostate cancer cells and xenografted tumor-bearing mice. The USPIO(Cy7.5)-BBN nanoparticles had a core diameter of 4.93 ± 0.31 nm and a hydrodynamic diameter of 35.56 ± 0.58 nm. The r2 relaxivity was measured to be 70.2 ± 2.5 s−1 mM−1 at 7T MRI. The Cy7.5-K-8AOC-BBN [7-14] peptide-to-nanoparticle ratio was determined to be 21:1. The in vitro GRPr inhibitory binding (IC50) value was 2.5 ± 0.7 nM, indicating a very high binding affinity of USPIO(Cy7.5)-BBN to the GRPr on PC-3 cells. In vivo MRI showed significant tumor-to-muscle contrast enhancement in the uptake group at 4 h (31.1 ± 3.4%) and 24 h (25.7 ± 2.1%) post-injection compared to the blocking group (4 h: 15.3 ± 2.0% and 24 h: −2.8 ± 6.8%; p < 0.005). In vivo and ex vivo near-infrared fluorescence (NIRF) imaging revealed significantly increased fluorescence in tumors in the uptake group compared to the blocking group. These findings demonstrate the high specificity of bimodal USPIO(Cy7.5)-BBN nanoparticles towards GRPr-expressing PC-3 cells, suggesting their potential for targeted imaging in aggressive prostate cancer.

The three-dimension (3D) inversion of magnetic data is an effective method of recovering underground magnetic susceptibility distributions using magnetic anomaly data. The conventional regularization inversion method has good data fitting; however, its inversion model has the problem of a poor model-fitting ability due to a low depth resolution. The 3D inversion method based on deep learning can effectively improve the model-fitting accuracy, but it is difficult to guarantee the data-fitting accuracy of the inversion results. The loss function of traditional deep learning 3D inversion methods usually adopts the metric of the absolute mean squared error (MSE). In order to improve the accuracy of the data fitting, we added a forward-fitting constraint term (FFit) on the basis of the MSE. Meanwhile, in order to further improve the accuracy of the model fitting, we added the Dice coefficient to the loss function. Finally, we proposed a multi-constraint deep learning 3D inversion method based on UNet++. Compared with the traditional single-constraint deep learning method, the multi-constraint deep learning method has better data-fitting and model-fitting effects. Then, we designed corresponding test models and evaluation metrics to test the effectiveness and feasibility of the method, and applied it to the actual aeromagnetic data of a test area in Suqian City, Jiangsu Province.

Cinnamomum camphora seed kernels (CCSKs) are rich in phytochemicals, especially plant extracts. Phytochemicals play a vital role in therapy due to their strong antioxidant and anti-inflammatory activities. Extracts from CCSK can be obtained through multiple steps, including pretreatment, extraction and purification, and the purpose of pretreatment is to separate the oil from other substances in CCSKs. However, C. camphora seed kernel extracts (CKEs) were usually considered as by-products and discarded, and their potential bioactive values were underestimated. Additionally, little has been known about the effect of pretreatment on CKE. This study aimed to investigate the effects of pretreatment methods (including the solvent extraction method, cold pressing method, aqueous extraction method and sub-critical fluid extraction method) on the extraction yields, phytochemical profiles, volatile compounds and antioxidant capacities of different CKE samples. The results showed that the CKE samples were rich in phenolic compounds (15.28–20.29%) and alkaloids (24.44–27.41%). The extraction yield, bioactive substances content and in vitro antioxidant capacity of CKE pretreated by the sub-critical fluid extraction method (CKE-SCFE) were better than CKEs obtained by other methods. CKE pretreated by the solvent extraction method (CKE-SE) showed the best lipid emulsion protective capacity. Moreover, the volatile substances composition of the CKE samples was greatly influenced by the pretreatment method. The results provided a fundamental basis for evaluating the quality and nutritional value of CKE and increasing the economic value of by-products derived from CCSK.

Increasing environmental and economic concerns necessitate the research for peat moss alternatives, aiming to balance ecological sustainability with cost-effectiveness. This study assessed whether biochar (BC) and hydrafiber (HF) could be a partial replacement for peat moss as substrate components. Twelve substrates were formulated by either mixing BC (20%, 40%, and 60%, by vol.) with HF (20%, 40%, and 60%, by vol.), with the remaining being peat moss or mixing BC (0%, 20%, 40%, and 60%, by vol.) with the commercial substrates (CS) to grow zinnia (Zinnia elegans) and snapdragon (Antirrhinum majus) plants in containers. The physical properties of the substrates, including container capacity, total porosity, air space, bulk density, and chemical properties including leachate pH and electrical conductivity (EC) were measured. Plant growth parameters including growth index (GI) and leaf greenness (indicated with SPAD), biomass, and number of flowers were measured biweekly. The results showed all the substrate mixes had similar air space, bulk density, and SPAD. Treatment with 20% BC and 80% CS yielded the highest GI, biomass, and numbers of flowers in both zinnia and snapdragon. In conclusion, BC could be used to partially (20%) replace commercial substrate mix for container-grown zinnia and snapdragon.

Introduction. Breast cancer management is complex, requiring personalised care from multidisciplinary teams. Research shows that there is unwarranted clinical variation in mastectomy rates between rural and metropolitan patients; that is, variation in treatment which cannot be explained by disease progression or medical necessity. This study aims to determine the clinical and nonclinical factors contributing to any unwarranted variation in breast cancer management in rural patients and to evaluate how these factors and variations relate to patient outcomes. Methods. Comprehensive data from patients who had primary breast cancer surgery from 2010 to 2014 in either a rural or metropolitan location in a single local health district was analysed (n = 686). Records were subset into two rurality groupings based on the postcode in which the patient resided, and the Modified Monash Model (MMM), an Australian system for classifying rurality. Statistical analysis was used to compare rural and metropolitan cohorts on treatments, patient characteristics, timeliness, and outcomes (recurrence and survival). Results. Objective. To evaluate the influence of screw-tightening methods on the immediate and long-term stability of dental implant screw joints. Methodology. A total of 150 implants of three different implant systems with different diameters were used in this study. Each group was divided into three subgroups (n = 5), according to the tightening methods (A—tightening with recommended torque and retorque after 10 min; B—tightening with recommended torque, then loosening and immediate retorque; C—tightening with recommended torque only once). The operating time of tightening the assemblies was recorded. Ten minutes later, the immediate removal torque (IRT) (Ncm) was measured. After retightening the assemblies, a dynamic load between 20 and 200 N was applied for 105 cycles, and the postloading removal torque (PRT) (Ncm) was measured. Scanning electron microscopy (SEM) was used to observe the surface topography of the screws. Results. For different types of implants, the IRTs were 11.92 ± 1.04–34.12 ± 0.36 Ncm for method A, 11.64 ± 0.57–33.96 ± 0.29 Ncm for method B, and 10.30 ± 0.41–31.62 ± 0.52 Ncm for method C, and the IRTs of methods A and B were 6.28%–21.58% higher than that of method C (P≤0.046). The PRTs were 4.08 ± 0.77–29.86 ± 0.65 Ncm for method A, 4.04 ± 0.40–29.60 ± 0.36 Ncm for method B, and 2.98 ± 0.26–26.38 ± 0.59 Ncm for method C, and the PRTs of methods A and B were 11.77%–44.87% higher than that of method C (P≤0.016). BackgroundHypertension is a significant global disease burden. Mobile health (mHealth) offers a promising means to provide patients with hypertension with easy access to health care services. Yet, its efficacy needs to be validated, especially in lower-income areas with a high-salt diet. ObjectiveThis study aims to assess the efficacy of an mHealth app–based intervention in supporting patients’ self-management of hypertension. MethodsA 2-arm randomized controlled trial was conducted among 297 patients with hypertension at the General Hospital of Ningxia Medical University, Ningxia Hui Autonomous Region, China. Participants selected via convenience sampling were randomly allocated into intervention and control groups. Intervention group participants were trained and asked to use an mHealth app named Blood Pressure Assistant for 6 months. They could use the app to record and upload vital signs, access educational materials, and receive self-management reminders and feedback from health care providers based on the analysis of the uploaded data. Control group participants received usual care. To explore the characteristics, influencing factors, and effect of different treatments on the survival in patients with first primary cervical cancer (CC) and second primary CC. Data of 33,934 eligible patients with CC were extracted from the Surveillance, Epidemiology, and End Results (SEER) database in 2004–2015. We also included 176 patients with CC from the Affiliated Dongyang Hospital of Wenzhou Medical University. Univariate and multivariate Cox proportional hazard models were used to screen the potential influencing factors associated with the survival in patients with hazard ratios (HRs) and 95 % confidence intervals (CIs). Subgroup analyses of age, American Joint Committee on Cancer (AJCC) stages, tumor grades and histologic types were conducted to explore the association between different treatments and survival in different populations. The 5-year mortality was 43.08 % for patients with first primary CC and that was 58.13 % for patients with second primary CC. Abstract Background Amputees suffer from symptomatic neuroma and phantom limb pain. Regenerative peripheral nerve interface (RPNI) has recently been regarded as an effective method to prevent neuroma after amputation. However, the verifications of RPNI efficacy are mostly based on subjective evaluation, lacking objective approaches. This study aims to unveil the effect of RPNI on preventing neuroma formation and provide evidence supporting the efficacy of RPNI based on ultrasound. Methods Amputees of lower limb at Peking University People’s Hospital from July 2020 to March 2022 were analyzed retrospectively. The clinical data collected consisted of general information, pathology of primary disease, history of limb-salvage treatment, amputation level of nerve, pain scales such as the Numerical Rating Scale (NRS) and the Manchester Foot Pain and Disability Index (MFPDI). Three months after amputation, the transverse diameter, anteroposterior diameter, and cross-sectional area of neuromas in stump nerves at the end of residual limbs were measured using ultrasound and compared to adjacent normal nerves. Results Fourteen patients were enrolled in the study, including 7 in the traditional amputation group (TA group) and 7 in the RPNI group. There was no significant difference in basic information and amputation sites between the two groups. The NRS and MFPDI scores of patients in RPNI group were significantly lower than those in TA group, and decreased with the follow-up time increasing, indicating that RPNI could reduce symptomatic neuroma pain. The comparison of preoperative ultrasound and postoperative pathology showed ultrasound could reflect the size of neuroma in vivo. Independent-sample t tests indicated that the ratios of anteroposterior diameter, transverse diameter and area of the cross section of both the neuroma and adjacent normal nerve obtained via ultrasound were significantly reduced in the RPNI group. Conclusion This study suggested that RPNI can effectively prevent the formation of symptomatic neuroma after amputation using ultrasound.

Abstract Background N6-methyladenosine (m6A) modification is the most abundant reversible methylation modification in eukaryotes, and it is reportedly closely associated with a variety of cancers progression, including colorectal cancer (CRC). This study showed that activated lipid metabolism and glycolysis play vital roles in the occurrence and development of CRC. However, only a few studies have reported the biological mechanisms underlying this connection. Methods Protein and mRNA levels of FTO and ALKBH5 were measured using western blot and qRT-PCR. The effects of FTO and ALKBH5 on cell proliferation were examined using CCK-8, colony formation, and EdU assays, and the effects on cell migration and invasion were tested using a transwell assay. m6A RNA immunoprecipitation (MeRIP) and RNA-seq was used to explore downstream target gene. RIP was performed to verify the interaction between m6A and HK2. The function of FTO and ALKBH5 in vivo was determined by xenograft in nude mice. Results In this study, FTO and ALKBH5 were significantly down-regulated in CRC patients and cells both in vivo and in vitro in a high-fat environment. Moreover, FTO and ALKBH5 over-expression hampered cell proliferation both in vitro and in vivo. Conversely, FTO and ALKBH5 knockdown accelerated the malignant biological behaviors of CRC cells. The mechanism of action of FTO and ALKBH5 involves joint regulation of HK2, a key enzyme in glycolysis, which was identified by RNA sequencing and MeRIP-seq. Furthermore, reduced expression of FTO and ALKBH5 jointly activated the FOXO signaling pathway, which led to enhanced proliferation ability in CRC cells. IGF2BP2, as a m6A reader, positively regulated HK2 mRNA in m6A dependent manner. Additionally, down-regulation of FTO/ALKBH5 increased METTL3 and decreased METTL14 levels, further promoting CRC progression. Conclusion In conclusion, our study revealed the FTO-ALKBH5/IGF2BP2/HK2/FOXO1 axis as a mechanism of aberrant m6A modification and glycolysis regulation in CRC.

Abstract Open-shell nanographenes appear as promising candidates for future applications in spintronics and quantum technologies. A critical aspect to realize this potential is to design and control the magnetic exchange. Here, we reveal the effects of frontier orbital symmetries on the magnetic coupling in diradical nanographenes through scanning probe microscope measurements and different levels of theoretical calculations. In these open-shell nanographenes, the exchange energy exhibits a remarkable variation between 20 and 160 meV. Theoretical calculations reveal that frontier orbital symmetries play a key role in affecting the magnetic coupling on such a large scale. Moreover, a triradical nanographene is demonstrated for investigating the magnetic interaction among three unpaired electrons with unequal magnetic exchange, in agreement with Heisenberg spin model calculations. Our results provide insights into both theoretical design and experimental realization of nanographene materials with different exchange interactions through tuning the orbital symmetry, potentially useful for realizing magnetically operable graphene-based nanomaterials.

Neottia bifidus, a new mycoheterotrophic orchid, found in Maolan National Nature Reserve in Guizhou Province, China, is described and illustrated here. The new species is close to N. nidus-avis, N. kiusiana and N. papilligera but differs in having a finely pubescent rachis with fewer flowers, a finely pubescent pedicel, and a fishtail-shaped lip that is deeply bilobed to the middle of the lip, with the lobes diverging at an acute angle (45°) to each other and mesochile with many papillae. Additionally, N. bifidus is well supported as a new species by molecular phylogenetic results based on ITS and chloroplast genome. The chloroplast genome of the novelty, which contains an LSC region of 33,819 bp, SSC region of 5,312 bp and IRs of 46,762 bp was assembled and annotated. A key to mycoheterotrophic Neottia species in China is also provided.

Abstract Increasing evidence indicates that long non-coding RNA (lncRNA) is one of the most important RNA regulators in the pathogenesis of neuroblastoma (NB). Here, we found that FAM201A was low expressed in NB and a variety of gain and loss of function studies elucidated the anti-tumor effects of FAM201A on the regulation of proliferation, migration and invasion of NB cells. Intriguingly, we identified the ability of FAM201A to encode the tumor-suppressing protein, NBASP, which interacted with FABP5 and negatively regulated its expression. In vivo assays also revealed NBASP repressed NB growth via inactivating MAPK pathway mediated by FABP5. In conclusion, our findings demonstrated that NBASP encoded by FAM201A played a tumor-suppressor role in NB carcinogenesis via down-regulating FABP5 to inactivate the MAPK pathway. These results extended our understanding of the relationship of lncRNA-encoded functional peptides and plasticity of tumor progression.

Abstract Background It has been manifested in several studies that age-related metabolic reprogramming is associated with tumor progression, in particular, colorectal cancer (CRC). Here we investigated the role of upregulated metabolites of the aged serum, including methylmalonic acid (MMA), phosphoenolpyruvate (PEP), and quinolinate (QA), in CRC. Methods Functional assays including CCK-8, EdU, colony formation and transwell experiments were used to ascertain which upregulated metabolite of elderly serum was related to tumor progression. RNA-seq analysis was conducted to explore the potential mechanisms of MMA-induced CRC progression. Subcutaneous tumorigenesis and metastatic tumor models were constructed to verify the function of MMA in vivo. Results Among three consistently increased metabolites of the aged sera, MMA was responsible for tumorigenesis and metastasis in CRC, according to functional assays. The promotion of Epithelial–mesenchymal transition (EMT) was observed in CRC cells treated with MMA, on the basis of protein expression of EMT markers. Moreover, combined with transcriptome sequencing, Wnt/β-catenin signaling pathway was activated in CRC cells treated with MMA, which was verified by western blot and qPCR experiments. Furthermore, animal assays demonstrated the pro-proliferation and promotion of metastasis role of MMA in vivo. Conclusion We have identified that age-dependent upregulation of MMA in serum promoted the progression of CRC via Wnt/β-catenin signaling pathway mediated EMT. These collective findings provide valuable insights into the vital role of age-related metabolic reprogramming in CRC progression and propose a potential therapeutic target for elderly CRC.

Tahitian bridal veil (Gibasis pellucida) and small-leaf spiderwort (Tradescantia fluminensis) are both invasive species in natural areas throughout Florida. However, very little is known regarding herbicide control. To provide land managers with herbicidal control options for both species, postemergence herbicides were evaluated for efficacy in a greenhouse to identify herbicide options that control both species under similar settings. Four herbicides, including triclopyr acid, triclopyr amine + 2,4-D amine, triclopyr amine, and glufosinate were applied at standard label rates and compared to a non-treated control group for efficacy. Visual control ratings were taken at 2, 4, and 8 weeks after treatment (WAT), and shoot dry weights (WAT 8) and regrowth dry weights (WAT 12) were determined. Triclopyr (acid and amine) generally provided the most consistent control of both species as evidenced by the visual control ratings and shoot dry weight data which showed reductions of 76% to 89% in shoot biomass at trial conclusion. Triclopyr + 2,4-D reduced shoot dry weights by 52% to 54% and was the least effective when considering the control of both species.

Tricholoma matsutake is a kind of ectomycorrhizal fungus. In addition to its vital influence on symbiotic plants, the impact of the soil microbial community on the growth and development of T. matsutake has been garnering attention. To clarify the differences in soil microflora between “T. matsutake Shangri-la” and “T. matsutake Nanhua”, and the effects of soil bacteria and fungi on the growth and development of T. matsutake, this study took the soil of “T. matsutake Shangri-la” and “T. matsutake Nanhua” at different developmental stages. A total of 7694 bacterial OTUs and 2170 fungal OTUs were obtained through microbial omics based on amplicon sequencing. The results indicate that the α diversity and composition of the soil microorganisms in the T. matsutake Shangri-la shiro were higher than those of the T. matsutake Nanhua. It is affected by species, geographical locations, and the growth period of matsutake. Matsutake mycelia also recruit certain types of bacteria and fungi in the stage of fruiting body development. Both bacteria and fungi positively and negatively regulate the fruiting body development of matsutake mushrooms. This study will provide a basis for the semi-artificial cultivation of matsutake.

This paper concentrates on the elevator passenger detection task, a pivotal element for subsequent elevator passenger tracking and behavior recognition, crucial for ensuring passenger safety. To enhance the accuracy of detecting passenger positions inside elevators, we improved the YOLOv8 network and proposed the SC-YOLOv8 elevator passenger detection network with soft-pooling and attention mechanisms. The main improvements in this paper encompass the following aspects: Firstly, we transformed the convolution module (ConvModule) of the YOLOv8 backbone network by introducing spatial and channel reconstruction convolution (SCConv). This improvement aims to reduce spatial and channel redundancy in the feature extraction process of the backbone network, thereby improving the overall efficiency and performance of the detection network. Secondly, we propose a dual-branch SPP-Fast module by incorporating a soft-pooling branch into the YOLOv8 network’s SPP-Fast module. This dual-branch SPP-Fast module can preserve essential information while reducing the impact of noise. Finally, we propose a soft-pooling and multi-scale convolution CBAM module to further enhance the network’s performance. This module enhances the network’s focus on key regions, allowing for more targeted feature extraction, thereby further improving the accuracy of object detection. Additionally, the attention module enhances the network’s robustness in handling complex backgrounds. We conducted experiments on an elevator passenger dataset. The results show that the precision, recall, and mAP of our improved YOLOv8 network are 94.32%, 91.17%, and 92.95%, respectively, all surpassing those of the original YOLOv8 network.

Positive temperature coefficient of electrical resistivity (PTCR) materials with low Curie temperature have been paid increasing attention lately. In this study, PTCR materials with a Curie temperature of approximately −15 °C were investigated by La3+ doping Ba0.55Sr0.45TiO3 ceramics. It could be expected to meet the requirements of thermal management systems for low-temperature control. In addition, a trace amount of Bi0.5Na0.5TiO3 (BNT) was employed to improve the resistivity and the PTCR performance. A significant PTCR effect was achieved with a high resistivity jump of nearly four orders of magnitude, a high temperature coefficient of ~28.76%/°C, and a narrow transition temperature span of 22 °C in the (Ba0.55Sr0.45)0.99875La0.00125Ti1.01O3-0.0025Bi0.5Na0.5TiO3 ceramics. The PTCR enhancement mechanism of BNT is discussed.

Exploiting novel materials with high specific capacities is crucial for the progress of advanced energy storage devices. Intentionally constructing functional heterostructures based on a variety of two-dimensional (2D) substances proves to be an extremely efficient method for capitalizing on the shared benefits of these materials. By elaborately designing the structure, a greatly escalated steadiness can be achieved throughout electrochemical cycles, along with boosted electron transfer kinetics. In this study, chemical vapor deposition (CVD) was utilized to alter the surface composition of multilayer Ti3C2Tx MXene, contributing to contriving various layered heterostructure materials through a precise adjustment of the reaction temperature. The optimal composite materials at a reaction temperature of 500 °C (defined as MX500), incorporating MXene as the conductive substrate, exhibited outstanding stability and high coulombic efficiency during electrochemical cycling. Meanwhile, the reactive sites are increased by using TiS2 and TiO2 at the heterogeneous interfaces, which sustains a specific capacity of 449 mAh g−1 after 200 cycles at a current density of 0.1 A g−1 and further demonstrates their exceptional electrochemical characteristics. Additionally, the noted pseudocapacitive properties, like MXene materials, further highlight the diverse capabilities of intuitive material design. This study illuminates the complex details of surface modification in multilayer MXene and offers a crucial understanding of the strategic creation of heterostructures, significantly impacting sophisticated electrochemical applications.

Cinnamomum camphora seed kernel protein isolate (CPI) has attracted increasing attention due to its sustainability and potential applications. This study aimed to investigate the effects of freeze-drying (FD), vacuum-drying (VD), and spray-drying (SD) on the physicochemical and functional properties of CPI. The morphology observation results showed that the SD-CPI, SD-CPI, and VD-CPI were spherical, lamellar, and massive, respectively. Compared to FD and SD, VD had more impact on the color, surface hydrophobicity, intermolecular disulfide bonds, intrinsic fluorescence, and thermal stability of CPI. Fourier transform infrared spectroscopy (FTIR) analyses showed that among three CPI samples, VD-CPI had the highest content of β-sheet but the lowest contents of α-helix and β-turn. At different pH values, the solubility, emulsification, and foaming properties of VD-CPI were inferior to those of FD-CPI and SD-CPI. These results provide useful information on the changes in the physicochemical and functional properties of CPI subjected to different drying methods, and offer theoretical guidance for the production and use of CPI in the food industry.

Rhizoma Panacis Japonici (RPJ) is an ancient herbal medicine from China that has long been employed for its medicinal benefits in relieving arthritis physical debility and diverse afflictions. The primary bioactive constituents found in RPJ are triterpene saponins, which exhibit numerous pharmacological actions, including anti-inflammatory, antioxidant, and immunomodulating effects. The present study established a straightforward and effective approach for characterizing triterpene saponins in RPJ. An offline HILIC × RP LC/QTOF-MS method was developed, along with a self-constructed in-house database containing 612 saponins reported in the Panax genus and 228 predicted metabolites. The approach achieved good chromatographic performance in isolating triterpene saponins of RPJ, with the HILIC column as the first dimension (1D) and the BEH C18 column as the second dimension (2D). The developed two-dimensional liquid chromatography system exhibited an orthogonality of 0.61 and a peak capacity of 1249. Detection was performed using a QTOF mass spectrometer in a data-independent manner (MSE) in a negative ion mode. Using the in-house database, the collected MS data were processed by an automatic workflow on UNIFI 1.8.2 software, which included data correction, matching of precursor and product ions, and peak annotation. In this study, 307 saponins were characterized from RPJ and 76 saponins were identified for the first time in Panax japonicus. This research not only enhances our understanding of the chemical characteristics of RPJ but also offers a simple and efficient method for analyzing the complex composition of herbal medicine.

Gravity inversion can be used to obtain the spatial structure and physical properties of subsurface anomalies through gravity observation data. With the continuous development of machine learning, geophysical inversion methods based on deep learning have achieved good results. Geophysical inversion methods based on deep learning often employ large-scale data sets to obtain inversion networks with strong generalization. They are widely used but face a problem of lacking information constraints. Therefore, a self-constrained network is proposed to optimize the inversion results, composed of two networks with similar structures but different functions. At the same time, a fine-tuning strategy is also introduced. On the basis of data-driven deep learning, we further optimized the results by controlling the self-constrained network and optimizing fine-tuning strategy. The results of model testing show that the method proposed in this study can effectively improve inversion precision and obtain more reliable and accurate inversion results. Finally, the method is applied to the field data of Gonghe Basin, Qinghai Province, and the 3D inversion results are used to effectively delineate the geothermal storage area.

Numerical modeling and industrial experiment were used to study the entire vacuum arc remelting (VAR) process of 660 mm GH4742 superalloy. In order to calculate the electromagnetic field, flow field, temperature field, and melt pool shape change throughout the entire VAR process, a 2D axisymmetric model with coupled multi-physical fields matching industry trials was constructed. The microstructure and elemental distribution conditions in the large-size GH4742 ingot were analyzed in combination with the industrial ingot dissection experiment, and the most likely locations of freckle defects were predicted. According to the research, the melt pool has two clockwise flow fields that are affected by buoyancy and Lorentz forces. The melt pool depth deepens gradually as melting proceeds, with a maximum melt pool depth of 0.32 m and a paste zone width of 0.16 m. Although the ingot's elemental distribution is largely consistent, there is microsegregation between the dendrites. Where the freckle is most likely to appear, at (0.322, 0.075), the Rayleigh has a great value of 1.1. For melt pool form and dendritic spacing throughout the entire melting process, the results of the industrial ingot dissection experiment and the simulation results show good agreement, proving the accuracy of the thermal history and solidification microstructure calculation for the entire melting process. Furthermore, the prospect of combining numerical simulation with a special material database for accurate defect prediction is proposed.
