Abstract Endoplasmic reticulum stress (ERS) is commonly induced by accumulating misfolded or unfolded proteins in tumor microenvironment. Long non-coding RNAs (lncRNAs) play important roles in ERS response and lung adenocarcinoma (LUAD) progression. However, the role of ERS-related lncRNAs in LUAD remains unknown. In this study, we aimed to identify ERS-associated lncRNAs with prognostic value in LUAD and characterize their clinical implications. Cox and least absolute shrinkage and selection operator regression analyses identified nine ERS-related lncRNAs with independent prognostic abilities, including five protective factors (CROCCP2, KIAA0125, LINC0996, RPARP-AS1 and TBX5-AS1) and four risk factors (LINC0857, LINC116, RP11-21L23.2 and RP11-295G20.2). We developed an ERS-related lncRNA risk prediction model in predicting overall survival of LUAD patients, which classified TCGA cohorts into high-risk (HS) and low-risk (LS) groups. Comprehensive bioinformatic analyses revealed HS patients featured with late-stage tumors, greater mutation burdens, weaker anti-tumor immunity/responses, and lower sensitivity to targeted drugs compared to LS patients, contributing to tumor progression and a poor prognosis. Functional enrichment analysis implicated these ERS-related lncRNAs in cell migration, cell death, and immunity. Furthermore, expression of the most significantly upregulated risk lncRNA, RP11-295G20.2, was validated at the mRNA level using clinical LUAD samples. Knockdown of RP11-295G20.2 obviously reduced ERS and suppressed proliferation, invasion, and migration of LUAD cells. This novel ERS-related lncRNA signature provides a new biomarker for prognostic prediction, and ERS-associated RP11-295G20.2 serves as a potential therapeutic target in LUAD.

Abstract Background Sepsis is a life-threatening condition caused by an excessive inflammatory response to an infection, associated with high mortality. However, the regulatory mechanism of sepsis remains unclear. Results In this study, bioinformatics analysis revealed the novel key biomarkers associated with sepsis and potential regulators. Three public datasets (GSE28750, GSE57065 and GSE95233) were employed to recognize the differentially expressed genes (DEGs). Taking the intersection of DEGs from these three datasets, GO and KEGG pathway enrichment analysis revealed 537 shared DEGs and their biological functions and pathways. These genes were mainly enriched in T cell activation, differentiation, lymphocyte differentiation, mononuclear cell differentiation, and regulation of T cell activation based on GO analysis. Further, pathway enrichment analysis revealed that these DEGs were significantly enriched in Th1, Th2 and Th17 cell differentiation. Additionally, five hub immune-related genes (CD3E, HLA-DRA, IL2RB, ITK and LAT) were identified from the protein–protein interaction network, and sepsis patients with higher expression of hub genes had a better prognosis. Besides, 14 drugs targeting these five hub related genes were revealed on the basis of the DrugBank database, which proved advantageous for treating immune-related diseases. Conclusions These results strengthen the new understanding of sepsis development and provide a fresh perspective into discriminating the candidate biomarkers for predicting sepsis as well as identifying new drugs for treating sepsis.

The gut microbiomes of arthropods have significant impact on key physiological functions such as nutrition, reproduction, behavior, and health. Spiders are diverse and numerically dominant predators in crop fields where they are potentially important regulators of pests. Harnessing spiders to control agricultural pests is likely to be supported by an understanding of their gut microbiomes, and the environmental drivers shaping microbiome assemblages. This study aimed to deciphering the gut microbiome assembly of these invertebrate predators and elucidating potential implications of key environmental constraints in this process. Here, we used high-throughput sequencing to examine for the first time how the assemblages of bacteria in the gut of spiders are shaped by environmental variables. Local drivers of microbiome composition were globally-relevant input use system (organic production vs. conventional practice), and crop identity (Chinese cabbage vs. cauliflower). Landscape-scale factors, proportion of forest and grassland, compositional diversity, and habitat edge density, also strongly affected gut microbiota. Specific bacterial taxa were enriched in gut of spiders sampled from different settings and seasons. These findings provide a comprehensive insight into composition and plasticity of spider gut microbiota. Understanding the temporal responses of specific microbiota could lead to innovative strategies development for boosting biological control services of predators.

The characteristics, formation mechanisms, and influences on physical properties of carbonate minerals in shale reservoirs of Wufeng-Longmaxi formations in Sichuan Basin are systematically investigated by utilizing electron probe microscope with spectrometer and energy spectrometer, combined with physical properties and whole rock X-diffraction and organic carbon data. The research yielded the findings that follow: First, the main carbonate minerals are calcite, dolomite, and ferriferous dolomite. Calcite is a single mineral that fills the siliceous shell cavity of radiolarians and exists between mineral particles. Ferriferous dolomite always rings dolomite, which is a single mineral that is present among mineral particles and aggregates. Second, calcite is produced by microorganisms that secrete calcium carbonate in the surface of seawater. The siliceous skeleton cavity of radiolarian and seawater both precipitate calcite, which partially dissolves while settling in seawater before depositing on the seabed and being preserved by burial. Thirdly, the dolomite is a diagenetic mineral formed on the water–sediment interface with physiological activities of sulfate bacteria, and the ferriferous dolomite is produced by methanogenic metabolism during the initial burial of muddy sediments. Fourthly, organic carbon, pyrite, quartz, and clay minerals are closely related to reservoir physical properties, while carbonate has no effect on porosity and permeability as a whole. Future research on shale reservoir diagenesis should make use of the in-situ detection and element area scanning, in particular with the spectrogram from electron probe microscope technology, which provides typical petrological evidences for the study of characteristics, formation mechanism, and influence on physical properties of carbonate minerals in shale reservoirs.

Mangrove forests inhabit tropical or subtropical intertidal zones and have remarkable abilities in coastline protection. Kandelia obovata is considered the most cold-tolerant mangrove species and has been widely transplanted to the north subtropical zone of China for ecological restoration. However, the physiological and molecular mechanisms of K. obovata under colder climate was still unclear. Here, we manipulated the typical climate of cold waves in the north subtropical zone with cycles of cold/recovery and analyzed the physiological and transcriptomic responses of seedlings. We found that both physiological traits and gene expression profiles differed between the first and later cold waves, indicating K. obovata seedlings were acclimated by the first cold experience and prepared for latter cold waves. 1,135 cold acclimation-related genes (CARGs) were revealed, related to calcium signaling, cell wall modification, and post-translational modifications of ubiquitination pathways. We identified the roles of CBFs and CBF-independent transcription factors (ZATs and CZF1s) in regulating the expression of CARGs, suggesting both CBF-dependent and CBF- independent pathways functioned in the cold acclimation of K. obovata. Finally, we proposed a molecular mechanism of K. obovata cold acclimation with several key CARGs and transcriptional factors involved. Our experiments reveal strategies of K. obovata coping with cold environments and provide prospects for mangrove rehabilitation and management.

ObjectivesTo compare patients with atrial fibrillation (AF) undergoing left atrial appendage closure (LAAC) with catheter ablation (CA) and those without CA.BackgroundThe CA of AF may cause ridge edema, which may affect the safety of LAAC.MethodsPatients with AF (N = 98) who underwent LAAC (combined CA + LAAC procedure group; N = 51) or alone (LAAC group; N = 47) received pre-procedural, intra-procedural, and 6 week post-procedural transesophageal echocardiography (TEE). The depth and ostial diameter of LAA, device compression, residual leak, and ridge thickness were evaluated in the patients who had undergone combined and alone procedures, as well as images of LAA and primary clinical characteristics.ResultsA residual leak was identified in 27 patients at 6 weeks after implantation by TEE (19 in the combined procedures group and eight in the alone group; p = 0.04). The combined procedure group had a significantly higher rate of a new residual leak than the alone group (25.5 vs. 8.5%; p = 0.03). Meanwhile, compared with at the time of implant, a smaller amount of device compression ratio was significant after 6 weeks (22.44 ± 3.90 vs. 19.59 ± 5.39; p = 0.03). There was no significant difference between both groups in all-cause mortality, cardiovascular mortality, and TIA/stroke/system embolism.ConclusionThe combined procedures of CA and LAAC for AF are feasible and safe; however, during the follow-up period, we found that the resolution of ridge edema caused by CA might cause an increased residual leak and a smaller device compression ratio.

Natural gas from shale gas reservoirs has been an important contributor for reserve growth, deliverability construction, and profits growth in natural gas industry in the world. Hydraulic fracturing is commonly required in the shale gas commercial development, and thus understanding the present-day in situ stress field is greatly significant for the hydraulic fracturing and efficient development in shale gas reservoirs. However, there are no systematic investigations on the present-day in situ stress field in the Haiba Block from the Sichuan Basin, South China. In this study, the present-day in situ stress orientations and magnitudes in shale reservoir of Haiba Block are investigated based on the well interpretations from borehole image log and geomechanical modeling. Then, the effects of stresses on hydraulic fracturing, horizontal wells, and natural fracture reactivation were discussed. The results indicate that the horizontal maximum principal stress (SHmax) orientation is mainly in the NE-SW-trending, NW-SE-trending, and WNW-ESE-trending in the Haiba Block. The magnitudes of horizontal maximum and minimum principal stresses are 13.5 MPa~85.5 MPa and 2.8 MPa~31.6 MPa, respectively. In the Haiba Block, the differential stress is generally low in the northern part, which indicates that complex hydraulic fracture networks may be produced. While the natural fractures are generally stable under the present-day in situ stress field. When the increase of pore pressure gradient is about 30 KPa/m, nearly all natural fractures in the Longmaxi Formation may be reactivated. The results can provide the insights into a better understanding of the present-day in situ stress distribution so as to optimize perforation orientation, hydraulic fracturing design, and enhance gas production in shale gas reservoirs.

Abstract Inter‐ and intra‐guild interactions are important in the coexistence of predators and their prey, especially in highly disturbed vegetable cropping systems with sporadic food resources. Assessing the dietary range of a predator taxon characterized by diverse foraging behavior using conventional approaches, such as visual observation and conventional molecular approaches for prey detection, has serious logistical problems. In this study, we assessed the prey compositions and compare the dietary spectrum of a functionally diverge group of predators—spiders—to characterize their trophic interactions and assess biological control potential in Brassica vegetable fields. We used high‐throughput sequencing (HTS) and biotic interaction networks to precisely annotate the predation spectrum and highlight the predator–predator and predator–prey interactions. The prey taxa in the gut of all spider families were mainly enriched with insects (including dipterans, coleopterans, orthopterans, hemipterans, and lepidopterans) with lower proportions of arachnids (such as Araneae) along with a wide range of other prey factions. Despite the generalist foraging behavior of spiders, the community structure analysis and interaction networks highlighted the overrepresentation of particular prey taxa in the gut of each spider family, as well as showing the extent of interfamily predation by spiders. Identifying the diverse trophic niche proportions underpins the importance of spiders as predators of pests in highly disturbed agroecosystems. More specifically, combining HTS with advanced ecological community analysis reveals the preferences and biological control potential of particular spider taxa (such as Salticidae against lepidopterans and Pisauridae against dipterans), and so provides a valuable evidence base for targeted conservation biological control efforts in complex trophic networks.

Abstract Background Restoration through planting is the dominant strategy to conserve mangrove ecosystems. However, many of the plantations fail to survive. Site and seeding selection matters for planting. The process of afforestation, where individuals were planted in a novel environment, is essentially human-controlled transplanting events. Trying to deepen and expand the understanding of the effects of transplanting on plants, we have performed a seven-year-long reciprocal transplant experiment on Kandelia obovata along a latitudinal gradient. Results Combined phenotypic analyses and next-generation sequencing, we found phenotypic discrepancies among individuals from different populations in the common garden and genetic differentiation among populations. The central population with abundant genetic diversity and high phenotypic plasticity had a wide plantable range. But its biomass was reduced after being transferred to other latitudes. The suppressed expression of lignin biosynthesis genes revealed by RNA-seq was responsible for the biomass reduction. Moreover, using whole-genome bisulfite sequencing, we observed modification of DNA methylation in MADS-box genes that involved in the regulation of flowering time, which might contribute to the adaptation to new environments. Conclusions Taking advantage of classical ecological experiments as well as multi-omics analyses, our work observed morphology differences and genetic differentiation among different populations of K. obovata, offering scientific advice for the development of restoration strategy with long-term efficacy, also explored phenotypic, transcript, and epigenetic responses of plants to transplanting events between latitudes.

High graphitization is responsible for low-resistivity shale development with poor reservoir quality. This paper provides an explanation of organic matter graphitization and determines the impact of high graphite content on low-resistivity shale reservoir quality at the Wufeng-Longmaxi Formation in the Southern Sichuan Basin. Fine veins are frequently developed at shale samples with Ro > 3.5%, graphitized organic matter > 25%, and resistivity < 5 Ω•m, which are dominated by three mineral assemblages: brunsvigite, barite-hyalophane-barium feldspar-potassium feldspar-anhydrite, and calcite-ankerite. These filling minerals are characterized by an Eu positive anomaly and high Ba, Fe, and Mn contents, suggesting that low-resistivity shale was modified by magmatic-related low-temperature hydrothermal fluid. Temperature measurements of brine inclusions and a semi-empirical geothermometer of chlorite show that low-temperature hydrothermal fluid experienced the chlorite stage (150–180 °C), the low-sulfidation stage (120–150 °C), and the low-temperature calcitization stage. Paleozoic fault systems and late Permian hydrothermal activities associated with the Emeishan mantle plume control the graphitization of low-resistivity shale. The water formation and seawater infiltrated into the deep crust along the Paleozoic basement faults under gravity, developing alkaline hot brine through mantle plume heating and then causing a water-rock reaction with basement rocks. They migrated upward along deep and large Paleozoic faults through convective thermal circulation in the Tiangongtang area, the Shuanglong-Luochang area, and the Xuyong area. Cation exchange and redox reactions occurred during the interaction between high-temperature hydrothermal fluid and cool wall rocks. The migration of alkaline hot brine via the Wufeng-Longmaxi shale introduced a subsequent water-rock reaction, resulting in the development of hydrothermal mineral assemblages that intricately filled fractures. It increased formation temperature and enhanced thermal maturity and graphitization of organic matter at the Wufeng-Longmaxi low-resistivity shale, resulting in a wide distribution of low-resistivity shale at the Changning Block.

Deep shale gas (burial depth > 3500 m) in the Longmaxi Formation of southern Sichuan Province will be the primary target for exploration and development in China for a relatively long period. However, the lack of a physical basis for the “sweet-spots” seismic and well-logging prediction is caused by uncertainty in the rock physical properties of deep shale gas in the research area. Acoustic and hardness measurements were performed on shale samples from a deep layer of the Longmaxi Formation in southern Sichuan. Microtextural characteristics of the shale samples were also analyzed by conventional optical microscopy and scanning electron microscopy. Based on these measurements, the rock physical properties of the shale samples and control factors are discussed. It is shown that the deep shale samples have similar properties to the shallow shale in mineral composition, microtexture, and pore type. However, the organic pore in deep shale samples is relatively undeveloped, while the dissolved pores are more developed. For high-quality shale samples (total organic content > 2%), crystal quartz of biological origin forms the framework of rock samples, resulting in effective dynamic and static properties, reflecting the elastic behavior of rigid quartz aggregates. For organic-lean samples (total organic content < 2%), orientated detrital clay particles take the role of load-bearing grains. Therefore, these shale samples’ overall rock physical properties are mainly controlled by the elastic properties of “soft” clay. The load-bearing grain variation from organic-rich shale samples to organic-lean samples results in an overturned “V”-type change in terms of velocity versus content. Organic-rich shale samples also show an apparent low Poisson’s ratio. Organic-rich shale has a slight velocity–porosity trend, while organic-lean shale shows a significant velocity–porosity trend. In addition, due to the difference in rock microtexture between organic-rich and organic-lean shale, these two kinds of reservoir rocks can be discriminated in cross plots of P-wave impedance versus Poisson’s ratio and Young’s modulus versus Poisson’s ratio. Change in hardness also reflects the control of microtexture, and shale samples with biological-origin quartz as load-bearing grains show higher hardness and brittleness. However, the variation in quartz content has less of an impact on hardness and brittleness in shale samples with clay as the load-bearing grain. Our results provide an experimental basis for the geophysical identification and prediction of deep shale gas layers.

Abstract Background Marine medaka (Oryzias melastigma) is considered as an important ecotoxicological indicator to study the biochemical, physiological and molecular responses of marine organisms towards increasing amount of pollutants in marine and estuarine waters. Results In this study, we reported a high-quality and accurate de novo genome assembly of marine medaka through the integration of single-molecule sequencing, Illumina paired-end sequencing, and 10X Genomics linked-reads. The 844.17 Mb assembly is estimated to cover more than 98% of the genome and is more continuous with fewer gaps and errors than the previous genome assembly. Comparison of O. melastigma with closely related species showed significant expansion of gene families associated with DNA repair and ATP-binding cassette (ABC) transporter pathways. We identified 274 genes that appear to be under significant positive selection and are involved in DNA repair, cellular transportation processes, conservation and stability of the genome. The positive selection of genes and the considerable expansion in gene numbers, especially related to stimulus responses provide strong supports for adaptations of O. melastigma under varying environmental stresses. Conclusions The highly contiguous marine medaka genome and comparative genomic analyses will increase our understanding of the underlying mechanisms related to its extraordinary adaptation capability, leading towards acceleration in the ongoing and future investigations in marine ecotoxicology.

Both complete and uncompleted radiolarian siliceous shells were developed at Wufeng-Longmaxi radiolarian siliceous shale laminae in Sichuan Basin. Micro- and ultra-micropetrological observation suggests that they were successively filled by calcite, pyrite and organic–silicon complex, where pyrite and organic–silicon complex filled dissolved pores associated with calcite during sedimentation. Calcite was derived from calcium carbonate produced by microbial activities at the seawater surface. The environment of radiolarian siliceous shell cavities, which was suitable for sulfate reducing bacterial growth or dissolved hydrogen sulfide reducing Fe3+, contributed positively to pyrite development. Organic–silicon complex development was related to microorganism metabolism that was an important silica source. Honeycomb-like organic pores were developed in cavities with complete shells, but were not developed in cavities with uncompleted shells. This is because the latter could not withstand overburden pressure compared with the former. The only approach to figure out organic pore carriers and understand sequences and development processes of minerals and organic matter is to select weakly compacted radiolarian siliceous shale laminae to carry out micro- and ultra-micropetrological observation and geochemical testing via various technologies.

Strobilanthes cusia (Nees) Kuntze is an important plant used to process the traditional Chinese herbal medicines “Qingdai” and “Nanbanlangen”. The key active ingredients are indole alkaloids (IAs) that exert antibacterial, antiviral, and antitumor pharmacological activities and serve as natural dyes. We assembled the S. cusia genome at the chromosome level through combined PacBio circular consensus sequencing (CCS) and Hi-C sequencing data. Hi-C data revealed a draft genome size of 913.74 Mb, with 904.18 Mb contigs anchored into 16 pseudo-chromosomes. Contig N50 and scaffold N50 were 35.59 and 68.44 Mb, respectively. Of the 32,974 predicted protein-coding genes, 96.52% were functionally annotated in public databases. We predicted 675.66 Mb repetitive sequences, 47.08% of sequences were long terminal repeat (LTR) retrotransposons. Moreover, 983 Strobilanthes-specific genes (SSGs) were identified for the first time, accounting for ~2.98% of all protein-coding genes. Further, 245 putative centromeric and 29 putative telomeric fragments were identified. The transcriptome analysis identified 2,975 differentially expressed genes (DEGs) enriched in phenylpropanoid, flavonoid, and triterpenoid biosynthesis. This systematic characterization of key enzyme-coding genes associated with the IA pathway and basic helix-loop-helix (bHLH) transcription factor family formed a network from the shikimate pathway to the indole alkaloid synthesis pathway in S. cusia. The high-quality S. cusia genome presented herein is an essential resource for the traditional Chinese medicine genomics studies and understanding the genetic underpinning of IA biosynthesis.

We developed a tool for targeted generation of singlet oxygen using light activation of a genetically encoded fluorogen-activating protein complexed with a unique dye molecule that becomes a potent photosensitizer upon interaction with the protein. By targeting the protein receptor to activate this dye in distinct subcellular locations at consistent per-cell concentrations, we investigated the impact of localized production of singlet oxygen on induction of cell death. We analyzed light dose-dependent cytotoxic response and characterized the apoptotic vs. necrotic cell death as a function of subcellular location, including the nucleus, the cytosol, the endoplasmic reticulum, the mitochondria, and the membrane. We find that different subcellular origins of singlet oxygen have different potencies in cytotoxic response and the pathways of cell death, and we observed that CT26 and HEK293 cell lines are differentially sensitive to mitochondrially localized singlet oxygen stresses. This work provides new insight into the function of type II reactive oxygen generating photosensitizing processes in inducing targeted cell death and raises interesting mechanistic questions about tolerance and survival mechanisms in studies of oxidative stress in clonal cell populations.

Based on various test data, the composition, texture, structure and lamina types of gas-bearing shale were determined based on Well Wuxi 2 of the Silurian Longmaxi Formation in the Sichuan Basin. Four types of lamina, namely organic-rich lamina, organic-bearing lamina, clay lamina and silty lamina, are developed in the Longmaxi Formation of Well Wuxi 2, and they form 2 kinds of lamina set and 5 kinds of beds. Because of increasing supply of terrigenous clasts and enhancing hydrodynamics and associated oxygen levels, the contents of TOC and brittle mineral reduce and content of clay mineral increases gradually as the depth becomes shallow. Organic-rich lamina, organic-rich + organic-bearing lamina set and organic-rich bed dominate the small layers 1-3 of Member 1 of the Longmaxi Formation, suggesting anoxic and weak hydraulic depositional setting. Organic-rich lamina, along with organic-bearing lamina and silty lamina, appear in small layer 4, suggesting increased oxygenated and hydraulic level. Small layers 1-3 are the best interval and drilling target of shale gas exploration and development. This paper mainly discusses the industrialization progress, “sweet spot” evaluation criterion, E&P technologies, success experiences, challenges and prospects of China's shale gas. Based on the geologic and engineering parameters of the Fuling, Changning and Weiyuan shale gas fields in the Sichuan Basin, this paper points out that China's shale gas has its particularity. The discoveries of super-giant marine shale gas fields with high evolution degree (Ro=2.0%−3.5%) and ultrahigh pressure (pressure coefficient=1.3−2.1) in southern China is of important scientific significance and practical value to ancient marine shale gas exploration and development to China and even the world. It's proposed that shale gas “sweet spots” must be characterized by high gas content, excellent frackability and good economy etc. The key indicators to determine the shale gas enrichment interval and trajectory of horizontal wells include “four highs”, that is high TOC (>3.0%), high porosity (>3.0%), high gas content (>3.0 m3/t) and high formation pressure (pressure coefficient>1.3), and “two well-developed” (well-developed beddings and well-developed micro-fractures). It's suggested that horizontal well laneway be designed in the middle of high pressure compartment between the Upper Ordovician Wufeng Formation and Lower Silurian Longmaxi Formation. The mode of forming “artificial shale gas reservoir” by “fracturing micro-reservoir group” is proposed and the mechanisms of “closing-in after fracturing, limiting production through pressure control” are revealed. Several key technologies (such as three-dimensional seismic survey and micro-seismic monitoring of fracturing, horizontal wells, “factory-like” or industrialized production mode, etc.) were formed. Some successful experiences (such as “sweet spot” selection, horizontal well laneway control, horizontal length optimization and “factory-like” production mode, etc.) were obtained. The four main challenges to realize large-scale production of shale gas in China include uncertainty of shale gas resources, breakthroughs in key technologies and equipment of shale gas exploration and development below 3 500 m, lower cost of production, as well as water resources and environment protection. It is predicted that the recoverable resources of the Lower Paleozoic marine shale gas in southern China are approximately 8.8×1012 m3, among which the recoverable resources in the Sichuan Basin are 4.5×1012 m3 in the favorable area of 4.0×104 km2. The productivity of (200−300)×108 m3/a is predicted to be realized by 2020 when the integrated revolution of “theory, technology, production and cost” is realized in Chinese shale gas exploration and development. It is expected in the future to be built “Southwest Daqing Oilfield (Gas Daqing)” in Sichuan Basin with conventional and unconventional natural gas production. The main factors controlling the enrichment and high yield of shale gas were analyzed based on the recent research progress of depositional model and reservoir characterization of organic-rich shale in China. The study determines the space-time comparison basis of graptolite sequence in the Upper Ordovician Wufeng Formation–Lower Silurian Longmaxi Formation and proposes the important depositional pattern of marine organic-rich shale: stable ocean basin with low subsidence rate, high sea level, semi-enclosed water body, and low sedimentation rate. Deposited in the stage of Late Ordovician-Early Silurian, the superior shale with thickness of 20−80 m and total organic carbon (TOC) content of 2.0%−8.4% was developed in large deep-water shelf environment which is favorable for black shale development. Based on the comparison among the Jiaoshiba, Changning and Weiyuan shale gas fields, it is believed that reservoirs of scale are mainly controlled by shale rich in biogenic silica and calcium, moderate thermal maturity, high matrix porosity, and abundant fracture. The shales in the Wufeng and Longmaxi formations are characterized by porosity of 3.0%−8.4%, permeability of 0.000 2×10−3−0.500 0×10−3 μm2, stable areal distribution of matrix pore volume and their constituents, great variation in fracture and pore characteristics among different tectonic regions as well as different well fields and different intervals in the same tectonic. The Cambrian Qiongzhusi shale features poor physical properties with the porosity of 1.5%−2.9% and the permeability of 0.001×10−3−0.010×10−3 μm2, resulted from the carbonization of organic matter, high crystallinity of clay minerals and later filling in bioclastic intragranular pores. Four factors controlling the accumulation and high production of shale gas were confirmed: depositional environment, thermal evolution, pore and fracture development, and tectonic preservation condition; two special features were found: high thermal maturity (Ro of 2.0%−3.5%) and overpressure of reservoir (pressure coefficient of 1.3−2.1); and two enrichment modes were summarized: “structural sweet spots” and “continuous sweet area”. Low-salinity water (LSW) flooding technology has obvious operational and economic advantages, so it is applied to practice in many oilfields. However, there are differences in the oil recovery efficiencies in different oilfields, the reasons for which need to be further studied and discussed. This paper studies the effect of different clay mineral compositions on low-salinity water flooding. For this purpose, three groups of core displacement experiments were designed with cores containing different clay mineral compositions for comparison. In the process of formation water and low-salinity water driving, the oil recovery and produced-water properties were measured. By comparing the two types of water flooding, it was found that the cores with the highest montmorillonite content had the best effect (5.7%) on low-salinity water flooding and the cores with the highest kaolinite content had the least effect (1.9%). This phenomenon is closely related to the difference in ion exchange capacity of the clay minerals. Moreover, after switching to low-salinity water flooding, the interfacial tension and wetting angle of the produced-water increased and the value of pH decreased, which are important mechanisms for enhancing oil recovery by low-salinity water flooding. This study reveals the influence of clay mineral composition on low-salinity water flooding and can provide more guidance for conventional and unconventional oilfield application of low-salinity water flooding technology.

The gut microbiomes of arthropods have significant impact on key physiological functions such as nutrition, reproduction, behavior, and health. Spiders are diverse and numerically dominant predators in crop fields where they are potentially important regulators of pests. Harnessing spiders to control agricultural pests is likely to be supported by an understanding of their gut microbiomes, and the environmental drivers shaping microbiome assemblages. This study aimed to deciphering the gut microbiome assembly of these invertebrate predators and elucidating potential implications of key environmental constraints in this process. Here, we used high-throughput sequencing to examine for the first time how the assemblages of bacteria in the gut of spiders are shaped by environmental variables. Local drivers of microbiome composition were globally-relevant input use system(organic production vs. conventional practice), and crop identity (Chinese cabbage vs. cauliflower). Landscape-scale factors, proportion of forest and grassland, compositional diversity, and habitat edge density, also strongly affected gut microbiota. Specific bacterial taxa were enriched in gut of spiders sampled from different settings and seasons. These findings provide a comprehensive insight into composition and plasticity of spider gut microbiota. Understanding the temporal responses of specific microbiota could lead to innovative strategies development for boosting biological control services of predators. [ABSTRACT FROM AUTHOR]
