Abstract Nuclear factor-kappaB (NF-ĸB) plays a crucial role in both innate and adaptive immune systems, significantly influencing various physiological processes such as cell proliferation, migration, differentiation, survival, and stemness. The function of NF-ĸB in cancer progression and response to chemotherapy has gained increasing attention. This review highlights the role of NF-ĸB in inflammation control, biological mechanisms, and therapeutic implications in cancer treatment. NF-ĸB is instrumental in altering the release of inflammatory factors such as TNF-α, IL-6, and IL-1β, which are key in the regulation of carcinogenesis. Specifically, in conditions including colitis, NF-ĸB upregulation can intensify inflammation, potentially leading to the development of colorectal cancer. Its pivotal role extends to regulating the tumor microenvironment, impacting components such as macrophages, fibroblasts, T cells, and natural killer cells. This regulation influences tumorigenesis and can dampen anti-tumor immune responses. Additionally, NF-ĸB modulates cell death mechanisms, notably by inhibiting apoptosis and ferroptosis. It also has a dual role in stimulating or suppressing autophagy in various cancers. Beyond these functions, NF-ĸB plays a role in controlling cancer stem cells, fostering angiogenesis, increasing metastatic potential through EMT induction, and reducing tumor cell sensitivity to chemotherapy and radiotherapy. Given its oncogenic capabilities, research has focused on natural products and small molecule compounds that can suppress NF-ĸB, offering promising avenues for cancer therapy.

Abstract The outcomes of patients with sepsis are influenced by the contractile function of the right ventricle (RV), but the impact of cardiopulmonary interaction in ICU-mortality of sepsis patients remains unclear. This study aims to investigate the ICU-mortality impact of right ventricular-pulmonary artery (RV-PA) coupling in patients with sepsis. We employed echocardiography to assess patients with sepsis within the initial 24 h of their admission to the ICU. RV-PA coupling was evaluated using the tricuspid annular plane systolic excursion (TAPSE) to pulmonary artery systolic pressure (PASP) ratio. A total of 92 subjects were enrolled, with 55 survivors and 37 non-survivors. TAPSE/PASP ratio assessed mortality with an area under the curve (AUC) of 0.766 (95% CI 0.670–0.862) and the optimal cutoff value was 0.495 mm/mmHg. We constructed a nomogram depicting the TAPSE/PASP in conjunction with IL-6 and Lac for the joint prediction of sepsis prognosis, and demonstrated the highest predictive capability (AUC = 0.878, 95% CI 0.809–0.948). In conclusion, the TAPSE/PASP ratio demonstrated prognostic value for ICU mortality in sepsis patients. The nomogram, which combines the TAPSE/PASP, IL-6, and LAC, demonstrated enhanced predictive efficacy for the prognosis of sepsis patients.

Abstract Thermosensation is vital for the survival, propagation, and adaption of all organisms, but its mechanism is not fully understood yet. Here, we find that TMC6, a membrane protein of unknown function, is highly expressed in dorsal root ganglion (DRG) neurons and functions as a Gαq-coupled G protein-coupled receptor (GPCR)-like receptor to sense noxious heat. TMC6-deficient mice display a substantial impairment in noxious heat sensation while maintaining normal perception of cold, warmth, touch, and mechanical pain. Further studies show that TMC6 interacts with Gαq via its intracellular C-terminal region spanning Ser780 to Pro810. Specifically disrupting such interaction using polypeptide in DRG neurons, genetically ablating Gαq, or pharmacologically blocking Gαq-coupled GPCR signaling can replicate the phenotype of TMC6 deficient mice regarding noxious heat sensation. Noxious heat stimulation triggers intracellular calcium release from the endoplasmic reticulum (ER) of TMC6- but not control vector-transfected HEK293T cell, which can be significantly inhibited by blocking PLC or IP3R. Consistently, noxious heat-induced intracellular Ca2+ release from ER and action potentials of DRG neurons largely reduced when ablating TMC6 or blocking Gαq/PLC/IP3R signaling pathway as well. In summary, our findings indicate that TMC6 can directly function as a Gαq-coupled GPCR-like receptor sensing noxious heat.

Abstract Objective This study aimed to develop and apply a nomogram with good accuracy to predict the risk of CRAB infections in neuro-critically ill patients. In addition, the difficulties and expectations of application such a tool in clinical practice was investigated. Methods A mixed methods sequential explanatory study design was utilized. We first conducted a retrospective study to identify the risk factors for the development of CRAB infections in neuro-critically ill patients; and further develop and validate a nomogram predictive model. Then, based on the developed predictive tool, medical staff in the neuro-ICU were received an in-depth interview to investigate their opinions and barriers in using the prediction tool during clinical practice. The model development and validation is carried out by R. The transcripts of the interviews were analyzed by Maxqda. Results In our cohort, the occurrence of CRAB infections was 8.63% (47/544). Multivariate regression analysis showed that the length of neuro-ICU stay, male, diabetes, low red blood cell (RBC) count, high levels of procalcitonin (PCT), and number of antibiotics ≥ 2 were independent risk factors for CRAB infections in neuro-ICU patients. Our nomogram model demonstrated a good calibration and discrimination in both training and validation sets, with AUC values of 0.816 and 0.875. Additionally, the model demonstrated good clinical utility. The significant barriers identified in the interview include “skepticism about the accuracy of the model”, “delay in early prediction by the indicator of length of neuro-ICU stay”, and “lack of a proper protocol for clinical application”. Conclusions We established and validated a nomogram incorporating six easily accessed indicators during clinical practice (the length of neuro-ICU stay, male, diabetes, RBC, PCT level, and the number of antibiotics used) to predict the risk of CRAB infections in neuro-ICU patients. Medical staff are generally interested in using the tool to predict the risk of CRAB, however delivering clinical prediction tools in routine clinical practice remains challenging.

Due to the uncertainty in output caused by environmental changes, significant discrepancies are expected between the surface flow velocities predicted using deep learning methods and the instantaneous flow velocities. In this paper, a two-stage deep learning flow velocity measurement algorithm is proposed. During the external calibration process, the upper and lower frames of the recorded water flow video are cyclically traversed to acquire predicted flow velocity values using the deep learning velocity measurement algorithm. Meanwhile, the pixel displacement is obtained using the sparse optical flow tracking method and then post-processed to derive the velocity calibration value and pixel calibration value. During the detection process, the deep learning-predicted flow velocity is internally calibrated using the velocity calibration value and the pixel calibration value to adapt to changes in water flows. Compared with the pre-improved algorithm, the method achieves the minimum root mean square error in five different flow velocity videos and maintains high accuracy when the flow velocity changes rapidly. The obtained results are very promising and can help improve the reliability of video flow rate assessment algorithms. HIGHLIGHTS New methods are needed to ensure the instability of the video-based surface velocity prediction due to environmental changes.; Avoid direct measurement of high velocity, using the slow to fast process to indirectly get accurate high velocity.; We provide a river data set for deep learning model training, which is convenient for researchers interested in this to expand their research.;

Since 2020, with in-depth research on the Shuangji structural belt in the Jimsar Depression, Junggar Basin and high-yield oil flow data obtained from the Triassic Jingjingzigou Formation in the Well Satan 1, it has been demonstrated that the Jimsar and Jinan depressions have promising potential for conventional oil and gas exploration. Objective To further study the geological characteristics and exploration potential of the Jimsar and Jinan depressions, Methods this paper comprehensively analyses data from core samples, logging, well logging, and seismic surveys; identifies regional unconformities; categorizes different tectonic stages; and establishes the correlation between tectonic and stratigraphic evolution. Based on this, the development process of each stratum is restored, and the coupling relationship between tectonics and the evolution of uplift and depression within the research area is established, and the prototype basin is restored. Results This study revealed five large unconformities in the Jimsar and Jinan depressions, which have undergone four episodes of tectonic activity. The stratigraphic evolution characteristics of the study area ranged from "north high and south low" in the Early Permian to "south high and north low" in the Palaeogene and Neogene. The original basin also changed from a "multiconvex and multiconcave" pattern in the Early Permian to a "few convex and large concave" structural pattern in the Palaeogene and Neogene. Conclusion The establishment of the coupling relationship between tectonics and the evolution of uplift and depression at different times and the restoration of the prototype basin during each period lays an important foundation for clarifying the basin characteristics of the Jimsar and Jinan depressions during different periods and helps to enhances oil and gas exploration in these areas.

Abstract Background Gastric cancer (GC) ranks fifth in global cancer incidence and third in mortality rate among all cancer types. Circular RNAs (circRNAs) have been extensively demonstrated to regulate multiple malignant biological behaviors in GC. Emerging evidence suggests that several circRNAs derived from FNDC3B play pivotal roles in cancer. However, the role of circFNDC3B in GC remains elusive. Methods We initially screened circFNDC3B with translation potential via bioinformatics algorithm prediction. Subsequently, Sanger sequencing, qRT-PCR, RNase R, RNA-FISH and nuclear-cytoplasmic fractionation assays were explored to assess the identification and localization of circ0003692, a circRNA derived from FNDC3B. qRT-PCR and ISH were performed to quantify expression of circ0003692 in human GC tissues and adjacent normal tissues. The protein-encoding ability of circ0003692 was investigated through dual-luciferase reporter assay and LC/MS. The biological behavior of circ0003692 in GC was confirmed via in vivo and in vitro experiments. Additionally, Co-IP and rescue experiments were performed to elucidate the interaction between the encoded protein and c-Myc. Results We found that circ0003692 was significantly downregulated in GC tissues. Circ0003692 had the potential to encode a novel protein FNDC3B-267aa, which was downregulated in GC cells. We verified that FNDC3B-267aa, rather than circ0003692, inhibited GC migration in vitro and in vivo. Mechanistically, FNDC3B-267aa directly interacted with c-Myc and promoted proteasomal degradation of c-Myc, resulting in the downregulation of c-Myc-Snail/Slug axis. Conclusions Our study revealed that the novel protein FNDC3B-267aa encoded by circ0003692 suppressed GC metastasis through binding to c-Myc and enhancing proteasome-mediated degradation of c-Myc. The study offers the potential applications of circ0003692 or FNDC3B-267aa as therapeutic targets for GC. Graphical abstract The mechanism of circ0003692 in suppressing metastasis of GC. Abstract Background Clear cell renal cell carcinoma (ccRCC) is the most prevalent kidney cancer with high aggressive phenotype and poor prognosis. Accumulating evidence suggests that circRNAs have been identified as pivotal mediators in cancers. However, the role of circRNAs in ccRCC progression remains elusive. Methods The differentially expressed circRNAs in 4 paired human ccRCC and adjacent noncancerous tissues ccRCC were screened using circRNA microarrays and the candidate target was selected based on circRNA expression level using weighted gene correlation network analysis (WGCNA) and the gene expression omnibus (GEO) database. CircPDHK1 expression in ccRCC and adjacent noncancerous tissues (n = 148) were evaluated along with clinically relevant information. RT-qPCR, RNase R digestion, and actinomycin D (ActD) stability test were conducted to identify the characteristics of circPDHK1. The subcellular distribution of circPDHK1 was analyzed by subcellular fractionation assay and fluorescence in situ hybridization (FISH). Immunoprecipitation-mass spectrometry (IP-MS) and immunofluorescence (IF) were employed to evaluate the protein-coding ability of circPDHK1. ccRCC cells were transfected with siRNAs, plasmids or lentivirus approach, and cell proliferation, migration and invasion, as well as tumorigenesis and metastasis in nude mice were assessed to clarify the functional roles of circPDHK1 and its encoded peptide PDHK1-241aa. RNA-sequencing, western blot analysis, immunoprecipitation (IP) and chromatin immunoprecipitation (ChIP) assays were further employed to identify the underlying mechanisms regulated by PDHK1-241aa. Results CircPDHK1 was upregulated in ccRCC tissues and closely related to WHO/ISUP stage, T stage, distant metastasis, VHL mutation and Ki-67 levels. CircPDHK1 had a functional internal ribosome entry site (IRES) and encoded a novel peptide PDHK1-241aa. Functionally, we confirmed that PDHK1-241aa and not the circPDHK1 promoted the proliferation, migration and invasion of ccRCC. Mechanistically, circPDHK1 was activated by HIF-2A at the transcriptional level. PDHK1-241aa was upregulated and interacted with PPP1CA, causing the relocation of PPP1CA to the nucleus. This thereby inhibited AKT dephosphorylation and activated the AKT-mTOR signaling pathway. Conclusions Our data indicated that circPDHK1-encoded PDHK1-241aa promotes ccRCC progression by interacting with PPP1CA to inhibit AKT dephosphorylation. This study provides novel insights into the multiplicity of circRNAs and highlights the potential use of circPDHK1 or PDHK1-241aa as a therapeutic target for ccRCC.

Abstract With an estimated one million new cases reported annually, gastric cancer (GC) ranks as the fifth most diagnosed malignancy worldwide. The early detection of GC remains a major challenge, and the prognosis worsens either when patients develop resistance to chemotherapy or radiotherapy or when the cancer metastasizes. The precise pathogenesis underlying GC is not well understood, which further complicates its treatment. Circular RNAs (circRNAs), a recently discovered class of noncoding RNAs that originate from parental genes through “back-splicing”, have been shown to play a key role in various biological processes in both eukaryotes and prokaryotes. CircRNAs have been linked to cardiovascular diseases, diabetes, hypertension, Alzheimer's disease, and the occurrence and progression of tumors. Prior studies have established that circRNAs play a crucial role in GC, impacting tumorigenesis, diagnosis, progression, and therapy resistance. This review aims to summarize how circRNAs contribute to GC tumorigenesis and progression, examine their roles in the development of drug resistance, discuss their potential as biotechnological drugs, and summarize their response to therapeutic drugs and microorganism in GC.

Abstract Background As stroke has become the leading cause of death and disability in China, it has induced a heavy disease burden on society, families, and patients. Despite much attention within the literature, the effect of multiple risk factors on length of stay (LOS) and inpatient costs in China is still not fully understood. Aim To analyse the association between the number of risk factors combined and inpatient costs among adults with stroke and explore the mediating effect of LOS on inpatient costs. Methods A retrospective cross-sectional study was conducted among stroke patients in a tertiary hospital in Nantong City from January 2018 to December 2019. Lifestyle factors (smoking status, exercise), personal disease history (overweight, hypertension, dyslipidemia, diabetes mellitus, atrial fibrillation), family history of stroke, and demographic characteristics were interviewed by trained nurses. Inpatient costs and LOS were extracted from electronic medical records. Hierarchical multiple linear regression models and mediation analysis were used to examine the direct and indirect effects of the number of risk factors combined for stroke on inpatient costs. Results A total of 620 individuals were included, comprising 391 ischaemic stroke patients and 229 haemorrhagic stroke patients, and the mean age was 63.2 years, with 60.32% being male. The overall mean cost for stroke inpatients was 30730.78 CNY ($ 4444.91), and the average length of stay (LOS) was 12.50 days. Mediation analysis indicated that the greater number of risk factors was not only directly related to higher inpatient costs (direct effect = 0.16, 95%CI:[0.11,0.22]), but also indirectly associated with inpatient cost through longer LOS (indirect effect = 0.08, 95% CI: [0.04,0.11]). Furthermore, patients with high risk of stroke had longer LOS than those in low-risk patients, which in turn led to heavier hospitalization expenses. Conclusions Both the greater number of risk factors and high-risk rating among stroke patients increased the length of stay and inpatient costs. Preventing and controlling risk behaviors of stroke should be strengthened.

Analcime plays a significant role in sandstone reservoirs as an authigenic diagenetic mineral in the Junggar Basin (northwestern China). However, the origin and controls on the reservoirs have received remarkably little attention. This study investigates the formation mechanism of analcime in the Middle Permian strata in the Jinan sag (southern Junggar Basin) through petrography and geochemistry. The results show that analcime is formed through early alkaline hydrolysis of volcanic materials under specific temperature and pressure conditions. The reservoir rocks primarily consist of various lithic sandstones, including volcanic debris such as basalt, andesite, and tuff. Analcime is characterized as rich in aluminium and poor in sodium, classified as low-silica analcime with a low Si-Al ratio (1.98–2.38). Furthermore, various other diagenetic minerals, such as glauconite, chlorite, albite, and calcite have been identified. The primary reservoir space chiefly consists of intragranular dissolved pores of analcime, while secondary pores are formed by intragranular pores of feldspar and lithic, along with some remaining intergranular pores. Cementation of analcime during early diagenesis changes primary pore structures and reduces reservoir properties. The low-silica analcime dissolves due to acidic pore fluids associated with three stages of oil and gas charging, transforming into albite and creating numerous secondary pores, thereby enhancing reservoir quality.

Bladder cancer is one of the most common malignancies of the urinary tract and a prevalent cancer worldwide, still requiring efficient therapeutic agents and approaches. 20-Hydroxyecdysone (20-HE), a steroid hormone, can be found in insects and few plants and mediate numerous biological events to control the progression of varying diseases; however, its impacts on bladder cancer remain unclear. In the study, we found that 20-HE treatments effectively inhibited the viability and proliferation of bladder cancer cells and induced apoptosis by activating Caspase-3. The migratory and invasive potential of bladder cancer cells was markedly repressed by 20-HE in a dose-dependent manner. The inhibitory effects of 20-HE on bladder cancer were confirmed in an established xenograft mouse model, as indicated by the markedly reduced tumor growth rates and limited lung and lymph node metastasis. High-throughput RNA sequencing was performed to explore dysregulated genes in bladder cancer cells after 20-HE treatment. We identified ubiquitin-specific protease 21 (USP21) as a key deubiquitinating enzyme for bladder cancer progression and a positive correlation between USP21 and nuclear factor-κB (NF-κB)/p65 in patients. Furthermore, 20-HE treatments markedly reduced USP21 expression, NF-κB/p65 mRNA, stability and phosphorylated NF-κB/p65 expression levels in bladder cancer cells, which were validated in animal tumor tissues. Mechanistic studies showed that USP21 directly interacted with and stabilized p65 by deubiquitinating its K48-linked polyubiquitination in bladder cancer cells, which could be abolished by 20-HE treatment, contributing to p65 degradation. Finally, we found that USP21 overexpression could not only facilitate the proliferation, migration, and invasion of bladder cancer cells, but also significantly eliminated the suppressive effects of 20-HE on bladder cancer. Notably, 20-HE could still perform its anti-tumor role in bladder cancer when USP21 was knocked down with decreased NF-κB/p65 expression and activation, revealing that USP21 suppression might not be the only way for 20-HE during bladder cancer treatment. Collectively, all our results clearly demonstrated that 20-HE may function as a promising therapeutic strategy for bladder cancer treatment mainly through reducing USP21/p65 signaling expression.

Artemisia argyi, a perennial herb of the genus Artemisia in the family Asteraceae, holds significant importance in Chinese traditional medicine, referred to as “Aicao”. Here, we report a high-quality reference genome of Artemisia argyi L. cv. beiai, with a genome size up to 4.15 Gb and a contig N50 of 508.96 Kb, produced with third-generation Nanopore sequencing technology. We predicted 147,248 protein-coding genes, with approximately 68.86% of the assembled sequences comprising repetitive elements, primarily long terminal repeat retrotransposons(LTRs). Comparative genomics analysis shows that A. argyi has the highest number of specific gene families with 5121, and much more families with four or more members than the other 6 plant species, which is consistent with its more expanded gene families and fewer contracted gene families. Furthermore, through transcriptome sequencing of A. argyi in response to exogenous MeJA treatment, we have elucidated acquired regulatory insights into MeJA’s impact on the phenylpropanoid, flavonoid, and terpenoid biosynthesis pathways of A. argyi. The whole-genome information obtained in this study serves as a valuable resource for delving deeper into the cultivation and molecular breeding of A. argyi. Moreover, it holds promise for enhancing genome assemblies across other members of the Asteraceae family. The identification of key genes establishes a solid groundwork for developing new varieties of Artemisia with elevated concentrations of active compounds.

BackgroundInsulin resistance (IR) is closely related to the development of cardiovascular diseases. Triglyceride-glucose-body mass index (TyG-BMI) has been proven to be a reliable surrogate of IR, but the relationship between TyG-BMI and acute myocardial infarction (AMI) is unknown. The present study aims to determine the effects of TyG-BMI on the clinical prognosis of critically ill patients with AMI.MethodsThe data of AMI patients were extracted from the Medical Information Mart for Intensive Care IV (MIMIC-IV) database. All patients were divided into four groups according to the TyG-BMI quartile. Outcomes were defined as 30-, 90-, 180-, and 365-day all-cause mortality. Kaplan–Meier (K-M) curve was used to compare survival rate between groups. Meanwhile, Cox regression analysis and restricted cubic splines (RCS) were used to explore the relationship between TyG-BMI index and outcome events.ResultsA total of 1,188 critically ill patients with AMI were included in this study. They were divided into four groups according to TyG-BMI quartiles, there were significant differences in 90-, 180-, and 365-day all-cause mortality while there was no difference in 30-day all-cause mortality. Interestingly, with the increase of TyG-BMI, the 90-, 180-, and 365-day survival rate increased first and then gradually decreased, but the survival rate after decreasing was still higher than that in the group with the lowest TyG-BMI. U-shaped relationships between TyG-BMI index and 90-, 180-, and 365-day all-cause mortality were identified using RCS curve and the inflection point was 311.1, 316.5, and 320.1, respectively, whereas the TyG-BMI index was not non-linearly associated with 30-day all-cause mortality. The results of Cox proportional hazard regression analysis are consistent with those of RCS analysis.ConclusionU-shaped relationships are existed between the TyG-BMI index and 90-, 180-, and 365-day all-cause mortality in critically ill patients with AMI, but not 30-day all-cause mortality. The TyG-BMI index can be used as an effective index for early prevention of critically ill patients with AMI.

Seeking the available precision limit of unknown parameters is a significant task in quantum parameter estimation. One often resorts to the widely utilized quantum Cramér–Rao bound (QCRB) based on unbiased estimators to finish this task. Nevertheless, most actual estimators are usually biased in the limited number of trials. For this reason, we introduce two effective error bounds for biased estimators based on a unitary evolution process in the framework of the quantum optimal biased bound. Furthermore, we show their estimation performance by two specific examples of the unitary evolution process, including the phase encoding and the SU(2) interferometer process. Our findings will provide an useful guidance for finding the precision limit of unknown parameters.

Abstract Background Hypertension is a main contributing factor of cardiovascular diseases; deregulated circular RNAs are involved in the pathogenesis of pulmonary arterial hypertension (PAH). Herein, we evaluated the function and mechanism of circST6GAL1 in PAH process. Methods Human pulmonary artery smooth muscle cells (HPASMCs) were cultured in hypoxic environment for functional analysis. The cell counting kit‐8, 5‐ethynyl‐2′‐deoxyuridine, wound healing, and flow cytometry assays were used to investigate cell proliferation, migration, and apoptosis. qRT‐PCR and Western blotting analyses were used for level measurement of genes and proteins. The binding between miR‐509‐5p and circST6GAL1 or multiple C2 and transmembrane domain containing 2 (MCTP2) was analyzed by dual‐luciferase reporter, RNA immunoprecipitation, and pull‐down assays. The monocrotaline (MCT)‐induced PAH mouse models were established for in vivo assay. Results CircST6GAL1 was highly expressed in PAH patients and hypoxia‐induced HPASMCs. Functionally, circST6GAL1 deficiency reversed hypoxia‐induced proliferation and migration, as well as apoptosis arrest in HPASMCs. Mechanistically, circST6GAL1 directly targeted miR‐509‐5p, and MCTP2 was a target of miR‐509‐5p. Rescue assays showed that the regulatory effects of circST6GAL1 deficiency on hypoxia‐induced HPASMCs were abolished. Moreover, forced expression of miR‐509‐5p suppressed HPASMC proliferation and migration and induced cell apoptosis under hypoxia stimulation, while these effects were abolished by MCTP2 overexpression. Moreover, circST6GAL1 silencing improved MCT‐induced pulmonary vascular remodeling and PAH. Conclusion CircST6GAL1 deficiency reversed hypoxia‐induced proliferation and migration, as well as apoptosis arrest in HPASMCs, and alleviated pulmonary vascular remodeling in MCT‐induced PAH mouse models through the miR‐509‐5p/MCTP2 axis, indicating a potential therapeutic target for PAH.

The Soil Plant Analysis Development (SPAD) is a vital index for evaluating crop nutritional status and serves as an essential parameter characterizing the reproductive growth status of winter wheat. Non-destructive and accurate monitorin3g of winter wheat SPAD plays a crucial role in guiding precise management of crop nutrition. In recent years, the spectral saturation problem occurring in the later stage of crop growth has become a major factor restricting the accuracy of SPAD estimation. Therefore, the purpose of this study is to use features selection strategy to optimize sensitive remote sensing information, combined with features fusion strategy to integrate multiple characteristic features, in order to improve the accuracy of estimating wheat SPAD. This study conducted field experiments of winter wheat with different varieties and nitrogen treatments, utilized UAV multispectral sensors to obtain canopy images of winter wheat during the heading, flowering, and late filling stages, extracted spectral features and texture features from multispectral images, and employed features selection strategy (Boruta and Recursive Feature Elimination) to prioritize sensitive remote sensing features. The features fusion strategy and the Support Vector Machine Regression algorithm are applied to construct the SPAD estimation model for winter wheat. The results showed that the spectral features of NIR band combined with other bands can fully capture the spectral differences of winter wheat SPAD during the reproductive growth stage, and texture features of the red and NIR band are more sensitive to SPAD. During the heading, flowering, and late filling stages, the stability and estimation accuracy of the SPAD model constructed using both features selection strategy and features fusion strategy are superior to models using only a single feature strategy or no strategy. The enhancement of model accuracy by this method becomes more significant, with the greatest improvement observed during the late filling stage, with R2 increasing by 0.092-0.202, root mean squared error (RMSE) decreasing by 0.076-4.916, and ratio of performance to deviation (RPD) increasing by 0.237-0.960. In conclusion, this method has excellent application potential in estimating SPAD during the later stages of crop growth, providing theoretical basis and technical support for precision nutrient management of field crops.

Digital surface model (DSM) is the fundamental data in various geoscience applications, such as city 3-D modeling and urban environment analysis. The freely available DSM often suffers from limited spatial resolution. Super-resolution (SR) is a promising technique to increase the spatial resolution of DSM. However, most existing SR models struggle to reconstruct spatial details, such as buildings, valleys, and ridges. This article proposes a novel DSM super-resolution (DSMSR) model that integrates high-resolution remote sensing imagery using generative adversarial networks. The generator in DSMSR contains three modules. The first DSM feature extraction module uses the residual-in-residual dense block to extract features from low-resolution DSM. The second multiscale attention feature extraction module employs the pyramid convolutional residual dense blocks to capture the spatial details of ground objects at multiple scales from remote sensing imagery. The third DSM reconstruction module uses a squeeze-and-excitation block to fuse the extracted features from low-resolution DSM and high-resolution remote sensing imagery for generating SR DSM. The discriminator of DSMSR uses the relativistic average discriminator for adversarial learning. The slope loss is further introduced to ensure the accurate representation of topographic features. We evaluate DSMSR on four different terrain regions in the U.K. to downscale the 30-m AW3D30 DSM to 5-m DSM. The experimental results indicate that DSMSR outperforms the traditional interpolation algorithms and four existing deep-learning-based SR models. The DSMSR restores more spatial detail of topographic features and generates more accurate image quality, elevation, and terrain metrics.

The rapid growth of the tourism industry has spurred extensive research into tourist route planning. However, existing studies primarily focus on route planning for individual tourists, leaving a notable gap in addressing multiple tourists planning scenarios. Traditional methods for multiple tourists planning, derived from single tourist frameworks, often prioritize the interests and benefits of tourists. This has led to problems like popularity-biased route planning, which exacerbates overtourism in favored destinations and under-tourism in lesser-known spots, thereby undermining sustainable tourism. To address these challenges, we introduce a multi-agent reinforcement learning (RL) framework specifically designed for multiple tourists route planning, incorporating consideration of tourists’ distribution. Our approach consists of two key components: firstly, a novel tourism RL environment that enables interaction with multiple tourists; secondly, a dual-congestion model that considers both local congestion at tourist spots and overall city-wide tourist’ distribution. This dual-congestion mechanism formulates the reward system in our multi-agent RL framework. We validate our framework with comprehensive experiments using real-world human mobility data from Kyoto, a globally renowned tourist city. The results show our model’s superiority over existing methods in optimizing route rewards and managing tourists’ distribution. Additionally, we conducted a user study to examine the effect of our method on tourists’ experience, serving as a reference for our future implementation. The findings indicate that our dual-congestion model slightly affects tourists who prefer popular spots. Our discussion highlights the typically non-cooperative relationship between tourism sustainability and tourist self-interest, underscoring the necessity for a trade-off between them in practical applications. Significantly, our model demonstrates the potential to transform this non-cooperative relationship into a cooperative dynamic.

Yadong trout (Salmo trutta fario) is an important and characteristic aquaculture fish species in Yadong, Xizang, China. Farmed Yadong trout has been produced on a large scale. Usually, two different-colored eggs (yellow and orange) are produced during Yadong trout farming and show differential reproductive performance. There has been very limited information on the nutrient composition of eggs with different colors, and relevant results have only been reported for rainbow trout. The present study aimed to analyze the nutrient compositions in different-colored Yadong trout eggs to elucidate the factors responsible for the different reproductive performances of the two egg types. The experimental samples were collected from Yadong Industry Park, where the broodstocks were reared with artificial compound feeds. Each egg type was analyzed in 3 replicates. Yellow eggs were produced by 2–3 year-old broodstocks, and orange eggs were produced from 3–5 year-old broodstocks. After spawning, the eggs were cryopreserved and transported to the laboratory by plane. The samples were lyophilized and used for the determination of proximate composition, fatty acids, amino acids, and other reproduction-related nutrients. The crude protein and crude lipid contents were analyzed using the Kjeldahl and Soxhlet extraction methods, respectively. The fatty acid composition (expressed as % total fatty acids) and amino acid composition (expressed as % dry matter) were assayed using gas chromatography and an automatic amino acid analyzer, respectively. The protein carbonyl and malondialdehyde concentration were assayed using commercial kits supplied by Nanjing Jiancheng Bioengineering Institute. The α-vitamin E and astaxanthin were assayed using national standard methods, and the α-vitamin E was measured from wet samples. The statistics of proximate composition, fatty acids, and amino acids data were determined using a t-test for independent samples. The results were expressed as mean ± standard error, and showed that yellow eggs had lower contents of crude protein and ash contents, but higher contents of crude lipid and moisture than orange eggs. There was no significant difference in carbohydrate and energy content between yellow and orange eggs. The saturated fatty acid (SFA), n-3 polyunsaturated fatty acid (n-3 PUFA), and docosahexaenoic acid (DHA) contents of yellow eggs were significantly lower than those of orange eggs, while the n-6 polyunsaturated fatty acids (n-6 PUFA) and EPA contents did not differ significantly between the two egg types. The C16:0 and C20:0 contents of yellow eggs were significantly lower than those of orange eggs, while the contents of monounsaturated fatty acids (MUFA), including C16:1n-7, C18:1n-9, C20:1n-9, and C22:1n-9, of yellow eggs were significantly higher than those of orange eggs. The contents of essential amino acids, non-essential amino acids, and total amino acids were slightly lower in yellow eggs compared to orange eggs. Among the essential amino acids, the contents of valine, methionine, leucine, lysine, and histidine in yellow eggs were significantly lower than those in orange eggs. Among the non-essential amino acids, except for glycine and alanine which were significantly lower in yellow eggs than in orange eggs, the contents of other amino acids did not differ significantly between the two egg types. The contents of protein carbonyl and malondialdehyde in yellow eggs were higher than those in orange eggs, while the contents of α-vitamin E and astaxanthin in yellow eggs were lower than those in orange eggs. In conclusion, orange eggs had higher contents of protein, some essential amino acids, DHA, α-vitamin E, and astaxanthin, but lower contents of MUFA, crude lipid, and peroxidation products (protein carbonyl and malondialdehyde) than yellow eggs. These differences in nutrient composition may contribute to the superior reproductive performance of orange eggs over yellow eggs.

This article proposes a multi-head attention flood forecasting model (MHAFFM) that combines a multi-head attention mechanism (MHAM) with multiple linear regression for flood forecasting. Compared to models based on Long Short-Term Memory (LSTM) neural networks, MHAFFM enables precise and stable multi-hour flood forecasting. First, the model utilizes characteristics of full-batch stable input data in multiple linear regression to solve the problem of oscillation in the prediction results of existing models. Second, full-batch information is connected to MHAM to improve the model's ability to process and interpret high-dimensional information. Finally, the model accurately and stably predicts future flood processes through linear layers. The model is applied to Dawen River Basin, and experimental results show that the MHAFFM, compared to three benchmarking models, namely, LSTM, BOA-LSTM (LSTM with Bayesian Optimization Algorithm for Hyperparameter Tuning), and MHAM-LSTM (LSTM model with MHAM in hidden layer), significantly improves the prediction performance under different lead time scenarios while maintaining good stability and interpretability. Taking Nash–Sutcliffe efficiency index as an example, under a lead time of 3 h, the MHAFFM model exhibits improvements of 8.85, 3.71, and 10.29% compared to the three benchmarking models, respectively. This research provides a new approach for flood forecasting. Aboveground biomass (AGB) is regarded as a critical variable in monitoring crop growth and yield. The use of hyperspectral remote sensing has emerged as a viable method for the rapid and precise monitoring of AGB. Due to the extensive dimensionality and volume of hyperspectral data, it is crucial to effectively reduce data dimensionality and select sensitive spectral features to enhance the accuracy of rice AGB estimation models. At present, derivative transform and feature selection algorithms have become important means to solve this problem. However, few studies have systematically evaluated the impact of derivative spectrum combined with feature selection algorithm on rice AGB estimation. To this end, at the Xiaogang Village (Chuzhou City, China) Experimental Base in 2020, this study used an ASD FieldSpec handheld 2 ground spectrometer (Analytical Spectroscopy Devices, Boulder, Colorado, USA) to obtain canopy spectral data at the critical growth stage (tillering, jointing, booting, heading, and maturity stages) of rice, and evaluated the performance of the recursive feature elimination (RFE) and Boruta feature selection algorithm through partial least squares regression (PLSR), principal component regression (PCR), support vector machine (SVM) and ridge regression (RR). Moreover, we analyzed the importance of the optimal derivative spectrum. The findings indicate that (1) as the growth stage progresses, the correlation between rice canopy spectrum and AGB shows a trend from high to low, among which the first derivative spectrum (FD) has the strongest correlation with AGB. (2) The number of feature bands selected by the Boruta algorithm is 19~35, which has a good dimensionality reduction effect. (3) The combination of FD-Boruta-PCR (FB-PCR) demonstrated the best performance in estimating rice AGB, with an increase in R² of approximately 10% ~ 20% and a decrease in RMSE of approximately 0.08% ~ 14%. (4) The best estimation stage is the booting stage, with R2 values between 0.60 and 0.74 and RMSE values between 1288.23 and 1554.82 kg/hm2. This study confirms the accuracy of hyperspectral remote sensing in estimating vegetation biomass and further explores the theoretical foundation and future direction for monitoring rice growth dynamics.

Clear cell renal cell carcinoma (ccRCC) is the most prevalent kidney cancer, with a highly aggressive phenotype and poor prognosis. RNA binding proteins (RBPs) play crucial roles in post-transcriptional gene regulation and have been implicated in tumorigenesis. RBPs have the potential to become a new therapeutic target for ccRCC. In this study, we screened and validated that insulin-like growth factor 2 mRNA binding protein 2 (IGF2BP2) as an RBP, was down-regulated in ccRCC tissues and cell lines. Functionally, we verified that IGF2BP2 significantly suppressed the migration and invasion ability of ccRCC in vitro and in vivo. Mechanistically, RIP-seq and actinomycin D experiments results showed that IGF2BP2 enhanced the expression of Creatine Kinase B (CKB) by binding to CKB mRNA and enhancing its mRNA stability. Thus, IGF2BP2 inhibited ccRCC metastasis through enhancing the expression of CKB. Taken together, these finding suggests that IGF2BP2 is a novel metastasis suppressor of ccRCC and may serve as a potential therapeutic target.

Thioredoxin reductase (TXNRD) is a selenoprotein that plays a crucial role in cellular antioxidant defense. Previously, a distinctive guiding bar motif was identified in TXNRD1, which influences the transfer of electrons. In this study, utilizing single amino acid substitution and Excitation-Emission Matrix (EEM) fluorescence spectrum analysis, we discovered that the guiding bar communicates with the FAD and modulates the electron flow of the enzyme. Differential Scanning Fluorimetry (DSF) analysis demonstrated that the aromatic amino acid in guiding bar is a stabilizer for TXNRD1. Kinetic analysis revealed that the guiding bar is vital for the disulfide reductase activity but hinders the selenocysteine-independent reduction activity of TXNRD1. Meanwhile, the guiding bar shields the selenocysteine residue of TXNRD1 from the attack of electrophilic reagents. We also found that the inhibition of TXNRD1 by caveolin-1 scaffolding domain (CSD) peptides and compound LCS3 did not bind to the guiding bar motif. In summary, the obtained results highlight new aspects of the guiding bar that restrict the flexibility of the C-terminal redox motif and govern the transition from antioxidant to pro-oxidant.

Summary: Background: Azvudine and nirmatrelvir/ritonavir are approved to treat mild-to-moderate coronavirus disease 2019 (COVID-19) in adults with a high risk for progression to severe infection. We sought to compare the antiviral effectiveness and clinical outcomes of elderly severe patients with COVID-19 receiving these two antiviral agents. Methods: In this observational study, we identified 249 elderly patients with severe COVID-19 infection who were admitted to the Second Medical Center of the People's Liberation Army General Hospital from December 2022 to January 2023, including 128 azvudine recipients, 66 nirmatrelvir/ritonavir recipients and 55 patients not received antiviral treatments. We compared the cycle threshold (Ct) value dynamic change of all three groups. The primary outcome was a composite outcome of disease progression, including all-cause death, intensive care unit admission, and initiation of invasive mechanical ventilation. The outcomes of all enrolled patients were followed up from the electronic medical record system. Kaplan–Meier and Cox risk proportional regression analyses were used to compare the clinical outcomes of all three groups. To more directly compare the effectiveness of the two antiviral drugs, we performed propensity-score matching between the two antiviral groups and compared antiviral efficacy and clinical outcomes in the matched population. Findings: Among 249 patients (mean age, 91.41 years), 77 patients died during the follow-up period. When compared to patients who did not receive any antivirals, neither nirmatrelvir/ritonavir nor azvudine demonstrated a survival benefit. The Cox analysis of the all-cause death of the three groups showed that the risk of death was 0.730 (0.423–1.262) in the azvudine group 0.802 (0.435–1.480) and in the nirmatrelvir/ritonavir group compared with the non-antiviral group. After propensity score matching, we included 58 azvudine recipients and 58 nirmatrelvir/ritonavir recipients. The fitted curve of the Ct value after matching illustrated that the rate of viral decline in the early stage of nirmatrelvir/ritonavir treatment seems to surpass that of azvudine, but there was no statistical significance. Azvudine was seemly associated with a lower risk of composite outcomes (HR:1.676, 95% CI:0.805–3.488) and short-term all-cause death (HR: 1.291, 95%CI: 0.546–3.051). Interpretation: Patients who received azvudine have a similar antiviral effectiveness and survival curve trend compared to nirmatrelvir/ritonavir. In this limited series, antiviral treatment was not associated with a significant clinical benefit. This lack of clinical benefit might be attributed to potential bias. As the “blood vessels” and “nerves” of the power grid, cables are important carriers for transmitting electric energy, and their failures will affect the safe and stable operation of the power grid. In this paper, a digital twin model of thermal characteristics is constructed for 10 kV YJV×400 cluster cable and its joint model. Based on the multi-physical field coupling analysis method, the fluid field, temperature field and current-carrying capacity of the cluster cable are analyzed. The temperature rise and airflow distribution characteristics of the joint and its external insulation when the defect of joint false connection occurs are analyzed. Based on the fuzzy analytic hierarchy process (FAHP), weights are assigned to the locations of multiple measurement points of the cable external insulation, and the temperature distribution of each measurement point is combined with the digital twin technology to calculate the hot spot temperature of the cable, so as to assess the defects of the joints of the cables. The digital twin platform is used for real-time monitoring of cable status and assessment of defects. The temperature rise experiments of cables under different carrying capacity and joint defects show that this method can accurately and efficiently calculate hot spot temperatures and assess defects of joints in cluster cables.

Fish meal and fish oil are important sources of protein and lipid in feeds. The lack of fish meal and fish oil supply has become an important limiting factor for the aquaculture industry. The proportion of fish oil and fish meal used in commercial feeds for farmed fish and crustaceans is continually decreasing, while the carbohydrate content is increasing. Carbohydrates are one of the three major nutrients that provide energy for the body. They have a much lower production price than protein and fat. Starch is a polymeric carbohydrate made from glucose molecules; it is the most commonly used in aquatic feeds and can replace fish meal and fish oil to reduce feed cost. Therefore, it is important to fully explore the nutritional function of carbohydrates for aquaculture. Hypoxia is a common stress in aquaculture, and acute hypoxia may lead to massive mortality of cultured fish in a short period of time resulting in serious economic losses to aquaculture. Therefore, it is crucial to find ways to improve the acute hypoxia tolerance of fish. Fish mainly use glucose for energy under acute hypoxia. Hypoxia inhibits oxidative phosphorylation in mitochondria and activates the anaerobic glycolysis pathway. Glucose degrades to produce lactate and ATP. Carnivorous fishes have limited absorption and utilization capacities to feed carbohydrates, unlike omnivorous fishes. Dextrin is an intermediate starch hydrolysis product, with a molecular weight between starch and glucose; it has good adhesion properties and is more easily digested and absorbed than starch. Therefore, we hypothesized that the use of easily digestible carbohydrate in the feed is an effective way to improve the acute hypoxia tolerance of fish. Takifugu rubripes is loved by Japanese and Korean consumers owing to its delicious taste and high nutritional value. It is a characteristic species of Chinese mariculture fish. It is mainly cultured in a high-density factory, and the dissolved oxygen in the water often relies on water exchange and an oxygenation pump; its gill cover is degraded and is at risk of acute hypoxia. This study determined the effect of the addition of corn starch or dextrin (corn starch hydrolysate) to the feed on growth performance, acute hypoxia survival rate, metabolite content, and the hypoxia inducible factor (HIF) signaling pathway of T. rubripes after 8 weeks of feeding. There were no significant differences in weight gain, feed conversion ratio, hepatosomatic index, viscerosomatic index, condition factor, and body composition between the starch group and the dextrin group (P > 0.05). However, the survival rate during acute hypoxia of the dextrin group was significantly higher than that of the starch group (P < 0.05). There were no significant differences in the liver glycogen and lactate contents, and lactate dehydrogenase A4 (ldha) gene expression between the starch group and dextrin group (P > 0.05) during normoxia. However, the lactate content and ldha gene expression were significantly higher in the liver of the dextrin group than those in the starch group (P < 0.05) during hypoxia. This indicated that feeding dextrin strongly activated anaerobic glycolysis to provide more energy under hypoxia. The serum triglyceride (TG) content significantly increased in the dextrin group compared with the normoxia groups, although the TG content in the serum and liver significantly decreased in the starch group after acute hypoxia (P < 0.05). This suggested that feeding starch promoted lipid catabolism and oxygen consumption. There was no significant difference in the total soluble protein content of the muscle between the starch group and the dextrin group (P > 0.05) during normoxia. However, the total soluble protein content of the muscle decreased in the dextrin group compared with the starch group (P < 0.05) after acute hypoxia. Meanwhile, the total muscle soluble protein content, and the gene expression of liver v-akt murine thymoma viral oncogene homolog 1 (akt1), and mechanistic target of rapamycin kinase (mtor) significantly decreased in the dextrin group after hypoxia compared with the normoxia group. However, the liver mtor gene expression significantly increased in the starch group after hypoxia (P < 0.05). This data demonstrated that protein synthesis was inhibited in the dextrin group under hypoxia. Hypoxia inducible factor (HIF) is the most critical transcription factor in cellular response to hypoxic stress. The dextrin group had higher hypoxia inducible factor 1 subunit alpha like (hif-3α) gene expression in the liver and lower hypoxia inducible factor 1 subunit alpha a (hif-1α) gene expression in the muscle (P < 0.05) compared with the starch group during normoxia. The gene expression of liver hif-1α and hif-3α and muscle hif-1α and vascular endothelial growth factor A (vegfa) significantly increased in the dextrin group compared with the starch group (P < 0.05) during hypoxia. This proved that feeding dextrin strongly activated the HIF signaling pathway under hypoxia. In summary, replacing starch with easily digestible dextrin in the feed did not affect the growth performance. Instead, it activated the HIF signaling pathway and anaerobic glycolysis to provide more energy for fish. Meanwhile, feeding dextrin inhibited lipid catabolism and protein synthesis, and reduced oxygen consumption to improve the acute hypoxia tolerance of T. rubripes. The study provides important guidance for the formulation design of hypoxia-tolerant feed and healthy development of aquaculture.

Objective The Jimsar Sag of the Junggar Basin is representative of successful shale oil exploration in China. However, the sedimentary facies in the Permian Lucaogou Formation hosting shale oil has long been disputed. Previous studies proposed two contradictory environments of shallow lakes and semideep to deep lakes in the interior lacustrine basin and identified sandstone interlays as deltaic distal sandbar deposition and beach and bar deposition. Methods The study combined core observations with analyses of sandstone granularity, logging facies, and reservoir properties to identify the development of sublacustrine fans in a semi-deep to deep lake background. Results Fine-grained turbidity currents played a significant role in the transportation and deposition of sediment in these fans. The lithofacies observed included lenticular-wave bedded muddy siltstone to siltstone (dominant type), parallel bedded siltstone, graded siltstone, and massive-graded bedded siltstone to fine-grained sandstone. The cumulative probability granularity curves exhibited smooth upper arches with a high suspension population, and the cross-spots on the C-M diagram were located in the grade suspension zone. The sublacustrine fans consisted of sedimentary microfacies of sheet-like lobes and gravity flow channel levees, showing fining- and coarsening-upwards successions, respectively. These fans contained numerous interlayers of siltstone and fine-grained sandstone, as well as terrigenous detrital particles that influenced the mineral content and enhanced shale oil reservoir properties by promoting the growth of solution pores. Conclusion The study concluded that sublacustrine fan deposits are thin-bedded and fine-grained, and they occur frequently. It also determined that desert reservoirs formed in the areas where sublacustrine fans were deposited, which has significant implications for shale oil exploration in the study area.

This paper proposes a novel welding process for ultrahigh-strength steel. The effects of welding parameters on the welding process and weld formation were studied to obtain the optimal parameter window. It was found that the metal transfer modes of solid wires were primarily determined by electrical parameters, while flux-cored wires consistently exhibited multiple droplets per pulse. The one droplet per pulse possessed better welding stability and weld formation, whereas the short-circuiting transfer or one droplet multiple pulses easily caused abnormal arc ignition that decreased welding stability, which could easily lead to a “sawtooth-shaped” weld formation or weld offset towards one side with more spatters. Thus, the electrical parameters corresponding to one droplet per pulse were identified as the optimal parameter window. Furthermore, the weld zone (WZ) was predominantly composed of AF, and the heat-affected zone (HAZ) primarily consisted of TM and LM. Consequently, the welded joint still exhibited excellent mechanical properties, particularly toughness, despite higher welding heat input. The average tensile strength reached 928 MPa, and the impact absorbed energy at −40 °C for the WZ and HAZ were 54 J and 126 J, respectively. In addition, the application of triple-wire welding for ultrahigh-strength steel (UHSS) demonstrated a significant enhancement in post-weld deposition rate, with increases of 106% and 38% compared to single-wire and twin-wire welding techniques, respectively. This process not only utilized flux-cored wire to enhance the mechanical properties of joints but also achieved high deposition rate welding.

Background: Infiltration of immune cells associated with tumor clinical results affects different cancers. However, the composition and the clinical significance of tumor-infiltrated immune cells in epithelial ovarian cancer has not been completely investigated. Methods: The metagene deconvolution algorithm (Cell type Identification by Estimating Relative Subsets of known RNA Transcripts (CIBERSORT)) was used to analyze gene expression profiles in public databases (GEO and TCGA) to infer the composition of 22 immune cell subgroups in ovarian tumors. The proportions obtained in this study were used to investigate the association between each cell type and the clinical outcomes for ovarian cancer diagnosis and prognosis. Quantitative real-time polymerase chain reaction (qRT-PCR) was used for detecting the expression levels of related genes. Results: The profiles of immune cells infiltration were altered in malignant ovarian neoplastic tissue. Ovarian cancer tissues contained higher proportion of T follicular helper cells (Tfh) and macrophages (M0 and M1) rather than the normal ovarian tissue. Meanwhile, lower proportion of monocytes and neutrophils was also observed in ovarian cancer tissues compared with normal tissues. The qRT-PCR test confirmed the conclusion that the contents of CD80 (M1 cells) and CD4+ (Tfh cells) were high in the interstitium of ovarian cancer tissue, while the contents of CD21 (B cells) and CD66b (neutrophil) were low. Interestingly, immune cell infiltration was observed to be correlated with the change in clinical outcome. The activated mast cell subpopulation was associated with poor prognosis, while the subpopulation of resting dendritic cells was correlated with pathological grade of the ovarian cancer. Conclusions: Our bioanalysis revealed that the composition of tumor-infiltrating immune cells was closely related to the clinical outcome of ovarian cancer, which was also validated in clinical samples. These results provide a new strategic basis for the prognosis and treatment of ovarian cancer.

Recombinant adeno-associated virus (rAAV) has emerged as a prominent vector for in vivo gene therapy, owing to its distinct advantages. Accurate determination of the rAAV genome titer is crucial for ensuring the safe and effective administration of clinical doses. The evolution of the rAAV genome titer assay from quantitative PCR (qPCR) to digital PCR (dPCR) has enhanced accuracy and precision, yet practical challenges persist. This study systematically investigated the impact of various operational factors on genome titration in a single-factor manner, aiming to address potential sources of variability in the quantitative determination process. Our findings revealed that a pretreatment procedure without genome extraction exhibits superior precision compared with titration with genome extraction. Additionally, notable variations in titration results across different brands of dPCR instruments were documented, with relative standard deviation (RSD) reaching 23.47% for AAV5 and 11.57% for AAV8. Notably, optimal operations about DNase I digestion were identified; we thought treatment time exceeding 30 min was necessary, and there was no need for thermal inactivation after digestion. And we highlighted that thermal capsid disruption before serial dilution substantially affected AAV genome titers, causing a greater than ten-fold decrease. Conversely, this study found that additive components of dilution buffer are not significant contributors to titration variations. Furthermore, we found that repeated freeze–thaw cycles significantly compromised AAV genome titers. In conclusion, a comprehensive dPCR titration protocol, incorporating insights from these impact factors, was proposed and successfully tested across multiple serotypes of AAV. The results demonstrate acceptable variations, with the RSD consistently below 5.00% for all tested AAV samples. This study provides valuable insights to reduce variability and improve the reproducibility of AAV genome titration using dPCR.

Leaf nitrogen concentration (LNC) is a primary indicator of crop nitrogen status, closely related to the growth and development dynamics of crops. Accurate and efficient monitoring of LNC is significant for precision field crop management and enhancing crop productivity. However, the biochemical properties and canopy structure of wheat change across different growth stages, leading to variations in spectral responses that significantly impact the estimation of wheat LNC. This study aims to investigate the construction of feature combination indices (FCIs) sensitive to LNC across multiple wheat growth stages, using remote sensing data to develop an LNC estimation model that is suitable for multiple growth stages. The research employs UAV multispectral remote sensing technology to acquire canopy imagery of wheat during the early (Jointing stage and Booting stage) and late (Early filling and Late filling stages) in 2021 and 2022, extracting spectral band reflectance and texture metrics. Initially, twelve sensitive spectral feature combination indices (SFCIs) were constructed using spectral band information. Subsequently, sensitive texture feature combination indices (TFCIs) were created using texture metrics as an alternative to spectral bands. Machine learning algorithms, including partial least squares regression (PLSR), random forest regression (RFR), support vector regression (SVR), and Gaussian process regression (GPR), were used to integrate spectral and texture information, enhancing the estimation performance of wheat LNC across growth stages. Results show that the combination of Red, Red edge, and Near-infrared bands, along with texture metrics such as Mean, Correlation, Contrast, and Dissimilarity, has significant potential for LNC estimation. The constructed SFCIs and TFCIs both enhanced the responsiveness to LNC across multiple growth stages. Additionally, a sensitive index, the Modified Vegetation Index (MVI), demonstrated significant improvement over NDVI, correcting the over-saturation concerns of NDVI in time-series analysis and displaying outstanding potential for LNC estimation. Spectral information outperforms texture information in estimation capability, and their integration, particularly with SVR, achieves the highest precision (coefficient of determination (R2) = 0.786, root mean square error (RMSE) = 0.589%, and relative prediction deviation (RPD) = 2.162). In conclusion, the sensitive FCIs developed in this study improve LNC estimation performance across multiple growth stages, enabling precise monitoring of wheat LNC. This research provides insights and technical support for the construction of sensitive indices and the precise management of nitrogen nutrition status in field crops.

Reinforcement soil slope with pile penetration is a new load bearing structure, which has a complex working mechanism, but few studies have been carried out. This paper aims to investigate the stability characteristics of this structure using model tests. The study investigates the lateral displacement and-pile bending moment caused by vertical loads and evaluates the influence of different factors, including the structure type (such as pile, cap of pile, and reinforcement material), number of reinforcing layers, spacing of reinforcement material, pile length, and slope rate on the load-carrying capacity of the pile penetration fill-reinforced load-bearing structure. The findings suggest that within a certain range, increasing the pile length and number of reinforcing layers, the limiting effect of the pile on the lateral displacement in the middle and at the bottom of the slope of the pile-penetrating reinforced structure is enhanced, which can reduce the extreme value of the bending moment and make the distribution of the bending moment of the pile more reasonable. The lateral limiting effect on the soil body can be maximized by appropriate reinforcement spacing. Within a certain range, the slope rate is reduced, which can reduce the extreme value of the bending moment, make the bending moment distribution of the pile more reasonable, and avoid the phenomenon of the localized force concentration of the pile.

BackgroundPrevious retrospective studies have shown a correlation between depression and increased risk of infections, including a moderate rise in sepsis likelihood associated with severe depression and anxiety. To investigate the potential causal links between depression, sepsis, and mortality risks, while considering confounding factors, we employed a Mendelian randomization (MR) approach.MethodsIn this two-sample Mendelian randomization study, we analyzed data from a large-scale genome-wide association study on depression, involving 807,553 European individuals (246,363 cases, 561,190 controls). We extracted SNP associations with sepsis and 28-day mortality from UK Biobank GWAS outcomes. The correlation analysis primarily employed the inverse-variance weighted method, supplemented by sensitivity analyses for heterogeneity and pleiotropy assessment.ResultsOur analysis revealed a potential causal link between depression and an increased risk of sepsis (OR = 1.246, 95% CI: 1.076-1.442, P = 0.003), but no causal association was found with sepsis-induced mortality risk (OR = 1.274, 95% CI: 0.891-1.823, P = 0.184). Sensitivity analyses confirmed the robustness of these findings.ConclusionsWe identified a potential causal association between depression and heightened sepsis risk, while no link was found with sepsis-induced mortality. These findings suggest that effective management of depression could be important in preventing sepsis.

This study aimed to evaluate the effects of combined replacement of fishmeal (FM) and fish oil (FO) with poultry byproduct meal (PBM) and mixed oil (MO, poultry oil: coconut oil = 1 : 1) on growth performance, body composition and muscle quality of tiger puffer (Takifugu rubripes). Fish with an average initial body weight of 14.29 g were selected for the feeding experiment. FM accounting for 0%, 5%, and 10% of the diet was replaced by PBM. For each grade of FM replacement, 5% FO or MO was used as added oil. The six experimental diets were designated as FO-FM, MO-FM, FO-5PBM, MO-5PBM, FO-10PBM, and MO-10PBM, respectively. Each treatment was performed in triplicate with 30 fish per replicate. The feeding period was 45 days. There was no significant difference in growth performance among the groups. Dietary supplementation of both PBM and MO had marginal effects on whole-fish proximate composition, except that dietary MO supplementation significantly increased the liver moisture content. In serum, there were no significant differences in contents of triglyceride, total cholesterol, total bile acid, and protein carbonyl among groups, but the malondialdehyde content was reduced by MO. The fatty acid composition in fish mirrored those in the diets, but the omega-3 sparing effects of saturated and monounsaturated fatty acid in MO can still be observed. Dietary PBM and MO had marginal effects on free amino acid composition and texture of fish muscle, but exerted complicated effects on the muscle volatile flavor compound composition. In conclusion, combined fishmeal (10% of the diet) and fish oil (5% of the diet) replacement with poultry byproduct and mixed oil (poultry oil + coconut oil) had no adverse effects on the growth performance and body proximate composition of farmed tiger puffer. However, these replacements changed the muscle flavor compound profile.

ABSTRACT This study aimed to develop and validate a simple-to-use nomogram model to assess the risk of infection caused by carbapenem-resistant Klebsiella pneumoniae (CRKP) in neurocritically ill patients. The clinical data of 544 patients with CRKP infection admitted to a neurointensive care unit (neuro-ICU) were retrospectively analyzed. The demographic data, laboratory test results, and clinical characteristics of patients in the neuro-ICU were collected. Subsequently, multivariate regression analysis was used to construct a nomogram to predict the risk of CRKP infection in these patients. The calibration ability, clinical effectiveness, and discriminative ability of the nomogram were evaluated. The incidence of CRKP infection was estimated to be 6.43%, and a majority of bacterial isolates causing the infection were found in sputum (74.3%). Multivariate regression analysis showed that the number of antibiotics of ≥2 [odds ratio (OR): 9.08, 95% confidence interval (CI): 2.78–29.71], undergoing surgery (OR: 3.84, 95% CI: 1.09–13.54), and long neuro-ICU stay (OR: 1.08, 95% CI: 1.01–1.14) were associated with CRKP infection in neurocritically ill patients. The nomogram model demonstrated good calibration and discrimination in both the training and validation sets, with area under the curve values of 0.860 and 0.907, respectively. This study developed and validated a nomogram that combines three easily accessed variables during clinical practice to predict the risk of nosocomial CRKP infection in neuro-ICU patients. The tool demonstrated a good predictive performance and discrimination, which might serve as a useful tool to facilitate early detection and reduction of the CRKP infection risk in neurocritically ill patients. IMPORTANCE Patients in neuro-ICU are at a high risk of developing nosocomial CRKP infection owing to complex conditions, critical illness, and frequent invasive procedures. However, studies focused on constructing prediction models for assessing the risk of CRKP infection in neurocritically ill patients are lacking at present. Therefore, this study aims to establish a simple-to-use nomogram for predicting the risk of CRKP infection in patients admitted to the neuro-ICU. Three easily accessed variables were included in the model, including the number of antibiotics used, surgery, and the length of neuro-ICU stay. This nomogram might serve as a useful tool to facilitate early detection and reduction of the CRKP infection risk of neurocritically ill patients.

BackgroundResearch has shown that the disordered serum lipid profile may be associated with the risk of differentiated thyroid cancer (DTC). Whether this association reflect causal effect is still unclear. The aim of this study was to evaluate the causality of circulating lipoprotein lipids on DTC.MethodsMendelian randomization (MR) analysis was conducted to evaluate the relationship between the circulating lipoprotein lipids and DTC risk using single-nucleotide polymorphisms (SNPs) from a genome-wide association (GWA) study containing a high-incidence Italian population of 690 cases samples with DTC and 497 controls.ResultsUnivariate and multivariate mendelian randomization analysis demonstrated that ‘total cholesterol’, ‘HDL cholesterol’, ‘apolipoprotein B’ and ‘ratio of apolipoprotein B to apolipoprotein A1’ were correlated with DTC. According to sensitivity analysis, our results were reliable. Furthermore, multivariate analysis revealed that there is no causative association between DTC and any of the many cause factors when they interact with one another, suggesting that there was a deep interaction between the four factors, which could affect each other. Finally, the mechanism of the related effects each other as well as the target genes with significant SNP regulatory effects in DTC was explored by conducting functional enrichment analysis and constructing the regulatory networks.ConclusionsWe obtained four exposure factors (total cholesterol, HDL cholesterol, apolipoprotein B and ratio of apolipoprotein B to apolipoprotein A1) closely related to DTC, which laid a theoretical foundation for the treatment of DTC.

Objective Forkhead box a2 (Foxa2) is proven to be an insulin-sensitive transcriptional regulator and affects hepatic steatosis. This study aims to investigate the mechanism by which Foxa2 affects nonalcoholic fatty liver disease (NAFLD). Methods Animal and cellular models of NAFLD were constructed using high-fat diet (HFD) feeding and oleic acid (OA) stimulation, respectively. NAFLD mice received tail vein injections of either an overexpressing negative control (oe-NC) or Foxa2 (oe-Foxa2) for four weeks. HepG2 cells were transfected with oe-NC and oe-Foxa2 for 48 h before OA stimulation. Histological changes and lipid accumulation were assessed using hematoxylin-eosin staining and oil red O staining, respectively. Expression of Foxa2, NF-κB/IKK pathway proteins, lipid synthesis proteins, and fatty acid β-oxidation protein in HFD mice and OA-induced HepG2 cells was detected using western blot. Results Foxa2 expression was downregulated in HFD mice and OA-induced HepG2 cells. Foxa2 overexpression attenuated lipid accumulation and liver injury, and reduced the levels of aspartate aminotransferase, alanine aminotransferase, total cholesterol, or triglyceride in HFD mice and OA-induced HepG2 cells. Moreover, Foxa2 overexpression decreased the expression of lipid synthesis proteins and increased fatty acid β-oxidation protein expression in the liver tissues. Furthermore, overexpression of Foxa2 downregulated the expression of p-NF-κB/NF-κB and p-IKK/IKK in OA-induced HepG2 cells. Additionally, lipopolysaccharide (NF-κB/IKK pathway activator) administration reversed the downregulation of lipid synthesis proteins and the upregulation of fatty acid β-oxidation protein. Conclusion Foxa2 expression is downregulated in NAFLD. Foxa2 ameliorated hepatic steatosis and inhibited the activation of the NF-κB/IKK signaling pathway.

The trade-off of electro-induced shape-changing hydrogels (EISCHs) between mechanical properties and deformation ability limited its further applications. In this work, the double network technique of reinforcing nondeformable hydrogels’ mechanical properties was introduced into EISCHs, and the NIPAAm/CNTs/Fe(phen)3 double network low-voltage-induced shape-changing hydrogel (DN) was synthesized. The experimental results indicated that the DN gained the balance between mechanical properties and deformation ability compared to the NIPAAm-Fe(phen)3 single network and NIPAAm-Fe(phen)3-CNTs nanocomposite single network hydrogels. The DN was found to have the ability to undergo swelling and shrinking as a result of the transition between Fe(phen)3 network iron (Ⅱ) and iron (Ⅲ) states under low-voltage stimulation. The hydrogen bonding and physical entanglement structure between CNTs and polymer chain segments, and physical entanglements between different network, which enable effective energy dissipation. Meanwhile, the special double network structure enabled the hydrogel could decrease the resistance to deformation stress by the slide between two networks.

Carbohydrates are a common and economical energy source in animal feeds. However, most fish show a persistent postprandial hyperglycemia after intake of a high-carbohydrate diet. Unfortunately, the mechanism of glucose metabolism in fish is still unclear. In the present study, tiger puffer (Takifugu rubripes) and turbot (Scophthalmus maximus) were intraperitoneally injected or orally administered with glucose or fructose (500 mg/kg body weight) to evaluate the ability of fish to utilize carbohydrates. Afterwards, serum glucose, fructose, pyruvate, insulin levels, and acute hypoxia tolerance were measured. Our results showed increased serum glucose level and then decreased post intraperitoneal injection with glucose, and reached a peak after 0.5 hours in turbot and 1 hour in tiger puffer. Tiger puffer had significantly lower liver glycogen, serum glucose, fructose, pyruvate, and insulin contents than turbot. Glucose and fructose only induced insulin secretion in turbot, but did not change serum insulin level in tiger puffer. Glucose was a stronger stimulator of insulin than fructose in the two marine species. Both intraperitoneal injection and oral fructose intake increased serum glucose level, while intraperitoneal or oral glucose also increased serum fructose level. Intraperitoneal injection of glucose promoted absorption and utilization of glucose in the blood more effectively than oral glucose intake. In addition, turbot and tiger puffer were intolerant to acute hypoxia, whereas supplementation with glucose or fructose improved hypoxia tolerance in the two marine fishes by activating anaerobic glycolysis. Taken together, our results provide important scientific information for understanding the mechanism for glucose and fructose utilization and improving hypoxia tolerance in fish.

The increasing concurrences of heatwaves and droughts in the context of global warming have attracted much attention from the scientific community given their devastating social and environmental impacts. In this study, the effects of heatwaves in each adjacent week of flash drought onset on the intensification rate of soil moisture were quantified through a meta-Gaussian-based conditional probability model. Results showed that both heatwaves and flash droughts have become more frequent since the middle of the 1990s. For the seasonal distributions, except for the southwestern region where flash droughts lagged behind heatwaves, there was a good synchronization between the two climate extremes. Strong correlations between heatwaves and flash droughts were found in the northeastern, northern, and southwestern regions. Heatwaves with varied timing of emergence behave differently on the formation of flash droughts, along with significant regional differences. Short-term impending hot conditions were crucial for the breakout of flash droughts, especially for the week when flash droughts were initiated, the emergence of heatwaves was likely to increase the intensification rate of soil moisture by 20% compared to those with no heatwaves in their development stage. HIGHLIGHTS The composite patterns of concurrent heatwaves and flash droughts are investigated.; Conditional probability is adopted to quantify the effect of heatwaves on drought.; Heatwaves increase the intensification rate of drought events by about 20%.;

Abstract Background Long working hours and effort-reward imbalance (ERI) among medical workers may contribute to poor mental health and reduced productivity. However, the potential mechanisms among them are not well understood. This study aimed to explore the role of depressive symptoms and ERI in the relationship between long working hours and presenteeism among village doctors. Methods We conducted a cross-sectional study in Jiangsu Province, eastern China. 705 village doctors were assessed for working hours, ERI (the Effort-Reward Imbalance Questionnaire, ERI questionnaire), presenteeism (6-item Stanford Presenteeism Scale, SPS-6 Scale), and depressive symptoms (12-item General Health Questionnaire, GHQ-12). A moderated mediation model was used to test the role of depressive symptoms (M), and ERI (W) in the relationship between long working hours (X) and presenteeism (Y). Results 45.11% of the village doctors worked more than 55 h per week, and 55.89% were exposed to ERI. The prevalence of depressive symptoms among Chinese village doctors was 40.85%. Abstract Background Insulin like growth factor II mRNA binding protein 3 (IGF2BP3) is an RNA binding protein with multiple roles in regulation of gene expression at the post-transcriptional level and is implicated in tumorigenesis and progression of numerous cancers including gastric cancer (GC). Circular RNAs (circRNAs) are a diverse endogenous noncoding RNA population that have important regulatory roles in cancer. However, circRNAs that regulate the expression of IGF2BP3 in GC is largely unknown. Methods CircRNAs that bound to IGF2BP3 were screened in GC cells using RNA immunoprecipitation and sequencing (RIP-seq). The identification and localization of circular nuclear factor of activated T cells 3 (circNFATC3) were identified using Sanger sequencing, RNase R assays, qRT-PCR, nuclear-cytoplasmic fractionation and RNA-FISH assays. CircNFATC3 expression in human GC tissues and adjacent normal tissues were measured by qRT-PCR and ISH. The biological role of circNFATC3 in GC was confirmed by in vivo and in vitro experiments. Furthermore, RIP, RNA-FISH/IF, IP and rescue experiments were performed to uncover interactions between circNFATC3, IGF2BP3 and cyclin D1 (CCND1). Results We identified a GC-associated circRNA, circNFATC3, that interacted with IGF2BP3. CircNFATC3 was significantly overexpressed in GC tissues and was positively associated with tumor volume. Functionally, the proliferation of GC cells decreased significantly after circNFATC3 knockdown in vivo and in vitro. Mechanistically, circNFATC3 bound to IGF2BP3 in the cytoplasm, which enhanced the stability of IGF2BP3 by preventing ubiquitin E3 ligase TRIM25-mediated ubiquitination, thereby enhancing the regulatory axis of IGF2BP3-CCND1 and promoting CCND1 mRNA stability. Conclusions Our findings demonstrate that circNFATC3 promotes GC proliferation by stabilizing IGF2BP3 protein to enhance CCND1 mRNA stability. Therefore, circNFATC3 is a potential novel target for the treatment of GC.

According to the latest drilling and the analysis of the burial history, source rock evolution history and hydrocarbon accumulation history, the sub-source hydrocarbon accumulation characteristics of the Permian reservoirs in the Jinan Sag, eastern Junggar Basin, are clarified, and the hydrocarbon accumulation model of these reservoirs is established. The results are obtained in four aspects. First, the main body of the thick salified lake basin source rocks in the Lucaogou Formation has reached the mature stage with abundant resource base. Large-scale reservoirs are developed in the Jingjingzigou, Wutonggou and Lucaogou formations. Vertically, there are multiple sets of good regional seals, the source-reservoir-caprock assemblage is good, and there are three reservoir-forming assemblages: sub-source, intra-source and above-source. Second, dissolution, hydrocarbon charging and pore-preserving effect, and presence of chlorite film effectively increase the sub-source pore space. Oil charging is earlier than the time when the reservoir becomes densified, which improves the efficiency of hydrocarbon accumulation. Third, buoyancy and source-reservoir pressure difference together constitute the driving force of oil charging, and the micro-faults within the formation give the advantage of “source-reservoir lateral docking” under the source rock. Microfractures can be critical channels for efficient seepage and continuous charging of oil in different periods. Fourth, the Jingjingzigou Formation experienced three periods of oil accumulation in the Middle–Late Permian, Middle–Late Jurassic and Late Neogene, with the characteristics of long-distance migration and accumulation in early stage, mixed charging and accumulation in middle stage and short-distance migration and high-position accumulation in late stage. The discovery and theoretical understanding of the Permian reservoirs in the Jinan Sag reveal that the thrust belt has good conditions for forming large reservoirs, and it is promising for exploration. The study results are of guidance and reference significance for oil and gas exploration in the Jinan Sag and other geologically similar areas.

Abstract Background Sertoli cell-only syndrome (SCOS) is the most serious pathological type of non-obstructive azoospermia. Recently, several genes related to SCOS have been identified, including FANCM, TEX14, NR5A1, NANOS2, PLK4, WNK3, and FANCA, but they cannot fully explain the pathogenesis of SCOS. This study attempted to explain spermatogenesis dysfunction in SCOS through testicular tissue RNA sequencing and to provide new targets for SCOS diagnosis and therapy. Methods We analyzed differentially expressed genes (DEGs) based on RNA sequencing of nine patients with SCOS and three patients with obstructive azoospermia and normal spermatogenesis. Abstract Salicylic acid (SA) and jasmonic acid (JA) are plant hormones that typically act antagonistically in dicotyledonous plants and SA and JA signaling is often manipulated by pathogens. However, in monocotyledonous plants, the detailed SA-JA interplay in response to pathogen invasion remains elusive. Here, we show that different types of viral pathogen can disrupt synergistic antiviral immunity mediated by SA and JA via OsNPR1 in the monocot rice. The P2 protein of rice stripe virus, a negative-stranded RNA virus in the genus Tenuivirus, promotes OsNPR1 degradation by enhancing the association of OsNPR1 and OsCUL3a. OsNPR1 activates JA signaling by disrupting the OsJAZ-OsMYC complex and boosting the transcriptional activation activity of OsMYC2 to cooperatively modulate rice antiviral immunity. Unrelated viral proteins from different rice viruses also interfere with the OsNPR1-mediated SA-JA interplay to facilitate viral pathogenicity, suggesting that this may be a more general strategy in monocot plants. Overall, our findings highlight that distinct viral proteins convergently obstruct JA-SA crosstalk to facilitate viral infection in monocot rice.

Abstract Background Rootstocks can improve the chilling tolerance of grafted cucumbers, but their effectiveness varies. Rootstocks with strong de-blooming capacity may result in lower chilling tolerance of grafted cucumbers compared to those with weak de-blooming capacity, while also reducing the silicon absorption. However, it remains unclear whether this reduction in chilling tolerance is due to differences in rootstock genotypes or the reduction in silicon absorption. Results The chilling tolerance of cucumber seedlings was improved by using rootstocks and silicon nutrition. Rootstocks had a more significant effect than silicon nutrition, and the weak de-blooming rootstock ‘Yunnan figleaf gourd’ was superior to the strong de-blooming rootstock ‘Huangchenggen No. 2’. Compared to self-rooted cucumber, twelve miRNAs were regulated by two rootstocks, including seven identical miRNAs (novel-mir23, novel-mir26, novel-mir30, novel-mir37, novel-mir46, miR395a and miR398a-3p) and five different miRNAs (novel-mir32, novel-mir38, novel-mir65, novel-mir78 and miR397a). Notably, four of these miRNAs (novel-mir38, novel-mir65, novel-mir78 and miR397a) were only identified in ‘Yunnan figleaf gourd’-grafted cucumbers. Furthermore, six miRNAs (miR168a-5p, miR390a-5p, novel-mir26, novel-mir55, novel-mir67 and novel-mir70) were found to be responsive to exogenous silicon. Target gene prediction for 20 miRNAs resulted in 520 genes. Functional analysis of these target genes showed that ‘Yunnan figleaf gourd’ improves the chilling tolerance of cucumber by regulating laccase synthesis and sulfate metabolism, while ‘Huangchenggen No. 2’ and exogenous silicon reduced chilling stress damage to cucumber by regulating ROS scavenging and protein protection, respectively. Conclusion Among the identified miRNAs, novel-mir46 and miR398a-3p were found in cucumbers in response to chilling stress and two types of rootstocks. However, no identical miRNAs were identified in response to chilling stress and silicon. In addition, the differential expression of novel-mir38, novel-mir65, novel-mir78 and miR397a may be one of the important reasons for the differences in chilling tolerance of grafted cucumbers caused by two types of rootstocks.

Astaxanthin (AST), functioning as an efficient antioxidant and pigment, is one of the most expensive additives in shrimp feeds. How to improve the uptake efficiency of dietary astaxanthin into farmed shrimp is of significance. The present study investigated the effects of lysophosphatidylcholine (LPC), an emulsifier, on dietary astaxanthin efficiency, growth performance, body color, body composition, as well as lipid metabolism of juvenile Pacific white shrimp (average initial body weight: 2.4 g). Three diets were prepared: control group, the AST group (supplemented with 0.02% AST), and the AST + LPC group (supplemented with 0.02% AST and 0.1% LPC). Each diet was fed to triplicate tanks, and each tank was stocked with 30 shrimp. The shrimp were fed four times daily for eight weeks. The AST supplementation improved the growth of white shrimp, while LPC further promoted the final weight of shrimp, but the whole-shrimp proximate composition and fatty acid composition were only slightly affected by AST and LPC. The LPC supplementation significantly increased the astaxanthin deposition in the muscle. The LPC supplementation significantly increased the shell yellowness of both raw and cooked shrimp compared to the AST group. Moreover, the dietary LPC increased the high-density lipoprotein-cholesterol content but decreased the low-density lipoprotein-cholesterol content in the serum, indicating the possible regulation of lipid and cholesterol transport. The addition of astaxanthin significantly up-regulated the expression of npc2 in the hepatopancreas compared to the control group, while the addition of LPC down-regulated the expression of mttp compared to the AST group. In conclusion, the LPC supplementation could facilitate the deposition of dietary astaxanthin into farmed shrimp and further enlarge the beneficial effects of dietary astaxanthin. LPC may also independently regulate shrimp body color and cholesterol transportation. This was the first investigation of the promoting effects of LPC on dietary astaxanthin efficiency.

The lattice Boltzmann method is employed in the current study to simulate the heat transfer characteristics of sinusoidal-temperature-distributed heat sources at the bottom of a square cavity under various conditions, including different amplitudes, phase angles, initial positions, and angular velocities. Additionally, a machine learning-based model is developed to accurately predict the Nusselt number in such a sinusoidal temperature distribution of heat source at the bottom of a square cavity. The results indicate that (1) in the phase angle range from 0 to π, Nu basically shows a decreasing trend with an increase in phase angle. The decline in Nu at an accelerated rate is consistently observed when the phase angle reaches 4π/16. The corresponding Nu decreases as the amplitude increases at the same phase angle. (2) The initial position of the sinusoidal-temperature-distributed heat source Lc significantly impacts the convective heat transfer in the cavity. Moreover, the decline in Nu was further exacerbated when Lc reached 7/16. (3) The optimal overall heat transfer effect was achieved when the angular velocity of the non-uniform heat source reached π. As the angular velocity increases, the local Nu in the square cavity exhibits a gradual and oscillatory decline. Notably, it is observed that Nu at odd multiples of π surpasses that at even multiples of π. Furthermore, the current work integrates LBM with machine learning, enabling the development of a precise and efficient prediction model for simulating Nu under specific operational conditions. This research provides valuable insights into the application of machine learning in the field of heat transfer.

Lard (LD) and Basa fish offal oil (BFO) have similar fatty acid profiles, both containing high contents of saturated fatty acids (SFA) and monounsaturated fatty acids (MUFA). The present study aimed to investigate the efficacy of partial or complete replacement of marine fish oil (MFO, herring oil) by LD or BFO in the diets of tiger puffer. The control diet contained 49.1% crude protein and 9.28% crude lipid content including 6% added MFO. In other diets, 1/3, 2/3, and 3/3 of the added MFO was replaced by LD or BFO, respectively. Each diet was fed to triplicate tanks of juvenile fish (initial body weight, 13.88 g). A 46-day feeding trial was conducted in a flow-through seawater system. Each diet was fed to triplicate 200-L rectangular polyethylene tanks, each of which was stocked with 30 fish. Fish were fed to satiation three times a day. The complete replacement of added MFO (replacing 65% of the total crude lipid) had no adverse effects on fish growth performance in terms of survival (>94%), weight gain (360–398%), feed intake (2.37–3.04%), feed conversion ratio (0.84–1.02), and somatic indices. The dietary LD or BFO supplementation also had marginal effects on fish body proximate composition, biochemical parameters, muscle texture, and water-holding ability, as well as the hepatic expression of lipid metabolism-related genes. Partial (2/3) replacement of added MFO by LD or BFO did not significantly reduce the muscle n-3 LC-PUFA content, indicating the n-3 LC-PUFA sparing effects of SFA and MUFA in LD and BFO. In general, dietary LD or BFO reduced the peroxidation level and led to significant changes in the muscle volatile flavor compound profile, which were probably attributed to the change in fatty acid composition. The results of this study evidenced that LD and BFO are good potential lipid sources for tiger puffer feeds.
