Irrigation water quality is important to sustain agricultural productivity. The primary irrigation water sources in Al-Ahsa Oasis, KSA, are groundwater wells, mixed with treated wastewater and agricultural drainage. This study sought to evaluate irrigation water quality in Al-Ahsa Oasis with the aid of using irrigation water quality indices (IWQIs). One hundred and forty-eight different water samples were collected from various irrigation water resources throughout Al-Ahsa Oasis. The investigated physiochemical characteristics include pH, temperature, TDS, EC, turbidity, free chlorine, total hardness, cations (Na, K, Ca, Mg), anions (Cl, CO3, HCO3, SO4), organic matter indices (DO, BOD, COD), and nutrients (NH4, NO3, PO4). The IWQIs used in this study include salinity hazard, sodium adsorption ratio (SAR), Kelly's ratio (KR), soluble sodium percentage (SSP), Permeability index (PI), residual sodium carbonate (RSC), and magnesium hazard (MH). The results indicated that treated wastewater mixed with groundwater is acceptable for irrigation. Spatial variations in irrigation water quality throughout Al-Ahsa are associated with water resources. For instance, groundwater mixed with agricultural drainage has the highest values of TDS, cations, and anions, whereas the lowest values were reported for treated wastewater, reflecting the good efficiency of wastewater treatment plants. The IWQI results revealed that 4.1% and 62.1% of the investigated irrigation water samples were considered good (class III) and satisfactory (class IV) for irrigation, respectively, whereas 33.8% of the collected water samples fall within the severe irrigation restrictions. Moreover, 79.7% of the investigated water samples were classified to have high to very high salinity hazards (C3, C4) and medium to high sodium hazards (S2, S3). Regular monitoring and assessment of treated water quality and wastewater treatment plant efficiency are important factors in achieving the sustainability of treated wastewater reuse in irrigation and consequently food security. Abstract Malaria remains a global health challenge, disproportionately affecting vulnerable communities. Despite substantial progress, the emergence of anti-malarial drug resistance poses a constant threat. The Greater Mekong Subregion (GMS), which includes Cambodia, China’s Yunnan province, Lao People's Democratic Republic, Myanmar, Thailand, and Viet Nam has been the epicentre for the emergence of resistance to successive generations of anti-malarial therapies. From the perspective of the World Health Organization (WHO), this article considers the collaborative efforts in the GMS, to contain Plasmodium falciparum artemisinin partial resistance and multi-drug resistance and to advance malaria elimination. The emergence of artemisinin partial resistance in the GMS necessitated urgent action and regional collaboration resulting in the Strategy for Malaria Elimination in the Greater Mekong Subregion (2015–2030), advocating for accelerated malaria elimination interventions tailored to country needs, co-ordinated and supported by the WHO Mekong malaria elimination programme. The strategy has delivered substantial reductions in malaria across all GMS countries, with a 77% reduction in malaria cases and a 97% reduction in malaria deaths across the GMS between 2012 and 2022. Notably, China was certified malaria-free by WHO in 2021. Countries' ownership and accountability have been pivotal, with each GMS country outlining its priorities in strategic and annual work plans. The development of strong networks for anti-malarial drug resistance surveillance and epidemiological surveillance was essential. Harmonization of policies and guidelines enhanced collaboration, ensuring that activities were driven by evidence. Challenges persist, particularly in Myanmar, where security concerns have limited recent progress, though an intensification and acceleration plan aims to regain momentum. Barriers to implementation can slow progress and continuing innovation is needed. Accessing mobile and migrant populations is key to addressing remaining transmission foci, requiring effective cross-border collaboration. In conclusion, the GMS has made significant progress towards malaria elimination, particularly in the east where several countries are close to P. falciparum elimination. New and persisting challenges require sustained efforts and continued close collaboration. The GMS countries have repeatedly risen to every obstacle presented, and now is the time to re-double efforts and achieve the 2030 goal of malaria elimination for the region.

Ecotourism has the potential to support protected areas (PAs) financing where PAs’ visitor centers (VCs) are mechanisms for raising public awareness, and revenue generation. Park agencies must shift from a sole emphasis on preservation to include management approaches that increase the benefits of ecotourism and promote partnerships, collaboration, and integration of tourist activities into protected areas core business. A strategy is needed for Wadi El Gemal National Park (WGNP) to enhance tourism practices in a manner that will protect ecosystems, benefit the local community, and promote sustainability. Visitor centers contribution to the development and sustainability of PAs and ecotourism is rarely investigated. This article sheds the light on PAs’ visitor centers and their role in ecotourism, conservation, and PAs financial sustainability. It defines the potential ecotourism products and services in WGNP and its Visitor Center, elucidates the impacts of COVID-19 on the park and the local community, and identifies the necessary tools and resources to implement a sustainable business and management model for WGNP Visitor Center. The results of the VC’s probability analysis and the financial indicators suggest that the Visitor Center is financially feasible and could present a model of sustainable revenue generation and ecotourism development in WGNP.

Antibody-oligonucleotide conjugates (AOCs) are a fast-expanding modality for targeted delivery of therapeutic oligonucleotides to tissues. Here, we present a protocol to generate, purify, and analyze AOCs from off-the-shelf antibodies. We describe steps to conjugate single/double-stranded oligonucleotides bearing amine handles to linkers and, then, to antibodies using well-established chemistry. In addition, we provide details regarding the purification techniques and analytical methods suitable for AOC. This protocol can be applied for several purposes where AOC is a modality of interest. Double-stranded RNAs (dsRNA)-based strategies appeared as promising therapies to induce an inflammation in the tumor microenvironment. However, currently described systems generally lack active targeting of tissues, and their clinical translation is thus limited to intratumoral injection. Herein, we developed an antibody-siRNA-5'triphosphate conjugate with multiple modes of action, combining cell surface EphA2-specific internalization, leading to a simultaneous gene silencing and activation of the receptor retinoic acid-inducible gene I (RIG-I). Recognition of cytosolic siRNA-5'triphosphate by RIG-I triggers the expression of interferons and pro-inflammatory cytokines, inducing an inflammation of the tumor environment and activating neighboring immune cells. In addition, these RIG-I-specific effects synergized with siRNA-mediated PLK1 silencing to promote cancer cell death by apoptosis. Cleavable linkers have become the subject of intense study in the field of chemical biology, particularly because of their applications in the construction of antibody-drug conjugates (ADC), where they facilitate lysosomal cleavage and liberation of drugs from their carrier protein. Due to lysosomes' acidic nature, acid-labile motifs have attracted much attention, leading to the development of hydrazone and carbonate linkers among several other entities. Continuing our efforts in designing new moieties, we present here a family of cyclic acetals that exhibit excellent plasma stability and acid lability, notably in lysosomes. Incorporated in ADC, they led to potent constructs with picomolar potency in vitro and similar in vivo efficacy as the commercially available ADC Kadcyla in mouse xenograft models.
