In a preregistered, cross-sectional study, we investigated whether olfactory loss is a reliable predictor of COVID-19 using a crowdsourced questionnaire in 23 languages to assess symptoms in individuals self-reporting recent respiratory illness. We quantified changes in chemosensory abilities during the course of the respiratory illness using 0-100 visual analog scales (VAS) for participants reporting a positive (C19+; n = 4148) or negative (C19-; n = 546) COVID-19 laboratory test outcome. Logistic regression models identified univariate and multivariate predictors of COVID-19 status and post-COVID-19 olfactory recovery. Both C19+ and C19- groups exhibited smell loss, but it was significantly larger in C19+ participants (mean ± SD, C19+: -82.5 ± 27.2 points; C19-: -59.8 ± 37.7). Smell loss during illness was the best predictor of COVID-19 in both univariate and multivariate models (ROC AUC = 0.72). Additional variables provide negligible model improvement. VAS ratings of smell loss were more predictive than binary chemosensory yes/no-questions or other cardinal symptoms (e.g., fever). Olfactory recovery within 40 days of respiratory symptom onset was reported for ~50% of participants and was best predicted by time since respiratory symptom onset. We find that quantified smell loss is the best predictor of COVID-19 amongst those with symptoms of respiratory illness. To aid clinicians and contact tracers in identifying individuals with a high likelihood of having COVID-19, we propose a novel 0-10 scale to screen for recent olfactory loss, the ODoR-19. We find that numeric ratings ≤2 indicate high odds of symptomatic COVID-19 (4 < OR < 10). Once independently validated, this tool could be deployed when viral lab tests are impractical or unavailable.

Recent anecdotal and scientific reports have provided evidence of a link between COVID-19 and chemosensory impairments, such as anosmia. However, these reports have downplayed or failed to distinguish potential effects on taste, ignored chemesthesis, and generally lacked quantitative measurements. Here, we report the development, implementation, and initial results of a multilingual, international questionnaire to assess self-reported quantity and quality of perception in 3 distinct chemosensory modalities (smell, taste, and chemesthesis) before and during COVID-19. In the first 11 days after questionnaire launch, 4039 participants (2913 women, 1118 men, and 8 others, aged 19-79) reported a COVID-19 diagnosis either via laboratory tests or clinical assessment. Importantly, smell, taste, and chemesthetic function were each significantly reduced compared to their status before the disease. Difference scores (maximum possible change ±100) revealed a mean reduction of smell (-79.7 ± 28.7, mean ± standard deviation), taste (-69.0 ± 32.6), and chemesthetic (-37.3 ± 36.2) function during COVID-19. Qualitative changes in olfactory ability (parosmia and phantosmia) were relatively rare and correlated with smell loss. Importantly, perceived nasal obstruction did not account for smell loss. Furthermore, chemosensory impairments were similar between participants in the laboratory test and clinical assessment groups. These results show that COVID-19-associated chemosensory impairment is not limited to smell but also affects taste and chemesthesis. The multimodal impact of COVID-19 and the lack of perceived nasal obstruction suggest that severe acute respiratory syndrome coronavirus strain 2 (SARS-CoV-2) infection may disrupt sensory-neural mechanisms.

Context: Dietary zinc oxide (ZnO) (a bitter antimicrobial chemical) in pigs is being gradually phased out due to pollution and antibiotic resistance. Bitter compounds like quinine hydrochloride (HCl) have shown potential to enhance growth and feed efficiency by slowing gastric emptying and intestinal passage rates in pigs. Aims: This study aimed to evaluate quinine's ability to improve performance in weanling pigs without ZnO. Methods: Two experiments were conducted. Experiment 1: 120 Landrace (LD) × Large White (LW) weaned piglets (initial BW 6.8 ± 0.1 kg) were randomly assigned to one of four diets in a 2 × 2 factorial design: with/without ZnO and copper sulfate (CuSO4) (3000 and 250 ppm, respectively) and two quinine levels (0 and 500 ppm). Parameters measured included average daily feed intake (ADFI), average daily gain (ADG), gain to feed ratio (G:F), and faecal score. Experiment 2: 1440 LD × LW piglets (initial BW 7.4 ± 0.2 kg) were housed in groups of 18 per pen and assigned the same four dietary treatments as in Experiment 1. Key results: Growth performance parameters were recorded and analysed, showing that ZnO/CuSO4 supplement improved growth and feed efficiency (P < 0.05) compared to the ZnO/CuSO4-free diet group. In Experiment 1, pigs supplemented with quinine in non-ZnO/CuSO4 diets showed similar (P > 0.05) performance levels to the ZnO/CuSO4 fed group. In addition, an interaction (P < 0.05) was found, indicating that adding quinine improved or worsened ADG and G:F depending on the absence or presence of ZnO/CuSO4 in the diet, respectively. In Experiment 2, quinine inclusions in non-ZnO diets improved (P < 0.05) ADG but did not affect (P > 0.05) ADFI and G:F. Conclusion: Our findings suggest that the anticipated deleterious effects of phasing out the use of dietary ZnO can be partially compensated by includingquinine in the diet of post-weaning pigs. The negative effect of quinine when provided together with ZnO is compatible with a competitive exclusion mechanism linked to both stimulating bitterness, a mechanism that warrants further investigation. Implications: Quinine shows potential as a partial replacement for ZnO in post-weaning pig diets, providing a promising alternative to maintain piglet health and growth while transitioning away from ZnO. Quinine shows promise in improving the sustainability of pig farming by enhancing piglet growth while addressing environmental concerns linked to zinc oxide. Our studies revealed that quinine, comparable to traditional supplements, not only boosts piglet growth but also does so independently of zinc oxide showing promise as a viable alternative. Simple Summary: This study examines the impact of replacing 50% of soybean meal (SBM) with untreated cottonseed meal (CSM), fermented CSM (FCSM), or electron beam-irradiated CSM (ICSM) on the growth performance and physiological characteristics of broiler chickens. The findings revealed that broilers fed ICSM achieved higher body weight than those fed CSM. Furthermore, substituting 50% of SBM with FCSM significantly improved growth performance and nutrient digestibility compared to CSM and ICSM diets. In addition, including FCSM in the diet enhanced digestive enzyme activity, balanced the microbiota population, and reduced ammonia emissions compared to other experimental diets. Overall, FCSM provided superior nutritional benefits compared to both untreated and irradiated CSM in broiler diets. Therefore, FCSM can be used as a suitable alternative to SBM in broiler chicken diets. This study was conducted to investigate and compare the effects of substituting soybean meal (SBM) with untreated cottonseed meal (CSM), fermented CSM (FCSM), or electron beam-irradiated CSM (ICSM) on the growth performance, cecal microbiota, digestive enzyme activity, apparent ileal digestibility (AID), and excreta gas emission of broiler chickens. A total of 384 one-day-old male broiler chickens were randomly assigned to four experimental diets, with eight replicates per diet and 12 birds per replicate, for six weeks. The experimental diets consisted of a control diet based on corn–SBM and three other diets in which 50% of the SBM (control) was substituted with CSM in its raw, irradiated, and fermented forms. The results showed that throughout the entire rearing period, feeding broiler chickens with ICSM significantly increased average daily gain (ADG) and body weight (BW) compared to the CSM diet (p < 0.05). Replacing 50% of SBM with FCSM led to a significant improvement in BW, ADG, and feed conversion ratio (FCR) compared to the CSM and ICSM diets (p < 0.05). Interestingly, no significant differences in BW, ADG, or FCR were observed between birds fed FCSM and those on the control diet (p > 0.05). Birds fed FCSM diets exhibited the lowest pH value in the crop, ileum, and ceca. Substituting SBM with FCSM significantly reduced Escherichia coli and Clostridium spp. counts in the ceca, while enhancing the presence of Lactobacillus spp. (p < 0.05). The AID of protein and ether extract was higher in the FCSM group than in the CSM and ICSM groups (p < 0.05). Compared to the CSM diet, ICSM feeding improved protein digestibility (p < 0.05). Broiler chickens on the FCSM diet exhibited higher intestinal amylase and protease activity than those on the other diets (p < 0.05). Furthermore, feeding diets containing FCSM significantly reduced ammonia emissions compared to the other diets (p < 0.05). Overall, our results indicated that microbial fermentation of CSM is a more effective approach than irradiation for enhancing the nutritional value of CSM. Therefore, FCSM is recommended as a viable alternative protein source that can safely replace up to 50% of SBM in broiler chicken diets, particularly during times of fluctuating SBM prices and availability issues. Aims: In 2019, the Australian Academy of Science in collaboration with the nutrition community published the decadal plan for the science of nutrition. This article aims to review progress towards each of its pillar goals (societal determinants, nutrition mechanisms, precision and personalised nutrition, and education and training) and two enabling platforms (a national data capability and a trusted voice for nutrition science), prioritise actions, and conceptualise program logic implementation models. This process also brought together public health nutrition researchers to reflect on societal determinants of health, and advise how the next 5 years of the decadal plan could reflect contemporary issues. Methods: Two engagement events, in 2023, brought together experienced and mid‐ and early‐career nutrition professionals for co‐creation of implementation logic models. Results: One hundred and nine early and mid‐career professionals were involved. A revised model for the decadal plan pillars emerged from synthesis of all logic models. This new model integrated the precision and personalised nutrition pillar with nutrition mechanisms pillar. These combined pillars build towards the national data capability enabling platform and created new cross‐cutting themes for education and training. The need arose for greater focus on respectful engagement with Aboriginal and Torres Strait Islander communities and sustained effort to build cross‐disciplinary collaboration to realise the plan's societal determinants goals. A new alliance for nutrition science is proposed to become a unified advocacy voice and build trust in nutrition professionals. Conclusions: A programmatic approach provides a road map for implementing the decadal plan for the final 5 years. [ABSTRACT FROM AUTHOR]

Excess dietary amino acids (AA) has been associated with reduced feed intake, increased satiation, and extended satiety in pigs. Recent ex vivo studies suggested that satiety peptide cholecystokinin (CCK) and insulinotropic glucagon-like peptide 1 (GLP-1), mediated the anorexigenic or insulinotropic effects of Lys, Glu, Phe, Ile, and Leu. However, the ex vivo model limitations require validation in vivo. The aim of the present study was to assess the effect of orally administered AA in vivo in pigs. It was hypothesized that oral Lys, Ile, and Leu have an anorexigenic effect via CCK, while Glu and Phe have an insulinotropic effect increasing circulating levels of GLP-1. Eight entire male pigs (Landrace × Large White) of 18.23 ± 1.06 kg of body weight were administered an oral gavage of water (control) or a 3 mmol/kg of Glu, Ile, Leu, Lys, Phe, or glucose (positive control for GLP-1 release) following an overnight fasting during 5 consecutive days using an incomplete latin square design. Blood samples were collected from the jugular vein before (−5 min, baseline value) and after the gavage (5, 15, 30, 60 and 90 min) to assess CCK and GLP-1 plasma levels. Pigs administered the oral gavage of Leu (P < 0.05), or Lys (P < 0.1) had increased levels of plasma CCK from 0 to 90 min post-gavage when compared to the control. A strong association (P < 0.001) was observed between GLP-1 plasma levels with Phe intake. The impact was significant starting 30 min post-gavage and was sustained until the end of the experiment (90 min post-gavage). Glucose administration increased GLP-1 early after the intake at the 5 min mark (P < 0.1). A positive correlation (P < 0.05, r = 0.89) driven by the impact of Phe at the 60 to 90 min post-gavage was identified between CCK and GLP-1 indicating feedback mechanisms between proximal and distal small intestine. In conclusion, oral gavages of Leu and Lys increased anorexigenic hormone CCK plasma levels in pigs. Phe caused a significant long-lasting increase in incretin GLP-1 plasma levels. Blood CCK and GLP-1 concentrations in Phe gavaged pigs were positively correlated indicating a potential feedback mechanism between proximal (CCK) and distal (GLP-1) small intestine. The present results are compatible with the known anorexigenic effects of excess dietary Leu and Lys, and the insulinotropic effect of Phe in pigs. Excess dietary amino acids (AA) may negatively affect feed intake in pigs. Previous results showed that Lys, Leu, Ile, Phe, and Glu significantly increased gut peptide secretion (i.e. cholecystokinin, glucagon-like peptide 1). However, the link between dietary AA and gut peptide secretion with changes in feeding behavior patterns has not been demonstrated to date in pigs. The aim of the present study was to determine the effect of Lys, Leu, Ile, Phe, and Glu, on feed intake and meal patterns in young pigs. Twelve male pigs (Landrace × Large White, body weight = 16.10 ± 2.69 kg) were administered an oral gavage of water (control) or Lys, Leu, Ile, Phe, Glu, or glucose (positive control) at 3 mmol.kg−1 following an overnight fasting. The experiment consisted in measuring individual feed disappearance and changes in meal pattern (including latency to first meal, first meal duration, intermeal interval, second meal duration, and number of meals) based on video footage. Compared to the control group Lys significantly (P ≤ 0.01) reduced feed intake during the first 30 min and up to 2.5 h post-gavage, including a reduction (P ≤ 0.05) in the first meal duration. Similarly, Leu and Ile also significantly decreased feed intake up to 3 h post-gavage on a cumulative count. However, the strongest (P ≤ 0.01) impacts on feed intake by the two branched chained AA were observed after the first- or second-hour post-gavage for Leu or Ile, respectively. In addition, Leu or Ile did not affect the first meal duration (P ≥ 0.05). Leu significantly increased (P ≤ 0.01) the intermeal interval while decreasing (P ≤ 0.05) the number of meals during the initial 2 h following the gavage when compared with the control group. In contrast, the oral gavages of Phe or Glu had no significant impact (P > 0.05) on the feeding behavior parameters measured relative to the control pigs. In turn, glucose had a short-lived effect on appetite by reducing (P < 0.05) feed intake for 30 min after the first-hour post-gavage. In conclusion, the impact of an oral gavage of Lys on feeding behavior is compatible with a stimulation of early satiation and an increased duration of satiety. The main impact of the oral gavages of Leu and Ile was an increase in the duration of satiety. The gastrointestinal mechanisms associated with non-bound dietary AA sensing and the impact on voluntary feed intake warrant further investigations. [ABSTRACT FROM AUTHOR]
