Objective: To evaluate the expression of matrix metalloproteinase-2 (MMP-2), MMP-9 and tissue inhibitor of metalloproteinase-2 (TIMP-2) in pituitary tumors and investigate their correlation with circulating plasma proteins and cavernous sinus invasion. In addition, the Ki-67 index was also assessed. Methods: Seventy-four patients (37 females) with pituitary adenomas were included, with preoperative peripheral blood collected in 29 cases. Tumor samples were evaluated for MMP-2, MMP-9, TIMP-2 and Ki-67 expression by immunohistochemistry. Protein plasma was semi-quantitatively detected using a commercial membrane antibody array. Results: Sixteen patients presented tumors invading the cavernous sinus. MMP-2 and TIMP-2 were slightly increased in these tumors compared to the noninvasive group, but the difference was not statistically significant. MMP-9 and TIMP-2 plasma concentrations did not correlate with tumor protein expression and also did not differ between the two groups. MMP-2 was not detected in plasma in any case. No statistically significant difference was observed when different tumor subtypes were considered. A significant difference was observed in tumor size (3.4 cm (2.8–4.9) vs 1.9 cm (1.3–2.6); P < 0.001) and in the Ki-67 index (1.8% (0.3–2.5) vs 0.5% (0.2–1.0); P = 0.01) between the invasive and noninvasive groups. Conclusions: In this cohort, we found no significant correlation between tissue and plasma levels of MMP-2, MMP-9 and TIMP-2 and cavernous sinus invasion in pituitary tumors. Further investigation is needed to elucidate the potential role of these markers in the invasiveness of pituitary tumors.

Abstract To connect the protection of natural resources to economic development, environmental rehabilitation is a promising way to repair and compensate for impacts on biodiversity and ecosystem services. Here, we aimed to compare and select potential indicators for the success of different rehabilitating ecosystems to quantify gains in biodiversity and ecosystem services within the Impact Mitigation Hierarchy. We sampled nine environmental variables along rehabilitation chronosequences from rehabilitating (i) iron mining waste piles, (ii) sand quarries, and (iii) compensation areas in the Carajás National Forest. From that, we computed the rehabilitation status, i.e., the proportion of environmental enhancements compared to the overall rehabilitation trajectory, and statistically validated the indicators that best described the status. With a mean rehabilitation status for the oldest rehabilitation stages from waste piles, sand quarries, and compensation areas of 52, 71, and 74%, respectively, we confirmed that rehabilitation activities were able to generate considerable gains in biodiversity. In all the cases, the Shannon diversity, phylogenetic diversity and Leaf Area Index performed better than did the other indices, encouraging the increased use of these indices for upscale monitoring activities. Consistent indicators across distinct projects highlight the importance of maximizing tree diversity and canopy closure in rehabilitation projects to increase biodiversity gains within Impact Mitigation Hierarchy.

Abstract Studies have underscored a growingdemand for innovative practices in the cultivation of seedlings from forest species, with a notable emphasis on the utilization of organic waste, inoculation with arbuscular mycorrhizal fungi (AMF), and phosphate fertilization. This study aimed to evaluate the impact of organic residues, inoculation with AMF, and phosphorus on the growth and quality of Peltophorum dubium (Spreng.) Taub. seedlings. Two independent experiments were conducted. In Experiment I, treatments included inoculation with various AMF species: control (without inoculation), Clareoideoglomus etunicatum, Rhizophagus heterosporum, Rhizophagus clarum, and MIX (a combination of the three AMF species), all in conjunction with varying doses of phosphorus (0, 60, 120, 180, and 240 mg kg-1 soil). In Experiment II, treatments comprised a control group (without AMF) and inoculation with Glomus clarum, Gigaspora margarita, Gigaspora albida, Clareoideoglomus etunicatum, and MIX (a combination of the four AMF species), each associated with four substrates: S1) soil + coarse sand – SCS, S2) SCS + poultry manure, S3) SCS + cattle manure, and S4) SCS + sheep manure. Peltophorum dubium seedlings exhibited heightened growth with the inoculation of R. heterosporum, R. clarum, and MIX. Positive responses were observed in seedlings when exposed to organic residues, particularly sheep manure, resulting in increased biomass production and enhanced Dickson quality index. The AMF inoculation, specifically with R. heterosporum, R. clarum, and MIX, provided optimal growth conditions for P. dubium seedlings. Remarkably, the utilization of organic residues, notably substrates with chicken manure and cattle manure, exerted substantial positive effects on both growth and quality of P. dubium seedlings.

Screening of Pap smear images continues to depend upon cytopathologists’ manual scrutiny, and the results are highly influenced by professional experience, leading to varying degrees of cell classification inaccuracies. In order to improve the quality of the Pap smear results, several efforts have been made to create software to automate and standardize the processing of medical images. In this work, we developed the CEA (Cytopathologist Eye Assistant), an easy-to-use tool to aid cytopathologists in performing their daily activities. In addition, the tool was tested by a group of cytopathologists, whose feedback indicates that CEA could be a valuable tool to be integrated into Pap smear image analysis routines. For the construction of the tool, we evaluate different YOLO configurations and classification approaches. The best combination of algorithms uses YOLOv5s as a detection algorithm and an ensemble of EfficientNets as a classification algorithm. This configuration achieved 0.726 precision, 0.906 recall, and 0.805 F1-score when considering individual cells. We also made an analysis to classify the image as a whole, in which case, the best configuration was the YOLOv5s to perform the detection and classification tasks, and it achieved 0.975 precision, 0.992 recall, 0.970 accuracy, and 0.983 F1-score.

Aquaculture produces more than 122 million tons of fish globally. Among the several economically important species are the Serrasalmidae, which are valued for their nutritional and sensory characteristics. To meet the growing demand, there is a need for automation and accuracy of processes, at a lower cost. Convolutional neural networks (CNNs) are a viable alternative for automation, reducing human intervention, work time, errors, and production costs. Therefore, the objective of this work is to evaluate the efficacy of convolutional neural networks (CNNs) in counting round fish fingerlings (Serrasalmidae) at different densities using 390 color photographs in an illuminated environment. The photographs were submitted to two convolutional neural networks for object detection: one model was adapted from a pre-trained CNN and the other was an online platform based on AutoML. The metrics used for performance evaluation were precision (P), recall (R), accuracy (A), and F1-Score. In conclusion, convolutional neural networks (CNNs) are effective tools for detecting and counting fish. The pre-trained CNN demonstrated outstanding performance in identifying fish fingerlings, achieving accuracy, precision, and recall rates of 99% or higher, regardless of fish density. On the other hand, the AutoML exhibited reduced accuracy and recall rates as the number of fish increased.

Abstract The main objective of this study is to propose different probabilistic models for adjusting the trend component, since it significantly influences the quality of the spatio-temporal interpolation of rainfalls. We used the monthly total precipitation data of the São Francisco River Basin (SFRB) for the period of 31 years, 1989–2019. The SFRB occupies 8% of the whole Brazilian territory, mostly located in the Northeast Brazilian region. For the trend component, we propose the fitted GAMLSS models by comparing different probability distribution families, which in most cases include the characteristics of these data. The results indicate the existence of a spatio-temporal pattern of the residues obtained from the adjustment of the trend with zero adjusted Gamma distribution for the accumulated monthly precipitation. The adjustment revealed a spatial dependence of up to 873 km between the pluviometric stations and temporal autocorrelation of approximately 1.6 months. The methodology used in this study enabled us to create rainfall maps, interpolating unobserved locations in differences years. The projection of these maps to the SFRB is considered extremely important for planning and implementing activities related to water resources across the river basin.

The N-acylhydrazone function has been reported as a pharmacophore group of molecules with diverse pharmacological activities, including anti-inflammatory effects. Therefore, this study was designed to evaluate the anti-inflammatory potential of the compound N′-(3-(1H-indol-3-yl)benzylidene)-2-cyanoacetohydrazide (JR19) in vivo. The study started with the carrageenan-induced peritonitis model, followed by an investigation of leukocyte migration using the subcutaneous air pouch test and an assessment of the antinociceptive profile using formalin-induced pain. A preliminary molecular docking study focusing on the crystallographic structures of NFκB, iNOS, and sGC was performed to determine the likely mechanism of action. The computational study revealed satisfactory interaction energies with the selected targets, and the same peritonitis model was used to validate the involvement of the nitric oxide pathway and cytokine expression in the peritoneal exudate of mice pretreated with L-NAME or methylene blue. In the peritonitis assay, JR19 (10 and 20 mg/kg) reduced leukocyte migration by 59% and 52%, respectively, compared to the vehicle group, with the 10 mg/kg dose used in subsequent assays. In the subcutaneous air pouch assay, the reduction in cell migration was 66%, and the response to intraplantar formalin was reduced by 39%, particularly during the inflammatory phase, suggesting that the compound lacks central analgesic activity. In addition, a reversal of the anti-inflammatory effect was observed in mice pretreated with L-NAME or methylene blue, indicating the involvement of iNOS and sGC in the anti-inflammatory response of JR19. The compound effectively and significantly decreased the levels of IL-6, TNF-α, IL-17, and IFN-γ, and this effect was reversed in animals pretreated with L-NAME, supporting a NO-dependent anti-inflammatory effect. In contrast, pretreatment with methylene blue only reversed the reduction in TNF-α levels. Therefore, these results demonstrate the pharmacological potential of the novel N-acylhydrazone derivative, which acts through the nitric oxide pathway and cytokine signaling, making it a strong candidate as an anti-inflammatory and immunomodulatory agent.

Abstract This study aims to evaluate the in silico genomic characteristics of Streptococcus infantarius subsp. infantarius, isolated from Coalho cheese from Paraíba, Brazil, with a view to application in lactic fermentations. rRNA sequences from the 16S ribosomal region were used as input to GenBank, in the search for patterns that could reveal a non-pathogenic behavior of S. infantarius subsp. infantarius, comparing mobile genetic elements, antibiotic resistance genes, pan-genome analysis and multi-genome alignment among related species. S. infantarius subsp. infantarius CJ18 was the only complete genome reported by BLAST/NCBI with high similarity and after comparative genetics with complete genomes of Streptococcus agalactiae (SAG153, NJ1606) and Streptococcus thermophilus (ST106, CS18, IDCC2201, APC151) revealed that CJ18 showed a low number of transposases and integrases, infection by phage bacteria of the Streptococcus genus, absence of antibiotic resistance genes and presence of bacteriocin, folate and riboflavin producing genes. The genome alignment revealed that the collinear blocks of S. thermophilus ST106 and S. agalactiae SAG153 have inverted blocks when compared to the CJ18 genome due to gene positioning, insertions and deletions. Therefore, the strains of S. infantarius subsp. infantarius isolated from Coalho cheese from Paraíba showed genomic similarity with CJ18 and the mobility of genes analyzed in silico showed absence of pathogenicity throughout the genome of CJ18, indicating the potential of these strains for the dairy industry.

Abstract Dryophthorinae is an economically important, ecologically distinct, and ubiquitous monophyletic group of pantropical weevils with more than 1,200 species in 153 genera. This study provides the first comprehensive phylogeny of the group with the aim to provide insights into the process and timing of diversification of phytophagous insects, inform classification and facilitate predictions. The taxon sampling is the most extensive to date and includes representatives of all five dryophthorine tribes and all but one subtribe. The phylogeny is based on secondary structural alignment of 18S and 28S rRNA totaling 3,764 nucleotides analyzed under Bayesian and maximum likelihood inference. We used a fossil‐calibrated relaxed clock model with two approaches, node‐dating and fossilized birth‐death models, to estimate divergence times for the subfamily. All tribes except the species‐rich Rhynchophorini were found to be monophyletic, but higher support is required to ascertain the paraphyly of Rhynchophorini with more confidence. Nephius is closely related to Dryophthorini and Stromboscerini, and there is strong evidence for paraphyly of Sphenophorina. We find a large gap between the divergence of Dryophthorinae from their sister group Platypodinae in the Jurassic‐Cretaceous boundary and the diversification of extant species in the Cenozoic, highlighting the role of coevolution with angiosperms in this group.

de Medeiros and Farrell test how insect-host interactions influence species divergence in phytophagous insects. Using ddRAD-seq data and hierarchical modelling, they demonstrate, surprisingly, that antagonistic interactions between insect and host do not lead to greater species diversification.

Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) is responsible for causing Coronavirus Disease-2019 (COVID-19), a heterogeneous clinical condition that manifests varying symptom severity according to the demographic profile of the studied population. While many studies have focused on the spread of COVID-19 in large urban centers in Brazil, few have evaluated medium or small cities in the Northeast region. The aims of this study were: (i) to identify risk factors for mortality from SARS-CoV-2 infection, (ii) to evaluate the gene expression patterns of key immune response pathways using nasopharyngeal swabs of COVID-19 patients, and (iii) to identify the circulating SARS-CoV-2 variants in the residents of a medium-sized city in Northeast Brazil. A total of 783 patients infected with SARS-CoV-2 between May 2020 and August 2021 were included in this study. Clinical-epidemiological data from patients who died and those who survived were compared. Patients were also retrospectively divided into three groups based on disease severity: asymptomatic, mild, and moderate/severe. Samples were added to a qPCR array for analyses of 84 genes involved with immune response pathways and sequenced using the Oxford Nanopore MinION technology. Having pre-existing comorbidity; being male; having cardiovascular disease, diabetes, and/or chronic obstructive pulmonary disease; and PCR cycle threshold (Ct) values under 22 were identified as risk factors for mortality. Analysis of the expression profiles of inflammatory pathway genes showed that the greater the infection severity, the greater the activation of inflammatory pathways, triggering the cytokine storm and downregulating anti-inflammatory pathways. Viral genome analysis revealed the circulation of multiple lineages, such as B.1, B.1.1.28, Alpha, and Gamma, suggesting that multiple introduction events had occurred over time. This study’s findings help identify the specific strains and increase our understanding of the true state of local health. In addition, our data demonstrate that epidemiological and genomic surveillance together can help formulate public health strategies to guide governmental actions.

Abstract Aiming to enrich the knowledge about the flora of savannas, this paper studied the composition and structure of the bryophyte community of Park Savanna areas in Marajó Island - PA. Biological material was collected within 60 100-m2 plots equally distributed in the dry season of 2016 and the rainy season of 2017 in five Park Savanna areas (SP-I to SP-V). The composition, density, richness and diversity of species and presence of indicator species were compared between the sampled areas and seasons. The species were classified according to the substrates colonized and ecological groups of light tolerance. Significant differences in SP-V indicated that the area was the main factor influencing the composition of bryophytes (p: 0.0001), with five indicator species. There were also significant differences in density (p = 0.0001168) and richness (p = 0.0001317) of bryophytes between seasons (p-value = 0.3393; p-value = 0.04065; p: 0.1081). There was a predominance of generalist (25 spp.) and corticolous (728 individuals) species, which were widely distributed in the sampled areas. Therefore, the structure of the bryophyte communities was not influenced by seasonality, and this indicates that these plants are adapted to the environmental conditions.

Introduction: Overweight is an emerging problem among children and adolescents that leads to the development of several morbidities and health risks. Overweight occurs differently in different populations, especially in vulnerable groups like the rural and quilombola communities (an African-descendant population). This study aimed to estimate the prevalence of overweight and to investigate the possible associated factors in rural adolescents living in both quilombola and non-quilombola communities in Northeast Brazil.Methods: This study is a population-based cross-sectional study with a household approach carried out in 2015 with 390 adolescents (age 10–19 years) living in rural quilombola and non-quilombola communities. The nutritional status was gauged using z-scores calculated for body mass index (BMI) and varies with gender and age. Prevalence ratios (PRs) and 95% confidence intervals (95% CIs) were used to establish associations between the results and explained variables. The multivariate analysis followed a model with a hierarchical entry of covariables controlled by gender and age.Results: The study showed that 18.5% of rural adolescents were overweight, of which 17.9% were quilombolas and 19.0% were non-quilombolas. A significant difference in overweight between the samples was not found. In the multivariate-adjusted model, age ≥16 years (PR: 0.51; 95% CI: 0.28–0.95), the habit of having regular breakfast (PR: 0.58; 95% CI: 0.35–0.98), and process of attending school (PR: 0.35; 95% CI: 0.17–0.71) were associated with a lower prevalence of overweight. Stationary screen time, in contrast, was associated with a higher prevalence (PR: 1.61; 95% CI: 1.05–2.46). The process of attending school was associated with a lower prevalence of overweight (PR: 0.26; 95% CI: 0.09–0.69), even for the quilombolas.Conclusions: A low prevalence of overweight was identified in rural adolescents. Overweight was significantly associated with the habit of having regular breakfast, older age, stationary screen time, and the process of attending school. The results reveal that school is a potential space for health promotion interventions, specifically in the most vulnerable rural regions, such as the quilombola communities. Besides, the study emphasizes the importance of adopting a healthy lifestyle early in life, including cultivating the habit of having regular breakfast and reducing stationary screen time.

Abstract Copaiba oil is a natural product used by Amazonian populations and recognized for its medicinal properties because it has significant antimicrobial activity for several pathogenic microorganisms. The present work aimed to evaluate and characterize the effect of natural oil produced by copaiba – Copaifera multijuga against multiresistant isolates of bubaline mastitis. The nitrocefin test was performed with isolates of Staphylococcus aureus from bubaline mastitis, which were 100% positive for beta-lactamase enzyme detection. Minimum Bactericidal Concentration (MBC) of 25% to 3.12% was obtained for Enterococcus faecalis and Escherichia coli and 50% and 25% for S. aureus, but Klebsiella pneumoniae and Bacillus subtilis were resistant. MBC with 12.5% and 6.25% oil were obtained for most multiresistant bubaline mastitis isolates from the states of Pernambuco, Ceará, Bahia and Alagoas. The results demonstrated the great potential of using copaiba natural oil in the treatment of buffalo mastitis.

Abstract Bryophytes play an important role in the process of ecological succession: conditioning the environment favourably for the emergence of subsequent groups. The objective of this study was to investigate the distribution of bryophyte communities in a cronossequence in the Caxiuanã National Forest, Pará, Brazil. To this end, biological material was collected in forest remnants with different successional stages based on regeneration age: Stage I (0 – 10 years), Stage II (10 - 25), Stage III (> 25) and Stage IV (primary forests). Density, richness and composition of species were compared between successional stages and the occurrence of possible indicator-species was investigated. The identified taxa were also classified by guilds of tolerance to solar radiation and colonized substrate. Composition of species was the variable that most contributed to understanding the distribution of bryophyte communities throughout successional stages, with eight species identified as potential indicators of some successional stages. Generalist species predominated in all stages. The richness of sun tolerants, in turn, decreased with the progress of succession, while shade tolerants increased. The land use history and land cover can influence the availability and quality of substrates and consequently their colonization by bryophytes in the different stages.

The compound (E)-2-cyano-N,3-diphenylacrylamide (JMPR-01) was structurally developed using bioisosteric modifications of a hybrid prototype as formed from fragments of indomethacin and paracetamol. Initially, in vitro assays were performed to determine cell viability (in macrophage cultures), and its ability to modulate the synthesis of nitrite and cytokines (IL-1β and TNFα) in non-cytotoxic concentrations. In vivo, anti-inflammatory activity was explored using the CFA-induced paw edema and zymosan-induced peritonitis models. To investigate possible molecular targets, molecular docking was performed with the following crystallographic structures: LT-A4-H, PDE4B, COX-2, 5-LOX, and iNOS. As results, we observed a significant reduction in the production of nitrite and IL-1β at all concentrations used, and also for TNFα with JMPR-01 at 50 and 25 μM. The anti-edematogenic activity of JMPR-01 (100 mg/kg) was significant, reducing edema at 2–6 h, similar to the dexamethasone control. In induced peritonitis, JMPR-01 reduced leukocyte migration by 61.8, 68.5, and 90.5% at respective doses of 5, 10, and 50 mg/kg. In silico, JMPR-01 presented satisfactory coupling; mainly with LT-A4-H, PDE4B, and iNOS. These preliminary results demonstrate the strong potential of JMPR-01 to become a drug for the treatment of inflammatory diseases.

In recent years, deep learning methods have outperformed previous state-of-the-art machine learning techniques for several problems, including image classification. Classifying cells in Pap smear images is very challenging, and it is still of paramount importance for cytopathologists. The Pap test is a cervical cancer prevention test that tracks preneoplastic changes in cervical epithelial cells. Carrying out this exam is important in that early detection. It is directly related to a greater chance of curing or reducing the number of deaths caused by the disease. The analysis of Pap smears is exhaustive and repetitive, as it is performed manually by cytopathologists. Therefore, a tool that assists cytopathologists is needed. This work considers 10 deep convolutional neural networks and proposes an ensemble of the three best architectures to classify cervical cancer upon cell nuclei and reduce the professionals’ workload. The dataset used in the experiments is available in the Center for Recognition and Inspection of Cells (CRIC) Searchable Image Database. Considering the metrics of precision, recall, F1-score, accuracy, and sensitivity, the proposed ensemble improves previous methods shown in the literature for two- and three-class classification. We also introduce the six-class classification outcome.
