Chickpea (Cicer arietinum) is a grain crop that is an important source of protein, vitamins, carbohydrates and minerals. It is highly sensitive to salt stress, and salt damage to cellular homeostasis and protein folding affects production. Plants have several mechanisms to prevent cellular damages under abiotic stresses, such as proteins in the endoplasmic reticulum (protein isulfide somerases (PDIs) and PDI-like proteins), which help prevent the build-up of mis-folded proteins that are damaged under abiotic stresses. In this study, we completed initial comprehensive genome-wide analysis of the chickpea PDI gene family. We found eight PDI genes are distributed on six out of eight chromosomes. Two pairs of paralogous genes were found to have segmental duplications. The phylogenetic analysis showed that the PDIs have a high degree of homology in C. arietinum, Cicer reticulatum, Lens culinaris, Phaseolus acutifolius, Pisum sativum and Oryza sativa. The gene structure analysis displayed that CaPDI1-CaPDI8 have 9-12 exons except for CaPDI5, which has 25 exons. Subcellular localisation indicated accumulation of CaPDIs in endoplasmic reticulum. Protein-conserved motifs and domain analysis demonstrated that thioredoxin domains of PDI family is present in all CaPDIs. CaPDI proteins have strong protein-protein interaction. In silico expression analysis showed that four out of eight PDI genes (CPDI2, CaPDI6, CaPDI7 and CaPDI8) were expressed under salt stress. Of these, expression of CaPDI2 and CaPDI8 was the highest. This work indicated that PDI genes are involved in salt stress tolerance in chickpea and the CaPDIs may be further studied for their role of inducing salt tolerance. [ABSTRACT FROM AUTHOR]

Plants have certain adaptation mechanisms to combat temperature extremes and fluctuations. The heat shock protein (HSP90A) plays a crucial role in plant defence mechanisms under heat stress. In silico analysis of the eight TaHSP90A transcripts showed diverse structural patterns in terms of intron/exons, domains, motifs and cis elements in the promoter region in wheat. These regions contained cis elements related to hormones, biotic and abiotic stress and development. To validate these findings, two contrasting wheat genotypes E-01 (thermo-tolerant) and SHP-52 (thermosensitive) were used to evaluate the expression pattern of three transcripts TraesCS2A02G033700.1, TraesCS5B02G258900.3 and TraesCS5D02G268000.2 in five different tissues at five different temperature regimes. Expression of TraesCS2A02G033700.1 was upregulated (2-fold) in flag leaf tissue after 1 and 4 h of heat treatment in E-01. In contrast, SHP-52 showed downregulated expression after 1 h of heat treatment. Additionally, it was shown that under heat stress, the increased expression of TaHSP90A led to an increase in grain production. As the molecular mechanism of genes involved in heat tolerance at the reproductive stage is mostly unknown, these results provide new insights into the role of TaHSP90A transcripts in developing phenotypic plasticity in wheat to develop heat-tolerant cultivars under the current changing climate scenario. Hyphae of arbuscular mycorrhizal fungi (AMF) in soil often form complex mycorrhizal networks among roots of same or different plant species for transfer of nutrients from one plant to another. However, the effect of soil nitrogen (N) availability on nutrient transfer between different plant species via common mycorrhizal networks (CMNs) has not been experimentally examined. In order to quantify CMN-mediated nutrient transfer between Leymus chinensis (LC) and Cleistogene squarrosa (CS), two systems, i.e. the CS–LC system (CS and LC were donor and recipient, respectively) and the LC–CS system (LC and CS were donor and recipient, respectively) were established. Stable isotopic 15N was applied to track N transfer between heterospecific seedlings connected by CMNs under three levels of soil N additions: no N addition control (N0), N addition with 7 mg/kg (N1) and N addition with 14 mg/kg (N2). In the CS–LC system, the highest rate of AMF colonization and hyphal length density (HLD) were found at N1. In contrast, maximum AMF colonization rate and HLD were recorded at N2 in LC–CS system. Consequently, plant biomass was significantly higher under N1 and N2 levels in CS–LC and LC–CS systems, respectively. Moreover, in CS–LC system, 15N transfer rate ranged from 16% to 61%, with maximum transfer rate at N1. For LC–CS system, 15N transfer rate was much lower, with the maximum occurring at N0. These findings suggest that CMNs could potentially regulate N transfer from a donor to recipient plant depending upon the strength of individual plant carbon sink. [ABSTRACT FROM AUTHOR]

Transgenic Arabidopsis thaliana (ecotype Columbia) was successfully transformed with the gene fructose-1,6-bisphosphatase (FBPas e) and named as AtFBPase plants. Transgenic plants exhibited stable transformation, integration and significantly higher expressions for the transformed gene. Morphological evaluation of transgenic plants showed increased plant height (35cm), number of leaves (25), chlorophyll contents (28%), water use efficiency (increased from 1.5 to 2.6μmol CO2 μmol-1 H2 O) and stomatal conductance (20%), which all resulted in an enhanced photosynthetic rate (2.7μmolm-2 s-1 ) compared to wild type plants. This study suggests the vital role of FBPase gene in the modification of regulatory pathways to enhance the photosynthetic rate, which can also be utilised for economic crops in future.

In the wake of the recent global pandemic of COVID-19, there has been increasing concern among the general public to improve their immune system. The causative agent of COVID-19 is SARS CoV-2, similar to its relative viruses SARS Cov-1 and MERS. Although vaccines have been developed now and are in public use, still the role of a healthy immune system is crucial as new strains of the virus are being discovered and along with emergence of new strains, short and long term side effects of vaccine cannot be excluded. Plants have been used in herbal medicines and many chemicals since ancient times and extracts in them are found to boost the immune system. Therefore, a consciously maintained diet consisting of plant-based immunity boosters is the need of the hour. Plants are known to provide many chemicals, natural therapeutics and vitamins which naturally enhance our immune system. Recently, vaccination research is also being carried out in transgenic plants.
