En este trabajo se detalla la importancia del oxígeno en el envejecimiento del vino en barrica de roble. Se estudia la relación entre la estructura de la madera y la cantidad de oxígeno que puede permear al vino (OTR), a su vez determina la calidad de la composición química y la calidad organoléptica del vino envejecido en barrica. Primeramente se determinó la dinámica de desgasificación de la madera y el consumo de oxígeno de los compuestos liberados por las diferentes estructuras anatómicas de la madera. Para ello se utilizaron unos nanosensores experimentales para conocer el papel de los elagitaninos como barrera activa al paso del oxígeno en el envejecimiento del vino en barrica de roble y visualizar y cuantificar el proceso de desgasificación de la madera. Esta metodología utilizada permitió visualizar la dinámica del oxígeno a partir del análisis de imágenes RGB de una solución sensora sensible al oxígeno, con una gran señal y con poco error, de alta resolución que permitió diferenciar entre áreas muy pequeñas. Es una metodología simple y económica, por lo que puede ser utilizada en muchos más campos de la ciencia. Observada la importancia de la estructura de la madera se validó una metodología no destructiva de clasificación de la madera para la construcción de barricas con tasa de transferencia de oxígeno conocida. Se construyeron 16 barricas de 225L de tostado medio, donde 8 eran de baja tasa de transferencia de oxígeno (LWOTR), 8 de alta tasa (HWOTR) y se fabricaron 4 más llamadas control, las cuales tienen el mismo tipo de producción, pero sin trazabilidad sobre las duelas. La mitad de cada lote se destinó a la validación de la metodología y se llenaron con solución hidroalcohólica previamente desgasificada. Para el análisis de estas se utilizó un tapón expandible modificado que permitió determinar la presión en el interior de la barrica y dejaba pasar dos fibras ópticas, con un sensor luminiscente en la punta, situados a una distancia del tapón de 21 y 42 cm que permitieron la medida y control del oxígeno en la solución. Este trabajo demostró que se puede clasificar la madera según su estructura anatómica para la construcción de barricas homogéneas con tasa de transferencia de oxígeno personalizada, viendo que el aporte de oxígeno en las barricas de alta tasa de transferencia es 2.3 veces superior que en las de baja. La otra mitad de los lotes se destinaron para al envejecimiento de un vino Ribera del Duero durante un año. Durante este tiempo se analizó cada tres meses el vino envejecido para el estudio de la evolución de las antocianinas y derivados y los parámetros básicos del vino. Los resultados muestran que los vinos envejecidos en barricas de alto OTR, tienen una mayor cantidad de antocianinas monoméricas y los vinos envejecidos en barricas de bajo OTR presentan una mayor intensidad de color. Además, por LC-MS se detectaron diferentes familias de derivados antocianidicos, incluyendo la tentativa de 2 nuevos pigmentos aldehídos-flavonol-metilpiranoantocinina. También se estudió la composición tánica de la madera y del vino envejecido durante un año en las barricas con OTR personalizado. Se analizó la concentración de 4 taninos, castalagina, vescalagina, grandinina y Rubirina E. Los resultados muestran que no existe una relación directa entre la concentración de elagitaninos en la madera y el OTR, no obstante, hay una tendencia de una mayor concentración en las barricas clasificadas con baja tasa de transferencia de oxígeno. En cuanto a los resultados al vino envejecido, siguen mostrando una gran homogeneidad entre muestras, siendo los vinos envejecidos en las barricas de bajo OTR, más ricos en composición tánica. Por último, se realizó una cata triangular con un panel experto donde se consiguió diferenciar entre los vinos envejecidos en barricas de alto y bajo OTR. Se confirma con estos estudios la homogeneidad de la clasificación, y que esta diferencia de contribución de oxígeno permite obtener vinos de diferente composición., Departamento de Química Analítica, Doctorado en Enología, Viticultura y Sostenibilidad

Oxidation of wine while in contact with oak wood is a well-known fact and recognized as an important process in wine ageing. The slow and continuous diffusion of oxygen from the oak wood entrapped in its porosity occurs and encourages the reactions usually related with wine aging. In this work, oxygen sensitive particles were used to study the oxygen dynamics from different anatomical regions of the oak wood and simultaneously the oxygen increasement because of the deoxygenation of the wood and the oxygen depletion due to its consumption by the tannins and other compounds released from the wood. To this end, core-shell-type PSPVP nanoparticles stained with Macrolex Fluorescence Yellow and Pt-TFPP were employed as reference and oxygen-sensitive particles respectively. Moreover, a Guppy Pro RGB camera was employed to monitor the particles performing ratiometric oxygen imaging, using the green and red channels to acquire the light emitted from the reference and the oxygen-sensitive particles respectively. Because the volume of nanosensors corresponding to each surface unit is very different from that of the wood region, different exposure times were chosen to obtain the images at each of the times. The results show the wood degassing process during the first minutes of the experiment, the oxygen release from different structural elements of the wood, its consumption by the released compounds from wood and the diffusion of the oxygen through the model wine.

The ageing of wines in oak barrels is a key stage in the production of high-quality red wines, with the type of oak chosen and the amount of oxygen received by the wine being the determining factors of the process. This work analyses the effect of ageing the same red wine in barrels with different oxygenation rates for one year (OTR), specifically the effect on the evolution of anthocyanins, their derivatives and the appearance of new pigments according to the oxygen dosage in barrels. Results show that wines aged in High-Wood-OTR barrels have a large quantity of monomeric anthocyanins and wine aged in Low-Wood-OTR barrels presents a major intensity of colour. Moreover, using LC&ndash, MS analysis, it was possible to detect and identify different families of anthocyanin derivatives, including the tentative identification of two new aldehyde-flavanol-methylpyranoanthocyanin pigments.

The process of aging red wines in barrels is a common practice that aims to improve the quality of the wine. Oxygen, together with the compounds released by the oak wood, is responsible for this improvement through interaction with the wine compounds during the process. Wood as a natural product presents a series of highly significant discontinuities in its characteristics, not only between trees of the same species, but even between different areas of the same log. It is not surprising that, despite the homogenization of the characteristics of the wood due to the number of staves in a barrel (around 30 pieces), barrels made from the same batch of wood show very different behavior when aging the same wine. This work presents the results of applying a new way of classifying wood in cooperage by means of image analysis of the staves, based on its anatomical characteristics besides grain and handling of the staves, in order to achieve the production of barrels with a desired oxygen transfer rate (OTR) potential and also with great homogeneity between manufacturing batches. The two batches of barrels built with high and low OTR proved to be very homogeneous and the oxygen dosage of the high OTR barrels was more than twice that of the low OTR barrels, which confirmed the success of the system developed.
