Abstract This study by Okuno et al. successfully identified eight exo‐miRNAs through exosome‐based discovery and established an exo‐miRNA‐based liquid biopsy assay for predicting therapeutic response in metastatic gastric cancer (mGC). Exosomes, a subpopulation of extracellular vesicles originating from endosomes, play a crucial role in this context. Liquid biopsy, analyzing blood for circulating tumor cells, extracellular vesicles, or cell‐free nucleic acid, has revolutionized cancer diagnosis and monitoring. It significantly contributes to early detection, staging, and relapse detection in various cancers. Numerous studies have highlighted the clinical significance of miRNA and lncRNA within extracellular vesicles. The authors developed a response‐prediction model for chemo‐responsiveness in mGC patients. This study's model predicts responses robustly, demonstrating its potential efficacy in clinical practice. It offers a non‐invasive and accessible method for therapeutic response prediction, crucial for precision medicine in mGC. Successful translation of these findings into clinical applications promises substantial benefits for patient care.

Lily is an important cut flower worldwide and prefers cool environments. The high summer temperature in China affects the quality and quantity of cut lily–flower production. In this study, the heat stress transcription factor HsfA2 was cloned from two cultivated commercial varieties, Lilium longiflorum 'White Heaven' and Lilium oriental 'Siberia', which have different responses to heat stress. An actin-interacting protein 3 (AIP3) domain was found in LlHsfA2 of Lilium longiflorum 'White Heaven' but did not appear in LoHsfA2 of Lilium oriental 'Siberia' using prediction from the simple modular architecture research tool (SMART) website. The genes LlACTIN and LoACTIN were cloned, and their amino acid sequences were found to be the same, so they were named LACTIN. There was an interaction between LlHsfA2 and LACTIN, whereas this interaction did not occur between LoHsfA2 and LACTIN based upon the bimolecular fluorescence complementary (BiFC) experiments. The silenced plants of LlHsfA2 and LACTIN were sensitive to heat stress treatment using barley stripe mosaic virus (BSMV) induced gene silencing in 'White Heaven', and LoHsfA2 silenced plants were also sensitive in 'Siberia'. In contrast, LACTIN silenced plants in 'Siberia' were normal. The expressions of LlHsfA2 or LACTIN were reduced in the LACTIN or LlHsfA2 silenced 'White heaven' plants, whereas the expressions of LoHsfA2 or LACTIN were normal in the LACTIN or LoHsfA2 silenced 'Siberia' plants. In conclusion, the diversity of protein interactions between HsfA2 and ACTIN may affect the distinctness of the heat stress response in different lily varieties, which provides new knowledge for further study on the heat stress response in lily.

Abstract Imaging-based spatial transcriptomics techniques provide valuable spatial and gene expression information at single-cell resolution. However, their current capability is restricted to profiling a limited number of genes per sample, resulting in most of the transcriptome remaining unmeasured. To overcome this challenge, we develop ENGEP, an ensemble learning-based tool that predicts unmeasured gene expression in spatial transcriptomics data by using multiple single-cell RNA sequencing datasets as references. ENGEP outperforms current state-of-the-art tools and brings biological insight by accurately predicting unmeasured genes. ENGEP has exceptional efficiency in terms of runtime and memory usage, making it scalable for analyzing large datasets.

Abstract Supplier selection is of great significance and role, which influence the quality of major product development, and economic security and life safety. However, there exists a variety of uncertain information as a result of evaluation experts’ strong subjective consciousness and complexity of decision environment in the process of supplier selection evaluation. To deal with these problems, by exploiting gray incidence analysis, cloud models and TOPSIS, we establish a multi-attribute decision-making supplier selection method for complex product based on gray group clustering and improved criteria importance through intercriteria correlation, and then a case verifies the validity and feasibility of the proposed method. The results show that (1) the proposed model can provide a better portrayal of the uncertainty of the evaluation process in terms of both the fuzziness of the semantic concept and the randomness of the affiliation degree, while taking into account the differences between the evaluated solution and the positive and negative ideal solutions. (2) The proposed model can fully voice the decision-maker’s attitude on the basis of available information, allowing the decision-making process to be better tailored to reality by taking into account the ambiguity and randomness of the evaluation process.

Background: Our objective was to assess the viability and oncological security of a gasless, transaxillary single-incision endoscopic procedure for performing total thyroidectomy and bilateral central neck dissection (TT + BCND). This study focused on patients diagnosed with bilateral papillary thyroid microcarcinoma (PTMC). Method: Between April 2020 and November 2021, 22 patients with bilateral PTMC underwent single-incision, gasless, transaxillary endoscopic TT + BCND. The patients’ clinicopathologic characteristics, surgical completeness and complications were analyzed. Result: Single-incision, gasless, transaxillary endoscopic TT + BCND was successful performed in all patients. The median (IQR) total surgical time was 143 (85–160) min. Only two patients experienced transient unilateral RLN palsy or transient hypocalcemia. All these complications resolved within 1 month after surgery. The median duration of hospital stay after surgery was 4 (3–4.5) days. The median hospitalization expense for these patients was 3848 (3781–4145) USD. The median number of lymph node yielded was 10.5 (8–15). The cosmetic outcomes were well-received by all individuals. Conclusion: In certain cases, gasless, transaxillary endoscopic TT + BCND procedure performed through a single incision proved to be a secure alternative for managing bilateral PTMC.

Abstract Background As digital medicine has exerted profound influences upon diagnosis and treatment of hepatobiliary diseases, our study aims to investigate the accuracy of three-dimensional visualization and evaluation (3DVE) system in assessing the resectability of hilar cholangiocarcinoma (hCCA), and explores its potential clinical value. Materials and methods The discovery cohort, containing 111 patients from April 2013 to December 2019, was retrospectively included to determine resectability according to revised criteria for unresectability of hCCA. 3D visualization models were reconstructed to evaluate resectability parameters including biliary infiltration, vascular involvement, hepatic atrophy and metastasis. Evaluation accuracy were compared between contrast-enhanced CT and 3DVE. Logistic analysis was performed to identify independent risk factors of R0 resection. A new comprehensive 3DVE classification of hCCA based on factors influencing resectability was proposed to investigate its role in predicting R0 resection and prognosis. The main outcomes were also analyzed in cohort validation, including 34 patients from January 2020 to August 2022. Results 3DVE showed an accuracy rate of 91% (95%CI 83.6–95.4%) in preoperatively evaluating hCCA resectability, significantly higher than 81% (95%CI 72.8–87.7%) of that of CT (p = 0.03). Multivariate statistical monitoring methods are proven to be effective for the dynamic tobacco strip manufacturing process. However, the traditional methods are not sensitive enough to small faults and the practical tobacco processing monitoring requires further root cause of quality issues. In this regard, this study proposed a unified framework of detection-identification-tracing. This approach developed a dissimilarity canonical variable analysis (CVA), namely, it integrated the dissimilarity analysis concept into CVA, enabling the description of incipient relationship among the process variables and quality variables. We also adopted the reconstruction-based contribution to separate the potential abnormal variable and form the candidate set. The transfer entropy method was used to identify the causal relationship between variables and establish the matrix and topology diagram of causal relationships for root cause diagnosis. We applied this unified framework to the practical operation data of tobacco strip processing from a tobacco factory. The results showed that, compared with traditional contribution plot of anomaly detection, the proposed approach cannot only accurately separate abnormal variables but also locate the position of the root cause. The dissimilarity CVA proposed in this study outperformed traditional CVA in terms of sensitiveness to faults. This method would provide theoretical support for the reliable abnormal detection and diagnosis in the tobacco production process.

The sensitivity of freshness indicator labels/films has become an important research direction of intelligent food packaging. In this study, a high-sensitivity indicator film containing gelatin and Fe2＋ was prepared by electrospinning using blueberry anthocyanins as the indicator and zein as the matrix for monitoring fish freshness. The validity and sensitivity of the indicator film for detecting the freshness of silver carp were tested and the potential mechanism was elucidated. The results of pH sensitivity, ammonia sensitivity and anthocyanin release showed that the addition of gelatin and Fe2+ improved the sensitivity of the indicator film to pH and ammonia, and contributed to better binding of the anthocyanins in the film. There was a strong correlation between the color response (P = (L* + a* + b* + R + G + B)/a*) of the film and the content of total volatile basic nitrogen (TVB-N) content in fish meat as a freshness indicator (R2 > 0.98). In conclusion, the prepared indicator film can effectively monitor fish freshness, and the hydrogen bond interactions between anthocyanins and gelatin/Fe2+ may affect the color response characteristics and sensitivity of the indicator film.

Abstract Background The role of echocardiography in the diagnostic and prognostic assessment of pulmonary hypertension (PH) has been widely studied recently. However, these findings have not undergone normative evaluation and may provide confusing evidence for clinicians. To evaluate and summarize existing evidence, we performed an umbrella review. Methods Systematic reviews and meta-analyses were searched in PubMed, Embase, Web of Science, and Cochrane Library from inception to September 4, 2022. The methodological quality of the included studies was assessed using Assessment of Multiple Systematic Reviews (AMSTAR), and the Grading of Recommendations Assessment, Development and Evaluation (GRADE) system was used to evaluate the quality of evidence. Results Thirteen meta-analyses (nine diagnostic and four prognostic studies) were included after searching four databases. The methodological quality of the included studies was rated as high (62%) or moderate (38%) by AMSTAR. The thirteen included meta-analyses involved a total of 28 outcome measures. The quality of evidence for these outcomes were high (7%), moderate (29%), low (39%), and very low (25%) using GRADE methodology. In the detection of PH, the sensitivity of systolic pulmonary arterial pressure is 0.85–0.88, and the sensitivity and specificity of right ventricular outflow tract acceleration time are 0.84. Pericardial effusion, right atrial area, and tricuspid annulus systolic displacement provide prognostic value in patients with pulmonary arterial hypertension with hazard ratios between 1.45 and 1.70. Meanwhile, right ventricular longitudinal strain has independent prognostic value in patients with PH, with a hazard ratio of 2.96–3.67. Conclusion The umbrella review recommends echocardiography for PH detection and prognosis. Systolic pulmonary arterial pressure and right ventricular outflow tract acceleration time can be utilized for detection, while several factors including pericardial effusion, right atrial area, tricuspid annular systolic displacement, and right ventricular longitudinal strain have demonstrated prognostic significance. Abstract Background The correlations between genetic risk for Alzheimer’s disease (AD) with comprehensive brain regions at a regional scale are still not well understood. We aim to explore whether these associations vary across different age stages. Methods This study used large existing genome-wide association datasets to calculate polygenic risk score (PRS) for AD in two populations from the UK Biobank (N ~ 23 000) and Adolescent Brain Cognitive Development Study (N ~ 4660) who had multimodal macrostructural and microstructural magnetic resonance imaging (MRI) metrics. We used linear mixed-effect models to assess the strength of the association between AD PRS and multiple MRI metrics of regional brain structures at different stages of life. Results Compared to those with lower PRSs, adolescents with higher PRSs had thinner cortex in the caudal anterior cingulate and supramarginal. In the middle-aged and elderly population, AD PRS had correlations with regional structure shrink primarily located in the cingulate, prefrontal cortex, hippocampus, thalamus, amygdala, and striatum, whereas the brain expansion was concentrated near the occipital lobe. Furthermore, both adults and adolescents with higher PRSs exhibited widespread white matter microstructural changes, indicated by decreased fractional anisotropy (FA) or increased mean diffusivity (MD). Conclusions In conclusion, our results suggest genetic loading for AD may influence brain structures in a highly dynamic manner, with dramatically different patterns at different ages. This age-specific change is consistent with the classical pattern of brain impairment observed in AD patients.
