With the rapid development of science and technology, Chinese urban rail transit has entered a newstage of intelligent development from the stage of rapid construction. Urban rail transit has the advantagesof high speed, large capacity and high density, which greatly facilitates modern life. However,urban rail transit is quite different from conventional ground transportation. Traditional informationtransmission methods cannot be well applied to urban rail transit. The maturity and commercial useof 5G mobile communication technology provide new ideas for solving the problem of informationtransmission in urban rail transit. Based on the salient features and key technologies of 5G communicationtechnology, this paper researches on key technonlgies 5G in rail transit scenarios to achievefast and efficient processing efficiency. It is a solution for intelligent operation and maintenance ofurban rail transit and high-definition video surveillance, and studies and analyzes the security of thesystem solution. This fills the gap of rail transit in these two fields, and provides dual guarantees ofsafety and service for urban rail transit construction. It provides a new direction for the applicationof future urban rail transit system.

This paper presents the conducted research on an innovative pultrusion beam made of NCF carbon fabric for an enhanced flexural behavior. This includes laminate design and optimization. By changing the ply thickness of each 0°-, ±45°- and 90°- orientations (while fixing the others), their influence on the strength and stiffness of the pultrusion beam was studied by the finite element method. 7 different combinations of layups were designed to analyze mechanical performance. The simulation results show the layup of 4/5/1 ply ratio is the best one for strength and stiffness; therefore, the carbon fabric pultrusion beam samples were manufactured according to this layup. Bending experimental testing was conducted for both CFRP pultrusion beam and aluminum beam under the design case of equal thickness to compare their strength and stiffness. The testing results show the strength of fabric pultrusion beam is 82.5% of that of aluminum beam, weight is 60% of the aluminum one, and the stiffness is 75% of it. From the results obtained from FEA and physical testing, it is shown the pultrusion beam with complex multi-cavities shapes possess high mechanical performance. This research concludes the CFRP NCF pultrusion beam entails great advantages for railway structure parts and other industries.

Vitamin D (VD) is one of the important nutrients required by livestock; however, VD deficiency is reported to be widespread. Earlier studies have suggested a potential role for VD in reproduction. Studies on the correlation between VD and sow reproduction are limited. The aim of the current study was aimed to determine the role of 1,25-dihydroxy vitamin D3 (1α,25(OH)2D3) on porcine ovarian granulosa cells (PGCs) in vitro to provide a theoretical basis for improving the reproductive efficiency of sows. We used chloroquine (autophagy inhibitor) and reactive oxygen species (ROS) scavenger N-acetylcysteine in conjunction with 1α,25(OH)2D3 to explore the effect on PGCs. The results showed that 10 nM of 1α,25(OH)2D3 increased PGC viability and ROS content. In addition, 1α,25(OH)2D3 induces PGC autophagy according to the gene transcription and protein expression levels of LC3, ATG7, BECN1, and SQSTM1 and promotes the generation of autophagosomes. 1α,25(OH)2D3-induced autophagy affects the synthesis of E2 and P4 in PGCs. We investigated the relationship between ROS and autophagy, and the results showed that 1α,25(OH)2D3-induced ROS promoted PGC autophagy. The ROS-BNIP3-PINK1 pathway was involved in PGC autophagy induced by 1α,25(OH)2D3. In conclusion, this study suggests that 1α,25(OH)2D3 promotes PGC autophagy as a protective mechanism against ROS via the BNIP3/PINK1 pathway.

The brain of the domestic pig (Sus scrofa domesticus) has drawn considerable attention due to its high similarities to that of humans. However, the cellular compositions of the pig brain (PB) remain elusive. Here we investigated the single-nucleus transcriptomic profiles of five regions of the PB (frontal lobe, parietal lobe, temporal lobe, occipital lobe, and hypothalamus) and identified 21 cell subpopulations. The cross-species comparison of mouse and pig hypothalamus revealed the shared and specific gene expression patterns at the single-cell resolution. Furthermore, we identified cell types and molecular pathways closely associated with neurological disorders, bridging the gap between gene mutations and pathogenesis. We reported, to our knowledge, the first single-cell atlas of domestic pig cerebral cortex and hypothalamus combined with a comprehensive analysis across species, providing extensive resources for future research regarding neural science, evolutionary developmental biology, and regenerative medicine.

Olfaction, the sense of smell, is a fundamental trait crucial to many species. The olfactory bulb (OB) plays pivotal roles in processing and transmitting odor information from the environment to the brain. The cellular heterogeneity of the mouse OB has been studied using single-cell RNA sequencing. However, the epigenetic landscape of the mOB remains mostly unexplored. Herein, we apply single-cell assay for transposase-accessible chromatin sequencing to profile the genome-wide chromatin accessibility of 9,549 single cells from the mOB. Based on single-cell epigenetic signatures, mOB cells are classified into 21 clusters corresponding to 11 cell types. We identify distinct sets of putative regulatory elements specific to each cell cluster from which putative target genes and enriched potential functions are inferred. In addition, the transcription factor motifs enriched in each cell cluster are determined to indicate the developmental fate of each cell lineage. Our study provides a valuable epigenetic data set for the mOB at single-cell resolution, and the results can enhance our understanding of regulatory circuits and the therapeutic capacity of the OB at the single-cell level.

BackgroundImmune cells play important roles in mediating immune response and host defense against invading pathogens. However, insights into the molecular mechanisms governing circulating immune cell diversity among multiple species are limited.MethodsIn this study, we compared the single-cell transcriptomes of 77□957 immune cells from 12 species using single-cell RNA-sequencing (scRNA-seq). Distinct molecular profiles were characterized for different immune cell types, including T cells, B cells, natural killer cells, monocytes, and dendritic cells.ResultsResults revealed the heterogeneity and compositions of circulating immune cells among 12 different species. Additionally, we explored the conserved and divergent cellular crosstalk and genetic regulatory networks among vertebrate immune cells. Notably, the ligand and receptor pair VIM-CD44 was highly conserved among the immune cells.ConclusionsThis study is the first to provide a comprehensive analysis of the cross-species single-cell atlas for peripheral blood mononuclear cells (PBMCs). This research should advance our understanding of the cellular taxonomy and fundamental functions of PBMCs, with important implications in evolutionary biology, developmental biology, and immune system disorders.

It is well known that approximately 99% of ovarian follicles in mammals suffer from a degenerative process known as atresia, which is a huge waste of genetic resource in female animals. Studies have shown that activin A (ACT-A) is located in ovarian granulosa cells and has different effects in granulosa cell depending on species. Although granulosa cells play a critical role during follicular atresia, the mechanism of action of ACT-A in bovine ovarian granulosa cells (BGC) is poorly understood. In this study, we first determined the apoptosis of BGCs isolated from growth follicles and atretic follicles respectively. Then, BGC isolated from atretic follicles were used as a model to elucidate the role of ACT-A in cattle ovary. The results showed that apoptosis occurred in both growing follicles and atretic follicles, and the percentage of apoptotic cells in atretic follicles was higher than that in growing follicles. The current results indicated that ACT-A can attenuate apoptosis of BGC by maintaining the function of BGC in atretic follicles. Increased ERβ induced by ACT-A promoted BGC autophagy but had no effect on apoptosis. In summary, this study suggests that ACT-A attenuates BGC apoptosis in atretic follicles by ERβ-mediated autophagy signalling.

SummarySystem-wide molecular characteristics of COVID-19, especially in those patients without comorbidities, have not been fully investigated. We compared extensive molecular profiles of blood samples from 231 COVID-19 patients, ranging from asymptomatic to critically ill, importantly excluding those with any comorbidities. Amongst the major findings, asymptomatic patients were characterized by highly activated anti-virus interferon, T/natural killer (NK) cell activation, and transcriptional upregulation of inflammatory cytokine mRNAs. However, given very abundant RNA binding proteins (RBPs), these cytokine mRNAs could be effectively destabilized hence preserving normal cytokine levels. In contrast, in critically ill patients, cytokine storm due to RBPs inhibition and tryptophan metabolites accumulation contributed to T/NK cell dysfunction. A machine-learning model was constructed which accurately stratified the COVID-19 severities based on their multi-omics features. Overall, our analysis provides insights into COVID-19 pathogenesis and identifies targets for intervening in treatment.

A few animals have been suspected to be intermediate hosts of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). However, a large-scale single-cell screening of SARS-CoV-2 target cells on a wide variety of animals is missing. Here, we constructed the single-cell atlas for 11 representative species in pets, livestock, poultry, and wildlife. Notably, the proportion of SARS-CoV-2 target cells in cat was found considerably higher than other species we investigated and SARS-CoV-2 target cells were detected in multiple cell types of domestic pig, implying the necessity to carefully evaluate the risk of cats during the current COVID-19 pandemic and keep pigs under surveillance for the possibility of becoming intermediate hosts in future coronavirus outbreak. Furthermore, we screened the expression patterns of receptors for 144 viruses, resulting in a comprehensive atlas of virus target cells. Taken together, our work provides a novel and fundamental strategy to screen virus target cells and susceptible species, based on single-cell transcriptomes we generated for domesticated animals and wildlife, which could function as a valuable resource for controlling current pandemics and serve as an early warning system for coping with future infectious disease threats.

Down syndrome (DS) is one of the most common human birth defects caused by trisomy 21 (T21), leading to a variety of cognitive impairments. The cellular composition of human brain has been explored using single cell RNA sequencing in both physiological and pathological conditions. However, the cellular heterogeneity of human brain with chromosome aneuploidy is largely unknown. Here, we profiled the transcriptome of 36046 cells in cerebral cortex of T21 human fetus, covering frontal lobe, parietal lobe, occipital lobe and temporal lobe. Intriguingly, we detected several genes positively associated with neurons maturation was dysregulated in T21 frontal cortex (HIC2, POU2F2, ZGLP1 and FOXK1). To share, explore and utilized the data resources of T21 cerebral cortex, we developed a comprehensive platform named T21atlas, composing of two functional modules (T21cluster and T21talk). Overall, our study provides, as far as we know, the first single cell atlas for T21 cerebral cortex, which could promote our understanding of the molecular mechanism of DS at an unprecedented resolution and could potentially facilitate the development of novel clinical therapeutics against T21.

Domestic pig (Sus scrofa domesticus) has drawn much attention from researchers worldwide due to its implications in evolutionary biology, regenerative medicine and agriculture. The brain atlas of Homo sapiens (primate), Mus musculus (rodent), Danio rerio (fish) and Drosophila melanogaster (insect) have been constructed at single cell resolution, however, the cellular compositions of pig brain remain largely unexplored. In this study, we investigated the single-cell transcriptomic profiles of five distinct regions of domestic pig brain, from which we identified 21 clusters corresponding to six major cell types, characterized by unique spectrum of gene expression. By spatial comparison, we identified cell types enriched or depleted in certain brain regions. Inter-species comparison revealed cell-type similarities and divergences in hypothalamus between mouse and pig, providing invaluable resources for the evolutionary exploration of brain functions at single cell level. Besides, our study revealed cell types and molecular pathways closely associated with several diseases (obesity, anorexia, bulimia, epilepsy, intellectual disability, and autism spectrum disorder), bridging the gap between gene mutations and pathological phenotypes, which might be of great use to the development precise therapies against neural system disorders. Taken together, we reported, so far as we know, the first single cell brain atlas of Sus scrofa domesticus, followed by comprehensive comparisons across brain region and species, which could throw light upon future evo-devo, regenerative medicine, and agricultural studies.

Retina is a crucial tissue for the capturing and processing of light stimulus. Characterization of the retina at single cell level is essential for the understanding of its biological functions. A variety of abnormalities in terms of morphology and function were reported in T21 retina. To evaluate the effects of chromosome aneuploidy on retina development, we characterized single cell transcriptional profiles of a T21 fetus and performed comprehensive bioinformatic analyses. Our data revealed the diversity and heterogeneity of cellular compositions in T21 retina. In total, we identified seven major cell types, and detected several subtypes within each cell type, followed by the detection of corresponding molecular markers including previously reported ones and a series of novel markers. Our analyses identified extensive communication networks between distinct cellular types, among which a few ligand-receptor interactions were associated with the development of retina and immunoregulatory interactions. Taken together, our data provided the first single cell transcriptome profile for human T21 retina which facilitates our understanding on the dosage effects of chromosome 21 on the development of retina.

BackgroundInvestigating cell fate decision and subpopulation specification in the context of the neural lineage is fundamental to understanding neurogenesis and neurodegenerative diseases. The differentiation process of neural-tube-like rosettesin vitrois representative of neural tube structures, which are composed of radially organized, columnar epithelial cells and give rise to functional neural cells. However, the underlying regulatory network of cell fate commitment during early neural differentiation remains elusive.ResultsIn this study, we investigated the genome-wide transcriptome profile of single cells from six consecutive reprogramming and neural differentiation time points and identified cellular subpopulations present at each differentiation stage. Based on the inferred reconstructed trajectory and the characteristics of subpopulations contributing the most towards commitment to the central nervous system (CNS) lineage at each stage during differentiation, we identified putative novel transcription factors in regulating neural differentiation. In addition, we dissected the dynamics of chromatin accessibility at the neural differentiation stages and revealed active c/s-regulatory elements for transcription factors known to have a key role in neural differentiation as well as for those that we suggest are also involved. Further, communication network analysis demonstrated that cellular interactions most frequently occurred among embryoid body (EB) stage and each cell subpopulation possessed a distinctive spectrum of ligands and receptors associated with neural differentiation which could reflect the identity of each subpopulation.ConclusionsOur study provides a comprehensive and integrative study of the transcriptomics and epigenetics of human early neural differentiation, which paves the way for a deeper understanding of the regulatory mechanisms driving the differentiation of the neural lineage.

Because of excellent electrical properties, epoxy resin is widely used in packaging and casting power equipment. Moisture and temperature in the environment are inclined to seriously affect the insulation tolerance of epoxy resin. This work focuses on the aging characteristics of epoxy resin in hygrothermal environment. The effect of hygrothermal aging on the properties of epoxy resin was deeply studied. Scanning electron microscopy images show that there are micro-crack, micro-slit and deterioration of resin in the hygrothermal aging process. The moisture adsorption process undergoes three equilibrium stages and it does not follow the Fick's second law. In the hygrothermal environment, the content of small free molecules produced by the oxidation and hydrolysis of the material and the absorbed water molecules dynamically changes, which leads to an increase-decrease-increase variation in e'. After the aged sample is dried, its e' descends, but because the internal structure is destroyed, its original dielectric properties cannot completely restored. It is found that after chemical moisture adsorption emerging, the equipment is damaged severely.

We propose a class of Alamouti-like Orthogonal Space-Time Block Codes (STBC) for two additional information bits without source such as power, bandwidth and time slot. To maintain the full rank property of the coding gain matrix, we propose a set of STBCs for 4 transmit antennas transmission to transmit one additional information bit achieving rate-9/8. Another orthogonal STBC code with rate-1 is proposed in this paper within the set. It shows by computer simulation results that by employing the set of STBCs, it achieves better bit error rate (BER) performance and throughput than that of [1] with a valid BER improvement at high SNR region above 20dB.

The mechanism of thermogenesis and thermolysis of transformer was analyzed in this paper, the position of the hot-spot temperature was also analyzed. By calculating winding average surface temperature rising, a hot-spot temperature calculating model of dry -type transformer was established, and the calculating model of insulation life loss of dry-type transformer was established based on the hot-spot temperature calculating model. In the end, according to a practical example, the winding hot-spot temperature was calculated, the ageing rate of transformer winding under rated load was calculated by the calculating model, and the insulation life loss of the transformer was evaluated. The result shows that the hot-spot temperature of LV coil is higher than HV coil; the winding has a smaller aging rate and insulation life loss when less than rated load.

The structural, magnetic and electronic properties of the polycrystalline La1−xAgxMnO3 are systematically investigated as a function of Ag-doping level. The result of the Rietveld refinement of X-ray powder diffraction shows that the samples are single rhombohedral ( R 3 C ) structure phase for x R 3 C perovskite phase and a nonmagnetic metal Ag phase. The temperature dependence of resistivity shows that all samples undergo a sharp insulator–metal (I–M) transition accompanying a paramagnetic to ferromagnetic transition with the decrease of temperature. Polycrystalline ordered double-perovskite Ca2FeMoO6 bulk samples with nanometer-scale grain size have been synthesized using a sol–gel method. A large magnetoresistance (MR) of 7.6% at a low magnetic field of 0.5 T and room temperature is obtained. The result of ac susceptibility shows that the sample has two magnetic transitions at TC1=374.6 K and TC2=336.4 K, which may correspond to the orthorhombic structure and the monoclinic structure phase, respectively. Due to the large MR at room temperature and well ferromagnetic conductivity of Ca2FeMoO6, it may be a desirable material for practical applications in future.

The author of the comment is too unilateral to associate double-peak behaviour (broad insulator–metal (I–M) transition below the Curie temperature ( T c )) and a sharp one near ( T c ) in rare-earth manganese perovskites only with the grain boundaries (GB) effect. Moreover, it is especially confused that the grain-size effect should be necessary to relate with this electronic transport behaviour. In this reply, we provide an argument in support of the statement that inhomogeneous electronic/magnetic phase separation is one of the most important reasons for the double-peak behaviour in the ρ ( T ) curve.
