We herein report an autopsy case of streptococcal toxic shock syndrome (STSS) with disseminated intravascular coagulation and multiple cerebral infarctions induced by Streptococcus dysgalactiae subsp. equisimilis in an 84-year-old male. Pathological examination revealed sepsis with hemophagocytosis in the reticular system and intravascular bacteria in multiple organs, originating from bacterial necrotizing fasciitis of the lower extremities. The brain MRI findings showed a diffusion weighted imaging-fluid-attenuated inversion-recovery mismatch, whereas the pathology was almost normal, thus supporting a hyperacute phase of cerebral infarction. The findings in this case help to elucidate the pathogenesis of STSS and develop appropriate treatment strategies.

Three novel coccoid-shaped strains, designated 21WXBC0057M1 T , 21WXBC0044M1 T and BJSWXB5TM5 T , were isolated from human breast milk in Wuxi, Jiangsu Province, China. These strains were facultative anaerobes, catalase-negative and Gram-positive. Through a comprehensive analysis of rRNA genes, protein-coding housekeeping genes and genomic phylogeny, we identified these strains as belonging to the genus Streptococcus . Specifically, strain 21WXBC0057M1 T was phylogenetically most closely related to Streptococcus infantis, strain 21WXBC0044M1 T was most closely related to Streptococcus oralis and strain BJSWXB5TM5 T displayed similarities to Streptococcus australis, Streptococcus peroris and S. infantis . The pairwise average nucleotide identity and digital DNA-DNA hybridization values for these three strains were below 95 and 70%, respectively, indicating that they occupied evolutionary branches distinct from all previously validly published Streptococcus species. Distinctive phenotypic characteristics discriminated these novel species from the type strains of their most closely related species. The major cellular fatty acids in the three strains were C 16 : 0 and C 18 : 0 . Genome annotation and a thorough examination of carbohydrate-active enzyme distribution highlighted the observation that all strains possessed extensive capabilities for carbohydrate metabolism, particularly human milk oligosaccharides utilization. Thus, based on these findings, we proposed the classification of the strains as representing three novel species within the genus Streptococcus: Streptococcus wuxiensis sp. nov. (type strain 21WXBC0057M1 T =GDMCC 1.4126 T =KCTC 25760 T ), Streptococcus jiangnanensis sp. nov. (type strain 21WXBC0044M1 T = GDMCC 1.4127 T =KCTC 25762 T ) and Streptococcus fermentans sp. nov. (type strain BJSWXB5TM5 T =GDMCC 1.4130 T =KCTC 25759 T ).

Streptococcus dysgalactiae (S. dysgalactiae ) is a common pathogen of humans and various animals. However, the phylogenetic position of animal S. dysgalactiae isolates and their zoonotic potential remain unclear. Most molecular epidemiological studies explicate beta-hemolytic streptococci according to their MLST and M protein gene (emm) types. Although human S. dysgalactiae isolates are relatively well characterized, the data concerning animal isolates are scarce. Here, we report the molecular characteristics and antimicrobial resistance of S. dysgalactiae strains recovered from sheep and their genetic relationship with isolates from other animal hosts and humans. Overall, 11 PFGE pulsotypes, five MLST sequence types (STs), and two emm types were distinguished, with ST248 and stL1376 being the most prevalent, indicating genetic diversity among tested 17 ovine isolates. Some isolates exhibited resistance to doxycycline (59%), erythromycin (6%), ciprofloxacin (6%), and trimethoprim/sulfamethoxazole (6%), harboring various resistance determinants. Phylogenetic analysis showed that studied ovine isolates grouped together with human S. dysgalactiae isolates from the cases of zoonotic infections. Moreover, some ovine isolates shared identical STs and emm gene sequences with human non-invasive and invasive S. dysgalactiae strains. Objective: Helicobacter pylori is known for colonizing the gastric mucosa and instigating severe upper gastrointestinal diseases such as gastritis, gastroduodenal ulcers, and gastric cancer. To date, there is no data available on the oral cavity as transmission site, whether H. pylori can survive in the oral cavity or in human saliva. The aim of the study was to investigate the influence of oral microorganisms and human saliva on the survival of H. pylori in human saliva., Methods: H. pylori strains KE, a motile derivate of type strain H. pylori 26695, and H. pylori SS1, a clinical isolate from a gastric biopsy, were grown in human pooled saliva (pooled from 4 healthy human donors, 0.22 μm filter-sterilized) or in BBF (Brucella browth formula; control) either as mono-cultures or in co-culture with Streptococcus mutans, Streptococcus oralis, Actinomyces naeslundii, Lacticaseibacillus casei and Candida dubliniensis. Bacterial survival of H. pylori and the oral microorganisms were investigated using colony forming units (CFU) assay and MALDI-TOF MS at baseline and after 24, 48 and 168 h., Results: In saliva, H. pylori KE demonstrated enhanced survival in co-culture with S. mutans, A. naeslundii, and C. dubliniensis, enduring for at least 48 h. In contrast, L. casei and S. oralis inhibited H. pylori KE in saliva. H. pylori KE could not be cultured after 168 h in saliva, neither in mono- nor co-culture. In contrast, H. pylori SS1 in saliva could be cultured after 168 h in co-culture with S. mutans and C. dubliniensis, but not in mono-culture. In BBF, H. pylori KE could be cultured after 168 h with S. mutans, L. casei and C. dubliniensis, and H. pylori SS1 with L. casei and C. dubliniensis, but not with S. mutans. Notably, the co-cultured microorganisms survived at high CFU numbers similar to those of the monocultures., Conclusion: The study suggests that H. pylori can transiently survive in human saliva and even with presence of certain oral microorganisms. However, it may not be a permanent resident of the oral microbiota. The genus Streptococcus is highly diverse and a core member of the primate oral microbiome. Streptococcus species are grouped into at least eight phylogenetically-supported clades, five of which are found almost exclusively in the oral cavity. We explored the dominant Streptococcus phylogenetic clades in samples from multiple oral sites and from ancient and modern-day humans and non-human primates and found that clade dominance is conserved across human oral sites, with most Streptococcus reads assigned to species falling in the Sanguinis or Mitis clades. However, minor differences in the presence and abundance of individual species within each clade differentiated human lifestyles, with loss of S. sinensis appearing to correlate with toothbrushing. Of the non-human primates, only baboons show clade abundance patterns similar to humans, suggesting that a habitat and diet similar to that of early humans may favor the growth of Sanguinis and Mitis clade species., Competing Interests: Competing interests: The authors declare no competing interests., (© 2025. Objective: Summaries of the relationships between the microbiota and liver cirrhosis and their conclusions are not consistent. This study describes microbial differences in patients with liver cirrhosis by performing a meta-analysis., Methods: We searched PubMed, Embase, Web of Science, and the Cochrane Library and collected related articles published before March 10, 2024. Ratio of autochthonous to non-autochthonous taxa was calculated as the cirrhosis dysbiosis ratio (CDR). Using a random-effects model, the standard mean deviation (SMD) and 95% confidence interval (CI) were calculated. We subsequently performed subgroup, sensitivity, and publication bias analyses. cirrhosis dysbiosis ratio., Results: A total of 53 eligible papers including 5076 participants were included. The pooled estimates revealed a moderately significant reduction in gut microbiome richness in patients with liver cirrhosis compared with controls, including the Shannon, Chao1, observed species, ACE, and PD indices, but no significant difference was observed for the Simpson index. Over 80% of the studies reported significant differences in β diversity. Families Enterobacteriaceae and Pasteurellaceae, belonging to the phylum Proteobacteria, along with the family Streptococcaceae and the genera Haemophilus, Streptococcus, and Veillonella, were significantly associated with liver cirrhosis compared to the control group. In contrast, the healthy group exhibited a higher abundance of the class Clostridia, particularly the families Lachnospiraceae and Ruminococcaceae, which are known for their diversity and role as common gut commensals. Furthermore, the class Bacilli, predominantly represented by the genus Streptococcus, was markedly enriched in the cirrhosis group., Conclusions: The microbiota richness of liver cirrhosis patients was lower than that of healthy controls. Three novel strains within the genus Streptococcus (29887 T , 29892 T and 29896 T ) were isolated from healthy pigs during routine veterinary physical exams. All three strains were non-motile and non-spore-forming Gram-positive cocci. The complete genome of each strain was attained, and phylogenetic analyses were performed. Comparison of the genomes of 29887 T , 29892 T and 29896 T to the genomes of other Streptococcus strains revealed digital DNA-DNA hybridization (dDDH) values between 21.2% and 53.9% and average nucleotide identity (ANI) values between 70.00% and 94.44%. Phylogenetic analyses suggested that each strain, 29896 T ( S. suivaginalis sp. nov.), 29887 T ( S. iners sp. nov.) and 29892 T ( S. iners subsp. hyiners subsp. nov.), may represent a novel species within the genus Streptococcus , while ANI analysis indicated that strains 29896 T ( S. suivaginalis sp. nov.) and 29887 T ( S. iners sp. nov.) represent novel species within the genus Streptococcus, and 29892 T ( S. iners subsp. hyiners subsp. nov.) represents a novel subspecies of 29887 T ( S. iners sp. nov.). Based upon the combined data presented in this study, two novel species, Streptococcus suivaginalis sp. nov. (type strain, 29896 T =NRRL B-65677 T =NCTC 14941 T ) and Streptococcus iners sp. nov. (type strain, 29887 T =NRRL B-65675 T =NCTC 14939 T ) are proposed, and one novel subspecies, Streptococcus iners subsp. hyiners subsp. nov. (type strain, 29892 T =NRRL B-65676 T =NCTC 14940 T ) is proposed.

Objective: We aimed to characterize integrative and conjugative elements (ICEs) in antimicrobial resistant Streptococcs uberis isolates from bovine milk in Chiba, Japan, based on whole-genome sequence (WGS) data., Results: Of the 101 isolates, we found the 36 isolates harboring erm(B)-tet(O), showing resistance to macrolides-lincosamides-tetracyclines. The 22 isolates were randomly selected and subject to WGS determination. The genomes measured 1.991-2.517 Mbp, with G + C contents of 35.8-36.9%. We used ResFinder-ICEfinder (web-based applications) to search for the antimicrobial resistant genes and ICEs. ResFinder detected combined erm(B)-tet(O)-ant(6)-Ia at the identical contig in each WGS. ICEfinder detected ICEs belonging to the same contigs, which contained erm(B)-tet(O)-ant(6)-Ia complete or partial sequences. Detection of putative ICEs using comparative genomic analysis was performed with identification of other streptococcal ICE resembling S. uberis ICEs. Using comparative genomic analysis (a reference WGS in NZ01 strain), putative ICE base size in UB37 isolate was 77,386-bp that was identical in other 13 isolates. Another similar streptococcal ICE was S. suis ICEnsui78-tet(O)-erm(B) mobile element. For ICE characterization in S. uberis with WGSs, a comparative genomic analysis is required with use of ICEfinder and other annotation tools., Competing Interests: Declarations. This study introduces an innovative on-site diagnostic method for rapidly detecting the Streptococcus bovis / Streptococcus equinus complex (SBSEC), crucial for livestock health and food safety. Through a comprehensive genomic analysis of 206 genomes, this study identified genetic markers that improved classification and addressed misclassifications, particularly in genomes labeled S. equinus and S. lutetiensis . These markers were integrated into a portable quantitative polymerase chain reaction (qPCR) that can detect SBSEC species with high sensitivity (down to 10 1 or 10 0 colony-forming units/mL). The portable system featuring a flat chip and compact equipment allows immediate diagnosis within 30 min. The diagnostic method was validated in field conditions directly from cattle udders, farm environments, and dairy products. Among the 100 samples, 51 tested positive for bacteria associated with mastitis. The performance of this portable qPCR was comparable to laboratory methods, offering a reliable alternative to whole-genome sequencing for early detection in clinical, agricultural, and environmental settings.

Introduction and Objectives: Infective endocarditis (IE) caused by oral streptococci is considered to be a 'more benign' IE than those caused by other microorganisms. Our aim is to analyse the changes in its incidence and its differential characteristics in our setting., Methods: Observational, single-centre, prospective cohort study including all cases of IE diagnosed in our centre between 1987 and 2023. IE caused by oral streptococci were identified and their characteristicsand mortality were compared with those of other IE., Results: Of 569 cases of IE, 104 (18.5%) were oral streptococcal IE, decreasing from 29.5% in 1987-1999 to 12.2% in 2000-2023 (p<0.001). The incidence of serious complications was lower in oral IE (65.3 vs. 78.9%; p=0.003), as was in-hospital mortality (14.4 vs. 29.9%; p=0.001)., Conclusions: Oral streptococcal IEs account for almost 20% of all IEs in our setting. They have a better prognostic clinical profile, with a lower incidence of serious complications, and their mortality is significantly lower than that of other IE. The Gram-positive bacteria Streptococcus pneumoniae is part of the Streptococcus mitis group (SMG) and causes life-threatening infections, such as pneumonia, sepsis, and meningitis. The closely related Streptococcus pseudopneumoniae has recently been shown to cause respiratory tract infections, as well as invasive infections, especially in patients with comorbidities. Due to the genetic and phenotypic similarities of species belonging to the SMG, the identification of S. pneumoniae and S. pseudopneumoniae is difficult and unreliable using phenotypic tests, as well as matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) and 16S rRNA sequencing. In this study, a loop-mediated isothermal amplification (LAMP)-based assay was developed using molecular markers specific for S. pneumoniae (SPN0001) and S. pseudopneumoniae (SPPN&#95;RS10375). The LAMP assay was evaluated using a collection of SMG clinical isolates, concluding that the method provides a correct, reliable, and fast identification of both S. pneumoniae and S. pseudopneumoniae clinical isolates., Competing Interests: The authors declare no conflict of interest.

Almost 25 years have now passed since the first identification of Streptococcus sinensis ( S. sinensis ). It can cause infections both in immunocompetent and immunocompromised hosts. However, it has been rarely described as an aetiology of infectious endocarditis. We herein report the case of a 75-year-old Italian gentleman who was admitted for shortness of breath, asthenia, weight loss, and an episode of loss of consciousness and who was subsequently diagnosed with S. sinensis endocarditis (the first reported in Italy). He was, therefore, treated with ceftriaxone for six weeks and underwent cardiac surgery. We performed a literature review on S. sinensis endocarditis cases and found 12 other reported cases. Demographics, clinical presentation, prognostic factors, treatment, and outcomes were summarized. Despite anecdotic cases being reported, S. sinensis endocarditis can occur and should be promptly and properly identified using accurate diagnostic methods. Continued research into its epidemiology, pathogenesis, antimicrobial resistance, and host interactions is essential for enhancing our knowledge and improving clinical management strategies.

Mastitis, a prevalent and economically important disease in the dairy industry, poses substantial challenges to dairy cow health, milk quality, and farm profitability worldwide. Mastitis is predominantly caused by bacterial infections. The objective of this study was to estimate the sensitivity (Se) and specificity (Sp) of bacterial culture and the VetMAX™ MastiType Multi Kit PCR in identified clinical mastitis pathogens. A total of 396 quarter-level milk samples were collected from 396 cows with clinical mastitis on 29 farms in the subtropical dairy region of Australia between March and December 2021. These samples were cultured and tested by PCR, and analysed using Bayesian latent class analysis under the assumption of one population two tests and also of three populations two tests, by dividing the population into subpopulations based on regions. Informative priors used in the analysis were calculated from published evidence. Models were compared using the Deviance Information Criterion (DIC). Sensitivity analysis was performed to evaluate the impact of changes in priors. The most common isolates cultured and detected by PCR were Streptococcus uberis (17.4 % and 27.3 %, respectively) and Escherichia coli (12.6 % and 25.0 %, respectively). Under the assumption of one population two tests, the Se of PCR (at cycle threshold (Ct) ≤ 37) was higher than that of bacterial culture for both pathogens: for E. coli, the Se was 50.2 % (95 % posterior probability interval (PPI): 37.4; 74.1) for bacterial culture, and 93.7 % (95 % PPI: 85.5; 98.4) for PCR. For S. uberis, the Se was 50.4 % (95 % PPI: 40.9; 61.3) for bacterial culture, and 81.5 % (73.0; 88.9) for PCR. Conversely, the Sp of bacterial culture was higher than that of PCR for both pathogens: for E. coli, the Sp was 99.2 % (97.8; 100) for bacterial culture, and 95.1 % (87.8; 99.4) for PCR. For S. uberis, the Sp was 99.2 % (95 % PPI: 97.6; 100) for bacterial culture, and 96.7 % (95 % PPI: 92.1; 99.2) for PCR. Bayesian latent class analysis with three populations two tests was only performed for S. uberis. For E. coli, this could not be performed because there were no PCR-positive results in one subpopulation. Under the assumption of three populations two tests, for S. uberis, the Se was 49.6 % (40.6; 59.4) for bacterial culture, and 81.1 % (72.6; 88.6) for PCR; and the Sp for bacterial culture was 99.1 % (97.7; 100), and for PCR was 96.9 % (93.0; 99.3). The DIC for the one population two tests model was lower than the DIC for the three populations two tests model. The sensitivity analysis for the one population two tests model demonstrated that a 10 % reduction in priors led to substantial changes in Se of both bacterial culture and PCR tests for E. coli and S. uberis, with overlap percentages ranging from 80.6 % to 92.2 %. In contrast, the Sp of bacterial culture and PCR tests remained relatively stable despite changes in priors, except for the Sp of PCR test for E. coli. In summary, the VetMAX™ MastiType Multi Kit demonstrated higher Se compared to bacterial culture, suggesting its potential as a routine test for identifying mastitis pathogens in milk samples from cows with clinical mastitis. Objectives: Among healthy people, we understand very little about the sociodemographic, lifestyle, and dental hygiene behaviours that shape their oral microbiota. This study investigates how sociodemographic, lifestyle and dental hygiene behaviours shape oral microbiota diversity and composition in an Australian population to better inform healthy oral microbiota donors for Oral Microbiota Transplantation (OMT)., Methods: The study comprised 93 healthy adults who underwent comprehensive oral examinations and questionnaires to assess their health status. Participants were excluded if they had any active systemic or oral disease. All completed a questionnaire containing information on socio-economic, lifestyle, behavioural, and oral health factors. Supragingival plaque was collected, and 16S ribosomal RNA (rRNA) amplicon sequencing was used to analyse microbial composition. Associations between the core microbiome, alpha- (within-sample), beta-diversity (between-sample) and an individual's co-variates were tested for statistical significance. A redundancy analysis (RDA), multivariate adonis, differential abundance and correlation analysis were performed to characterise which factors drive the variation in the healthy oral microbiome., Results: Streptococcus and Corynebacterium were the most prevalent and abundant genera among healthy Australians. The alpha and beta diversity were higher among unemployed non-Australian-born students who consumed low carbohydrates, fat, and sugar and had not visited the dentist for over 12 months. Additionally, beta diversity was significantly higher among daily flossers who abstained from fluoride treatment and had high salivary pH, although no single factor explained >4 % of the total variation (R 2 = 0.042). Alloprevotella, Lachnosporacea, and Parvimonas were significantly abundant among non-Australians who did not visit the dentist within a year. The RDA analysis revealed associations between microbiome composition and factors such as high carbohydrate, sugar, and fat consumption, low fibre intake, and regular dental checks among Australian-born individuals., Conclusion: These findings indicate that alpha and beta diversity of the oral microbiome varied significantly with sociodemographic, lifestyle, and dietary factors, including non-Australian birthplaces, unemployment, diet, and infrequent dental visits., Clinical Significance: These findings underscore the importance of considering diverse sociodemographic, lifestyle, and dietary factors in oral health management. Streptococci are well-known opportunistic bacterial abortifacients in mares. Colonization of the pregnant uterus is considered to happen after transcervical migration of bacteria from the lower genital tract mucosa. Streptococcus ovis is a pathogen mainly associated with inflammatory lesions in sheep. This species has not been reported in association with disease in horses. In the present case, S. ovis was isolated in monoculture from the lung of an 8-months-old equine fetus and was associated with development of acute suppurative bronchopneumonia, umbilical cord cellulitis and placentitis in the cervical star region of the allantochorion. The mare had been in a pasture together with sheep. One week prior to abortion, a double-guarded uterine swab had been inserted into the cervical canal by a veterinarian, who was unaware of the mare being pregnant. Necrotizing fasciitis is a fatal, soft tissue infection of the skin that requires prompt treatment. Historically, most cases have been attributed to group A beta-hemolytic Streptococcus infection. However, in recent years, other bacteria have been identified as causing necrotizing fasciitis. In the current study, we analyzed cases of necrotizing fasciitis and examined the significant differences in symptoms caused by pathogenic bacteria. We included 79 patients (43 males and 36 females, mean age 65.4 years) diagnosed with necrotizing fasciitis who visited our hospital between April 2004 and July 2023. The patients were classified into five groups based on the identified pathogen: group A beta-hemolytic Streptococcus; group B beta-hemolytic streptococcus; group G beta-hemolytic Streptococcus; mixed infection, including anaerobic bacteria; and Staphylococcus (S) aureus. The clinical characteristics of patients, including treatment duration and laboratory values, were analyzed. Group G beta-hemolytic Streptococcus was more common in older patients (Bonferroni method, p < 0.05). Patients with S. aureus tended to be hyperglycemic (Bonferroni method, p < 0.05), had a higher rate of bacteremia (Fisher's direct probability test, p < 0.05), and had a longer treatment duration than the other examined groups (Bonferroni method, p = 0.0132). Although the five groups did not differ in the mortality rate, overall survival was shorter in the mixed infection group than in the other groups (log-rank test, p < 0.05). The legs were the most common site of infection in the non-mixed infection group; in the mixed infection group, the pubic area was identified as the most common site of infection, accompanied by a poor prognosis. The present study investigates the prevalence and etiology of subclinical mastitis in Țurcana sheep flocks located in south-western Romania. Milchtest and California Mastitis Test (CMT), were used for the detection of subclinical mastitis. A number of 360 milk samples across three lactation phases were analyzed. Subclinical mastitis was identified in 146 (40.6%) of sheep milk samples. Only milk samples (n = 146) collected from sheep diagnosed with subclinical mastitis were considered for microbiological analyses. Thus, Staphylococcus aureus (129/146; 88.4%) was identified as the predominant pathogen followed by Streptococcus spp. (14/146; 9.6%) Enterococcus spp. (7/146; 4.8%), Pseudomonas aeruginosa (6/146; 4.1%) and Klebsiella pneumoniae (5/146; 3.4%). Antimicrobial susceptibility of isolated strains of Staphylococcus aureus, Streptococcus spp. and Enterococcus spp. was also tested. The results showed varying resistance patterns depending on the farm and microorganism. In heard A the highest resistance of Staphylococcus aureus was identified to polymyxin B (79.7%) followed by novobiocin (73.4%) and tetracycline (45.3%); Streptococcus spp. to streptomycin (100%), novobiocin (100%), tetracycline (100%) and Enterococcus spp. to tylosin (100%), streptomycin (75.0%) and tetracycline (75.0%). In herd B, Staphylococcus aureus showed high resistance rates to polymyxin B (86.3%), novobiocin (74.5%) and cloxacillin (39.2%); Streptococcus spp. strains to streptomycin (100%), novobiocin (100%), tetracycline (100%) and Enterococcus spp. to gentamicin (100%), streptomycin (66.7%) and erythromycin (66.7%). The obtained preliminary results indicate a potential risk that future treatments of sheep mastitis may not be effective, and demonstrated that raw unpasteurized milk can harbor antimicrobial resistance pathogens posing a threat to public health. Background: To investigate the difference of salivary microbiome composition in type 2 diabetes mellitus with different stages of periodontitis., Methods: 105 patients diagnosed with T2DM were divided into four groups according to the Stage of periodontitis.The 16 S rDNA hypervariable regions V3-V4 were amplified by polymerase chain reaction (PCR) after saliva DNA was extracted. The library was constructed and Illumina sequencing was performed. The sequencing results were compared with the silva 138 database, and the salivary microbiome of the four groups of patients was studied by species composition analysis, species difference analysis, principal coordinate analysis, and correlation analysis., Results: There were significant differences in alpha diversity of salivary microbiome among the four groups, but no difference in Beta diversity. At the phylum level, there was one differential bacteria-Firmicutes, while at the genus level, there were 16 differential bacteria. Spearman's correlation analysis identified significant correlations between periodontal health indices (PLI, SBI, AL and PD) and the abundances of over 13 genera, including Streptococcus, Gemella and Capnocytophaga, etc. Additionally, the duration of diabetes exhibited a positive correlation with AL and PD., Conclusions: There are differences in salivary microbial composition in patients with type 2 diabetes mellitus with different stages of periodontitis. Introduction: Changes in the human gut microbiome have been linked to various chronic diseases, including chronic obstructive pulmonary disease (COPD). While substantial knowledge is available on the genomic features of fecal communities, little is known about the microbiome's transcriptional activity. Here, we analyzed the metatranscriptomic (MTR) abundance of MetaCyc pathways, SuperPathways, and protein domain families (PFAM) represented by the gut microbiome in a cohort of non-small cell lung cancer (NSCLC) patients with- or without COPD comorbidity., Methods: Fecal samples of 40 NSCLC patients with- or without COPD comorbidity were collected at the time of diagnosis. Data was preprocessed using the Metaphlan3/Humann3 pipeline and BioCyc © to identify metabolic SuperPathways. LEfSe analysis was conducted on Pathway- and PFAM abundance data to determine COPD- and non-COPD-related clusters., Results: Key genera Streptococcus, Escherichia, Gemella , and Lactobacillus were significantly more active transcriptionally compared to their metagenomic presence. LEfSe analysis identified 11 MetaCyc pathways that were significantly overrepresented in patients with- and without COPD comorbidity. According to Spearman's rank correlation, Smoking PY showed a significant negative correlation with Glycolysis IV , Purine Ribonucleoside Degradation and Glycogen Biosynthesis I , and a significant positive correlation with Superpathway of Ac-CoA Biosynthesis and Glyoxylate cycle , whereas forced expiratory volume in the first second (FEV1) showed a significant negative correlation with Glycolysis IV and a significant positive correlation with Glycogen Biosynthesis I . Furthermore, COPD patients showed a significantly increased MTR abundance in ~60% of SuperPathways, indicating a universally increased MTR activity in this condition. FEV1 showed a significant correlation with SuperPathways Carbohydrate degradation , Glycan biosynthesis , and Glycolysis . Taxonomic analysis suggested a more prominent MTR activity from multiple Streptococcus species, Enterococcus (E.) faecalis, E. faecium and Escherichia (E.) coli than expected from their metagenomic abundance. Streptococcus dysgalactiae subspecies equisimilis (SDSE) is a Gram-positive bacterial pathogen that infects humans and is closely related to group A streptococcus (GAS). Compared with GAS, far less is known about SDSE pathobiology. Increased rates of invasive SDSE infections have recently been reported in many countries. One SDSE emm type ( stG62647 ) is known to cause severe diseases, including necrotizing soft-tissue infections, endocarditis, and osteoarticular infections. To increase our understanding of the molecular pathogenesis of stG62647 SDSE isolates causing human infections, we sequenced to closure the genomes of 120 stG62647 SDSE isolates. The genomes varied in size from 2.1 to 2.24 Mb pairs. The great majority of stG62647 isolates had IS1548 integrated into the silB gene, thereby inactivating it. Regions of difference, such as mobile genetic elements, were the largest source of genomic diversity. All 120 stG62647 isolates were assayed for virulence using a well-established mouse model of necrotizing myositis. An unexpectedly wide range of virulence was identified (20% to 95%), as assessed by near-mortality data. To explore the molecular mechanisms underlying virulence differences, we analyzed RNAseq transcriptome profiles for 38 stG62647 isolates (comprising the 19 least and most virulent) grown in vitro . Genetic polymorphisms were identified from whole-genome sequence data. Collectively, the results suggest that these SDSE isolates use multiple genetic pathways to alter virulence phenotype. The data also suggest that human genetics and underlying medical conditions contribute to disease severity. Our study integrates genomic, mouse virulence, and RNAseq data to advance our understanding of SDSE pathobiology and its molecular pathogenesis., Importance: This study integrated genomic sequencing, mouse virulence assays, and bacterial transcriptomic analysis to advance our understanding of the molecular mechanisms contributing to Streptococcus dysgalactiae subsp. equisimilis emm type stG62647 pathogenesis. We tested a large cohort of genetically closely related stG62647 isolates for virulence using an established mouse model of necrotizing myositis and discovered a broad spectrum of virulence phenotypes, with near-mortality rates ranging from 20% to 95%. This variation was unexpected, given their close genetic proximity. Transcriptome analysis of stG62647 isolates responsible for the lowest and highest near-mortality rates suggested that these isolates used multiple molecular pathways to alter their virulence. In addition, some genes encoding transcriptional regulators and putative virulence factors likely contribute to SDSE emm type stG62647 pathogenesis. Background. Commensal streptococci are common inhabitants of the oral microbiome and regulate its structure and function in beneficial ways for human health. They can, however, also be opportunistic pathogens and act as a reservoir of resistance genes that can be passed on to other bacteria, including pathogens. Little is known about the prevalence of these commensals in parents and their children and their antimicrobial susceptibilities in the Belgian general population. Gap Statement. The macrolide susceptibility of commensal oral Streptococci in Belgium is unknown. Methods. We assessed the prevalence and azithromycin susceptibility of commensal streptococcal species in the parents ( n =38) and children ( n =50) of 35 families in Belgium. Results. The most frequently detected taxonomic grouping was Streptococcus mitis/oralis , which was detected in 78/181 (43.1%) of the children's isolates and 66/128 (51.6%) of the parents' isolates. Of the 311 isolates collected in this study, 282 isolates (90.7%) had an azithromycin MIC value greater than the breakpoint of 0.25 mg l -1 and 146 isolates (46.9%) had azithromycin MICs greater than 2 mg l -1 . There was no difference in the azithromycin MIC distribution of all streptococcal isolates between children and parents. All individuals were colonized by streptococci with azithromycin MICs greater than 0.25 mg l -1 , and 87.5% of individuals had streptococci with MICs greater than 2 mg l -1 . Interpretation. The most prevalent species identified in both age groups was S. mitis/oralis . All individuals harboured streptococci with macrolide resistance. This highlights the need for additional antimicrobial stewardship initiatives to reduce the consumption of macrolides in the general population.

Two Gram-stain-positive, oxidase-negative, non-motile, facultative anaerobic, α-hemolytic, coccus-shaped bacteria (zg-86 T and zg-70) were isolated from the respiratory tracts of marmots (Marmota Himalayana) on the Qinghai-Tibet Plateau of China. Phylogenetic analysis of the 16S rRNA gene and 545 core genes revealed that these two strains belong to the Streptococcus genus. These strains were most closely related to Streptococcus respiraculi HTS25 T , Streptococcus cuniculi CCUG 65085 T , and Streptococcus marmotae HTS5 T . The average nucleotide identity (ANI) and digital DNA‒DNA hybridization (dDDH) were below the threshold for species delineation. The predominant cellular fatty acids (CFAs) in this novel species were C 16:0 , C 18:0 , and C 18:1 ω9c, whereas the primary polar lipids were phosphatidylglycerol (PG), phosphatidylethanolamine (PE) and an unknown phosphoglycolipid (PGL). The optimal growth conditions for the strains were 37 °C, pH 7.0, and 0.5% (w/v) NaCl on brain-heart infusion (BHI) agar supplemented with 5% defibrinated sheep blood. Comparative genomics analyses revealed the potential pathogenicity of strain zg-86 T through comparisons with suis subclade strains in terms of virulence factors, pathogen-host interactions (PHIs) and mobile genetic factors (MGEs). Based on the phenotypic characteristics and phylogenetic analyses, we propose that these two isolates represent novel species in the genus Streptococcus, for which the names Streptococcus zhangguiae sp. nov. Background: Streptococcus dysgalactiae subsp equisimilis (SDSE) is an emerging pathogen causing pharyngitis and post-streptococcal sequelae like S. pyogenes. SDSE was initially considered a commensal microorganism inhabiting the upper respiratory tract and skin. However, recently it has gained attention due to an increase in the invasive SDSE infections, which were reported in the early 20th century., Objectives: The aim of this review is to bring awareness of SDSE in the medical microbiologists because often its ignorance leads to the under reporting or misdiagnosis of SDSE. This also highlights the clinical spectrum of infections and the molecular epidemiology of SDSE., Content: Diagnosis of SDSE in clinical laboratories is challenging, because SDSE can be seen expressing either of the three Lancefield antigen Groups, Group A, C, and G. While MALDI-TOF (Matrix-Assisted Laser Desorption/Ionization Time-of-Flight) is a discriminatory method for identifying SDSE, its high cost can limit its use in many laboratories. Aims: To assess agreement of bacterial culture results from samples taken from nasal discharge, the nasal cavity and nasal biopsy from dogs and cats with nasal disease., Methods: Nineteen dogs and 21 cats with different nasal diseases (chronic rhinitis, n = 30; neoplasia, n = 7; sinonasal aspergillosis, n = 3) were prospectively enrolled in the study. Nasal swabs were taken bilaterally from nasal discharge at the nares, the nasal cavity, and one nasal mucosal biopsy per side. All samples were subjected to aerobic bacterial culture. Kappa statistics were used to evaluate agreement for the most prevalent bacterial species between sampling sites., Results: A positive culture result for at least one bacterial species was detected in 80% of samples from nasal discharge/nares, 92% of nasal cavity samples, and 75% of biopsy samples. The mean agreement between the three sampling sites for positive vs. negative culture results was never greater than moderate and the precision of the estimates of agreement varied widely.The most frequently isolated bacterial species in dogs were Staphylococcus pseudintermedius , Staphylococcus spp. and Streptococcus spp. In cats, Pasteurella spp. and Staphylococcus felis were the bacterial species cultured most frequently.For the most prevalent cultured species, Staphylococcus spp., mean agreement between sites was never greater than fair and the precision again varied widely., Conclusion: This study indicates that bacterial culture results in feline and canine nasal disease are site-specific and there was no evidence from this study for consistency between sites within a patient for many bacterial species. Consequently, if bacterial culture results from nasal swabs are used to guide therapeutic antimicrobial choice, different treatments may be selected depending on the site of culture. As a consequence, there is no evidence from this study that nasal bacterial cultures should be recommended as a routine diagnostic measure.

Background: Bacterial contamination of medical equipment is a significant risk in both human and veterinary medicine, leading to potential cross-contamination between clinicians, technicians, patients, and healthcare professionals. Pathogens can persist on surfaces, resulting in healthcare-associated infections. Tonometry used to measure intraocular pressure for diagnosing conditions like glaucoma and uveitis, is crucial in both human and veterinary care. However, there is limited information on bacterial transmission through reused tonometer probes in veterinary patients. This pilot study aimed to explore the potential for cross-contamination of bacteria via a rebound tonometer probe using bacterial isolates linked to bacterial ulcerative keratitis in dogs., Aim: To evaluate the potential for bacterial transmission via rebound tonometry using the ICare ® TONOVET Plus in a controlled laboratory environment. The study focused on two bacterial isolates commonly implicated in canine bacterial ulcerative keratitis: Pseudomonas aeruginosa and beta-haemolytic Streptococcus., Methods: Bacterial cultures were inoculated on blood agar and MacConkey agar plates. Tonometer probes were used to perform multiple corneal rebounds (CRs) readings on known bacterial samples, followed by multiple CR readings on agar growth plates, incubation, and re-examination of the agar plates for evidence of bacterial transfer. Controls included plates where the probes were only used on sterile agar., Results: Bacterial transmission was detected with distinct bacterial colonies corresponding to individual CR readings on the plates inoculated with Pseudomonas and beta-haemolytic Streptococcus . A control plate exhibited a single bacterial colony, likely due to inadvertent contamination., Conclusion: The results indicate that bacterial transmission via rebound tonometer probes is possible. Although this was an in vitro study, the findings raise concerns about cross-contamination in clinical practice. We isolated three novel strains, S1 T , S2 T , and S5 T , from human oral cavities and identified them as distinct novel species. All these strains are facultatively anaerobic, Gram-stain-positive, and non-flagellated bacteria. Their optimal growth conditions for these strains were observed in Columbia broth (CB) at 37 °C, pH 7.0, and in the absence of NaCl. Phylogenetic analyses, employing the 16S rRNA gene and whole-genome sequencing, confirmed that all three strains belong to the genus Streptococcus. The 16S rRNA gene sequences of strains S1 T , S2 T , and S5 T showed the highest similarities to Streptococcus parasanguinis, 98.57%, 99.05%, and 99.05%, respectively, and the orthologous average nucleotide identity (OrthoANI) values between the three strains and S. parasanguinis were 93.82%, 93.67%, and 94.04%, respectively. The pairwise OrthoANI values between the novel strains were 94.37% (S1 T -S2 T ), 95.03% (S2 T -S5 T ), and 94.71% (S1 T -S5 T ). All strains had C 20:1 ω9c and summed feature 8 (C 18:1 ω7c and/or C 18:1 ω6c) as major cellular fatty acids. Additionally, diphosphatidylglycerol (DPG) and hydroxyphosphatidylethanolamine (OH-PE) were identified as major polar lipids. Menaquinone was undetected in all strains. The results from the phylogenetic, phenotypic, chemotaxonomic, and genotypic analyses collectively indicated that strains S1 T , S2 T , and S5 T represent three distinct novel species within the genus Streptococcus, and we propose the names Streptococcus dentalis sp. nov. for strain S1 T (= KCTC 21234 T  = JCM 36526 T ), Streptococcus gingivalis sp. nov. for strain S2 T (= KCTC 21235 T  = JCM 36527 T ), and Streptococcus lingualis sp. nov. for strain S5 T (= KCTC 21236 T  = JCM 36528 T )., Competing Interests: Declarations. Background: Primary biliary cholangitis (PBC) is associated closely with the gut microbiota. This study aimed to explore the characteristics of the gut microbiota after the progress of PBC to cirrhosis., Method: This study focuses on utilizing the 16S rRNA gene sequencing method to screen for differences in gut microbiota in PBC patients who progress to cirrhosis. Then, we divided the data into training and verification sets and used seven different machine learning (ML) models to validate them respectively, calculating and comparing the accuracy, F1 score, precision, and recall, and screening the dominant intestinal flora affecting PBC cirrhosis., Result: PBC cirrhosis patients showed decreased diversity and richness of gut microbiota. Additionally, there are alterations in the composition of gut microbiota in PBC cirrhosis patients. The abundance of Faecalibacterium and Gemmiger bacteria significantly decreases, while the abundance of Veillonella and Streptococcus significantly increases. Furthermore, machine learning methods identify Streptococcus and Gemmiger as the predominant gut microbiota in PBC patients with cirrhosis, serving as non-invasive biomarkers (AUC = 0.902)., Conclusion: Our study revealed that PBC cirrhosis patients gut microbiota composition and function have significantly changed. Infective endocarditis (IE) is a life-threatening disease that affects the endocardial surface of the heart. Although heart valves are commonly involved in IE, in rare cases, vegetation is attached to the cardiac walls without valvular endocardial involvement, which is referred to as mural IE. In this case, a 60-year-old female presented with a seven-day history of fever associated with worsening pain in the right shoulder and left hip. Streptococcus dysgalactiae subsp. Equisimilis was detected in both blood and joint fluid cultures. Although transthoracic echocardiography revealed no mass, transesophageal echocardiography revealed a mobile mass in the fossa ovalis of the right atrium. She was subsequently diagnosed with mural IE and successfully treated with antibiotics without cardiac surgery. To our knowledge, only a few reports have described mural IE with vegetation in or around the fossa ovalis of the right atrium. This case highlights the importance of transesophageal echocardiography in diagnosing mural IE. Introduction: Streptococcus porcinus is considered a zoonotic opportunistic pathogen for several animal species, including swine, and can cause systemic clinical conditions. There are morphological similarities between streptococcal species, leading to possible incorrect diagnosis and inappropriate treatment choice. Here, we describe the identification, and genotypic and genomic characterization of S. porcinus isolated from 5 adult sick pigs from Minas Gerais and São Paulo (Brazil) between 2010-2017., Methodology: Streptococcus spp. strains were isolated from vaginal discharge, joint abscess, and brain tissue received for routine diagnosis. They were identified as S. porcinus by mass spectrometry and partial sequencing of the groEL gene. Genotypic characterization by amplified fragment length polymorphism was performed. In addition, the genome of one of the S. porcinus invasive strains was sequenced and comparative analyses were performed., Results: Genotyping revealed that the nervous system and joint abscess invasive strains had higher genetic similarity and clustered separately from vaginal discharge strains. Genome sequencing of one of the invasive strains revealed the presence of genes conferring resistance to erythromycin, tetracycline, lincosamides, and macrolides. A high level of similarity of the Brazilian strain genome with British and American strains was found. Objective: To investigate the pathogen species, composition, and distribution characteristics of infectious keratitis pathogens in Shandong Province and its surrounding areas. Methods: In this cross-sectional study, patients with keratitis who underwent corneal sampling and microbiological culture at the Shandong Eye Hospital from January 1, 2018 to December 31, 2022 were included. Under topical anesthesia, the edge of the lesion was scraped by an experienced physician. The samples were inoculated on blood agar and Sabouraud dextrose agar plates, separately for bacterial and fungal culture and identification. If necessary, the samples were inoculated on a non-nutrient agar medium with Escherichia coli for Acanthamoeba culture. Bacterial isolates were identified using Vitek 2 compact or matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Fungal isolates were identified based on morphological characteristics or sent to a company for sequencing in cases of difficult identification. The epidemiological data of the patients, pathogen species and number (counting as 1 strain if the same strain was isolated from multiple corneal specimens of the same patient), culture positivity rate, and seasonal distribution were recorded. Differences in pathogen positivity rates among different seasons were analyzed using the chi-square test. Results: Among the 4, 024 patients with infectious keratitis during the study period, there were 2 510 males (62.3%) and 1 514 females (37.6%), aged from 46 days to 94 years. Positive microbial culture results were achieved in 2, 363 patients (58.7%), including 906 cases (38.3%) with bacterial positivity, 1 231 cases (52.1%) with fungal positivity, 28 cases (1.2%) with Acanthamoeba positivity, and 198 cases (8.4%) with mixed fungal and bacterial infections. A total of 2 561 strains were isolated, including 1 104 bacterial strains. The most common bacteria were coagulase-negative Staphylococcus spp. (623/1 104, 56.4%), followed by Streptococcus spp. (131/1 104, 11.9%) and Pseudomonas aeruginosa (68/1 104, 6.2%). The most common fungi were Fusarium spp. (634/1 429, 44.4%), followed by Aspergillus spp. (279/1 429, 19.5%) and Alternaria spp. (229/1 429, 16.0%). Bacterial keratitis was more common in summer and autumn, while fungal keratitis was more common in autumn and winter. Conclusions: Among infectious keratitis cases in Shandong Eye Hospital, Fusarium species were predominant fungal pathogens, while coagulase-negative Staphylococcus predominated in bacterial pathogens. Both fungal and bacterial corneal infections showed seasonal variations.

Subclinical mastitis is an asymptomatic inflammatory condition that can be difficult to define and diagnose. In the dairy industry, subclinical mastitis is diagnosed by milk somatic cell counts (SCCs) of ≥250,000 cells mL -1 . In this pilot study, we assessed the efficacy of this index to identify human subclinical mastitis by comparing SCC levels with the inflammatory response [interleukin-8 (IL-8) levels] in 37 samples from asymptomatic and 10 clinical mastitis (CM) lactating women. The milk microbiota was determined by 16S rRNA gene sequencing. The SCC of CM samples ranged from 310,000 to 6,600,000 cells mL -1 . However, 14 of 37 (37.8%) asymptomatic samples had high SCC (250,000-460,000 cells mL -1 ), indicating subclinical mastitis. SCC levels significantly ( P < 0.001) and positively correlated with milk IL-8 levels reflecting the escalating inflammatory response across subclinical and clinical mastitis samples. Samples with an SCC of ≥250,000 cells mL -1 showed significant increases in IL-8 responses when compared with milk samples from healthy women. The milk microbiome of CM samples was dominated by streptococcal and staphylococcal species (89.9% combined median relative abundance). In contrast, the combined median streptococcal/staphylococcal relative levels were 75.4% and 66.3% in milks from asymptomatic (subclinical mastitis) and healthy groups, respectively. The Streptococcus genus was increased in samples with an SCC of ≥250,000, although this should be interpreted with caution. Thus, the index of ≥250,000 somatic cells mL -1 could be a reliable indicator of subclinical mastitis in humans and should aid future studies investigating the impact of subclinical mastitis on maternal health, breastfeeding behaviors, infant health, and development., Importance: This pilot study suggests that SCC at a level of (greater than or equal to) 250,000 cells mL -1 , as used in the dairy industry, is a suitable index to identify asymptomatic subclinical mastitis in lactating women since it reflects a significant increase in the inflammatory response compared to milk samples from healthy women. Reports of novel species of α-hemolytic Streptococcus have increased recently. However, limited information exists regarding the pathogenicity of these species, with the exception of Streptococcus pneumoniae and Streptococcus pseudopneumoniae . In this study, a quinolone-resistant α- Streptococcus strain, MTG105, was isolated from the sputum of a patient with pneumonia. This strain was first identified as S. pneumoniae at the hospital laboratory; however, it exhibited unique genetic features upon further analysis. Digital DNA-DNA hybridization and average nucleotide identity based on BLAST values from whole-genome sequencing revealed MTG105 to be a novel species closely related to S. pseudopneumoniae . Although MTG105 carried two copies of the pneumolysin gene, similar to S. pseudopneumoniae , this isolate exhibited susceptibility to optochin under both aerobic and 5% CO 2 conditions. Notably, no biochemical features could be used to definitively identify this species. In an infection assay using organotypic lung tissue models, MTG105 induced epithelial damage comparable to that of S. pneumoniae and S. pseudopneumoniae , possibly suggesting its potential as a pathogenic α- Streptococcus . The natural transformation abilities of Streptococcus species facilitate their exchange of genes within the same genus, resulting in the existence of species with increasingly more diverse genome structures. Therefore, the identification of this species highlights the importance of monitoring the emergence of novel species exhibiting virulence and/or multidrug resistance. This isolate was proposed as a novel species, designated Streptococcus nakanoensis sp. nov. Background: Empyema is a serious infection in pleural space. Finding out seasonal variations of empyema and its pathogens can help in providing preventive measures, and implicating future researches., Methods: This study is a 10-year observational study in a single center. Patients with empyema thoracis receiving thoracoscopic decortication between January 2012 and December 2021 were included in the study., Results: There were 1082 empyema patients enrolled in this study. No seasonal variation was noted (spring = 25.7 %, summer =25.5 %, autumn = 24.8 %, winter = 24.0 %). However, we observed seasonal variations in pathogens. Streptococcus species had slightly higher prevalence in winter and spring than summer and autumn (54.3 % vs. 45.7 %) without significant difference (p = 0.251). On the contrary, Staphylococcus species occurred more often in summer and autumn than winter and spring (61.5 % vs. 38.5 %) (p = 0.035). Klebsiella species were more likely found in autumn (34.9 %) (p = 0.050), and Pseudomonas species showed no peak prevalence in any season (p = 0.423). The incidence of Streptococcus species increased over the years., Conclusions: Although no seasonal variation was found in severe empyema patients requiring surgery, there were seasonal variations for the pathogens in Taiwan. Purpose: Patients with non-beta-hemolytic streptococcal bacteremia (NBHSB) are at risk of infective endocarditis (IE). Patients with cardiac implantable electronic device (CIED) have been described to have an increased risk of IE. The aim of the study was to describe a population-based cohort of patients with NBHSB and CIED and variables associated with IE and recurrent NBHSB., Methods: All episodes with NBHSB in blood culture from 2015 to 2018 in a population of 1.3 million inhabitants were collected from the Clinical Microbiology Laboratory, Lund, Sweden. Through medical records, patients with CIED during NBHSB were identified and clinical data were collected. Patients were followed 365 days after NBHSB., Results: Eighty-five episodes in 79 patients with CIED and NBHSB constituted the cohort. Eight patients (10%) were diagnosed with definite IE during the first episode, five of whom also had heart valve prosthesis (HVP). In 39 patients (49%) transesophageal echocardiography (TEE) was performed of which six indicated IE. Four patients had the CIED extracted. Twenty-four patients did not survive (30%) the study period. Four patients had a recurrent infection with NBHSB with the same species, three of whom had HVP and had been evaluated with TEE with a negative result during the first episode and diagnosed with IE during the recurrency., Conclusion: The study did not find a high risk of IE in patients with NBHSB and CIED. Introduction. Streptococcus uberis is a common cause of mastitis in cattle, leading to significant economic losses. The widespread use of antimicrobials has contributed to the emergence of resistance, which poses a severe challenge in controlling S. uberis infection. Aim. The objective of this study was to gain insights into the antimicrobial resistance (AMR) and epidemiological typing of S. uberis isolated from milk collected from bovine mastitis on dairy farms in Thuringia. Methodology. In this study, 84 S. uberis isolates were obtained from cattle with clinical mastitis in Thuringia, their phenotypic and genotypic AMR were analyzed and their phylogenetic relationship was explored using whole-genome sequencing. Results. Genetically heterogeneous strains were found on the farms, but clusters of highly similar strains also circulated within the same farms. All isolates were sensitive to ampicillin, penicillin, ceftiofur, and vancomycin. However, 42.9%, 42.9%, 22.6%, 19.0%, and 13.0% were resistant to tetracycline, doxycycline, clindamycin, pirlimycin, and erythromycin, respectively. Thirty-nine strains were phenotypically resistant to two or more tested antibiotics. We identified a plasmid associated with macrolide and lincosamide resistance in 12% of the strains. Conclusion. The emergence of S. uberis strains resistant to multiple antibiotics highlights the importance of S. uberis surveillance and the prudent use of antimicrobials.

We present a case of Streptococcus iniae causing leptomeningitis, pyocephalus, and subdural empyema in an elderly male from India. There have been only a handful of cases of S. iniae infection reported worldwide, and none of them have been from India. In this case, an elderly diabetic patient presenting with backache, headache, and fever with severe neurological worsening was diagnosed with severe invasive S. iniae infection. He had hydrocephalus that needed ventriculoperitoneal shunting. Objective: The relationship between the in vitro detected virulence factors biofilm and capsule formation of Streptococcus (S.) uberis isolates of clinical mastitis in dairy cows and the bacteriological cure rate after antibiotic therapy was investigated in order to better understand the importance of these virulence factors for the bacteriological cure rate., Material and Methods: A total of 111 clinical mastitis (CM) cases were collected, in which S. uberis was bacteriologically detected. All mastitis cases were treated in accordance with the approval conditions of the antibiotic udder tubes used. Individual cow information including age, number of lactations, current lactation mastitis and antimicrobial treatment received was recorded. The microtiter plate test was used to detect biofilm formation and Anthony capsule staining was used to detect capsular capacity. Statistical analyses were performed to characterize the correlation between in vitro virulence factors and bacteriological cure (BC) rate., Results: 30.5% (n=29) of the S. uberis isolates of bacteriologically cured cases and 34.5% (n=10) of the isolates of bacteriologically non-cured mastitis cases were found to have the ability to produce capsules in vitro . 70.7% (n=58) of the S. uberis isolates from bacteriologically cured mastitis cases had the ability to produce biofilm in vitro , whereas 58.6% (n=17) of the isolates of non-cured mastitis cases showed ability in producing biofilm. No correlation was found between the in vitro ability of S. uberis to form capsules and biofilms and the BC rate after antibiotic treatment of bovine mastitis., Conclusion(s): The present work has shown that the investigated in vitro virulence factors are not associated with the BC after antibiotic therapy. Further studies on the role of S. uberis virulence factors are needed to complete the missing knowledge on the difficulties in curing S. uberis mastitis., Clinical Relevance: This study is of great clinical relevance since it enhances the understanding of the occurrence of BC in S. uberis mastitis cases. Bovine mastitis poses a significant threat to global dairy production, resulting in substantial losses in milk production. Streptococcus bacteria, particularly Streptococcus uberis, Streptococcus agalactiae, and Streptococcus dysgalactiae, are commonly implicated in this condition. An accurate diagnosis is crucial for implementing effective treatment and minimizing its impact on production. This study examined 115 Streptococcus strains isolated from bovine mastitis cases in Uruguay using PCR for species identification. Additionally, the resistance to tetracycline, erythromycin, and penicillin was assessed in 81 of the bacterial strains under study. Significant disparities between phenotypic and genotypic detection were evident across all three species, with only 31% of strains identified phenotypically aligning with PCR results. Phenotypic prevalence indicated S. dysgalactiae as the most prevalent (44.35%), followed by S. uberis (24.34%) and S. agalactiae (6.09%). However, the genotypic identification revealed S. uberis as the most prevalent, followed by S. dysgalactiae, while S. agalactiae remained the least prevalent. The high sensitivity and speed of PCR suggest its potential routine implementation for diagnosing bovine mastitis caused by Streptococcus in any laboratory. Although, penicillin resistance was practically nonexistent, tetracycline and erythromycin exhibit higher resistance levels across all three species studied. The objective of this study was to analyze the in vitro antimicrobial resistance (AMR) of Streptococcusdysgalactiae, Strep. agalactiae, and Strep. canis over a 10-yr period from 2012 to 2022 against the most commonly used antimicrobial agents. For this purpose, all quarter milk samples (QMS) submitted to the milk laboratory of the Bavarian Animal Health Service were analyzed. Each QMS was tested using the California Mastitis Test and categorized as negative, subclinical, or clinical mastitis if the milk character was abnormal. Samples with Strep. dysgalactiae, Strep. agalactiae, or Strep. canis were included and a subset of isolates were further tested for in vitro AMR by breakpoint analysis with broth microdilution. Streptococcus dysgalactiae (61%, n = 65,750) was the most abundant pathogen among those 3 species, followed by Strep. agalactiae (28%, n = 30,486), and Strep. canis (11%, n = 11,336). All 3 species showed the highest resistance to the same 4 antimicrobial agents: erythromycin, marbofloxacin, pirlimycin, and cefalexin/kanamycin with varying degrees of resistance. Throughout the study period, Strep. dysgalactiae, Strep. agalactiae, and Strep. canis were largely susceptible to the remaining antimicrobial agents tested (penicillin, amoxicillin-clavulanate, oxacillin, cefazolin, cefoperazone, cefquinome). Only less than 14% of isolates of Strep. dysgalactiae and Strep. canis were resistant against any of the antimicrobials tested. Streptococcus agalactiae was the species with the highest percentage of resistant isolates. Although the percentage of resistant isolates from Strep. canis and Strep. dysgalactiae decreased, the percentage of resistant Strep. agalactiae isolates increased after 2017. In summary, most isolates were not resistant to the most commonly used antimicrobial agents for mastitis therapy, including β-lactam antibiotics and penicillin should remain the first-choice therapy against streptococcal mastitis., (The Authors. Four Gram-stain-positive bacterial strains (designated 475-2 T , 46-6B T , 778-2 T and A810-3), isolated from traditional Chinese pickle, were characterized using a polyphasic taxonomic approach. Strain 475-2 T was most closely related to the type strain of Lapidilactobacillus achengensis, having 99.9% 16S rRNA gene sequence similarity, 94.1-95.1% average nucleotide identity (ANI) and 57.6% digital DNA-DNA hybridization (dDDH) values. Strain 46-6B T was most closely related to the type strain of Secundilactobacillus similis, having 99.8% 16S rRNA gene sequence similarity, 94.3-94.9% ANI and 58.9-59.2% dDDH values. Strains 778-2 T and A810-3 were phylogenetically related to the type strains of Streptococcus salivarius, Streptococcus thermophilus and Streptococcus vestibularis, having 99.7-99.9% 16S rRNA gene sequence similarities, 89.1-94.4% ANI and 39.0-55.5% dDDH values. Based upon the data obtained in the present study, three novel species, Lapidilactobacillus salsurivasis sp. nov., Secundilactobacillus muriivasis sp. nov. and Streptococcus parasalivarius sp. nov., are proposed and the type strains are 475-2 T (= JCM 36613 T  = CCTCC AB 2023258 T  = LMG 33412 T ), 46-6B T (= JCM 36612 T  = CCTCC AB 2023259 T  = LMG 33411 T ) and 778-2 T (= JCM 36614 T  = CCTCC AB 2023257 T  = LMG 33413 T ), respectively., (© 2024. Purpose: Streptococcus dysgalactiae subsp. equisimilis (SDSE) has increasingly been recognised as a significant pathogen that causes a myriad of infections, ranging from cellulitis to invasive infections, including bacteraemia and even toxic shock syndrome. The aim of this study was to examine the epidemiology and disease manifestations of bacteraemia caused by SDSE., Methods: We retrospectively reviewed cases of SDSE bacteraemia in adults aged ≥ 18 years admitted to four public hospitals in Western Sydney, Australia, between January 2015 and December 2020. We reviewed demographics, comorbidities, disease manifestations, management, and outcomes., Results: There were 108 patients with SDSE bacteraemia over a six-year period. The median age of individuals with SDSE bacteraemia was 70 years (interquartile range, IQR, 58-85 years). Cardiovascular disease (46%), chronic skin conditions (44%) and diabetes (37%) were the most common comorbidities. Ten patients (9%) with SDSE bacteraemia had healthcare-acquired infections. Skin and skin structure infections (SSTIs) were the most common presentations (59%), while bone and joint infections (BJIs) represented 13% of the cases. Twenty patients (19%) had septic shock on presentation. Fifteen patients (14%) were prescribed clindamycin, while one patient received intravenous immunoglobulin (IVIg). Infective endocarditis (IE) was present in 3% of patients; however, only 44% of the total patients had an echocardiogram. The 30-day mortality rate was 13%, but it was greater in those aged > 75 years (21%). The average length of hospital stay for patients who survived was 15 days, and the average duration of intravenous therapy was 12 days., Conclusion: SDSE bacteraemia is typically a community-onset infection with a fifth of patients in our cohort presenting with septic shock. To explore the potential storage and safety of drinking leftover bottled tea beverages from various manufacturers after direct drinking from bottles, we conducted a screening experiment on the growth of salivary bacteria in plastic bottles of tea. The diluted saliva samples from 10 participants were inoculated into the test bottled beverages, which resulted in bacteria, particularly former members of the genus Lactobacillus, growing in some green tea beverages with a neutral pH. Bovine mastitis is an inflammation of the mammary gland, and it is the most common infectious disease in dairy cattle. Mastitis reduces milk yield and quality, costing dairy farmers millions of dollars each year. The aim of this study was to develop a point-of-need test for identifying mastitis pathogens that is field portable, cost-effective and can be used with minimal training. Using a proprietary polymer-based milk sample preparation method to rapidly extract pathogen DNA in milk samples, we demonstrated quantitative Polymerase Chain Reaction (qPCR) assays for six common bovine bacterial mastitis pathogens: Staphylococcus aureus, Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, Mycoplasma bovis and Escherichia coli. We also implemented this sample preparation method on a prototype point-of-need system in a proof-of-concept field trial to evaluate user experience. Importantly, the protype system enabled a sample-to-result turnaround time of within 70 min to quantitatively detect all six target pathogens. The key advantage of our point-of-need prototype system is being culture-independent yet providing automated milk sample preparation for molecular identification of key mastitis pathogens by non-expert users. Our point-of-need prototype system showed a good correlation to laboratory-based qPCR for target pathogen detection outcomes, thus potentially removing the need for milk samples to be transported off-site for laboratory testing. Diapers, being one of the most used items for infant care, it is imperative that they are always free of contamination from microorganisms. The objective of this study is to determine the rate of bacterial contamination and the antibiotics profile of isolated bacteria from unused diapers sold in markets in Enugu Metropolis, southeast Nigeria. The study also investigated the effectiveness of the Nigerian Consumer protection laws towards maintaining the standard of care for infants and toddlers. Fifty pieces of different brands of diapers were sampled for bacterial contamination using standard bacteriological procedures. The results show that out of 50 samples analyzed, bacterial growths were identified in 9 (18%), of which five different bacterial species were isolated. The most prevalent being Escherichia coli and Staphylococcus aureus 3 (33.3%). Other bacteria isolated were Lactobacillus spp, Klebsiella spp, and Streptococcus spp 1 (11.1%). There was no statistically significant difference in the distribution of the bacterial contamination of diapers across the different brands(p>0.05). Streptococcal toxic shock syndrome (STSS) is an uncommon disorder characterised by hypotension and multiorgan failure in the setting of streptococcal infection. Recurrent STSS is rare and has been due to recurrence of the same streptococcal species. Strangles is a globally widespread, commonly diagnosed and important infectious disease of equids caused by Streptococcus equi subsp. equi. We performed whole genome sequencing of 19 S. equi isolates collected from imported horses at the Japanese border. Of these isolates, 15 isolates were obtained from clinical cases and 4 were from subclinical cases. The 19 isolates were grouped into 3 Bayesian analysis of population structure (BAPS) groups by the core genome single nucleotide polymorphism analysis corresponding to exporting country, SeM typing, or exporter of the horses. The 19 isolates possessed same pathogenic genes regardless of clinical status in imported horses and no antimicrobial resistance genes. The disease status of the horses may rather reflect the prior exposure of animals with sub-clinical infection to S. equi.

The long-read sequencing platform MinION, developed by Oxford Nanopore Technologies, enables the sequencing of bacterial genomes in resource-limited settings, such as field conditions or low- and middle-income countries. For this purpose, protocols for extracting high-molecular-weight DNA using nonhazardous, inexpensive reagents and equipment are needed, and some methods have been developed for gram-negative bacteria. However, we found that without modification, these protocols are unsuitable for gram-positive Streptococcus spp., a major threat to fish farming and food security in low- and middle-income countries. Multiple approaches were evaluated, and the most effective was an extraction method using lysozyme, sodium dodecyl sulfate, and proteinase K for lysis of bacterial cells and magnetic beads for DNA recovery. We optimized the method to consistently achieve sufficient yields of pure high-molecular-weight DNA with minimal reagents and time and developed a version of the protocol which can be performed without a centrifuge or electrical power. The suitability of the method was verified by MinION sequencing and assembly of 12 genomes of epidemiologically diverse fish-pathogenic Streptococcus iniae and Streptococcus agalactiae isolates. The combination of effective high-molecular-weight DNA extraction and MinION sequencing enabled the discovery of a naturally occurring 15 kb low-copy number mobilizable plasmid in S. iniae, which we name pSI1. We expect that our resource-limited settings-adapted protocol for high-molecular-weight DNA extraction could be implemented successfully for similarly recalcitrant-to-lysis gram-positive bacteria, and it represents a method of choice for MinION-based disease diagnostics in low- and middle-income countries., (© 2024 The Author(s). Purpose: To analyze the correlation between the changes in salivary ion concentration, Streptococcus and Bifidobacterium in children with dental caries and the severity of the disease., Methods: Eighty children with dental caries treated from May 2022 to April 2023 were selected as the experimental group. According to the DMFT, they were divided into mild group (DMFT≤15%, n=35) and severe group (DMFT>15%, n=45). Another 80 children without dental caries were selected as the control group. The calcium ion concentration in saliva was detected by methanol-permeabilized calciometric method, the level of Streptococcus and Bifidobacterium was detected by real-time single-quantity PCR technology, and the correlation between DMFT and salivary calcium ion concentration, Streptococcus and Bifidobacterium was analyzed by Pearson correlation analysis. The diagnostic value of salivary calcium ion concentration, Streptococcus and Bifidobacterium for the severity of dental caries in children was evaluated by the receiver operating characteristic curve(ROC), and the area under the curve(AUC) was calculated. SPSS 22.0 software package was used for statistical analysis., Results: The level of salivary calcium ion concentration, Streptococcus mutans, Streptococcus sobrinus and Bifidobacterium in the experimental group was significantly higher than that in the control group(P＜0.05), and significantly higher in the severe group than in the mild group(P＜0.05). DMFT was significantly positively correlated with salivary calcium ion concentration, Streptococcus and Bifidobacterium (P＜0.05). Based on DMFT, ROC curve was prepared, and AUCs of salivary calcium ion concentration, Streptococcus mutans, Streptococcus sobrinus and Bifidobacterium for assessing the severity of dental caries in children were 0.760, 0.658, 0.718 and 0.705, respectively., Conclusions: Increased salivary ion concentration, Streptococcus sobrinus, Streptococcus mutans and Bifidobacterium levels are related to dental caries in children, and are positively correlated with the severity of the disease, which has a certain value for assessing the severity of the disease.

Many of the reproductive tract infections in the bitches are caused by bacteria that can normally be present on the vaginal mucosa. These bacteria also might have an important role as the cause responsible for pregnancy loss and fetal deaths. The choice of antibiotic therapy for the pregnant animal is narrow and represents a severe problem in veterinary practice, especially due to increased antimicrobial resistance. Due to incorrect antimicrobials use in breeding kennels, the aim of the present study was to assess the occurrence of the bacterial flora isolated from the pregnant bitches and their antibiotic sensitivity. The study was carried out at the private Veterinary clinic in Novi Sad, Serbia. The vaginal swabs were taken from 60 bitches diagnosed with pregnancy and were sent to be laboratory tested. Based on the results, the most common isolated pathogens were Staphylococcus pseudintermedius (20%) and beta-hemolytic streptococci (18.33%). Furthermore, significant resistance to antibiotics from beta-lactams group was detected.
