Ultrasonic approach to the synthesis of the first selenium-containing derivatives of chitin and chitosan has been developed. The synthetic procedure is simple, provides high yields, does not require harsh conditions, and uses water as the reaction medium. The elaborated chitin and chitosan derivatives and their based nanoparticles are non-toxic and possess high antibacterial and antifungal activity. Their antimicrobial activity exceeds the effect of the classic antibiotics (Ampicillin and Gentamicin) and the antifungal drug Amphotericin B. The obtained selenium-containing cationic chitin and chitosan derivatives exhibit a high transfection activity and are promising gene delivery vectors. Nanoparticles of the synthesized polymers are highly efficient catalysts for the oxidation of 1-phenylethyl alcohol to acetophenone by bromine at room temperature. In this work, we demonstrate that the thiol-yne click reaction could be efficiently mediated by ultrasonic irradiation and implement the ultrasound-assisted thiol-yne click reaction to chitosan chemistry as a polymer-analogous transformation. We optimize power and frequency of ultrasound to preserve selectivity of the click reaction and avoid ultrasonic degradation of the chitosan polymer chain. Thus, we obtain a new water-soluble betaine. Using ionic gelation of the obtained betaine derivatives of chitosan, we prepare nanoparticles with a unimodal size distribution. Furthermore, we present results of antibacterial and transfection activity tests for the chitosan derivatives and their based nanoparticles. The derivative with a medium molecular weight and a high degree of substitution demonstrated the best antibacterial effect. It derived nanoparticles with a size of ca. 100 nm and ζ-potential of ca. +69 mV revealed even higher antibacterial activity, slightly superior to commercial antibiotics ampicillin and gentamicin. On the contrary, the obtained polymers possess a much more pronounced transfection activity as compared with their based nanoparticles and species with a low degree of substitution acts as the most efficient transfecting agent. Poly(β-amino ester)s (PAEs) have been widely developed for gene delivery, and hydrophobic modification can further enhance their gene transfection efficiency. However, systematic manipulation of amphiphilicity of PAEs through copolymerization with hydrophobic monomers is time-consuming and, to some extent, uncontrollable. Here, a modular strategy is developed to manipulate the amphiphilicity of the PAE/DNA polyplexes. A hydrophobic polymer (DD-C12-122) and a hydrophilic polymer (DD-90-122) are synthesized separately and used as a hydrophobic module and a hydrophilic module, respectively. The amphiphilicity of polyplexes could be manipulated by changing the ratio of the hydrophobic module and hydrophilic module. Using the modular strategy, the PAE/DNA polyplexes with the highest gene transfection efficiency and safety profile as well as possible mechanisms are identified. The modular strategy provides a novel way to engineer the hydrophobicity of PAEs to improve their gene transfection and can be easily generalized and potentially extended to other polymeric gene delivery systems.

The presented work describes the synthesis and characterization of a novel magnetic cationic phospholipid (MCP) system with a stable dopamine anchor as well as its transfection activity study. The synthesized architectural system increases the biocompatibility of iron oxide and promises applications of magnetic nanoparticles in living cells. The MCP system is soluble in organic solvents and can be easily adapted to prepare magnetic liposomes. We created complexes with liposomes containing MCP and other functional cationic lipids and pDNA as gene delivery tools, which possessed the ability to enhance the efficiency of transfection, particularly the process of interaction with cells by inducing a magnetic field. The MCP is able to create iron oxide nanoparticles and has the potential for the materials to prepare the system for site-specific gene delivery with the application of an external magnetic field.

Cholesterol derivatives M1 – M6 as synthetic cationic lipids were designed and the biological evaluation of the cationic liposomes based on them as non-viral gene delivery vectors were described. Plasmid pEGFP-N1, used as model gene, was transferred into 293T cells by cationic liposomes formed with M1 – M6 and transfection efficiency and GFP expression were tested. Cationic liposomes prepared with cationic lipids M1 – M6 exhibited good transfection activity, and the transfection activity was parallel ( M2 and M4 ) or superior ( M1 and M6 ) to that of DC-Chol derived from the same backbone. Among them, the transfection efficiency of cationic lipid M6 was parallel to that of the commercially available Lipofectamine2000. The optimal formulation of M1 and M6 were found to be at a mol ratio of 1:0.5 for cationic lipid/DOPE, and at a N/P charge mol ratio of 3:1 for liposome/DNA. Under optimized conditions, the efficiency of M1 and M6 is greater than that of all the tested commercial liposomes DC-Chol and Lipofectamine2000, even in the presence of serum. The results indicated that M1 and M6 exhibited low cytotoxicity, good serum compatibility and efficient transfection performance, having the potential of being excellent non-viral vectors for gene delivery. The chemical structure of methylated poly(1-vinylimidazole) (PVIm-Me) has been tuned for gene carrier properties, which are cell viability, the stability of its DNA polyion complexes, and gene expression. The resulting PVIm-Me with lower methylation degree (6 and 20 mol%) exhibited no significant cytotoxicity, as compared with 40 mol% PVIm-Me, in spite of the same concentration and chemical structure of positive charges. The DNA complex with 20 mol% PVIm-Me stably retained the DNA, as compared with 6 or 40 mol% PVIm-Me, which was examined by competitive exchange with dextran sulfate. As a result, the DNA complex with 20 mol% PVIm-Me mediated most efficient gene delivery, where the resulting transfection activity was higher than that mediated by a positive control, poly(ethylenimine). These results suggest that the optimization of the balance of the methylimidazolium and the imidazole groups of PVIm-Me is essential for gene carrier design. Copyright © 2014 John Wiley & Sons, Ltd. Abstract: The purpose of the study was to investigate the influence of cationic polymer structure on the formation of DNA–polycation complexes and their transfection activity. Primary, tertiary, and quaternary polyamines with molecular masses ranging from 8000 to 200,000 were investigated. DNA–cationic polymer interaction was characterized by low gradient viscometry, dynamic light scattering, circular dichroism, UV spectrometry, flow birefringence, DNA electrophoresis, and electron microscopy. Transfection activity of the complexes was evaluated by the expression of reporter gene (β-galactosidase) and using synthetic FITC-labelled oligonucleotides. Complex formation was found to be dependent on the structure and molecular weight of the polymer and the ionic strength of the solution. Secondary DNA structure in complexes was not disrupted, and DNA was protected from protonation. Cell lines of different origin were used for testing of transfection activity of the complexes. The sensitivity of the cells to transfection was established to be highly dependent on the cell line. DNA–polycation complexes are non-toxic according to MTT. Polyallylamine, and polydimethylaminoethylmethacrylate were found to be the most promising polycations for gene delivery. Transfection efficacy of their complexes with DNA to T-98G cells reaches up to 90–100%. It was found that optimal molecular mass of polydimethylaminoethylmethacrylate is in the range of 8000–50,000Da. [Copyright &y& Elsevier]

Abstract: We synthesized new cationic lipids, analogue to N-[1-(2,3-dioleoyloxy)propyl]-N,N,N-trimethylammonium chloride (DOTMA) and 1,2-dimyristyloxypropyl-3-dimethyl-hydroxyethylammonium bromide (DMRIE), in order to compare those containing a dodecyl chain with those having a relatively long chain with two or five double bonds, such as squalenyl and dihydrofarnesyl derivatives, or complex saturated structures, such as squalane derivatives. The fusogenic helper lipid dioleoylphosphatidylethanolamine (DOPE) was added to cationic lipids to form a stable complex. Liposomes composed of 50:50 w/w cationic lipid/DOPE were prepared and incubated with plasmidic DNA at various charge ratios and the diameter and zeta potential of the complexes were measured. The surface charge of the DNA/lipid complexes can be controlled by adjusting the cationic lipid/DNA ratio. Finally, we tested the in vitro transfection efficiency of the cationic lipid/DNA complexes using different cell lines. The transfection efficiency was highest for the dodecyloxy derivative containing a single hydroxyethyl group in the head, followed by the dodecyloxy and the farnesyloxy trimethylammonium derivatives. An optimization of ultrasonic irradiation parameters allowed us to (i) accelerate the rate of the reactions dramatically, (ii) achieve high selectivity, and (iii) preserve integrity of the polysaccharide backbone avoiding its depolymerization. We evaluated antibacterial/antifungal and transfection activity of 8 different derivatives of chitosan and their based nanoparticles in vitro. Moreover, we studied antibacterial activity of the most efficient polymer and their based nanoparticles in vivo.
