The growing world population and global increases in the standard of living both result in an increasing demand for food, feed and other plant-derived products. In the coming years, plant-based research will be among the major drivers ensuring food security and the expansion of the bio-based economy. Crop productivity is determined by several factors, including the available physical and agricultural resources, crop management, and the resource use efficiency, quality and intrinsic yield potential of the chosen crop. This review focuses on intrinsic yield potential, since understanding its determinants and their biological basis will allow to maximize the plant's potential in food and energy production. Yield potential is determined by a variety of complex traits that integrate strictly regulated processes and their underlying gene regulatory networks. Due to this inherent complexity, numerous potential targets have been identified that could be exploited to increase crop yield. These encompass diverse metabolic and physical processes at the cellular, organ and canopy level. We present an overview of some of the distinct biological processes considered to be crucial for yield determination that could further be exploited to improve future crop productivity.

With the provoked environmental constraints under extreme climatic events, a better understanding of plant responses to these environmental stresses helps in obtaining sustainable productivity. Wheat is a significant cereal crop for the burgeoning population; its yield is significantly limited by too little water in the rhizosphere. The ramifications of water deficiency on the wheat crop can be reduced by the application of vermicompost. With the objective to cope with drought stress, a wire-house experiment was established where seedlings of two cultivars, viz., Faisalabad-08 and Galaxy-13 (drought-tolerant and -sensitive cultivar, respectively) were grown in pots and exposed to mild drought (D1, 45% field capacity) and severe drought stress (D2, 30% field capacity). A control with well-watered condition (70% field capacity) was kept for comparison. Various application rates of wheat straw vermicompost (control (VT0), 4 t ha−1 (VT1), 6 t ha−1 (VT2), and 8 t ha−1 (VT3)), were used in soil-filled pots under drought and control treatments. Our data depicted that compared with control, drought treatments recorded a significant reduction in morpho-physiological and biochemical attributes with maximum reduction under severe drought conditions. Nonetheless, it was observed that soil application of vermicompost, particularly at a high rate, ameliorated the negative effects of drought. Under severe drought conditions, a significant and positive influence on morphological and physiological traits was recorded for VT3 treatment, which increased root and shoot length by 27.55 and 27.85%, root and shoot fresh weight by 26.98 and 28.20%, root and shoot dry weight by 40 and 50.05%, and photosynthesis and transpiration rate by 27.65 and 49.25%, respectively, on average of two cultivars. Similarly, VT3 also significantly ameliorated the adverse effect of drought by enhancing the antioxidant enzyme activities as it increased superoxide dismutase activity by 14.28%, peroxidase by 27.28%, and catalase by 50% compared to the control treatment. Among cultivars, Faisalabad-2008 showed comparatively more resistance against drought stress. The findings of this work revealed that drought drastically reduced the growth and productivity of wheat; however, soil-applied vermicompost positively influenced the performance of wheat cultivars.

Selenium (Se) is found in plants both in inorganic and organic forms as selenoamino acids and methylated complexes. Se influences growth and physiological parameters, viz. root and shoot growth, starch accumulation, water status regulation, respiration, germination promotion, nitrogen assimilation and delaying senescence in plants. Se deficiency causes many human medical conditions, viz. cancer, liver disease, cardiovascular disease, thyroid disease, Keshan disease and central nervous system disorders. Plants need Se to alleviate abiotic stresses, viz. high/low temperatures, drought, salinity, light, UV-B radiation and toxic metals/metalloids. Ameliorative role of Se, applied in relatively low doses, against abiotic stresses was mediated via enzymatic and non-enzymatic antioxidation. Chemical form, application method, edaphic presence and bioavailability of Se and its interaction with other elements regulate Se biofortification and abiotic stress tolerance in food and medicinal crops. Popular scientific search engines, viz. PubMed, Pubget, ScienceDirect, Medline, Scopus, SpringerLink, Mendeley, EMBASE, Google Scholar and JSTOR were searched to retrieve published articles by using pre-determined search strings involving selenium, plants, stress, metals, metalloids, etc. The retrieved literature encompasses the role of Se biofortification in enhancing dietary Se in crops and its bioavailability. The review also presents recent trends in Se-mediated amelioration of toxic metal and metalloid stresses via maintaining cell membrane integrity, functioning of photosynthetic machinery, inhibition against uptake and translocation of toxic metals and metalloids, control over the uptake and distribution of mineral nutrition and conversion of toxic metals and metalloids to non-toxic Se-metal complexes. Furthermore, current research on the genetic engineering and omics studies on Se biofortification and Se-mediated abiotic stress tolerance are also discussed. The present review comprehensively elucidates the beneficial role Se biofortification in developing resilience in crops against toxic metals and metalloids. Among cereal crops, wheat has been identified as a major source for human food consumption. Wheat breeders require access to new genetic diversity resources to satisfy the demands of a growing human population for more food with a high quality that can be produced in variable environmental conditions. The close relatives of domesticated wheats represent an ideal gene pool for the use of breeders. The genera Aegilops and Triticum are known as the main gene pool of domesticated wheat, including numerous species with different and interesting genomic constitutions. According to the literature, each wild relative harbors useful alleles which can induce resistance to various environmental stresses. Furthermore, progress in genetic and biotechnology sciences has provided accurate information regarding the phylogenetic relationships among species, which consequently opened avenues to reconsider the potential of each wild relative and to provide a context for how we can employ them in future breeding programs. In the present review, we have sought to represent the level of genetic diversity among the wild relatives of wheat, as well as the breeding potential of each wild species that can be used in wheat-breeding programs.

International audience; The Nitrogen Use Efficiency (NUE) of grain cereals depends on nitrate (NO3-) uptake from the soil, translocation to the aerial parts, nitrogen (N) assimilation and remobilization to the grains. Brachypodium distachyon has been proposed as a model species to identify the molecular players and mechanisms that affects these processes, for the improvement of temperate C3 cereals. We report on the developmental, physiological and grain-characteristic responses of the Bd21-3 accession of Brachypodium to variations in NO3- availability. As previously described in wheat and barley, we show that vegetative growth, shoot/root ratio, tiller formation, spike development, tissue NO3- and N contents, grain number per plant, grain yield and grain N content are sensitive to pre- and/or post-anthesis NO3- supply. We subsequently described constitutive and NO3--inducible components of both High and Low Affinity Transport Systems (HATS and LATS) for root NO3- uptake, and BdNRT2/3 candidate genes potentially involved in the HATS. Taken together, our data validate Brachypodium Bd21-3 as a model to decipher cereal N nutrition. Apparent specificities such as high grain N content, strong post-anthesis NO3- uptake and efficient constitutive HATS, further identify Brachypodium as a direct source of knowledge for crop improvement.

Salinity is affecting plant growth and development. Low concentration of hydrogen peroxide (H2O2) has shown to be effective against various stress factors. In this study, effect of different H2O2 priming methods on growth, physiological and biochemical parameters in three wheat varieties (NKU Lider, Sultan-95, and Tosunbey) under salt stress were investigated. Salt stress (0 and 160 mM NaCl) was applied gradually to 100 mu M H2O2 applied (-H2O2: negative control, no application; H2O2: positive control, 100 mu M H2O2 applied; 1xH2O2: 100 mu M H2O2 applied one year ago; 2xH2O2: 100 mu M H2O2 applied second time after one year) wheat seedlings. Biochemical results showed that the lowest H2O2 level was in NKU Lider variety and in-H2O2 and 1xH2O2 groups. The lowest thiobarbituric acid reactive substances (TBARS) level was in Tosunbey variety and 2xH2O2 group. The highest superoxide dismutase (SOD) activity was in NKU Lider variety, all H2O2 pre-treatment caused an increase in SOD activity and 2xH2O2 pre-treatment caused the highest SOD activity. However, H2O2 and TBARS levels increased in all application groups except 2xH2O2 group, while the H2O2 amount increased and TBARS level decreased in 2xH2O2 group. MnSOD was not detected in any groups. CuZnSOD increased in all groups except 2xH2O2 groups under salt stress in Sultan-95 variety compared to FeSOD. H2O2 pre-treatment better tolerated salt stress, and second-applied H2O2 pre-treatment eliminated the stress and improved plant growth. In conclusion, it was determined that H2O2 re-pre-treatment to wheat seeds resulted in improvement of plant growth in tolerant varieties exposed to salt stress.

Wheat is the major staple food in Western Europe and an important source of energy, protein, dietary fibre, minerals, B vitamins and phytochemicals. Plant breeders have been immensely successful in increasing yields to feed the growing global population. However, concerns have been expressed that the focus on increasing yield and processing quality has resulted in reduced contents of components that contribute to human health and increases in adverse reactions. We review the evidence for this, based largely on studies in our own laboratories of sets of wheats bred and grown between the 18(th)century and modern times. With the exception of decreased contents of mineral micronutrients, there is no clear evidence that intensive breeding has resulted in decreases in beneficial components or increases in proteins which trigger adverse responses. In fact, a recent study of historic and modern wheats from the UK showed increases in the contents of dietary fibre components and a decreased content of asparagine in white flour, indicating increased benefits for health.

Plant regeneration is essential for survival upon wounding and is, hence, considered to be a strong natural selective trait. The capacity of plant tissues to regenerate in vitro, however, varies substantially between and within species and depends on the applied incubation conditions. Insight into the genetic factors underlying this variation may help to improve numerous biotechnological applications that exploit in vitro regeneration. Here, we review the state of the art on the molecular framework of de novo shoot organogenesis from root explants in Arabidopsis, which is a complex process controlled by multiple quantitative trait loci of various effect sizes. Two types of factors are distinguished that contribute to natural regenerative variation: master regulators that are conserved in all experimental systems (e.g., WUSCHEL and related homeobox genes) and conditional regulators whose relative role depends on the explant and the incubation settings. We further elaborate on epigenetic variation and protocol variables that likely contribute to differential explant responsivity within species and conclude that in vitro shoot organogenesis occurs at the intersection between (epi) genetics, endogenous hormone levels, and environmental influences.

We report a large-scale plant phosphoproteome under the control of higher ambient temperature, and expose early signalling events associated with a mild temperature increase in wheat., Wheat (Triticum ssp.) is one of the most important human food sources. However, this crop is very sensitive to temperature changes. Specifically, processes during wheat leaf, flower, and seed development and photosynthesis, which all contribute to the yield of this crop, are affected by high temperature. While this has to some extent been investigated on physiological, developmental, and molecular levels, very little is known about early signalling events associated with an increase in temperature. Phosphorylation-mediated signalling mechanisms, which are quick and dynamic, are associated with plant growth and development, also under abiotic stress conditions. Therefore, we probed the impact of a short-term and mild increase in temperature on the wheat leaf and spikelet phosphoproteome. In total, 3822 (containing 5178 phosphosites) and 5581 phosphopeptides (containing 7023 phosphosites) were identified in leaf and spikelet samples, respectively. Following statistical analysis, the resulting data set provides the scientific community with a first large-scale plant phosphoproteome under the control of higher ambient temperature. This community resource on the high temperature-mediated wheat phosphoproteome will be valuable for future studies. Our analyses also revealed a core set of common proteins between leaf and spikelet, suggesting some level of conserved regulatory mechanisms. Furthermore, we observed temperature-regulated interconversion of phosphoforms, which probably impacts protein activity.

Proper carbon (C) supply is essential for nitrogen (N) assimilation especially when plants are grown under ammonium (NH4+) nutrition. However, how C and N metabolic fluxes adapt to achieve so remains uncertain. In this work, roots of wheat (Triticum aestivum L.) plants grown under exclusive NH4+ or nitrate (NO3−) supply were incubated with isotope-labelled substrates (15NH4+, 15NO3−, or [13C]Pyruvate) to follow the incorporation of 15N or 13C into amino acids and organic acids. Roots of plants adapted to ammonium nutrition presented higher capacity to incorporate both 15NH4+ and 15NO3− into amino acids, thanks to the previous induction of the NH4+ assimilative machinery. The 15N label was firstly incorporated into [15N]Gln vía glutamine synthetase; ultimately leading to [15N]Asn accumulation as an optimal NH4+ storage. The provision of [13C]Pyruvate led to [13C]Citrate and [13C]Malate accumulation and to rapid [13C]2-OG consumption for amino acid synthesis and highlighted the importance of the anaplerotic routes associated to tricarboxylic acid (TCA) cycle. Taken together, our results indicate that root adaptation to ammonium nutrition allowed efficient assimilation of N thanks to the promotion of TCA cycle open flux modes in order to sustain C skeleton availability for effective NH4+ detoxification into amino acids.

Responses of wheat (Triticum aestivum) to complete submergence are not well understood as research has focused on waterlogging (soil flooding). The aim of this study was to characterize the responses of 2 wheat cultivars differing vastly in submergence tolerance to test if submergence tolerance was linked to shoot carbohydrate consumption as seen in rice. Eighteen-day-old wheat cultivars Frument (intolerant) and Jackson (tolerant) grown in soil were completely submerged for up to 19 days while assessing responses in physiology, gene expression, and shoot metabolome. Results revealed 50% mortality after 9.3 and 15.9 days of submergence in intolerant Frument and tolerant Jackson, respectively, and significantly higher growth in Jackson during recovery. Frument displayed faster leaf degradation as evident from leaf tissue porosity, chlorophyll a, and metabolomic fingerprinting. Surprisingly, shoot soluble carbohydrates, starch, and individual sugars declined to similarly low levels in both cultivars by day 5, showing that cultivar Jackson tolerated longer periods of low shoot carbohydrate levels than Frument. Moreover, intolerant Frument showed higher levels of phytol and the lipid peroxidation marker malondialdehyde relative to tolerant Jackson. Consequently, we propose to further investigate the role of ethylene sensitivity and deprivation of reactive O 2 species in submerged wheat.

We investigated the impact of the particle size of wheat bran on gut dysbiosis and inflammation induced by a fructose overload. Mice received drinking water with or without fructose (30%) and a standard diet supplemented with or without 5% of wheat bran fractions characterized by different average particle sizes (1690 μm versus 150 μm) for 8 weeks. Fructose increased Enterobacteriaceae associated with higher expression of key inflammatory genes in the liver. The two wheat bran fractions differently affected specific gut bacteria known to be involved in the regulation of the gut barrier function and/or inflammatory processes. Moreover, wheat bran with small particle size was the sole fibre that reduced hepatic and systemic inflammatory markers upon high fructose intake. The framework of root system architecture is shaped by the three-dimensional (3D) trajectory of the axile roots. However, methods for characterising root trajectory in the field are lacking. Our aim was to develop a new method for quantifying the spatial distribution of axile roots of maturing plants. The trajectory of axile roots of two maize cultivars was measured using a 3D digitiser in situ in the field, in parallel with diameter measurements along the same roots based on scanned images. The measured diameters were then mapped onto the digitised trajectory. We show that inter-plant distances did not influence root trajectory in either cultivar. The two cultivars had different initial angles from the vertical and presented slightly different patterns of root angle distribution. The reorientation of an axile root downwards was related to its angle and diameter, which can serve as a valuable resource for the parameterisation of root architecture models. The method is suitable for accurate determination of the 3D architecture of axile roots of mature maize plants under field conditions. The value of acquiring a relative complete root trajectory is highlighted.

High-density single nucleotide polymorphism (SNP) genotyping arrays are a powerful tool for studying genomic patterns of diversity, inferring ancestral relationships between individuals in populations and studying marker-trait associations in mapping experiments. We developed a genotyping array including about 90 000 gene-associated SNPs and used it to characterize genetic variation in allohexaploid and allotetraploid wheat populations. The array includes a significant fraction of common genome-wide distributed SNPs that are represented in populations of diverse geographical origin. We used density-based spatial clustering algorithms to enable high-throughput genotype calling in complex data sets obtained for polyploid wheat. We show that these model-free clustering algorithms provide accurate genotype calling in the presence of multiple clusters including clusters with low signal intensity resulting from significant sequence divergence at the target SNP site or gene deletions. Assays that detect low-intensity clusters can provide insight into the distribution of presence-absence variation (PAV) in wheat populations. A total of 46 977 SNPs from the wheat 90K array were genetically mapped using a combination of eight mapping populations. The developed array and cluster identification algorithms provide an opportunity to infer detailed haplotype structure in polyploid wheat and will serve as an invaluable resource for diversity studies and investigating the genetic basis of trait variation in wheat. © 2014 The Authors Plant Biotechnology Journal published by Society for Experimental Biology and The Association of Applied Biologists and John Wiley & Sons Ltd.

Pollutants can have detrimental effects on living organisms. They can cause toxicity, damaging cells, tissues and organs because of their high concentrations or activities. Plants provide a useful system for screening and monitoring environmental pollutants. Among pollutants, aluminum is considered as a primary growth limiting factor for plants resulting in decreased plant growth and development. Although considered to be a non-essential and highly toxic metal ion for growth and development, aluminum (Al) is easily absorbed by plants. Urticaceae family members have high nutrient requirements demonstrated by leaves containing high levels of calcium (Ca), iron (Fe), magnesium (Mg), and nitrogen (N). Urtica pilulifera is one of the important traditional medicinal plants in Turkey. In this study, U. pilulifera was used as a bioindicator to investigate the possible differences in the absorption and accumulation of mineral nutrients at different levels of the Al exposure and examine the mineral nutrition composition of U. pilulifera under Al stress. Also, some growth parameters (leaf-stem fresh and dry weights, root dry weights, stem lengths and leaf surface area) were investigated. U. pilulifera seedlings were grown for two months in growth-room conditions and watered with spiked Hoagland solution, which contained 0, 100, and 200 μM aluminium chloride (AlCl3). It was observed that macro- and micro-nutritional status of roots and leaves was altered by Al exposure. The concentrations of some macro- and micronutrients were reduced while concentrations of others were increased by excess of Al. Some macro- and micronutrients were increased at low level of Al whereas reductions were observed at high level of Al, and vice versa. The patterns were dependent on the macro- or micronutrient and the plant part.

Wheat is a cereal grain and one of the world's major food crops. Recent advances in wheat genome sequencing are by now facilitating its genomic and proteomic analyses. However, little is known about possible differences in total protein levels of hexaploid versus tetraploid wheat cultivars, and also knowledge of phosphorylated wheat proteins is still limited. Here, we performed a detailed analysis of the proteome of seedling leaves from two hexaploid wheat cultivars (Triticum aestivum L. Pavon 76 and USU-Apogee) and one tetraploid wheat (T. turgidum ssp. durum cv. Senatore Cappelli). Our shotgun proteomics data revealed that, whereas we observed some significant differences, overall a high similarity between hexaploid and tetraploid varieties with respect to protein abundance was observed. In addition, already at the seedling stage, a small set of proteins was differential between the small (USU-Apogee) and larger hexaploid wheat cultivars (Pavon 76), which could potentially act as growth predictors. Finally, the phosphosites identified in this study can be retrieved from the in-house developed plant PTM-Viewer (bioinformatics.psb.ugent.be/webtools/ptm_viewer/), making this the first searchable repository for phosphorylated wheat proteins. This paves the way for further in depth, quantitative (phospho) proteome-wide differential analyses upon a specific trigger or environmental change.

The introduction of Reduced height (Rht)-B1b and Rht-D1b into bread wheat (Triticum aestivum) varieties was a key component of the 'green revolution' and today these alleles are the primary sources of semi-dwarfism in wheat. The Rht-1 loci encode DELLA proteins, which are transcription factors that affect plant growth and stress tolerance. In bread wheat, Rht-D1b and Rht-B1b influence resistance to the disease Fusarium Head Blight. To identify Rht-1 variants, locus specific primers were developed and used to sequence the entire open reading frame (ORF) and 1.7 kb of the 5' and 0.5 kb of the 3' flanking regions of Rht-A1 (Rht-A1+f), Rht-B1 (Rht-B1+f), and Rht-D1 (Rht-D1+f) in bread wheat (36 sequences from each genome) and tetraploid and diploid wheat (TDW) (one to three sequences from each genome). Among the bread wheat accessions, the Rht-A1+f and Rht-D1+f sequences contained relatively low genetic diversity and few haplotypes relative to the Rht-B1+f sequences. The TDW accessions were relatively rich in genetic diversity and contained the majority of the polymorphic sites. Novel polymorphisms, relative to 'Chinese Spring', discovered among the accessions include 160 and 197 bp insertions 5' of Rht-B1 and a frameshift in the Rht-B1 ORF. Quantitative real-time PCR using shoot and leaf tissue from 5-day-old seedlings of genotypes lacking or containing the 5' insertions revealed no major effect on Rht-B1 transcript accumulation. This research provides insights into the genetic diversity present at the Rht-1 loci in modern bread wheat and in relation to ancestral wheat accessions.

Consumers' interest in organic foods is increasing and so is the need for robust analytical tools for their authentication. This review focuses on the most promising biomarkers/analytical approaches that are available for the authentication of organic produce. Food products have been subdivided into two groups: foods of plant origin (crops) and foods of animal origin (meat, milk and dairy products, eggs and fish). For each food category the most suitable biomarkers are presented and their potential for authentication is discussed. In the light of current knowledge, it is unlikely that the authentication of organic food products can be attained by the measurement of a single marker. Analytical approaches based on the measurement of multiple markers and/or complex chemical or physical profiles/fingerprints supported by multivariate statistical analysis seem considerably more promising in this respect. For the development of robust classification models, well-designed experimental studies must be performed that rely on data sets that are both well balanced and of sufficient size to ensure that all relevant sources of variation for the target biomarkers are included in the reference database. © 2012 Society of Chemical Industry.

Improved mapping, multi-environment quantitative trait loci (QTL) analysis and dissection of allelic effects were used to define a QTL associated with grain yield, thousand grain weight and early vigour on chromosome 3BL of bread wheat (Triticum aestivum L.) under abiotic stresses. The QTL had pleiotropic effects and showed QTL x environment interactions across 21 diverse environments in Australia and Mexico. The occurrence and the severity of water deficit combined with high temperatures during the growing season affected the responsiveness of this QTL, resulting in a reversal in the direction of allelic effects. The influence of this QTL can be substantial, with the allele from one parent (RAC875) increasing grain yield by up to 12.5 % (particularly in environments where both heat and drought stress occurred) and the allele from the other parent (Kukri) increasing grain yield by up to 9 % in favourable environments. With the application of additional markers and the genotyping of additional recombinant inbred lines, the genetic map in the QTL region was refined to provide a basis for future positional cloning.

As the first organ in plants to sense water-deficit in the soil, roots have important roles for improving crop adaption to water limited environments. Stem water soluble carbohydrates (WSC) are a major carbon source for grain filling under drought conditions. The contributions of root WSC during grain filling under drought has not been revealed. Wheat parental lines of Westonia, Kauz and their derived four double haploid (DH) lines, namely, DH 125, DH 139, DH 307, and DH 338 were used in a field drought experiment with four replications. Through measurements of the root and stem WSC components, and the associated enzyme activities during grain filling, we identified that the levels of root WSC and fructan were one third of the levels in stems. In particular, root glucose and 6-kestose levels were one third of the stem, while the root fructose and bifurcose level were almost half of the stem and sucrose level was two third of the stem. The accumulation and the degradation patterns of root fructan levels were similar to that in the stem, especially under drought. Correlations between root fructan levels and grain assimilation were highly significant, indicating that under terminal drought, root WSC represents a redistributed carbon source for grain filling rather than deep rooting. The significantly higher root sucrose levels under drought suggest that sucrose may act as a signal under drought stress. As compared with stem fructose levels, the earlier increased root fructose levels in DH 307, DH 139, and DH 338 provided agile response to drought stress. Our root results further confirmed that β-(2-6) linkages predominate in wheat with patterns of 6-kestose being closely correlated with overall fructan patterns. Background In wheat, the vernalization requirement is mainly controlled by the VRN genes. Different species of hexaploid and tetraploid wheat are widely used as genetic source for new mutant variants and alleles for fundamental investigations and practical breeding programs. In this study, VRN-A1 and VRN-B1 were analysed for 178 accessions representing six tetraploid wheat species (Triticum dicoccoides, T. dicoccum, T. turgidum, T. polonicum, T. carthlicum, T. durum) and five hexaploid species (T. compactum, T. sphaerococcum, T. spelta, T. macha, T. vavilovii). Results Novel allelic variants in the promoter region of VRN-A1 and VRN-B1 were identified based on the change in curvature and flexibility of the DNA molecules. The new variants of VRN-A1 (designated as Vrn-A1a.2, Vrn-A1b.2 – Vrn-A1b.6 and Vrn-A1i) were found to be widely distributed in hexaploid and tetraploid wheat, and in fact were predominant over the known VRN-A1 alleles. The greatest diversity of the new variants of VRN-B1 (designated as VRN-B1.f, VRN-B1.s and VRN-B1.m) was found in the tetraploid and some hexaploid wheat species. For the first time, minor differences within the sequence motif known as the VRN-box of VRN1 were correlated with wheat growth habit. Thus, vrn-A1b.3 and vrn-A1b.4 were revealed in winter wheat in contrast to Vrn-A1b.2, Vrn-A1b.5, Vrn-A1b.6 and Vrn-A1i. It was found that single nucleotide mutation in the VRN-box can influence the vernalization requirement and growth habit of wheat. Our data suggest that both the A-tract and C-rich segment within the VRN-box contribute to its functionality, and provide a new view of the hypothesised role of the VRN-box in regulating transcription of the VRN1 genes. Specifically, it is proposed that combination of mutations in this region can modulate vernalization sensitivity and flowering time of wheat. Conclusions New allelic variants of the VRN-A1 and VRN-B1 genes were identified in hexaploid and tetraploid wheat. Mutations in A-tract and C-rich segments within the VRN-box of VRN-A1 are associated with modulation of the vernalization requirement and flowering time. New allelic variants will be useful in fundamental investigations into the regulation of VRN1 expression, and provide a valuable genetic resource for practical breeding of wheat. International audience; There is an amazing diversity of cell wall composition and cell wall polysaccharide structure in wheat grain that is only partly explained by the complexity of plant tissues and cell type functions present in this organ. This review presents the state of the art on the structure and diversity of cell wall polymers in mature wheat grain and recent knowledge on cell wall metabolism with a focus on development-associated changes in wheat grain cell wall polysaccharides and genes controlling their biosynthesis. To pursue genetic improvement of lily, efficiency of both regeneration and transformation from callus cultures induced from different explants were evaluated in multiple cultivars. Thirty-five callus lines induced from filaments or styles and one control callus line derived from bulb scales of in total twenty lily cultivars representing Lilium longiflorum, Oriental × Trumpet and Longiflorum × Asiatic hybrids were maintained on a medium with 8.3 μM picloram (PIC). In this study, they were tested for their regeneration potential by transferring them onto a regeneration medium supplemented with 0.4 μM PIC and 0.044 μM 6-benzyladenine. Regeneration was obtained in all cultivars examined and the percentage varied from zero to 89 % in the 36 callus lines. Regeneration frequency was significantly influenced by the genotype (cultivar). Subculturing the calli every 4 weeks by refreshing the regeneration medium contributed positively to bulblet formation, when compared to an eight week subculture frequency. It was found that the regeneration ability generally decreased with an increasing age of the callus cultures for all cultivars. The origin of the callus (style or filament) did not lead to significant differences in regeneration frequency, but there was an interaction between callus origin and genotype. Calli of eight randomly chosen cultivars were co-cultivated with Agrobacterium tumefaciens strain AGL0 carrying binary vectors with the gus gene as reporter and putative transgenic plants were produced. GUS histochemical assays demonstrated transient and stable expression of the gus gene in both calli and regenerated lily plants. Transient expression frequencies ranged from 0.3 to 20.6 % while stable transformation was much lower, only 1.4 % as the maximum.

It is estimated that more than 95% of organic production is based on crop varieties that were bred for the conventional high-input sector. Recent studies have shown that such varieties lack important traits required under organic and low-input production conditions. This is primarily due to selection in conventional breeding programmes being carried out in the background of high inorganic fertilizer and crop protection inputs. Also, some of the traits (e.g., semi-dwarf genes) that were introduced to address problems like lodging in cereals in high-input systems were shown to have negative side-effects (reduced resistance to diseases such as Septoria, lower protein content and poorer nutrient-use efficiency) on the performance of varieties under organic and low-input agronomic conditions. This review paper, using wheat, tomato and broccoli as examples, describes (1) the main traits required under low-input conditions, (2) current breeding programmes for organic, low-input agriculture, (3) currently available breeding and/or selection approaches, and (4) the benefits and potential negative side-effects of different breeding methodologies and their relative acceptability under organic farming principles. © 2010 Royal Netherlands Society for Agricultural Sciences. Wheat quality is an important target trait. Previous studies mainly focus on storage protein, but their contribution to quality is partial, and most loci for quality are still undetected. Wild species of wheat are valuable resources for wheat improvement and introgression lines (ILs) are the ideal materials for detecting quantitative trait loci (QTL). In this study, a set of 82 BC5 F2-6 ILs, carrying a range of introgressed chromosome segments from a synthetic hexaploid wheat Am3 (Triticum carthlicum x Aegilops tauschii), was developed and genotyped with 170 microsatellite markers. QTL analysis was performed for 14 parameters, sodium dodecyl sulfate sedimentation volume, grain protein content (GPC), grain hardness and 11 mixograph parameters, associated with end-use quality of wheat, using the materials harvested in three environments. This led to the detection of 116 QTL, with c. 95% of the positive alleles contributed by Am3. Six important and novel genomic regions for bread-making quality were found on chromosomes 2D, 3A, 4A, 4B, 5A and 6A. These loci for bread-making quality showed pleiotropy and had large positive effects on several quality parameters with no or very weak negative effect on grain yield, thus demonstrating the value of synthetic wheat as a source of useful genetic variation for the improvement of bread wheat quality.

Celiac disease is a T-cell mediated immune response in the small intestine of genetically susceptible individuals caused by ingested gluten proteins from wheat, rye, and barley. In the allohexaploid bread wheat (Triticum aestivum), gluten proteins are encoded by multigene loci present on the homoeologous chromosomes 1 and 6 of the three homoeologous genomes A, B, and D. The effect of deleting individual gluten loci was analyzed in a set of deletion lines of T. aestivum cv. Chinese Spring with regard to the level of T-cell stimulatory epitopes (Glia-α9 and Glia-α20) and to technological properties of the dough including mixing, stress relaxation, and extensibility. Deletion of loci encoding ω-gliadins, γ-gliadins, and LMW-glutenins located on the short arm of chromosome 1D, reduced the number of T-cell stimulatory epitopes and caused minor deterioration of dough quality by increase of elasticity. Deletion of loci encoding α-gliadins located on the short arm of chromosome 6D, resulted in a significant decrease in T-cell stimulatory epitopes. In parallel, the dough became more stiff and less elastic, which is an improvement for ‘Chinese Spring’ dough. We demonstrated that α-gliadins from wheat can largely be compensated by addition of avenins from oat to the flour to meet technological requirements.

In an attempt to shed light on the role of root systems in differential responses of wheat genotypes to long-term water limitation, transcriptional differences between two wheat genotypes (Triticum aestivum L., cv. Plainsman V and landrace Kobomugi) were identified during adaptation to moderate water stress at the tillering stage. Differences in organ sizes, water-use efficiency and seed production were detected in plants grown in soil, and root functions were characterised by expression profiling. The molecular genetic background of the behaviour of the two genotypes during this stress was revealed using a cDNA macroarray for transcript profiling of the roots. During a 4-week period of moderate water deficit, a set of up-regulated genes displaying transiently increased expression was identified in young plantlets, mostly in the second week in the roots of Kobomugi, while transcript levels remained constantly high in roots of Plainsman V. These genes encode proteins with various functions, such as transport, protein metabolism, osmoprotectant biosynthesis, cell wall biogenesis and detoxification, and also regulatory proteins. Oxidoreductases, peroxidases and cell wall-related genes were induced significantly only in Plainsman V, while induction of stress- and defence-related genes was more pronounced in Kobomugi. Real-time qPCR analysis of selected members of the glutathione S-transferase gene family revealed differences in regulation of family members in the two genotypes and confirmed the macroarray results. The TaGSTZ gene was stress-activated only in the roots of Kobomugi.

The loss of variation in crops due to the modernization of agriculture has been described as genetic erosion. The current paper discusses the different views that exist on the concept of genetic erosion in crops. Genetic erosion of cultivated diversity is reflected in a modernization bottleneck in the diversity levels that occurred during the history of the crop. Two stages in this bottleneck are recognized: the initial replacement of landraces by modern cultivars; and further trends in diversity as a consequence of modern breeding practices. Genetic erosion may occur at three levels of integration: crop, variety and allele. The different approaches in the recent literature to measure genetic erosion in crops are reviewed. Genetic erosion as reflected in a reduction of allelic evenness and richness appears to be the most useful definition, but has to be viewed in conjunction with events at variety level. According to the reviewed literature, the most likely scenario of diversity trends during modernization is the following: a reduction in diversity due to the replacement of landraces by modern cultivars, but no further reduction after this replacement has been completed.

In recent years, an increasing number of papers has been published on the genetic diversity trends in crop cultivars released in the last century using a variety of molecular techniques. No clear general trends in diversity have emerged from these studies. Meta analytical techniques, using a study weight adapted for use with diversity indices, were applied to analyze these studies. In the meta analysis, 44 published papers were used, addressing diversity trends in released crop varieties in the twentieth century for eight different field crops, wheat being the most represented. The meta analysis demonstrated that overall in the long run no substantial reduction in the regional diversity of crop varieties released by plant breeders has taken place. A significant reduction of 6% in diversity in the 1960s as compared with the diversity in the 1950s was observed. Indications are that after the 1960s and 1970s breeders have been able to again increase the diversity in released varieties. Thus, a gradual narrowing of the genetic base of the varieties released by breeders could not be observed. Separate analyses for wheat and the group of other field crops and separate analyses on the basis of regions all showed similar trends in diversity.

International audience; Powdery mildew (Blumeria graminis f. sp. tritici) is one of the major diseases of wheat (Triticum aestivum). Adult plant resistance (APR) to powdery mildew is considered more durable than resistance conferred by major race-specific resistance genes. The objective of the present study was a better understanding of the genetic basis of APR in RE714 by means of QTL analysis of several resistance scores along the growing season. A population of 160 recombinant inbred lines obtained from the cross between RE714 and Hardi (susceptible) was assessed for APR under natural infection conditions during 3 years and a genetic map with whole genome coverage was developed with microsatellite and AFLP markers in this population. Two major QTL on chromosomes 5D and 6A were detected each year, and 6 minor QTL were detected only in 1 or 2 years. The QTL on chromosome 5D was detected during all the growing season each year and its R 2 value varied between 8.5 and 56.3%, whereas the QTL on chromosome 6A was detected at 1–4 scoring dates in the 3 years, and its R 2 value varied between 6.1 and 20.5%. The two QTL explained between 24.4 and 52.1% of the phenotypic variance for AUDPC, depending on the year. The models including QTL and cofactors in the composite interval mapping explained between 29 and 72% of the variance. The molecular markers linked to the two major QTL could be used in marker-assisted selection for adult plant resistance to powdery mildew.

International audience; Fusarium pathogens cause two major diseases in cereals, Fusarium crown rot (FCR) and head blight (FHB). A large-effect locus conferring resistance to FCR disease was previously located to chromosome arm 3BL (designated as Qcrs-3B) and several independent sets of near isogenic lines (NILs) have been developed for this locus. In this study, five sets of the NILs were used to examine transcriptional changes associated with the Qcrs-3B locus and to identify genes linked to the resistance locus as a step towards the isolation of the causative gene(s). Of the differentially expressed genes (DEGs) detected between the NILs, 12.7% was located on the single chromosome 3B. Of the expressed genes containing SNP (SNP-EGs) detected, 23.5% was mapped to this chromosome. Several of the DEGs and SNP-EGs are known to be involved in host-pathogen interactions, and a large number of the DEGs were among those detected for FHB in previous studies. Of the DEGs detected, 22 were mapped in the Qcrs-3B interval and they included eight which were detected in the resistant isolines only. The enrichment of DEG, and not necessarily those containing SNPs between the resistant and susceptible isolines, around the Qcrs-3B locus is suggestive of local regulation of this region by the resistance allele. Functions for 13 of these DEGs are known. Of the SNP-EGs, 28 were mapped in the Qcrs-3B interval and biological functions for 16 of them are known. These results provide insights into responses regulated by the 3BL locus and identify a tractable number of target genes for fine mapping and functional testing to identify the causative gene(s) at this QTL.

Organic anion exudation by roots as a mechanism of aluminium (Al) resistance has been intensively studied lately. In the present study, we evaluated qualitative and quantitative aspects of root exudation of organic anions in maize genotypes of distinct sensitivity to Al in response to Al exposure. Roots of maize seedlings were grown axenically in nutrient solution and root exudates were collected along the whole seminal root axis for a short period ( 4 h) using a divided-root-chamber technique. In root exudates collected from 10-mm long root apices, citrate accounted for 67% of the total organic anions found, followed by malate (29%), trans-aconitate (3%), fumarate (

Organic agriculture needs spring barley varieties that are adapted to organic growing conditions and have good and stable grain yield across years, even under less favourable growing conditions. The aim of this study was to compare how varieties differ in yield and yield stability under conventional and organic management conditions. The results help to decide under which growing conditions selection of genotypes for organic farming can be most effective. Grain yield and yield components of 10 varieties were estimated in field trials for three years at four sites: two conventionally and two organically managed sites. Varieties differed in stability: some varieties had high yield under conventional conditions and relatively high and stable yield under organic conditions. Heritabilities for yield and yield components were lower under organic (especially in the field with low weed control) than under conventional conditions. Heritabilities for yield components were lower than those for yield itself. Selection for yield components, therefore, may be less effective than selection directly for grain yield. Our data showed that generally the top performing cultivars under conventional conditions also performed as the best under organic conditions, but there were also exceptions. Therefore we conclude that selection of genotypes for organic farming may take place under conventional conditions, but that a final testing should be conducted under organic conditions to confirm the suitability of the selected varieties for cultivation on organic farms.

Quantitative trait locus (QTL) analysis of nitrogen use efficiency (NUE) of barley (Hordeum vulgare L.) was conducted on data generated from two pot experiments carried out in 2005 (using four nitrogen rates) and 2008 (with three rates) with AFLP markers and 94 recombinant inbred lines (RILs) of the Prisma × Apex mapping population. In total 41 QTLs were detected on 6 chromosomes and for 18 traits in both trials. About 95 % of the detected QTLs were with major additive effects. The percentage of variance accounted for by individual QTLs in the multiple QTL mapping model ranged from 8.4 to 54.4 % across all mapped traits in both years. Fifteen QTLs were related to NUE and its components; most of these QTLs were detected at lower nitrogen rates and none at the highest rate in both trials. These QTLs were found on Chromosomes 3(3H) and 7(5H) in 2005 and Chromosome 2(2H) in 2008. Except for the QTLs of plant height and NUE based on grain yield, none of the QTLs which were detected for a given trait in 2005, expressed themselves in 2008 irrespective of the nitrogen levels. QTLs controlling some traits were co-located in each year, and QTLs for many traits were detected on the same chromosome and close to the denso locus. Further research is needed to investigate the possibility to reduce nitrogen fertilizer requirements through breeding while maintaining high yield of barley.

Increasing crop yield and water use efficiency (WUE) in dryland farming requires a quantitative understanding of relationships between crop yield and the water balance over many years. Here, we report on a long-term dryland monitoring site at the Loess Plateau, Shanxi, China, where winter wheat was grown for 30 consecutive years and soil water content (0-200 cm) was measured every 10 days. The monitoring data were used to calibrate the AquaCrop model and then to analyse the components of the water balance. There was a strong positive relationship between total available water and mean cereal yield. However, only one-third of the available water was actually used by the winter wheat for crop transpiration. The remaining two-thirds were lost by soil evaporation, of which 40 and 60% was lost during the growing and fallow seasons, respectively. Wheat yields ranged from 0.6 to 3.9 ton/ha and WUE from 0.3 to 0.9 kg/m(3). Results of model experiments suggest that minimizing soil evaporation via straw mulch or plastic film covers could potentially double wheat yields and WUE. We conclude that the relatively low wheat yields and low WUE were mainly related to (i) limited rainfall, (ii) low soil water storage during fallow season due to large soil evaporation, and (iii) poor synchronisation of the wheat growing season to the rain season. The model experiments suggest significant potential for increased yields and WUE.

Consumers' interest in organic foods is increasing and so is the need for robust analytical tools for their authentication. This review focuses on the most promising biomarkers/analytical approaches that are available for the authentication of organic produce. Food products have been subdivided into two groups: foods of plant origin (crops) and foods of animal origin (meat, milk and dairy products, eggs and fish). For each food category the most suitable biomarkers are presented and their potential for authentication is discussed. In the light of current knowledge, it is unlikely that the authentication of organic food products can be attained by the measurement of a single marker. Analytical approaches based on the measurement of multiple markers and/or complex chemical or physical profiles/fingerprints supported by multivariate statistical analysis seem considerably more promising in this respect. For the development of robust classification models, well-designed experimental studies must be performed that rely on data sets that are both well balanced and of sufficient size to ensure that all relevant sources of variation for the target biomarkers are included in the reference database. © 2012 Society of Chemical Industry.

Since the dawn of wheat cytogenetics, chromosome 3B has been known to harbor a gene(s) that, when removed, causes chromosome desynapsis and gametic sterility. The lack of natural genetic diversity for this gene(s) has prevented any attempt to fine map and further characterize it. Here, gamma radiation treatment was used to create artificial diversity for this locus. A total of 696 radiation hybrid lines were genotyped with a custom mini array of 140 DArT markers, selected to evenly span the whole 3B chromosome. The resulting map spanned 2,852 centi Ray with a calculated resolution of 0.384 Mb. Phenotyping for the occurrence of meiotic desynapsis was conducted by measuring the level of gametic sterility as seeds produced per spikelet and pollen viability at booting. Composite interval mapping revealed a single QTL with LOD of 16.2 and r 2 of 25.6 % between markers wmc326 and wPt-8983 on the long arm of chromosome 3B. By independent analysis, the location of the QTL was confirmed to be within the deletion bin 3BL7-0.63-1.00 and to correspond to a single gene located ~1.4 Mb away from wPt-8983. The meiotic behavior of lines lacking this gene was characterized cytogenetically to reveal striking similarities with mutants for the dy locus, located on the syntenic chromosome 3 of maize. This represents the first example to date of employing radiation hybrids for QTL analysis. The success achieved by this approach provides an ideal starting point for the final cloning of this interesting gene involved in meiosis of cereals.

The potential for improving wheat grain quality by management strategies involving crop rotation, catch crops, and organic manure was tested in organic long-term experiments in Denmark and Austria. Growing grass clover in a four-year rotation resulted in a higher wheat yield increase that could not be achieved by including leguminous catch crops in the rotation. Yield was also higher with a pre-crop of pea than of lucerne. The average protein concentration was 132 g kg−1 for grains from the Austrian experiments while the Danish grains held 85 g kg−1. Protein was generally much less affected by the experimental conditions than grain yield. None of the tested management parameters affected grain protein concentrations in the Danish experiment. In the Austrian trial, a significant pre-crop × treatment interaction reflected a positive effect of the animal manure treatment on protein and dry gluten in wheat following pre-crop pea. Danish grains generally contained more soluble polymers of less interest for the baking process than the Austrian ones. The study emphasizes the challenges in improving the quality of organically grown wheat beyond what is predetermined by environmental growth conditions and cultivar. However, baking quality appeared better than could be expected from the quality parameters determined. KEY MESSAGE : Wheat low-molecular-weight-glutenin and α-gliadin were accumulated in the endoplasmic reticulum and formed protein body-like structures in tobacco cells, with the participation of BiP chaperone. Possible interactions between these prolamins were investigated. Wheat prolamins are the major proteins that accumulate in endosperm cells and are largely responsible for the unique biochemical properties of wheat products. They are accumulated in the endoplasmic reticulum (ER) where they form protein bodies (PBs) and are then transported to the storage vacuole where they form a protein matrix in the ripe seeds. Whereas previous studies have been carried out to determine the atypical trafficking pathway of prolamins, the mechanisms leading to ER retention and PB formation are still not clear. In this study, we examined the trafficking of a low-molecular-weight glutenin subunit (LMW-glutenin) and α-gliadin fused to fluorescent proteins expressed in tobacco cells. Through transient transformation in epidermal tobacco leaves, we demonstrated that both LMW-glutenin and α-gliadin were retained in the ER and formed mobile protein body-like structures (PBLS) that generally do not co-localise with Golgi bodies. An increased expression level of BiP in tobacco cells transformed with α-gliadin or LMW-glutenin was observed, suggesting the participation of this chaperone protein in the accumulation of wheat prolamins in tobacco cells. When stably expressed in BY-2 cells, LMW-glutenin fusion was retained longer in the ER before being exported to and degraded in the vacuole, compared with α-gliadin fusion, suggesting the involvement of intermolecular disulphide bonds in ER retention, but not in PBLS formation. Co-localisation experiments showed that gliadins and LMW-glutenin were found in the same PBLS with no particular distribution, which could be due to their ability to interact with each other as indicated by yeast two-hybrid assays.

Modern wheat (Triticum aestivum L.) varieties in Western Europe have mainly been bred, and selected in conditions where high levels of nitrogen-rich fertilizer are applied. However, high input crop management has greatly increased the risk of nitrates leaching into groundwater with negative impacts on the environment. To investigate wheat nitrogen tolerance characteristics that could be adapted to low input crop management, we supplied 196 accessions of a wheat core collection of old and modern cultivars with high or moderate amounts of nitrogen fertilizer in an experimental network consisting of three sites and 2 years. The main breeding traits were assessed including grain yield and grain protein content. The response to nitrogen level was estimated for grain yield and grain number per m(2) using both the difference and the ratio between performance at the two input levels and the slope of joint regression. A large variability was observed for all the traits studied and the response to nitrogen level. Whole genome association mapping was carried out using 899 molecular markers taking into account the five ancestral group structure of the collection. We identified 54 main regions involving almost all chromosomes that influence yield and its components, plant height, heading date and grain protein concentration. Twenty-three regions, including several genes, spread over 16 chromosomes were involved in the response to nitrogen level. These chromosomal regions may be good candidates to be used in breeding programs to improve the performance of wheat varieties at moderate nitrogen input levels.

International audience; Since the domestication of crop species, humans have derived specific varieties for particular uses and shaped the genetic diversity of these varieties. Here, using an interdisciplinary approach combining ethnobotany and population genetics, we document the within-variety genetic structure of a population-variety of bread wheat (Triticum aestivum L.) in relation to farmers’ practices to decipher their contribution to crop species evolution. Using 19 microsatellites markers, we conducted two complementary graph theory-based methods to analyze population structure and gene flow among 19 sub-populations of a single population-variety [Rouge de Bordeaux (RDB)]. The ethnobotany approach allowed us to determine the RDB history including diffusion and reproduction events. We found that the complex genetic structure among the RDB sub-populations is highly consistent with the structure of the seed diffusion and reproduction network drawn based on the ethnobotanical study. This structure highlighted the key role of the farmer-led seed diffusion through founder effects, selection and genetic drift because of human practices. An important result is that the genetic diversity conserved on farm is complementary to that found in the genebank indicating that both systems are required for a more efficient crop diversity conservation.

Dasypyrum villosum (L.) Candargy (sin. Haynaldia villosa) is an annual wild diploid grass species (2n = 2x = 14; genome VV) belonging to the Poaceae family, which is considered to be an important source of biotic and abiotic stress resistance genes for wheat breeding. Enhanced characterization of D. villosum chromosomes can facilitate exploitation of its gene pool and its use in wheat breeding programs. Here we present the cytogenetic identification of D. villosum chromosomes on slide by fluorescent in situ hybridization (FISH), with the GAA simple sequence repeat (SSR) as a probe. We also describe the isolation and the flow cytometric analysis of D. villosum chromosomes in suspension, resulting in a distinguished flow karyotype. Chromosomes were flow sorted into three fractions, according their DNA content, one of which was composed of a single type of chromosome, namely 6 V, sorted with over 85% purity. Chromosome 6 V is known to carry genes to code for important resistance and seed storage characteristics, and its isolation represents a new source of genetic traits and specific markers useful for wheat improvement.

Over the last three decades substantial attention has been given to the role of dietary fiber in health and disease, in particular diabetes, cardiovascular disease, intestinal health, and some types of cancer. As a result the food industry started to add back fiber to refined foods and develop fiber rich foods. Scientists suggested that whole grain foods are superior to foods enriched with fibers obtained/synthesized using enzyme treatment, and thermal or chemical processing because the content of bioactive components and micronutrients in whole grain is more abundant. This triggered interest in how to isolate the micronutrient rich aleurone fiber fraction from wheat. Aleurone is a single cell layer at the inner site of the bran. It contains most of the minerals, vitamins, phenolic antioxidants, and lignans of the wheat grain. Novel milling and dry-fractionation techniques have recently allowed for full-scale separation of aleurone cells from the other layers of wheat bran, yielding a fiber rich concentrate which potentially contains many of the "whole grain kernel bioactives," which recently have been used in a variety of studies. The present review highlights available data on aleurone isolation, composition, intestinal physiology, and its metabolism and potential health benefits as well as its use in food.

Resistance (R) genes protect plants very effectively from disease, but many of them are rapidly overcome when present in widely grown cultivars. To overcome this lack of durability, strategies that increase host resistance diversity have been proposed. Among them is the use of multilines composed of near-isogenic lines (NILs) containing different disease resistance genes. In contrast to classical R-gene introgression by recurrent backcrossing, a transgenic approach allows the development of lines with identical genetic background, differing only in a single R gene. We have used alleles of the resistance locus Pm3 in wheat, conferring race-specific resistance to wheat powdery mildew (Blumeria graminis f. sp. tritici), to develop transgenic wheat lines overexpressing Pm3a, Pm3c, Pm3d, Pm3f or Pm3g. In field experiments, all tested transgenic lines were significantly more resistant than their respective nontransformed sister lines. The resistance level of the transgenic Pm3 lines was determined mainly by the frequency of virulence to the particular Pm3 allele in the powdery mildew population, Pm3 expression levels and most likely also allele-specific properties. We created six two-way multilines by mixing seeds of the parental line Bobwhite and transgenic Pm3a, Pm3b and Pm3d lines. The Pm3 multilines were more resistant than their components when tested in the field. This demonstrates that the difference in a single R gene is sufficient to cause host-diversity effects and that multilines of transgenic Pm3 wheat lines represent a promising strategy for an effective and sustainable use of Pm3 alleles.

Various international initiatives have emerged to develop new sets of biodiversity indicators for wild biodiversity but little progress has been made for crop diversity. This study proposes a new methodology to assess the diversity of cultivated species. We developed a new integrative indicator H-T* that takes account of varietal richness, spatial evenness, between-variety genetic diversity, and within-variety genetic diversity. This index is compared to existing indexes using a comprehensive historical dataset from a French territory. The study reveals that more varieties (the varietal richness factor) can mean less diversity when (i) their genetic structure is more similar (the effect of between-variety genetic diversity), or (ii) when more diverse landraces are replaced by many homogeneous lines (the effect of within-variety genetic diversity) or (iii) when one or a few varieties become hegemonic in the landscape (the spatial evenness effect). Furthermore, an increased evenness in variety distribution (varietal evenness) can also mean less diversity when varieties are genetically related. This is why indicators which do not take into account the full range of factors (varietal richness, between-variety genetic diversity, spatial evenness and within-variety genetic diversity) may provide a misleading assessment of the state and trend of crop biodiversity. New integrative indicators such as HT. are critically needed. We discuss potential further development such as the use of this indicator for monitoring and the perspective of a broader indicator to assess genetic diversity at the multi-species scale and landscape level. International audience; In France, a stagnation of bread wheat yield has been observed since the middle of the 1990s, and the possibility of a decline in breeding progress has been questionned. To investigate this hypothesis, we have considered 2 independent long time series of yield data: one corresponding to the registration trials conducted in France from 1976 to 2010, and the other corresponding to trials from the INRA bread wheat breeding program along the 1970-2010 period. These raw data related to potential bread wheat yield in France, exhibited the same trend to stagnation as observed on national yield in the farms. However, after correction of the "year" effects, the corrected yields appeared to have increased regularly from the 1970s up to now, with quite a high rate in intensive conditions (from 0.065 t ha(-1) yr(-1) to 0.137 t ha(-1) yr(-1)). This linear trend for genetic progress was even higher without fungicide (from 0.097 t ha(-1) yr(-1) to 0.158 t ha(-1) yr(-1)), which could indicate that breeding efforts for productivity and for resistance to diseases tended to have additive results. The corrected yields also enabled us to point out a marked degradation of the agro-climatic potential of the different years. As our registration and breeding trials were as free as possible from agronomic limiting factors, it appeared that climatic factors constituted the main explanation to this degradation. Thus, our study demonstrated that since the end of the 1980s, genetic progress has been partly or totally counterbalanced by the adverse effects of climate change. (C) 2012 Elsevier B.V. In the past two decades, the wheat community has made remarkable progress in developing molecular resources for breeding. A wide variety of molecular tools has been established to accelerate genetic and physical mapping for facilitating the efficient identification of molecular markers linked to genes and QTL of agronomic interest. Already, wheat breeders are benefiting from a wide range of techniques to follow the introgression of the most favorable alleles in elite material and develop improved varieties. Breeders soon will be able to take advantage of new technological developments based on Next Generation Sequencing. In this paper, we review the molecular toolbox available to wheat scientists and breeders for perfomiing fundamental genomic studies and breeding. Special emphasis is given on the production and detection of single nucleotide polymorphisms (SNPs) that should enable a step change in saturating the wheat genome for more efficient genetic studies and for the development of new selection methods. The perspectives offered by the access to an ordered full genome sequence for further marker development and enhanced precision breeding is also discussed. Northwestern European consumers like their bread to be voluminous and easy to chew. These attributes require a raw material that is rich in protein with, among other characteristics, a suitable ratio between gliadins and glutenins. Achieving this is a challenge for organic growers, because they lack cultivars that can realise high protein concentrations under the relatively low and variable availability of nitrogen during the grain-filling phase common in organic farming. Relatively low protein content in wheat grains thus needs to be compensated by a high proportion of high-quality protein. Organic farming therefore needs cultivars with genes encoding for optimal levels of glutenins and gliadins, a maximum ability for nitrogen uptake, a large storage capacity of nitrogen in the biomass, an adequate balance between vegetative and reproductive growth, a high nitrogen translocation efficiency for the vegetative parts into the grains during grain filling and an efficient conversion of nitrogen into high-quality proteins. In this perspective paper the options to breed and grow such varieties are discussed.

Genome wide association (GWA) analysis of yield, yield components, developmental, physiological and anatomical traits was conducted for a barley germplasm collection consisting of 185 cultivated (Hordeum vulgare L.) and 38 wild (Hordeum spontaneum L.) genotypes, originating from 30 countries of four continents. Phenotypic evaluations were performed at a dry (Breda) and wet (Tel Hadya) location in Syria. Genome wide association study was done with 816 markers comprised of 710 diversity array technology (DArT), 61 single nucleotide polymorphism (SNP) and 45 microsatellite or simple sequence repeat (SSR) markers. Diversity analysis revealed 5 groups of germplasm, related to origin (Middle East, North Africa), structural information (two-rows), and domestication (wild versus domesticated). Linkage disequilibrium (LD) decayed after 3 cM with a few exceptions at 10 and 15 cM. Although a few QTLs were identified that differed between the dry and wet site, these QTLs explained low phenotypic variation and could not unequivocally be related to drought tolerance when compared to earlier linkage mapping based QTL analysis studies. Therefore, GWA analysis seems to be not very effective for identification of major QTLs for complex traits like drought tolerance in highly structured germplasm collections.

In this paper, the effect of root oxygen loss on energy recovery of the plant microbial fuel cell (PMFC) is described. In this manner, advanced understanding of competing processes within the rhizosphere-anode interface was provided. A microscopic model was developed on the basis of exudation, oxygen loss, biological oxidation, and biological current generation. The model was successfully validated by comparison to oxygen concentration profiles, volatile fatty acid profiles, and chemical oxygen demand profiles measured in the anode compartment. The model predicted oxic zones around roots in the anode of the plant microbial fuel cell. Results show no direct link between current generation and photosynthesis. This was consistent with the model which predicted that current was generated via hydrolysis of root-derived organic compounds. This result means that to optimize energy recovery of a PMFC, the plant selection should focus on high root biomass production combined with low oxygen loss.
