This work evaluated the effects of food frequencies and self-feeding on the behavior, performance, and blood biochemistry of Colossoma macropomum. Juveniles weighing 3.63 ± 0.05 g were distributed in 27 aquariums (40 L), with six fish per aquarium, to evaluate nine treatments in three replications in a 12L:12D photoperiod. The treatments were 24-h self-feeding (SF); daytime self-feeding at will (SFD); nighttime self-feeding at will (SFN); and feeding at fixed frequencies of two (2D), four (4D), and six times (6D) daytime and twice (2 N), four (4 N), and six times (6 N) nightly. Aims/hypothesis We hypothesised that the insulin-sensitising effect of physical activity depends on the timing of the activity. Here, we examined cross-sectional associations of breaks in sedentary time and timing of physical activity with liver fat content and insulin resistance in a Dutch cohort. Methods In 775 participants of the Netherlands Epidemiology of Obesity (NEO) study, we assessed sedentary time, breaks in sedentary time and different intensities of physical activity using activity sensors, and liver fat content by magnetic resonance spectroscopy (n=256). Participants were categorised as being most active in the morning (06:00–12:00 hours), afternoon (12:00–18:00 hours) or evening (18:00–00:00 hours) or as engaging in moderate-to-vigorous-physical activity (MVPA) evenly distributed throughout the day. Most active in a certain time block was defined as spending the majority (%) of total daily MVPA in that block. We examined associations between sedentary time, breaks and timing of MVPA with liver fat content and HOMA-IR using linear regression analyses, adjusted for demographic and lifestyle factors including total body fat. Associations of timing of MVPA were additionally adjusted for total MVPA. Results The participants (42% men) had a mean (SD) age of 56 (4) years and a mean (SD) BMI of 26.2 (4.1) kg/m2. Total sedentary time was not associated with liver fat content or insulin resistance, whereas the amount of breaks in sedentary time was associated with higher liver fat content. Total MVPA (−5%/h [95% CI −10%/h, 0%/h]) and timing of MVPA were associated with reduced insulin resistance but not with liver fat content. Compared with participants who had an even distribution of MVPA throughout the day, insulin resistance was similar (−3% [95% CI −25%, 16%]) in those most active in morning, whereas it was reduced in participants who were most active in the afternoon (−18% [95% CI −33%, −2%]) or evening (−25% [95% CI −49%, −4%]). Conclusions/interpretation The number of daily breaks in sedentary time was not associated with lower liver fat content or reduced insulin resistance. Moderate-to-vigorous activity in the afternoon or evening was associated with a reduction of up to 25% in insulin resistance. Further studies should assess whether timing of physical activity is also important for the occurrence of type 2 diabetes. Graphical abstract

PURPOSE OF REVIEW The low fat diet (LFD) is currently the first choice to treat nonalcoholic fatty liver disease (NAFLD) alongside with physical activity. However, low carbohydrate diets (LCDs) and ketogenic diets have gained attention lately, thanks to their favourable impact in reducing intrahepatic triglyceride content. We therefore aimed at providing an update on recent evidence evaluating the hepatoprotective effects of such dietary interventions. RECENT FINDINGS Novel findings confirmed previous evidence by showing beneficial effects on liver fat content reduction for both LFDs and LCDs. The further restriction of carbohydrates to less than 50 g/day, usually leading to ketosis, confirmed to produce an improvement in NAFLD, with very low-calorie ketogenic diets possibly proving particularly beneficial thanks to the significant weight loss that can be obtained. SUMMARY Most of the latest evidence shows that carbohydrate restriction plays a fundamental role in the modulation of lipid metabolism leading to similar efficacy in improving NAFLD compared with LFDs. The hepatoprotective role of carbohydrate restriction appears to be boosted when ketogenesis is induced, when the total calorie intake is extremely reduced, or, possibly, when dietary interventions have reduced content in free sugars, making such interventions valuable tools to deal with NAFLD.

In this study, the effect of dough salt content, frying temperature and time on the conjugated diene values, polymer triglyceride content, total polar material content, viscosity, and color values of the sunflower oil during frying of leavened doughs was determined using response surface methodology. Fifty repeated frying operations were applied in the same day at 160–200 °C for 1–5 min and doughs with 0–2% salt content. According to the results of the study, frying temperature, frying time and dough salt content were significantly (p 0.05). To minimize the oxidation products of frying oil, the frying process can be applied at 160 °C for 1 min using dough with 1.97% salt content.

A new monacolin derivative, namely (1S,2S,5′R,6R,8aR)-7-hydroxy-2,6-dimethyl-8-prenyl-1,2,6,8atetrahydronaphthalene-4′,5′-dihydro-2H-pyran-1′-one (1), and five known monacolin derivatives (2–6) were separated from the highland barley Monascus purpureus Went. for the first time. Their structures were elucidated on the basis of 1D and 2D NMR, HR-ESI-MS, and relevant references. Compounds 1–6 were tested for their hypolipidemic activities by measuring the triglyceride content in HepG2 cells. Among them, compounds 1 and 2 (10 μM) exhibited moderate hypolipidemic activity.

Introduction Electrical impedance myography (EIM) provides insight into muscle composition and structure. We sought to evaluate its use in a mouse obesity model characterized by myofiber atrophy. Methods We applied a prediction algorithm, i.e., the least absolute shrinkage and selection operator (LASSO), to surface, needle array, and ex vivo EIM data from db/db and WT mice and assessed myofiber cross-sectional area (CSA) histologically and triglyceride (TG) content biochemically. Results EIM data from all three modalities provided acceptable predictions of myofiber CSA with average root mean squared error (RMSE) of 15% in CSA (i.e., ± 209 μm2 for a mean CSA of 1439 μm2 ) and TG content with RMSE of 30% in TG content (i.e., ± 7.3 nmoles TG/mg muscle for a mean TG content of 25.4 nmoles TG/mg muscle). Discussion EIM combined with a predictive algorithm provides reasonable estimates of myofiber CSA and TG content without the need for biopsy.

A new lactone, named 9-allyl-5-methyl-1H-difuro-isochromene-1,8-dione (1), together with nine known lactone derivatives (2–10), was isolated from highland barley Monascus for the first time. Compounds 1–10 were studied by spectroscopic methods and comparison with references and evaluated for their hypolipidemic activities by measuring the triglyceride content in HepG2 cells with simvastatin as positive control. Among them, compounds 1 and 7 (10 μM) exhibited moderate hypolipidemic activities.

Caenorhabditis elegans serves as a model for understanding adiposity and its connections to aging. Current methodologies do not distinguish between fats serving the energy needs of the parent, akin to mammalian adiposity, from those that are distributed to the progeny, making it difficult to accurately interpret the physiological implications of fat content changes induced by external perturbations. Using spectroscopic coherent Raman imaging, we determine the protein content, chemical profiles and dynamics of lipid particles in live animals. We find fat particles in the adult intestine to be diverse, with most destined for the developing progeny. In contrast, the skin-like epidermis contains fats that are the least heterogeneous, the least dynamic and have high triglyceride content. These attributes are most consistent with stored somatic energy reservoirs. These results challenge the prevailing practice of assessing C. elegans adiposity by measurements that are dominated by the intestinal fat content.

Significance Ketogenic diet is an effective treatment for nonalcoholic fatty liver disease (NAFLD). Here, we present evidence that hepatic mitochondrial fluxes and redox state are markedly altered during ketogenic diet-induced reversal of NAFLD in humans. Ketogenic diet for 6 d markedly decreased liver fat content and hepatic insulin resistance. These changes were associated with increased net hydrolysis of liver triglycerides and decreased endogenous glucose production and serum insulin concentrations. Partitioning of fatty acids toward ketogenesis increased, which was associated with increased hepatic mitochondrial redox state and decreased hepatic citrate synthase flux. These data demonstrate heretofore undescribed adaptations underlying the reversal of NAFLD by ketogenic diet and highlight hepatic mitochondrial fluxes and redox state as potential treatment targets in NAFLD., Weight loss by ketogenic diet (KD) has gained popularity in management of nonalcoholic fatty liver disease (NAFLD). KD rapidly reverses NAFLD and insulin resistance despite increasing circulating nonesterified fatty acids (NEFA), the main substrate for synthesis of intrahepatic triglycerides (IHTG). To explore the underlying mechanism, we quantified hepatic mitochondrial fluxes and their regulators in humans by using positional isotopomer NMR tracer analysis. Ten overweight/obese subjects received stable isotope infusions of: [D7]glucose, [13C4]β-hydroxybutyrate and [3-13C]lactate before and after a 6-d KD. IHTG was determined by proton magnetic resonance spectroscopy (1H-MRS). The KD diet decreased IHTG by 31% in the face of a 3% decrease in body weight and decreased hepatic insulin resistance (−58%) despite an increase in NEFA concentrations (+35%). These changes were attributed to increased net hydrolysis of IHTG and partitioning of the resulting fatty acids toward ketogenesis (+232%) due to reductions in serum insulin concentrations (−53%) and hepatic citrate synthase flux (−38%), respectively. The former was attributed to decreased hepatic insulin resistance and the latter to increased hepatic mitochondrial redox state (+167%) and decreased plasma leptin (−45%) and triiodothyronine (−21%) concentrations. These data demonstrate heretofore undescribed adaptations underlying the reversal of NAFLD by KD: That is, markedly altered hepatic mitochondrial fluxes and redox state to promote ketogenesis rather than synthesis of IHTG.

This study investigated the effects of free-range days on growth performance, carcass traits, meat quality, lymphoid organ indices, and blood biochemical parameters of Wannan Yellow chickens. A total of 1,000 one-day-old male Wannan Yellow chickens were reared to 56 D. At 56 D of age, 960 birds with similar body weight (BW) were randomly allocated to free-range treatment at 56, 70, 84, and 98 D of age (assigned to free-range treatment for 42, 28, 14, and 0 D, respectively); 6 replicates with 40 chickens per treatment. In the free-range system, chickens were reared in indoor floor pens with an outdoor free-range paddock measuring 4 × 7 m (28 m2, 1.5 birds/m2). Results showed that BW of birds decreased significantly in the first 2 wk after birds were assigned to free-range treatment compared with those in the conventional treatment (P < 0.05). Average daily gain (ADG) and average daily feed intake (ADFI) decreased significantly (P < 0.05) for chickens assigned to free-range treatment from 56 to 70 D of age, while feed conversion ratio increased significantly (P < 0.05). Breast yield increased linearly with increasing free-range days (P < 0.05), whereas leg and foot yields decreased linearly (P < 0.05). Drip loss and L* value of thigh muscle decreased linearly with increasing free-range days (P < 0.05), while shear force improved linearly (P < 0.05). Additionally, the absolute thymus weight and thymus to BW ratio showed significant increasing and then decreasing quadratic responses to increasing free-range days (P < 0.05). Furthermore, serum glucose, total protein, cholesterol, and triglyceride content declined linearly, while high-density lipoprotein cholesterol (HDL-C) content increased linearly with increasing free-range days (P < 0.05). In conclusion, increasing free-range days had positive effects on breast yield, shear force, thymus weight, and HDL-C content, but negatively affected leg yield, foot yield, drip loss, L* value of thigh muscle, glucose, total protein, cholesterol, and triglyceride levels of Wannan Yellow chickens.

Nonalcoholic fatty liver disease (NAFLD) is a progressive condition that includes steatosis (NAFL) and nonalcoholic steatohepatitis (NASH). In the U.S., Hispanics (HIS) are afflicted with NAFLD at a higher rate and severity compared to other ethnicities. To date, the mechanisms underlying this disparity have not been elucidated. In this pilot study, we compared untargeted plasma metabolomic profiles for primary metabolism, complex lipids, choline and related compounds between a group of HIS (n =7) and White Caucasian (CAU, n =8) subjects with obesity and biopsy-characterized NAFL to ethnicity-matched lean healthy controls (n =14 HIS and 8 CAU). We also compared liver and plasma metabolomic profiles in a group of HIS and CAU subjects with obesity and NASH of comparable NAFLD Activity Scores, to BMI-matched NASH-free subjects in both ethnicities. Results highlight signs of metabolic dysregulation observed in HIS, independent of obesity, including higher plasma triglycerides, acylcarnitines, and free fatty acids. With NASH progression, there were ethnicity-related differences in the hepatic profile, including higher free fatty acids and lysophospholipids seen in HIS, suggesting lipotoxicity is involved in the progression of NASH. We also observed greater hepatic triglyceride content, higher plasma triglyceride concentrations and lower hepatic phospholipids with signs of impaired hepatic mitochondrial β-oxidation. These findings provide preliminary evidence indicating ethnicity-related variations that could potentially modulate the risk for progression of NALD to NASH. Highlights Hispanic subjects show signs of metabolic dysregulations independent of obesity. In nonalcoholic fatty liver, plasma metabolomic profiles are differentially altered between ethnicities. In nonalcoholic steatohepatitis, the hepatic lipidomic profiles are differentially altered between ethnicities. In Hispanics, nonalcoholic steatohepatitis shows higher triglycerides and lipotoxic lipids with signs of impaired hepatic mitochondrial β-oxidation. Our results indicate evidence of ethnicity-related variations in the pathogenesis of nonalcoholic fatty liver disease. Graphical Abstract

Nicotinamide mononucleotide increases muscle insulin sensitivity in prediabetic women In evaluating any randomized clinical trial, it is important to determine whether baseline differences between groups could have affected the primary outcome. In our study, muscle insulin sensitivity, which was identical in both groups at baseline, improved after nicotinamide mononucleotide (NMN), not placebo, therapy. Differences in baseline intrahepatic triglyceride content between groups do not negate the effects of NMN observed in muscle.

H-1-MR spectroscopy of skeletal muscle provides insight into metabolism that is not available noninvasively by other methods. The recommendations given in this article are intended to guide those who have basic experience in general MRS to the special application of H-1-MRS in skeletal muscle. The highly organized structure of skeletal muscle leads to effects that change spectral features far beyond simple peak heights, depending on the type and orientation of the muscle. Specific recommendations are given for the acquisition of three particular metabolites (intramyocellular lipids, carnosine and acetylcarnitine) and for preconditioning of experiments and instructions to study volunteers.

Three new phenanthraquinones (1–3), together with seven known phenanthraquinone derivatives (4–10) were isolated from Heleocharis dulcis for the first time. Their structures were established on extensive NMR, MS, UV, IR experiments in conjunction with their references. Compounds (1–10) were evaluated for their hypolipidemic activities for the first time. Compounds 1, 3, 7, and 9 (10 μM) exhibited significant hypolipidemic activities by measuring the triglyceride content in HepG2 cells with simvastatin as positive control. The chemotaxonomic significance of compounds (1–10) was also discussed.

Background Fatty liver is the leading cause of chronic liver diseases and increases the risk of cardiovascular disease. Besides alcohol consumption, energy-containing nonalcoholic beverages may contribute to liver fat accumulation. Objective We aimed to study the consumption of alcoholic and nonalcoholic beverages and their mutual replacement in relation to hepatic triglyceride content (HTGC) in middle-aged men and women. Methods In this cross-sectional analysis, HTGC was assessed by proton magnetic resonance spectroscopy. Habitual consumption of alcoholic and nonalcoholic beverages was assessed using a validated food-frequency questionnaire. All beverages were converted to standard servings and to percentage of total energy intake (En%). We performed linear regression to examine the association of alcoholic and nonalcoholic beverages with HTGC, adjusted for age, sex, smoking, education, ethnicity, physical activity, total energy intake, and total body fat. We studied replacement of alcoholic beverages with nonalcoholic beverages per 1 serving/d and per 5 En%/d. Results After exclusion of individuals with missing values, 1966 participants (47% men) were analyzed, with a mean ± SD age of 55 ± 6 y, BMI of 26 ± 4 kg/m2, and HTGC of 5.7% ± 7.9%. Each extra alcoholic serving per day was associated with more liver fat (1.09 times; 95% CI: 1.05, 1.12). Replacing 5 En% of alcoholic beverages with milk was associated with less liver fat (0.89 times; 95% CI: 0.81, 0.98), whereas replacement with 5 En% of sugar-sweetened beverages was associated with liver fat to an extent similar to alcoholic beverages (1.00 times; 95% CI: 0.91, 1.09). Conclusion In a population-based cohort, consumption of each extra daily alcoholic beverage was associated with more liver fat. In isocaloric replacement of alcoholic beverages, milk was associated with less liver fat, whereas sugar-sweetened beverages were equally associated with liver fat. This suggests that intake of alcohol and sugars may contribute to liver fat accumulation. This trial was registered at clinicaltrials.gov as NCT03410316.

AimsTo investigate the association between overweight/obesity and fatty liver index (FLI) on the odds of incident prediabetes/type 2 diabetes and non-alcoholic fatty liver disease (NAFLD) in 2020 participants after 10 years follow up.MethodsAt baseline (in 2001) 2020 participants, males and females, aged 24-39 years, were stratified according to body mass index (BMI), normal weight (2), overweight (≥25-2), or obese (≥30 kg/m2) and FLI (as high FLI ≥60 or low FLI Results514 and 52 individuals developed prediabetes and type 2 diabetes during follow-up. Such individuals were older, with higher BMI, serum glucose, insulin, alanine aminotransferase (ALT) and triglyceride (TG) concentrations than those who did not develop prediabetes or type 2 diabetes (n = 1454). The additional presence of high FLI significantly increased the risk of developing prediabetes and type 2 diabetes above the risk of being overweight/obese. Compared with normal weight, low FLI participants, the odds of prediabetes were ∼2-fold higher and the odds of type 2 diabetes were 9-10-fold higher respectively in the overweight/obese, high FLI group. No difference was observed between normal weight, low FLI and overweight/obese and low FLI groups.ConclusionsAn increased FLI significantly increases the odds of incident prediabetes, type 2 diabetes and NAFLD in individuals with overweight/obese highlighting the contributory role of liver fat accumulation in the pathophysiology of prediabetes/type 2 diabetes.Key messagesObesity is a risk factor for non-alcoholic fatty liver disease (NAFLD), prediabetes and type 2 diabetes.Additionally, NAFLD is more prevalent in people with prediabetes and type 2 diabetes when compared to age- and BMI-matched individuals.The presence of a raised fatty liver index (FLI) confers a significantly increased risk of developing prediabetes, type 2 diabetes and NAFLD above that conferred by being overweight/obese.The degree of elevation of FLI can risk stratify for incident prediabetes and type 2 diabetes in people with obesity.

The cardioprotective association of high-density lipoprotein cholesterol (HDL-C) may vary by menopause stage or estradiol level. We tested whether associations of comprehensive HDL metrics (HDL subclasses, phospholipid and triglyceride content, and HDL cholesterol efflux capacity [HDL-CEC]) with coronary artery calcium (CAC) score and density vary by menopause stage or estradiol level in women transitioning through menopause. Participants (N = 294; mean age [SD]: 51.3 [2.9]) had data on HDL metrics and CAC measures at one or two time points during the menopause transition. Generalized estimating equations were used for analyses. Effect modifications by menopause stage or estradiol level were tested in multivariable models. In adjusted models, menopause stage modified the associations of specific HDL metrics with CAC measures. Higher small HDL particles (HDL-P) concentrations (p-interaction = 0.008) and smaller HDL size (p-interaction = 0.02) were associated with greater odds of CAC presence in late perimenopause than in pre/early perimenopause stage. Women in the highest estradiol tertile, but not the lower tertiles, showed a protective association of small HDL-P with CAC presence (p-interaction = 0.007). Lower large HDL-P concentrations (p-interaction = 0.03) and smaller HDL size (p-interaction = 0.03) were associated with lower CAC density in late perimenopause than in postmenopause stage. Associations of HDL phospholipid and triglyceride content and HDL-CEC with CAC measures did not vary by menopause stage or estradiol level. We concluded that HDL subclasses may impact the likelihood of CAC presence and the stability of coronary plaque differently over the menopause transition. Endogenous estradiol levels may contribute to this observation.

The objective of this study was to investigate the effects of dietary supplementation with different n-6/n-3 polyunsaturated fatty acid (PUFA) ratios on growth performance, meat quality, and fatty acid profile in Heigai pigs. A total of 54 Heigai finishing pigs (body weight: 71.59 &plusmn, 2.16 kg) were randomly divided into three treatments with six replications (three pigs per replication) and fed diets containing different n-6/n-3 PUFA ratios: 8:1, 5:1, and 3:1. Pigs fed the dietary n-6/n-3 PUFA ratio of 8:1 had the highest feed to gain ratio (p &lt, 0.01), carcass weight (p &lt, 0.05), redness a* (p &lt, 0.01), and yellowness b* (p &lt, 0.01). Fatty acid compositions in longissimus dorsi muscle (LDM) and subcutaneous adipose tissue (SAT) were significantly changed (p &lt, 0.01). Notably, the meat from the pigs fed with the low dietary n-6/n-3 PUFA ratio had higher n-3 PUFA contents (p &lt, 0.01) and lower n-6/n-3 PUFA ratio (p &lt, 0.01). The triglyceride and total cholesterol contents were significantly decreased in SAT from the pigs fed with dietary n-6/n-3 PUFA ratios of 5:1 (p &lt, 0.05) and 3:1 (p &lt, 0.01). Reducing n-6/n-3 PUFA ratio upregulated the expression of HSL (p &lt, 0.05), CPT1 (p &lt, 0.01), and FABP4 (p &lt, 0.01) but downregulated ATGL (p &lt, 0.01) expression. These results demonstrate that the lower n-6/n-3 PUFA ratio regulates meat quality and enhances the deposition of n-3 PUFA in Heigai pigs.

Background: Obesity affects 25% of pregnant women and is associated with a higher risk of neonatal complications, such as macrosomia and increased adiposity. The placenta may contribute to neonatal adiposity by accumulating and transferring excess lipid in response to maternal hyperinsulinemia. We previously found that insulin promotes a 3-fold increase in placental triglyceride (TG) content in lean women. We hypothesized that obese women have higher placental insulin resistance compared to lean women[FC1] with respect to TG content. To learn more about the mechanisms of human dietary fat perception, 398 human twins rated fattiness and liking for six types of potato chips that differed in triglyceride content (2.5, 5, 10, and 15% corn oil); reliability estimates were obtained from a subset (n = 50) who did the task twice. Some chips also had a saturated long-chain fatty acid (hexadecanoic acid, 16:0) added (0.2%) to evaluate its effect on fattiness and liking. We computed the heritability of these measures and conducted a genome-wide association study (GWAS) to identify regions of the genome that co-segregate with fattiness and liking. Perceived fattiness and liking for the potato chips were reliable (r = 0.31-0.62, p < 0.05) and heritable (up to h2 = 0.29, p < 0.001, for liking). Adding hexadecanoic acid to the potato chips significantly increased ratings of fattiness but decreased liking. Twins with the G allele of rs263429 near GATA3-AS1 or the G allele of rs8103990 within ZNF729 reported more liking for potato chips than did twins with the other allele (multivariate GWAS, p < 1×10-5), with results reaching genome-wide suggestive but not significance criteria. Person-to-person variation in the perception and liking of dietary fat was (a) negatively affected by the addition of a saturated fatty acid and (b) related to inborn genetic variants. These data suggest liking for dietary fat is not due solely to fatty acid content and highlight new candidate genes and proteins within this sensory pathway.

This study aimed to evaluate the effects of supplementation with polyunsaturated oil on lipid and mineral blood biomarkers in two different groups of horses. Was used 6 healthy horses in maintenance (MT-g) and 10 healthy horses in marcha gait training (GT-g). The MT-g and the GT-g received 100mL and 300mL/day/animal of combination of polyunsaturated oils, respectively, during 8 weeks. Blood was collected in pre-test and after 4 and 8 weeks, after overnight fasting and it was measured: total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride, NEFA, LPS, glycerol, Ca, P, Mg, and Cl. The supplementation produced significant increase in NEFA and reduction in lipase in both groups. In addition, the LPS content in both groups had a large decrease (~35%) from the pre-test to the 8th week of supplementation. In the MT-g, the LDL cholesterol and glycerol increased significantly after 4 weeks but returned to the pre-test levels after 8 weeks, and the triglyceride content decreased (~62%) after 4 weeks. The total cholesterol concentration in the GT-g increased after 4 weeks (~70%), but after 8 weeks it decreased to levels similar to those observed in the pre-test. The mineral biomarkers increased in the MT-g and GT-g, after 8 weeks. In the MT-g and in the GT-g, Ca decreased. In conclusion, supplementation with a combination of polyunsaturated oils increased NEFA concentration and reduced LPS activity in both groups, unlike the concentrations of the other biomarkers that showed different variations between groups, indicating that both NEFA and LPS the changes in their concentrations during supplementation may be more sensitive and more indicated for these types of evaluation. In addition, some blood mineral concentrations reduced significantly in both groups of horses, and thus, they need to be evaluated in horses under chronic oil supplementation Resumo O objetivo deste estudo foi avaliar os efeitos da suplementação com uma combinação de óleos poliinsaturado nos biomarcadores lipídicos e minerais sanguíneos em dois diferentes grupos de equinos. Foram utilizados 6 equinos saudáveis em manutenção (MT-g) e 10 equinos saudáveis em treinamento de marcha (GT-g). O MT-g e o GT-g receberam 100,0mL e 300mL/dia/animal de uma combinação de óleos poliinsaturado, respectivamente, durante 8 semanas. O sangue foi coletado no pré-teste e após 4 e 8 semanas, após a noite, medidas: colesterol total, colesterol HDL, colesterol LDL, triglicérides, NEFA, LPS, glicerol, Ca, P, Mg e Cl. A suplementação produziu aumento significativo de NEFA e redução da lipase nos dois grupos. Além disso, o conteúdo de LPS em ambos os grupos ocorreu um decréscimo (~ 35%) desde o pré-teste até a 8ª semana de suplementação. Na MT-g, o colesterol LDL e o glicerol aumentaram significativamente após 4 semanas, mas retornaram aos níveis pré-teste após 8 semanas, e o conteúdo de triglicérides diminuiu (~ 62%) após 4 semanas. A concentração total de colesterol no GT-g aumentou após 4 semanas (~ 70%), mas após 8 semanas diminuiu para valores semelhantes aos observados no pré-teste. Os biomarcadores minerais aumentaram no MT-g e GT-g, após 8 semanas. No MT-g e no GT-g, o Ca diminuiu. Em conclusão, a suplementação com uma combinação de óleos poliinsaturado elevou a concentração do NEFA e reduziu a atividade de LPS em ambos os grupos estudados, diferentemente das concentrações das demais biomarcadores que apresentaram variações diferentes entre os grupos, indicando que tanto o NEFA como a LPS podem ser mais sensíveis as modificações em suas concentrações durante a suplementação e mais indicados para esse tipo de avaliação. Além disso, a concentração sanguínea de alguns minerais reduziu significativamente em ambos os grupos de equinos e, portanto, precisam ser avaliados em cavalos sob suplementação crônica de óleos.

The aim of this work was to evaluate two functional feeds for the gilthead seabream, Sparus aurata, containing low inclusion of two microalgae-based products (LB-GREENboost, LBGb; and LB-GUThealth, LBGh). Fish (12–13 g) were fed for 13 weeks a control diet or one of the four diets supplemented with both products at 0.5% or 1%. LBGb and LBGh did not affect specific growth rate or survival, but increased feed efficiency by decreasing feed intake and enlarging the intestines. LBGb increased hepatosomatic index and reduced cortisol levels in plasma, while both products lowered plasma lactate. Extensive metabolite and metabolic enzyme profiling revealed that microalgae supplementations, especially 1% LBGh: (i) decrease plasma lactate and increase hepatic glycogen, (ii) reduce hepatic gluconeogenesis, (iii) enhance hepatic lipogenic activity and lipid secretion, (iv) led fish to double triglyceride content in muscle and to stimulate its lipid oxidative capacity, and (v) increase the content of monounsaturated fatty acids and the omega-3 alpha-linolenic acid in muscle. This study demonstrates that both microalgae-based products are suited to improve feed efficiency and orchestrate significant changes in the intermediary metabolism in gilthead seabream juveniles.

Microplastics are an emerging environmental issue as a result of their ubiquity, persistence, and intrinsic toxic potential. In addition, their ability to sorb and transport a wide variety of environmental pollutants (i.e. “Trojan Horse” effect) exerts significant adverse impacts upon ecosystems. The toxicological evaluation of the single and combined effects produced by polyethylene microplastics and two polychlorinated biphenyl congeners was performed on the human hepatoma cell line HepG2 by cell viability assessment and an untargeted lipidomic study. The cell lethality evaluation evinced that MPs did not induce relevant cell lethality at any of the concentration range tested, while both PCBs presented a hormetic behavior. The lipidomic analysis suggested that both single PCB exposures induced significant lipidomic changes, especially for glycerophospholipids and glycerolipids. In contrast, for MPs single exposure, the most remarkable change was the substantial enhancement of triglyceride content. Regarding combined exposures, results showed that MPs could induce even more harmful effects than those produced intrinsically as a result of desorbing previously sorbed toxic pollutants. To the best of our knowledge, this is the first study assessing the toxicity of microplastics and their possible “Trojan Horse” effect by applying an untargeted lipidomic methodology., The research leading to these results has received funding from the Spanish Ministry of Science and Innovation (MCI, Grant CTQ2017-82598-P). Intramyocellular triglyceride (imTG) in skeletal muscle plays a significant role in metabolic health, and an infusion of [(13)C(16)]palmitate can be used to quantitate the in vivo fractional synthesis rate (FSR) and absolute synthesis rate (ASR) of imTGs. However, the extramyocellular TG (emTG) pool, unless precisely excised, contaminates the imTG pool, diluting the imTG-bound tracer enrichment and leading to underestimation of FSR. Because of the difficulty of excising the emTGs precisely, it would be advantageous to be able to calculate the imTG synthesis rate without dissecting the emTGs from each sample. Here, we tested the hypothesis that the ASR of total TGs (tTGs), a combination of imTGs and emTGs, calculated as “FSR × tTG pool,” reasonably represents the imTG synthesis. Muscle lipid parameters were measured in nine healthy women at 90 and 170 min after the start of [(13)C(16)]palmitate infusion. While the measurements of tTG content, enrichment, and FSR did not correlate (P > 0.05), those of the tTG ASR were significantly correlated (r = 0.947, P < 0.05). These results demonstrate that when imTGs and emTGs are pooled, the resulting underestimation of imTG FSR is balanced by the overestimation of the imTG content. We conclude that imTG metabolism is reflected by the measurement of the tTG ASR.

Devices that analyze body composition of rodents by time-domain nuclear magnetic resonance (TD-NMR) are becoming popular in research centers that study metabolism. Theoretically, TD-NMR devices can also evaluate lipid content in isolated tissues. However, the accuracy of TD-NMR to determine hepatic steatosis in the liver of small laboratory animals has not been evaluated in detail. We observed that TD-NMR was able to detect increased lipid content in the liver of rats consuming high-fat diet (HFD) for 12 weeks and in genetically obese (Lepob/ob and Leprdb/db) mice. The lipid content determined by TD-NMR showed a positive correlation with triglyceride content measured by colorimetric assays. In contrast, TD-NMR did not detect hepatic steatosis in C57BL/6 mice consuming HFD for 4 or 12 weeks, despite their obesity and increased liver triglyceride content. These findings indicate that tissue mass and the severity of hepatic steatosis affect the sensitivity of TD-NMR to detect liver lipid content.

Background Renal steatosis (fatty kidney) is a potential biomarker for obesity-related renal disease; however, noninvasive assessment of renal fat content remains a technical challenge. Purpose To evaluate reproducibility and explore clinical application of renal metabolic imaging for the quantification of renal triglyceride content (TG) using proton magnetic resonance spectroscopy (1 H-MRS). Study type Reproducibility and clinical cohort study. Population Twenty-three healthy volunteers (mean age 30.1 ± 13.4 years) and 15 patients with type 2 diabetes mellitus (T2DM) (mean age 59.3 ± 7.0 years). Field strength/sequence 3T, single-voxel point resolved spectroscopy (PRESS). Assessment Intra- and interexamination reproducibility of renal TG was assessed in healthy volunteers, and compared to T2DM patients. Intraexamination differences were obtained by repeating the 1 H-MRS measurement directly after the first 1 H-MRS without repositioning of the subject or changing surface coil and measurement volumes. Interexamination variability was studied by repeating the scan protocol after removal and replacement of the subject in the magnet, and subsequent repositioning of body coil and measurement volumes. Statistical tests Reproducibility was determined using Pearson's correlation and Bland-Altman analyses. Differences in TG% between healthy volunteers and T2DM patients were assessed using the Mann-Whitney U-test. Results After logarithmic (log) transformation, both intraexamination (r = 0.91, n = 19) and interexamination (r = 0.73, n = 9) measurements of renal TG content were highly correlated with the first renal TG measurements. Intraexamination and interexamination limits of agreement of renal log TG% were respectively [-1.36%, + 0.84%] and [-0.77%, + 0.62%]. Backtransformed limits of agreement were [-0.89%,+0.57%] and [-0.55%, + 0.43%] multiplied by mean TG for intra- and interexamination measurements. Overall median renal TG content was 0.12% [0.08, 0.22; 25th percentile, 75th percentile] in healthy volunteers and 0.20% [0.13, 0.22] in T2DM patients (P = 0.08). PurposeAccumulation of triglycerides in nonadipose tissue (e.g. ectopic lipids) is characteristic of metabolic derangements and is linked to insulin resistance and cardiovascular disease. Although the detrimental effects of the total amount of ectopic fat has been established, the role of composition of the ectopic lipid stores is unknown. In this study we used proton magnetic resonance spectroscopy (H-1-MRS) homonuclear spectral editing to characterize lipid stores in adipose tissue and skeletal muscle at 4T. MethodsA MEGA-sLASER sequence was used to selectively detect lipid resonances that are scalar coupled to the methine resonance at 5.31ppm and that can be used to estimate saturated fatty acid and mono- and polyunsaturated fatty acids. Phantom experiments were performed to empirically determine correction factors for editing efficiency of the different lipid groups. ResultsThe spectral editing approach enabled the estimation of saturated, mono-unsaturated, and polyunsaturated fatty acid contributions in phantoms and in vivo. These estimations are in the same order as reported in studies using invasive biopsies. ConclusionsIn this study, we have shown the feasibility of spectral editing techniques for ectopic lipid store characterization with H-1-MRS, regardless of spectral resolution (e.g., B(0-)field strength). Exercise training reduces intrahepatic lipid (IHL) content in people with elevated liver fat content. It is unclear, however, whether exercise training reduces IHL content in people with normal liver fat content. Here, we measured the effect of exercise training on IHL content in people with and people without nonalcohol fatty liver. We further measured changes in insulin sensitivity and hepatic energy metabolism. Eleven males with nonalcoholic fatty liver (NAFL) and 11 body mass index-matched individuals without nonalcoholic fatty liver (CON) completed a 12-wk supervised exercise training program. IHL content (proton magnetic resonance spectroscopy), maximal oxidative capacity (V̇o2max, spiroergometry), total muscle strength, body composition, insulin sensitivity (hyperinsulinemic-euglycemic clamp), hepatic ATP-to-total phosphorus ratio, and the hepatic phosphomonoester-to-phosphodiester (PME/PDE) ratio (phosphorus magnetic resonance spectroscopy) were determined. IHL content reduced with exercise training ( P = 0.014) in the whole study population. The relative reduction in IHL content was comparable in NAFL (−34.5 ± 54.0%) and CON (−28.3 ± 60.1%) individuals ( P = 0.800). V̇o2max ( P < 0.001), total muscle strength ( P < 0.001), and skeletal muscle insulin sensitivity ( P = 0.004) increased, whereas adipose tissue ( P = 0.246) and hepatic ( P = 0.086) insulin sensitivity did not increase significantly. Hepatic ATP-to-total phosphorus ratio ( P = 0.987) and PME/PDE ratio ( P = 0.792) did not change. Changes in IHL content correlated with changes in body weight ( r = 0.451, P = 0.035) and changes in hepatic PME/PDE ratio ( r = 0.569, P = 0.019). In conclusion, exercise training reduced intrahepatic lipid content in people with nonalcoholic fatty liver and in people with normal intrahepatic lipid content, and the percent reduction in intrahepatic lipid content was similar in both groups.

Objectives An assessment of methods for the accurate measurement of low-density lipoprotein cholesterol (LDL-C) at decreased concentrations has not yet been carried out. We evaluated the performance of the Friedewald equation, a direct enzymatic assay, and a novel equation for determining LDL-C levels in a pediatric population with elevated triglycerides and reduced LDL-C levels. Methods LDL-C concentrations of 127 pediatric patients were determined by the Friedewald equation, a direct enzymatic assay, and a novel equation. The bias of each approach was assessed at selected LDL-C cutoffs and after stratifying samples by triglyceride content. The concordance of each approach, relative to the reference method, was determined at LDL-C cut-points of less than 70, 70 to 99, and 100 to 129 mg/dL. Results The Friedewald equation substantially underestimated pediatric LDL-C concentrations below 100 mg/dL in the presence of elevated triglycerides. The Ortho Clinical Diagnostics (Raritan, NJ) direct LDL assay was positively biased at low LDL-C levels. The novel equation most effectively reduced the bias of the Friedewald equation at all LDL-C concentrations and increased the concordance of sample classification to the reference method. Conclusions The novel equation should be used for accurate measurement of pediatric LDL-C when the concentration is below 100 mg/dL in the presence of elevated triglycerides (150-399 mg/dL).

Purpose Epicardial (EAT) and pericardial (PAT) fat masses and myocardial triglyceride content (MTC) are enlarged in obesity and insulin resistance. We studied whether the high-intensity interval training (HIIT) and moderate-intensity continuous training (MICT) similarly decrease ectopic fat in and around the heart and whether the decrease is similar in healthy subjects and subjects with defective glucose tolerance (DGT). Methods A total of 28 healthy men (body mass index = 20.7–30.0 kg·m−2, age = 40–55 yr) and 16 men with DGT (body mass index = 23.8–33.5 kg·m−2, age = 43–53 yr) were randomized into HIIT and MICT interventions for 2 wk. EAT and PAT were determined by computed tomography and MTC by 1H-MRS. Results At baseline, DGT subjects had impaired aerobic capacity and insulin sensitivity and higher levels of whole body fat, visceral fat, PAT, and EAT (P < 0.05, all) compared with healthy subjects. In the whole group, HIIT increased aerobic capacity (HIIT = 6%, MICT = 0.3%; time × training P = 0.007) and tended to improve insulin sensitivity (HIIT = 24%, MICT = 8%) as well as reduce MTC (HIIT = −42%, MICT = +23%) (time × training P = 0.06, both) more efficiently compared with MICT, and without differences in the training response between the healthy and the DGT subjects. However, both training modes decreased EAT (−5%) and PAT (−6%) fat (time P < 0.05) and not differently between the healthy and the DGT subjects. Conclusion Whole body fat, visceral fat, PAT, and EAT masses are enlarged in DGT. Both HIIT and MICT effectively reduce EAT and PAT in healthy and DGT subjects, whereas HIIT seems to be superior as regards improving aerobic capacity, whole-body insulin sensitivity, and MTC.

The average blood urea nitrogen content in T1 13.99, 17.26, 17.47 and 14.28, 17.32, 18.41 in T2 (mg/dl). The average total serum lipid content in T1 272.51, 278.70, 283.60 and 277.02, 300.38, 315.53 (mg/dl) in T2. Average total serum cholesterol content in T1- 59.92, 70.80, 74.95 and T2- 60.94, 70.29, 76.27 (mg/dl). Average total serum triglyceride content in T1- 16.07, 17.26, 17.48 and T2- 16.22, 17.98, 17.89 (mg/dl) in the month of February, April and June, respectively.

This experiment was conducted to study the effects of pantothenic acid on the growth performance, slaughter performance, lipid metabolism, and antioxidant function of one-to four-week-old Wulong geese and determine the appropriate supplemental level of pantothenic acid. A total of 360 one-day-old Wulong geese were randomly divided into 6 groups with 6 replicates per group and 10 geese (5 males and 5 females) per replicate. The geese in group Ⅰ (control group) were fed a basal diet, and the geese in groups Ⅱ to Ⅵ (experimental groups) were given the basal diet supplemented with 8, 15, 30, 60, and 120 mg/kg pantothenic acid, respectively. The experiment lasted for 4 weeks. Based on the results of unrelated comparative analysis, the body weight was the highest when the dietary pantothenic acid level was 27.57 mg/kg. When the dietary pantothenic acid level was 26.17 mg/kg, the average daily gain peaked. When the dietary pantothenic acid level was 15.50 mg/kg, the feed:gain ratio was the lowest. In this issue of the journal , Dr. Yasuki Ito from Denka-Seiken, along with colleagues, describes the development of a novel direct assay for the measurement of the triglyceride content of LDL (LDL-TG2; 1). It has been known for over 100 years, since the pioneering work of Dr. N. Anitschkow in 1913 (2), that plasma cholesterol plays a central role in the pathogenesis of atherosclerosis. Much later in the mid-1950s, Dr. John Gofman first showed that, by separating lipoproteins via analytical ultracentrifugation, cholesterol on LDL (LDL-C) is positively related to cardiovascular disease risk, whereas cholesterol on HDL (HDL-C) is inversely related (3). Most recently, however, there has been a growing awareness of the importance of the compositional complexity of lipoproteins, and the possibility that other lipid and protein components of lipoproteins besides cholesterol can also serve as biomarkers (4). In fact, in a 2004 study by Dr. Winfried Marz and colleagues, it was first reported that LDL-TG was superior to LDL-C as a cardiovascular disease risk predictor (5). Intoduction Excess visceral and liver fat are known risk factors for cardiometabolic disorders. Metabolomics might allow for easier quantification of these ectopic fat depots, instead of using invasive and costly tools such as MRI or approximations such as waist circumference. Objective We explored the potential use of plasma metabolites as biomarkers of visceral adipose tissue (VAT) and hepatic triglyceride content (HTGC). Methods We performed a cross-sectional analysis of a subset of the Netherlands Epidemiology of Obesity study. Plasma metabolite profiles were determined using the Biocrates AbsoluteIDQ p150 kit in 176 individuals with normal fasting plasma glucose. VAT was assessed with magnetic resonance imaging and HTGC with proton-MR spectroscopy. We used linear regression to investigate the associations of 190 metabolite variables with VAT and HTGC. Results After adjustment for age, sex, total body fat, currently used approximations of visceral and liver fat, and multiple testing, three metabolite ratios were associated with VAT. The strongest association was the lysophosphatidylcholines to total phosphatidylcholines (PCs) ratio [− 14.1 (95% CI − 21.7; − 6.6) cm2 VAT per SD of metabolite concentration]. Four individual metabolites were associated with HTGC, especially the diacyl PCs of which C32:1 was the strongest at a 1.31 (95% CI 1.14; 1.51) fold increased HTGC per SD of metabolite concentration. Conclusion Metabolomics may be a useful tool to identify biomarkers of visceral fat and liver fat content that have added diagnostic value over current approximations. Replication studies are required to validate the diagnostic value of these metabolites.

Objective: SGLT2 inhibitors reduce body weight (BW) due to negative energy balance. Although previous reports suggested that SGLT2 inhibitor improved liver dysfunction and steatosis, the efficacy of SGLT2 inhibitor on intrahepatic triglyceride (IHTG) were merely quantified. Methods: The study was an open label randomized controlled trial. Fifty-two overweight patients (BMI > 23) who had inadequate glycemic control with oral antidiabetic agents were randomly assigned to dapagliflozin (Dapa) or standard treatment (Con) and followed for 24 weeks. The Primary endpoint was the change in BW from baseline to week 24 between the two groups. The secondary endpoint was IHTG which was assessed by magnetic resonance spectroscopy (MRS). Results: The change in BW was significantly larger in Dapa group compared to Con. The mean difference in changes between two groups was -1.72 kg (95% CI: -2.85, -0.59, p = 0.004) which was comparable with the previous studies of Caucasians. IHTG measured with MRS was significantly reduced in Dapa group (p = 0.033). Conclusion: Dapagliflozin treatment for 24 weeks resulted in reduction of BW and IHTG. These findings suggest the therapeutic potential of dapagliflozin for patients with NAFLD. Clinical study registration number: UMIN000020239. Disclosure K. Background: Obesity affects 25% of pregnant women and is associated with a higher risk of neonatal complications, such as macrosomia and increased adiposity. In addition to transporting maternal nutrients to the fetus, the placenta receives maternal hormone signals and synthesizes paracrine hormones which influence nutrient flux to the fetus. Obese women were shown to have higher placental lipid content compared to lean women, but the regulation of placental lipid metabolism is unknown. We hypothesized that insulin regulates lipid metabolism in the placenta by activating IR-B, which is the insulin receptor (IR) isoform characterized as having metabolic activity. Methods: Healthy, lean women (n=7) of mean age 34.4 ± 1.4 years and BMI 21.5 ± 0.5 kg/m2 consented for placental collection at elective c-section. Full thickness placental samples from multiple quadrants were obtained. Villous explants were cultured for 24 hours. After 4-hour starvation, explants were treated for 48 hours with insulin 100nM, IGF-1 100nM, IGF-2 100nM, or vehicle. Lipids were extracted from homogenized explants using a chloroform-methanol separation protocol. Triglyceride (TG) content was quantified by colorimetric spectrophotometer and normalized to explant weight. Data were analyzed by one-way ANOVA with a Tukey’s multiple comparisons test. Results: In placenta explants from lean women, insulin induced a 2.5-fold increase in triglyceride content, relative to vehicle (19.8 ± 3 vs 7.3 ± 1.5 mcg/mg, p= 0.002). Similarly, IGF-2 promoted a 3.4-fold increase compared to vehicle-treated placental tissue (24.9 ± 3.3 mcg/mg, p= 0.0002). In contrast, IGF-1 did not significantly alter TG levels (12.7 ± 2.9 vs 10.2 ± 2.6 mcg/mg, p= NS). Conclusions: We show that placental triglyceride content is equally regulated by maternal insulin and placenta-synthesized IGF-2. Since IGF-2 has a high affinity for IR-A and minimal affinity for IR-B, these data indicate that IR-A likely plays a key role in placental lipid metabolism. These findings are novel since IR-A activity is generally characterized as mitogenic, rather than metabolic. Notably, IGF-1, which activates IGF-1R, does not play a role in placental lipid metabolism in vitro. Hyperinsulinemia due to maternal obesity may alter placental triglyceride storage and thus promote neonatal adiposity through greater availability, independent of circulating maternal triglyceride levels. Laboratory analytical techniques employed for triglyceride quantification in oleaginous biomass (e.g., microalgae and oilseeds) involve multiple steps and typically require use of volatile organic solvents. Here we describe a single-step approach for measurement of triglycerides using thermogravimetry (TG). We have observed that triglycerides undergo complete volatilization over a narrow temperature interval of 370-450 °C, with negligible solid residue under inert atmosphere, whereas other constituents of oleaginous biomass (such as proteins and carbohydrates) primarily degrade below 350 °C. As a result, triglyceride content of biomass can be estimated using TG by determining the mass loss of the sample in the temperature interval of 370-450 °C.

Feeding of untreated soybean straw (T 2 ) and 2 per cent urea treated soybean straw (T 3 ) was evaluated on the performance of lactating cows in reference to conventional jowar straw diet (T 1 ). It was found within 120 days trial that, the Hb levels showed a increase from the initial level of 9.83 to 9.96, 9.76 to 10.08 and 9.86 to 10.80 g/dl/cow, blood glucose levels increased from 48.68 to 48.89, 45.89 to 53.36 and 45.16 to 56.24 mg/dl in T 1 , T 2 and T 3 groups, respectively The STP levels increase from initial content was 5.89 to 6.05, 5.76 to 6.38 and 5.70 to 6.58 g/dl, Significantly more BUN (17.82 mg/dl) was noticed in cows offered with T3 followed by T 2 (13.85 mg/dl) and T 2 (16.66 mg/dl) while blood urea pooled means were noticed as 29.65, 38.13 and 35.67 mg/dl in cows offered T 1 , T 2 and T 3 groups, respectively The pooled means of STL were 278.15, 298.22 and 302.30 mg/d, the pooled means of STC content were 69.18, 68.64 and 77.06mg/dl and the serum triglyceride content of blood increased were 16.72, 16.79 and 17.57mg/dl, in cows offered T 1 , T 2 and T 3 groups, respectively. The results do suggest that 2 per cent urea treated SBS can find a place in the ration of lactating cows without any adverse effect on health of cows.
