In order to explore the floor failure characteristics of the passing anticline section of the working face, taking the 16091 working face of Zhaogu No.1 mine as an example, the passing anticline section and the normal mining section are divided according to the actual geological conditions of the working face and the field microseismic monitoring data, and the distribution characteristics of microseismic events of the passing anticline section and the normal mining section are compared and analyzed. The failure characteristics of the floor of the working face passing anticline section under the microseismic angle are revealed. Then the failure law of coal mining floor under anticlinal structure is revealed by similar simulation test. The results show that the anticlinal axis of B1 is a region of high energy core density when the working face passes through the anticlinal section; the failure depth of the floor in the early and late anticlinal sections of the working face is 1.12 and 1.08 times that of the normal mining section, respectively, and the failure boundary of the floor touches the interior of L8 limestone. In the process of coal mining on deep confined water body, the tectonic stress in the passing anticlinal section has a significant effect on the instability failure of floor.

Abstract Cryptosporidium parvum is a zoonotic parasite of the intestine and poses a threat to human and animal health. However, it is difficult to obtain a large number of oocysts for genome sequencing using in vitro culture. To address this challenge, we employed the strategy of whole-genome amplification of 10 oocysts followed by long-read sequencing and obtained a high-quality genome assembly of C. parvum IIdA19G1 subtype isolated from a pre-weaning calf with diarrhea. The assembled genome was 9.13 Mb long and encompassed eight chromosomes with six capped by telomeric sequences at one or both ends. In total, 3,915 protein-coding genes were predicted, exhibiting a high completeness with 98.2% single-copy BUSCO genes. To our current knowledge, this represents the first chromosome-level genome assembly of C. parvum achieved through the combined use of whole-genome amplification of 10 oocysts and long-read sequencing. This achievement not only advances our understanding of the genomic landscape of this zoonotic intestinal parasite, but also provides valuable resources for comparative genomics and evolutionary analyses within the Cryptosporidium clade.

This paper reviews recent developments and key advances in terahertz (THz) science, technology, and applications, focusing on 3 core areas: astronomy, telecommunications, and biophysics. In THz astronomy, it highlights major discoveries and ongoing projects, emphasizing the role of advanced superconducting technologies, including superconductor–insulator–superconductor (SIS) mixers, hot electron boundedness spectroscopy (HEB), transition-edge sensors (TESs), and kinetic inductance detectors (KIDs), while exploring prospects in the field. For THz telecommunication, it discusses progress in solid-state sources, new communication technologies operating within the THz band, and diverse modulation methods that enhance transmission capabilities. In THz biophysics, the focus shifts to the physical modulation of THz waves and their impact across biological systems, from whole organisms to cellular and molecular levels, emphasizing nonthermal effects and fundamental mechanisms. This review concludes with an analysis of the challenges and perspectives shaping the future of THz technology.

Abstract Hydrothermal carbonization (HTC) stands out as an eco-friendly, cost-effective method for generating renewable carbon-based materials from biomass. The HTC process yields products such as hydrochars and carbon dots (CDs), possessed of notable photocatalytic capabilities due to their unique physicochemical features. Additionally, pairing traditional photocatalysts with hydrochar derivatives elevates their performance, rendering them more effective. Recent times have witnessed a surge in interest in these hydrochar based photocatalysts (HC-photocatalysts). Their appeal stems from multiple attributes: impeccable performance, adaptability to visible light, and adjustable physicochemical properties. This review delves deep into the evolving landscape of these HC-photocatalysts, segmenting them into three distinct categories: hydrochars, hydrochar-based CDs (HC-CDs), and hydrochar-based composites (HC-composites). For each category, we dissect their synthesis routes, unravel the photocatalytic mechanisms, and explore various enhancement strategies. We further traverse their versatile applications, spanning environmental treatment, disinfection, energy conversion, and organic synthesis. In the end, we spotlight the prevailing challenges and uncharted territories in the domain of HC-photocatalysts. In essence, this review serves as a guide, furnishing a theoretical foundation and steering directions for future explorations and tangible implementations of HC-photocatalysts. Graphical Abstract

BackgroundSelf-narratives about traumatic experiences and symptoms are informative for early identification of potential patients; however, their use in clinical screening is limited. This study aimed to develop an automated screening method that analyzes self-narratives of early adolescent earthquake survivors to screen for PTSD in a timely and effective manner.MethodsAn inquiry-based questionnaire consisting of a series of open-ended questions about trauma history and psychological symptoms, was designed to simulate the clinical structured interviews based on the DSM-5 diagnostic criteria, and was used to collect self-narratives from 430 survivors who experienced the Ya’an earthquake in Sichuan Province, China. Meanwhile, participants completed the PTSD Checklist for DSM-5 (PCL-5). Text classification models were constructed using three supervised learning algorithms (BERT, SVM, and KNN) to identify PTSD symptoms and their corresponding behavioral indicators in each sentence of the self-narratives.ResultsThe prediction accuracy for symptom-level classification reached 73.2%, and 67.2% for behavioral indicator classification, with the BERT performing the best.ConclusionsThese findings demonstrate that self-narratives combined with text mining techniques provide a promising approach for automated, rapid, and accurate PTSD screening. Moreover, by conducting screenings in community and school settings, this approach equips clinicians and psychiatrists with evidence of PTSD symptoms and associated behavioral indicators, improving the effectiveness of early detection and treatment planning.

Abstract Growing interest in Earth Orientation Parameters (EOP) resulted in various approaches to the EOP prediction algorithms, as well as in the exploitation of distinct input data, including the observed EOP values from various operational data centers and modeled effective angular momentum functions. Considering these developments and recently emerged new methodologies, the Second Earth Orientation Parameters Prediction Comparison Campaign (2nd EOP PCC) was pursued in 2021–2022. The campaign was led by Centrum Badań Kosmicznych Polskiej Akademii Nauk in cooperation with Deutsches GeoForschungsZentrum and under the auspices of the International Earth Rotation and Reference Systems Service. This paper provides the analysis and evaluation of the polar motion predictions submitted during the 2nd EOP PCC with the prediction horizons between 10 and 30 days. Our analysis shows that predictions are highly reliable with only a few occasional discrepancies identified in the submitted files. We demonstrate the accuracy of EOP predictions by (a) calculating the mean absolute error relative to polar motion observations from September 2021 through December 2022 and (b) assessing the stability of the predictions in time. The analysis shows unequal results for the x and y components of polar motion (PMx and PMy, respectively). Predictions of PMy are usually more accurate and have a smaller spread across all submitted files when compared to PMx. We present an analysis of similarity between the participants to indicate what methods and input data give comparable output. We also prepared the ranking of prediction methods for polar motion summarizing the achievements of the campaign.

Abstract In recent years, many governments and companies have gradually launched robotaxi projects to help make transportation systems smarter, improve travel efficiency, and reduce travel costs. Robotaxi is a new mode of travel that replaces human driving with machines, freeing up social labour and enriching people’s travel choices. This study employs the Extended Unified Theory of Acceptance and Use of Technology (UTAUT2) to understand the influencing factors of users’ adoption and usage of robotaxis in China to facilitate the broader integration of robotaxis into urban transportation systems. This study surveyed the preferences of 2048 respondents and analysed the data through structural equation modelling. The results indicate that performance expectancy, hedonic motivation, and price value are the factors influencing users’ behavioural intentions, while effort expectancy and social influence affect use behaviour. In contrast, habit is an important factor that affect both behavioural intention and actual use behaviour. Based on the findings, we have proposed practical strategies to improve robotaxi services and updated the UTAUT2 model in the context of robotaxi. We suggest that robotaxi operators can promote user acceptance and use by reducing the difficulty of use, improving the cost performance and the ride experience, and making appropriate publicity and guidance.

Abstract Background The 2018/2023 ESC/ESH Guidelines underlined a gap how baseline cardiovascular disease (CVD) risk predicted blood pressure (BP) lowering benefits. Further, 2017 ACC/AHA Guideline and 2021 WHO Guideline recommended implementation studies about intensive BP control. Now, to bridge these guideline gaps, we conducted a post hoc analysis to validate whether the baseline CVD risk influences the effectiveness of the intensive BP control strategy, which was designed by China Rural Hypertension Control Project (CRHCP). Methods This is a post hoc analysis of CRHCP, among which participants were enrolled except those having CVD history, over 80 years old, or missing data. Subjects were stratified into quartiles by baseline estimated CVD risk and then grouped into intervention and usual care group according to original assignment in CRHCP. Ciliated microeukaryotes (ciliates) are distinguished by their cilia, which are rich in tubulin. We developed a method for tubulin staining in ciliate cells that involves using live-cell tubulin-staining dyes instead of antibodies thereby streamlining the staining process, which is effective across diverse ciliate lineages. Moreover, our method allows integration with immunofluorescence staining using antibodies when needed. The potential applications of this technique extend to cell biology and ciliate morphological and ecological studies.

Abstract Balantidium ctenopharyngodoni is identified as the sole ciliate species that exclusively resides within the hindgut of grass carp with high prevalence and intensity. In this study, the successful cultivation of B. ctenopharyngodoni enabled us to collect enough cells for genome sequencing. Consequently, we acquired a high-quality genome assembly spanning 68.66 Mb, encompassing a total of 22,334 nanochromosomes. Furthermore, we predicted 29,348 protein-coding genes, and 95.5% of them was supported by the RNA-seq data. The trend of GC content in the subtelomeric regions of single-gene chromosomes was similar to other ciliates containing nanochromosomes. A large number of genes encoding carbohydrate-binding modules with affinities for starch and peptidoglycans was identified. The identification of mitochondrion-related organelles (MROs) within genome indicates its well-suited adaptation to the anaerobic conditions in the hindgut environment. In summary, our results will offer resources for understanding the genetic basis and molecular adaptations of balantidia to hindgut of herbivorous fish.

Abstract The Tibetan Plateau supplies water to nearly 2 billion people in Asia, but climate change poses threats to its aquatic microbial resources. Here, we construct the Tibetan Plateau Microbial Catalog by sequencing 498 metagenomes from six water ecosystems (saline lakes, freshwater lakes, rivers, hot springs, wetlands and glaciers). Our catalog expands knowledge of regional genomic diversity by presenting 32,355 metagenome-assembled genomes that de-replicated into 10,723 representative genome-based species, of which 88% were unannotated. The catalog contains nearly 300 million non-redundant gene clusters, of which 15% novel, and 73,864 biosynthetic gene clusters, of which 50% novel, thus expanding known functional diversity. Using these data, we investigate the Tibetan Plateau aquatic microbiome’s biogeography along a distance of 2,500 km and >5 km in altitude. Microbial compositional similarity and the shared gene count with the Tibetan Plateau microbiome decline along with distance and altitude difference, suggesting a dispersal pattern. The Tibetan Plateau Microbial Catalog stands as a substantial repository for high-altitude aquatic microbiome resources, providing potential for discovering novel lineages and functions, and bridging knowledge gaps in microbiome biogeography.

Abstract As an emerging business model, merchants are selling surplus food in blind boxes, which provides new opportunities for reducing waste and promoting sustainable development. Surplus food blind boxes, however, have not been extensively researched in terms of design and marketing strategies. Therefore, we are examining how to increase consumers’ purchase intention regarding surplus food blind boxes as well as the relationship between purchase intention, continuous intention, and recommendation intention. In this paper, two studies are conducted. As part of study 1, We used factor analysis to explore the reasons that influence consumers to purchase. In study 2, we utilized structural equation modeling to examine the impact of purchase intention on continuous intention and recommendation intention. Results indicate that food quality, perceived sustainability, and conspicuous consumption are the most important positive factors. The most important negative factors are perceived food risk, resistance to sales techniques, taste anxiety, and inadequate marketing strategies. Based on our findings, we constructed a path relationship between purchase intention, continuous intention, and recommendation intention. The purchase intention of the surplus food blind box users directly influences the recommendation intention and the continuous intention, while the recommendation intention indirectly influences the continuous intention. This study did not demonstrate that gender had a moderating effect. Meanwhile, it provides a reference for the actual marketing management of surplus food blind box merchants.

Abstract The peptidyl-prolyl cis-trans isomerase Pin1 is a pivotal therapeutic target in cancers, but the regulation of Pin1 protein stability is largely unknown. High Pin1 expression is associated with SUMO1-modified protein hypersumoylation in glioma stem cells (GSCs), but the underlying mechanisms remain elusive. Here we demonstrate that Pin1 is deubiquitinated and stabilized by USP34, which promotes isomerization of the sole SUMO E2 enzyme Ubc9, leading to SUMO1-modified hypersumoylation to support GSC maintenance. Pin1 interacts with USP34, a deubiquitinase with preferential expression and oncogenic function in GSCs. Such interaction is facilitated by Plk1-mediated phosphorylation of Pin1. Disruption of USP34 or inhibition of Plk1 promotes poly-ubiquitination and degradation of Pin1. Furthermore, Pin1 isomerizes Ubc9 to upregulate Ubc9 thioester formation with SUMO1, which requires CDK1-mediated phosphorylation of Ubc9. Combined inhibition of Pin1 and CDK1 with sulfopin and RO3306 most effectively suppresses orthotopic tumor growth. Our findings provide multiple molecular targets to induce Pin1 degradation and suppress hypersumoylation for cancer treatment.

Porcine epidemic diarrhea virus (PEDV) is a highly contagious coronavirus that infect pigs’ intestinal epithelial cells, causing high morbidity and mortality. Due to the rapid mutation of PEDV, vaccine efficacy is uncertain, prompting exploration of alternative treatments. Nanobodies, also known as variable heavy chain domains of heavy chain-only antibodies (VHHs), offer significant potential in biomedical applications due to their small size and high specificity. In this study, yeast two-hybrid technology was employed to screen for eight specific VHH sequences targeting the PEDV S protein from a synthetically constructed nanobody yeast library. The VHH genes were then cloned into expression plasmids for recombinant protein production, and the resulting VHHs (termed PEDV S-VHHs) were purified. Indirect immunofluorescence assay (IFA) and Western blotting analysis confirmed that these VHHs specifically bind to both PEDV and its S protein. Neutralization assays demonstrated that seven PEDV S-VHHs exhibited potent neutralizing activity against PEDV. Additionally, a combination of these seven antibodies showed enhanced antiviral effects. Preliminary predictions were also made regarding the binding sites between these VHHs and PEDV. The PEDV S-VHHs described in this study hold potential as candidates for the prevention and treatment of PEDV infection.

Abstract Co-culture systems of rice and aquatic animals can contribute to the ecological intensification of agriculture by reducing nutrient loss and the need for N fertilizer application and by enhancing nutrient-use efficiency. However, the input of high-protein diets into paddy fields, to facilitate the growth of aquatic animals, has been found to increase N pollution and acidification of the soil. Although soil amendments have been widely used to ameliorate acidic soils, reduce N2O emissions, and improve agronomic production, the relationship between soil amendments and aquatic animal remains unclear. Therefore, this study investigated the effects of calcined dolomite (hereafter referred to as dolomite) as an acidic soil amendment and Ca–Mg supplement in rice–crab co-culture using Eriocheir sinensis crabs (Chinese mitten crabs). High-throughput sequencing was used to examine crab bacterial community composition and crab hepatopancreas biology. Although the water pH was significantly increased in the dolomite group, the number, composition, and diversity of bacteria identified in crab gut microbiome did not vary significantly between the dolomite and control groups. In the dolomite group, the probiotic agents Candidatus Hepatoplasma and Lactobacillus were highly abundant in the crab gut, and immune- and retinol metabolism-related genes were significantly upregulated in the crab hepatopancreas. Overall, dolomite application increased crab health and water pH. Dolomite is a low-cost amendment, with better stability, compared to other soil amendments, thus making it ideal for sustainable and clean rice–aquatic animal co-culture.

New energy sources are transforming the automotive market. This shift has also expanded the possibilities for in-vehicle interaction. Through a literature review, this study categorizes the in-vehicle interaction activities into three types: driving tasks, comfort tasks, and entertainment tasks. This study conducted empirical survey of 377 users to understand their preferences of in-vehicle interaction input methods inside new energy vehicles. The results show that gender, educational level, income, driver's license type and driving experience have significant influence on the perception and preference of the in-vehicle interaction input methods. However, age and experience with new energy vehicle didn't show significant results. The findings of this study can assist manufacturers in developing targeted solutions and meeting the personalized needs of users in future vehicle market segments.

Abstract Understanding the relationship between plant diversity and invasibility is essential in invasion ecology. Species‐rich communities are hypothesized to be more resistant to invasions than species‐poor communities. However, while soil microorganisms play a crucial role in regulating this diversity–invasibility relationship, the effects of plant competition mode and soil nutrient status on their role remain unclear. To address this, we conducted a two‐stage greenhouse experiment. Soils were first conditioned by growing nine native species separately in them for 1 year, then mixed in various configurations with soils conditioned using one, three, or six species, respectively. Next, we inoculated the mixed soil into sterilized substrate soil and planted the alien species Rhus typhina and native species Ailanthus altissima as test plants. We set up two competition modes (intraspecific and interspecific) and two nutrient levels (fertilization using slow‐release fertilizer and nonfertilization). Under intraspecific competition, regardless of fertilization, the biomass of the alien species was higher in soil conditioned by six native species. By contrast, under interspecific competition, the biomass increased without fertilization but remained stable with fertilization in soil conditioned by six native species. Analysis of soil microbes suggests that pathogens and symbiotic fungi in diverse plant communities influenced R. typhina growth, which varied with competition mode and nutrient status. Our findings suggest that the soil microbiome is pivotal in mediating the diversity–invasibility relationship, and this influence varies according to competition mode and nutrient status.

Biomethylation is an effective means of arsenic detoxification by organisms living in aquatic environments. Ciliated protozoa (including Tetrahymena species) play an important role in the biochemical cycles of aquatic ecosystems and have a potential application in arsenic biotransformation. This study compared arsenic tolerance, accumulation, methylation, and efflux in 11 Tetrahymena species. Nineteen arsenite (As(III)) S-adenosylmethionine (SAM) methyltransferase (arsM) genes, of which 12 are new discoveries, were identified, and protein sequences were studied. We then constructed recombinant cell lines based on the Tetrahymena thermophila (T. thermophila) wild-type SB210 strain and expressed each of the 19 arsM genes under the control of the metal-responsive the MTT1 promoter. In the presence of Cd2+ and As(V), expression of the arsM genes in the recombinant cell lines was much higher than in the donor species. Evaluation of the recombinant cell line identified one with ultra-high arsenic methylation enzyme activity, significantly higher arsenic methylation capacity and much faster methylation rate than other reported arsenic methylated organisms, which methylated 89% of arsenic within 6.5 h. It also had an excellent capacity for the arsenic detoxification of lake water containing As(V), 56% of arsenic was methylated at 250 μg/L As(V) in 48 h. This study has made a significant contribution to our knowledge on arsenic metabolism in protozoa and demonstrates the great potential to use Tetrahymena species in the arsenic biotransformation of aquatic environments.

The construction of floor pumping roadway in flooding mining area can affect the water resistance of floor strata, increase the risk of coal mining and bring challenges to the floor grouting reinforcement technology. So, the study on the influence of roadway on the water resistance of floor strata under far field stresses and the optimization of control technology was carried out. According to the axial and cross section characteristics of the floor pumping roadway, an analysis model of the bending resistance of key rock strata under the far field principal stresses was established and the theoretical solutions of the deformation and the bending moment of the siltstone under the floor failure area with and without a floor pumping roadway were obtained. Furthermore, the application effect of grout filled casing pipes on the reinforcement of rock strata is studied and verified numerically. First, according to the far field principal stresses, the stress component σ′ acting on siltstone II is obtained in the local coordinate system. According to Levy solution of elastic rectangular thin plate single trigonometric series, the precise solutions of rock deformation and bending moment are obtained by using the generalized simply supported edge theory and superposition method. The exact solution of the deflection equation for the thin plate of strata penetrated by the gas roadway with two adjacent edges free and two adjacent edges fixed is got, while the ratio of mining distance to working face length is 0.50, 0.75, 1.00, 1.50 and 2.00, respectively. The calculation results show that the displacement of siltstone II at the intersection of roadway under the influence of roadway under the elastic condition is the largest, which is larger than the maximum displacement of the four sides fixed with the same geometric size. The maximum bending moment of the fixed side of siltstone II under the influence of floor pumping roadway is one order of magnitude higher than that of the fixed side. It illustrates that the siltstone II influenced by gas roadway is easier to be deformed than that without roadway, and its resistance to deformation is reduced to some extent. Based on this, an optimization method for grouting reinforcement of floor rock strata is proposed. The vertical depth of grout filling casing pipes entering into siltstone III is designed to be greater than 2 m, which can play the role of fixed end and increase constraints on free edge of siltstone II. The long slurry casing pipes can improve the boundary conditions of siltstone II and increase the resistance strength due to the deformation difference within the reinforcement range.The numerical results verify the actual effect of grout filling casing on the siltstone deformation.

Based on Zhaogu No. 1 Mine’s characters that are the overlying thick alluvium, multi-aquifers (groups) and thin bedrock, the water pressure of the gravel aquifer under the alluvial layer reaches 4.0 MPa, defined a high-pressure aquifer. To determine the influence of bedrock properties on roof water inrush and sand bursting, and ensure the normal mining around the thin bedrock area under groups, there were tests, point loading, dry saturated water absorption rate and indoor disintegration, of bedrock samples taken from hydrological survey holes to determine those properties and influence on retaining sand-proof pillars by analyzing the variation curves of various indexes of them with depth. The experiments’ results showed that the weathering depth of bedrock exceeds 20 m; the dry saturated water absorption rate of mudstone in the vertical depth ranging of 0−6.5 m from the bottom interface of the alluvial layer is greater than 15%. The mudstone exposed to water features muddy disintegration, broken rock fragments and mud blocks, which means it is good water-proof performance of effective bridging mining cracks and a protective layer for waterproof coal pillars; as the strength of weathered mudstone below the alluvial layer 0 to 11.4 m is lower than it of the fine gravel aquifer in the lower that of 4.0 MPa, the sand control coal pillar’s protective layer that is greater more than 11.4 m is cannot be entirely composed of weathered mudstone; due to strong resistance to disintegration and lower dry saturated water absorption rate of sandstone, the protective layer cannot be entirely composed of weathered sandstone. The compressive strength of weathered sandstone, when it is higher than 4.0 MPa, can effectively resist the overlying water head pressure.

Although most species have two sexes, multisexual (or multi-mating type) species are also widespread. However, it is unclear how mating-type recognition is achieved at the molecular level in multisexual species. The unicellular ciliate Tetrahymena thermophila has seven mating types, which are determined by the MTA and MTB proteins. In this study, we found that both proteins are essential for cells to send or receive complete mating-type information, and transmission of the mating-type signal requires both proteins to be expressed in the same cell. We found that MTA and MTB form a mating-type recognition complex that localizes to the plasma membrane, but not to the cilia. Stimulation experiments showed that the mating-type-specific regions of MTA and MTB mediate both self- and non-self-recognition, indicating that T. thermophila uses a dual approach to achieve mating-type recognition. Our results suggest that MTA and MTB form an elaborate multifunctional protein complex that can identify cells of both self and non-self mating types in order to inhibit or activate mating, respectively.

In-situ photocatalytic H2O2 production has been receiving increasing attention in recent years for sustainable H2O2 synthesis. Graphitic carbon nitride (g-C3N4) is regarded as one of the most promising semiconductor photocatalysts for H2O2 evolution. Introducing N defects in g-C3N4 has been proved to be an effective strategy to enhance photocatalytic activity. However, the photocatalytic mechanism of the N vacancies is ambiguous and different types of N vacancies in g-C3N4 may exhibit different effects on photocatalytic activity. Herein, we develop a facile sodium persulfate eutectic polymerization method to prepare the g-C3N4 with abundant three coordinate nitrogen (N3C) vacancies. This type of nitrogen vacancy has not been studied in g-C3N4 for photocatalytic H2O2 production. Our results show that the introduction of N3C vacancies in the g-C3N4 successfully broadens the light absorption range, and inhibits the photoexcited charge recombination with enhanced O2 adsorption to promote oxygen activation. The photocatalytic H2O2 evolution from the N3C-rich g-C3N4 is 4.5 times higher than that of the pristine g-C3N4. This study demonstrates a novel strategy to introduce N3C vacancies in g-C3N4, which offers a new method to develop active catalysts for photocatalytic H2O2 evolution.

Background: Hepatic lipid disorder impaired mitochondrial homeostasis and intracellular redox balance, triggering development of non-alcohol fatty liver disease (NAFLD), while effective therapeutic approach remains inadequate. Ginsenosides Rc has been reported to maintain glucose balance in adipose tissue, while its role in regulating lipid metabolism remain vacant. Thus, we investigated the function and mechanism of ginsenosides Rc in defending high fat diet (HFD)-induced NAFLD. Methods: Mice primary hepatocytes (MPHs) challenged with oleic acid & palmitic acid were used to test the effects of ginsenosides Rc on intracellular lipid metabolism. RNAseq and molecular docking study were performed to explore potential targets of ginsenosides Rc in defending lipid deposition. Wild type and liver specific sirtuin 6 (SIRT6, 50721) deficient mice on HFD for 12 weeks were subjected to different dose of ginsenosides Rc to determine the function and detailed mechanism in vivo. Results: We identified ginsenosides Rc as a novel SIRT6 activator via increasing its expression and deacetylase activity. Ginsenosides Rc defends OA&PA-induced lipid deposition in MPHs and protects mice against HFD-induced metabolic disorder in dosage dependent manner. Ginsenosides Rc (20mg/kg) injection improved glucose intolerance, insulin resistance, oxidative stress and inflammation response in HFD mice. Ginsenosides Rc treatment accelerates peroxisome proliferator activated receptor alpha (PPAR-α, 19013)-mediated fatty acid oxidation in vivo and in vitro. Hepatic specific SIRT6 deletion abolished ginsenoside Rc-derived protective effects against HFD-induced NAFLD. Conclusion: Ginsenosides Rc protects mice against HFD-induced hepatosteatosis by improving PPAR-α-mediated fatty acid oxidation and antioxidant capacity in a SIRT6 dependent manner, and providing a promising strategy for NAFLD.

Abstract Background Spinal cord injury (SCI) is a debilitating event that often results in long‐term physical damage, disability, and a significant impact on a patient's quality of life. Past research has noted an age‐dependent decline in regeneration post‐SCI. This study seeks to identify potential biomarkers and enriched pathways in young and aged SCI mouse models. Methods We retrieved the microarray data of spinal cord samples from SCI mice and control mice from the Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) were identified using the R software and the Linear Models for Microarray Data (limma) package. The Gene Set Enrichment Analysis (GSEA) determined enrichment differences among gene sets. The Weighted Gene Co‐expression Network Analysis (WGCNA) pinpointed clinically significant modules and hub genes in SCI. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis was employed to uncover significantly related pathways of crucial genes in SCI. Results We found 2560 DEGs in the young SCI group, comprised of 1733 upregulated RNAs and 827 downregulated RNAs. In the aged SCI group, 3048 DEGs were revealed including 1856 upregulated and 1192 downregulated genes. The GSEA revealed 12 enriched signaling pathways in the young SCI group, such as IL6/JAK/STAT3 signaling, interferon alpha response, and interferon gamma response, and 21 signaling pathways in the aged SCI group such as IL6/JAK/STAT3 signaling, E2F targets, and angiogenesis‐related pathways. The WGCNA identified the turquoise module as significantly associated with the clinical traits of both young and aged SCI, and revealed 3181 hub genes. Ultimately, 1858 significant genes in SCI were found, with associated signaling pathways including epithelial‐mesenchymal transition (EMT), interferon gamma response, and KARS signaling. Conclusion Our study explored the RNA expression patterns and enriched signaling pathways in young and aged SCI mice. These findings may provide potential biomarkers for targeted SCI therapy.

Study region: Shandong Province in China. Study focus: Multi-water supply can alleviate regional water scarcity. However, the regional water usage weakens ecological water requirements owing to cost management. And the simplified optimal allocation model usually considers regional external water purchase cost or departmental water consumption cost individually, while neglecting differences of multi-water sources in water quality, quantity, and overall costs, resulting in unreasonable utilization. The present work calculated both rigid and flexible water requirements based on different ecological objectives. Meanwhile, the improved model considering water quantity, cost, and quality was constructed, and in comparison to simplified model and actual allocation scheme. New hydrological insights for the region: Taking Shandong Province in 2020 as a case study, the rigid and flexible ecological water requirements are 1.35 and 1.74 times higher than the actual usage, respectively, reflecting the insufficient actual ecological water use. Compared to the simplified models that do not distinguish water source, the improved model can not only consider departmental water consumption costs, but also regional external water purchase costs, and result in a more reasonable water allocation. Compared to the actual water usage scheme, the present scheme reduces the water usage of the Yellow River and increases the water usage of Eastern Route, resulting in a more reasonable water allocation overall improved efficiency.

In order to overcome the pandemic of COVID-19, messenger RNA (mRNA)-based vaccine has been extensively researched as a rapid and versatile strategy. Herein, we described the immunogenicity of mRNA-based vaccines for Beta and the most recent Omicron variants. The homologous mRNA-Beta and mRNA-Omicron and heterologous Ad5-nCoV plus mRNA vaccine exhibited high-level cross-reactive neutralization for Beta, original, Delta, and Omicron variants. It indicated that the COVID-19 mRNA vaccines have great potential in the clinical use against different SARS-CoV-2 variants.

After the first Earth Orientation Parameters Prediction Comparison Campaign (1st EOP PCC), the traditional method using least-squares extrapolation and autoregressive (LS + AR) models was considered as one of the polar motion prediction methods with higher accuracy. The traditional method predicts individual polar motion series separately, which has a single input data and limited improvement in prediction accuracy. To address this problem, this paper proposes a new method for predicting polar motion by combining the difference between polar motion series. The X, Y, and Y-X series were predicted separately using LS + AR models. Then, the new forecast value of X series is obtained by combining the forecast value of Y series with that of Y-X series; the new forecast value of Y series is obtained by combining the forecast value of X series with that of Y-X series. The hindcast experimental comparison results from January 1, 2011 to April 4, 2021 show that the new method achieves a maximum improvement of 12.95% and 14.96% over the traditional method in the X and Y directions, respectively. The new method has obvious advantages compared with the differential method. This study tests the stability and superiority of the new method and provides a new idea for the research of polar motion prediction.

Objective: Spontaneous iliac vein rupture (SIVR) is a rare but life-threatening condition with limited understanding regarding its clinical presentation, pathogenesis, diagnosis, management, and risk factors for mortality. This study aims to address this gap by providing comprehensive insights into SIVR through personal case reports and a systematic review of previous cases. Methods: We detail a case of right SIVR caused by inappropriate positioning of the stent in the inferior vena cava and systematically reviewed previous cases. Logistic regression analysis was used to identify risk factors for mortality. Results: Our SIVR case was successfully managed with percutaneous mechanical thrombectomy and covered stent placement. In the systematic review, 68 patients were included in the analysis with an average age of 62.01 ± 13.25 years; 86.76% were female, 91.17% had left iliac vein rupture, 55.88% presented hemodynamic instability, 76.47% had lower abdomen or iliac fossa pain, 67.64% had deep venous thrombosis (DVT), and 32.35% had May‒Thurner syndrome (MTS). The mortality rates of conservative treatment and open surgery were 2.94% and 17.65%, respectively. All 12 patients receiving endovascular treatment survived. The factors associated with a worse outcome were younger age (52.86 ± 12.96 years, OR: 1.085, 95% CI: 1.002–1.174) and SIVR patients without DVT (OR:10.111, 95% CI: 1.637–62.443). Conclusion: This first systematic review on SIVR shows that SIVR should be highly suspected in elderly females who develop lower extremity DVT and concurrent lower abdominal pain, particularly those with a retroperitoneal mass and unstable hemodynamics. Thrombosis secondary to MTS may be the main cause of SIVR. Angiography and endovascular therapy should be prioritized for DVT patients with unexplained retroperitoneal hematoma. This study classifies SIVR into two types: iliac vein rupture alone and iliac vein rupture with DVT. These findings provide critical insights for clinicians to accurately diagnose and manage SIVR, thereby improving patient outcomes.

COVID-19 has adversely affected public access to public green spaces. As a means of interacting with nature, parks and green spaces are an important aspect of residents' daily lives. In this study, the focus is on new digital solutions, such as the experience of painting in virtual natural settings through virtual reality technologies. This study examines factors that affect the user's perceived playfulness and continuance intention to paint in a virtual environment. A total of 732 valid samples were collected through a questionnaire survey, and a theoretical model was developed through structural equation model by analyzing attitude, perceived behavioral control, behavioral intention, continuance intention, and perceived playfulness. Results demonstrate that perceived novelty and perceived sustainability increase the positive attitude of users toward VR painting functions, whereas perceived interactivity and aesthetics have no impact on it within VR painting context. As users use VR painting, they are more concerned about time and money than equipment compatibility. This makes resource facilitating conditions a more influential factor for perceived behavior control than technology facilitating conditions.

In this paper, polylactide (PLA)/wood flour (WF)/calcium carbonate (CC) composites with different content of poly(maleic anhydride -co- vinyl acetate) (PMAVA) were prepared. PMAVA was obtained by radical solution polymerization and identified by FTIR. The torque - time curves showed that the balance torques of PLA/WF/CC composites with PMAVA were improved by the adding of PMAVA. SEM photographs showed that after the addition of PMAVA, the compatibility and interfacial adhesion between the filler (CC or WF) and PLA in each composite were increased. DSC heating curves showed that two cold crystallization temperature (Tcc) peaks appeared in each PLA/WF/CC composite, and the Tcc peak at high temperature and Tm peak in the composite containing PMAVA were split into two peaks, showing that the effects of WF and CC on the crystal of PLA were uncorrelated, and the presence of PMAVA decreased the crystal integrity of PLA. The tensile test results showed that the tensile strength and modulus of the composite were increased with the addition of PMAVA, and the composite with 0.5 wt% of PMAVA owned the best tensile performance. The hydrolysis results showed that the use of low content of PMAVA in the composite was beneficial to improve its hydrolysis rate.

This article focuses on the psychocultural perceptions and social interactions among a sample of 58 Chinese immigrant women in the Maricopa County, the metro area of the city of Phoenix, Arizona, and the manner in which they are able to negotiate multiple identity markers that in part influence and define their capacity to achieve and maintain self-referential growth. The sample of Chinese women living in the Phoenix area not only apply the metaphor of the phoenix to themselves, but also reference this mythical bird in their social media ID, clubs names, and themed events, and include it in oral traditions passed on to children. In comparison, they reject, negotiate, or resist the stigma and stereotypes attached to the “dragon” symbol which often convey qualities of overpowering and irrational oppression. Instead, they associate themselves with the heuristic of the phoenix as a tool for self-empowerment, virtue, well-being, and ethnic- self-representation. These spontaneous reconstructions of ethnic symbols and metaphors based on traditional cultural consensus allowed immigrants to develop cultural self-confidence because they believed that they had eliminated the possibility of discrimination, and eventually contribute to feasible solutions of silent symbolic violence.

Microbial communities are essential components of aquatic ecosystems and are widely employed for the detection, protection, and restoration of water ecosystems. The polyurethane foam unit (PFU) method, an effective and widely used environmental monitoring technique, has been improved with the eDNA-PFU method, offering efficiency, rapidity, and standardization advantages. This research aimed to explore the colonization process of microbial communities within PFUs using eDNA-PFU technology. To achieve this, we conducted ten-day monitoring and sequencing of microbial communities within PFUs in a stable and controlled artificial aquatic ecosystem, comparing them with water environmental samples (eDNA samples). Results showed 1065 genera in eDNA-PFU and 1059 in eDNA, with eDNA-PFU detecting 99.95% of eDNA-identified species. Additionally, the diversity indices of bacteria and eukaryotes in both methods showed similar trends over time in the colonization process; however, relative abundance differed. We further analyzed the colonization dynamics of microbes in eDNA-PFU and identified four clusters with varying colonization speeds. Notably, we found differences in colonization rates between bacteria and eukaryotes. Furthermore, the Molecular Ecological Networks (MEN) showed that the network in eDNA-PFU was more modular, forming a unique microbial community differentiated from the aquatic environment. In conclusion, this study, using eDNA-PFU, comprehensively explored microbial colonization and interrelationships in a controlled mesocosm system, providing foundational data and reference standards for its application in aquatic ecosystem monitoring and beyond.

Remote ischemic perconditioning (RIPerC) could improve neuronal damage and inhibit inflammation and apoptosis. We conducted an in-depth exploration of the protective mechanism of RIPerC in cerebral ischaemia injury. In this study, a middle cerebral artery occlusion (MCAO) mouse model was built. According to whether to undergo RIPerC treatment and the duration of cerebral infarction, mice were divided into 5 groups: Sham group, MCAO 3.0 h group, MCAO 4.5 h group, MCAO 3.0 h + RIPerC group, and MCAO 4.5 h + RIPerC group. Overexpressed or silenced miR-153-5p was transfected into the cells to analyse the effects of oxygen-glucose deprivation (OGD) treatment on Neuro-2a cell viability, apoptosis, and related gene expressions by performing quantitative real-time polymerase chain reaction (qRT-PCR), MTT assay, flow cytometry, and Western blot. Bioinformatics analysis, qRT-PCR, dual-luciferase experiment, and RNA immunoprecipitation (RIP) were used to screen and verify the miRNA and downstream mRNA-targeted Toll-like receptor 4 (TLR4). The rescue test further verified the effects of the above target genes and miR-153-5p on the apoptosis of OGD-injured cells, apoptosis-related proteins, and the p65/IkBa pathway. The plasma levels of miR-153-5p in 68 patients with ischaemic stroke were detected within 6 hours of onset, and the patients were followed up for 3 months. We found that, in in vivo studies, RIPerC treatment inhibits cerebral infarction volume and neurological damage, and promotes the expression of miR-153-5p in the MCAO animal model. The expression of miR-153-5p in OGD cells was inhibited, and its upregulation protected Neuro-2a cells. TLR4 was predicted to be the target gene of miR-153-5p and could offset the effect of miR-153-5p mimic on OGD cell protection after up-regulating TLR4. TLR4 overexpression promoted the activation of OGD on the p65/IkBa pathway. Compared with the high plasma miR-153-5p group, the 3-month overall survival rate of patients with ischaemic stroke in the low plasma miR-153-5p group was significantly lower (c2 = 5.095, p = 0.024). In conclusion, RIPerC intervention inhibits the damage caused by cerebral ischaemia partially through the miR-153-5p/TLR4/p65/IkBa signalling pathway.

Abstract Background Osteoarthritis (OA) is a multifaceted condition that affects both the subchondral bones and the articular cartilage. Animal models are widely used as an effective supplement and simulation for human OA studies in investigating disease mechanisms and pathophysiology. This study is aimed to evaluate the temporal changes of bone and cartilage in surgically and chemically induced osteoarthritis using micro-computed tomography and histology. Methods Thirty rabbits underwent either anterior cruciate ligament transection (ACLT) procedure or injected intraarticularly with monosodium iodoacetate (MIA, 8 mg) at the right knee joint. The subchondral bones were scanned via micro-CT, and articular cartilage was assessed histologically at 4-, 8- and 12-week post-induction. Results Based on bone micro-architecture parameters, the surgically induced group revealed bone remodelling processes, indicated by increase bone volume, thickening of trabeculae, reduced trabecular separation and reduced porosity. On the other hand, the chemically induced group showed active bone resorption processes depicted by decrease bone volume, thinning of trabeculae, increased separation of trabecular and increased porosity consistently until week 12. Histologically, the chemically induced group showed more severe articular cartilage damage compared to the surgically induced group. Conclusions It can be concluded that in the ACLT group, subchondral bone remodelling precedes articular cartilage damage and vice versa in the MIA group. The findings revealed distinct pathogenic pathways for both induction methods, providing insight into tailored therapeutic strategies, as well as disease progression and treatment outcomes monitoring.

Additionally, NPCs were found to be effective in improving learning outcomes through emotional support.DiscussionAn interactive education system may be able to enhance the perceived playfulness of learning in order to enhance the learning experience.

Protists, which include eukaryotic algae and protozoa, are crucially important for biogeochemical cycling and the aquatic food web but are sensitive to environmental fluctuations. Despite investigations in the 1970s, protist diversity and biogeographic characteristics in the Tibetan Plateau are poorly understood. Within the framework of a regional survey of protist diversity, we used high-throughput amplicon sequencing to investigate the composition and spatiotemporal turnover of protist communities along the Yarlung Zangbo River, a representative aquatic habitat of the Tibetan Plateau. In total, we detected 9,959 amplicon sequence variants in protist communities along the river and associated lakes and wetlands. The alpha diversity of protists was highest in spring and declined in the autumn. Moreover, alpha diversity was much higher in the middle and upstream reaches and tributaries than in the connected lakes and wetlands. We discovered spatiotemporal heterogeneity in the composition of protist communities across the seasons. Bacillariophyta predominated in protist communities in cold seasons (spring and autumn) and colder (high-altitude) areas, whereas Ciliophora and Cercozoa species predominated in the warm season (summer) and warmer (low-altitude) areas. Of the environmental factors, altitude, turbidity and salinity correlated with spatiotemporal dissimilarities among protist communities. The breadth of the habitat niche was restricted by turbidity and altitude in river areas and by salinity in lake and wetland areas. Co-occurrence network analysis suggested that protist communities in the Yarlung Zangbo River were substantially more complex in the warm season than in the cold seasons, and that amplicon sequence variants belonging to Ochrophyta, Chlorophyta, and Ciliophora contributed greatly to the stability of the co-occurrence network. In summary, this study provides a comprehensive overview of the spatiotemporal dynamics of protist diversity in the Yarlung Zangbo River and improves our understanding of the environmental adaptation of protists living in aquatic habitats of the Tibetan Plateau.

Mobile interactive video advertising is a relatively new type of media. A combination of interactivity and narrative is used to tell the video story to the audience. This study studied in three steps on mobile interactive video advertising to determine the design basis that should be paid attention to. Firstly, the research framework is proposed through expert interview and qualitative coding of the interview text. Secondly, research samples are made through surveys and conference discussions. The samples were designed into 30 types containing various interactive narrative and involvement. Finally, the qualitative results are verified in a quantitative way. The results show that in order to improve the audience's purchase intention, more attention should be paid to interactive narrative, subjective video quality assessment, immersion, satisfaction, and product involvement.

With the rapid spread of mobile devices and the Internet, mobile interactive video advertising has become an increasingly popular means of accessing advertising information for a large number of users. Interactive narratives are advertisements that require collaboration between consumers and designers to complete the story. Interactive narratives influence marketing impact and the advertising experience. Building on previous research, this study delves deeper into the design methods of interactive narratives for mobile video advertisements. We developed various interactive narrative samples by controlling video quality parameters, content, and product involvement, and then measured consumer perceptions of these samples in a laboratory environment. The results indicate that six design methods for interactive narratives foster positive perceptions, immersion, and satisfaction in advertisements with low product involvement. For ads with a high degree of product involvement, two design methods can achieve positive consumer perceptions of interactive narratives. This study offers insights for businesses and interaction designers aiming to advance the commercial use of mobile interactive video advertising. At the same time, we propose a design method for mobile interactive video advertising that can also serve as an entry point for theoretical research on interactive narratives.

This study aims to explore the utilization of user-generated content for product improvement and decision-making processes. In the era of big data, the channels through which enterprises obtain user feedback information are transitioning from traditional methods to online platforms. The original data for this study were obtained from customer reviews of cordless hairdryers on JD.com. The specific process is as follows: First, we used the Python Requests package to crawl 20,157 initial comments. Subsequently, the initial data were cleaned, resulting in 1405 valid comments. Next, the cleaned and valid comments were segmented into Chinese words using the HanLP package. Finally, the Latent Dirichlet Allocation (LDA) method was applied for topic modeling. The visualization of the topic clustering was generated using pyLDAvis, and three optimal topics were identified. These topics were named “User Experience”, “Product Evaluation”, and “Product Features”, respectively. Through data analysis and expert consultation, this study developed product design improvement strategies based on online reviews and verified the validity of the developed cordless hairdryer design index system through a questionnaire survey, providing practical references and innovative theoretical foundations for future product design assessments.

BackgroundTraditional open gastric cancer surgery has evolved from porous to reduced-hole, single-hole, or even natural cavity surgery to laparoscopic surgery, due to the continuous development of minimally invasive concepts and medical technologies, as well as awareness for the concept of rapid recovery. Conventional laparoscopic radical gastrectomy is quite mature in age at the moment, but how to progress to minimally invasive surgery without increasing the difficulty of surgery while ensuring clinical safety and feasibility is worth further investigation. Therefore, the clinical safety and feasibility of reduced port laparoscopic radical gastrectomy were assessed in this study.MethodsInformation on the clinical data of patients undergoing laparoscopic radical gastric cancer surgery in a single centre between May 2020 and May 2022 was collected, and a total of 232 patients were included in this study according to the study protocol design. The clinical data of 232 patients with gastric cancer treated by two different surgical methods, namely, reduced port laparoscopic surgery (RPLS) or conventional laparoscopic surgery (CLS), were retrospectively analysed. The intraoperative indices, postoperative pathological indices, and short-term postoperative complications (within 30 days) of the two different surgical methods were evaluated, as well as the surgical methods’ feasibility and short-term postoperative recovery effect.ResultsThere was no significant difference between the general data of patients with RPLS and CLS (P > 0.05). Compared with CLSG, the operation time, digestive tract reconstruction time and lymph node dissection time of RPLSG are shorter. The intraoperative blood loss was less, and the incision was minimally invasive (P 0.05).ConclusionsThe treatment of gastric cancer with RPLS has good safety, feasibility and short-term postoperative effects, which is in line with the implementation of the modern concept of rapid rehabilitation surgery.

BackgroundThe method of operation and the range of resection for Siewert II adenocarcinoma of the esophagogastric junction (AEG) remain controversial. This study aims to evaluate the safety, feasibility, and short-term postoperative effect of total laparoscopic versus laparoscopic-assisted transabdominal posterior mediastinal digestive tract reconstruction in the treatment of Siewert II AEG.MethodsTotal laparoscopic or laparoscopic-assisted gastrointestinal reconstruction through abdominal posterior mediastinum was performed in 108 patients with Siewert II AEG from October 2017 to February 2019. This study evaluated the loss of intraoperative blood, the number of lymph nodes, the marginal of the tumor, short-term postoperative complications (within 30 days), the rate of survival at follow-up, and the economic cost, feasibility, and effect of short-term postoperative recovery for patients who received these two operations.ResultThere were no significant differences in general data between the total laparoscopic group and the laparoscopic-assisted group (P > 0.05). However, the total laparoscopic group cost more time on the surgical procedure and digestive tract reconstruction, lost less intraoperative blood, and had more mediastinal lymph nodes compared with the laparoscopic-assisted group (P 0.05).ConclusionsThe safety, feasibility, and short-term effect of total laparoscopic transabdominal posterior mediastinal digestive tract reconstruction in the treatment of Siewert II AEG were better than those for the laparoscopic-assisted group.

Hand tool design should integrate the concept of Inclusive Design to be accessible to most users. However, current Inclusive Design strategies of product development are mostly used in post-design evaluation. The retention of inclusive properties in product when new functions are incorporated is essential. Fine operation-aid screwdrivers are designed according to user requirements to address frequently-encountered problems when using screwdrivers namely-insufficient lighting and difficulty in properly installing screws respectively. TRIZ method is applied, comprised the improving parameters solving the problems and worsening parameters which prevents the original inclusive design factors from being damaged into the contradiction matrix, and obtains a set of innovation principles. Eight experts were consulted for their design ideas and developed two fine operation-aid screwdrivers embracing the concept of Inclusive Design. Furthermore, factors regarding the two major operating problems were added to an existing hand tools Inclusive Design Scale. After correlation analysis, the inclusive fine operation-aid screwdriver evaluation scale was established. In addition, two more screwdrivers were selected with the same functions and high reviews on the market as control samples, and 39 users were recruited using a quota sampling strategy to participate in Inclusive Design evaluations. The results revealed that the fine operation-aid screwdrivers evidently solved the two major operating problems in terms of the five dimensions including functionality, comfort, professionality, safety, and usability in the inclusive fine operation-aid screwdriver evaluation scale, thereby affirming the rationality and reliability of our hand tool development approach.

Ciliates contain two kinds of nuclei: the germline micronucleus (MIC) and the somatic macronucleus (MAC) in a single cell. The MAC usually have fragmented chromosomes. These fragmented chromosomes, capped with telomeres at both ends, could be gene size to several megabases in length among different ciliate species. So far, no telomere-to-telomere assembly of entire MAC genome in ciliate species has been finished. Development of the third generation sequencing technologies allows to generate sequencing reads up to megabases in length that could possibly span an entire MAC chromosome. Taking advantage of the ultra-long Nanopore reads, we established a simple strategy for the complete assembly of ciliate MAC genomes. Using this strategy, we assembled the complete MAC genomes of two ciliate species Tetrahymena thermophila and Tetrahymena shanghaiensis, composed of 181 and 214 chromosomes telomere-to-telomere respectively. The established strategy as well as the high-quality genome data will provide a useful approach for ciliate genome assembly, and a valuable community resource for further biological, evolutionary and population genomic studies.

Abstract Background The cell wall and chloroplast are two fundamental structures determining plant mechanical strength and grain yield. Therefore, understanding mechanisms that improve plants’ ability to develop a robust cell wall and well-developed chloroplast is of utmost importance for agricultural activities. Results In this study, we report the functional characterization of a novel rice mutant, brittle stem and zebra leaf (bz1), which displays altered cell wall composition and collapsed chloroplast membrane. Molecular and biochemical analysis revealed that BZ1 encodes a functional UDP-galactose/glucose epimerase (UGE) and is ubiquitously expressed with higher expression in stem and leaf tissues. Multiple techniques analyses, including immunoblots, immuno-gold, and cryogenic scanning electron microscopy, demonstrated a significantly impaired glycosylation of arabinogalactan proteins (AGPs) and disordered cellulose microfibril deposition in bz1. Lipid profiling assay showed that the amount of monogalactosyldiacylglycerols (MGDG), a major chloroplast membrane glycolipid, was significantly decreased in bz1. Taken together, these results strongly demonstrate that BZ1 participates in UDP-galactose supply for the sugar chains biosynthesis of AGPs and MGDG, which thereby, respectively, results in altered cell wall and abnormal chloroplast development. Due to inferior mechanical strength and reduced photosynthesis, bz1 plants displayed detrimental agronomic traits, whereas BZ1 overexpressing lines showed enhanced plant growth. Transcriptome analysis of stems and leaves further showed that numerous key genes involved in AGPs biosynthesis and photosynthesis metabolism were substantially suppressed in bz1. Conclusions Our finding identifies BZ1 as a dual-targeting UGE protein for glycosylation of AGPs and MGDG and suggests a strategy for breeding robust elite crops.

Oxidative stress-associated endothelial damage is the initiation factor of cardiovascular disease, and protein posttranslational modifications play critical roles in this process. Bcl-2-associated athanogene 3 (BAG3) is a molecular chaperone regulator of the BAG family, which interacts with various proteins and influences cell survival by activating multiple pathways. BAG3 undergoes posttranslational modifications; however, research evaluating BAG3 acetylation and its regulatory mechanism is lacking. In addition, the interacting protein and regulatory mechanism of BAG3 in oxidative stress-associated endothelial damage remain unclear. Here, key molecular interactions and protein modifications of BAG3 were identified in oxidative stress-associated endothelial damage. Endothelial-specific BAG3 knockout in the mouse model starkly enhances oxidative stress-associated endothelial damage and vascular remodeling, while BAG3 overexpression in mice significantly relieves this process. Mechanistically, poly(ADP-ribose) polymerase 1 (PARP1), causing oxidative stress, was identified as a novel physiological substrate of BAG3. Indeed, BAG3 binds to PARP1's BRCT domain to promote its ubiquitination (K249 residue) by enhancing the E3 ubiquitin ligase WWP2, which leads to proteasome-induced PARP1 degradation. Furthermore, we surprisingly found that BAG3 represents a new substrate of the acetyltransferase CREB-binding protein (CBP) and the deacetylase Sirtuin 2 (SIRT2) under physiological conditions. CBP/SIRT2 interacted with BAG3 and acetylated/deacetylated BAG3's K431 residue. Finally, deacetylated BAG3 promoted the ubiquitination of PARP1. This work reveals a novel regulatory system, with deacetylation-dependent regulation of BAG3 promoting PARP1 ubiquitination and degradation via enhancing WWP2, which is one possible mechanism to decrease vulnerability of oxidative stress in endothelial cells.

Carbohydrates are the main source of nutrition for B. coli, supplying energy for cell growth and development. The research aimed at investigating the mechanism of starch on the growth and replication of B. coli. Single-cell separation was used to isolate single trophozoites of B. coli under a stereomicroscope, transcriptomic profiling was conducted based on the SMART-seq2 single-cell RNA-seq method. Comparative genomic analysis was performed on B. coli and eight other ciliates to obtain specific and expanded gene families of B. coli. GO and KEGG enrichment analysis were used to analyze the key genes of B. coli under the action of starch in the present study. The results of single-cell RNA-seq depicts starch affected the growth and replication of B. coli in two ways: (1) the cell cycle was positively promoted by the activation of the cAMP/PKA signaling pathway via glycolysis; (2) the cell autophagy was suppressed through the PI3K/AKT/mTOR pathway. Genes involved in endocytosis, carbohydrate utilization, and the cAMP/PKA signaling pathway were highly enriched in both specific and expanded gene families of B. coli. Starch can be ingested and hydrolyzed into glucose, in turn affecting various biological processes of B. coli. The molecular mechanism of the effect of starch on the growth and proliferation of B. coli by promoting cell cycle and inhibiting the autophagy of trophozoites has been elucidated in our study.

Background Animal models are significant for understanding human osteoarthritis (OA). This study compared the synovial fluid proteomics changes in surgical and chemical induced OA models. Methods Thirty rabbits either had anterior cruciate ligament transection (ACLT) procedure or injected intra-articularly with monosodium iodoacetate (MIA, 8 mg) into the right knee. The joints were anatomically assessed, and the synovial fluid proteins analyzed using two-dimensional polyacrylamide gel electrophoresis (2DGE) and MALDI TOF/TOF mass spectrometry analysis at 4, 8 and 12 weeks. The proteins’ upregulation and downregulation were compared with control healthy knees. Results Seven proteins (histidine-rich glycoprotein, beta-actin-like protein 2 isoform X1, retinol-binding protein-4, alpha-1-antiproteinase, gelsolin isoform, serotransferrin, immunoglobulin kappa-b4 chain-C-region) were significantly expressed by the surgical induction. They characterized cellular process (27%), organization of cellular components or biogenesis (27%), localization (27%) and biological regulation (18%), which related to synovitis, increased cellularity, and subsequently cartilage damage. Three proteins (apolipoprotein I-IV precursor, serpin peptidase inhibitor and haptoglobin precursor) were significantly modified by the chemical induction. They characterized stimulus responses (23%), immune responses (15%), biological regulations (15%), metabolism (15%), organization of cellular components or biogenesis (8%), cellular process (8%), biological adhesions (8%) and localization (8%), which related to chondrocytes glycolysis/death, neovascularization, subchondral bone necrosis/collapse and inflammation. Conclusions The surgical induced OA model showed a wider range of protein changes, which were most upregulated at week 12. The biological process proteins expressions showed the chemical induced joints had slower OA progression compared to surgical induced joints. The chemical induced OA joints showed early inflammatory changes, which later decreased.
