Silicon‐based materials are regarded as the most promising negative electrode materials for next‐generation high‐energy lithium ion batteries due to their high theoretical capacity, low lithiation plateau and low cost, but they still suffer from dramatic volume variation during charge/discharge and sluggish kinetics, substantially restricting their practical applications. Recently, 2D MXenes are demonstrated to improve the electrochemical performances of Si‐based materials owing to their metallic conductivity, large surface aspect ratio, rich surface chemistry, good mechanical strength, etc. Serving as conductive host, MXenes can alleviate the volume expansion of Si during lithiation, enhance the electronic conductivity of the composite and facilitate the electron/ion transport, thus making the resultant Si/MXene composites exhibit excellent performance as negative electrodes of secondary batteries. Herein, an overview toward the recent advances of Si/MXene nanocomposites is presented for lithium storage applications. The preparation strategies and structural characteristics of the Si/MXene nanocomposites are introduced. The applications of diverse Si/MXene nanostructures such as powders, 3D architectures, films and fibers, in lithium ion batteries with corresponding structure‐performance relationships are discussed. Finally, the challenges and perspectives for the future research of Si/MXene nanocomposites are highlighted, aiming to provide a fundamental reference in designing advanced Si/MXene nanocomposites for high‐energy lithium ion batteries. Deep learning has demonstrated notable success in mechanical signal processing with a large amount labelled data. However, the systems of the Jiaolong deep‐sea submersible prone to malfunction are typically diverse, due to the high complexity of its structure and operational environment. Consequently, this diversity gives rise to variations in the types of sensor signals and their associated data distributions that require analysis. Unsupervised domain adaptation (UDA) uses transferable knowledge derived from the source domain and applies it to an unlabelled target domain in order to improve the reusability of pre‐existing models and data. Inspired by the diffusion models that have the robust capabilities to transform data distributions across a large gap, we propose a novel diffusion‐based unsupervised domain adaptation (diffusion‐UDA) model, which leverages contrastive learning to alleviate the challenges of cross‐domain analysis for fault diagnosis within different systems of the deep‐sea submersible. Chimeric antigen receptor (CAR) T-cell-associated coagulopathy can cause bleeding events. To explore risk factors for hemorrhage after CAR T-cell therapy, we retrospectively analyzed routine indicators in 56 patients with non-Hodgkin lymphoma and B-cell acute lymphoblastic leukemia who received anti-CD19 CAR T-cell therapy. Disturbance of coagulation occurred mainly within one month post infusion, especially on day 7 and 14. The cumulative incidence of bleeding events within one month was 32.8%, with the median onset of 7 (range, 0–28) days. All bleeding events were grade 1–3. Patients who experienced bleeding events within one month had longer prothrombin time, higher IL-6, higher IL-10, and lower platelets before lymphodepletion. There were also correlations among coagulation-, inflammatory-, and tumor burden-related markers. Multi-variate analysis showed IL-10 (> 7.98 pg/mL; adjusted odds ratio [OR], 13.84; 95% confidence interval [CI], 2.03–94.36; P = 0.007) and the endothelial activation and stress index (EASIX, defined as dehydrogenase [U/L] × creatinine [mg/dL] / platelets [×109 cells/L]; >7.65; adjusted OR, 7.06; 95% CI, 1.03–48.23; P = 0.046) were significant risk factors for bleeding events. IL-10 plus the EASIX defined three risk groups for bleeding events with cumulative incidence of 100% (hazard ratio [HR], 14.47; 95% CI, 2.78–75.29; P < 0.0001), 38.5% (HR, 3.68; 95% CI, 0.82–16.67; P = 0.089), and 11.8% (reference), respectively. Objective: This work was aimed to analyze the effect of vitamin energy drink on muscle fatigue by surface electromyography (SEMG) and ultrasonic bioimaging (USBI). Methods: 20 healthy men were selected to do increasing load fatigue test. Surface electromyographic signals and ultrasonic biological images were collected based on wavelet threshold function with improved thresholds. Time domain and frequency domain characteristic integrated electromyography (IEMG), root mean square amplitude (RMS), average power frequency (MPF), and surface and deep muscle morphological changes were analyzed. Hemoglobin concentration (HB), red blood cell number (RBC), mean volume of red blood cell (MCV), blood lactic acid (BLA), malondialdehyde (MDA), and phosphocreatine kinase (CK) were measured. Results: 1) the Accuracy (94.10%), Sensitivity (94.43%), Specificity (93.75%), and Precision (94.07%) of the long and short-term memory (LSTM) specificity for muscle fatigue recognition were higher than those of other models. 2) Compared with the control group, the levels of BLA, MDA, and CK in the experimental group were decreased and HB levels were increased after exercise (P < 0.05). 3) IEMG and RMS of the experimental group were higher than those of the control group, and increased with time (P < 0.05). 4) The mean amplitude of the response signal decreased with time. Compared with the control group, the surface muscle thickness, deep muscle thickness, total muscle thickness, contrast, and homogeneity (HOM) decreased in the experimental group; while the angular second moment (ASM) and contrast increased, showing great differences (P < 0.05). Conclusion: Surface electromyographic signal and ultrasonic biological image can be used as auxiliary monitoring techniques for muscle fatigue during exercise. At high cutting speeds, the internal microstructure of the difficult-to-machine material Inconel 718 will change. Alterations in the internal microstructure lead to macroscopic deformations, which in turn result in the formation of adiabatic shear bands. In order to better study the formation of adiabatic shear bands during cutting Inconel 718, cutting experiments, SHPB experiments, and finite element simulations were carried out. The stored energy model for adiabatic shear bands was developed by combining the overall dislocation density and the variation of the stress field in this paper. Based on the strain gradient theory, the subroutine was compiled for secondary development to achieve multi-scale finite element simulation that can visualize the stored energy. The experiment and simulation results were combined to derive the change in stress, temperature, and overall dislocation density and to analyze the adiabatic shear bands storage energy based on the change. The analytical results are also combined with dynamic recrystallization and periodic fracture theories to characterize the formation mechanism of adiabatic shear bands. It was found that the overall dislocation density, stress, and adiabatic shear bands storage energy follow the same trend, while the temperature and shear zone storage energy follow opposite trends. Electric vehicles have been considered into effective solutions to address energy problems in urban traffic systems for their remarkable performance in energy costs and carbon emissions reduction. However, the energy-saving effect of electric vehicles in urban traffic systems is restricted due to the complexities arising from mixed traffic conditions. To improve energy efficiency, this paper proposes an eco-driving-based mixed vehicular platoon control model for successive signalized intersections with connected and autonomous vehicles (CAVs), connected vehicles (CVs) and manned vehicles (MVs). Firstly, the dynamics model and spacing policy are analyzed, then the optimal platoon size and speed are calculated according to different traffic scenarios. Secondly, the mixed vehicular platoon control algorithm is established by considering traffic timing, energy consumption and string stability. Thirdly, an actual eco-driving-based platoon control system for tested electric vehicles is designed. The field test results demonstrate that all platoons can pass through intersections during green phase with minimum energy consumption, meanwhile the string stability can be guaranteed. Diabetes mellitus (DM) is a disease that affects millions of individuals worldwide and is characterized by abnormal glucose metabolism that can induce severe damage to numerous organs throughout the body. Sex differences have been demonstrated in a number of factors associated with diabetes and its complications, such as diabetic kidney disease and diabetic liver disease. To investigate the sex differences in DM further, the changes in the weight, food and water intake, and blood sugar of mice were recorded for 8 weeks in the present study. Hematoxylin and eosin staining, Masson's trichrome staining and transmission electron microscopy were used to observe the pathological changes of liver and kidney tissues. There is no significant difference in the water intake and blood glucose concentration between db/db female and male mice was observed. However, sex differences in liver and kidney damage including glomerular injury and hepatic fibrosis were found. In conclusion, the present study characterized the features of liver and kidney damage in db/db mice and indicated that sex differences should be taken into account in experiments using female and male experimental animals. • Epitope molecularly imprinted polymer was prepared by autoxidation of caffeic acid. • High adsorption capacity and selectivity toward ovalbumin were achieved. • The polymer was successfully applied to enrich ovalbumin in egg white samples. Food allergy can lead to severe allergic reactions that are potentially fatal for human, hence the detection of food allergens such as ovalbumin (OVA) is important. In this study, a poly(caffeic acid)-coated epitope molecularly imprinted polymer (EMIP) was prepared by chelation and autoxidation of caffeic acid with hexamethylenediamine. EMIP has not only imprinted cavities highly matched with OVA in size and spatial structure, but also externally abundant hydrophilic groups, resulting in few non-specific binding and good hydrophilicity. With high specificity, significant paramagnetism, and great reusability, EMIP can distinguish OVA from other proteins and selectively enrich OVA in egg white samples, which opens up a promising route to the determination of allergens in food products. Electrochemical micromachining (EMM) has become a popular technology for fabricating microscale structures. To obtain higher machining localization and minor stray corrosion, a new hybrid EMM process combining air assistance and ultrasonic assistance was introduced in this paper. Simulations and experimental investigations were conducted to study the influencing mechanism and the influencing characteristics of the assisting process energies. In the multi-physics model, a reactant transport model is included, and the direct coupling between the assisting processes and the electrochemical process is achieved. Results indicate that, when the two assisting processes are applied separately, the assisting effect tend to act on the entire workpiece surface, either promotes the overall dissolution or inhibits it, usually leading to unsatisfactory results; by combining the two assisting processes, synergistic effect occurs: dissolution in the central area is promoted by the ultrasonic assistance, while stray corrosion in the margin area is hindered by the air assistance. Thus, microstructures with enhanced aspect ratio and reduced stray corrosion can be fabricated. The results also show that the surrounding airflow can produce an electrical insulating effect on both the workpiece and the cathode. When the air pressure is low, an additional concentrating effect on the electric field can be observed; however, a high air pressure leads to a greatly hindered dissolution. When the applied amplitude is appropriate, ultrasonic assistance promotes the electrochemical dissolution through disturbance effect and cavitation phenomena. But if the amplitude is too large, massive cavitation bubbles will inhibit the dissolution greatly. [ABSTRACT FROM AUTHOR]

Abstract Jet electrochemical (Jet-EC) milling, using a movable nozzle as the cathode, can fabricate complicated structures flexibly. With the dissolution restricted to a limited area by the electrolyte jet, high removal localization and good surface quality can be obtained. However, the edge of the machined groove is usually subject to stray corrosion because of the low current density in the edge area. In this paper, it is confirmed that the jet shape, which is determined by the working parameters, has a notable influence on the edge condition. Through experiments and analysis, the interrelation between the working parameters, jet shape and edge condition in Jet-EC groove milling is established. It is found that the stray corrosion on the edge can be reduced significantly by adjusting the jet shape, and the necessary conditions to completely eliminate the stray corrosion are established. Furthermore, based on the conclusions, a groove with both a sharp edge and a smooth surface is fabricated via a two-pass process. Abstract: Very little is known regarding whether structural hubs of human brain networks that enable efficient information communication may be classified into different categories. Using three multimodal neuroimaging data sets, we construct individual structural brain networks and further identify hub regions based on eight widely used graph‐nodal metrics, followed by comprehensive characteristics and reproducibility analyses. We show the three categories of structural hubs in the brain network, namely, aggregated, distributed, and connector hubs. Spatially, these distinct categories of hubs are primarily located in the default‐mode system and additionally in the visual and limbic systems for aggregated hubs, in the frontoparietal system for distributed hubs, and in the sensorimotor and ventral attention systems for connector hubs. These categorized hubs exhibit various distinct characteristics to support their differentiated roles, involving microstructural organization, wiring costs, topological vulnerability, functional modular integration, and cognitive flexibility; moreover, these characteristics are better in the hubs than nonhubs. Finally, all three categories of hubs display high across‐session spatial similarities and act as structural fingerprints with high predictive rates (100%, 100%, and 84.2%) for individual identification. Collectively, we highlight three categories of brain hubs with differential microstructural, functional and, cognitive associations, which shed light on topological mechanisms of the human connectome. Age prediction is an important field in forensic and aging research. Traditional methods used DNA methylation, telomere shortening, and mitochondrial DNA mutations to conduct age prediction models. Sex chromosomes, like the Y chromosome, have a significant role in aging as previously reported in hematopoietic disease and many non-reproductive cancers. Until now, there is no age predictor based on the percentage of loss of Y chromosome (LOY). LOY has been previously revealed to be correlated with Alzheimer's disease, short survival, and higher risk of cancer. The possible correlation of LOY between normal aging was not fully explored. In this study, we conducted age prediction by measuring LOY percentage by droplet digital PCR (ddPCR), based on 232 healthy male samples, including 171 blood samples, 49 saliva samples, 12 semen samples. The age group of samples ranges from 0 to 99 years, with two individuals in almost every single age. Pearson correlation method was performed to calculate the correlation index. The result indicated a correlation index of 0.21 (p = 0.0059) between age and LOY percentage in blood samples, with the regression formula being y = − 0.016823 + 0.001098x. The correlation between LOY percentage and age is obvious only when the individuals were divided into different age groups (R = 0.73, p = 0.016). In the studied saliva and semen samples, p-values of the correlation are 0.11 and 0.20, respectively, showing no significant association between age and LOY percentage in these two biological materials. For the first time, we investigated male-specific age predictor based on LOY. The study showed that LOY in leukocytes can be regarded as a male-specific age predictor for age group estimation in forensic genetics. Electrochemical drilling (ECD) is a useful method to produce small holes with a large aspect ratio on hard-to-machine materials. Recently, instead of using an acid electrolyte, more and more research has focused on neutral salt electrolytes because they are less harmful to the environment. However, more problems have been caused to the removal of waste products, especially in deep-hole drilling. Traditionally, an electrolyte flow with constant pressure control is adopted in ECD, which might result in the poor removal of waste products at depth because of pressure loss. In this paper, an ECD method with a constant electrolyte flow has been proposed to obtain a consistent ability to remove insoluble waste products from machining gap. Experiments of ECD with both constant electrolyte pressure and constant flow are conducted on workpieces of Inconel 718. Machining currents and the profile of machined holes are compared between the two methods. The results show that when a constant electrolyte flow rate is applied, the machining currents are much smoother and the profile of machined holes is also uniform. In a composite beam, transverse web stiffeners are often provided to the steel section at regular intervals along the beam span to eliminate local shear buckling as well as to reduce web distortion in the steel section. These transverse stiffeners are also found to be effective in providing intermediate lateral restraints from the stiff concrete slab to the compressive steel flange, forming a 'discrete inverted U-frame' action so that the resistance of the composite beam against distortional lateral buckling is significantly enhanced. In general, the load carrying capacity of a continuous composite beam depends on a number of factors such as initial imperfections, residual stresses, and material nonlinearity. A general design method to evaluate the contribution of transverse web stiffeners to the capacity of a continuous composite beam, is however yet unavailable.This paper presents an advanced numerical investigation into the structural behaviour of composite beams under the discrete inverted U-frame action. Based on a comprehensive finite element study, the following key parameters affecting the load carrying capacity of the beam are studied: i) the flexural stiffness of the concrete slab, ii) the stiffness of the web stiffeners, iii) the ratio of the depth to thickness of the steel web, and iv) the ratio of the spacing of the web stiffeners to the beam span. It has been verified that in the presence of properly designed web stiffeners, the buckling moment resistances of composite beams will be significantly increased while the lateral deflection of the compression flanges is greatly reduced. Abstract: A comparative study of prestressed continuous steel–concrete composite beams is carried out based on an experimental investigation. Four full-scale continuous composite beams were tested, among which two were two-span beams and two were three-span beams. One of the two-span beams was a conventional non-prestressed composite beam, and the other was a composite beam prestressed with external tendons in both the positive and negative moment regions. One of the three-span composite beams was non-prestressed, and the other was prestressed only in the negative moment regions with external high-strength tendons. The cracking behaviors, distortional lateral and local buckling as well as the load carrying capacity of the beams were investigated experimentally. Full plasticity was developed at the mid-span section of each beam; however, the maximum moments attained at the internal supports were governed by local buckling, which is related to the slenderness of the composite section. It is found that in hogging moment regions, the ultimate moment resistance of a composite beam prestressed with external tendons is governed by either distortional lateral buckling or local buckling, or an interactive mode composed of distortional lateral and local buckling. The results show that exerting prestressing on a continuous composite beam with external tendons increases the degree of the internal force and moment redistribution in the beam. A design proposal based on moment redistribution to evaluate the load carrying capacity of continuous composite beams with external tendons is proposed. [Copyright &y& Elsevier]

Brain extraction (a.k.a. skull stripping) is a fundamental step in the neuroimaging pipeline as it can affect the accuracy of downstream preprocess such as image registration, tissue classification, etc. Most brain extraction tools have been designed for and applied to human data and are often challenged by non-human primates (NHP) data. Amongst recent attempts to improve performance on NHP data, deep learning models appear to outperform the traditional tools. However, given the minimal sample size of most NHP studies and notable variations in data quality, the deep learning models are very rarely applied to multi-site samples in NHP imaging. To overcome this challenge, we used a transfer-learning framework that leverages a large human imaging dataset to pretrain a convolutional neural network (i.e. U-Net Model), and then transferred this to NHP data using a small NHP training sample. The resulting transfer-learning model converged faster and achieved more accurate performance than a similar U-Net Model trained exclusively on NHP samples. We improved the generalizability of the model by upgrading the transfer-learned model using additional training datasets from multiple research sites in the Primate Data-Exchange (PRIME-DE) consortium. Our final model outperformed brain extraction routines from popular MRI packages (AFNI, FSL, and FreeSurfer) across a heterogeneous sample from multiple sites in the PRIME-DE with less computational cost (20 s~10 min). We also demonstrated the transfer-learning process enables the macaque model to be updated for use with scans from chimpanzees, marmosets, and other mammals (e.g. pig). Human brain connectomes include sets of densely connected hub regions. However, the consistency and reproducibility of functional connectome hubs have not been established to date and the genetic signatures underlying robust hubs remain unknown. Here, we conduct a worldwide harmonized meta-connectomic analysis by pooling resting-state functional MRI data of 5212 healthy young adults across 61 independent cohorts. We identify highly consistent and reproducible connectome hubs in heteromodal and unimodal regions both across cohorts and across individuals, with the greatest effects observed in lateral parietal cortex. These hubs show heterogeneous connectivity profiles and are critical for both intra- and inter-network communications. Using post-mortem transcriptome datasets, we show that as compared to non-hubs, connectome hubs have a spatiotemporally distinctive transcriptomic pattern dominated by genes involved in the neuropeptide signaling pathway, neurodevelopmental processes, and metabolic processes. These results highlight the robustness of macroscopic connectome hubs and their potential cellular and molecular underpinnings, which markedly furthers our understanding of how connectome hubs emerge in development, support complex cognition in health, and are involved in disease.A worldwide harmonized meta-connectomic analysis of 5212 healthy young adults across 61 independent cohorts provides consistent, reproducible, and transcriptionally relevant functional connectome hubs in the resting human brain. Rapidly mutating Y-STRs (RM Y-STRs) harbor great potential to distinguish male relatives and achieve male identification. However, forensic applications were greatly limited by the small number of the initially identified 14 RM Y-STRs. Recently, with the emergence of 12 novel RM Y-STRs, an integrated panel named RMplex was introduced, which contains all 26 RM Y-STRs and four fast mutating Y-STRs (FM Y-STRs). To obtain the first data on the mutation rates and father-son differentiation rates of the 30 newly proposed Y-STRs in Chinese populations, we performed an empirical mutation study on 307 DNA-confirmed Chinese paternal pairs. Previously reported mutation rates for 14 RM Y-STRs in Chinese and European populations were pooled and merged with our data. The highest meiosis number for the two groups reached 4771 and 2687, respectively. Five loci showed significant differences between the populations (DYS570, DYS399S1, DYS547, DYS612, and DYF403S1b). For the new panel covering 30 Y-STR loci, our results show extensive differences in the mutation rates between the two populations, as well. 10 RM Y-STR loci showed relatively low mutation rates (10–3–10–2 per meiosis) and 2 FM Y-STR loci had rapid mutation rates (> 10–2 per meiosis) in the Chinese population. Several-fold differences in mutation rates were found in nine Y-STR loci between the Chinese and reference populations, with two loci having significantly higher mutation rates and one locus with a significantly lower mutation rate in the Chinese population (P < 0.05). Eighteen RM Y-STRs (> 10–2 per meiosis), 8 FM Y-STR loci (5×10–3-10–2 per meiosis), 3 moderately mutating Y-STRs (MM Y-STRs, 10–3-5×10–3 per meiosis), and one locus with no observed mutation events were identified in the Chinese population. 40.06% of the Chinese paternity pairs were discriminated with RMplex while only 20.84% with the initial 14 RM Y-STRs, indicating that RMplex is beneficial for distinguishing paternally related males. Impacts of zinc sulphate and cupric chloride on characteristics and antioxidant capacities of glycated products (GPs) derived from casein hydrolysate (CH) and xylose were studied. The browning degree of xylose–CH GPs was the largest at zinc sulphate addition of 15 mg L−1, while its browning degree increased with cupric chloride extended from 0 to 15 mg L−1. Loss of free amino groups, fluorescence intensity and the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging capacity of xylose–CH GPs increased with the increase of cupric chloride addition, but loss of amino groups, fluorescence intensity and DPPH radical scavenging capacity were largest at the zinc sulphate addition of 15 mg L−1. The addition of zinc sulphate and cupric chloride promoted production of higher and smaller molecular substances in xylose–CH GPs and CH. Therefore, zinc sulphate and cupric chloride promoted glycated reaction of xylose and CH and enhanced antioxidant activity of xylose–CH GPs. [ABSTRACT FROM AUTHOR]

Micro-structure on metal surface can be created with high precision and good surface quality by ultrasonic-assisted electrochemical micromachining (USEMM). One of the prevalent material removal mechanisms in ultrasonic machining (UM) is cavitation erosion. However, the mechanism of material erosion is not clear and worth investigating. This study of the mechanical and chemical effects of the ultrasonic vibration on the 6061 aluminum alloy is targeted to reveal the material processing mechanism in USEMM. Based on the built model, the velocity of micro-jet produced near the workpiece surface by ultrasonic cavitation reaches up to 350 m/s when bubble collapses computed by software MATLAB. The impact of micro-jet produces plastic micro-pits on the metal surface and the convex peak around the edge of the pits, which is verified in ABAQUS software. The metallographic microscope and curves of the electrochemical polarization behavior results indicate a significant grain refinement and a marked increase of anodic dissolution current, as well as a weaker resistance than the original workpiece in NaNO3 electrolyte during UM. The recently proposed q-rung picture fuzzy set (q-RPFSs) can describe complex fuzzy and uncertain information effectively. The Hamy mean (HM) operator gets good performance in the process of information aggregation due to its ability to capturing the interrelationships among aggregated values. In this study, we extend HM to q-rung picture fuzzy environment, propose novel q-rung picture fuzzy aggregation operators, and demonstrate their application to multi-attribute group decision-making (MAGDM). First of all, on the basis of Dombi t-norm and t-conorm (DTT), we propose novel operational rules of q-rung picture fuzzy numbers (q-RPFNs). Second, we propose some new aggregation operators of q-RPFNs based on the newly-developed operations, i.e., the q-rung picture fuzzy Dombi Hamy mean (q-RPFDHM) operator, the q-rung picture fuzzy Dombi weighted Hamy mean (q-RPFDWHM) operator, the q-rung picture fuzzy Dombi dual Hamy mean (q-RPFDDHM) operator, and the q-rung picture fuzzy Dombi weighted dual Hamy mean (q-RPFDWDHM) operator. Properties of these operators are also discussed. Third, a new q-rung picture fuzzy MAGDM method is proposed with the help of the proposed operators. Finally, a best project selection example is provided to demonstrate the practicality and effectiveness of the new method. Uncontrolled inflammation is a pathological state that underlies many diseases. Despite the development of numerous anti-inflammatory agents, the treatment of uncontrolled inflammation remains a challenging task. We developed a targeted delivery system for [5-(p-fluorophenyl)-2-ureido]thiophene-3-carboxamide (TPCA-1), a potent inhibitor of the NF-κB signaling pathway. The system comprises TPCA-1-loaded nanoparticles (NPs) functionalized with a monoclonal antibody (mAb) that specifically binds to the break point of the IgD6 region of the platelet/endothelial cell adhesion molecule-1 (PECAM-1) extracellular segment that is overexposed on the injured endothelium and activated macrophages during the pathogenesis of inflammation. In vitro binding and cellular uptake experiments revealed that the mAb modification on NPs could significantly enhance uptake by both Raw264.7 and HUVEC compared with unmodified NPs. In studies conducted at the cellular level focusing on anti-inflammatory and antioxidant effects, this formulation was found to effectively inhibit M1 polarization of macrophages, downregulate the secretion of pro-inflammatory cytokines, and reduce the production of reactive oxygen species (ROS) and nitric oxide (NO). In an animal model of vascular endothelial injury with acute inflammation, these NPs were capable of delivering TPCA-1 to inflammatory lesions in a targeted manner. Compared with the free agent-treated group, the NP-treated group exhibited reduced infiltration of inflammatory cells. Background/Aims: To explore the protective effects and therapeutic mechanism of Esomeprazole (PPI), polaprezinc granule (PZ), and PPI + PZ on reflux esophagitis (RE) in the rat model. Methods: Wistar rats were randomly divided into 9 groups, which contain the control group, the acid cessation group (0.7% HCl, Q3D × 4), and the acid persistence group (0.7% HCl, Q3D × 11). PPI was administered by gavage at 8 mg·kg−1 body weight and PZ was administered by gavage at 120 mg·kg−1 body weight once a day for 15 days. The gastric cardia tissue of the feeding tube was observed under the light microscope, and the levels of interleukin-8 (IL-8) and prostaglandin E2 (PGE2) were measured by ELISA. The expression of EGFR, Akt, p-Akt, and p-mTOR was detected by Western blot. Results: The ELISA results showed that the levels of IL-8 and PGE2 were significantly increased in the model group, but decreased in all groups after treatment. In the acid cessation group, PZ treatment had the most significant effect on reducing IL-8 levels and PPI + PZ treatment had the most significant effect on reducing PGE2 levels. In the acid persistence group, the PPI treatment had the most significant effect on reducing the levels of IL-8 and PGE2, and the PZ treatment could also significantly reduce their levels, close to the normal value. Western blot results showed that the expression of PI3K/Akt/mTOR pathway protein was increased in the model group, while its expression was decreased after treatment. Conclusions: Polaprezinc has a significant therapeutic effect on RE in rats, which can reduce the levels of IL-8 and PGE2 and downregulate the expression of PI3K/Akt/mTOR signal pathway protein. The efficacy of polaprezinc in the treatment of reflux esophagitis is comparable to that of PPI, and the combination of them is more effective in the reflux esophagitis treatment. Combination therapy is being actively explored to improve the efficacy and safety of anti-CD19 chimeric antigen receptor T-cell (CART19) therapy, among which Bruton tyrosine kinase inhibitors (BTKIs) are highly expected. BTKIs may modulate T-cell function and remodel the tumor micro-environment (TME), but the exact mechanisms involved and the steps required to transform different BTKIs into clinical applications need further investigation. We examined the impacts of BTKIs on T-cell and CART19 phenotype and functionality in vitr o and further explored the mechanisms. We evaluated the efficacy and safety of CART19 concurrent with BTKIs in vitro and in vivo. Moreover, we investigated the effects of BTKIs on TME in a syngeneic lymphoma model. Here we identified that the three BTKIs, ibrutinib, zanubrutinib and orelabrutinib, attenuated CART19 exhaustion mediated by tonic signaling, T-cell receptor (TCR) activation and antigen stimulation. Mechanistically, BTKIs markedly suppressed CD3-ζ phosphorylation of both chimeric antigen receptor and TCR and downregulated the expression of genes associated with T-cell activation signaling pathways. Moreover, BTKIs decreased interleukin 6 and tumor necrosis factor alpha release in vitro and in vivo. In a syngeneic lymphoma model, BTKIs reprogrammed macrophages to the M1 subtype and polarized T helper (Th) cells toward the Th1 subtype. Our data revealed that BTKIs preserved T-cell and CART19 functionality under persistent antigen exposure and further demonstrated that BTKI administration was a potential strategy for mitigating cytokine release syndrome after CART19 treatment. Our study lays the experimental foundation for the rational application of BTKIs combined with CART19 in clinical practice. BTKIs attenuate T-cell and CART19 exhaustion mediated by sustained TCR stimulation and CAR tonic signaling and impede release of IL-6 and TNF-α by macrophages. The AGCU Expressmarker 20 + 20Y Kit is a newly devised short tandem repeat (STR) multiplex system that simultaneously analyses a set of 19 autosomal STR loci, 20 Y chromosomal STR loci and the amelogenin locus with six-dye fluorescent labelling. Here, the AGCU Expressmarker 20 + 20Y system was validated following the guidelines published by the Scientific Working Group on DNA Analysis Methods (SWGDAM), including PCR-condition, sensitivity, mixtures, species specificity, models of inhibition, precision, stutter percentage, concordance, population genetic studies, performance on three kinds of degraded DNA and a type of casework samples. The results indicated that the kit had high sensitivity when there was a small amount of DNA (0.0625 ng), more than one male (minor: major=1:19), or a mixture of males and females (male: female=1:32), models of inhibition (250 μM hematin, 1500 ng/μL humic acid and more than 100 ng/μL tannic acid) and degraded samples. The kit showed high precision level with standard deviation of allele size no more than 0.0930. Furthermore, this system was also tested in 444 random male samples of Chinese Han and Hui population, showing its high discrimination capability in Han and Hui population. Meanwhile, the system was applicable to the case of the AMELY abnormality. Y-chromosomal short tandem repeats (Y-STRs) are widely applied to evolutionary, genealogical, and kinship analyses of male linages in forensic studies, but these low to midrange mutated Y-STRs typically fail to separate related males from the same paternal lineage. Recently, rapidly mutating Y-STRs (RM Y-STRs) have been demonstrated to improve the differentiation of male relatives and individuals. The Microreader™ RM-Y ID System is a new RM Y-STR kit that is capable of simultaneously amplifying 17 RM Y-STRs. Herein, to verify the efficiency and accuracy of the Microreader™ RM-Y ID System, developmental validation was conducted, including PCR-based studies, sensitivity, stability, species specificity, mixture, stutter percentage, and precision studies. Full profiles could be obtained when the hematin concentration was 250 μM, humic acid concentration was 1500 ng/μl, and tannic acid concentration was 200 ng/μl. Full profiles of the mixture of males/males could be detected up to a ratio of 19:1, and full profiles of females/males could always be detected even at ratios up to 24,000:1. Moreover, the forensic characteristics of 250 DNA-confirmed father-son pairs were analysed. The results showed that these 17 RM Y-STRs had high power for forensic discrimination (HD = 1) in the Chinese Han population, and the mutation rates were in the range of 4 × 10−3 (95% CI 1.00 × 10−4 to 2.21 × 10−2, DYS464) to 8.8 × 10−2 (95% CI 5.60 × 10−2 to 1.30 × 10−1, DYF399S1), indicating that the kit was effective for RM Y-STR studies and absolute individualisation of interrelated male individuals. The droplet digital Polymerase Chain Reaction (ddPCR) has garnered recognition for its distinctive attribute of absolute quantification. And it has found practical utility in age prediction through DNA methylation profiles. However, a prevalent limitation in current ddPCR methodologies is the restricted capacity to detect only two targets concurrently in most instruments, leading to high costs, sample wastage, and labor-intensive procedures. To address the limitations, a novel high-throughput ddPCR system allowing for the simultaneous detection of eight targets was developed. Through the implementation of a new 8-plex ddPCR assay, coupled with comprehensive linear regression analyses involving primers and probes ratios, diverse inputs of single CpG sites with distinct primers and probes, and varying plex assay configurations, stable DNA methylation values for four CpGs and stable measurement precisions for distinct multiplex systems were consistently observed. These findings pave the way for advancing the field of chemistry science by enabling more efficient and cost-effective methods. Furthermore, the comparative validation of ddPCR and SNaPshot demonstrated a remarkable concordance in results, and the system also displayed well in the field of various aspects, including species specificity, DNA input, and aged samples. In this study, the recommended input of bisulfite-converted DNA was determined to be 10–50 ng due to the double-positive droplets. Notably, the Pearson correlation coefficient squared values of four CpGs were 0.4878 (ASPA), 0.4832 (IGSF1), 0.6881 (COL1A1), and 0.6475 (MEIS1-AS3). Introduction: Cancer patients are prone to infections, but the mortality of fatal infections remains unclear. Understanding the patterns of fatal infections in patients with cancer is imperative. In this study, we report the characteristics, incidence, and predictive risk factors of fatal infections among a population-based cancer cohort. Methods: A total of 8,471,051 patients diagnosed with cancer between 1975 and 2016 were retrospectively identified from the Surveillance, Epidemiology, and End Results (SEER) program. The primary outcome was dying from fatal infections. Mortality rates and standardized mortality ratios (SMRs) adjusted for age, sex, race, and calendar year were calculated to characterize the relative risks of dying from fatal infections and to compare with the general population. Furthermore, cumulative mortality rates and the Cox regression models were applied to identify predictive risk factors of fatal infections. Results: In cancer patients, the mortality rate of fatal infections was 260.1/100,000 person-years, nearly three times that of the general population [SMR, 2.92; 95% (confidence interval) CI 2.91–2.94]. Notably, a decreasing trend in mortality rate of fatal infections was observed in recent decades. SMRs of fatal infections were highest in Kaposi sarcoma (SMR, 162.2; 95% CI 159.4–165.1), liver cancer (SMR, 30.9; 95% CI 30.0–31.8), acute lymphocytic leukemia (SMR, 19.1; 95% CI 17.0–21.4), and acute myeloid leukemia (SMR, 13.3; 95% CI 12.4–14.3). Patients aged between 20 and 39 years old exhibited a higher cumulative mortality rate in the first few years after cancer diagnosis, whereas the cumulative mortality rate of those > 80 years old was rapidly increasing and became the highest approximately 3 years post-cancer diagnosis. Predictive risk factors of dying from fatal infections in cancer patients were the age of 20–39 or > 80 years, male sex, black race, diagnosed with cancer before 2000, unmarried status, advanced cancer stage, and not receiving surgery and radiotherapy, but receiving chemotherapy. Conclusion: Cancer patients were at high risks of dying from infectious diseases. Certain groups of cancer patients, including those aged between 20 and 39 or > 80 years, as well as those receiving chemotherapy, should be sensitized to the risk of fatal infections. In general, a two-dimensional (2D) moving trajectory can be reconstructed using data collected from a 6-axis IMU (inertial measurement unit), which consists of a 3-axis gyroscope and a 3-axis accelerometer. Since the power consumption of a gyroscope is significantly larger than that of an accelerometer, to be energy-efficient, it is more preferable to use two gyro-free IMUs as alternatives. The existing gyro-free trajectory reconstruction approach is an angular velocity (i.e. ω) -based method. The design of an ω-free gyro-free approach remains as a research gap, which motivates this research work. In this paper, an ω-free accelerometer pair (OFAP) method is proposed, which can yield a lower accumulated error, has a lower power consumption, is more scalable, and can deliver the same trajectory reconstruction performance as the existing approach. The chronnectome of the human brain represents dynamic connectivity patterns of brain networks among interacting regions, but its organization principle and related transcriptional signatures remain unclear. Using task-free fMRI data from the Human Connectome Project (681 participants) and microarray-based gene expression data from the Allen Institute for Brain Science (1791 brain tissue samples from six donors), we conduct a transcriptome-chronnectome association study to investigate the spatial configurations of dynamic brain networks and their linkages with transcriptional profiles. We first classify the dynamic brain networks into four categories of nodes according to their time-varying characteristics in global connectivity and modular switching: the primary sensorimotor regions with large global variations, the paralimbic/limbic regions with frequent modular switching, the frontoparietal cortex with both high global and modular dynamics, and the sensorimotor association cortex with limited dynamics. Such a spatial layout reflects the cortical functional hierarchy, microarchitecture, and primary connectivity gradient spanning from primary to transmodal areas, and the cognitive spectrum from perception to abstract processing. Importantly, the partial least squares regression analysis reveals that the transcriptional profiles could explain 28% of the variation in this spatial layout of network dynamics. The top-related genes in the transcriptional profiles are enriched for potassium ion channel complex and activity and mitochondrial part of the cellular component. These findings highlight the hierarchically spatial arrangement of dynamic brain networks and their coupling with the variation in transcriptional signatures, which provides indispensable implications for the organizational principle and cellular and molecular functions of spontaneous network dynamics. Microdimples as a typical surface texture has been widely used for improving the properties in tribology and heat transfer fields. Ultrasonically assisted electrochemical micromachining (USEMM) is a popular method for generating microdimples as it can reduce the overcutting and enhance electrolyte renewal to improve the machining localization and promote the material removal. However, the fundamental understanding of the process and particularly the relationship between the machining performance and gap pressure is limited. In this paper, radial ultrasonicrolling electrochemical micromachining (RUREMM) is employed to generate microdimples on SS304 surface. Effects of gap pressure on the material removal rate, machining accuracy, and roughness are studied and discussed in detail. Results show that RUREMM can fabricate the microdimples and the existing ultrasonic vibration in the inter-electrode gap is more effective to enhance the machining performance. By using the selected parameters in RUREMM, the current density during machining has a 6.6% increase in RUREMM than in EMM. Moreover, the width of the microdimple is decreased by 14.6%, the depth of the microdimple is increased by 19.2%, the cross-sectional area of the machined dimple is expanded by 31.2%, the roughness of the bottom is decreased 23.9% and also, better surface quality for the side wall could be obtained. The role of cerebellum and cerebro‐cerebellar system in neural plasticity induced by cerebral gliomas involving language network has long been ignored. Moreover, whether or not the process of reorganization is different in glioma patients with different growth kinetics remains largely unknown. To address this issue, we utilized preoperative structural and resting‐state functional MRI data of 78 patients with left cerebral gliomas involving language network areas, including 46 patients with low‐grade glioma (LGG, WHO grade II), 32 with high‐grade glioma (HGG, WHO grade III/IV), and 44 healthy controls. Spontaneous brain activity, resting‐state functional connectivity and gray matter volume alterations of the cerebellum were examined. We found that both LGG and HGG patients exhibited bidirectional alteration of brain activity in language‐related cerebellar areas. Brain activity in areas with increased alteration was significantly correlated with the language and MMSE scores. Structurally, LGG patients exhibited greater gray matter volume in regions with increased brain activity, suggesting a structure–function coupled alteration in cerebellum. Furthermore, we observed that cerebellar regions with decreased brain activity exhibited increased functional connectivity with contralesional cerebro‐cerebellar system in LGG patients. Together, our findings provide empirical evidence for a vital role of cerebellum and cerebro‐cerebellar circuit in neural plasticity following lesional damage to cerebral language network. Moreover, we highlight the possible different reorganizational mechanisms of brain functional connectivity underlying different levels of behavioral impairments in LGG and HGG patients. Human cognition and behaviors depend upon the brain's functional connectomes, which vary remarkably across individuals. However, whether and how the functional connectome individual variability architecture is structurally constrained remains largely unknown. Using tractography- and morphometry-based network models, we observed the spatial convergence of structural and functional connectome individual variability, with higher variability in heteromodal association regions and lower variability in primary regions. We demonstrated that functional variability is significantly predicted by a unifying structural variability pattern and that this prediction follows a primary-to-heteromodal hierarchical axis, with higher accuracy in primary regions and lower accuracy in heteromodal regions. We further decomposed group-level connectome variability patterns into individual unique contributions and uncovered the structural-functional correspondence that is associated with individual cognitive traits. These results advance our understanding of the structural basis of individual functional variability and suggest the importance of integrating multimodal connectome signatures for individual differences in cognition and behaviors. Nanostructured gold substrates provide chemically stable, signal-enhancing substrates for the sensitive detection of a variety of compounds through surface-enhanced Raman spectroscopy (SERS). Recent developments in advanced fabrication methods have enabled the manufacture of SERS substrates with repeatable surface nanostructures that provide reproducible quantitative analysis, historically a weakness of the SERS technique. Here, we describe the novel use of gold-sputtered Blu-ray disc surfaces as SERS substrates. The unique surface features and composition of the Blu-ray disc recording surface lead to the formation of gold nano-islands and nanogaps following simple gold sputtering, without any background peaks from the substrate. The SERS performance of this substrate is strong and reproducible with an enhancement factor (EF) of 10 for melamine. A limit of detection (LOD) for this compound of 70 ppb and average reproducibility of ±12 % were achieved. Gold-sputtered Blu-ray discs thus offer an excellent alternative to more exotic gold SERS substrates prepared by advanced, time-consuming and expensive methods. Functional near-infrared spectroscopy (fNIRS), a promising noninvasive imaging technique, has recently become an increasingly popular tool in resting-state brain functional connectivity (FC) studies. However, the corresponding software packages for FC analysis are still lacking. To facilitate fNIRS-based human functional connectome studies, we developed a MATLAB software package called “functional connectivity analysis tool for near-infrared spectroscopy data” (FC-NIRS). This package includes the main functions of fNIRS data preprocessing, quality control, FC calculation, and network analysis. Because this software has a friendly graphical user interface (GUI), FC-NIRS allows researchers to perform data analysis in an easy, flexible, and quick way. Furthermore, FC-NIRS can accomplish batch processing during data processing and analysis, thereby greatly reducing the time cost of addressing a large number of datasets. Extensive experimental results using real human brain imaging confirm the viability of the toolbox. • Microwave is best treatment on transglutaminase (TGase)-induced whey protein isolate. • Ultrasound-microwave promoted the reaction of whey protein isolate and TGase. • Ultrasound-microwave enhanced properties of TGase-induced whey protein isolate. In the present study, ultrasound (400 W, U), microwave heating (75 ℃ for 15 min, M) and ultrasound synergized with microwave heating (UM) pretreatments of whey protein isolate (WPI) were applied to investigate and compare their influence on structure, physicochemical and functional characteristic of transglutaminase (TGase)-induced WPI. From the results of size exclusion chromatography, it could be seen that all three physical pretreatments could promote the formation of polymers in TGase cross-linked WPI, whose polymer amounts were increased by the order of U, UM and M pretreatment. Among three physical methods, M pretreatment had the strongest effect on structure and functional characteristics of TGase-induced WPI. Furthermore, compared with TGase-induced WPI, α-helix and β-turn of M−treated TGase-induced WPI (M−WPI−TGase) were reduced by 7.86% and 2.93%, whereas its β-sheet and irregular curl were increased by 15.37% and 7.23%. Zeta potential, emulsion stability and foaming stability of M−WPI−TGase were increased by 7.8%, 59.27% and 28.95%, respectively. This experiment exhibited that M was a more effective pretreatment method than U, UM for WPI, which could promote its reaction with TGase and improve its functional properties. The MNI CIVET pipeline for automated extraction of cortical surfaces and evaluation of cortical thickness from in-vivo human MRI has been extended for processing macaque brains. Processing is performed based on the NIMH Macaque Template (NMT), as the reference template, with the anatomical parcellation of the surface following the D99 and CHARM atlases. The modifications needed to adapt CIVET to the macaque brain are detailed. Results have been obtained using CIVET-macaque to process the anatomical scans of the 31 macaques used to generate the NMT and another 95 macaques from the PRIME-DE initiative. It is anticipated that the open usage of CIVET-macaque will promote collaborative efforts in data collection and processing, sharing, and automated analyses from which the non-human primate brain imaging field will advance. Neuroimaging non-human primates (NHPs) is a growing, yet highly specialized field of neuroscience. Resources that were primarily developed for human neuroimaging often need to be significantly adapted for use with NHPs or other animals, which has led to an abundance of custom, in-house solutions. In recent years, the global NHP neuroimaging community has made significant efforts to transform the field towards more open and collaborative practices. Here we present the PRIMatE Resource Exchange (PRIME-RE), a new collaborative online platform for NHP neuroimaging. PRIME-RE is a dynamic community-driven hub for the exchange of practical knowledge, specialized analytical tools, and open data repositories, specifically related to NHP neuroimaging. The effects of different concentrations of safflower yellow (SY) on the structure and emulsifying properties of whey protein isolate (WPI)-SY complexes were investigated at 25 °C for 0, 3 and 6 days. About 92.6%, 93.75%, and 95.47% of the free sulfhydryl groups from the WPI-SY complexes were lost over 0, 3 and 6 days, respectively, in the presence of 2% SY. Meanwhile, the fluorescence intensity of the WPI-SY complexes also weakened obviously, indicating that the tryptophan residues micro-environment underwent certain changes. Furthermore, fourier transform infrared spectroscopy demonstrated that SY increased the proportion of WPI β-sheets and decreased the proportion of β-turn structures. The particle size of the WPI-SY complexes increased as the SY concentration was increased from 1 to 4%. Compared with SY or WPI, the antioxidant activity of the WPI-SY complexes was enhanced with SY ranged from 0.25 to 4%. SY triggered emulsifying activity of WPI initially increased and then decreased. The effect of high pressure homogenization (HPH) pretreatments (0, 20, 40, 60, 80, 100, and 120 MPa for 4 cycles) on structure, physicochemical properties and functionality of laccase-treated α-lactalbumin (α-LA) was examined in this experimental study. Mass spectrometry and gel filtration chromatography revealed that monomers of laccase-treated α-LA decreased remarkably, and oligomers (dimers, trimers and tetramers) and polymers were formed with homogenization pressures ranging from 0 to 120 MPa. The HPH treatment exposed more α-LA tyrosine residues, which were further catalyzed by laccase to form dityrosine. Additionally, fourier transform infrared spectroscopy indicated that there were lower α-helix and higher random coil contents in laccase-treated α-LA with increasing homogenization pressure. The HPH pretreatment increased the emulsifying and gel properties of laccase-treated α-LA. Thus, the HPH pretreatment enhanced the crosslinking degree of laccase-treated α-LA as well as its functionality. Impacts of ultrafiltration and hydrolysis on antioxidant abilities of Maillard reaction products (MRPs) derived from whey protein isolate (WPI) and galactose (Gal), were evaluated in this study. Fraction (MW > 50 kDa)of WPI-Gal MRPs prepared with ultrafiltration, had the strongest antioxidant activity. The trichloroacetic acid (TCA, 2–8%) treatment could effectively enhanced antioxidant activities of WPI-Gal MRPs. WPI-Gal MRPs had higher degrees of hydrolysis and antioxidant activity than heated WPI. With hydrolysis time of pepsin and trypsin extended up, degrees of hydrolysis and antioxidant activities of WPI-Gal MRPs and WPI gradually increased (P < 0.05).
