Altered expression of miR-182 has been observed in various types of human cancer. The purpose of this study was to investigate the expression of miR-182 and its role in prostate cancer (PCa). Expression of miR-182 and ST6GALNAC5 in tumor tissues and the Du145 PCa cell line was analyzed. Cell proliferation assay, colony formation assay, transwell assay, and wound healing assay were performed. The impact of miR-182 on tumor growth was investigated using a xenograft model. The results indicated that expression of miR-182 was higher in PCa tissues and cell lines, while ST6GALNAC5 was decreased. Downregulating miR-182 significantly inhibited the capacities of proliferation and invasion of PC3 and Du145 cells. ST6GALNAC5 was demonstrated to be a target of miR-182 by luciferase assay, and western blot results indicated PI3K/Akt pathway was involved in miR-182 associated effects on PC3 and Du145 cells. The animal experiment suggested that knockdown of miR-182 inhibited tumor growth. Our study proved that miR-182 participated in the proliferation and invasion of PCa cells via mediating expression of ST6GALNAC5 and established a miR-182/ST6GALNAC5/PI3K/AKT axis in regulation of tumor progression. Our investigation provided a basis for further exploration of the application of miR-182 or ST6GALNAC5-associated therapies for PCa patients.

Patients and methods: We used the Surveillance, Epidemiology and End Results (SEER) database (1973–2014) to identify metastatic ACC patients (stage IV by using European Network for the Study of Adrenal Tumors stage classification). Correlated variables, including age, sex, race, tumor laterality, treatment modality, lymph node dissection, surgery of metastatic site, tumor size, and tumor stage, were extracted. Univariate and multivariate Cox regression analyses were used to define the efficacy of surgery on survival outcomes, including overall survival and cancer-specific survival of ACC. Results: There were 290 metastatic ACC patients identified from the database. The overall median survival time was 7 (95% CI, 6–8) months. Among these patients, 118 patients received primary site surgery and 172 patients did not. The mode localization phenomenon in mistuned cyclically periodic structures has been a hot spot in the past few decades. According to numbers of documents, the fundamental concepts, mechanism, main research methods, and the main results of mode localization are introduced; the current research status of the mode localization is summarized. The possiblity of mode localization in a telescope reflector model has been analyzed by using Finite Element Method. We also illustrate the problems for further studies, including: the unified modeling and analysis theory with more extensive applicability and better analytical accuracy; determining how to properly introduce mistuning parameters that can reflect the true state of the structure; constructing mode localization factor which can sensitively and effectively reflect the effect of mode localization; developing new damage identification method considering the influence of mode localization; how to make use of the localization characteristic to control the vibration level of the important sub-structure and improve the sensitivity of the periodic structure to the random detuning.

Optimal sensor placement technique plays a key role in structural health monitoring and structural vibration control. Based on the advantages and disadvantages of effective independence (EI) and modal kinetic energy (MKE) methods, two improved optimal sensor placement methods which are Effective Independence - Average Acceleration Amplitude (EI-AAA) method and Effective Independence - Modal Kinetic Energy (EI-MKE) method are proposed in this paper. Firstly the formulas are deduced from modal expansion of multiple-input-multiple-output (MIMO) displacement frequency response function matrix. Then a computational simulation of steel cross beam structure has been implemented to demonstrate the feasibility of the two improved methods above. The obtained optimal sensor locations using the two improved methods are compared with those gained by EI method and MKE method. Finally six classical comparison criteria are employed to demonstrate the advantage and disadvantage of these four methods. The results showed that some innovations proposed in this paper are effective and reliable. The two improved optimal sensor placement methods (EI-AAA method and EI-MKE method) can not only make the truncated mode shapes as linearly independent as possible but also enable the measured modal kinetic energy to maintain the maximum value.

The wireless sensor network is a highly active area of research. Much progress has been made in wireless sensor network research for structural monitoring, but the research of the long-term monitoring system based on wireless sensor network in civil engineering structures is still at the beginning stage. The paper proposed to use wireless sensor network to realize intensive measurement long-term monitoring, in order to improve the effect of structural health monitoring. It introduced the characteristics and deficiencies of the wireless technology application in the structure health monitoring system, and proposed the system design principle of the densely measured points long-term monitoring system based on wireless sensor network. We has carried on the detailed argumentation and design from system architecture, the terminal node, monitoring method three aspects, and put forward a set of solutions.

Based on numerical and experimental methods, a systematic structural evaluation of a steel natatorium in service was carried out in detail in this paper. Planning of inspection tasks was proposed firstly according to some national codes in China in order to obtain the economic and reliable results. The field visual inspections and static computation were conducted in turn under in-service environmental conditions. Further a three-dimensional finite element model was developed according to its factual geometry properties obtained from the field inspection. An analytical modal analysis was performed to provide the analytical modal properties. The field vibration tests on the natatorium were conducted and then two different system identification methods were used to obtain the dynamic characteristics of the natatorium. A good correlation was achieved in results obtained from the two system identification methods and the finite element method (FEM). The numerical and experimental results demonstrated that the main structure of the natatorium in its present status is safe and it still satisfies the demand of the national codes in China. But the roof system such as purlines and skeletons must be removed and rebuilt completely. Moreover the system identification results showed that field vibration test is sufficient to identify the reliable dynamic properties of the natatorium. The constructive suggestion on structural evaluation of the natatorium is that periodic assessment work must be maintained to ensure the natatorium`s safety in the future.

In the present study, we aimed to investigate the expression and distribution of transient receptor potential melastatin (TRPM)- and vanilloid (TRPV)- related channels in rat spermatogenic cells and spermatozoa. Spermatogenic cells and spermatozoa were obtained from male Sprague-Dawley rats. Reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of all TRPM and TRPV channel members with specific primers. Western blot analysis was applied for detecting the expression of TRPM and TRPV channel proteins. Immunohistochemistry staining for TRPM4, TRPM7 and TRPV5 was also performed in rat testis. The mRNAs of TRPM3, TRPM4, TRPM7 and TRPV5 were detected in the spermatogenic cells and spermatozoa in rat. Western blot analysis verified the expression of TRPM4, TRPM7 and TRPV5 in the rat spermatogenic cells and spermatozoa. Immunocytochemistry staining for TRPM and TRPV channel families indicated that TRPM4 and TRPM7 proteins were highly expressed in different stages of spermatogenic cells and spermatozoa, while TRPV5 protein was lowly expressed in these cells. Our results demonstrate that mRNAs or proteins for TRPM3, TRPM4, TRPM7 and TRPV5 exist in rat spermatogenic cells and spermatozoa. These data presented here may assist in elucidating the possible physiological function of TRPM and TRPV channels in spermatogenic cells and spermatozoa.

Optimal sensor placement technique plays a key role in structural health monitoring of spatial lattice structures. This paper considers the problem of locating sensors on a spatial lattice structure with the aim of maximizing the data information so that structural dynamic behavior can be fully characterized. Based on the criterion of optimal sensor placement for modal test, an improved genetic algorithm is introduced to find the optimal placement of sensors. The modal strain energy (MSE) and the modal assurance criterion (MAC) have been taken as the fitness function, respectively, so that three placement designs were produced. The decimal two-dimension array coding method instead of binary coding method is proposed to code the solution. Forced mutation operator is introduced when the identical genes appear via the crossover procedure. A computational simulation of a 12-bay plain truss model has been implemented to demonstrate the feasibility of the three optimal algorithms above. The obtained optimal sensor placements using the improved genetic algorithm are compared with those gained by exiting genetic algorithm using the binary coding method. Further the comparison criterion based on the mean square error between the finite element method (FEM) mode shapes and the Guyan expansion mode shapes identified by data-driven stochastic subspace identification (SSI-DATA) method are employed to demonstrate the advantage of the different fitness function. The results showed that some innovations in genetic algorithm proposed in this paper can enlarge the genes storage and improve the convergence of the algorithm. More importantly, the three optimal sensor placement methods can all provide the reliable results and identify the vibration characteristics of the 12-bay plain truss model accurately.

This paper adopts autoregressive (AR) model to simulate the wind velocity of spatial three-dimensional fields in accordance with the time and space dependent characteristics of the 3-D fields. Based on the built MATLAB programming, this paper discusses in detail the issues of the AR model deduced by matrix form in the simulation and proposes the corresponding solving methods: the over-relaxation iteration to solve the large sparse matrix equations produced by large number of degrees of freedom of structures; the improved Gauss formula to calculate the numerical integral equations which integral functions contain oscillating functions; the mixed congruence and central limit theorem of Lindberg-Levy to generate random numbers. This paper also develops a method of ascertaining the rank of the AR model. The numerical examples show that all those methods are stable and reliable, which can be used to simulate the wind velocity of all large span structures in civil engineering.

The occurrence of the mode localization is studied in a structure of a single-layer spherical reticulated shell. The matrix perturbation method is used to analyze the mechanism of the occurrence of mode localization. The dynamical characteristics of this structure, especially the effects of the variation of the stiffness parameter on the vibration modes of the structure, were analyzed by numerical simulations. An amplitude significance coefficient is proposed as a measure of the degree of the mode localization. The results show that small deviations of the stiffness parameter can cause significant changes to the structural dynamics characteristics; the mode localization does occur for some particular modes of this structure in the case of random stiffness mistuning. Preliminary analysis found that the mode can be considered as strongly localized when the amplitude significance coefficient is greater than 8.

To evaluate in vitro effect of abnormal savda munziq (ASMq) on the proliferation and apoptosis of human hypertrophic scar fibroblasts (HSFs).HSFs were divided into six groups to receive different treatments as group A (blank control group), group B-E (ASMq in different concentration), and group F(5-Fu). Each group contains six specimens. The HSFs were cultured in vitro. After culture for 48 hours, the CCK8 test and flow cytometry methods were used to detect the proliferation, cell cycle and apoptosis.The proliferation of HSFs in the B, C, D and E groups was inhibited at G2/M period, while it was inhibited at G0/S period in group F (P0.05). The inhibition effect of ASMq (0.1-1.0 mg/ml) on the fibroblasts enhanced in a concentration-dependent manner. Flow cytometry analysis with annexin V-FITC and PI staining confirmed the apoptotic. When HSFs were exposed to ASMq at 1.0 mg/ml (group E) for 48 h, the percentage of apoptotic cells increased to (43.7 +/- 2.58)%, which was significantly higher than that of blank control group (2.2 +/- 0.59)%. The induced apoptosis effect was also increased in a concentration-dependent manner.ASMq has a inhibitory effect on the proliferation and an enhancement effect on the apoptosis of fibroblast. ASMq could be used as an effective drug for treatment of hypertrophic scar.

Modal identification with output-only measurements plays a key role in vibration-based damage detection, model updating, and structural health monitoring in civil engineering. This paper addresses the application of modal identification method to a triangle steel tube truss natatorium using the field measurement data. To obtain dynamic characteristics of the spatial structure, four different output-only system identification methods are employed. They are natural excitation technique–eigensystem realization algorithm, data-driven stochastic subspace identification method, frequency-domain decomposition/frequency-spatial domain decomposition method, and half spectra/rational fractional orthogonal polynomial method. First an analytical modal analysis was performed on the three-dimensional finite element model according to the factual layout design to obtain the calculated frequencies and mode shapes. Then the whole procedure of the field vibration tests on the natatorium was presented. Finally, practical issues and efficiency of the four output-only modal identification techniques are investigated, and compared with the results from a finite element model. The system identification results demonstrate that both methods can provide reliable information on dynamic characteristics of the spatial structure. The frequency-domain methods, however, can quickly identify the modal parameters, but the leakage error introduced by power-spectral density estimation is existent due to the limited length of data. And the time-domain methods can avoid the leakage error, but the computational modes and the computational cost are the main two drawbacks in application. The conclusion is that several system identification methods should be consulted to ensure the accuracy of the estimated modal parameters.

To study the expression of the mRNAs of transient receptor potential (TRP) gene subfamily TRPV and TRPM in rat testes.Normal SD rat testes were collected and the expression of TRPV and TRPM mRNAs were detected by routine RT-PCR.The TRPV4, TRPV5, TRPV6, TRPM3, TRPM4 and TRPM8 mRNAs were detected in the rat testes, but the other members of TRPV and TRPM family were not detected.TRPV4, TRPV5, TRPV6, TRPM3, TRPM4 and TRPM8 are expressed in rat testes. This finding provides the basis for exploring the functions of TRPV and TRPM in the testes and the relation between testis diseases and the TRP family.

To investigate the expression of transient receptor potential melastatin (TRPM) and transient receptor potential vanilloid (TRPV) channel family genes in rat spermatogenic cells.Rat spermatogenic cells were isolated by a mechanical procedure and the total RNA was extracted using TRIzol reagent. TRPM and TRPV channel family genes were amplified by RT-PCR and the presence of the target genes was detected by agarose gel electrophoresis. The relative gene expression levels were measured by real-time quantitative RT-PCR.TRPV5, TRPM3, TRPM4 and TRPM7 mRNAs were expressed in rat spermatogenic cells, but TRPV1, TRPV2, TRPV3, TRPV4, TRPV6, TRPM1, TRPM2, TRPM5, TRPM6, TRPM7 and TRPM8 mRNAs were not detected. The relative expressions of TRPM and TRPV mRNA were determined by quantitative real-time RT-PCR. TRPM7 expression was the highest among all the TRPM subtypes in rat spermatogenic cells, at a level equivalent to (0.0430-/+0.0034)% of beta-actin expression. TRPM3 and TRPM4 were also highly expressed, but their expression levels were only approximately 56% and 63% of that of TRPM7, respectively. For the TRPV subfamily, only TRPV5 mRNA was abundantly expressed at the level of (0.0157-/+0.0029)% relative to that of beta-actin.TRPV5, TRPM3, TRPM4 and TRPM7 mRNAs were coexpressed in spermatogenic cells in rats, among which TRPM4 and TRPM7 mRNA were expressed at high levels. TRPM4 and TRPM7 channels may be involved in the regulation of growth, differentiation and maturation of rat spermatogenic cells and are associated with the generation of the sperms.

To investigate the best dose and the long-term effect of the human insulin-like growth factor-1 (hIGF-1) gene injection into the penis of aged rats.Included in this study were 10 young (4 months old) and 40 aged (24 months old) Sprague-Dawley male rats, the latter equally divided into a PBS control and a 10 microg, a 100 microg and a 1 000 microg hIGF-1 injection group. Electrical stimulation was conducted 4 and 8 weeks after hIGF-1 injection into the penile corpus cavernous of the rats to detect the intracavernous pressure (ICP) and mean arterial pressure (MAP). Dose - and time -associated therapeutic results were analyzed and the mRNA expression of hIGF-1 determined by RT - PCR.ICP, MAP and total ICP were significant decreased by electrical stimulation in the aged rats as compared with the young ones (P0.05), statistically increased in the three hIGF-1 dose groups in comparison with the PBS controls (P0.05), and showed no obvious difference between the young rats and the latter two dose groups at 4 and 8 weeks. Although less obvious effect was achieved in the 10 microg group than in the young rats, the therapeutic result was still of significance. The mRNA expression of the hIGF-1 gene was confirmed in all the hIGF-1 treated rats.The hIGF-1 therapy can improve erectile function in aged rats, 100 microg suffices for effective erection and the effect may last at least 8 weeks for a single dose.

To evaluate the incidence of capsular contracture in breast augmentation with textured versus smooth mammary Implants.Randomized controlled trials (RCTs) and clinical controlled trials (CCTs) were collected from Medline, Embase from May 1966 to May 2006, cochrane library (Issue 2, 2005), and CBM disc from May 1979 to May 2006. We handsearched Chinese Journal of Plastic Surgery (from establishment to May 2006) and Journal of Plastic and Reconstructive Surgery of America (from establishment to May 2006). RCTs were included. Data were extracted by two reviewers with designed extraction form RevMan. 4.2.8 software was used for data analysis.Six RCTs were included. The combined results of meta analysis showed that patients with textured implants had a lower tendency to develop capsules contracture than those with smooth implants either at one year or ten years after the breast augmentation.Compared with smooth mammary implants, textured surface implants significantly reduce the incidence of capsular contracture after breast augmentation.
