Abstract With climate change and anthropic influence on the coastal ecosystems, mangrove ecosystems are disappearing at an alarming rate. Accordingly, it becomes important to track, study, record and store the mangrove microbial community considering their ecological importance and potential for biotechnological applications. Here, we provide information on mangrove fungal community composition and diversity in mangrove ecosystems with different plant species and from various locations differing in relation to anthropic influences. We describe twelve newly assembled genomes, including four chromosomal-level genomes of fungal isolates from the mangrove ecosystems coupled with functional annotations. We envisage that these data will be of value for future studies including comparative genome analysis and large-scale temporal and/or spatial research to elucidate the potential mechanisms by which mangrove fungal communities assemble and evolve. We further anticipate that the genomes represent valuable resources for bioprospecting related to industrial or clinical uses.

Abstract Synthetic auxotrophy in which cell viability depends on the presence of an unnatural amino acid (unAA) provides a powerful strategy to restrict unwanted propagation of genetically modified organisms (GMOs) in open environments and potentially prevent industrial espionage. Here, we describe a generic approach for robust biocontainment of budding yeast dependent on unAA. By understanding escape mechanisms, we specifically optimize our strategies by introducing designed “immunity” to the generation of amber-suppressor tRNAs and developing the transcriptional- and translational-based biocontainment switch. Solanum torvum, a wild relative of eggplant in the Solanaceae family, is widely used as a rootstock to prevent various soil-borne diseases. Despite this valued trait, S. torvum seeds exhibit strong seed dormancy, resulting in low-frequency germination and uneven emergence rates during agricultural production. In this study, exogenous gibberellin (GA) was applied to induce the germination of seeds, thereby enabling the application of transcriptome analysis to explore the effects of exogenous GA on germination. A total of 9723 genes were identified to be involved in GA-induced germination of S. torvum seeds. These GA-associated genes were enriched in amino sugar and nucleotide sugar metabolism, glycolysis, the citrate cycle, pyruvate metabolism, hormone biosynthesis, and signaling transduction. Further analysis of these genes reveals that exogenous GA impaired the endogenous biosynthesis of GA. Analysis of the 285 key genes influenced by GA, auxin, abscisic acid (ABA), and other hormones suggests mutual crosstalk in the signaling of various hormones, enabling the joint regulation of the germination of S. torvum seeds. Additionally, a total of 440 genes encoding transcription factors were identified to be responsible for transcriptional reprogramming during GA-induced seed germination, and putative candidate genes responsible for S. torvum seed dormancy and germination, including ABI5, DOG1, DRM1, LEC1, and PIF, were significantly down-regulated in germinated S. torvum seeds. These findings provide the first insights into transcriptome regulation during GA-induced germination of S. torvum seeds.

In this study, a multilevel drought-induced resistance and resilience analysis (MDRRA) approach was developed to investigate the stability of vegetation in the Yellow River Basin (YRB). MDRRA was quantified by utilizing the Normalized Difference Vegetation Index (NDVI). It was applied to YRB to assess vegetation resistance and resilience to various levels of drought by utilizing precipitation and NDVI data from 2000 to 2019. The results reveal that vegetation resistance and resilience in YRB are affected by drought severity. Monthly and annual changes in SPI over the warm–temperate humid zone of the YRB show a decreasing trend, with rates of 0.001 per decade and 0.034 per decade, respectively; however, the other climatic subregions exhibit an increasing trend, with rates ranging from 0.002 per decade to 0.82 per decade. Over 77.56% of the downstream areas show increases in the annual SPI averages. Drought severity differs across subregions in the YRB. More severe drought events occur in its upper and middle reaches, while less severe ones happen in its lower reaches. As the drought severity increases, the arid and semiarid regions of the mesothermal zone exhibit a decrease in the resistance and resilience indices. MDRRA can help improve the stability and resilience of the ecosystem in the YRB.

Background The temporal progression states of the molecular and structural substrate in atrial fibrillation (AF) are not well understood. We hypothesized that these can be detected by AF electrograms and magnetic resonance imaging parametric mapping. Methods and Results AF was induced in 43 dogs (25–35 kg, ≥1 year) by rapid atrial pacing (RAP) (3–33 weeks, 600 beats/min), and 4 controls were used. We performed high‐resolution epicardial mapping (UnEmap, 6 atrial regions, both atria, 130 electrodes, distance 2.5 mm) and analyzed electrogram cycle length, dominant frequency, organization index, and peak‐to‐peak bipolar voltage. Implantable telemetry recordings were used to quantify parasympathetic nerve activity over RAP time. Magnetic resonance imaging native T1, postcontrast T1, T2 mapping, and extracellular volume fraction were assessed (1.5T, Siemens) at baseline and AF. Abstract The oral cavity harbors highly diverse communities of microorganisms. However, the number of isolated species and high-quality genomes is limited. Here we present a Cultivated Oral Bacteria Genome Reference (COGR), comprising 1089 high-quality genomes based on large-scale aerobic and anaerobic cultivation of human oral bacteria isolated from dental plaques, tongue, and saliva. COGR covers five phyla and contains 195 species-level clusters of which 95 include 315 genomes representing species with no taxonomic annotation. The oral microbiota differs markedly between individuals, with 111 clusters being person-specific. Genes encoding CAZymes are abundant in the genomes of COGR. Members of the Streptococcus genus make up the largest proportion of COGR and many of these harbor entire pathways for quorum sensing important for biofilm formation. Several clusters containing unknown bacteria are enriched in individuals with rheumatoid arthritis, emphasizing the importance of culture-based isolation for characterizing and exploiting oral bacteria.

Addressing the challenges for detecting underwater damage to the structures with pile foundation, this study presents the design of an adsorption-operated robotic system. Initially, a prototype is proposed for an adsorption-operated robot that is equipped with an automatic movement mechanism. To account for the disturbance of the drag force in the control system, a hydrodynamic numerical model for the detection robot is established. The numerical results of the drag force are then integrated into the control system, leading to the design of a force-redeemed active disturbance rejection controller. Furthermore, a novel detection algorithm based on the image segmentation network UNet is developed for the automatic identification of damages. The performance of the controller and detection algorithm is evaluated against traditional methods. The results indicate that the proposed force-redeemed active disturbance rejection controller outperforms traditional PID and ADR controllers in terms of overshoot and adjustment time. Moreover, the developed detection algorithm exhibits superior performance in precision, F1 score, and mean intersection over union compared to the traditional algorithms of FCN, PSPnet, Deeplabv3, and UNet. Overall, this paper advances the technology of autonomous robots for underwater detection in the structures with pile foundation.

The maximum detection distance is usually the primary concern of magnetic anomaly detection (MAD). Intuition tells us that larger object size, stronger magnetization and finer measurement resolution guarantee a further detectable distance. However, the quantitative relationship between detection distance and the above determinants is seldom studied. In this work, unmanned aerial vehicle-based MAD field experiments are conducted on cargo vessels and NdFeB magnets as typical magnetic objects to give a set of visualized magnetic field flux density images. Isometric finite element models are established, calibrated and analyzed according to the experiment configuration. A maximum detectable distance map as a function of target size and measurement resolution is then obtained from parametric sweeping on an experimentally calibrated finite element analysis model. We find that the logarithm of detectable distance is positively proportional to the logarithm of object size while negatively proportional to the logarithm of resolution, within the ranges of 1 m~500 m and 1 pT~1 μT, respectively. A three-parameter empirical formula (namely distance-size-resolution logarithmic relationship) is firstly developed to determine the most economic sensor configuration for a given detection task, to estimate the maximum detection distance for a given magnetic sensor and object, or to evaluate minimum detectable object size at a given magnetic anomaly detection scenario.

Astragalus mongholicus is a traditional Chinese medicine (TCM) with important medicinal value and is widely used worldwide. Heat shock (HSF) transcription factors are among the most important transcription factors in plants and are involved in the transcriptional regulation of various stress responses, including drought, salinity, oxidation, osmotic stress, and high light, thereby regulating growth and developmental processes. However, the HFS gene family has not yet been identified in A. mongholicus, and little is known regarding the role of HSF genes in A. mongholicus. This study is based on whole genome analysis of A. mongholicus, identifying a total of 22 AmHSF genes and analyzing their physicochemical properties. Divided into three subgroups based on phylogenetic and gene structural characteristics, including subgroup A (12), subgroup B (9), and subgroup C (1), they are randomly distributed in 8 out of 9 chromosomes of A. mongholicus. In addition, transcriptome data and quantitative real time polymerase chain reaction (qRT-PCR) analyses revealed that AmHSF was differentially transcribed in different tissues, suggesting that AmHSF gene functions may differ. Red and blue light treatment significantly affected the expression of 20 HSF genes in soilless cultivation of A. mongholicus seedlings. AmHSF3, AmHSF3, AmHSF11, AmHSF12, and AmHSF14 were upregulated after red light and blue light treatment, and these genes all had light-corresponding cis-elements, suggesting that AmHSF genes play an important role in the light response of A. mongholicus. Although the responses of soilless-cultivated A. mongholicus seedlings to red and blue light may not represent the mature stage, our results provide fundamental research for future elucidation of the regulatory mechanisms of HSF in the growth and development of A. mongholicus and its response to different light conditions.

Background: Suppressor APC domain containing 2 (SAPCD2) is involved in cell cycle regulation and its mRNA levels are higher in cancer tissues. But, the function of SAPCD2 in cancer development remains unclear.Objective: To generate mouse monoclonal antibodies (mAbs) specific to SAPCD2 and thus clarify the function of SAPCD2 in the development of gastric carcinoma (GC).Methods: Purified SAPCD2-GST immunized BALB/c mouse spleen cells were collected and fused with myeloma cells to obtain monoclonal antibody hybridoma. A group of monoclonal antibodies exhibiting high specificity and sensitivity against SAPCD2 has been generated and characterized by IHC, WB, IP, IF, and ELISA. By immunohistochemical analysis, the SAPCD2 expression was evaluated in 228 clinical samples of gastric mucosal lesions, including precancerous lesions and GC samples.Results: We identified a highly specific and sensitive clone of s12 in eukaryotic cells and performed an IHC analysis. Rechargeable aqueous zinc metal battery has been one of the next‐generation energy storage systems with a promising future due to the advantages of aqueous electrolyte and metallic zinc anode. Among many cathode materials, vanadium‐based materials with considerable specific capacity, adjustable crystal structure, and multiple valence states are very promising cathodes, but the electrochemical performance of vanadium‐based zinc metal batteries faces severe challenges such as the limited and declining capacity and sluggish ion/e− transport kinetics process. Herein, taking VO2 as an example, the crystal structure and energy storage mechanism of VO2 cathode are briefly introduced. The challenges of aqueous Zn/VO2 battery are analyzed in depth. Significantly, the current advanced modification strategies for aqueous Zn/VO2 battery are summarized and discussed in detail, and the future development is also prospected. This article can provide some significant references for the development of aqueous Zn/VO2 battery and the guiding significance for other metal ion battery.

Abstract The skin is the largest organ in the human body. Various skin environments on its surface constitutes a complex ecosystem. One of the characteristics of the skin micro‐ecosystem is low biomass, which greatly limits a comprehensive identification of the microbial species through sequencing. In this study, deep‐shotgun sequencing (average 21.5 Gigabyte (Gb)) from 450 facial samples and publicly available skin metagenomic datasets of 2069 samples to assemble a Unified Human Skin Genome (UHSG) catalog is integrated. The UHSG encompasses 813 prokaryotic species derived from 5779 metagenome‐assembled genomes, among which 470 are novel species covering 20 phyla with 1385 novel assembled genomes. Based on the UHSG, the core functions of the skin microbiome are described and the differences in amino acid metabolism, carbohydrate metabolism, and drug resistance functions among different phyla are identified. Furthermore, analysis of secondary metabolites of the near‐complete genomes further find 1220 putative novel secondary metabolites, several of which are found in previously unknown genomes. Single nucleotide variant (SNV) reveals a possible skin protection mechanism: the negative selection process of the skin environment to conditional pathogens. UHSG offers a convenient reference database that will facilitate a more in‐depth understanding of the role of skin microorganisms in the skin.

Here, the authors present an expanded version of the Cultivated Genome Reference (CGR), termed CGR2, a catalog that includes 3324 high-quality draft genomes based on gut bacterial isolates from Chinese individuals, and classifies 527 species from 8 phyla, including 179 previously unidentified species, and provides information of secondary metabolite biosynthetic gene clusters and gut phage-bacteria interactions.

IntroductionLongxi bacon is a traditional fermented meat from Gansu province, China. The ripening process of the bacon is crucial for quality and flavor. The aim of this study was to gain deeper knowledges on the bacterial and fungal community diversity and the changes of chemical components including fatty acids and volatile compounds at different time points during the ripening of the bacon and to understand the relationship between microbial profiles and the chemical components related the bacon flavor.MethodsBacon samples were collected from days 0, 15, 30, 60 and 90. The bacterial and fungal compositions were analyzed with next generation sequencing targeting the 16S rDNA loci for bacteria and ITS loci for fungi. The fatty acids and the volatile components were analyzed by headspace solid phase micro extraction followed by gas chromatography/mass spectrometry (HS-SPME-GC/MS).ResultsWe found that the abundance of bacteria in bacon was higher than that of fungi, and Psychrobacter, Brochothrix, Phoma and Trichoderma was the dominant bacon’s population. The largest contributors of volatiles were aldehydes, ketones and esters, and the main fatty acids were palmitic, oleic and linoleic acids. Pearson correlation analysis between microbial succession and key flavor substances showed that the production of Longxi bacon flavor is the result of a combination of bacteria and fungi. Ten bacteria genera and six fungi genera were determined as functional core microbiota for the flavor production based their dominance and functionality in microbial community. In addition, bacteria and fungi are involved in the oxidation and hydrolysis of fatty acids during the ripening of bacon, which also contributes to the formation of bacon flavor.DiscussionThis study provides a comprehensive analysis of the key microbiota involved in shaping bacon’s distinctive flavor. Here, the results presented should provide insight into the influence of the microenvironment on the microbial community in bacon and lay a foundation for further investigations into the food ecology of bacon.

With the increasing demand for biomarker detection in wearable electronic devices, flexible biosensors have garnered significant attention. Additionally, graphene field-effect transistors (GFETs) have emerged as key components for constructing biosensors, owing to their high sensitivity, multifunctionality, rapid response, and low cost. Leveraging the advantages of flexible substrates, such as biocompatibility, adaptability to complex environments, and fabrication flexibility, flexible GFET sensors exhibit promising prospects in detecting various biomarkers. This review provides a concise summary of design strategies for flexible GFET biosensors, including non-encapsulated gate without dielectric layer coverage and external gate designs. Furthermore, notable advancements in sensing applications of biomolecules, such as proteins, glucose, and ions, are highlighted. Finally, we discuss the future challenges and prospects in this field, aiming to inspire researchers to address these issues in their further investigations.

Abstract Background Increasing studies have demonstrated potential disproportionate functional and ecological contributions of rare taxa in a microbial community. However, the study of the microbial rare biosphere is hampered by their inherent scarcity and the deficiency of currently available techniques. Sample-wise cross contaminations might be introduced by sample index misassignment in the most widely used metabarcoding amplicon sequencing approach. Although downstream bioinformatic quality control and clustering or denoising algorithms could remove sequencing errors and non-biological artifact reads, no algorithm could eliminate high quality reads from sample-wise cross contaminations introduced by index misassignment, making it difficult to distinguish between bona fide rare taxa and potential false positives in metabarcoding studies. Results We thoroughly evaluated the rate of index misassignment of the widely used NovaSeq 6000 and DNBSEQ-G400 sequencing platforms using both commercial and customized mock communities, and observed significant lower (0.08% vs. 5.68%) fraction of potential false positive reads for DNBSEQ-G400 as compared to NovaSeq 6000. Significant batch effects could be caused by stochastically introduced false positive or false negative rare taxa. These false detections could also lead to inflated alpha diversity of relatively simple microbial communities and underestimated that of complex ones. Further test using a set of cow rumen samples reported differential rare taxa by different sequencing platforms. Correlation analysis of the rare taxa detected by each sequencing platform demonstrated that the rare taxa identified by DNBSEQ-G400 platform had a much higher possibility to be correlated with the physiochemical properties of rumen fluid as compared to NovaSeq 6000 platform. Community assembly mechanism and microbial network correlation analysis indicated that false positive or negative rare taxa detection could lead to biased community assembly mechanism and identification of fake keystone species of the community. Conclusions We highly suggest proper positive/negative/blank controls, technical replicate settings, and proper sequencing platform selection in future amplicon studies, especially when the microbial rare biosphere would be focused.

In this paper, quinoa dough was used as raw material to investigate the effects of different salt (NaCl, KCl, sodium caseinate) on the texture and rheological properties of quinoa dough and noodles such as hardness, chewiness, viscosity and structural recovery. Sensory evaluation showed that the smoothness and elasticity of quinoa dough were better when the amount of water added was 50%. NaCl, KCl and sodium caseinate could affect the viscosity, elasticity, hardness and chewiness of the quinoa dough. The quinoa dough prepared with 1.5% sodium caseinate+1% KCl in quinoa flour had moderate viscosity, good hardness and elasticity. The rheological experiment results showed that the addition of NaCl, KCl and sodium caseinate could improve the G'', G', viscosity of the dough. The addition of 1.5% sodium caseinate+1% KCl could increase the G'', G', and viscosity of quinoa dough. Adding NaCl and sodium caseinate to quinoa noodles could significantly improve the water absorption of quinoa noodles, NaCl could reduce the dry matter loss rate of quinoa noodles, KCl had the opposite effect, and sodium caseinate combined with 1% KCl could increase its water absorption while reducing dry matter. The material loss rate reached the optimum under the condition of 2.0% sodium caseinate and 1% KCl, and the recovery ability was the best. The scanning of noodles morphology showed that sodium caseinate and 1% KCl could increase the density of the surface lamellae of quinoa noodles. The results showed that the elasticity, hardness and chewiness of quinoa dough reached the maximum when the amount of sodium caseinate and KCl was 1.5% and 1%. Meanwihle, the results quinoa noodles showed that 2.0% sodium caseinate and 1% KCl could improve the cooking loss and dry matter loss rate, reduce the turbidity of noodle soup, and increase the hardness of quinoa noodles.

Global pandemics such as COVID-19 have resulted in significant global social and economic disruption. Although polymerase chain reaction (PCR) is recommended as the standard test for identifying the SARS-CoV-2, conventional assays are time-consuming. In parallel, although artificial intelligence (AI) has been employed to contain the disease, the implementation of AI in PCR analytics, which may enhance the cognition of diagnostics, is quite rare. The information that the amplification curve reveals can reflect the dynamics of reactions. Here, we present a novel AI-aided on-chip approach by integrating deep learning with microfluidic paper-based analytical devices (µPADs) to detect synthetic RNA templates of the SARS-CoV-2 ORF1ab gene. The µPADs feature a multilayer structure by which the devices are compatible with conventional PCR instruments. During analysis, real-time PCR data were synchronously fed to three unsupervised learning models with deep neural networks, including RNN, LSTM, and GRU. Of these, the GRU is found to be most effective and accurate. Based on the experimentally obtained datasets, qualitative forecasting can be made as early as 13 cycles, which significantly enhances the efficiency of the PCR tests by 67.5% (∼40 min). Also, an accurate prediction of the end-point value of PCR curves can be obtained by GRU around 20 cycles. To further improve PCR testing efficiency, we also propose AI-aided dynamic evaluation criteria for determining critical cycle numbers, which enables real-time quantitative analysis of PCR tests. The presented approach is the first to integrate AI for on-chip PCR data analysis. It is capable of forecasting the final output and the trend of qPCR in addition to the conventional end-point Cq calculation. It is also capable of fully exploring the dynamics and intrinsic features of each reaction. This work leverages methodologies from diverse disciplines to provide perspectives and insights beyond the scope of a single scientific field. It is universally applicable and can be extended to multiple areas of fundamental research.

Small extracellular vesicle (sEV) is an emerging source of potential biomarkers of Alzheimer's disease (AD), but the role of microRNAs (miRNAs) in sEV is not well understood. In this study, we conducted a comprehensive analysis of sEV-derived miRNAs in AD using small RNA sequencing and coexpression network analysis. We examined a total of 158 samples, including 48 from AD patients, 48 from patients with mild cognitive impairment (MCI), and 62 from healthy controls. We identified an miRNA network module (M1) that was strongly linked to neural function and showed the strongest association with AD diagnosis and cognitive impairment. The expression of miRNAs in the module was decreased in both AD and MCI patients compared to controls. Conservation analysis revealed that M1 was highly preserved in the healthy control group but dysfunctional in the AD and MCI groups, suggesting that changes in the expression of miRNAs in this module may be an early response to cognitive decline prior to the appearance of AD pathology. We further validated the expression levels of the hub miRNAs in M1 in an independent population. The functional enrichment analysis showed that 4 hub miRNAs might interact with a GDF11-centered network and play a critical role in the neuropathology of AD. In summary, our study provides new insights into the role of sEV-derived miRNAs in AD and suggests that M1 miRNAs may serve as potential biomarkers for the early diagnosis and monitoring of AD.

This study aimed to identify potential circular RNA (circRNA), microRNA (miRNA) and mRNA biomarkers as well as their underlying regulatory mechanisms in papillary thyroid carcinoma (PTC). Three microarray datasets from the Gene Expression Omnibus database as well as expression data and clinical phenotype from The Cancer Genome Atlas (TCGA) were downloaded, followed by differential expression, functional enrichment, protein–protein interaction (PPI), and module analyses. The support vector machine (SVM)-recursive feature elimination (RFE) algorithm was used to screen the key circRNAs. Finally, the mRNA-miRNA-circRNA regulatory network and competitive endogenous RNA (ceRNA) network were constructed. The prognostic value and clinical correlations of key mRNAs were investigated using TCGA dataset, and their expression was validated using the UALCAN database. A total of 1039 mRNAs, 18 miRNAs and 137 circRNAs were differentially expressed in patients with PTC. A total of 37 key circRNAs were obtained using the SVM-RFE algorithm, whereas 46 key mRNAs were obtained from significant modules in the PPI network. A total of 11 circRNA-miRNA pairs and 40 miRNA-mRNA pairs were predicted. Based on these interaction pairs, 46 circRNA-miRNA-mRNA regulatory pairs were integrated, of which 8 regulatory pairs in line with the ceRNA hypothesis were obtained, including two circRNAs (circ_0004053 and circ_0028198), three miRNAs (miR-199a-5p, miR-199b-5p, and miR-7-5p), and five mRNAs, namely APOA2, CCL20, LPAR5, MFGE8, and TIMP1. Survival analysis showed that LPAR5 expression was associated with patient survival. APOA2 expression showed significant differences between metastatic and non-metastatic tumors, whereas CCL20, LPAR5, MFGE8 and TIMP1 showed significant differences between metastatic and non-metastatic lymph nodes. Overall, we identified several potential targets and regulatory mechanisms involved in PTC. APOA2, CCL20, LPAR5, MFGE8, and TIMP1 may be correlated with PTC metastasis.

ObjectiveLiquid metal (LM) nowadays is considered a new biomedical material for medical treatment. The most common application of LM in medical therapy is taking LM as a carrier for oncology therapeutics. However, the feasibility and direct effect of LM in tumor treatment are still unknown, and how to delineate the negative resection margin (NRM) of the tumor is also a crucial problem in surgery. We aimed to inject LM into interstitial channels of extremities of mice to overlay the surface of the primary tumor to investigate the effect of LM on inhibiting tumor growth and highlight the NRM of the tumor.MethodsIn this study, all 50 BALB/c-nude female mice were used to construct the transplanted HepG2-type hepatocellular carcinoma model. One week after the establishment of the model, the mice were divided into three groups, named LM group, PBS group and Control group by injecting different liquid materials into the forelimb interstitial channel of the mice. T2WI image on MRI and Magneto-acoustic tomography (MAT) were used to show the distribution of LM and PBS in vivo. The group comparisons of tumor growth and blood tests were evaluated by one-way ANOVA and post-hoc analysis. And the biocompatibility of LM to BALB/c nude mice was evaluated by histopathological analysis of LM group and control group.ResultsThe volume change ratio of tumor was significantly lower in LM group than in PBS and Control group after 10 days of grouping. Compared with PBS and Control group, the main indexes of blood tests in LM group were significantly lower and close to normal level. In addition, the distribution of LM in vivo could be clearly observed under T2WI anatomic images and the crossprofile of the tumor in MAT. LM also has a obvious contrast in MRI T2WI and enhanced the amplitude of imaging signal in MAT.ConclusionLM may inhibit the growth of transplanted hepatoma tumor through tumor encapsulation. In vivo, tumor imaging and LM distribution imaging were achieved by MRI T2WI, which verified that LM injected with interstitial injection made the NRM of tumor more prominent and had the potential of being MRI contrast agent. At the same time, LM could also be a new conductive medium to improve the imaging quality of MAT. Moreover, LM performed mild biocompatibility.

Magnetic current imaging is deemed an emerging powerful technique for visualizing electrical currents in electronic devices. However, the existing magnetic-field-based Fourier Transform back-evolution method is limited by its mono-function of imaging the magnitude of current density in devices under test, and subject to background noise distortion. Here, we developed a novel vectorial current density imaging method based on the detection of the magnetic field gradient generated by current carrying conductors. A closed form solution of current density inversion was analytically derived and numerically verified. Experiments were conducted by scanning tri-axial fluxgate sensor over different shapes of electrical wires. The results show that a current density resolution of 24.15 mA/mm2, probe-to-sample separation of 2 mm, and spatial resolution of 0.69 mm were achieved over a maximum scanning area of 300 mm × 300 mm. Such a method is verified to be capable of simultaneously imaging both magnitude and directions of current density, which is a promising technique for in situ noninvasive inspection for the power electronic and semiconductor industry.

Abstract Background The sequences of several important mitochondrion-encoded genes involved in respiration in higher plants are interrupted by introns. Many nuclear-encoded factors are involved in splicing these introns, but the mechanisms underlying this splicing remain unknown. Results We isolated and characterized a rice mutant named floury shrunken endosperm 5 (fse5). In addition to having floury shrunken endosperm, the fse5 seeds either failed to germinate or produced seedlings which grew slowly and died ultimately. Fse5 encodes a putative plant organelle RNA recognition (PORR) protein targeted to mitochondria. Mutation of Fse5 hindered the splicing of the first intron of nad4, which encodes an essential subunit of mitochondrial NADH dehydrogenase complex I. The assembly and NADH dehydrogenase activity of complex I were subsequently disrupted by this mutation, and the structure of the mitochondria was abnormal in the fse5 mutant. The FSE5 protein was shown to interact with mitochondrial intron splicing factor 68 (MISF68), which is also a splicing factor for nad4 intron 1 identified previously via yeast two-hybrid (Y2H) assays. Conclusion Fse5 which encodes a PORR domain-containing protein, is essential for the splicing of nad4 intron 1, and loss of Fse5 function affects seed development and seedling growth.

There are currently rising applications of battery electric buses in public transportation system, given their energy efficiency and societal benefits. However, limited range and time-consuming charging processes significantly reduce the operational efficiency of battery electric buses system. Thanks to fast charging technology, battery electric buses can run continuously through rational planning of fast charging system. Therefore, to achieve an effective and economic electric transit route network, electric transit route network and fast charging stations should be simultaneously and systematically deployed. This study develops a bi-level programming framework to formulate the electric transit route network design problem. Where, the upper-level model is to determine the route structure and charging station location to optimize the total costs of both user and operator, the lower-level passenger assignment model is to calculate user cost for upper-level model. Then, the proposed model is solved by a modified genetic algorithm (GA), which can search all possible route structures through specifically designed genetic operators. A numerical example from Swiss road network is employed to demonstrate the applicability of bi-level model and performance of modified GA.

ObjectiveThis study analyzed the differences in the cerebral blood flow (CBF) between unilateral Sudden Sensorineural Hearing Loss (SSNHL) patients and healthy controls (HCs). We also investigated CBF differences in auditory-related areas in patients with left- and right-sided SSNHL (lSSNHL and rSSNHL) and HCs. We further explore the correlation between unilateral SSNHL characteristics and changes in the CBF.Methods36 patients with unilateral SSNHL (15 males and 21 females, 40.39 ± 13.42 years) and 36 HCs (15 males and 21 females, 40.39 ± 14.11 years) were recruited. CBF images were collected and analyzed using arterial spin labeling (ASL). CereFlow software was used for the post-processing of the ASL data to obtain the CBF value of 246 subregions within brainnetome atlas (BNA). The Two-sample t-test was used to compare CBF differences between SSNHL patients and HCs. One-way ANOVA or Kruskal-Wallis test was used to compare the CBF difference of auditory-related areas among the three groups (lSSNHL, rSSNHL, and HCs). Then, the correlation between CBF changes and specific clinical characteristics were calculated.ResultsThe SSNHL patients exhibited decreased CBF in the bilateral middle frontal gyrus (MFG, MFG_7_1 and MFG_7_3), the contralateral precentral gyrus (PrG, PrG_6_3) and the bilateral superior parietal lobule (SPL, bilateral SPL_5_1, SPL_5_2, and ipsilateral SPL_5_4), p < 0.0002. Compared with HCs, unilateral SSNHL patients exhibited increased rCBF in the bilateral orbital gyrus (OrG, OrG_6_5), the bilateral inferior temporal gyrus (ITG, contralateral ITG_7_1 and bilateral ITG_7_7), p < 0.0002. lSSNHL showed abnormal CBF in left BA21 caudal (p = 0.02) and left BA37 dorsolateral (p = 0.047). We found that the CBF in ipsilateral MFG_7_1 of SSNHL patients was positively correlated with tinnitus Visual Analog Scale (VAS) score (r = 0.485, p = 0.008).ConclusionOur preliminary study explored CBF pattern changes in unilateral SSNHL patients in auditory-related areas and non-auditory areas, suggesting that there may exist reduced attention and some sensory compensation in patients with SSNHL. These findings could advance our understanding of the potential pathophysiology of unilateral SSNHL.

To address the fracture control problem of X80 unequal-diameter tee used in China-Russia Eastern Gas Pipeline under low temperature environment (－45 ℃), the finite element model for X80 unequal-diameter tee with defects was built taking into consideration the influence of wall thickness effect and structural stress concentration of the tee. The crack initiation toughness of X80 tee body and welds was calculated by applying the failure evaluation methods of Guide to methods for assessing the acceptability of flaws in metallic structures (BS 7910-2019) and Fitness-For-Service (API 579-1/ASME FFS-1-2016). Charpy impact toughness index of X80 tee at －45 ℃ was determined through the relation between fracture toughness and Charpy impact energy. The results show that the fracture toughness at the tee weld should be greater than 37.79 MPa·m1/2, and the minimum value of each Charpy impact energy should not be less than 32.22 J. The stress intensity factor of the inner surface at the shoulder of the tee body is the largest, with the fracture toughness greater than 42.46 MPa·m1/2, and the minimum value of each Charpy impact energy should not be less than 38.85 J. According to the requirements of domestic and foreign pipe fittings standards, the low temperature toughness index of the D 1 422×1 219 mm X80 unequal-diameter tee in China-Russia Eastern Gas Pipeline Project was determined.

Objective Two experiments were conducted to investigate the effects of dietary sucralose on diet preference and growth performance of weaned piglets, and a third experiment was a 28-d safety study to examine if high-dose sucralose could affect the health state of weaned piglets. Methods In experiment one, 48 piglets had free access to a corn-soybean based diet and the same diet supplemented with 150 mg/kg sucralose for 15 d. In experiment two, 180 piglets were blocked into 5 treatments with 6 replications. They were fed basal diets supplemented with 0, 75, 150, 225, and 300 mg/kg sucralose for 28 days. In experiment three, 108 piglets were randomly assigned to 3 treatments and fed diets supplemented with 0, 150 (suitable level), and 1,500 (ten-fold suitable level) mg/kg sucralose for 28 d. Quaternary biogenic gas reservoirs are extensively developed in the Sanhu Depression of the Qaidam Basin, where the largest domestic biogenic gas production base has been built up. In recent years, however, the exploration of biogenic gas there encounters a variety of difficulties, such as the identification of true and false seismic abnormality, the determination of micro-relief structure and the identification and description of lithologic traps, which are the bottlenecks restricting its exploration breakthrough. In this paper, hydrocarbon accumulation patterns of biogenic gas were studied. Then, based on fine structure interpretation results, combined with laboratory experiments, the sealing capacity, distribution range, formation time and genesis of salt crust were discussed, and the salt crust covered hydrocarbon accumulation pattern of biogenic gas was put forward. Finally, combined with the conditions of source rocks and reservoirs, the exploration prospect of salt crust covered biogenic gas reservoirs was predicted. And the following research results were obtained. First, the Quaternary in the Sanhu Depression is composed of sandstone–mudstone interbeds of shore-shallow lake facies, with superior conditions of source rocks and reservoirs. Many source–reservoir–caprock assemblages are vertically formed. The biogenic gas has the characteristics of dynamic hydrocarbon accumulation, i.e., continuous migration, accumulation, diffusion and re-accumulation. Second, under the effect of Himalayan movement, the climate changes frequently and the salinity of the ancient lake is zoned in the Sanhu Depression. A freshwater area is formed at the inlet of the river in the south and a brine area is formed in the north. Under the influence of evaporation, the phreatic water in the north is salinized continuously, and a set of extensively distributed salt crust with steady thickness and a strong sealing capacity is formed at the eastern part of Lingjian fault. Third, the biogenic gas generated by dark mudstone of lacustrine facies in the depression center migrates laterally and vertically to Lingjian fault zone and gets accumulated under the sealing of salt crust. In conclusion, high-quality source rocks and reservoirs and the salt crust with a strong sealing capacity in the Sanhu Depression constitute a good spatial–temporal configuration relationship of source rocks, reservoirs and cap rocks, so salt crust covered biogenic gas reservoirs can be formed to provide abundant natural gas resources with a broad exploration prospect. Operating short turning line is an efficient strategy to satisfy the unevenly distributed demand during peak periods while reducing operational cost. However, for the battery electric bus (BEB) system, the application of the strategy is challenging due to the disadvantages of BEBs, such as limited driving mileage and long charging time. Improper vehicle configuration and charging scheduling may dramatically increase the operational cost and cut the benefits of these strategies. In this work, we propose a general framework to design an effective short turning strategy for the BEB system at a tactical planning level. First, the trade-off relationship between the battery capacity and the average trip time is identified by modeling the BEBs operations. Second, a microeconomic model is formulated to jointly optimize the frequencies and charging schedules of the whole bus line and the short turning line, to effectively minimize passengers’ waiting time and operational cost. Finally, numerical experiments have been carried out for an illustrative linear line to demonstrate the potential benefits of the sub-line operating strategy compared with the normal operation.

The COVID-19 pandemic has resulted in significant global social and economic disruption. The highly transmissive nature of the disease makes rapid and reliable detection critically important. Point-of-care (POC) tests involve performing diagnostic tests outside of a laboratory that produce a rapid and reliable result. It therefore allows the diagnostics of diseases at or near the patient site. Paper-based POC tests have been gaining interest in recent years as they allow rapid, low-cost detection without the need for external instruments. In this review, we focus on the development of paper-based POC devices for the detection of SARS-CoV-2. The review first introduces the principles of detection methods that are available to paper-based devices. It then summarizes the state-of-the-art paper devices and their analytical performances. The advantages and drawbacks among methods are also discussed. Finally, limitations of the existing devices are discussed, and prospects are given with the hope to identify research opportunities and directions in the field. We hope this review will be helpful for researchers to develop a clinically useful and economically efficient paper-based platform that can be used for rapid, accurate on-site diagnosis to aid in identifying acute infections and eventually contain the COVID-19 pandemic.

Subcellular mRNA localization is an evolutionarily conserved mechanism to spatially and temporally drive local translation and, in turn, protein targeting. Hence, this mechanism achieves precise control of gene expression and establishes functional and structural networks during cell growth and development as well as during stimuli response. Since its discovery in ascidian eggs, mRNA localization has been extensively studied in animal and yeast cells. Although our knowledge of subcellular mRNA localization in plant cells lags considerably behind other biological systems, mRNA localization to the endoplasmic reticulum (ER) has also been well established since its discovery in cereal endosperm cells in the early 1990s. Storage protein mRNA targeting to distinct subdomains of the ER determines efficient accumulation of the corresponding proteins in different endosomal storage sites and, in turn, underlies storage organelle biogenesis in cereal grains. The targeting process requires the presence of RNA localization elements, also called zipcodes, and specific RNA-binding proteins that recognize and bind these zipcodes and recruit other factors to mediate active transport. Here, we review the current knowledge of the mechanisms and functions of mRNA localization to the ER in plant cells and address directions for future research.

Vibrio parahaemolyticus has emerged as a significant enteropathogen in human and marine habitats worldwide, notably in regions where aquaculture products constitute a major nutritional source. It is a growing cause of diseases including gastroenteritis, wound infections, and septicemia. Serotyping assays use commercially available antisera to identify V. parahaemolyticus strains, but this approach is limited by high costs, complicated procedures, cross-immunoreactivity, and often subjective interpretation. By leveraging high-throughput sequencing technologies, we developed an in silico method based on comparison of gene clusters for lipopolysaccharide (LPSgc) and capsular polysaccharide (CPSgc) by firstly using the unique-gene strategy. The algorithm, VPsero, which exploits serogroup-specific genes as markers, covers 43 K and all 12 O serogroups in serotyping assays. VPsero is capable of predicting serotypes from assembled draft genomes, outputting LPSgc/CPSgc sequences, and recognizing possible novel serogroups or populations. Our tool displays high specificity and sensitivity in prediction toward V. parahaemolyticus strains, with an average sensitivity in serogroup prediction of 0.910 for O and 0.961 for K serogroups and a corresponding average specificity of 0.990 for O and 0.998 for K serogroups.

With the development of underwater exploration, underwater networking is in urgent demand. At present, underwater wireless optical communication (UWOC) is primarily based on line-of-sight (LOS) communication links. However, the underwater environment is so complicated that LOS communication links are easily affected by a couple of factors such as air bubbles, turbidity, oceanic turbulence, and so on. We put forward novel UWOC links using the total reflection at the air-water interface, which can mitigate those phenomena. This paper aims to investigate a UWOC link based on the total reflection at the air-water interface. In our work, we achieved the maximum data rate of 300 Mb/s and a bit error rate (BER) of 3.10 × 10−3 under the forward error correction (FEC) with the total reflection angle of 7°. Furthermore, we verified the performance of the total reflection-based UWOC links at the air-water interface in the presence of waves and evaluated the impact on the UWOC links when the frequency and amplitude of the waves varied.

Mangroves are one of the most productive and biologically diverse ecosystems, with unique plants, animals, and microorganisms adapted to the harsh coastal environments. Although fungi are widely distributed in the mangrove ecosystem and they are playing an important role in the decomposition of organic matter, their genomic profiles are still poorly understood. In this study, we isolated seven Ascomycota fungi (Westerdykella dispersa F012, Trichoderma lixii F014, Aspergillus tubingensis F023, Penicillium brefeldianum F032, Neoroussoella solani F033, Talaromyces fuscoviridis F034, and Arthrinium marii F035) from rhizospheres of two mangroves of Kandelia obovata and Acanthus ilicifolius. We sequenced and assembled the whole genome of these fungi, resulting in size ranging from 29 to 48 Mb, while contig N50 from 112 to 833 Kb. We generated six novel fungi genomes except A. tubingensis, and the gene completeness and genome completeness of all seven genomes are higher than 94%. Comparing with non-mangrove fungi, we found Carbohydrate-Binding Modules (CBM32), a subfamily of carbohydrate active enzymes, only detected in two mangrove fungi. Another two subfamilies, Glycoside Hydrolases (GH6) and Polysaccharide Lyases (PL4), were significantly different in gene copy number between K. obovata and A. ilicifolius rhizospheres (P-value 0.041 for GH6, 0.047 for PL4). These findings may indicate an important influence of mangrove environments or hosts on the ability of decomposition in rhizosphere fungi. Secondary metabolite biosynthesis gene clusters were detected and we found the mangrove fungi averagely contain 18 Type I Polyketide (t1pks) synthase, which was significantly higher than 13 in non-mangrove fungi (P-value 0.048), suggesting their potential roles in producing bioactive compounds that important for fungi development and ecology. We reported seven mangrove-associated fungal genomes in this study and compared their carbohydrate active enzymes and secondary metabolites (SM) genes with those of non-mangrove fungi, and the results suggest that there are differences in genetic information among fungi in different habitats.

Abstract Background Massively-parallel-sequencing, coupled with sample multiplexing, has made genetic tests broadly affordable. However, intractable index mis-assignments (commonly exceeds 1%) were repeatedly reported on some widely used sequencing platforms. Results Here, we investigated this quality issue on BGI sequencers using three library preparation methods: whole genome sequencing (WGS) with PCR, PCR-free WGS, and two-step targeted PCR. BGI’s sequencers utilize a unique DNA nanoball (DNB) technology which uses rolling circle replication for DNA-nanoball preparation; this linear amplification is PCR free and can avoid error accumulation. We demonstrated that single index mis-assignment from free indexed oligos occurs at a rate of one in 36 million reads, suggesting virtually no index hopping during DNB creation and arraying. Furthermore, the DNB-based NGS libraries have achieved an unprecedentedly low sample-to-sample mis-assignment rate of 0.0001 to 0.0004% under recommended procedures. Conclusions Single indexing with DNB technology provides a simple but effective method for sensitive genetic assays with large sample numbers.

Abstract Background Whole exome sequencing (WES) has been widely used in human genetics research. BGISEQ-500 is a recently established next-generation sequencing platform. However, the performance of BGISEQ-500 on WES is not well studied. In this study, we evaluated the performance of BGISEQ-500 on WES by side-to-side comparison with Hiseq4000, on well-characterized human sample NA12878. Results BGISEQ demonstrated similarly high reproducibility as Hiseq for variation detection. Also, the SNVs from BGISEQ data is highly consistent with Hiseq results (concordance 96.5%~ 97%). Variation detection accuracy was subsequently evaluated with data from the genome in a bottle project as the benchmark. Both platforms showed similar sensitivity and precision in SNV detection. While in indel detection, BGISEQ showed slightly higher sensitivity and lower precision. The impact of sequence depth and read length on variation detection accuracy was further analyzed, and showed that variation detection sensitivity still increasing when the sequence depth is larger than 100x, and the impact of read length is minor when using 100x data. Conclusions This study suggested that BGISEQ-500 is a qualified sequencing platform for WES.

Abstract Objective In this study, crucial genes and microRNAs (miRNAs) associated with the progression, staging, and prognosis of papillary thyroid cancer (PTC) were identified. Methods Four PTC datasets, including our own mRNA-sequencing (mRNA-seq) dataset and three public datasets downloaded from Gene Expression Omnibus and The Cancer Genome Atlas, were used to analyze differentially expressed genes (DEGs) and miRNAs (DEMs) between PTC tumor tissues and paired normal tissues (control). Gene ontology (GO) terms and pathways associated with these DEGs were identified, and protein–protein interactions (PPIs) were analyzed. Additionally, an miRNA-mRNA regulatory network was constructed and the functions of DEMs were explored. Finally, miRNAs/mRNAs associated with tumor staging and prognosis were identified. The expression levels of several key genes and miRNAs were validated by qRT-PCR. Results Numerous DEGs and DEMs were identified between tumor and control groups in four datasets. The DEGs were significantly enriched in cell adhesion and cancer-related GO terms and pathways. In the constructed PPI network, ITGA2, FN1, ICAM1, TIMP1 and CDH2 were hub proteins. In the miRNA-mRNA negative regulatory networks, miR-204-5p regulated the largest number of target genes, such as TNFRSF12A. miR-146b, miR-204, miR-7-2, and FN1 were associated with tumor stage in PTC, and TNFRSF12A and CLDN1 were related to prognosis. Conclusions Our results suggested the important roles of ITGA2, FN1, ICAM1, TIMP1 and CDH2 in the progression of PTC. miR-204-5p, miR-7-2, and miR-146b are potential biomarkers for PTC staging and FN1, CLDN1, and TNFRSF12A may serve as markers of prognosis in PTC.

Co-barcoded reads originating from long DNA fragments (mean length >30 kbp) maintain both single base level accuracy and long-range genomic information. We propose a pipeline, stLFRsv, to detect structural variation using co-barcoded reads. stLFRsv identifies abnormal large gaps between co-barcoded reads to detect potential breakpoints and reconstruct complex structural variants (SVs). Haplotype phasing by co-barcoded reads increases the signal to noise ratio, and barcode sharing profiles are used to filter out false positives. We integrate the short read SV caller smoove for smaller variants with stLFRsv. The integrated pipeline was evaluated on the well-characterized genome HG002/NA24385, and 74.5% precision and a 22.4% recall rate were obtained for deletions. stLFRsv revealed some large variants not included in the benchmark set that were verified by long reads or assembly. For the HG001/NA12878 genome, stLFRsv also achieved the best performance for both resource usage and the detection of large variants. Our work indicates that co-barcoded read technology has the potential to improve genome completeness.

Summary: TxNIP (Thioredoxin-interacting protein) is considered as a potential drug target for type 2 diabetes. Although TxNIP expression is correlated with hyperglycemia and glucotoxicity in pancreatic β cells, its regulation in liver cells has been less investigated. In the current study, we aim at providing a better understanding of Txnip regulation in hepatocytes in response to physiological stimuli and in the context of hyperglycemia in db/db mice. We focused on regulatory pathways governed by ChREBP (Carbohydrate Responsive Element Binding Protein) and FoxO1 (Forkhead box protein O1), transcription factors that play central roles in mediating the effects of glucose and fasting on gene expression, respectively. Studies using genetically modified mice reveal that hepatic TxNIP is up-regulated by both ChREBP and FoxO1 in liver cells and that its expression strongly correlates with fasting, suggesting a major role for this protein in the physiological adaptation to nutrient restriction.

The emergence of the novel human coronavirus, SARS-CoV-2, causes a global COVID-19 (coronavirus disease 2019) pandemic. Here, we have characterized and compared viral populations of SARS-CoV-2 among COVID-19 patients within and across households. Our work showed an active viral replication activity in the human respiratory tract and the co-existence of genetically distinct viruses within the same host. The inter-host comparison among viral populations further revealed a narrow transmission bottleneck between patients from the same households, suggesting a dominated role of stochastic dynamics in both inter-host and intra-host evolutions.

Objective: To evaluate the efficacy and safety of preoperative embolization and vertebroplasty in the treatment of aggressive hemangioma. Methods: A retrospective clinical review of patients diagnosed with aggressive vertebral hemangiomas was conducted. All the patients were assigned to three groups according to the treatment strategies: patients in Group A underwent embolization and decompression with internal fixation, patients in Group B underwent vertebroplasty and decompression with internal fixation, patients in Group C received all three treatments. Clinical indexes were compared within three groups. Results: There were 16 patients received embolization and decompression (Group A), 19 patients underwent decompression with vertebroplasty (Group B) and 16 patients in Group C. The operative duration of patients in group A (198.33 ± 38.43 min) were less than another two groups ( p = 0.001). The intraoperative blood loss of patients in group C was 713.33 ± 165.13 mL, which was significantly less than group A and group B ( p = 0.045). Patients in group C exhibited the lowest volume of drainage on POD 1 (178.33 ± 66.76 mL), which showed significant difference compared with group A (368.33 ± 191.15 mL, p = 0.01). There was no significant difference of preoperative and postoperative VAS and JOA score among three groups, as well as drainage on POD 2, total volume and hospital duration. Conclusion: Both embolization and vertebroplasty are efficient and safe measures to reduce blood loss in the surgical treatment of aggressive vertebral hemangiomas, combination of all three methods is also competent.

Mental fatigue is a common psychobiological state elected by prolonged cognitive activities. Although, the performance and the disadvantage of the mental fatigue have been well known, its connectivity among the multiareas of the brain has not been thoroughly studied yet. This is important for the clarification of the mental fatigue mechanism. However, the common method of connectivity analysis based on EEG cannot get rid of the interference from strong noise. In this paper, an adaptive feature extraction model based on stacked denoising autoencoder has been proposed. The signal to noise ratio of the extracted feature has been analyzed. Compared with principal component analysis, the proposed method can significantly improve the signal to noise ratio and suppress the noise interference. The proposed method has been applied on the analysis of mental fatigue connectivity. The causal connectivity among the frontal, motor, parietal, and visual areas under the awake, fatigue, and sleep deprivation conditions has been analyzed, and different patterns of connectivity between conditions have been revealed. The connectivity direction under awake condition and sleep deprivation condition is opposite. Moreover, there is a complex and bidirectional connectivity relationship, from the anterior areas to the posterior areas and from the posterior areas to the anterior areas, under fatigue condition. These results imply that there are different brain patterns on the three conditions. This study provides an effective method for EEG analysis. It may be favorable to disclose the underlying mechanism of mental fatigue by connectivity analysis.

The effect of hot working parameters on the microstructure and texture evolution of the hot-deformed Zr-45Ti-5Al-3V alloy was studied by the electron backscatter diffraction (EBSD) technique. It was found that a high density of dislocations were generated when the alloy was deformed at 700 °C/0.001 s−1 and 800 °C/1 s−1. With the increment in hot-deformation temperature and the decrease in strain rate, the dislocation density decreased due to the increase in dynamic recrystallization (DRX) degree. The discontinuous dynamic recrystallization (DDRX) and continuous dynamic recrystallization (CDRX) mechanisms co-existed during the hot working of the Zr-45Ti-5Al-3V alloy at a true strain of 0.7. The texture evolution of the alloy during hot working was characterized and the texture component mainly consisted of {001}, {011}, {110}, and {112} textures. The volume fractions of {001} and {011} textures obviously rose with the reduction in strain rate, whereas those of {110} and {112} textures gradually decreased. At a given strain rate, an increase trend in the volume fraction of the {001} texture was observed with rising hot-deformation temperature, while the volume fraction of the {011} texture first increased and then decreased. An opposite trend was visible in the {112} and {110} texture compared with {011} textures.

To prevent the occurrence of temperature cracking, the induced joints of dams are typically arranged to reduce the tensile strength and orderly guide the crack to form at an induced joint, which can protect the dam structure. The state of the joint surface and the equivalent strength and arrangement of the induced joints have important practical significance for preventing the disorderly cracking of the dam. In this study, the concrete strength, inducer spacing, and concrete age were considered as variable conditions to perform axial tension tests on roller compacted concrete (RCC) bidirectional induced joints. A finite element software was used for numerical simulation, and the simulation results were compared with the experimental results. The equivalent strength of the RCC induced joints increased with the spacing of the induced joints, but the rate of increase gradually declined. The equivalent strength of the RCC induced joints rapidly increased in the early stage, but the strength weakening degree was different at different concrete ages. The strength weakening degree of the induced joint section decreased as the concrete strength increased. These results provide a reference for the setting of induced joints in practical engineering.

The dynamic recrystallization (DRX) behavior of 47Zr-45Ti-5Al-3V alloy was studied by using the experiment and numerical simulation method based on DEFORM-3D software and cellular automata (CA) over a range of deformation temperatures (850 to 1050 °C) and strain rates (10−3 to 100 s−1). The results reveal that the DRX behavior of 47Zr-45Ti-5Al-3V alloy strongly depends on hot-working parameters. With rising deformation temperature (T) and decreasing strain rate (ε˙), the grain size (dDRX) and volume fraction (XDRX) of DRX dramatically boost. The kinetics models of the dDRX and XDRX of DRX grains were established. According to the developed kinetics models for DRX of 47Zr-45Ti-5Al-3V alloy, the distributions of the dDRX and XDRX for DRX grains were predicted by DEFORM-3D. DRX microstructure evolution is simulated by CA. The correlation of the kinetics model is verified by comparing the dDRX and XDRX between the experimental and finite element simulation (FEM) results. The nucleation and growth of dynamic recrystallization grains in 47Zr-45Ti-5Al-3V alloy during hot-working can be simulated accurately by CA simulation, comparing with FEM.

Various isoforms of myocyte enhancer factor 2 (MEF2) have been shown to play a role in the activation of rat hepatic stellate cells (HSCs) in culture. The signals that regulate MEF2 in HSCs are unknown. In addition, whether MEF2s regulate the activation of human HSCs (H-HSCs) is unclear. Here, we studied the expression and function of MEF2s in H-HSCs. Our data showed that the levels of MEF2A, C, and D proteins were high in liver tissues from patients with cirrhosis and increased during culture-induced activation of primary H-HSCs. Exposure of H-HSCs to transforming growth factor beta 1 (TGF-β1) led to a significant increase in MEF2A and C protein levels and enhanced MEF2 activity. Interestingly, TGF-β1 did not further enhance MEF2D levels. Furthermore, TGF-β1 activated p38 mitogen-activated protein kinase (MAPK) and led to increased phosphorylation of MEF2C at its p38 recognition site. Inhibition of p38 MAPK inhibited both TGF-β1- and culture-induced activation of MEF2. The activity of collagen I reporter in H-HSCs was significantly reduced when MEF2A and MEF2C were blocked with overexpression of dominant negative MEF2 mutants. Salvianolic-acid B (SA-B), a water-soluble element of Salvia miltiorrhiza known to have anti-fibrosis effects, attenuated both basal and TGF-β1-induced increased levels of MEF2A and C mRNA and protein. In addition, SA-B inhibited MEF2 activity, which correlated with reduced expression of the HSC activation markers, α-smooth muscle actin (α-SMA), and collagen I. Administration of SA-B reduced MEF2A in vivo, which was accompanied by reduced levels of α-SMA in a model of dimethylnitrosamine-induced rat liver fibrosis. We concluded that the MEF2 transcription factor was stimulated by TGF-β1 in H-HSCs. Antagonizing TGF-β1-induced activation of the MEF2 signaling pathway may account in part for the anti-fibrosis effects of SA-B.

The dynamic recrystallization (DRX) behavior in the hot working of TB8 titanium alloy was studied by using the experiment and finite element simulation (FEM) method. The results showed that the DRX behavior of TB8 titanium alloys was drastically affected by the hot processing parameters. The rising deformation temperature and reducing strain rate led to an augmentation in the grain size (dDRX) and volume fraction (XDRX) of DRX grains. In view of the true stress–strain curves gained from the experiment, the dDRX and XDRX models of DRX grains were constructed. Based on the developed models for DRX of TB8 titanium alloy, the isothermal forging process of the cylindrical samples was simulated by the DEFORM-3D software. The distributions of the effective strain and XDRX for DRX were analyzed. A comparison of the dDRX and XDRX of DRX grains in the central regions of the samples between the experimental and FEM results was performed. A good correlation between the experimental and simulation results was obtained, indicating that the established FEM model presented good prediction capabilities.

Metabolism is a highly integrated process that is coordinately regulated between tissues and within individual cells. FoxO proteins are major targets of insulin action and contribute to the regulation of gluconeogenesis, glycolysis, and lipogenesis in the liver. However, the mechanisms by which FoxO proteins exert these diverse effects in an integrated fashion remain poorly understood. We report that FoxO proteins also exert important effects on intrahepatic lipolysis and fatty acid oxidation via the regulation of adipose triacylglycerol lipase (ATGL), which mediates the first step in lipolysis, and its inhibitor, the G0/S1 switch 2 gene (G0S2). We also find that ATGL-dependent lipolysis plays a critical role in mediating diverse effects of FoxO proteins in the liver, including effects on gluconeogenic, glycolytic, and lipogenic gene expression and metabolism. These results indicate that intrahepatic lipolysis plays a critical role in mediating and integrating the regulation of glucose and lipid metabolism downstream of FoxO proteins.

Technological innovation and increased affordability have contributed to the widespread adoption of genome sequencing technologies in biomedical research. In particular large cancer research consortia have embraced next generation sequencing, and have used the technology to define the somatic mutation landscape of multiple cancer types. These studies have primarily utilised the Illumina HiSeq platforms. In this study we performed whole genome sequencing of three malignant pleural mesothelioma and matched normal samples using a new platform, the BGISEQ-500, and compared the results obtained with Illumina HiSeq X Ten. Germline and somatic, single nucleotide variants and small insertions or deletions were independently identified from data aligned human genome reference. The BGISEQ-500 and HiSeq X Ten platforms showed high concordance for germline calls with genotypes from SNP arrays (>99%). The germline and somatic single nucleotide variants identified in both sequencing platforms were highly concordant (86% and 72% respectively). These results indicate the potential applicability of the BGISEQ-500 platform for the identification of somatic and germline single nucleotide variants by whole genome sequencing. The BGISEQ-500 datasets described here represent the first publicly-available cancer genome sequencing performed using this platform.

This paper analyzes the impact of capacity drop on commuters’ travel choice behaviors under uncertainty. For clarity, we assume that the capacity drop is triggered by the queue forming at the bottleneck under the hypercongestion circumstances, and the stochasticity of the drop could not be neglected. Considering the uncertainty of travel time, we establish a bottleneck model with commuters choosing their departure time according to the mean travel cost. From the proposed model, analytical solutions are achieved and therefore several properties are presented, including monotonicity of travel cost and departure rate, and the relationship between dispersion degree and length of peak period. To alleviate traffic congestion at the bottleneck and avoid capacity drop, we design a time-varying toll scheme and a step toll scheme. Evolution of queue length in equilibrium is discussed based on the Laih model. Numerical examples are also presented to demonstrate the established model and the effectiveness of the proposed toll schemes.
