Abstract Background Premature ovarian insufficiency (POI) is an important cause of female infertility and seriously impacts the physical and psychological health of patients. Human umbilical cord mesenchymal stem cell-derived exosomes (HucMSCs-Exs, H-Exs) have exhibited protective effects on ovarian function with unclear mechanisms. Methods A comprehensive analysis of the Gene Expression Omnibus (GEO) database were used to identify POI-associated circRNAs and miRNAs. The relationship between HucMSC-derived exosomal circBRCA1/miR-642a-5p/FOXO1 axis and POI was examined by RT-qPCR, Western blotting, reactive oxygen species (ROS) staining, senescence-associated β-gal (SA-β-gal) staining, JC-1 staining, TEM, oxygen consumption rate (OCR) measurements and ATP assay in vivo and in vitro. RT-qPCR detected the expression of circBRCA1 in GCs and serum of patients with normal ovarian reserve function (n = 50) and patients with POI (n = 50); then, the correlation of circBRCA1 with ovarian reserve function indexes was analyzed. Results Herein, we found that circBRCA1 was decreased in the serum and ovarian granulosa cells (GCs) of patients with POI and was associated with decreased ovarian reserve. H-Exs improved the disorder of the estrous cycles and reproductive hormone levels, reduced the number of atretic follicles, and alleviated the apoptosis and senescence of GCs in rats with POI. Moreover, H-Exs mitigated mitochondrial damage and reversed the reduced circBRCA1 expression induced by oxidative stress in GCs. Mechanistically, FTO served as an eraser to increase the stability and expression of circBRCA1 by mediating the m6A demethylation of circBRCA1, and exosomal circBRCA1 sponged miR-642a-5p to block its interaction with FOXO1. CircBRCA1 insufficiency aggravated mitochondrial dysfunction, mimicking FTO or FOXO1 depletion effects, which was counteracted by miR-642a-5p inhibition. Conclusion H-Exs secreted circBRCA1 regulated by m6A modification, directly sponged miR-642a-5p to upregulate FOXO1, resisted oxidative stress injuries in GCs and protected ovarian function in rats with POI. Exosomal circBRCA1 supplementation may be a general prospect for the prevention and treatment of POI. Graphical Abstract

Abstract Background Intrauterine adhesion (IUA) as a prevalent gynecological disease is developed from infection or trauma. However, therapeutic strategies to repair damaged endometrium are relatively limited. Emerging studies have shed light on the crucial role of endometrial stromal cells (EnSCs) in the process of uterine endometrial regeneration. EnSCs isolated from the uterine endometrium have similar characteristics to mesenchymal stem cells (MSCs). However, it is still unknown whether EnSCs could be used as donor cells to treat IUA. The aim of this study was to evaluate the potential efficacy of EnSCs in treating rat IUA. Methods Human EnSCs were isolated from the endometrial tissue of healthy female donors and subjected to extensive expansion and culture in vitro. Immunofluorescence, flow cytometry, cell proliferation assay, trilineage differentiation experiment, and decidualization assay were used to characterize the biological properties of EnSCs. We evaluated the immunoregulatory potential of EnSCs by analyzing their secreted cytokines and conducting bulk RNA sequencing after IFN-γ treatment. After EnSCs were transplanted into the uterine muscle layer in IUA rats, their therapeutic effects and underlying mechanisms were analyzed using histological analysis, Q-PCR, fertility and pregnancy outcome assay, and transcriptome analysis. Results We successfully isolated EnSCs from the endometrium of human donors and largely expanded in vitro. EnSCs exhibited characteristics of mesenchymal stem cells and retained responsiveness to sex hormones. Following IFN-γ stimulation, EnSCs upregulated the anti-inflammatory cytokines and activated immunosuppressive molecules. Xenogeneic transplantation of EnSCs successfully repaired injured endometrium and significantly restored the pregnancy rate in IUA rats. Mechanistically, the therapeutic effects of EnSCs on IUA endometrium functioned through anti-inflammation, anti-fibrosis and the secretion of regeneration factor. Conclusions Due to their large expansion ability, immunoregulatory properties, and great potential in treating IUA, EnSCs, as a valuable source of donor cells, could offer a potential treatment avenue for injury-induced IUA.

Abstract Objectives The rising of antibiotic resistance has sparked a renewed interest in mycobacteriophage as alternative therapeutic strategies against mycobacterial infections. So far, the vast majority of mycobacteriophages have been isolated using the model species Mycobacterium smegmatis, implying an overwhelming majority of mycobacteriophages in the environment remain uncultured, unclassified, and their specific hosts and infection strategies are still unknown. This study was undertaken to isolate and characterize novel mycobacteriophages targeting Mycobacterium septicum. Data description Here a novel mycobacteriophage WXIN against M. septicum was isolated from soil samples in Wuhan, China. Whole genome analysis indicates that the phage genome consists of 115,158 bp with a GC content of 61.9%. Of the 260 putative open reading frames, 46 may be associated with phage packaging, structure, lysis, lysogeny, genome modification/replication, and other functional roles. The limited genome-wide similarity, along with phylogenetic trees constructed based on viral proteome and orthologous genes show that phage WXIN represents a novel cluster distantly related to cluster J mycobacteriophages (genus Omegavirus). Overall, these results provide novel insights into the genomic properties of mycobacteriophages, highlighting the great genetic diversity of mycobacteriophages in relation to their hosts.

In the current study, the corrosion behavior of HSLA-65 steel was investigated in a simulated marine environment inoculated with P. aeruginosa. In the electrochemical measurements, the negative shift in OCP, lower values of corrosion potential, pitting potential, and increased corrosion current density of the HSLA-65 steel in the biotic medium signified its low corrosion resistance in the presence of P. aeruginosa. The observation of the surface morphology revealed biofilm formation on the steel surface in the P. aeruginosa-inoculated culture medium, which significantly accelerated the pitting corrosion of the steel. The pit observed on the steel surface in the biotic medium was about 3.9 times deeper than in the sterile medium. In addition, the weight loss of the steel in the biotic medium was about 2.4 times higher compared to that in the sterile medium. These findings suggest that the P. aeruginosa biofilm significantly accelerated the corrosion of HSLA-65 steel in the simulated marine environment.

Abstract Background Saline lakes are home to various archaea that play special and crucial roles in the global biogeochemical cycle. The Qinghai-Tibet Plateau hosts a large number of lakes with diverse salinity ranging from 0.1 to over 400 g/L, harboring complex and diverse archaea. To the best of our knowledge, the formation mechanisms and potential ecological roles of archaea in Qinghai-Tibetan Plateau saline lakes remain largely unknown. Results Using High-throughput Illumina sequencing, we uncovered the vastly distinct archaea communities between two typical saline lakes with significant salinity differences on the Qinghai Tibet Plateau (Qinghai saline lake and Chaka hypersaline lake) and suggested archaea played different important roles in methanogenesis-related and nitrate reduction-related functions of these two lakes, respectively. Rather than the individual effect of salinity, the composite effect of salinity with diverse environmental parameters (e.g., temperature, chlorophyll a, total nitrogen, and total phosphorus) dominated the explanation of the variations in archaeal community structure in different habitats. Based on the network analysis, we further found the correlations between dominant archaeal OTUs were tight but significantly different between the two habitats, implying that archaeal interactions may also largely determine the shape of archaeal communities. Conclusion The present study improved our understanding of the structure and function of archaea in different saline lakes on the Qinghai-Tibet Plateau and provided a new perspective on the mechanisms underlying shaping their communities.

Abstract The evaluation of grouting effects constitutes a critical aspect of grouting engineering. With the maturity of the grouting project, the workload and empirical characteristics of grouting effect evaluation are gradually revealed. In the context of the Qiuji coal mine’s directional drilling and grouting to limestone aquifer reformation, this study thoroughly analyzes the influencing factors of grouting effects from geological and engineering perspectives, comparing these with various engineering indices associated with drilling and grouting. This led to the establishment of a “dual-process, multi-parameter, and multi-factor” system, employing correlation analysis to validate the selected indices’ reasonableness and scientific merit. Utilizing the chosen indices, eight high-performing machine learning models and three parameter optimization algorithms were employed to develop a model for assessing the effectiveness of directional grouting in limestone aquifers. The model’s efficacy was evaluated based on accuracy, recall, precision, and F-score metrics, followed by practical engineering validation. Results indicate that the “dual-process, multi-parameter, multi-factor” system elucidates the relationship between influencing factors and engineering parameters, demonstrating the intricacy of evaluating grouting effects. Analysis revealed that the correlation among the eight selected indicators—including the proportion of boreholes in the target rock strata, drilling length, leakage, water level, pressure of grouting, mass of slurry injected, permeability properties of limestone aquifers before being grouted, permeability properties of limestone aquifers after being grouted—is not substantial, underscoring their viability as independent indicators for grouting effect evaluation. Comparative analysis showed that the Adaboost machine learning model, optimized via a genetic algorithm, demonstrated superior performance and more accurate evaluation results. Engineering validation confirmed that this model provides a more precise and realistic assessment of grouting effects compared to traditional methods.

It is difficult to fully develop coal resources in the north of the Yellow River due to the influence of mine water disaster. Grouting technology is an important means to solve the problem of mine water disasters at present. The in-depth study of grouting technology can not only liberate a large number of coal resources threatened by water disasters, but also have positive significance for the protection of groundwater resources. Laboratory tests were conducted on limestone samples obtained from Qiuji Coal Mine in the Yellow River North Coalfield to observe the microscopic characteristics of the limestone. The mechanical properties and grouting reinforcement characteristics of limestone with different lengths of cracks under fluid solid coupling were obtained. Through theoretical analysis, the variation law of the cracking strength of limestone with the change of prefabricated crack length was obtained, and the grouting reinforcement mechanism was analyzed. The test results show that the strength and toughness of the rock after grouting have been significantly improved, the limestone before grouting has obvious brittleness characteristics, and the brittleness of the limestone after grouting has decreased and the ductility has increased; The damage degree of grouting plus solid is more and more severe with the increase of crack length. When the crack length is large, it is accompanied by horizontal fracture development; The pre-peak permeability of grouting plus solid decreases slightly, and the post-peak permeability decreases significantly. The maximum stage of limestone permeability occurs in the post-peak stage, while the minimum stage of permeability occurs in the linear elastic stage of the stress-strain curve. The longer the fracture length, the easier the double-peak phenomenon of permeability appears. Based on the theory of continuous medium mechanics, the formulas for calculating the crack initiation strength and fracture toughness of grouting plus solid are derived. It is theoretically proved that the strength of grouting plus solid is greater than that of the original rock sample; The greater the difference between the elastic modulus of rock and the elastic modulus of the combination formed by grouting, the weaker the impact of grouting on rock strength.

Abstract Generally, when optimizing a rolling stock schedule, the locations of the depots, or places in the network where the composition changes and maintenance occurs, are assumed known. The locations where maintenance is performed naturally influence the quality of any resulting rolling stock schedules. In this paper, the problem of selecting new depot locations and their corresponding capacities is considered. A two-stage mixed integer programming approach for rolling stock scheduling with maintenance requirements is extended to account for depot selection. First, a conventional flow-based model is solved, ignoring maintenance requirements, to obtain a variety of rolling stock schedules with multiple depot locations and capacity options. Then, a maintenance feasible rolling stock schedule can be obtained by solving a series of assignment problems by using the schedules found in the first stage. The proposed methodology is tested on real-life instances, and the numerical experiments of different operational scenarios are discussed.

Background Symptom networks offer a theoretical basis for developing personalised and precise symptom management strategies. However, symptom networks in lymphoma patients during chemotherapy have been rarely reported. This study intends to establish contemporaneous symptom networks in lymphoma patients during chemotherapy and explore the centrality indices and density in these symptom networks.Methods and analysis This is a single-centre prospective cross-sectional study. A total of 315 lymphoma patients admitted to the Lymphoma Department of Shanxi Bethune Hospital since 1 June 2024 will be selected as the study subjects. The patient-reported outcome measures of General Data Questionnaire and Lymphoma Symptom Assessment Scale will be assessed. R package will be used to construct a contemporaneous symptom network, explore the relationship between core and analysed symptoms and analyse the predictive role of network density on patient prognosis.Ethics and dissemination This study adheres to the principles of the Declaration of Helsinki and relevant ethical guidelines. Ethical approval has been obtained from Shanxi Bethune Hospital Ethics Committee (approval number: YXLL-2023-186). The final outcomes will be published in a peer-reviewed journal and disseminated through a conference.

Summary: Background: The 5-year survival rate of oesophageal squamous cell carcinoma (ESCC) is approximately 20%. The prognosis and drug response exhibit substantial heterogeneity in ESCC, impeding progress in survival outcomes. Our goal is to identify a signature for tumour subtype classification, enabling precise clinical treatments. Methods: Utilising pre-treatment multi-omics data from an ESCC dataset (n = 310), an enhancer methylation-eRNA-target gene regulation network was constructed and validated by in vitro experiments. Four machine learning methods collectively identified core target genes, establishing an Enhancer Demethylation-Regulated Gene Score (EDRGS) model for classification. The molecular function of EDRGS subtyping was explored in scRNA-seq (n = 60) and bulk-seq (n = 310), and the EDRGS's potential to predict treatment response was assessed in datasets of various cancer types. Findings: EDRGS stratified ESCCs into EDRGS-high/low subtypes, with EDRGS-high signifying a less favourable prognosis in ESCC and nine additional cancer types. EDRGS-high exhibited an immune-hot but immune-suppressive phenotype with elevated immune checkpoint expression, increased T cell infiltration, and IFNγ signalling in ESCC, suggesting a better response to immunotherapy. Notably, EDRGS outperformed PD-L1 in predicting anti-PD-1/L1 therapy effectiveness in ESCC (n = 42), kidney renal clear cell carcinoma (KIRC, n = 181), and bladder urothelial carcinoma (BLCA, n = 348) cohorts. EDRGS-low showed a cell cycle-activated phenotype with higher CDK4 and/or CDK6 expression, demonstrating a superior response to the CDK4/6 inhibitor palbociclib, validated in ESCC (n = 26), melanoma (n = 18), prostate cancer (n = 15) cells, and PDX models derived from patients with pancreatic cancer (n = 30). Interpretation: Identification of EDRGS subtypes enlightens ESCC categorisation, offering clinical insights for patient management in immunotherapy (anti-PD-1/L1) and CDK4/6 inhibitor therapy across cancer types. Tumor necrosis factor receptor-associated factor 6 (TRAF6) serves as a critical factor in the initiation of the NF-κB pathway and holds significance in modulating the inflammatory response. However, the regulatory mechanism of TRAF6 in teleost fish is poorly understood. We demonstrate Miichthys miiuy nucleoporin 43 (mm-nup43), a member of the nuclear pore complex (NPC), is a negative regulator of the NF-κB pathway and inhibits the expression of inflammatory cytokines. Mechanistically, mm-nup43 interacts with TRAF6 and inhibits it via the ubiquitin-proteasome pathway. Meanwhile, mm-nup43 decreased p65 phosphorylation, thereby negatively regulating NF-κB signaling. In summary, these results classify that mm-nup43 inhibits the TRAF6-mediated NF-κB pathway and prevents excessive inflammation.

Changes in physio-biochemical metabolism, phenolics and antioxidant capacity during germination were studied in eight different wheat varieties. Results showed that germination enhanced sprout growth, and caused oxidative damage, but enhanced phenolics accumulation. Ferulic acid and p-coumaric acid were the main phenolic acids in wheat sprouts, and dihydroquercetin, quercetin and vitexin were the main flavonoids. The phenolic acid content of Jimai 44 was the highest on the 2th and 4th day of germination, and that of Bainong 307 was the highest on the 6th day. The flavonoid content of Hei jingang was the highest during whole germination. The enzymes activities of phenylalanine ammonia lyase (PAL), cinnamic acid 4-hydroxylase (C4H) and 4-coumarate coenzyme A ligase (4CL) were up-regulated. The activities of catalase, polyphenol oxidase and peroxidase were also activated. Antioxidant capacity of wheat sprouts was enhanced. The results provided new ideas for the production of naturally sourced phenolic rich foods.

Abstract The objective of this study was retrospectively to analyze the clinical characteristics and 18F-FDG PET/CT findings in Meigs syndrome (MS) patients. A total of 21 patients with MS induced by ovarian stromal tumors and 69 patients with pseudo-MS caused by ovarian cancer (OC-PMS) were subjected to evaluation using 18F-FDG PET/CT. Visual and semi-quantitative methods were employed to analyze the PET/CT findings. Visual analysis included recording whether the density of the primary tumor was uniform, whether there were cystic changes and calcifications, and the location of serous fluid accumulation. Semi-quantitative analysis involved the measurement of the tumor size, SUVmax, and SUVmean. No significant difference was observed in the size and density of primary tumors between the MS group and the OC-PMS group. However, the SUVmax and SUVmean of tumors in the MS group were found to be significantly lower than those in the OC-PMS group. The amount of serous cavity effusion caused by ovarian sex cord stromal tumors was found to be unrelated to the size of the tumor, SUVmax, and SUVmean but was positively correlated with the level of Ca125. MS patients have both benign ovarian tumors and ascites and/or pleural effusion, which may be accompanied by elevated Ca125 levels. This should be considered as one of the differential diagnoses for ovarian cancer. Understanding the PET/CT features of MS can facilitate the attainment of an accurate diagnosis before surgery.

Abstract Mutant KRAS (KRASMUT) is often exploited by cancers to shape tumor immunity, but the underlying mechanisms are not fully understood. Here we report that tumor-specific cytotoxic T lymphocytes (CTLs) from KRASMUT cancers are sensitive to activation-induced cell death (AICD). circATXN7, an NF-κB-interacting circular RNA, governs T cell sensitivity to AICD by inactivating NF-κB. Mechanistically, histone lactylation derived from KRASMUT tumor cell-produced lactic acid directly activates transcription of circATXN7, which binds to NF-κB p65 subunit and masks the p65 nuclear localization signal motif, thereby sequestering it in the cytoplasm. Clinically, circATXN7 upregulation in tumor-specific CTLs correlates with adverse clinical outcomes and immunotherapeutic resistance. Genetic ablation of circAtxn7 in CD8+ T cells leads to mutant-selective tumor inhibition, while also increases anti-PD1 efficacy in multiple tumor models in female mice. Furthermore, targeting circATXN7 in adoptively transferred tumor-reactive CTLs improves their antitumor activities. These findings provide insight into how lymphocyte-expressed circRNAs contribute to T-cell fate decisions and anticancer immunotherapies.

The evaluation of grouting effect is one of the crucial links in grouting water control project. As grouting technology matures, traditional methods of evaluating grouting effects are more subjective and fewer objective, which leads to the drawback that the evaluation results deviate from reality. Taking the grouting project of 8901 working face in Baizhuang Coal Mine of Shandong Province as the background, combining existing research and field experience, the grouting final pressure, permeability coefficient, water absorption rate and the number of dry material per ton of water are selected as the evaluation indexes, and the comprehensive evaluation system of grouting effect is established. Based on the comprehensive evaluation theory, a comprehensive subjective and objective weighting method is proposed, which based on analytic hierarchy process(AHP) combined with CRITIC objective weighting method. And the objectivity ofpf indicator assignment is improved by combining subjective and objective weights. Based on the combination weighting method, a comprehensive evaluation model of grouting effect is established, which is sorted by technique for order preference by similarity to an ideal solution (TOPSIS) and assisted by rank sum ratio (RSR). The evaluation results are converted into visual partition maps and compared with the actual evaluation results of field engineering and the results of the testing process after grouting, and the results show that the model has superior accuracy and rationality.

This paper introduces a four-band tunable perfect absorber built on Al-Cu-Fe quasicrystal. The operating frequency range is 4.75 THz to 9.25 THz, which is in the terahertz band. The absorber exhibits multiple narrow bands, polarization incoherence, high sensitivity, and tunability. The structure comprises a gold layer, a silica dielectric layer, and a top layer of Al-Cu-Fe quasicrystal from bottom to top. The Al-Cu-Fe quasicrystal structure at the top consists of a circular ring left after being cut by a 45° rotation of the cruciform and a cruciform design containing a central circular cavity. Simulation results reveal four absorption peaks attributed to cavity resonance and surface plasmon resonance, with absorption efficiencies of 0.998, 0.994, 0.989, and 0.998, respectively. Furthermore, changing the radius of the inner ring in the top layer structure allows for varying the number of absorption peaks. We also propose a new fabrication process for Al-Cu-Fe quasicrystals absorbers. The calculated results demonstrate that the designed absorber, characterized by a simple structure and high sensitivity of 2620 GHz/RIU, holds significant potential for applications in optical switches, optical modulators, biomedicine, and sensing.

Gastrointestinal stromal tumors (GISTs) are the most common mesenchymal tumors affecting the gastrointestinal tract. Typically, GISTs are solitary; however, in rare cases, they may be multiple and appear in one or more organs. Multiple GISTs can appear in familial GISTs, children, or certain tumor syndromes such as neurofibromatosis type 1, Carney syndrome, and Carney-Stratakis syndrome. However, the diagnosis of primary multiple sporadic GISTs is often more difficult than that of these diseases. Herein, we report a case of multiple primary sporadic GISTs in a 64-year-old man, affecting the abdominal cavity and retroperitoneum, as identified through dual-time point positron emission tomography (PET) with 18F-labeled fluoro-2-deoxyglucose (18F-FDG) and computed tomography (18F-FDG PET/CT). Notably, the dual-time-point PET/CT revealed the migration of masses near the lower abdomen into the abdominal cavity. Furthermore, a significant increase in radioactive uptake of the mass 3 h after 18F-FDG injection compared with that 1 h after injection may be an important cue for its diagnosis.

Abstract Parkinson’s disease (PD) is the second most common neurodegenerative disease after Alzheimer’s disease (AD). It is characterized by a progressive loss of dopaminergic neurons in the substantia nigra pars compacta (SNc) and the formation of Lewy bodies (LBs). Although PD is primarily considered a gray matter (GM) disease, alterations in white matter (WM) have gained increasing attention in PD research recently. Here we review evidence collected by magnetic resonance imaging (MRI) techniques which indicate WM abnormalities in PD, and discuss the correlations between WM changes and specific PD symptoms. Then we summarize transcriptome and genome studies showing the changes of oligodendrocyte (OLs)/myelin in PD. We conclude that WM abnormalities caused by the changes of myelin/OLs might be important for PD pathology, which could be potential targets for PD treatment.

Abstract Identifying changes in coal permeability with gas pressure and accurately codifying mean effective stresses in laboratory samples are crucial in predicting gas-flow behavior in coal reservoirs. Traditionally, coal permeability to gas is assessed using the steady-state method, where the equivalent gas pressure in the coal is indexed to the average of upstream and downstream pressures of the coal, while ignoring the nonlinear gas pressure gradient along the gas flow path. For the flow of a compressible gas, the traditional method consistently underestimates the length/volume-averaged pressure and overestimates mean effective stress. The higher the pressure differential within the sample, the greater the error between the true mean pressure for a compressible fluid and that assumed as the average between upstream and downstream pressures under typical reservoir conditions. A correction coefficient for the compressible fluid pressure asymptotes to approximately 1.3%, representing that the error in mean pressure and effective stress can be on the order of approximately 30%, particularly for highly pressure-sensitive permeabilities and compressibilities, further amplifying errors in evaluated reservoir properties. We utilized this volume-averaged pressure and effective stress to correct permeability and compressibility data reported in the literature. Both the corrected initial permeability and the corrected pore compressibility were found to be smaller than the uncorrected values, due to the underestimation of the true mean fluid pressure, resulting in an overestimation of reservoir permeability if not corrected. The correction coefficient for the initial permeability ranges from 0.6 to 0.1 (reservoir values are only approximately 40% to 90% of laboratory values), while the correction coefficient for pore compressibility remains at approximately 0.75 (reservoir values are only approximately 25% of laboratory value). Errors between the uncorrected and corrected parameters are quantified under various factors, such as confining pressure, gas sorption, and temperature. By analyzing the evolutions of the initial permeability and pore compressibility, the coupling mechanisms of mechanical compression, adsorption swelling, and thermal expansion on the pore structure of the coal can be interpreted. These findings can provide insights that are useful for assessing the sensitivity of coal permeability to gas pressure as truly representative of reservoir conditions.

Summary: Circular RNAs (circRNAs) are crucial regulators of β-cell function and are involved in lipotoxicity-induced β-cell damage in type 2 diabetes mellitus (T2DM). We previously identified that circGlis3, a circRNA derived from exon 4 of the diabetes susceptibility gene Glis3, was upregulated in lipotoxic β cells. However, the functional role and molecular mechanism of circGlis3 in β cells remain largely unknown. Here, we revealed that the splicing factor CUGBP Elav-Like Family Member 1 (CELF1) facilitated the biogenesis of circGlis3. Moreover, we established a transgenic mouse model and confirmed that the overexpression of circGlis3 impaired β-cell function. Mechanistically, circGlis3 bound to heterogeneous nuclear ribonucleoprotein F (hnRNPF) and blocked its nuclear translocation, thereby reducing Sirt1 levels. Additionally, circGlis3 encoded a 348aa protein that interacted with GLIS3 and inhibited its transcriptional activity. Our data uncover a critical role of circGlis3 in β-cell dysfunction, suggesting that circGlis3 may be a potential therapeutic target for T2DM.

Osteoarthritis (OA) has been a leading cause of disability in the elderly and there remains a lack of effective therapeutic approaches as the mechanisms of pathogenesis and progression have yet to be elucidated. As OA progresses, cellular metabolic profiles and energy production are altered, and emerging metabolic reprogramming highlights the importance of specific metabolic pathways in disease progression. As a crucial part of glucose metabolism, glycolysis bridges metabolic and inflammatory dysfunctions. Moreover, the glycolytic pathway is involved in different areas of metabolism and inflammation, and is associated with a variety of transcription factors. To date, it has not been fully elucidated whether the changes in the glycolytic pathway and its associated key enzymes are associated with the onset or progression of OA. This review summarizes the important role of glycolysis in mediating cellular metabolic reprogramming in OA and its role in inducing tissue inflammation and injury, with the aim of providing further insights into its pathological functions and proposing new targets for the treatment of OA.

This paper focuses on optimizing the management of delayed trains in operational scenarios by scientifically categorizing train delay levels. It employs static and dynamic models grounded in real-world train delay data from high-speed railways. This classification aids dispatchers in swiftly identifying and predicting delay extents, thus enhancing mitigation strategies' efficiency. Key indicators, encompassing initial delay duration, station impacts, average station delay, delayed trains' cascading effects, and average delay per affected train, inform the classification. Applying the K-means clustering algorithm to standardized delay indicators yields an optimized categorization of delayed trains into four levels, reflecting varying risk levels. This static classification offers a comprehensive overview of delay dynamics. Furthermore, utilizing Markov chains, the study delves into sequential dynamic analyses, accounting for China's railway context and specifically addressing fluctuations during the Spring Festival travel rush. This research, combining static and dynamic approaches, provides valuable insights for bolstering railway operational efficiency and resilience amidst diverse delay scenarios.

Abstract Objectives Emerging evidence has demonstrated that tumour budding (TB) is negatively associated with T‐lymphocyte infiltration in CRC. Despite extensive research, the molecular characteristics of immunologically ‘hot’ TB remain poorly understood. Methods We quantified the number of TB by haematoxylin–eosin (H&E) sections and the densities of CD3+ and CD8+ T‐lymphocytes by immunohistochemistry in a CRC cohort of 351 cases who underwent curative resection. We analysed the differential expression and T‐lymphocyte infiltration score of 37 human epithelial keratins in CRC using RNA sequencing from the TCGA dataset. In 278 TB‐positive cases, KRT17 expression was evaluated in tumour centre (TC) and TB with a staining score. Patient demographic, clinicopathological features and survival rates were analysed. Results In a CRC cohort of 351 cases, low‐grade TB was associated with high CD3+ and CD8+ T‐cell densities in the invasive margin (IM) but not in the TC. Of 37 human epithelial keratins, only KRT17 expression in TB had an apparent association with TB‐grade and T‐lymphocyte infiltration. In 278 TB‐positive cases, high KRT17 expression in TB (KRT17TB) was negatively associated with low‐grade TB and positively associated with high CD3+ and CD8+ T‐cell densities in IM. High KRT17TB predicted early tumour grade, absence of lymph node metastasis and absence of tumour deposits. Additionally, patients with high KRT17TB had good overall survival and disease‐free survival. Notably, low KRT17TB can specifically identify those patients with a poor prognosis among colorectal cancer patients with low TB and high T‐lymphocyte infiltration. Conclusions KRT17 can be employed as a new indicator for distinguishing different immunological TBs.

Directional regional grouting to transform aquifers is one of the effective ways to prevent and control mine water hazards. In order to study the diffusion law of grouting slurry in directional areas, a three-dimensional simulation test system for directional area grouting has been independently developed. The system is mainly composed of test bench, grouting system, water injection system, data monitoring and other subsystems, which can realize directional drilling grouting simulation, three-dimensional slurry diffusion simulation, high pressure water simulation and regional grouting simulation. The system can independently control the grouting hole unit to achieve porous and multi sequence grouting. With the help of six grouting holes, the slurry flow process is simulated under various parameters such as different grouting pressure, slurry viscosity, grouting rate, and water cement ratio. It can also achieve real-time monitoring of the slurry diffusion process in dynamic/static (high) water pressure environments. The paper takes the simulated naked hole lengths of 200 mm, 250 mm, and 300 mm as the research objects, and preliminarily explores the characteristics and effects of slurry diffusion in naked hole grouting under directional grouting. The study found that: in the case where only the naked hole length is a variable, the slurry diffusion range does not synchronously increase linearly with the linear increase of naked hole length; The slurry stone body of the short open hole is cylindrical with the drilling open hole as the axis, while the slurry stone body of the long open hole is circular with the drilling open hole as the axis; Increasing the same length of the bare hole results in a larger difference in the diffusion range of the slurry at both ends of the bare hole. At the same time, the more pressure loss there is, the smaller the diffusion distance of the slurry. Comparing the diffusion radius of the slurry obtained from the experiment with that calculated by the Maag formula, the difference between the two values is only 2.8% when the length of the grouting open hole is 200 mm. The experimental results comply with the Maag spherical diffusion theory and verified the scientificity and rationality of this experimental system.

Digestive system cancer and COVID-19 significantly affect the digestive system, but the mechanism of interaction between COVID-19 and the digestive system cancers has not been fully elucidated. We downloaded the gene expression of COVID-19 and seven digestive system cancers (oral, esophageal, gastric, colorectal, hepatocellular, bile duct, pancreatic) from GEO and identified hub differentially expressed genes. Multiple verifications, diagnostic efficacy, prognostic analysis, functional enrichment and related transcription factors of hub genes were explored. We identified 23 common DEGs for subsequent analysis. CytoHubba identified nine hub genes (CCNA2, CCNB1, CDKN3, ECT2, KIF14, KIF20A, KIF4A, NEK2, TTK). TCGA and GEO data validated the expression and excellent diagnostic and prognostic ability of hub genes. Functional analysis revealed that the processes of cell division and the cell cycle were essential in COVID-19 and digestive system cancers. Furthermore, six related transcription factors (E2F1, E2F3, E2F4, MYC, TP53, YBX1) were involved in hub gene regulation. Via in vitro experiments, CCNA2, CCNB1, and MYC expression was verified in 25 colorectal cancer tissue pairs. Our study revealed the key biomarks and common pathogenesis of digestive system cancers and COVID-19. These may provide new ideas for further mechanistic research.

This study aims to develop equipment for the preparation of composite films and successfully implement a film thickness prediction function. During the research process, we segmented the mechanical structure of the composite thin film preparation equipment into distinct modules, completed the structural design of the core module, and validated the stability of the process chamber, as well as the reasonableness of the strength and stiffness through simulation. Additionally, we devised a regression model for predicting the film thickness of composite films. The input features for the model included the sputtering air pressure, sputtering current, and sputtering time for magnetron sputtering process samples, as well as the evaporation volume and evaporation current for vacuum evaporation process samples. Simultaneously, the output features were the film thickness for both process samples. Subsequently, we established the designed composite film preparation equipment and conducted experimental verification. During the experiments, we successfully prepared Cr-Al composite films and utilized AFM for surface morphology analysis. The results confirmed the excellent performance of the Cr-Al composite films produced by the equipment, demonstrating the reliability of the equipment.

Abstract Background Colon cancer (CC) is a common tumor that causes significant harm to human health. Bacteria play a vital role in cancer biology, particularly the biology of CC. Genes related to bacterial response were seldom used to construct prognosis models. We constructed a bacterial response-related risk model based on three Molecular Signatures Database gene sets to explore new markers for predicting CC prognosis. Methods The Cancer Genome Atlas (TCGA) colon adenocarcinoma samples were used as the training set, and Gene Expression Omnibus (GEO) databases were used as the test set. Differentially expressed bacterial response-related genes were identified for prognostic gene selection. Univariate Cox regression analysis, least absolute shrinkage and selection operator-penalized Cox regression analysis, and multivariate Cox regression analysis were performed to construct a prognostic risk model. The individual diagnostic effects of genes in the prognostic model were also evaluated. Moreover, differentially expressed long noncoding RNAs (lncRNAs) were identified. Finally, the expression of these genes was validated using quantitative polymerase chain reaction (qPCR) in cell lines and tissues. Results A prognostic signature was constructed based on seven bacterial response genes: LGALS4, RORC, DDIT3, NSUN5, RBCK1, RGL2, and SERPINE1. Patients were assigned a risk score based on the prognostic model, and patients in the TCGA cohort with a high risk score had a poorer prognosis than those with a low risk score; a similar finding was observed in the GEO cohort. These seven prognostic model genes were also independent diagnostic factors. Finally, qPCR validated the differential expression of the seven model genes and two coexpressed lncRNAs (C6orf223 and SLC12A9-AS1) in 27 pairs of CC and normal tissues. Differential expression of LGALS4 and NSUN5 was also verified in cell lines (FHC, COLO320DM, SW480). Conclusions We created a seven-gene bacterial response‐related gene signature that can accurately predict the outcomes of patients with CC. This model can provide valuable insights for personalized treatment.

Feed efficiency (FE), an important economic trait in sheep production, is indirectly assessed by residual feed intake (RFI). However, RFI in sheep is varied, and the molecular processes that regulate RFI are unclear. It is thus vital to investigate the molecular mechanism of RFI to developing a feed-efficient sheep. The miRNA-sequencing (RNA-Seq) was utilized to investigate miRNAs in liver tissue of 6 out of 137 sheep with extreme RFI phenotypic values. In these animals, as a typical metric of FE, RFI was used to distinguish differentially expressed miRNAs (DE_miRNAs) between animals with high (n = 3) and low (n = 3) phenotypic values. A total of 247 miRNAs were discovered in sheep, with four differentially expressed miRNAs (DE_miRNAs) detected. Among these DE_miRNAs, three were found to be upregulated and one was downregulated in animals with low residual feed intake (Low_RFI) compared to those with high residual feed intake (High_RFI). The target genes of DE_miRNAs were primarily associated with metabolic processes and biosynthetic process regulation. Furthermore, they were also considerably enriched in the FE related to glycolysis, protein synthesis and degradation, and amino acid biosynthesis pathways. Six genes were identified by co-expression analysis of DE_miRNAs target with DE_mRNAs. These results provide a theoretical basis for us to understand the sheep liver miRNAs in RFI molecular regulation.

Abstract Active thermal radiation control of an object requires adaptive and flexible strategies to cope with emissivity engineering and its temperature dependence. On the other hand, the concept of digital coding allows a high degree of freedom in manipulating the interacting physical quantities based on local discretized features. Learning from this idea, this study put forward the manipulation of spatial infrared emission based on tungsten (W)‐doped VO2 films toward thermal coding. Here, 1‐bit coding worksheets created by low‐emissivity, that is, code ‘0′, and high‐emissivity, that is, code ‘1′, shift with materials metal‐insulator phase transition to encode emissive information in the temperature domain. It is shown experimentally that this concept has marked implications on thermal radiation control at Mid‐infrared, among which are as follows: digital anti‐counterfeiting by in‐plane spatial coding of a group of VO2 thin films with various W‐doping density and infrared camouflage by out‐of‐plane spatial coding of them. With a W‐doping gradient of 0%, 0.8%, and 1.7%, the three‐phase transition points in the temperature domain, that is, 72, 63, and 45 °C, give a remarkable modulation range of 27 °C. These phenomena pave the way for new classes of emissivity engineering for more accurate, flexible, and adaptive thermal radiation control.

Myeloid sarcoma (MS) is a rare extramedullary parenchymal tumor composed of immature myeloid cells, occurring mainly in the lymph nodes, skin, soft tissue, testicles, bones, peritoneum, and gastrointestinal tract, and rarely in the pancreas. Herein, we report the case of a 68-year-old female patient who visited our hospital for medical assistance due to acute abdominal pain. Abdominal computed tomography (CT) and magnetic resonance imaging showed a mass approximately 8 cm in diameter in the pancreatic tail, which was suspected to be a malignant tumor. To further assess the presence of distant metastases, the patient underwent fluorine-18-fluorodeoxyglucose positron emission tomography (18F-FDG PET)/CT, which revealed an increased 18F-FDG uptake in the corresponding lesions. Subsequently, the patient underwent surgical treatment, and postoperative pathology and immunohistochemistry revealed that the mass was MS. Moreover, we reviewed the clinical features, imaging findings, and histopathology of pathologically confirmed pancreatic MS in the published literature.

Code Reuse Attacks (CRA) represent a type of control flow hijacking that attackers exploit to manipulate the standard program execution path, resulting in abnormal processor behaviors. In response to the security concern, proposals for Control Flow Integrity (CFI) verification have emerged. The CFI scheme diligently monitors program jumps during execution, effectively restraining abnormal program execution and robustly safeguarding against CRA. This paper provides a comprehensive analysis and synthesis of the current state of hardware-based CFI implementations. In this survey, we initially discuss common attack methods and variations of predominant CRA, elucidating the general procedural steps intrinsic to such attacks. We delve into the protective capacities inherent in contemporary hardware-based CFI implementations. By conducting a thorough examination and organization of diverse research endeavors on hardware-based CFI, we systematically classify CFI based on implementation methodologies, including label verification, instruction encryption, stack edge detection, instruction tracing, sensitive data isolation, and basic block validation. We provide comprehensive explanations and critical evaluations for each category followed by comparative analyses while offering personal insights on the evolution of hardware-based CFI.

Aquatic vegetation is an important component of aquatic ecosystems; therefore, the classification and mapping of aquatic vegetation is an important aspect of lake management. Currently, the decision tree (DT) classification method based on spectral indices has been widely used in the extraction of aquatic vegetation data, but the disadvantage of this method is that it is difficult to fix the threshold value, which, in turn, affects the automatic classification effect. In this study, Sentinel-2 MSI data were used to produce a sample set (about 930 samples) of aquatic vegetation in four inland lakes (Lake Taihu, Lake Caohai, Lake Honghu, and Lake Dongtinghu) using the visual interpretation method, including emergent, floating-leaved, and submerged vegetation. Based on this sample set, a DL model (Res-U-Net) was used to train an automatic aquatic vegetation extraction model. The DL model achieved a higher overall accuracy, relevant error, and kappa coefficient (90%, 8.18%, and 0.86, respectively) compared to the DT method (79%, 23.07%, and 0.77) and random forest (78%,10.62% and 0.77) when utilizing visual interpretation results as the ground truth. When utilizing measured point data as the ground truth, the DL model exhibited accuracies of 59%, 78%, and 91% for submerged, floating-leaved, and emergent vegetation, respectively. In addition, the model still maintains good recognition in the presence of clouds with the influence of water bloom. When applying the model to Lake Honghu from January 2017 to October 2023, the obtained temporal variation patterns in the aquatic vegetation were consistent with other studies. The study in this paper shows that the proposed DL model has good application potential for extracting aquatic vegetation data.

The hydrogenation of C=C bonds in styrene−butadiene rubber (SBR), catalyzed by RhH(P(i-Pr)3)3, was experimentally investigated. Tris(triisopropylphosphine)hydridorhodium(I), RhH(P(i-Pr)3)3 (i-Pr=CH(CH3)2) was prepared by using rhodium chloride (RhCl3), tetrahydrofuran (THF), triisopropylphosphine (P(i-Pr)3) and a sodium mercury amalgam. The effect of catalyst/polymer ratio, reaction temperature, and hydrogen pressure on the reactivity of the catalytic system has been studied. The optimal experimental condition was obtained. The hydrogenated styrene-butadiene rubber (HSBR) was analyzed by FT-IR and 1H-NMR. In the absence of any additives, the conversion of C=C bonds in SBR could easily reach 95% in a short period of time, and no obvious cross-linking was observed. The dynamic properties of SBR did not change after the hydrogenation of the unsaturated C=C bonds. A preliminary reaction mechanism was also proposed. This study provides a new route, not only for the chemical modification of SBR by using a rhodium complex but also for the hydrogenation of other unsaturated polymers, such as diene-based rubbers.

The development of stiffness theory is constrained by its contradiction with engineering experience. Several easily overlooked details of stiffness theory were clarified, and a qualitative evaluation formula for the risk of coal burst was provided. Then a novel structure factor called uneven stiffness coal seam structure (USCS), which consists of high stiffness zone (HSZ), low stiffness zone (LSZ), and contiguous roof and floor, was proposed. Many areas prone to coal bursts, such as thinning zones, bifurcating areas, magmatic intrusion areas, and remnant pillar affected areas of coal seam, are the HSZs of USCSs. Comparative analysis of the uneven stiffness coal seam under different roof conditions and examination of the simplified trisection model of the USCS were conducted. Then 6 groups of 14 simplified 2D models using COMSOL5.2 was constructed based on controlled variable method to simulate different responses of the USCS with varying parameters under same working conditions. The results demonstrate the following: (1) coal bursts occur only when both the failure criterion and the stiffness criterion are simultaneously satisfied, the risk of coal burst (rCB) is the product of the risk of failure (rF) and the risk of instability (rI). (2) The pressure concentration function of USCS facilitates stress concentration from LSZ to HSZ, thus raising the rF in HSZ. The stiffness reduction function of USCS reduces the local mine stiffness (LMS) of the HSZ, allowing the system to meet the stiffness criterion even with a hard roof, thereby raising the rI in HSZ and reconciling the contrast between stiffness theory and engineering experience. Failures within HSZ of the USCS enables the roof strata to release bending deformation energy without roof breakage. (3) The normal stress of HSZ is positively correlates with the value of ERHRKHSL/KLSH; The LMS of the HSZ is positively correlated with the value of ERKL/KHHRSLSH. The USCS boasts significant advantages in integrating and harmonizing various existing theories and explaining multiple specific types of coal bursts. By applying relevant USCS findings, new explanations can be provided for engineering phenomena such as the time-delayed coal bursts, the inefficient pressure relief in ultra thick coal seams, and the “microseism deficiency” observed prior to certain coal bursts.

The blast-induced vibration during excavation by the drilling and blasting method has an important impact on the surrounding buildings/structures and auxiliary equipment. In particular, with the development of tunnel engineering, the impact of blasting vibration on tunnel construction has attracted extensive attention. Based on literature data statistics, this paper first explored the performance of several commonly used empirical equations in predicting the propagation and attenuation characteristics of blasting vibration on adjacent tunnels. Secondly, the relationships between the empirical parameters of the blasting vibration prediction equation and the geological strength index (GSI) of tunnel surrounding rock were discussed, and two new blasting vibration prediction equations based on site rock GSI were established to approximately predict blast-induced vibration on adjacent tunnels. Finally, the application feasibility of the established prediction equation in practical engineering was discussed based on field test data. The research results show that under the condition of multiple groups of data, the prediction performance of various prediction models does not differ significantly. With the increase of the GSI of the surrounding rock mass of the adjacent tunnel, the site coefficients β and k of the blasting vibration prediction equation in predicting PPV (peak particle velocity, resultant velocity) both show a decreasing trend as a whole. The site coefficient k is generally within 3,000. Two new empirical prediction equations of blasting vibration propagation and attenuation on adjacent tunnels under different site conditions were established: Eq. (14) for PPV and Eq. (15) for PPVi (max) (maximum value of the three component velocities; i = x, y, z represent peak component particle velocity). The verification analysis of five sites shows that these two equations have a certain practical application value. Compared with other empirical equations, these two equations do not need regression fitting blasting vibration data, they only used the GSI of the site rock mass, and they are more easy to use in the field when there is a lack of monitoring data.

The imbalance of capacitor voltage on the DC side of the inverter will cause the third-order current harmonics and the device will be damaged greatly with the increase of voltage stress. The mechanism of imbalance capacitors voltage and the third-order current harmonics generated by the double frequency fluctuation are analyzed. This paper presents a feedforward capacitor differential pressure control strategy based on PR controller, which is aimed at the single-phase three-level NPC inverter and considers the cause of double modulation wave. The PR controller is used in the current loop to suppress the third-order harmonic by static-free tracking of the AC signal, and the voltage balance is realized by the feedforward DC-link capacitor voltage difference to offset the DC component of the output side. The simulation model and 1kW prototype driven by the control strategy have been established to demonstrate that the capacitor voltage of the inverter is balanced and the third-order harmonic is suppressed efficiently.

As coal mines continue to deepen high-intensity mining, the two disasters of coal seam rock burst and spontaneous combustion obviously have a trend of compound occurrence. How to achieve the integrated prevention and control of the two disasters is an important guarantee to realize coal mine safety. As a result, it is very important to select a coal seam water injection additive that can not only reduce the bursting liability, but also prolong the spontaneous combustion period of coal. In this paper, the chelating wetting agent was chosen to soak the coal, and the mass changed and carbon-containing functional groups during the coal oxidation process were studied by thermogravimetric and X-ray photoelectron spectroscopy, and tested the strength of coal by uniaxial compression experiment, and the scanning electron microscopy and low-temperature N2 adsorption were used to analyze the pore structure of coal. The experiment results show that the mass of the raw coal, the mass of the coal soaked in FeSO4 solution, and the mass of the coal soaked in the chelating wetting agent were reduced by 30.01%, 34.62% and 19.00%, respectively. The amount of the carbon–carbon bond and carbon–hydrogen bond decrease and the amount of carbon–oxygen bond increase in the chelating wetting agent soaking coal were less than in the water soaking coal and FeSO4 solution soaking coal. After soaking in the chelating wetting agent, the mechanical properties of the coal were changed, and the uniaxial compressive strength was reduced by 52.55%. The fractal dimension of the coal surface increased from 1.8448 to 1.8575, the average pore diameter in the coal was increased, the specific surface area was reduced, and the number of mesopores also increased. The above results can provide an important theoretical basis for the integrated prevention and control of rock burst-spontaneous combustion compound disaster.

Biomimetic nanocomposites are widely used in the biomedical field because they can effectively solve the problems existing in the current cancer treatment by realizing multi-mode collaborative treatment. In this study, we designed and synthesized a multifunctional therapeutic platform (PB/PM/HRP/Apt) with unique working mechanism and good tumor treatment effect. Prussian blue nanoparticles (PBs) with good photothermal conversion efficiency were used as nuclei and coated with platelet membrane (PM). The ability of platelets (PLTs) to specifically target cancer cells and inflammatory sites can effectively enhance PB accumulation at tumor sites. The surface of the synthesized nanocomposites was modified with horseradish peroxidase (HRP) to enhance the deep penetration of the nanocomposites in cancer cells. In addition, PD-L1 aptamer and 4T1 cell aptamer AS1411 were modified on the nanocomposite to achieve immunotherapy and enhance targeting. The particle size, UV absorption spectrum and Zeta potential of the biomimetic nanocomposite were determined by transmission electron microscope (TEM), Ultraviolet-visible (UV-Vis) spectrophotometer and nano-particle size meter, and the successful preparation was proved. In addition, the biomimetic nanocomposites were proved to have good photothermal properties by infrared thermography. The cytotoxicity test showed that it had a good killing ability of cancer cells. Finally, thermal imaging, tumor volume detection, immune factor detection and Haematoxilin-Eosin (HE) staining of mice showed that the biomimetic nanocomposites had good anti-tumor effect and could trigger immune response in vivo. Therefore, this biomimetic nanoplatform as a promising therapeutic strategy provides new inspiration for the current diagnosis and treatment of cancer.

The European Space Agency (ESA) developed the Sentinel-2 Multispectral Imager (MSI), which offers a higher spatial resolution and shorter repeat coverage, making it an important source for the remote-sensing monitoring of water bodies. Atmospheric correction is crucial for the monitoring of water quality. To compare the applicability of seven publicly available atmospheric correction processors (ACOLITE, C2RCC, C2XC, iCOR, POLYMER, SeaDAS, and Sen2Cor), we chose complex and diverse lakes in Qinghai Province, China, as the research area. The lakes were divided into three types based on the waveform characteristics of Rrs: turbid water bodies (class I lakes) represented by the Dabusun Lake (DBX), clean water bodies (class II lakes) represented by the Qinghai Lake (QHH), and relatively clean water bodies (class III lakes) represented by the Longyangxia Reservoir (LYX). Compared with the in situ Rrs, it was found that for the DBX, the Sen2Cor processor performed best. The POLYMER processor exhibited a good performance in the QHH. The C2XC processor performed well with the LYX. Using the Sen2Cor, POLYMER, and C2XC processors for classes I, II, and III, respectively, compared with the Sentinel-3 OLCI Level-2 Water Full Resolution (L2-WFR) products, it was found that the estimated Rrs from the POLYMER had the highest consistency. Slight deviations were observed in the estimation results for both the Sen2Cor and C2XC.

Quality degradation due to the formation and growth of ice crystals caused by temperature fluctuations during storage, transportation, or retailing is a common problem in frozen surimi. While commercial antifreeze is used as an ingredient in frozen surimi, its high sweetness does not meet the contemporary consumer demand for low sugar and low calories. Therefore, the development of new green antifreeze agents to achieve an enhanced frozen-thawed stability of surimi has received more attention. The aim of this study was to develop a cryoprotectant (a mixture of citric acid and trehalose) to enhance the frozen-thawed stability of surimi by inhibiting the oxidative denaturation and structural changes of frozen-thawed mirror carp (Cyprinus carpio L.) surimi myofibrillar protein (MP). The results showed that the amounts of free amine, sulfhydryl, α-helix, intrinsic fluorescence intensity, and thermal stability in the control significantly decreased after five F-T cycles, while the Schiff base fluorescence intensity, amounts of disulfide bonds and surface hydrophobicity significantly increased (p < 0.05). Compared to sucrose + sorbitol (SS), the natural deep eutectic solvents (NADES) effectively inhibited protein oxidation. After five F-T cycles, the α-helix content and Ca2+-ATPase activity of the NADES samples were 4.32% and 80.0%, respectively, higher, and the carbonyl content was 17.4% lower than those of the control. These observations indicate that NADES could inhibit oxidative denaturation and enhance the structural stability of MP.

Capacitive equipment refers to its insulation design using the principle of capacitance of electrical equipment, mainly by a variety of different capacitive components in series. Most of the equipment in the substation is capacitive equipment. Once an insulation failure occurs, it will lead to extremely serious consequences. Monitoring grid overvoltage and insulation degradation of capacitive equipment is an effective means to ensure the stable operation of the power system. Therefore, in order to enhance the health management of capacitive equipment, including transformers, bushings, and current transformers, and to mitigate the risk of severe failures, it is imperative to conduct broad-spectrum frequency-domain online monitoring of overvoltages, dielectric losses, and partial discharge. However, the current monitoring work requires the utilization of multiple detection apparatuses. Aiming at the disadvantage that the existing inspection is not well integrated and requires a combination of multiple devices. This paper proposes a smart grid overvoltage identification system that utilizes partial discharge (PD) signals in correlation with dielectric loss detection. The system achieves synchronous detection of dielectric loss and high-frequency partial discharge by synchronously and in real-time acquiring four current signals from the power grid, enhancing the integration level of the hardware system.

Abstract Circular RNAs have been reported to play essential roles in the tumorigenesis and progression of various cancers. However, the biological processes and mechanisms involved in hepatocellular carcinoma (HCC) remain unclear. Initial RNA-sequencing data and qRT-PCR results in our cohort showed that hsa_circ_0072309 (also called circLIFR) was markedly downregulated in HCC tissues. Kaplan–Meier analysis indicated that higher levels of circLIFR in HCC patients correlated with favorable overall survival and recurrence-free survival rates. Both in vitro and in vivo experiments indicated that circLIFR inhibited the proliferation and invasion abilities of HCC cells. We therefore conducted related experiments to explore the mechanism of circLIFR in HCC. Fluorescence in situ hybridization results revealed that circLIFR was mainly located in the cytoplasm, and RNA immunoprecipitation assays indicated that circLIFR was significantly enriched by Ago2 protein. These results suggested that circLIFR may function as a sponge of miRNAs to regulate HCC progression. We further conducted bioinformatics prediction as well as dual-luciferase reporter assays, and the results of which showed that circLIFR could sponge miR-624-5p to stabilize glycogen synthase kinase 3β (GSK-3β) expression. Loss and gain of function experiments demonstrated that regulation of the expression of miR-624-5p or GSK-3β markedly affected HCC progression induced by circLIFR. Importantly, we also proved that circLIFR could facilitate the degradation of β-catenin and prevent its translocation to the nucleus in HCC cells. Overall, our study demonstrated that circLIFR acts as a tumor suppressor in HCC by regulating miR-624-5p and inactivating the GSK-3β/β-catenin signaling pathway.

Abstract The roles of oncogenic KRAS in tumor immune evasion remain poorly understood. Here, mutant KRAS is identified as a key driver of tumor immune evasion in colorectal cancer (CRC). In human CRC specimens, a significant reduction in cytotoxic CD8+ T‐cell tumor infiltration is found in patients with mutant versus wild type KRAS. This phenomenon is confirmed by preclinical models of CRC, and further study showed KRAS mutant tumors exhibited poor response to anti‐PD‐1 and adoptive T‐cell therapies. Mechanistic analysis revealed lactic acid derived from mutant KRAS‐expressing tumor cells sensitized tumor‐specific cytotoxic CD8+ T‐cells to activation‐induced cell death via NF‐κB inactivation; this may underlie the inverse association between intratumoral cytotoxic CD8+ T‐cells and KRAS mutation. Importantly, KRAS mutated tumor resistance to immunotherapies can be overcome by inhibiting KRAS or blocking lactic acid production. Together, this work suggests the KRAS‐mediated immune program is an exploitable therapeutic approach for the treatment of patients with KRAS mutant CRC.

In animal husbandry, feed efficiency is a crucial economic trait. In this study, the general linear model was used to perform association analysis for various genotypes and feed conversion ratio (FCR)-related traits. Reverse transcription-quantitative PCR (RT-qPCR) was used to detect the expression of SHISA3 and RFC3 mRNA levels in 10 tissues from 6 sheep. The results showed that SNPs in the NC_040257.1:c.625 T > C and NC_040261.1:g.9905 T > C were analyzed whether they were associated to feed efficiency parameters in Hu sheep (body weight, feed intake, average daily growth, and feed conversion ratio). NC_040257.1:c.625 T > C was shown to be significantly associated with body weight at 80, 100, and 120 days as well as feed conversion ratio (P < 0.05), whereas NC_040261.1:g.9905 T > C was found to be significantly associated with average daily weight gain from 80-140 days (ADG80-140) and FCR (P < 0.05). In Hu sheep, the CC genotypes of SHISA3 and RFC3 were the most common genotypes related to feed efficiency traits. Furthermore, the feed conversion ratio of the combined genotypes TTSHISA3-CCRFC3, TTSHISA3-CTRFC3, TTSHISA3-TTRFC3, CTSHISA3-CCRFC3 and CTSHISA3-CTRFC3 was significantly better than the FCR of CCSHISA3-TTRFC3. RT-qPCR results showed that the expression levels of SHISA3 were lower in the lung than in spleen, kidney, muscle and lymph (P < 0.05), and RFC3 was the lung had a highly significant higher expression level than the heart, liver, spleen, and muscle (P < 0.01). In conclusion, SHISA3 and RFC3 polymorphisms can be used as genetic markers for improving feed conversion efficiency in Hu sheep.

Researchers use underwater acoustic equipment to explore the unknown ocean environment, which is one of the important means to understand and utilize the ocean. For underwater acoustic equipment, the application of underwater acoustic metamaterials is the premise to ensure and improve the performance of underwater acoustic communication, acoustic stealth, and sonar detection. Due to the limitations of mass density law and high hydrostatic pressure, traditional underwater acoustic materials cannot effectively absorb low-frequency sound waves and have low efficiency of elastic energy conversion. The sound absorption effect is poor under low frequency and high hydrostatic pressure. In recent years, with the development of acoustic metamaterials technology, all kinds of underwater acoustic metamaterials have also been proposed. Compared with sound waves propagating in the air, underwater sound is more difficult to control than air sound with the same frequency, so the design of underwater acoustic metamaterials is more complicated. This paper reviews the basic characteristics, development history of sound absorption, sound insulation decoupling, and underwater acoustic guided metamaterials, then the existing problems and the future development direction of underwater acoustic metamaterials are discussed.

IQ modulation enjoys great popularity in wireless communication. Its main advantage is the symmetric convenience of combining independent signal components into a single composite signals in the transmitter. And it splits the composite signal into its independent components in the receiver. However, due to the limitations of communication facilities and imperfect channel environments, the symbols will be distorted. In this paper, we propose several signal processing schemes based on deep learning (DL) neural networks (i.e., long short-term memory (LSTM)) to process signal in an end-to-end manner, known as DLA, EDLA and PDNet. These schemes combine with advanced DL architectures and data-driven models for complex-valued signal processing. Enlightened by IQ demodulation ideas, we adopt LSTMs to develop several schemes to implement different functions (i.e., signal detection and peak to average power ratio (PAPR) reduction). The simulation results show that the schemes have significant performance improvement in bit error rate (BER) while reducing the probability of high PAPR.

To understand the ultra-early reaction of normal organ lipids during irradiation, we investigated the response of lipids, including polyunsaturated fatty acid (PUFA) chains, which are particularly susceptible to damage by ROS, in mice’s kidneys, lungs, brains, and livers within 5 min of single high-dose irradiation. In this study, we set up three groups of C56BL/6 male mice and conducted whole-body irradiation with 0 Gy, 10 Gy, and 20 Gy single doses. Kidney, lung, brain, and liver tissues were collected within 5 min of irradiation. PUFA-targeted and whole lipidomic analyses were conducted using liquid chromatography–tandem mass spectrometry (LC-MS/MS). The results showed that PUFA chains of kidney phosphatidylcholine (PC), phosphatidylethanolamine (PE), and triacylglycerol (TG) significantly increased within 5 min of 10 Gy and 20 Gy irradiation. The main components of increased PUFA chains in PC and PE were C18:2, C20:4, and C22:6, and in TG the main component was C18:2. The kidney lipidomes also showed significant changes from the perspective of lipid species, mainly dominated by an increase in PC, PE, TG, and signal lipids, while lipidomes of the lung, brain, and liver were slightly changed. Our results revealed that acute PUFA chains increase and other lipidomic changes in the kidney upon whole-body irradiation within 5 min of irradiation. The significantly increased lipids also showed a consistent preference for possessing PUFA chains. The lipidomic changes varied from organ to organ, which indicates that the response upon irradiation within a short time is tissue-specific.

In this work, we design a micro-vibration platform, which combined with the traditional metal-assisted chemical etching (MaCE) to etch silicon nanowires (SiNWs). The etching mechanism of SiNWs, including in the mass-transport (MT) and charge-transport (CT) processes, was explored through the characterization of SiNW’s length as a function of MaCE combined with micro-vibration conditions, such as vibration amplitude and frequency. The scanning electron microscope (SEM) experimental results indicated that the etching rate would be continuously improved with an increase in amplitude and reached its maximum at 4 μm. Further increasing amplitude reduced the etching rate and affected the morphology of the SiNWs. Adjusting the vibration frequency would result in a maximum etching rate at a frequency of 20 Hz, and increasing the frequency will not help to improve the etching effects.
