Leptospiral organisms have long been presumed to be associated with the presence of equine recurrent uveitis. This project was undertaken to determine the presence of Leptospira spp. in the aqueous humor of horses with uveitis to determine if there was an association with inflammation. Thirty horses were determined to have recurrent uveitis based on clinical evaluation or history. Sixteen horses were judged clinically and historically to be free of uveitis and were used as controls. Aqueous humor samples were cultured and evaluated by PCR for the presence of Leptospira DNA. Serum was collected and evaluated for the presence of antibodies against five serovars in a leptospirosis panel. Twenty-one of 30 horses with recurrent uveitis and one of 16 uveitis-free horses were positive by PCR for the presence of Leptospira DNA. Six of these 21 horses with uveitis were culture positive for leptospires from the aqueous humor. Serologic results did not correlate well with the presence of Leptospira DNA or organisms in the aqueous humor. Leptospira spp. are present in a high percentage of horses with naturally occurring recurrent uveitis.

Male and female strain A/J mice were exposed to a mixture of cigarette sidestream and mainstream smoke at a chamber concentration of total suspended particulates of 82.5 mg/m3. Exposure time was 6 h/day, 5 days/week for 5 months. The animals were allowed to recover for another 4 months in filtered air before sacrifice and lung tumor count. Male animals were fed either 0.2% N-acetylcystein (NAC) or 0.05% phenethyl isothiocyanate (PEITC) in diet AIN-76A with 5% corn oil added. Female animals received normal laboratory chow and were given a 1.25% extract of green tea in the drinking water. Corresponding control groups were fed diets without NAC or PEITC or given plain tap water. Exposure to tobacco smoke increased lung tumor multiplicity to 1.1-1.6 tumors/lung, significantly higher than control values (0.5-1.0 tumors/lung). None of the putative chemopreventive agents (NAC, PEITC or green tea extract) had a protective effect. In positive control experiments, PEITC significantly reduced both lung tumor multiplicity and incidence in mice treated with the tobacco smoke-specific carcinogen 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK). In mice treated with three different doses of urethan and fed NAC in the diet, a significant reduction in lung tumor multiplicity was found only at one dose level. Green tea extract did not reduce lung tumor multiplicity in animals treated with a single dose of NNK. It was concluded that successful chemoprevention of tobacco smoke-induced lung tumorigenesis might require administration of several chemopreventive agents rather than just a single one. Spaying female and castrating male dogs, hereinafter referred to as neutering, is a US convention for the first year in the dog's life. Research on 35 breeds of dogs revealed that early neutering increases risks of joint disorders, such as hip dysplasia (HD), elbow dysplasia (ED), or cranial cruciate ligament (CCL) tear, or cancers, such as lymphosarcoma (LSA), mast cell tumor (MCT), hemangiosarcoma (has), or osteosarcoma (OSA), for some breeds. Joint disorder risks are heightened for some larger breeds and for mixed-breed dogs weighing more than 20 kg. Some breeds had elevated risks for cancers. Several other research teams have reported health complications associated with neutering. The study goal includes using the same methodology for data collection and analyses as in the study on 35 breeds for five additional dog breeds weighing at least 20 kg. The breeds were: German Short/Wirehaired Pointer, Mastiff, Newfoundland, Rhodesian Ridgeback, and Siberian Husky. Major differences among breeds appeared in vulnerability to joint disorders and cancers with early neutering: male and female Pointer breeds had elevated joint disorders and increased cancers; male Mastiff breeds had increased CCL and LSA and females had non-significant elevated CCL risks; female Newfoundland breeds had heightened risks for joint disorders and males had non-significant elevated risks; female Ridgeback breeds had heightened MCT with very early neutering; and Siberian Huskies showed no significant effects on joint disorders or cancers, but female breeds showed a non-significant but elevated CCL. Updated guidelines cover 40 dog breeds. The November 2018 Camp Fire, a devastating wildfire in Northern California, occurred during the peak of breeding season for field monkeys at the California National Primate Research Center (CNPRC). Effects of environmental stressors, such as wildfires, on birth outcomes in primates, and in humans, are poorly understood. Additionally, wildfires are of growing concern due to their increasing frequency and severity. The objective was to examine the impact of wildfire smoke on fertility, timing of birth, and pregnancy loss for field monkeys. A unique case-control study to investigate birth outcomes in rhesus macaques (Macaca mulatta) was conducted at the CNPRC. All females in the study were maintained in outdoor fields during a period of elevated ambient wildfire smoke from November 8-22, 2018. In addition to ambient air quality evaluations, the effects on fertility, timing to birth, and pregnancy loss were documented. Archival records of approximately 5,000 conceptions from the previous nine years served as control data. During the Camp Fire, ambient fine particulate (PM 2.5 ) levels exceeded the 24 -h National Ambient Air Quality Standard (35 μg/m 3 ) of the United States Environmental Protection Agency, reaching levels as high as 185 μg/m 3 . A statistically significant association was observed between birth loss and the 2018-2019 CNPRC breeding season. Plant eating by domestic cats is of interest to veterinarians and cat owners, especially with the current trend to keep cats totally indoors. Feline grass gardens are commonly provided to such cats as a reflection of cat owners believing in the need or desire of cats for eating plants. Two surveys with 1000 to 2000 returns from cat owners were launched over 10 years to test different hypotheses regarding plant eating. These hypotheses are that plant eating: (1) is a response to the cat feeling ill; (2) induces vomiting; (3) is a means of expelling hair balls from consumed hair. Additionally, a perspective acquired from observations of wild felids is that plant eating reflects an innate predisposition acquired from the ancestral cat. In this study, very few cats showed signs of illness before eating plants. However, 27 to 37 percent of cats, respectively in the two surveys, frequently vomited after eating plants, indicating that gastrointestinal disturbance may be related to vomiting in some cats. Young cats consumed plants more frequently than older cats and appeared ill and vomited less frequently in association with plant eating. Short-haired cats ate plants as frequently as long-haired cats, arguing against the hairball expelling hypothesis. Some guidelines for cat owners with indoor cats are provided.

The early neutering of male and female dogs and its relationship to an increased risk of joint disorders and some cancers has recently become a concern, raising questions about the standard practice in the U.S. and much of Europe of neutering by 6 months of age. A noteworthy recent finding from this center is that there are major breed differences with small-dog breeds generally showing little vulnerability to neutering compared with breeds of larger body size. These findings on purebreds raise questions for dog owners and veterinarians about mixed-breed dogs. The purpose of this study was to examine a sample of mixed breed dogs of five weight categories using the same veterinary hospital database and diagnostic criteria for joint disorders and cancers as used in the newly published paper on 35 breeds and previous papers on the Golden Retriever, Labrador Retriever, and German Shepherd Dog. The weight categories were <10 kg (739 cases), 10-19 kg (546 cases), 20-29 kg (992 cases), 30-39 kg (604 cases), and over 40 kg (258 cases). Males and females were analyzed separately, as were various ages at neutering. The joint disorders examined were hip dysplasia, cranial cruciate ligament tear or rupture, and elbow dysplasia. The cancers were lymphoma, mast cell tumor, hemangiosarcoma, and osteosarcoma. There was no significant increased occurrence of one or more cancers, compared with intact dogs, in any weight category. However, in the three categories of dogs weighing 20 kg or more, neutering before 1 year generally was significantly associated with risks of one or more joint disorders above that of dogs left intact, commonly to 3 times the level of intact dogs, with sex differences in the degrees of joint disorders associated with neutering. For the dogs in the two weight categories <20 kg, no increased risks were found for joint disorders. Neutering (including spaying) of male and female dogs in the first year after birth has become routine in the U.S. and much of Europe, but recent research reveals that for some dog breeds, neutering may be associated with increased risks of debilitating joint disorders and some cancers, complicating pet owners' decisions on neutering. The joint disorders include hip dysplasia, cranial cruciate ligament tear or rupture, and elbow dysplasia. The cancers include lymphoma, mast cell tumor, hemangiosarcoma, and osteosarcoma. In previous studies on the Golden Retriever, Labrador Retriever and German Shepherd Dog, neutering before a year of age was associated with increased risks of one or more joint disorders, 2-4 times that of intact dogs. The increase was particularly seen with dogs neutered by 6 months of age. In female Golden Retrievers, there was an increase in one or more of the cancers followed to about 2-4 times that of intact females with neutering at any age. The goal of the present study was to expand and use the same data collection and analyses to cover an additional 29 breeds, plus three varieties of Poodles. There were major breed differences in vulnerability to neutering, both with regard to joint disorders and cancers. In most cases, the caregiver can choose the age of neutering without increasing the risks of these joint disorders or cancers. Small-dog breeds seemed to have no increased risks of joint disorders associated with neutering, and in only two small breeds (Boston Terrier and Shih Tzu) was there a significant increase in cancers. To assist pet owners and veterinarians in deciding on the age of neutering a specific dog, guidelines that avoid increasing the risks of a dog acquiring these joint disorders or cancers are laid out for neutering ages on a breed-by-breed and sex basis., (Copyright © 2020 Hart, Hart, Thigpen and Willits.)

Assistance dogs' roles have diversified to support people with various disabilities, especially in the U.S. Data presented here are from the U.S. and Canada non-profit facilities (including both accredited and candidate members that fulfilled partial requirements: all here termed "accredited") of Assistance Dogs International (ADI) and the International Guide Dog Federation (IGDF), and from non-accredited U.S. assistance dog training facilities, on the numbers and types of dogs they placed in 2013 and 2014 with persons who have disabilities. ADI categories of assistance dogs are for guide, hearing, and service (including for assistance with mobility, autism, psychiatric, diabetes, seizure disabilities). Accredited facilities in 28 states and 3 provinces responded; accredited non-responding facilities were in 22 states and 1 province (some in states/provinces with responding accredited facilities). Non-accredited facilities in 16 states responded. U.S./Canada responding accredited facilities (55 of 96: 57%) placed 2,374 dogs; non-accredited U.S. facilities (22 of 133: 16.5%) placed 797 dogs. Accredited facilities placed similar numbers of dogs for guiding ( n = 918) or mobility ( n = 943), but many more facilities placed mobility service dogs than guide dogs. Autism service dogs were third most for accredited ( n = 205 placements) and U.S. non-accredited ( n = 72) facilities. Psychiatric service dogs were fourth most common in accredited placements ( n = 119) and accounted for most placements ( n = 526) in non-accredited facilities. Other accredited placements were for: hearing ( n = 109); diabetic alert ( n = 69), and seizure response ( n = 11). Responding non-accredited facilities placed 17 hearing dogs, 30 diabetic alert dogs, and 18 seizure response dogs. Non-accredited facilities placed many dogs for psychiatric assistance, often for veterans, but ADI accreditation is required for veterans to have financial reimbursement. Twenty states and several provinces had no responding facilities; 17 of these states had no accredited facilities. In regions lacking facilities, some people with disabilities may find it inconvenient living far from any supportive facility, even if travel costs are provided. Despite accelerated U.S./Canada placements, access to well-trained assistance dogs continues to be limited and inconvenient for many people with disabilities, and the numerous sources of expensive, poorly trained dogs add confusion for potential handlers., (Copyright © 2019 Walther, Yamamoto, Thigpen, Willits and Hart.)

Although studies involving pet dogs and cats, and human adults and children, have been reported, the specific interactions between cats and children have not. This study sought information from parents about the cat's role in families that have at least one child 3-12 years of age and at least one cat. Demographic data on cat source, breed, gender/neuter status, was sought as well as information on adults and children in the families and on affectionate, aggressive, fearful, and playful responses of the cats to children. A convenience sample was recruited via listservs for pet owners and parents. Using a pilot tested web survey, descriptive statistics were based on 865 respondents. Multi-variate statistical analyses were conducted on data from 665 respondents with complete responses for all items, including respondents' locations and whether cats were adopted as kittens. Multi-variate analyses included consideration of demographic data, geographic region of respondents, behavioral characteristics of the cats, and responses of the children to the cats. From descriptive statistics, cats' affection was more typical with adults than young children. Neuter status or gender was unrelated to cats' aggression or affection. Being the family's only cat was associated with heightened aggression and reduced affection. Younger cats were more likely to be affectionate. Multivariate analysis revealed three primary factors accounting for children's compatibility with the specified cat: positive interactions of the cat, aggression/fearfulness of cat, and the cat's playfulness and children's reaction to the cats. Positive child-cat relationships were more typical with two or more adults and multiple cats in the home. Old cats were the least satisfactory. A breeder or shelter was a better source than as a feral, from a newspaper ad, or another source. European respondents rated their cats' interactions with children more favorably than in U.S./Canada. This difference may reflect the European adoptions more frequently being of kittens, often purebred, assuring more early handling within the family. A noteworthy finding was that all family participants, humans, and pets alike, affect the cat-child relationship, and these results reveal that many variables can play a role in achieving a desirable relationship for a cat and child.

Mental and physical benefits of dogs have been reported for adults and children with special needs, but less is known about benefits of cats for children. A cat that can be held by a child could provide important therapeutic companionship for children with severe or less severe autism spectrum disorder (ASD) who otherwise may lack prosocial behaviors. Because relatively little is known about the behavior of cats around children, we conducted this study. Phase 1 gathered web-survey data from families having an adult cat and a child with ASD ( n  = 64). In Phase 2, there were direct telephone interviews of parents having a child with severe ASD ( n  = 16) or less severe ASD ( n  = 11), or typical development ( n  = 17). From the Phase 1 web survey of families with ASD children (full range of severities), affectionate interactions of the cats with children were common. Most parents with ASD children volunteered positive comments regarding the cat, such as calming the child, being a soothing protector or a guardian. In the interviews in Phase 2, for all three groups, most parents characterized cats as at least moderately affectionate toward the child. However, cats living with severe ASD children were reported to exhibit less affection than those living with typically developing children or children with less severe ASD. A minority of cats in each group showed some aggression to the specified child; this was not elevated with ASD children. Responses suggested that the cats adopted as kittens were more affectionate and less aggressive to all categories of children than those adopted as adults. Overall, participants reported that ASD children's behaviors indicated that they valued the relationship with the cat, similar to typically developing children, pointing to the importance and potential usefulness of selecting affectionate and compatible cats for ASD children.

Sexually abused children providing essential testimony regarding crimes in forensic interviews now sometimes are provided facility dogs or therapy dogs for comfort. Facility dogs are extensively trained to work with forensic interviewers; when using therapy dogs in interviews, volunteers are the dog handlers. Interviews can impact child welfare workers' mental health causing secondary traumatic stress (STS). To investigate this stress, first data were gathered on stress retrospectively for when interviewers initially started the job prior to working with a dog, and then currently, from forensic interviewers using a facility dog, a therapy or pet dog, or no dog. These retrospective and secondary traumatic stress scale (STSS) data compared job stress among interviewers of children using: a certified, workplace facility dog ( n  = 16), a volunteer's trained therapy dog or the interviewer's pet dog ( n  = 13/3), or no dog ( n  = 198). Retrospective scores of therapy dog and no dog interviewers' stress were highest for the first interviewing year 1 and then declined. Extremely or very stressful retrospective scores differed among the three groups in year 1 ( p  < 0.038), and were significantly elevated for the therapy dog group as compared with the facility dog group ( p  < 0.035). All interviewing groups had elevated STSS scores; when compared with other healthcare groups that have been studied, sub-scores were especially high for Avoidance: a psychological coping mechanism to avoid dealing with a stressor. STSS scores differed among groups ( p  < 0.016), primarily due to Avoidance sub-scores ( p  < 0.009), reflecting higher Avoidance scores for therapy dog users than no dog users ( p  < 0.009). Facility dog users more consistently used dogs during interviews and conducted more interviews than therapy/pet dog users; both groups favored using dogs. Interviewers currently working with therapy dogs accompanied by their volunteers reported they had experienced heightened stress when they began their jobs; their high stress levels still persisted, indicating lower inherent coping skills and perhaps greater empathy among interviewers who later self-selected to work with therapy dogs. Results reveal extreme avoidant stress for interviewers witnessing children who are suffering and their differing coping approaches.

Dogs' roles to support people with disabilities are increasing. Existing U.S. laws and regulations pertaining to the use of dogs for people with disabilities are only minimally enforced. Pushback legislation against some aspects of uses of assistance dogs currently is being passed or proposed in several states. Further, the U.S. Department of the Army and the Veterans' Administration support only dogs trained by an Assistance Dogs International (ADI) or International Guide Dog Federation (IGDF) accredited facility. Lacking a mandatory national process for screening the selection, training, and placement of assistance dogs with persons who have disabilities, the U.S. offers a creative but confusing opportunity for people to train their own dogs for any disability. While no U.S. surveillance system monitors assistance dogs, other countries generally have a legislated or regulatory process for approving assistance dogs or a cultural convention for obtaining dogs from accredited facilities. We conducted an online survey investigating current demographics of assistance dogs placed in 2013 and 2014 with persons who have disabilities, by facilities worldwide that are associated with ADI or IGDF and by some non-accredited U.S. facilities. Placement data from ADI and IGDF facilities revealed that in most countries aside from the U.S., guide dogs were by far the main type of assistance dog placed. In the U.S., there were about equal numbers of mobility and guide dogs placed, including many placed by large older facilities, along with smaller numbers of other types of assistance dogs. In non-accredited U.S. facilities, psychiatric dogs accounted for most placements. Dogs for families with an autistic child were increasing in all regions around the world. Of dog breeds placed, accredited facilities usually mentioned Labrador Retrievers and Golden Retrievers, and sometimes, German Shepherd Dogs. The facilities bred their dogs in-house, or acquired them from certain breeders. Non-accredited facilities more often used dogs from shelters or assisted people in training their own dogs. Facilities in Europe and the U.S. place dogs in all roles; other parts of the world primarily focus on guide dogs. Expansion of assistance dogs in many roles is continuing, with numbers of dogs placed accelerating internationally.

German Shepherd Dogs are important in police and military work, and are a popular family pet. The debilitating joint disorders of hip dysplasia, cranial cruciate ligament tear (CCL) and elbow dysplasia can shorten a dog's useful working life and impact its role as a family member. For this study, veterinary hospital records were examined over a 14.5-year period on 1170 intact and neutered (including spaying) German Shepherd Dogs for joint disorders and cancers previously associated with neutering. The diseases were followed through 8 years of age, with the exception of mammary cancer (MC) in females that was followed through 11 years. The cancers followed, apart from mammary, were osteosarcoma, lymphoma, hemangiosarcoma and mast cell tumour. In intact males, 7% were diagnosed with one or more joint disorders, while in males neutered prior to a year of age, a significantly higher 21% were diagnosed with one or more joint disorders. In intact females, 5% were diagnosed with one or more joint disorders, while in females neutered prior to a year of age, this measure was significantly increased to 16%. The increased joint disorder incidence mostly associated with early neutering was CCL. MC was diagnosed in 4% of intact females compared with less than 1% in females neutered before 1 year. The occurrence of the other cancers followed through 8 years of age was not higher in the neutered than in the intact dogs. Urinary incontinence, not diagnosed in intact females, was diagnosed in 7% of females neutered before 1 year, a significant difference. These findings, profiling the increase in joint disorders associated with early neutering, should help guide the timing of neutering for this breed.

Objectives: To evaluate the effects of a multiple daily dose methadone regimen in pregnancy on neonatal outcomes., Methods: Although methadone maintenance has been the standard for the treatment of opioid dependence in pregnancy, there is no consensus on proper dosing. Single daily dosing is the most common strategy. Because of accelerated metabolism of methadone in pregnancy, this regimen may expose mother and fetus to daily episodes of withdrawal and possibly contribute to more severe Neonatal Abstinence Syndrome (NAS). This study reports on a protocol that increased both methadone dose and dose frequency in response to maternal reports of withdrawal., Results: Treatment of NAS was needed in 29% of neonates, compared to a published rate of 60% to 80%. The mean methadone dose was 152 mg at delivery, divided into 2 to 6 doses per day. Ninety-two percent of mothers were free of illicit drug use at delivery. There was no relationship between methadone dose and treatment of NAS. Female babies had a treatment rate of 16% versus 38% for male babies. Beyond abstinence symptoms, cohort outcomes in terms of gestational age, birth weight, prematurity, Caesarian sections, and breastfeeding equaled or approximated US population norms., Conclusions: The protocol was associated with low rates of treatment of NAS and high rates of maternal recovery. High rates of treatment for NAS reported in methadone-exposed neonates might relate in part to iatrogenic factors and be reduced through the use of divided daily doses and protocols that minimize maternal withdrawal.

Our recent study on the effects of neutering (including spaying) in Golden Retrievers in markedly increasing the incidence of two joint disorders and three cancers prompted this study and a comparison of Golden and Labrador Retrievers. Veterinary hospital records were examined over a 13-year period for the effects of neutering during specified age ranges: before 6 mo., and during 6-11 mo., year 1 or years 2 through 8. The joint disorders examined were hip dysplasia, cranial cruciate ligament tear and elbow dysplasia. The cancers examined were lymphosarcoma, hemangiosarcoma, mast cell tumor, and mammary cancer. The results for the Golden Retriever were similar to the previous study, but there were notable differences between breeds. In Labrador Retrievers, where about 5 percent of gonadally intact males and females had one or more joint disorders, neutering at <6 mo. doubled the incidence of one or more joint disorders in both sexes. In male and female Golden Retrievers, with the same 5 percent rate of joint disorders in intact dogs, neutering at <6 mo. increased the incidence of a joint disorder to 4-5 times that of intact dogs. The incidence of one or more cancers in female Labrador Retrievers increased slightly above the 3 percent level of intact females with neutering. In contrast, in female Golden Retrievers, with the same 3 percent rate of one or more cancers in intact females, neutering at all periods through 8 years of age increased the rate of at least one of the cancers by 3-4 times. In male Golden and Labrador Retrievers neutering had relatively minor effects in increasing the occurrence of cancers. Comparisons of cancers in the two breeds suggest that the occurrence of cancers in female Golden Retrievers is a reflection of particular vulnerability to gonadal hormone removal.

Objective: To evaluate the thermal antinociceptive and sedative effects and duration of action of tramadol hydrochloride after oral administration to American kestrels (Falco sparverius)., Animals: 12 healthy 3-year-old American kestrels., Procedures: Tramadol (5, 15, and 30 mg/kg) and a control suspension were administered orally in a masked randomized crossover experimental design. Foot withdrawal response to a thermal stimulus was determined 1 hour before (baseline) and 0.5, 1.5, 3, 6, and 9 hours after treatment. Agitation-sedation scores were determined 3 to 5 minutes before each thermal stimulus test., Results: The lowest dose of tramadol evaluated (5 mg/kg) significantly increased the thermal foot withdrawal thresholds for up to 1.5 hours after administration, compared with control treatment values, and for up to 9 hours after administration, compared with baseline values. Tramadol at doses of 15 and 30 mg/kg significantly increased thermal thresholds at 0.5 hours after administration, compared with control treatment values, and up to 3 hours after administration, compared with baseline values. No significant differences in agitation-sedation scores were detected between tramadol and control treatments., Conclusions and Clinical Relevance: Results indicated oral administration of 5 mg of tramadol/kg significantly increased thermal nociception thresholds for kestrels for 1.5 hours, compared with a control treatment, and 9 hours, compared with baseline values; higher doses resulted in less pronounced antinociceptive effects. Additional studies with other types of stimulation, formulations, dosages, routes of administration, and testing times would be needed to fully evaluate the analgesic and adverse effects of tramadol in kestrels and other avian species.

Objective: To evaluate antinociceptive effects and pharmacokinetics of butorphanol tartrate after IM administration to American kestrels (Falco sparverius)., Animals: Fifteen 2- to 3-year-old American kestrels (6 males and 9 females)., Procedures: Butorphanol (1, 3, and 6 mg/kg) and saline (0.9% NaCl) solution were administered IM to birds in a crossover experimental design. Agitation-sedation scores and foot withdrawal response to a thermal stimulus were determined 30 to 60 minutes before (baseline) and 0.5, 1.5, 3, and 6 hours after treatment. For the pharmacokinetic analysis, butorphanol (6 mg/kg, IM) was administered in the pectoral muscles of each of 12 birds., Results: In male kestrels, butorphanol did not significantly increase thermal thresholds for foot withdrawal, compared with results for saline solution administration. However, at 1.5 hours after administration of 6 mg of butorphanol/kg, the thermal threshold was significantly decreased, compared with the baseline value. Foot withdrawal threshold for female kestrels after butorphanol administration did not differ significantly from that after saline solution administration. However, compared with the baseline value, withdrawal threshold was significantly increased for 1 mg/kg at 0.5 and 6 hours, 3 mg/kg at 6 hours, and 6 mg/kg at 3 hours. There were no significant differences in mean sedation-agitation scores, except for males at 1.5 hours after administration of 6 mg/kg., Conclusion and Clinical Relevance: Butorphanol did not cause thermal antinociception suggestive of analgesia in American kestrels. Sex-dependent responses were identified. Further studies are needed to evaluate the analgesic effects of butorphanol in raptors.

Platelet-rich plasma (PRP) products may be useful for treatment of joint disease in horses, but may contain undesirable pro-inflammatory cytokines in addition to growth factors. This study investigated whether autologous PRP increases synovial fluid growth factor and cytokine concentrations when injected into normal equine metacarpophalangeal and metatarsophalangeal (fetlock) joints. Fetlock joints of seven healthy horses received one of four treatments: saline, resting PRP, CaCl2-activated PRP or thrombin-activated PRP. Synovial fluid was sampled prior to injection and at 6, 24, 48 and 96 h post-injection. Platelet-derived growth factor (PDGF-BB), transforming growth factor β1 (TGFβ1), interleukin (IL)-6 and tumor necrosis factor α (TNFα) concentrations in synovial fluid and PRP were measured by ELISA. Synovial fluid PDGF-BB, TGFβ1, IL-6, TNFα and IL-1 concentrations were also measured in vitro after incubation for 6h with resting PRP only. Growth factor concentrations, but not cytokine concentrations, were significantly higher in activated PRP than in resting PRP samples. After intra-articular injection with resting or thrombin-activated PRP, synovial TGFβ1 increased significantly compared to baseline levels. TNFα and IL-6 were significantly increased in synovial fluid after thrombin-activated PRP injection. In vitro, growth factor concentrations increased significantly in synovial fluid after mixing with PRP, indicating that exogenous activation of PRP for intra-articular injection may be unnecessary, whereas cytokine levels did not. In conclusion, thrombin-activated PRP induced an inflammatory cytokine response in joints, whereas resting or CaCl2-activated PRP did not. Synovial growth factor levels were low overall; the reported benefits of intra-articular PRP may not be attributable to changes in local PDGF or TGFβ1 concentrations., (Copyright © 2013 Elsevier Ltd. Objective: To evaluate the antinociceptive and sedative effects and duration of action of hydromorphone hydrochloride after IM administration to American kestrels (Falco sparverius)., Animals: 11 healthy 2-year-old American kestrels., Procedures: Hydromorphone (0.1, 0.3, and 0.6 mg/kg) and an equivalent volume of saline (0.9% NaCl) solution (control treatment) were administered IM to kestrels in a masked randomized complete crossover study design. Foot withdrawal response to a thermal stimulus was determined 30 to 60 minutes before (baseline) and 0.5, 1.5, 3, and 6 hours after treatment administration. Agitation-sedation scores were determined 3 to 5 minutes before each thermal test., Results: Hydromorphone at 0.6 mg/kg, IM, significantly increased the thermal foot withdrawal threshold, compared with the response after administration of saline solution, for up to 3 hours, and hydromorphone at 0.1, 0.3, and 0.6 mg/kg, IM, significantly increased withdrawal responses for up to 6 hours, compared with baseline values. No significant differences in mean sedation-agitation scores were detected between hydromorphone and saline solution treatments; however, appreciable sedation was detected in 4 birds when administered 0.6 mg of hydromorphone/kg., Conclusions and Clinical Relevance: Hydromorphone at the doses evaluated significantly increased the thermal nociception threshold for American kestrels for 3 to 6 hours. Additional studies with other types of stimulation, formulations, dosages, routes of administration, and testing times are needed to fully evaluate the analgesic and adverse effects of hydromorphone in kestrels and other avian species and the use of hydromorphone in clinical settings.

Seasonal variation in dengue virus transmission in northwestern Thailand is inversely related to the magnitude of diurnal temperature fluctuations, although mean temperature does not vary significantly across seasons. We tested the hypothesis that diurnal temperature fluctuations negatively influence epidemiologically important life-history traits of the primary dengue vector, Aedes aegypti (L.), compared with a constant 26 degrees C temperature. A large diurnal temperature range (DTR) (approximately equals 18 degrees C daily swing) extended immature development time (>1 d), lowered larval survival (approximately equals 6%), and reduced adult female reproductive output by 25% 14 d after blood feeding, relative to the constant 26 degreesC temperature. A small DTR (approximately equal 8 degrees C daily swing) led to a negligible or slightly positive effect on the life history traits tested. Our results indicate that there is a negative impact of large DTR on mosquito biology and are consistent with the hypothesis that, in at least some locations, large temperature fluctuations contribute to seasonal reduction in dengue virus transmission.

In many animals, exposure to cues in a natal habitat increases disperser preferences for those cues (natal habitat preference induction [NHPI]), but the proximate and ultimate bases for this phenomenon are obscure. We developed a Bayesian model to study how different types of experience in the natal habitat and survival to the age/stage of dispersal interact to affect a disperser's estimate of the quality of new natal-type habitats. The model predicts that the types of experience a disperser had before leaving its natal habitat will affect the attractiveness of cues from new natal-type habitats and that favorable experiences will increase the level of preference for natal-type habitats more than unfavorable experiences will decrease it. An experimental study of NHPI in Drosophila melanogaster provided with "good" and "bad" experiences in their natal habitats supports these predictions while also indicating that the effects of different types of natal experience on NHPI vary across genotypes. If habitat preferences are modulated by an individual's experience before dispersal as described in this study, then NHPI may have stronger effects on sympatric speciation, metapopulation dynamics, conservation biology, and pest management than previously supposed.

The present work describes a two-stage approach to analyzing combustion-generated samples for their potential to produce oxidant stress. This approach is illustrated with the two commonly encountered transition metals, copper and iron. First, their abilities to generate hydroxyl radical were measured in a cell-free, phosphate-buffered saline solution containing ascorbate and/or citrate. Second, their abilities to induce heme oxygenase-1 in cultured human epidermal keratinocytes were assessed in cell culture. Combustion-generated copper oxide nanoparticles were active in both assays and were found to be soluble in culture medium. Depletion of glutathione in the cells or loading the cells with ascorbate greatly increased heme oxygenase-1 induction in the presence of copper. By contrast, iron oxide nanoparticles were active in the phosphate-buffered saline but not in cell culture, and they aggregated in culture medium. Soluble salts of copper and iron exhibited the same contrast in activities as the respective combustion-generated particles. The results suggest that the capability of combustion-generated environmental samples to produce oxidant stress can be screened effectively in a two step process, first in phosphate-buffered saline with ascorbate and subsequently in epithelial cell culture for those exhibiting activity initially. The results also point to an unanticipated interaction in cells of oxidant stress-generating metals with an antioxidant (ascorbate) that is usually missing in culture medium formulations. Thus, ascorbate supplementation of cultured human cells is likely to improve their ability to model the in vivo effects of particulate matter containing copper and other redox-active metals.

Koi herpesvirus (KHV) has been associated with devastating losses of common carp (Cyprinus carpio carpio) and koi (Cyprinus carpio koi) in North America, Europe, Israel and Asia. A comparison of virion polypeptides and genomic restriction fragments of seven geographically diverse isolates of KHV indicated that with one exception they represented a homogeneous group. A principal environmental factor influencing the onset and severity of disease is water temperature. Optimal growth of KHV in a koi fin cell line occurred at temperatures from 15-25 degrees C. There was no growth or minimal growth at 4, 10, 30 or 37 degrees C. Experimental infections of koi with KHV at a water temperature of 23 degrees C resulted in a cumulative mortality of 95.2 %. Disease progressed rapidly but with lower mortality (89.4-95.2 %) at 28 degrees C. Mortality (85.0 %) also occurred at 18 degrees C but not at 13 degrees C. Shifting virus-exposed fish from 13-23 degrees C resulted in the rapid onset of mortality.

Using Western-blot analysis and enzyme-linked immunosorbent assay (ELISA) of N-deglycosylated samples, we have observed that plasma levels of fibronectin (FN) bearing the alternatively spliced EIIIB segment (EIIIB(+) FN) increase in patients after admission to the intensive-care unit (ICU) for acute major trauma. Although not increased at the first sampling ("0 hour"), taken within 24 hours of ICU admission, levels measured 24, 48, and 72 hours later were significantly increased compared with levels obtained in healthy controls. Furthermore, average concentrations at these latter time points were significantly increased relative to the 0-hour sampling. EIIIB(+) FN levels then decreased in plasma samples taken 1 month after hospital discharge, such that no significant difference was found between ELISA-measured values at this time and 0 hour or control values. On the basis of comparisons with previous measurements in these samples, it is apparent that after acute major trauma, circulating levels of soluble EIIIB(+) FN exhibit temporal changes that are qualitatively similar to those encountered for FN isoforms bearing the alternatively spliced EIIIA segment (EIIIA(+) FN), yet different from those observed for the total pool of circulating FN. This is the first report of measurement of FN in clinical plasma samples with antibodies specific for the highly conserved EIIIB segment. Like EIIIA(+) forms of FN, EIIIB(+) FNs are recognized as soluble bloodborne markers for vascular tissue injury.

Objective: To evaluate a dorsoproximal extra-articular approach for insertion of 8.25-mm, solid-titanium, intramedullary (IM) interlocking nails into ostectomized foal third metacarpal (MC3) and third metatarsal (MT3) bones; to compare the monotonic mechanical properties of IM nail constructs with paired intact bones; and to determine the effects of age, body weight, fore- or hindlimb, and left or right limb on the mechanical testing variables., Animal or Sample Population: Twenty bone pairs (10 MC3, 10 MT3) collected from 10 foals of various weights and ages., Methods: One bone from each pair was randomly selected to be ostectomized and stabilized using an 8.25-mm, solid-titanium IM nail, and four 3.7-mm titanium interlocking screws (construct). Constructs and contralateral intact bone specimens were tested in axial compression and palmaro-/plantarodorsal (PD) 4-point bending. Monotonic mechanical properties were compared between intact specimens and constructs with an ANOVA; significance was set at P <.05., Results: Nail insertion caused bone failure in 6 MC3 and 2 MT3. In general, mean mechanical testing values indicated that intact specimens were significantly stronger and stiffer than constructs for all age and weight ranges when tested in compression and PD 4-point bending (P <.05). Bone strength and stiffness of intact specimens tested in compression and bending tended to increase linearly with age and weight., Conclusion: IM interlocking nail fixation of gap-ostectomized MC3 and MT3 with 8.25-mm IM nails and 3.7-mm interlocking screws did not achieve sufficient strength or stiffness to be recommended as the sole means of repair for comminuted MC3 and MT3 fractures in young foals., Clinical Relevance: IM interlocking nail fixation of foal cannon bone fractures may be useful to decrease soft-tissue disruption at the fracture site; however, there is a risk of bone failure associated with extra-articular insertion. Objective: To compare drilling, tapping, and screw-insertion torque, force, and time for the 4.5-mm AO and 6.5-mm Acutrak Plus (AP) bone screws, and to compare the mechanical shear strength and stiffness of a simulated complete lateral condylar fracture of the equine third metacarpal bone (MC3) stabilized with either an AO or AP screw., Study Design: In vitro biomechanical assessment of screw-insertion variables, and shear failure tests of a bone-screw-stabilized simulated lateral condylar fracture., Sample Population: Eight pairs of cadaveric equine MC3s, Methods: Metacarpi were placed in a fixture and centered on a biaxial load cell in a materials-testing system to measure torque, compressive force, and time for drilling, tapping, and screw insertion. Standardized simulated lateral condylar fractures were stabilized by either an AO or AP screw and tested in shear until failure. A paired t test was used to assess differences between screws, with significance set at P < .05., Results: Insertion and mechanical shear testing variables were comparable for AO and AP insertion equipment and screws., Conclusion: The 6.5-mm tapered AP screw can be inserted in equine third metacarpal condyles and is mechanically comparable with the 4.5-mm AO screw for fixation of a simulated lateral condylar fracture., Clinical Relevance: Considering the comparable mechanical behavior, the potential for less-persistent soft-tissue irritation with the headless design, and the ability to achieve interfragmentary compression by inserting the screw in one hole drilled perpendicular to the fracture plane, the 6.5-mm tapered AP screw may be an attractive alternative for repair of incomplete lateral condylar fractures in horses., (Copyright 2001 by The American College of Veterinary Surgeons)

The bronchiolar injury/repair response to naphthalene (NA) in mice includes acute distal airway epithelial injury that is followed by epithelial proliferation and redifferentiation, which result in repair of the epithelium within 14 days. To test whether prior exposure to aged and diluted sidestream cigarette smoke (TS) would alter the injury/repair response of the airway epithelium, adult mice were exposed to either filtered air (FA) or smoke for 5 days before injection with either corn oil carrier (CO) or naphthalene. Mice were killed 1 and 14 days after naphthalene injury. Lung and lobar bronchus were examined and measured using high-resolution epoxyresin sections. The control group (FACOFA) that was exposed to filtered air/corn oil/filtered air contained airway epithelium similar to untreated controls at all airway levels. The group exposed to tobacco smoke/corn oil/filtered air (TSCOFA) contained some rounded cells in the small airways and some expansion of the lateral intercellular space in the larger airways. Necrotic or vacuolated cells were not observed. As expected, the epithelium in the group exposed to filtered air/naphthalene/filtered air (FANAFA) contained many light-staining vacuolated Clara cells and squamated ciliated cells within distal bronchioles during the acute injury phase. Repair (including redifferentiation of epithelial cells and restoration of epithelial thickness) was nearly complete 14 days after injury. The extent of Clara cell injury, as assessed in lobar bronchi, was not different between the four groups. Although the FANAFA group contained greater initial injury in the distal airways at 1 day, the group exposed to tobacco smoke/naphthalene/filtered air (TSNAFA) had the least amount of epithelial repair at 14 days after naphthalene treatment; many terminal bronchioles contained abundant squamated undifferentiated epithelium. We conclude that tobacco smoke exposure prior to injury (1) does not change the target site or target cell type of naphthalene injury, since Clara cells in terminal bronchioles are still selectively injured; (2) results in slightly diminished acute injury from naphthalene in distal bronchioles; and (3) delays bronchiolar epithelial repair.

Objective: To develop an objective, accurate method for quantifying forelimb ground reaction forces in horses by adapting a human in-shoe pressure measurement system and determine the reliability of the system for shod and unshod horses., Animals: 6 adult Thoroughbreds., Procedure: Horses were instrumented with a human in-shoe pressure measurement system and evaluated at a trot (3 m/s) on a motorized treadmill. Maximum force, stance time, and peak contact area were evaluated for shod and unshod horses. Three trials were performed for shod and unshod horses, and differences in the measured values were examined with a mixed model ANOVA for repeated measures. Sensor accuracy was evaluated by correlating measured variables to clinically observed lameness and by a variance component analysis., Results: 4 of 6 horses were determined to be lame in a forelimb on the basis of clinical examination and measured values from the system. No significant differences were observed between shod and unshod horses for maximum force and stance time. A significant decrease in peak contact area was observed for shod and unshod horses at each successive trial. Maximum force measurements provided the highest correlation for detecting lameness (r = 0.91, shod horses; r = 1.0, unshod horses). A variance component analysis revealed that 3 trials provided a variance of 35.35 kg for maximum force (+/- 5.78% accuracy), 0.007 seconds for stance time (+/- 2.5% accuracy), and 8.58 cm2 for peak contact area (+/- 11.95% accuracy)., Conclusions and Clinical Relevance: The in-shoe pressure measurement system provides an accurate, objective, and effective method to evaluate lameness in horses.

Objective: To evaluate the effects of dimethyl sulfoxide (DMSO) on equine articular cartilage matrix metabolism., Study Design: Using a cartilage explant culture system, proteoglycan (PG) synthesis, PG release, lactate metabolism, chondrocyte viability, and metabolism recovery were determined after cartilage exposure to DMSO., Sample Population: Cartilage harvested from metacarpophalangeal and metatarsophalangeal joints of 12 horses (age range, 1 to 10 years)., Methods: Explants were exposed to concentrations of DMSO (1% to 20%) for variable times (3 to 72 hours). PG synthesis and release were determined by a radiolabel incorporation assay and dimethylmethylene blue (DMMB) dye assay, respectively. Lactate released into culture media was measured, and chondrocyte viability was assessed using the Formizan Conversion Assay and a paravital staining protocol. Metabolism recovery was assessed in explants that were allowed to recover in maintenance media after exposure to DMSO., Results: PG synthesis and lactate metabolism were inhibited in a dose- and time-dependent manner after exposure to DMSO concentrations > or = 5%; there was no significant alteration in PG release. No change in chondrocyte viability was detected after incubation with DMSO. PG synthesis and lactate metabolism returned to baseline rates when allowed a recovery period after exposure to DMSO., Conclusions: DMSO concentrations > or = 5% suppress equine articular cartilage matrix metabolism. Suppression of PG synthesis and lactate metabolism is reversible and does not appear to be the result of chondrocyte death., Clinical Relevance: Equine clinicians adding DMSO to intraarticular lavage solutions should be aware that DMSO may have deleterious effects on equine articular cartilage matrix metabolism.

Objectives: To compare the biomechanical properties, in full limb preparations, of intact second phalanx and a simulated comminuted second phalangeal fracture stabilized with either two bone plates or a custom Y-plate., Study Design: In vitro biomechanical assessment of intact limbs and of paired limbs with a simulated second phalangeal fracture stabilized by one of two fixation methods. Animal Population-Thirteen pairs of equine cadaveric forelimbs., Methods: A comminuted second phalangeal fracture was created in six paired cadaveric limbs. For each limb pair, the fracture was stabilized with two plates in one limb, and with a Y-plate in the contralateral limb. These limbs and seven pairs of intact limbs were subjected to axial compression in a single cycle until failure. Mechanical properties were compared with a mixed-model ANOVA and post hoc contrasts. Joint contact pressure, screw insertion torque, and final screw torque remaining after mechanical testing were also evaluated for constructs., Results: No significant differences in mechanical testing variables were detected between construct types. However, the Y-Plate construct had significantly greater yield load, yield displacement and yield energy, and failure load and stiffness values than those for intact specimens, whereas the double-plate construct only had greater stiffness than intact specimens. There were no significant differences in joint contact pressures for both constructs. The final screw torque for proximal phalangeal screws was significantly greater for the Y-plate constructs than for double-plate constructs., Conclusions: The Y-plate was as effective as the double-plate technique for stabilization of simulated comminuted second phalangeal fractures in monotonically tested equine cadaveric forelimbs., Clinical Relevance: This investigation supports evaluation of the Y-plate for repair of comminuted second phalangeal fractures in equine patients. Its specific design may facilitate repair of second phalangeal fractures, and may provide increased stability by allowing the proximal fragments of the second phalanx to be fixed with three screws placed through the plate.

Objective: To document natural bacterial flora on the ventral aspect of the equine abdomen, to compare 2 preparation techniques, and to identify potential risk factors that may contribute to incisional drainage., Design: Prospective study., Animals: 53 horses undergoing exploratory celiotomy., Procedure: Group-1 horses (n = 26) were prepared with povidone-iodine and alcohol. Group-2 horses (27) were prepared with a film-forming iodophor complex. Numbers of bacterial colony-forming units (CFU) were measured before and after surgical scrub, following skin closure, and after recovery from general anesthesia. Swab specimens to identify normal skin bacterial flora and potential pathogens were obtained by swabbing a 4 x 4-cm area. Variables that might affect incisional drainage were also investigated., Results: For both techniques, there was a significant reduction in bacterial numbers after skin preparation. Incisional drainage was observed in 14 (26%) horses (8 group-1 and 6 group-2 horses). Preexisting dermatitis, poor intraoperative drape adherence, high number of bacterial CFU obtained after recovery from anesthesia, and high number of CFU obtained from the surgery room environment were the main risk factors associated with subsequent incisional drainage. Bacillus spp, nonhemolytic Staphylococcus spp, Micrococcus spp, Corynebacterium spp, Streptomyces spp, other nonenteric genera, and nonhemolytic Streptococcus spp were the most common isolates obtained before surgical scrub., Conclusions and Clinical Relevance: Both skin preparation techniques were equally effective in reducing numbers of bacterial CFU by 99%, and a significant difference was not found in incisional drainage rate between groups. Protection of the wound during recovery from anesthesia and the immediate postoperative period may reduce incisional drainage after abdominal surgery in horses.

Objective: To identify factors associated with various arterial partial pressures of oxygen (Pao2) in anesthetized horses., Design: Retrospective study., Animals: 1,450 horses anesthetized a total of 1,610 times with isoflurane or halothane., Procedure: Anesthesia records, particularly results of blood gas analyses, were reviewed, and horses were grouped on the basis of lowest Pao2., Results: For horses with lowest Pao2 < 120 mm Hg, those with low pulse pressure, that underwent anesthesia on an emergency basis, or that were males were more likely to have Pao2 < 80 mm Hg. For horses with lowest Pao2 < 250 mm Hg, those that were positioned in dorsal recumbency, that underwent anesthesia on an emergency basis, or that had a shorter duration of anesthesia were more likely to have lowest Pao2 < 120 mm Hg. For horses with lowest Pao2 < 400 mm Hg, those that were positioned in dorsal recumbency, that underwent anesthesia on an emergency basis, that had a shorter duration of anesthesia, that were older, that were heavier, or that were being ventilated mechanically were more likely to have lowest Pao2 < 250 mm Hg., Conclusions and Clinical Relevance: Low pulse pressure, emergency case status, dorsal recumbency, and short duration of anesthesia were significantly related with lower Pao2 in anesthetized horses.

Objective: To describe the prevalence, characteristics, and severity of soft-tissue and osseous lesions in the caudal portion of the thoracic and lumbosacral vertebral column and pelvis in Thoroughbred racehorses., Animals: 36 Thoroughbred racehorses that died or were euthanatized at California racetracks between October 1993 and July 1994., Procedure: Lumbosacral and pelvic specimens were collected and visually examined for soft-tissue and osseous lesions., Results: Acute sacroiliac joint injury was observed in 2 specimens. Signs of chronic laxity or subluxation of the sacroiliac joint were not observed in any specimens. Impingement of the dorsal spinous processes and transverse processes was observed in 92 and 97% of specimens, respectively. Thoracolumbar articular processes had variable degrees of degenerative change in 97% of specimens. Degenerative changes were observed at lumbar intertransverse joints and sacroiliac articulations in all specimens. Some degenerative changes were widespread and severe., Conclusions: Numerous degenerative changes affected vertebral processes, intervertebral articulations, and sacroiliac joints in these Thoroughbred racehorses., Clinical Relevance: Various types of vertebral and pelvic lesions need to be considered during clinical evaluation of the back and pelvis in horses. Undiagnosed vertebral or pelvic lesions could be an important contributor to poor performance and lameness in athletic horses.

This study sought to determine if extensive trimming of the front hooves of foals results in a higher incidence of palmar process fractures compared to untrimmed foals, and to characterise the clinical course of foals with palmar process fractures with physical findings, hoof measurements and radiography. Twenty foals age 4-8 weeks of multiple breeds were examined every 2 weeks over a 12 week period. Ten foals had both front hooves extensively trimmed every 4 weeks, while 10 foals remained untrimmed. Palmar process fractures occurred in 4 trimmed and 3 untrimmed foals. Four foals with fractures had brief lameness (Grades I-II/V) and hoof tester sensitivity was found inconsistently with fractures. All foals with fractures were sound at the end of the study. Fractures healed based on radiographic evaluations in an average of 8.4 weeks. Two of 3 foals with fractures developed club-footed conformation. None of the measured hoof parameters significantly differed between foals with and without fractures. We conclude that extensive trimming of the heels did not appear to affect the occurrence of palmar process fractures in this group of foals.

Objective: To determine anatomic landmarks for a lateral approach for arthrocentesis of the proximopalmarolateral aspect of the distal interphalangeal (DIP) joint in horses and the likelihood of entering synovial structures other than the DIP joint through this approach., Design: Prospective study., Sample Population: Paired forelimbs of 8 cadavers and 12 horses., Procedure: Anatomic preparations were used to determine anatomic landmarks. Positive-contrast arthrography was used to determine which structures were entered., Results: Landmarks for the lateral approach included a depression in the proximal border of the lateral ungular cartilage and the palmar border of the middle phalanx. Use of the lateral approach for arthrography resulted in deposition of contrast material exclusively in the DIP joint in only 13 of 20 limbs, whereas use of the dorsal approach resulted in deposition of contrast material exclusively in the DIP joint in 20 of 20 limbs., Clinical Implications: The lateral approach is an alternative to the conventional dorsal approach to the DIP joint in horses, however, inadvertent entry into adjacent synovial structures is a possible complication. The lateral approach provides an additional portal for through-and-through lavage and arthroscopic access to the palmar aspect of the DIP joint.

The objective of this study was to examine several simple ex vivo loading conditions for the equine metacarpus, and to evaluate their ability to reproduce the mid-diaphyseal bone surface strain distributions previously reported in vivo at the walk and trot. Distributed axial compressive loads, and 9 different axial compressive point loads at -7.5 kN and -15 kN were applied to metacarpal-distal carpal bone preparations from 6 Thoroughbred horses, aged 1-5 years. The resulting dorsal, medial, palmar, and lateral mid-diaphyseal bone surface axial and shear strains were compared with previously reported in vivo surface strain distributions using a root mean square error (RMSE) protocol. The effects of loading condition and load magnitude on RMSE were assessed with a mixed-model analysis of variance. There were significant differences between loading conditions, and, in most cases, between load magnitudes, in the fit of the ex vivo to the in vivo strain distributions. In vivo mid-diaphyseal bone surface strain distributions at the walk can be well approximated ex vivo by a distributed axial compressive load, or by a point load positioned 0.5 cm medial to the sagittal midline, at -7.5 kN loads. In vivo mid-diaphyseal bone surface strain distributions at the trot can be well approximated by the -15 kN loads applied to the same locations. These simplified loading conditions can be used in designing biologically relevant loading protocols for ex vivo mechanical testing studies, as well as in developing boundary conditions for finite element analysis. As such, these loading conditions may be considered as tools to be used as a means of replicating in vivo loading conditions during the initial design and testing stages in the development of fracture fixation devices, as well as in the theoretical mechanical analysis of the metacarpal structure.

Objective: To describe the incidence and types of gross osseous developmental variations and ages of physeal closure in the caudal portion of the thoracic and lumbosacral spine and the pelvis in a sample of Thoroughbred racehorses., Animals: Thoroughbred racehorses (n = 36) that died or were euthanatized at California racetracks between October 1993 and July 1994., Procedure: Lumbosacropelvic specimens were collected, and all soft tissues were removed. The osseous specimens were visually examined., Results: Only 22 (61%) specimens had the expected number of 6 lumbar and 5 sacral vertebrae. Eight (22%) specimens had thoracolumbar transitional vertebrae, and 13 (36%) had sacrocaudal transitional vertebrae. Articular process asymmetries were present at 1 or more vertebral segments in 30 (83%) specimens. Intertransverse joints (2 to 4 pairs/specimen) were bilaterally distributed in the caudal portion of the lumbar spine and the lumbosacral joint in 31 (86%) specimens. Five (14%) specimens had asymmetric distribution of the intertransverse joints. Intertransverse joint ankylosis was found in 10 (28%) specimens. Lumbosacral vertebral body physeal closure occurred between 4.9 and 6.7 years of age; pelvic physeal closure occurred between 5.2 and 5.8 years of age. Iliac crest and ischial arch epiphyseal formation was evaluated, using a grading system, and fusion to the underlying bone occurred at 7.2 years and 5.4 years of age, respectively., Conclusions: Numerous vertebral anatomic variations were commonly found in a sample of Thoroughbred racehorses., Clinical Relevance: Normal anatomic variations and ages of skeletal maturity need to be considered in clinical evaluation of the equine spine and pelvis for differentiation from pathologic findings.

Social interactions in red-backed salamanders, Plethodon cinereusare complex and often depend upon the sex and reproductive status of individuals. In the population at Mountain Lake Biological Station, Virginia, adult males outnumber sexually active (gravid) females by approximately 2:1, because males court annually while females accept mates only biennially. Field observations of male P. cinereus suggest that males maintain feeding territories and allow non-gravid (not sexually active) females access to their territories for foraging, whereas they defend their territories against intrusion by males. We used a game-theoretic analysis of models of male fitness to examine conditions that would favour the evolution of this 'permissive' behaviour. Under a series of increasingly realistic behavioural assumptions, we found that the critical factor in the evolution of male permissive behaviour is female preference for permissive males. If non-gravid females are more likely to return to mate with a male that allows access to his territory, then permissive behaviour is likely to evolve. Furthermore, the evolution of permissive behaviour is facilitated when energetic losses due to territorial defence exceed those due to allowing females to forage.1997The Association for the Study of Animal Behaviour

The distribution of material properties within the equine third metacarpal bone (MC3), and its possible effect on the mechanics of the structure, was quantitatively evaluated using single-load-to-failure compressive materials testing of specimens from ten horses. Bone samples from six regions within five proximodistal levels of MC3 were milled into right cylinders and compressed at a strain rate of 0.01 s-1. Diaphyseal MC3 bone material was stiffer, stronger, deformed less to yield and failure, and absorbed more energy to yield, than metaphyseal cortical bone material. Lateral and medial MC3 cortical bone material was stiffer and deformed less to yield and failure, than dorsal and palmar material. This distribution of material properties appears to increase the structural compliance in the sagittal plane, and may serve to enhance the predictability of the strain distribution during normal locomotion, as is provided in other bones by a sagittal curvature.

The effects of weight gain and subsequent weight loss on glucose tolerance and insulin response were evaluated in 12 healthy cats. Intravenous glucose tolerance tests (IVGTT) were performed at entry into the study, after a significant gain of body weight induced by feeding palatable commercial cat food ad libitum, after a significant loss of body weight induced by feeding a poorly palatable purified diet to discourage eating and promote fasting, and after recovery from fasting when body weight had returned to pre-study values and cats were eating commercial foods. A complete physical examination with measurement of body weight was performed weekly, a CBC and serum biochemistry panel were evaluated at the time of each IVGTT, and a liver biopsy specimen obtained 2 to 4 days after each IVGTT was evaluated histologically for each cat. Mean serum glucose and insulin concentrations after glucose infusion and total amount of insulin secreted during the second 60 minutes and entire 120 minutes after glucose infusion were significantly (P < .05) increased after weight gain, as compared with baseline. At the end of weight loss, cats had hepatic lipidosis and serum biochemical abnormalities consistent with feline hepatic lipidosis. There was a significant (P < .05) increase in mean serum glucose concentration and t1/2, and a significant (P < .05) decrease in mean serum insulin concentration and the glucose disappearance coefficient (K) after glucose infusion for measurements obtained after weight loss, compared with those obtained after weight gain and at baseline. Insulin peak response, insulinogenic index, and total amount of insulin secreted during the initial 10 minutes, 20 minutes, and 60 minutes after glucose infusion were decreased markedly (P < .05), compared with measurements obtained after weight gain and at baseline. In addition, the total amount of insulin secreted for 120 minutes after glucose infusion was decreased markedly (P < .05) in measurements obtained after weight loss, compared with those obtained after weight gain. At the end of recovery, all cats were voluntarily consuming food, serum biochemical abnormalities identified after weight loss had resolved, the number and size of lipid vacuoles in hepatocytes had decreased, and results of IVGTT were similar to those obtained at baseline. These findings confirmed the reversibility of obesity-induced insulin resistance in cats, and documented initial deterioration in glucose tolerance and insulin response to glucose infusion when weight loss was caused by severe restriction of caloric intake.

Stress associated with road transport is believed to be a significant contributor to the pathogenesis of post transport respiratory disease in horses. To determine the effects of road transport on pulmonary function, pulmonary aerosol clearance rates were measured in 4 horses 24 h before, and immediately after, 24 h of road transport by delivering aerosolised 99mtechnetium-labelled diethylenetriaminepentacetate (99mTc-DTPA) to the lungs and monitoring its washout. Each horse was transported twice, once while the trailer was equipped with a leaf-spring suspension and bias-ply tyres (trailer's original equipment, smooth ride) and once while the trailer was equipped with a torsion-bar suspension and normal pressure radial tyres (rough ride) in order to generate different ride characteristics. Before transport, blood was drawn from each horse for haematology and measurement of serum cortisol concentration; 24 h rates of hay and water intake and faecal output were recorded for each horse. Horses were then transported, 2 at a time, over a 128 km circular route of predominantly rural freeways at a constant speed of 72 km/h for 24 h. Horses were rested by stopping the trailer every 3.75 h for 0.25 h. During transport, heart rates (continuous 1 min averages), rates of hay and water intake and rates of faecal output were measured. Ammonia (NH3) and carbon monoxide (CO) concentrations were measured within the trailer and temperatures (wet bulb [WB], dry bulb [DB] and black globe [BG]) within the trailer were recorded each minute. Immediately after each experiment blood was drawn for haematology and measurement of pulmonary aerosol clearance rates were measured. For control studies, horses were housed in their stalls while heart rates were measured for 24 h. Slopes calculated from the 99mTc-DTPA clearance curves for pretransport horses were not significantly different from post transport clearance slopes. Pretransport mean 99mTc-DTPA clearance half-lives (T50, left lung mean +/- s.d. 41.7 +/- 15.8 min, right lung 44.6 +/- 19.1 min) were not significantly different from post transport T50 (left lung 53.5 +/- 14.0 min, right lung 52.0 +/- 11.6 min). Heart rates during transport were not affected by suspension type or trip order (the horse's first or second transport experiment) and were not significantly different from stall controls after the first 120 min of the experiment. Horses had increased red blood cell count, packed cell volume, haemoglobin, plasma protein and cortisol concentrations, and decreased body weights immediately post transport, indicating slight dehydration. Water and hay intake rates were significantly lower during transport than pretransport. Temperatures within the trailer were highest in the midafternoon and lowest in the early morning hours, but all temperatures measured in the trailer were within the comfort zone for large homeotherms. Ammonia and CO concentrations in the trailer during the transport period were within acceptable limits for human exposure. However, respirable articulates in the atmosphere were elevated above safe concentrations for human exposure.

The torsional monotonic structural material properties of equine metacarpi with or without, either a 5/16 inch or 3/8 inch diameter bicortical lateromedial middiaphyseal hole were assessed to determine the effect of a hole on metacarpal strength. Torsional stiffness was not significantly effected by the presence of a bicortical hole, whereas yield and failure angles, torques and energies of metacarpi with a hole were 51% to 97% of those of intact bones. Significant differences were not apparent for yield and failure mechanical properties between metacarpi with a 5/16 inch diameter hole and metacarpi with a 3/8 inch diameter hole; however, postyield mechanical properties were lower for metacarpi with a 3/8 inch hole. Whereas some metacarpi with a 5/16 inch hole were capable of plastic deformation before failure, metacarpi with a 3/8 inch diameter hole appeared to have sufficient stress concentration to propagate complete fracture on structural yield.

Most experiments conducted to determine the optimal essential amino acid (EAA) nitrogen to total nitrogen (E:T) ratio in rats, chicks and other species have shown that weight gain and nitrogen retention are optimal when the ratio is between 0.5 and 0.65. Two experiments were conducted to determine if weight gains of kittens fed EAA as a sole source of dietary nitrogen were equivalent to those of kittens fed a control diet that contained equal amounts of EAA and dispensable amino acids (DAA). In the first experiment, kittens fed diets that contained only EAA lost weight. Amino acid analysis of plasma showed that the concentration of methionine was 9.1 times that of controls, supporting the premise that the weight loss that occurred was the result of an adverse effect of excess methionine. Also, plasma threonine and arginine were elevated whereas proline and asparagine were very low. In the second experiment, feeding lower concentrations of methionine and arginine in a diet containing only EAA resulted in weight gains and nitrogen retention that were not significantly different from results for kittens fed the control diet (E:T ratio = 0.5). The addition of proline and asparagine to this EAA diet or their removal from the control diet did not improve or reduce weight gain or nitrogen retention. It is concluded that the decreased weight gain found in kittens (and probably other species) fed only EAA is the result of an adverse effect of excesses of methionine and possibly other EAA, and not the inability to synthesize DAA. If excesses of certain EAA are avoided, near-maximal weight gain can be achieved without any DAA in the diet.

Objective: To study the effects of inhalation anesthetic agents on the response of horses to 3 hours of hypoxemia., Design: Controlled crossover study., Animals: Five healthy adult horses., Procedure: Horses were anesthetized twice: once with halothane, and once with isoflurane in O2. Anesthetized horses were positioned in left lateral recumbency. Constant conditions for the study began at 2 hours of anesthesia. A constant agent dose of 1.2 minimum alveolar concentration, PaO2 of 50 +/- 5 mm of Hg, and PaCO2 of 45 +/- 5 mm of Hg were maintained for 3 hours. Circulatory measurements were made at 0.5, 1, 2, and 3 hours of hypoxemia (anesthesia hours 2.5, 3, 4, and 5). Blood was collected from horses for biochemical analyses before anesthesia, within a few minutes after standing, and at 1, 2, 4, and 7 days after anesthesia., Results: Cardiac index was greater (P = 0.018) during isoflurane than halothane anesthesia. Cardiac index remained constant during the 3 hours of hypoxemia during halothane anesthesia, whereas it decreased from the baseline during isoflurane anesthesia. Marginally nonsignificant P values for an agent difference were detected for arterial O2 content (P = 0.051), and oxygen delivery (P = 0.057). Serum activities of aspartate transaminase (P = 0.050) and sorbitol dehydrogenase (P = 0.017) were higher in halothane-anesthetized horses than in isoflurane-anesthetized horses. Circulatory function was better in hypoxemic horses anesthetized with isoflurane than with halothane. Isoflurane resulted in less muscular injury in hypoxemia horses than did halothane anesthesia. Halothane anesthesia and hypoxemia were associated with hepatic insult., Conclusion: Isoflurane is better than halothane for hypoxemic horses.

This is the first in a series of experiments to study the fatigue properties of equine cannon (third metacarpal) bone specimens from Thoroughbred racehorses. Monotonic and fatigue tests to failure were performed in four-point bending on diaphyseal specimens in a 37 degrees C saline bath to answer three initial questions. (a) Will a linear variable differential transducer yield the same elastic modulus as strain gauges? (b) Will fatigue results depend on whether the periosteal or endosteal side of the beam is in tension? (c) Are there regional variations in the monotonic and fatigue properties of the cannon bone midshaft? Eighteen left-right pairs of specimens from six horses were used. One beam of each pair was fitted with strain gauges. Fatigue tests were conducted on 24 specimens under load control at 2 Hz; an initial range of 0-10,000 microstrain was used so as to produce failure in a reasonable period of time. There were no left-right differences in the fatigue or monotonic properties, and the presence of a gauge had no effect on modulus measured by a linear variable differential transducer. However, gauge-measured moduli were about 1 GPa less than transducer-measured values. Fatigue life was independent of which side of the beam was in tension, and there were significant variations in mechanical properties around the cortex. The lateral region was stiffer than the dorsal region but the latter had a longer fatigue life. The fixed cylindrical supports used in this experiment eventually produced slight wear grooves, causing artifactual stiffening at the end of the load cycle in some specimens. A second experiment using roller supports confirmed the reason for this stiffening. It also showed that fatigue life was shorter when roller supports were used but regional differences were similar.

Paired metacarpi obtained at necropsy from 100 horses ranging in age from term fetus to 35 years were examined to estimate the prevalence and sites of metacarpal fusion. Metacarpal fusion was seen in 192 of 200 metacarpi, and 78% of all horses 2 years or older had 2 or more fusions. Fusion of the second metacarpal bone to the third metacarpal bone was significantly (P < 0.001) more common than was fusion of the fourth to the third metacarpal bone. Fusions appeared for the most part in pairs and were bilaterally symmetric. Rooney-Prickett type-A carpometacarpal joint configurations (in which there is no measurable articulation between the third carpal and second metacarpal bones) were rare in this population, and Rooney-Prickett type-B configurations (in which there is a measurable articulation between the third carpal and second metacarpal bones) were observed in 98.5% of metacarpi. Medial metacarpal fusion was positively correlated with age, occupation, and proportion of the proximal projection of the carpometacarpal distal joint surface that was taken by the second metacarpal bone. Lateral metacarpal fusion was positively correlated with age and the proportion of the proximal projection of the carpometacarpal distal joint surface taken by the fourth metacarpal bone. Horses in performance careers (racing, race training, or show ring occupations) had an earlier development of the first 2 fusions than did horses in other or unknown occupations; development of the third and fourth fusions were not significantly different between occupation groups. The rate of metacarpal fusion per horse-year appeared to be at least 10 times higher than a clinically evident rate. A variety of gross morphologic features was observed in the fusions from this sample, some of which were small, subtle, and possibly difficult to detect in vivo. It is hypothesized that many instances of metacarpal fusion may be a result of functional adaptation of the metacarpus to increased or changed loading conditions, rather than a response to isolated traumatic events.

To evaluate the efficacy of penicillin or penicillin and dexamethasone for treatment of infectious bovine keratoconjunctivitis, 6- to 8-month-old beef heifers with clinical signs of infectious bovine keratoconjunctivitis were randomly assigned to 1 of 3 treatment groups: penicillin only, penicillin and dexamethasone, or control. Cattle assigned to the penicillin group (n = 18) were treated with 3 daily subconjunctival injections of procaine penicillin G. Cattle assigned to the penicillin/dexamethasone group (n = 13) were treated with 3 daily subconjunctival injections of procaine penicillin G and dexamethasone sodium phosphate. Control cattle (n = 14) were not treated. Healing times and frequency of recurrence for corneal ulcers; severity, diameter, and surface area measurements of corneal ulcers; and clinical scores did not differ among the 3 groups. Frequency of Moraxella bovis isolation from specimens of ocular secretions from ulcerated and non-ulcerated eyes was similar in all groups. Minimum inhibitory concentration of penicillin G for 95 of the 102 tested M bovis isolates was 0.3 U/ml, and for 7 others was 0.03 U/ml. When first and last specimens from 42 of 45 calves with isolation of M bovis on serial microbial cultures were compared, the susceptibility of each last isolate was similar to that of the corresponding first isolate.

The metabolic responses of equine articular cartilage to incubation with bacterial lipopolysaccharide (LPS) were studied, using explant cultures of articular cartilage obtained from the metatarsophalangeal joints of 15 horses, age of which ranged from 3 months to 20 years. For comparison, explants were also established from the metatarsophalangeal joints of 3 calves. Explants were cultured for 3 days in medium containing various concentrations of LPS from 0 (control) to 100 micrograms/ml. Glycosaminoglycan (GAG) released during the 3-day incubation was determined by a spectrophotometric assay, using the dye 1,9-dimethylmethylene blue. Newly synthesized GAG content was assayed by measuring [35S]sulfate incorporation during a 3-hour pulse labeling period. In addition, prostaglandin E2 (PGE2) synthesis was quantified, using a [3H]PGE2 radioimmunoassay kit and magnetic separation. Finally, explants from 3 animals were used to evaluate the effect of supplementing culture medium with 5% serum on the response of explants to LPS, and explants from 1 horse were used to compare responses to stimulation with LPS derived from 2 bacterial sources. Equine explants cultured with bacterial LPS had a dose-dependent decrease in synthesis and increase in release of GAG, and these responses were significantly (P < 0.0001) greater in explants from younger horses. In addition, equine explants had a significant (P = 0.0001) dose-dependent increase in concentration of PGE2 released into the culture medium in response to incubation with LPS. Comparison of data for GAG synthesis from equine and bovine explants revealed a significant (P = 0.025) difference in responsiveness to LPS between the 2 species. Equine explants tended to have a greater suppression of GAG synthesis in response to incubation with increasing concentrations of LPS than did age-corrected bovine samples. However, similar analysis of data on GAG release did not indicate any difference in sensitivity between the 2 species for this response. There was no evidence that the presence or absence of serum supplementation or the use of LPS derived from different bacterial sources made a significant difference in the response of explants to incubation with LPS.

Six foals were deprived of colostrum for the first 36 hours after birth and, instead, received reconstituted powdered milk. Five control foals suckled their dams naturally. Blood samples were obtained from all the foals after birth and at approximately weekly intervals until at least 5.5 months of age. Sera were analyzed for hemolytic complement activity, complement component C3, and correlating IgG concentration. Hemolytic complement (P = 0.0145) and C3 (P = 0.0002) values were significantly higher in colostrum-deprived foals (CDF) than in naturally nursed foals at 2 to 5 days of age. In addition, significantly (P = 0.0149) higher IgG concentration was found in CDF than in naturally nursed foals between 3 and 5.5 months of age. It was concluded that the observed high complement activity in CDF within 2 to 5 days of age may provide an alternative in immune defense for IgG-deprived foals after failure of colostral transfer.
