The experiment aimed to establish a method for quantitatively measuring the concentration of bacterial endotoxins in cat food using the dynamic turbidity method. Bacterial endotoxins in cat food samples were extracted with water, centrifuged, filtered, and the filtrate was appropriately diluted. Quantitative determination was carried out using the dynamic turbidity method on a bacterial endotoxin measuring instrument. The results showed good linearity of bacterial endotoxin within the range of 0.040 to 5.000 EU/mL (correlation coefficient r=-0.995 3, >0.980). The standard curve was effective, and the relative standard deviation (RSD) of repeated measurements ranged from 8.9% to 14.0% ( n=6). The recovery rates of interference tests were between 50% and 200% when the dilution factor was ≥1 000. The limit of quantification (LOQ) of the method was determined as 0.04 EU/mg. The study indicates that the method is simple, reliable, accurate, and exhibits good reproducibility, making it effective for detecting bacterial endotoxin content in cat food samples, with promising application prospects. The unique thermogenic capacity of brown adipocyte makes it an attractive target for antiobesity treatments. Several epigenetic regulators can control brown adipocyte development. In this study, we show that SIRT5, a member of the sirtuins, is required for brown adipocyte differentiation and essential for brown adipogenic gene activation in vitro. Furthermore, we find out that knockdown of SIRT5 reduces intracellular α-ketoglutarate concentration, which leads to elevated H3K9me2 and H3K9me3 levels at promoter regions of Pparγ and Prdm16 loci. Finally, we discover that SIRT5 knockout mice on the Sv129 background exhibit less browning capacity in subcutaneous white adipose tissue compared with controls and show apparent cold intolerance, suggesting that SIRT5 can modulate the browning process in vivo. Thus, our study uncovers a new biological function of SIRT5 in brown adipocyte differentiation and a mechanism by which SIRT5 regulates brown adipogenic gene activation at least partly through an indirect effect on histone modifications. AMP-activated protein kinase (AMPK) is an energy sensor of metabolism that is an attractive therapeutic target for type 2 diabetes mellitus and metabolic syndrome. Using a homogeneous scintillation proximity assay (SPA), we identified a new small-molecule AMPK activator, ZLN024, which allosterically stimulated active AMPK heterotrimers and the inactive α1 subunit truncations α1 (1–394) and α1 (1–335) but not α1 (1–312). AMPK activation by ZLN024 requires the pre-phosphorylation of Thr-172 by at least one upstream kinase and protects AMPK Thr-172 against dephosphorylation by PP2Cα. ZLN024 activated AMPK in L6 myotubes and stimulated glucose uptake and fatty acid oxidation without increasing the ADP/ATP ratio. ZLN024 also activated AMPK in primary hepatocytes, decreased fatty acid synthesis and glucose output. Treatment of db/db mice with 15 mg/kg/day ZLN024 improved glucose tolerance; liver tissue weight, triacylglycerol and the total cholesterol content were decreased. The hepatic transcriptional level of G6Pase, FAS and mtGPAT were reduced. The transcription of genes involved in fatty acid oxidation and the mitochondrial biogenesis of muscle tissue were elevated. The ACC phosphorylation was increased in muscle and liver. This study provides a novel allosteric AMPK activator for functional study in vitro and in vivo and demonstrates that AMPK allosteric activators could be a promising therapeutic approach for type 2 diabetes mellitus and metabolic syndrome. Heterogeneousreactions on mineral aerosols remain an importantsubject in atmospheric chemistry because of their role in alteringthe properties of particles and the budget of trace gases. Yet, therole of coadsorption of trace gases onto mineral aerosols and potentialsynergistic effects are largely uncertain, especially synergisticeffects between inorganic and organic gas-phase pollutants. In thisstudy, synergistic effects between HCOOH and SO2were investigatedfor the first time using in situ diffuse-reflectance infrared Fouriertransform spectroscopy (DRIFTS). It was found that the heterogeneousreaction of HCOOH is hindered significantly by coexisting SO2. The total amount of formate decreased, whereas the total amountof sulfate was not affected during coadsorption on the surface ofα-Fe2O3. Futhermore, part of the formateon the surface was catalytically decomposed to CO2by α-Fe2O3with the help of SO2. Despite increased awareness of the role played by heterogeneousreactions of formic acid on mineral aerosol, the experimental determinationof how these atmospheric reaction rates vary with temperature remaina crucially important part of atmosphere science. Here we report thefirst measurement of heterogeneous uptake of formic acid on α-Al2O3as a function of temperature (T= 240–298 K) at ambient pressure using diffuse reflectanceinfrared Fourier transform spectroscopy (DRIFTS). From the analysisof the spectral features, crystalline HCOOH was identified at lowtemperature besides common product (formate ions) on the surface.It was also interesting to find that crystalline HCOOH can continueto react with α-Al2O3. The reaction mechanismsat both room and low temperature were discussed. Furthermore, thereactive uptake coefficients were acquired and found to increase withdecreasing temperature. Abstract: Two novel adsorbents to remove excess arsenate and arsenite in the drinking water were prepared for the first time by grafting monoamine and diamine, respectively, and then coordinating Fe3+ on silica gel that was obtained using sol–gel method with two-step acid-base catalysis. It was found that both adsorbents had mesoporous structure, large specific surface, and high amino and iron content according to N2 adsorption isotherms, FTIR, XPS, and NMR analysis. The removal ability and adsorption rate of the adsorbents were very high for both As(V) and As(III). Langmuir and Freundlich models were used to fit the adsorption isotherm and investigate the adsorption mechanism. The effects of chloride and sulfate anion on the removal of arsenate and arsenite for the two adsorbents were also studied. [Copyright &y& Elsevier]

Background: Painless gastroscopy is a widely used diagnostic and therapeutic technology in clinical practice. Propofol combined with opioids is a common drug for painless endoscopic sedation and anaesthesia. In clinical work, adverse drug reactions of anaesthesia schemes are often one of the important areas of concern for doctors and patients. With the increase in propofol dosage, the risk of serious adverse drug reactions, such as respiratory depression and hypotension, increases significantly; the use of opioids often causes gastrointestinal reactions in patients after examination, such as nausea, vomiting, delayed recovery of gastrointestinal function and other complications, which seriously affect their quality of life., Aim: To observe the effect of wrist-ankle acupuncture therapy on the anaesthesia regimen and anaesthesia-related complications during and after painless gastroscopy examination., Methods: Two hundred patients were selected and randomly divided into a treatment group ( n = 100) and a control group ( n = 100). Both groups were routinely anaesthetized with the nalbuphine and propofol regimen, gastroscopy began after the patient lost consciousness, and given supportive treatment and vital sign monitoring. If the patient interrupted the surgery due to intraoperative torsion, intravenous propofol was used to relieve his or her discomfort. The treatment group received wrist-ankle acupuncture on this basis., Results: The general data before treatment, American Society of Anesthesiologist (ASA) grade and operation time between the two groups was no significant difference. The Wakeup time, and the Self-ambulation time in the treatment group was significantly faster than that in the control group ( P < 0.05). The total dose of propofol in the treatment group was 109 ± 8.17 mg, significantly lower than that in the control group ( P < 0.05). The incidence of respiratory depression and hypotension was not significantly different, but the incidence of hiccups was significantly lower than that in the control group ( P < 0.05). After the examination, the incidence of nausea, vomiting, abdominal distension, and abdominal pain was 11%, 8%, 6%, and 5%, respectively, which was significantly lower than that in the control group ( P < 0.05). SH-SY5Y cell line treated with 6-hydroxydopamine (6-OHDA) is a classical Parkinson's disease (PD). In the present study, synchrotron-based Fourier transform infrared (FTIR) microspectroscopy was used to analyze the biochemical composition of SH-SY5Y cell line treated with 6-OHDA. The detailed spectral analyses show the significant changes in cellular compositions such as lipids, proteins and nucleic acids in SH-SY5Y cells treated with 6-OHDA compared to control SH-SY5Y cells. As a result, the unsaturation levels of phospholipids decrease in SH-SY5Y cells treated with 6-OHDA compared to control cells, the analysis of protein secondary structure shows the significantly higher ratio of beta-sheet in PD cells compared to that of control cells, and the content of nuclear acid is highly decreased compared to that of control cells, suggesting that 6-OHDA induces the serious oxidative damage in SH-SY5Y cells. These findings suggest that SR-FTIR is an effective and precise technical tool to probe the biochemical changes of cells and then evaluate the pathological damage in cells.

Objective: To observe the clinical effect and mechanism of migraine treated with pine needle moxibustion., Methods: Forty-three patients were randomly divided into 2 groups of pine needle moxibustion group (group A, 21 cases) and medicated thread moxibustion of Zhuang medicine group (group B, 22 cases). Same acu-points were selected as the main points for both groups, namely Fengchi (GB 20), Baihui (GV 20), Taiyang (EX-HN 5), Shuaigu (GB 8), Cuanzhu (BL 2), Sizhukong (TE 23), Yanglingquan (GB 34), Waiguan (TE 5) and Ashi points. Pine needle moxibustion was applied for the group A. Processed lighted pine needles were banded together with one even head to do moxibustion on acupoints or pain points. While, lighted medicated thread was adopted for the group B to do moxibustion. Therapeutic effects, the content of high-sensitivity C-reactive protein (hs-CRP), Mg2+ in the serum and the degree of pain of the two groups were compared., Results: (1) In the group A, the complete remission rate was 47.6% (10/21) and the effective rate was 90.5% (19/21). In the group B, the complete remission rate was 13.6% (3/22) and the effective rate was 50.0% (11/22). The two rates in the group A were superior to those in the group B (both P < 0.01); (2) In the group A, the content of hs-CRP was decreased from (4.29 +/- 0.98) mg/L to (2.11 +/- 0.87) mg/L, and Mg2+ was increased from (0.67 +/- 0.28) mmol/L to (1.07 +/- 0.16) mmol/L. In the group B, the contents of the above mentioned indices were respectively decreased from (4.30 +/- 1.07) mg/L to (3.38 +/- 1.15) mg/L and increased from (0.68 +/- 0.21) mmol/L to (0.88 +/- 0.25) mmol/L. There were significant differences between the two groups after treatment (both P < 0.05). (3) The degree of pain in the group A after treatment was much lower than that in the group B (P < 0.05)., Conclusion: The clinical effect of migraine treated with pine needle moxibustion is better than that with the medicated thread moxibustion of Zhuang medicine. And it has obvious regulation effect on the levels of hs-CRP and Mg2+ in the serum of patients.

Multiphase acid-catalyzed oxidation by hydrogen peroxide has been suggested to be a potential route to secondary organic aerosol (SOA) formation from isoprene and its gas-phase oxidation products, but the kinetics and chemical mechanism remain largely uncertain. Here we report the first measurement of uptake of methacrolein into aqueous solutions of sulfuric acid and hydrogen peroxide in the temperature range of 253-293 K. The steady-state uptake coefficients were acquired and increased quickly with increasing sulfuric acid concentration and decreasing temperature. Propyne, acetone, and 2,3-dihydroxymethacrylic acid were suggested as the products. The chemical mechanism is proposed to be the oxidation of carbonyl group and C═C double bonds by peroxide hydrogen in acidic environment, which could explain the large content of polyhydroxyl compounds in atmospheric fine particles. These results indicate that multiphase acid-catalyzed oxidation of methacrolein by hydrogen peroxide can contribute to SOA mass in the atmosphere, especially in the upper troposphere.

Synchrotron radiation based-Fourier transform infrared microspectroscopy (SR-FTIR) was used to preliminarily investigate the biochemical composition of the hippocampal neurons for 6-hydroxydopamine-lesioned rats and normal rats. Spectral analysis showed that in PD samples, the CH2 asymmetric and symmetric vibrational absorption of integral area at 2 924 and 2 850 cm(-1) and the intensity of C=O vibrational absorption at 1 736 cm(-1) (assigned to the lipid functional group) increase compared to normal samples, which indicate that lipid content increased in PD sample; the PO2 asymmetric and symmetric vibrational absorption decrease compared to normal samples (assigned to the nucleic acid functional group; However no clear difference of the vibrational fingerprinting of protein between PD and normal samples was noticed. The present results suggest that the changes in biochemical composition in hippocampal neurons in PD rats probed by synchrotron radiation based-FTIR may contribute to the elucidation of PD pathology.

The aromatic ring of the benzyl group is almost perpendicular to the N-heterocyclic ring that is directly connected to it [dihedral angles = 83.2 (2) and 77.3 (3)°].

In the title compound, C(17)H(14)N(2)O, the dihedral angle between the benzene ring and naphthalene ring system is 11.0 (3)°. The azo group adopts an anti configuration and an intra-molecular N-H⋯O hydrogen bond exists. Mol-ecules are packed by π-π inter-actions between adjacent mol-ecule (closest approach between centroids of benzene and naphthalene rings of 3.501 Å).

Objective: To evaluate the effect of elemene on the survival of normal human umbilical endothelial cells (HUVECs) and the protection of elemene on injured HUVECs induced by hydrogen peroxide (H2O2)., Methods: Normal HUVECs were treated with elemene 1-100 microg/ml for 24-72 hours, the survival rate of HUVECs was determined by tetrazolium assay (MTT). To evaluate the protective effect of elemene on HUVECs from H2O2 injury, HUVECs were injured by 1 mmol/L H2O2 and then different final concentrations of elemene were added before the injury. After culturing 1 hour, then detecting the index of MDA, T-AOC, SOD, CAT, GSH-Px and NO., Results: Elemene could inhabit the proliferation of VEC and it presented dose-dependent, while on the side of anti-oxidization injury, it also presented dose-dependent. MDA content and the effect of H2O2 to antioxidase activity were decreased, NO content in cell was increased, and the amount of apoptosis was reduced., Conclusion: Elemene has dual effects on the survival rate of normal HUVECs in vitro, which is related to the concentration and the duration of drug exposure. Elemene has protective effects on the injured endothelial cells injured by oxidization through the function of anti-lipid oxidization.

The title compound, [Cd(NO(3))(2)(C(9)H(12)N(4))(2)](n), has a one-dimensional double-bridged chain polymer structure with a 16-membered macrometallacyclic tetragonal structural motif. The Cd(II) ion occupies a crystallographic inversion centre and is coordinated by four equatorial N atoms from four distinct bis(2-methylimidazol-1-yl)methane ligands and two apical nitrate O atoms to form a slightly distorted octahedral coordination geometry.

Objective: To observe the photoprotective effect and possible mechanisms of resveratrol for ultraviolet A (UVA) irradiated HaCaT cells., Methods: HaCaT cells under UVA irradiation with 5J/cm(2) were interfered with 0.01 mmol/L and 0.1 mmol/L resveratrol. The testing objects were divided into a control and a UVA irradiation group, and then we detected the proliferation capacity with methylthiazdyl tetrazolium (MTT) and superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activity, content of maleic dialdehyde (MDA) with hydroxylamine, colorimetric, thiobarbituric acid (TBA) methods. The ultrastructure was observed under electron microscope., Results: Resveratrol could enhance the proliferation activity, SOD, GSH-Px activity of HaCaT cells under UVA irradiation, decrease the content of MDA in dose-dependent manner (P<0.05). The electron microscope revealed that resveratrol could relieve the injury of HaCaT cells' ultrastructure., Conclusion: Resveratrol can relieve the inhibition to HaCaT cell proliferation,injury of their ultrastructure and oxidation by UVA irradiation. The protection is dose-dependent. That resveratrol raises the oxidase activity and clears the oxyradical may account for these results.
