Abstract

Oxidative decolonization of indigo carmine by bipolar pulsed DBD in the water-air mixture was studied using the dynamic UV-Vis spectrum of the solution. Effects of various parameters such as gas flow rate, solution conductivity, pulse repetitive rate and etc., on color removal efficiency of dying wastewater were investigated. Concentrations of gas phase O/sub 3/ and aqueous phase H/sub 2/O/sub 2/ under various conditions were measured. Experimental results show that air bubbling facilitates the breakdown of water and promotes generation of chemically active species. Color removal efficiency of IC solution can be greatly improved by the air aeration under various solution conductivities. Decolonization efficiency increases with the increase of the gas flow rate, and decreases with the increase of the initial solution conductivity. A higher pulse repetitive rate and a larger pulse capacitor Cp are favorable for the decolonization process. Ozone and hydrogen peroxide formed decreases with the increase of initial solution conductivity. In addition, preliminary analysis of the decolonization mechanisms is given.

21. Anti-VEGF agents confer survival advantages to tumor-bearing mice by improving cancer-associated systemic syndrome.

Abstract

The underlying mechanism by which anti-VEGF agents prolong cancer patient survival is poorly understood. We show that in a mouse tumor model, VEGF systemically impairs functions of multiple organs including those in the hematopoietic and endocrine systems, leading to early death. Anti-VEGF antibody, bevacizumab, and anti-VEGF receptor 2 (VEGFR-2), but not anti-VEGFR-1, reversed VEGF-induced cancer- associated systemic syndrome (CASS) and prevented death in tumor- bearing mice. Surprisingly, VEGFR2 blockage improved survival by rescuing mice from CASS without significantly compromising tumor growth, suggesting that "off-tumor" VEGF targets are more sensitive than the tumor vasculature to anti-VEGF drugs. Similarly, VEGF-induced CASS occurred in a spontaneous breast cancer mouse model overexpressing neu. Clinically, VEGF expression and CASS severity positively correlated in various human cancers. These findings define novel therapeutic targets of anti-VEGF agents and provide mechanistic insights into the action of this new class of clinically available anti-VEGF cancer drugs [ABSTRACT FROM AUTHOR]

Abstract

Pepsin digestion prior to mass analysis increases the spatial resolution of hydrogen exchange mass spectrometry experiments. Online digestion with immobilized pepsin is advantageous for several reasons including better digestion efficiency. We have found that certain immobilized pepsin columns cause substantial deuterium back-exchange, rendering the data unusable. When pepsin immobilized on a POROS support was used for online digestion, back-exchange was within the expected range and was similar to the back-exchange of deuterated peptides produced by in-solution pepsin digestion. However, when pepsin immobilized onto selected polystyrene-divinylbenzene supports was used for online digestion with the same system, deuterium loss was extremely high. 28. Purified Haemoglobin Preparations in the Evaluation of HbA1cDetermination by Ion Exchange Chromatography

Abstract

The use of ion exchange resins for the estimation of HbA1cin clinical samples rests on the assumption that HbA1cis effectively and efficiently separated from other N-terminally modified haemoglobins and from HbAo. To test this assumption, we applied highly purified preparations of HbA1a+1b, HbA1cand HbAoto ion exchange minicolumns, using conditions of application simulating actual blood samples and the first and second elution buffers provided by the manufacturer. The authenticity and purity of the applied haemoglobin preparations were documented by high performance liquid chromatography, gel electrophoresis and carbohydrate content. About 40% of the applied HbA1a+1beluted in the first fraction; 45% eluted in the second fraction, and 10% to 15% required 1 mol/L NaCl to elute from the column. Of the applied HbA1c, 65–80% eluted where expected in the second fraction, about 20% required 1 mol/L NaCl to elute from the column, and the remainder eluted with HbA1a+1b. Some 3–6% of pure HbAoapplied to minicolumns emerged in the second fraction, with the remainder eluting as expected after making the buffer 1 mol/L in NaCl. The results indicate that the fraction eluting from ion exchange minicolumn chromatography that is designated ‘HbA1c’ contains HbA1a+1b, and that a substantial portion of the HbA1cin an applied sample does not elute in this fraction.
