o-Cresol shows high toxicity and stability. To explore a better degradation method of o-cresol, the degradation of o-cresol by three kinds of microalgae (Selenastrum capricornutum, Scenedesmus obliquus, and Microcystis aeruginosa) was researched in this paper. The growth status and degradation rate were used to reflect the tolerance and degradation effect of microalgae. The effects of the medium’s initial pH, microalgal density, and different exogenous pollutants on the degradation of o-cresol by Selenastrum capricornutum were investigated. The results showed that Selenastrum capricornutum had the best degradation effect on o-cresol. microalgal density increased after adaptation to different concentrations of o-cresol for some time. At pH 7.0 as the initial condition, the microalgal exhibited the best results of degradation. When the microalgal density OD680 was 0.20, o-cresol was the first to be completely degraded within 5 days. At higher initial concentrations of o-cresol, the microalgae preferentially degraded glucose to promote the growth of the microalgae under mixotrophic cultivation. Selenastrum capricornutum could degrade phenol and o-cresol at the same time, and the degradation was completed within 8 days when the initial concentration of o-cresol and phenol were 100 and 120 mg/L. It was proven that the degradation of o-cresol by Selenastrum capricornutum is feasible under suitable conditions.

Lymphoma relapse is very common in clinical work, but lineage switch at relapse is rare. Although some cases have reported acute lymphocytic leukemia (ALL) switch to acute myeloid leukemia (AML) or myeloid sarcoma upon relapse, phenotype switch seldom occurs in other types of lymphoma. Here we report six cases with lineage switch from lymphoma to myeloid neoplasms. In our cohort, three cases were mantle cell lymphoma (MCL), and the other three cases were T-cell lymphoblastic lymphoma (T-LBL), B-cell lymphoblastic lymphoma (B-LBL), and diffuse large B-cell lymphoma (DLBCL) at the initial diagnosis. When linage switch occurred, most cases were AML M5 phenotypes, and only one case was myelodysplastic syndrome (MDS) phenotype. 11q23/mixed-lineage leukemia (MLL) rearrangement was negative in all cases. Although intensive therapy and stem cell transplantation have been applied in most cases, the poor outcome cannot be reversed. Therefore, we found that lineage switch could occur not only from ALL to AML or vice versa, but also from MCL or DLBCL to AML. Moreover, the incidence of MLL rearrangement in lineage switch is lower in adult hematologic malignancies as compared with pediatric patients.

Understanding the acid coagulation of milk is the core of producing different fermented dairy products. The formation of the gelled structure includes the decreased stability of casein micelles, aggregation, and the gradual development of the bonding between proteins during acidification and cold storage. The coagulation behavior of casein micelles and the physical properties of the final gels can be modified by processing techniques. Exopolysaccharides (EPS) produced by starter culture during fermentation also contribute significantly to the microstructure and texture of acid gels. This chapter discusses the mechanisms of acid-induced gelation of milk based on the modified nanocluster model of casein micelles. The recent findings of heating, high-pressure treatment, ultrasonication, and enzymatic modification on the acid gelation behavior of milk are described. The influence of different ingredients such as polysaccharides (endogenous and exogenous) and phenolic compounds on the physical properties of acid gels are also summarized.

SEMA6A is a multifunctional transmembrane semaphorin protein that participates in various cellular processes, including axon guidance, cell migration, and cancer progression. However, the role of SEMA6A in clear cell renal cell carcinoma (ccRCC) is unclear. Based on high-throughput sequencing data, here we report that SEMA6A is a novel target gene of the VHL-HIF-2α axis and overexpressed in ccRCC. Chromatin immunoprecipitation and reporter assays revealed that HIF-2α directly activated SEMA6A transcription in hypoxic ccRCC cells. Wnt/β-catenin pathway activation is correlated with the expression of SEMA6A in ccRCC; the latter physically interacted with SEC62 and promoted ccRCC progression through SEC62-dependent β-catenin stabilization and activation. Depletion of SEMA6A impaired HIF-2α-induced Wnt/β-catenin pathway activation and led to defective ccRCC cell proliferation both in vitro and in vivo. SEMA6A overexpression promoted the malignant phenotypes of ccRCC, which was reversed by SEC62 depletion. Collectively, this study revealed a potential role for VHL-HIF-2α-SEMA6A-SEC62 axis in the activation of Wnt/β-catenin pathway. Thus, SEMA6A may act as a potential therapeutic target, especially in VHL-deficient ccRCC.

Background: Recent studies have suggested association between the ABO blood group and inflammation, which was a crucial pathological process of primary knee osteoarthritis. The aim of this study was to investigate the association between the ABO blood group and primary knee osteoarthritis ​and the severity of primary knee osteoarthritis evaluated by the Kellgren/Lawrence score, as well as the histopathologic association in a subgroup of patients. Methods: We performed a retrospective review of patients with primary knee osteoarthritis that served as the case group ​and a random sampling of healthy blood donors that served as the control group. The severity of knee osteoarthritis at the first outpatient visit was evaluated by the Kellgren/Lawrence scoring system. Further study was performed to investigate the expression of blood group antigens in synovial tissue of the knee in both cases and controls. Results: A total of 1126 cases and 30299 controls were involved. The proportion of AB blood group was higher in the case group than in the control group (9.7% vs. 7.8%), and logistic regression revealed that the AB blood group was a risk factor of primary knee osteoarthritis (P ​= ​0.025 and 0.048 for univariate and multivariate analysis, respectively), independent of age (P ​= ​0.973) and sex (P ​= ​0.520). Patients of the blood group AB had a higher Kellgren/Lawrence score (P ​= ​0.017). The immunohistochemical study indicated association between LeY antigen and primary knee osteoarthritis (P ​= ​0.029). Conclusions: This study suggested that the blood group AB was associated with primary knee osteoarthritis, as well as its radiological severity. Further study indicated that LeY antigen, which was related to the blood group, was associated with primary knee osteoarthritis. Translational potential of this article: This study revealed that blood group AB and LeY antigen was associated with primary knee osteoarthritis, which shed new light on the nature of osteoarthritis, and the development of novel therapy for osteoarthritis. In this study, the foamability and foam stability of nitrogen-infused cold brew coffee, as affected by coffee variety (Arabica and Robusta), degree of roast (light, medium, dark), brewing temperature (4, 20, 35 °C), brew ratio (1:5-1:15 w/w; coffee/water), ground particle size (712, 647 and 437 μm volume mean diameter) and beverage temperature (4, 20 and 35 °C), were investigated.Dynamic surface tension of cold brew, as determined from bubble tensiometry, decreased from 65-70 mN mThe foamability and foam stability of cold brew coffee are significantly affected by coffee variety, degree of roast, brewing temperature, ground particle size, and beverage temperatures. Forward osmosis membranes have a wide range of applications in the field of water treatment. However, the application of seawater desalination is restricted, so the research of forward osmosis membranes for seawater desalination poses new challenges. In this study, zeolitic imidazolate framework-8 (ZIF-8) was synthesized by a mechanical stirring method, and its crystal structure, surface morphology, functional group characteristics, thermochemical stability, pore size distribution and specific surface area were analyzed. The cellulose acetate (CA)/ZIF-8 mixed matrix forward osmosis membrane was prepared by using the synthesized ZIF-8 as a modified additive. The effects of the additive ZIF-8 content, coagulation bath temperature, mixing temperature and heat treatment temperature on the properties of the CA/ZIF-8 forward osmosis membrane were systematically studied, and the causes were analyzed to determine the best membrane preparation parameters. The structure of the CA membrane and CA/ZIF-8 mixed matrix forward osmosis membranes prepared under the optimal conditions were characterized by Fourier Transform infrared spectroscopy (FTIR), Scanning electron microscopy (SEM), contact angle and Atomic force microscope (AFM). Finally, the properties of the HTI membrane (Membrane manufactured by Hydration Technology Innovations Inc.), CA forward osmosis membrane and CA/ZIF-8 mixed matrix forward osmosis membrane were compared under laboratory conditions. For the CA membrane, the water flux and reverse salt flux reached 48.85 L·m−2·h−1 and 3.4 g·m−2·h−1, respectively. The reverse salt flux and water flux of the CA/ZIF-8 membrane are 2.84 g·m−2·h−1 and 50.14 L·m−2·h−1, respectively. ZIF-8 has a promising application in seawater desalination.

Abstract Zein-based materials were used to remove Trypan blue from water under flow conditions and in batch tests. In flow tests, zein dissolved at pH = 13 was injected in sand columns and subsequently coagulated with CaCl2, to create an adsorbent filter which removed over 99% of Trypan blue. Batch tests were conducted using zein powder, zein dissolved at pH = 13 and coagulated with CaCl2, Fe2Cl3 or citric acid, and zein dissolved in ethanol and then coagulated with water. The highest Trypan blue removal was achieved with zein powder (4000 mg Trypan blue/kg sorbent, as determined through spectrophotometry), followed by zein coagulated with Fe2Cl3 (500 mg Trypan blue/kg sorbent) and with other salts (140 mg Trypan blue/kg sorbent). Differences in the sorption efficiency are attributed to differences in the surface area. The sorption isotherm of Trypan blue onto zein-based sorbents was a Type II isotherm, suggesting physisorption. Desorption of Trypan blue was limited when zein-based coagulated sorbents were immersed in pure water. Trypan blue could be degraded by free laccase in water, as determined through spectrophotometry and electrospray ionization mass spectroscopy (ESI-MS). Trypan blue could also be degraded by laccase when zein-based laccase-containing sorbents were prepared at pH = 10, using Fe2Cl3 as coagulant. Graphic abstract

Background Disparities in multiple myeloma (MM) prognosis based on sociodemographic factors may exist. We investigated whether education level at diagnosis influenced Chinese MM patient outcomes. Methods We performed a multicenter retrospective analysis of data from 773 MM patients across 9 centers in China from 2006 to 2019. Sociodemographic and clinical factors at diagnosis and treatment regimens were recorded, and univariate and multivariate analyses were performed. Results Overall, 69.2% of patients had low education levels. Patients with low education levels differed from those with high education levels in that they were more likely to be older, and a higher proportion lived in rural areas, were unemployed, had lower annual incomes and lacked insurance. Additionally, compared to patients with high education levels, patients with low education levels had a higher proportion of international staging system (ISS) stage III classification and elevated lactate dehydrogenase (LDH) levels and underwent transplantation less often. Patients with high education levels had a median progression-free survival (PFS) of 67.50 (95% confidence interval (CI): 51.66–83.39) months, which was better than that of patients with low education levels (30.60 months, 95% CI: 27.38–33.82, p p p p Conclusions Low education levels may independently predict poor survival in MM patients in China.

As tyrosine kinase inhibitors (e.g., Imatinib, IM) fail to induce long-term response in some chronic myeloid leukemia (CML), novel therapies targeting leukemia-dysregulated pathways are necessary. Nuclear-cytoplasmic trafficking of proteins play a key role in the development of leukemia and drug resistance. KPT-330 (Selinexor), an inhibitor of chromosome region maintenance 1 (CRM1, nuclear receptor exportin 1, XPO1), demonstrated activities against a few hematological malignancies. We examined the anti-leukemic efficacy of KPT-330 in IM-resistant CML. Cell viability was examined by MTS assay. Apoptosis and cell cycle were assessed by flow cytometry. CRM1 mRNA was detected by PCR. Expression of CRM1 protein and its cargo proteins were determined by western blot or immunofluorescent staining. Furthermore, we engrafted nude mice subcutaneously with IM-resistant CML K562G. Mice were treated with IM, KPT-330 alone or in combination. Expression of CRM1 in CML were markedly higher than control. KPT-330 inhibited proliferation, induced cell cycle arrest and apoptosis of K562 and K562G. IC50 of IM on K562G was reduced by KPT-330. Mechanistically, KPT-330 inhibited CRM1 and increased the nuclear/cytoplasm ratio of BCR-ABL and P27. p-AKT was downregulated while p-STAT1 and caspase-3 were upregulated. Furthermore, KPT-330 showed anti-leukemic effect in primary IM-resistant CML with T315I mutation in CRM1-dependent manner. In K562G xenograft mice model, KPT-330 inhibited tumor growth and sensitized K562G to IM in vivo. To conclude, KPT-330 showed anti-leukemic activity and sensitized CML to IM in CRM1-dependent manner in vitro and in vivo. KPT-330 represents an alternative therapy for IM-refractory CML, warranting further investigation of CRM1 as therapeutic target.

Forward osmosis (FO) membrane is a promising membrane technique. However, its application in desalination is limited. Thus, we synthesized MIL-53(Fe)@γ-Al2O3 via solvent-thermal method to fabricate MIL-53(Fe)@γ-Al2O3/cellulose acetate (CA) composite membranes of high desalination performance. MIL-53(Fe)@γ-Al2O3 and MIL-53(Fe) particles were studied using XRD, FITR, SEM, N2 adsorption-desorption isotherms, and AFM. The structural features of MIL-53(Fe)@γ-Al2O3/CA membrane and MIL-53(Fe)/CA membrane were analyzed using SEM, AFM and water contact angle. The reverse salt flux and water flux of the MIL-53(Fe)@γ-Al2O3/CA membrane reached 1.78 g m-2 h-1 and 37.1 L m-2 h-1, respectively, when the deionized water and 1 M NaCl solution were the feed solution and traction solution, respectively. Compared to the MIL-53(Fe)/CA membrane, the MIL-53(Fe)@γ-Al2O3/CA membrane demonstrated better perviousness and selectivity properties of the membrane. This study shows a potential advantage of MOF composites as additives for improvement in the desalination performance of forward osmosis membranes.

Two CO2 formation pathways (chlorogenic acid (CGA) degradation and Maillard reaction) during coffee roasting were investigated. CGA is shown not a major contributor to CO2 formation, as heating of this compound under typical roasting conditions did not release a large quantity of CO2. However, heating of a CGA moiety, caffeic acid, resulted in high yield of CO2 (>98%), suggesting that CGA hydrolysis could be the rate limiting step for CO2 formation from CGA. A large amount of CO2 was detected from glycine-sucrose model system under coffee roasting conditions, implying the importance of Maillard reactions in CO2 formation. Further studies on the heating of various components isolated from green coffee beans showed that CO2 was generated from various green coffee components, including water insoluble proteins and polysaccharides. Around 50% of CO2 was formed from thermal reactions of lower molecular weight compounds that represent ∼25% by weight in green coffee.

As one of the most consumed beverages in the world, coffee plays many major socioeconomical roles in various regions. Because of the wide coffee varieties available in the marketplaces, and the substantial price gaps between them (e.g., Arabica versus Robusta; speciality versus commodity coffees), coffees are susceptible to intentional or accidental adulteration. Therefore, there is a sustaining interest from the producers and regulatory agents to develop protocols to detect fraudulent practices. In general, strategies to authenticate coffee are based on targeted chemical profile analyses to determine specific markers of adulterants, or nontargeted analyses based on the “fingerprinting” concept. This paper reviews the literature related to chemometric approaches to discriminate coffees based on nuclear magnetic resonance spectroscopy, chromatography, infrared/Raman spectroscopy, and array sensors/indicators. In terms of chemical profiling, the paper focuses on the detection of diterpenes, homostachydrine, phenolic acids, carbohydrates, fatty acids, triacylglycerols, and deoxyribonucleic acid. Finally, the prospects of coffee authentication are discussed.

Artesunate (ART) is a sesquiterpene lactone isolated from the leafy portions of the Chinese herb Artemisia annua. Here, we evaluated the effect of ART on the prostate cancer (PCa) cell lines DU145 and LNCaP and explored its potential mechanisms. ART inhibited the viability and mobility of DU145 and LNCaP cells. Mechanistically, we found that UCA1, one of the most important lncRNAs in malignancies of the urinary system, may be a potential mediator contributing to the tumor suppressor function of ART. First, the UCA1 level was reduced significantly after being exposed to ART. In addition, UCA1 was up-regulated in prostate cancer tissues compared to hyperplastic prostatic tissues, and a higher UCA1 level predicted poor prognosis in PCa patients. Furthermore, reintroduction of UCA1 into PCa cells reversed the effect of ART on apoptosis and metastatic ability. Then we determined that the miR-184/Bcl-2 axis might be the downstream signaling pathway of UCA1 upon ART treatment. UCA1 binds to miR-184 through its seed sequences and may function as a sponge for miR-184.

The spacer geometries have a crucial effect on the hydrodynamics of spacer-filled channels in electrodialysis (ED) process, which are beneficial to mass transfer enhancement. Herein, a three-dimensional, isothermal, and steady flow model was developed by computational fluid dynamics (CFD) to illuminate the impacts of the spacer geometrical configurations on hydrodynamic behaviors in the spacer-filled channel for ED applications. Simultaneously, the finite element analysis method was employed to investigate the effects of spacer geometries on the velocity and pressure profiles. Furthermore, the normalized expenditure (NEXP) was introduced to acquire one comparable cost estimation index, which determined the optimal spacer geometries subserving the efficiency and economic competitiveness of ED process. Theoretical and techno-economic analysis validated that the optimal mesh parameters within the range of the test parameters were a cylindrical mesh, a filament diameter of 0.8 mm, a filament interval of 1.0 mm, a flow attack angle of 30°. Therefore, this work can provide a feasible strategy to visualize and quantitate the hydrodynamics in the spacer-filled channel and insight into the optimization of ED stack, potentially reducing the expenditure related to materials usage, energy consumption and test time.

From its legendary discovery in Abyssinia (today Ethiopia) to becoming one of the most consumed beverages in the world, coffee has captivated the enthusiasts for centuries due to its unique aroma and taste, as well as its effects as a stimulant in enhancing mental performance (e.g., alertness, concentration, attention). This article provides a brief overview on the production and processing of coffee, focusing on the Coffea arabica and Coffea canephora var. Robusta, also known as Arabica and Robusta, respectively. Differences in chemical compositions (e.g., lipid, sucrose, trigonelline, diterpenes, caffeine, chlorogenic acids) of bean variety contribute to desirable/undesirable sensory attributes, as well as the health implications of the final brew products. Roasting of green beans, which is typically carried out at 170–230 °C for 10–15 min, causes the degradations of polysaccharides, sugars, amino acids, chlorogenic acids, and so on. Concomitantly, a myriad of aroma volatiles and complex condensed products are formed, mainly due to Maillard reaction, Strecker degradation and pyrolytic reactions. The effects of roast time–temperature profiles on a number of key physicochemical phenomena are discussed, including changes in microstructural, formation of aroma species, development of color, and generation of CO2 during roasting. Optimal storage conditions and packaging are important in delaying product staling and to mitigate CO2 degassing issues. These aspects, along with other factors that affect the shelf-life of coffee, are discussed. Finally, a brief literature review on the health implications of coffee consumption is presented, highlighting the importance of several bioactive components (e.g., caffeine, chlorogenic acids, melanoidins, trigonelline, acrylamide, and diterpenes).

BACKGROUND Many studies have demonstrated that the long non-coding RNA (lncRNA), NEAT1_1, plays critical roles in various human tumor entities and is related to the survival of patients with malignancies. However, the role of NEAT1_1 in diffuse large B cell lymphoma (DLBCL) remains unclear. The aim of this study was to investigate the role of NEAT1_1 in DLBCL. METHODS The expression of NEAT1_1 was evaluated in paraffin-embedded tissues from 64 DLBCL patients and 15 lymphnoditis patients using the ISH method. The correlations between the expression levels of NEAT1_1 and clinical-pathological features and patients' survival were also analyzed. After knocking down NEAT1_1 using shRNA in the DLBCL cell lines OCI-Ly1 and SUDHL-4, cell viability, apoptosis and migration were assessed by performing CCK8 assays, annexin V-FITC/PI double staining assays and migration filter assays, respectively. RESULTS NEAT1_1 expression was increased in DLBCL tissue compared to lymphnoditis tissue samples (P< 0.001). The NEAT1_1 level was positively related to stage (P= 0.031), IPI (P= 0.017), extranodal site involvement (P= 0.042) and drug response (P= 0.040). Kaplan-Meier analysis showed that high expression levels of NEAT1_1 were correlated with a poor prognosis in DLBCL patients. After shRNA-NEAT1_1 was transfected into OCI-Ly1 and SUDHL-4 for 24 h, the NEAT1_1 level decreased to approximately one-third the level of the control. Moreover, the viability and migration ability of the DLBCL cell lines were significantly suppressed. shRNA-NEAT1_1 induced apoptosis in both DLBCL cell lines. CONCLUSIONS Our results showed that NEAT1_1 plays an oncogenic role in DLBCL. NEAT1_1 expression may serve as a predictive marker for DLBCL patients.

Hexavalent chromium is one of the main heavy metal pollutants. As the environmental legislation becomes increasingly strict, seeking new technology to treat wastewater containing hexavalent chromium is becoming more and more important. In this research, a novel modified ultrafiltration membrane that could be applied to adsorb and purify water containing hexavalent chromium, was prepared by polyvinylidene fluoride (PVDF) blending with 2-aminobenzothiazole via phase inversion. The membrane performance was characterized by evaluation of the instrument of membrane performance, infrared spectroscopy (FTIR), scanning electron microscope (SEM), and water contact angle measurements. The results showed that the pure water flux of the PVDF/2-aminobenzothiazole modified ultrafiltration membrane was 231.27 L/m2·h, the contact angle was 76.1°, and the adsorption capacity of chromium ion was 157.75 µg/cm2. The PVDF/2-aminobenzothiazole modified ultrafiltration membrane presented better adsorption abilities for chromium ion than that of the traditional PVDF membrane.

The improved passive immune thrombocytopenia (ITP) mouse model has been extensively utilized for the study of ITP. However, how closely this model matches the human inflammation state and immune background is unclear. Our study aimed to explore the profile of Th cytokines and Th17/Treg cells in the model.We induced the ITP mouse model by dose-escalation injection of MWReg30. The serum levels of cytokines (IFN-γ, IL-2, IL-4, IL-10, IL-17A, and TGF-β1) were measured by enzyme-linked immunosorbent assay and the frequency of Th17 and Treg cells was measured by flow cytometry. The mRNA expression of Foxp3 and RORrt was measured by real-time PCR.The serum levels of cytokines IFN-γ, TGF-β1, IL-4, and IL-10 were significantly lower in ITP mice. The secretion of serum proinflammatory cytokines IL-2 and IL-17A and the percentage of Th17 cells showed no statistically significant increase. In ITP mice the frequency of Treg cells and mRNA expression of Foxp3 was significantly lower in splenocytes.Our data suggest that the improved passive ITP mouse model does not mimic the autoimmune inflammatory process of human ITP. Compared with human ITP, this model has a similar change in frequency of Treg cells, which may directly or indirectly result from antibody-mediated platelet destruction due to attenuated release of TGF-β.

CO2 is one of the major gases formed during coffee roasting, which has important implications on coffee's quality and packaging requirements. In this study, the residual CO2 content and CO2 degassing behavior of an Arabica coffee processed using a fluidized bed roaster, as affected by the roasting temperature–time conditions, were investigated. The results showed that positive correlations existed between the degree of roast (expressed as lightness value) and residual CO2, implying that lightness could be used as an indicator of initial CO2 content in roasted coffee. At the same degree of roast, coffee roasted with high-temperature–short-time process had significantly higher CO2 degassing rate than those with low-temperature–long-time process. Moreover, the CO2 releasing rate increased with the degree of roast. The degassing rate of CO2 in ground coffee was highly dependent on the grind size and roasting temperature, but less dependent on the degree of roast. The different degassing behaviors observed between roasted coffee samples were explained on the basis of chemical composition and microstructural differences.

This study describes the preparation and characterisation of MIL-53(Fe) and its effect on the performance of forward osmosis (FO) membranes. The cellulose acetate (CA)/MIL-53(Fe) hybrid membranes were fabricated using the method of phase inversion by dispersing MIL-53(Fe) in a CA casting solution. To improve the selectivity and permeability performance of FO membranes, the effects of MIL-53(Fe) content, casting solution temperature, coagulation bath temperature, and annealing temperature were studied. Results from scanning electron microscopy, membrane porosity, atomic force microscopy, and water contact angle tests of all the membranes show that the structural properties of the CA/MIL-53(Fe) hybrid membranes were optimised. The results of the FO performance tests, including the values of water flux and reverse salt flux, indicate improvement in the selectivity and permeability properties of the CA/MIL-53(Fe) hybrid membranes. Under the conditions of deionised water as the feed solution and 1 M NaCl solution as the draw solution, the water flux and reverse salt flux of the CA/MIL-53(Fe) hybrid membranes reached 34.9 L/(m2·h) and 2.02 g/(m2·h), respectively, in comparison with CA membranes. This study demonstrates that MIL-53(Fe) can improve the desalination performance and structural properties of FO membranes.

AZD6244, an ATP-uncompetitive inhibitor of mitogen-activated protein kinase 1/2 (MEK1/2), has shown activity in several malignant tumours. However, whether AZD6244 has a function in lymphoma cells is not known. We report that AZD6244 treatment represses the growth of Raji and MOLT4 cells by inducing apoptosis and G1-phase arrest. Using miRNAs array and quantitative RT-PCR, miR-92a was downregulated byAZD6244 treatment through the ERK1/2-AP1 signalling pathway. Overexpression of miR-92a abrogated AZD6244-induced apoptosis and G1-phase arrest, indicating that it is involved in the cytotoxicity of AZD6244 in lymphoma cells. A luciferase reporter assay showed that miR-92a directly targetsthe 3'-UTRs of Bim. Overexpression of miR-92a mimics downregulated Bim mRNA and protein expression level, indicating that miR-92a negatively regulates its expression at both levels. Silencing Bim decreases AZD6244-induced apoptosis and G1-phase arrest, suggesting that Bim contributes to the growth arrest. Thus, miR-92a mediates AZD6244-induced cytotoxicity of lymphoma cells by targeting Bim. Downregulation of miR-92a by AZD6244 is mediated by the ERK1/2-AP1 signalling pathway.
