Background Precision oncology pharmacotherapy relies on precise patient-specific alterations that impact drug responses. Due to rapid advances in clinical tumor sequencing, an urgent need exists for a clinical support tool that automatically interprets sequencing results based on a structured knowledge base of alteration events associated with clinical implications. Results Here, we introduced the Oncology Pharmacotherapy Decision Support System (OncoPDSS), a web server that systematically annotates the effects of alterations on drug responses. The platform integrates actionable evidence from several well-known resources, distills drug indications from anti-cancer drug labels, and extracts cancer clinical trial data from the ClinicalTrials.gov database. A therapy-centric classification strategy was used to identify potentially effective and non-effective pharmacotherapies from user-uploaded alterations of multi-omics based on integrative evidence. For each potentially effective therapy, clinical trials with faculty information were listed to help patients and their health care providers find the most suitable one. Conclusions OncoPDSS can serve as both an integrative knowledge base on cancer precision medicine, as well as a clinical decision support system for cancer researchers and clinical oncologists. It receives multi-omics alterations as input and interprets them into pharmacotherapy-centered information, thus helping clinicians to make clinical pharmacotherapy decisions. Sodium borohydride reduction in aprotic solvents has always been an unsolved problem. In this work a ion-pair-catalyzed sodium borohydride reduction in aprotic solvents was kinetically and mechanistically studied. Finally sodium borohydride reduction could be carried out in common aprotic solvents efficiently under mild conditions. A large quantity of water or methanol as the solvent or co-solvent has been avoided. The generation of stoichiometric or excess organic ammonium waste has also been avoided. The normal acidic work-up is not necessary and the catalyst was recycled.

Background: Recent studies have shown that vitiligo is a T-cell mediated autoimmune disease. Skin-homing cytotoxic T lymphocytes expressing cutaneous lymphocyte-associated antigen (CLA) have been suggested to be responsible for the destruction of melanocytes in vitiligo. An aberration in the suppressive function of regulatory T cells (Tregs) has been reported in vitiligo patients. However, whether the weakened suppressive ability of the Tregs contributes to hyper-activated skin homing CD8+CLA+ T cells remains to be determined. Objectives: To investigate the inhibition of circulating Tregs on the proliferation of autologous CD8+CLA+ T cells in non-segmental vitiligo patients. Methods: CD8+CLA+ T cells and Tregs were obtained from the peripheral blood of 13 non-segmental vitiligo patients and 7 controls. The proliferative responses of CD8+CLA+ T cells were assessed in the absence or presence of autologous Tregs, and the levels of Transforming Growth Factor β1(TGF-β1) and IL-10 in culture supernatants were detected by enzyme-linked immunosorbent assay. Results: The proliferative responses of circulating CD8+CLA+ T cells in the presence of Tregs were significantly higher in the active vitiligo than in the stable vitiligo and control groups. Tregs from active vitiligo patients exhibited a lower inhibitory effect on proliferation of CD8+CLA+ T cells. The levels of TGF-β1 produced by Tregs were significantly lower in active vitiligo than other groups and anti-TGF-β1 antibodies could abrogate the suppressive function of Tregs. Conclusions: The functional activity of Tregs is compromised in active vitiligo patients. TGF-β1 plays an important role in the autoimmune mechanism of the disease.

The paper studied characteristic of the morphology, substance and formation mechanism of Suo Duoxi deposit body in the Su Walong hydropower station reservoir area, focused on analysis of substance composition and formation at front area of deposit body (zoneII) and finally inferred the geological formation process and reached the formation mechanism . The results show that the block stones are main part of material of Suo Duoxi deposit and sandwiches gravel soils, but it is different from other four zones that the zone II takes gravel with less viscous as the skeleton and mixed up with uneven sizes and irregularly shape stones. The forming reason of deposit is the landslides at left bank of Jinsha River and rockfall of the steep mountain back of the deposit body on the right back with the effect of weathering, rainfall and glacial erosion and so on. Geological formation process of deposit determined the geomorphic features, but also changed paleocurrent of Jinsha River.

Blades are important components of aircraft engines. Free-form surface blade machining generally adopts sub-regional processing and cutting parameters vary greatly. Rational optimization of the cutting parameters can improve blade processing quality and efficiency in aerospace industry. To control the processing deformation of the blades, a method using artificial neural network and genetic algorithm is presented to optimize the cutting parameters. The method can reduce cutting parameter selection time and improve blade processing quality.

In this paper, linking with the basic principle of FCM (Fuzzy c-means clustering) algorithm, on the basis of theory research, a method of the cluster analysis of FCM is proposed. Firstly, the approximate optimal solution obtained by the improved FCM algorithm is taken as the original value of the FCM algorithm, then carrying on the local search to obtain the global optimal solution, the final segmentation result is achieved at last. The experiment results prove that in the view of the flame image segmentation, this method shows the good clustering performance and fast convergence rate, and has the widespread serviceability, so it is the practical method in image segmentation.

The asymmetric conjugate addition of α-substituted cyanoacetates to N-substituted maleimides has been developed. A number of cinchona alkaloids and amine thioureas were evaluated as catalysts. Takemoto’s catalyst was found to be the most efficient for the transformation. Chiral succinimides with two adjacent quaternary and tertiary stereogenic carbon centers were obtained in good yields, enantioselectivities, and diastereoselectivities. The products were converted to chiral γ-lactams conveniently without a loss in the enantioselectivity.

The article used the large span and heavy load cross-rail of some machine tools plant’s heavy-type numerical control milling planer for research object, by using the finite element software to build the model and force analysis. It also researched the force distribution and deformation condition of the large span motion girder under the actual working condition. It considered the slide carriage superposition effect to build the describing method and establishing process of the large span load curve by contacting analysis and restraining analysis, and got the knifepoint trajectory deformation. The research provided the sufficient theory basis and simulation method for designing improvement of cross-rail structure and got a perfect arch in advance.
