The medicinal plant Tirpitzia sinensis has been used by the Zhuang ethnic people in mountainous areas of Southwest China to stop bleeding, invigorate blood circulation, and treat inflammation and wounds. In order to further explore its traditional medicinal uses, the phytochemical constituents of this species were examined. Three new compounds, the lignan tirpitzin (1), the flavonoid tirpitzoside (2), and the furan-glycoside tirpitziol (3), along with five known compounds were isolated from the aerial part of T. sinensis for the first time. The structures of these compounds were elucidated by 1D and 2D NMR, LC/MS, IR spectrometric methods and compared with published data. The results of an in silico pharmacophore-based analysis showed potential targets of the new compounds, including ERBB2, IRAK4, LCK, JAK2, MAPK14, and MMP-12. These targets suggested that 1–3 may be involved with wound-healing and/or inflammation, leading to an in vitro assay of nitric oxide (NO) inhibition assays with lipopolysaccharide-induced BV-2 cells. All three new compounds displayed moderate NO inhibitory activity with the IC50 values of 14.97 ± 0.87, 26.63 ± 1.32, and 17.09 ± 2.3 μM, respectively.

Objective To investigate whether salidroside (SAL) has protective and anti-oxidative effects on astrocytes. Methods Firstly, SAL was extracted from the roots of Rhodiola rosea with 70% ethanol and butanol to obtain crude phenylethyl alcohol glycosides which have been known as bioactive part of R. rosea ; Secondly, WST-1 assay was carried out to assess the cell viability of astrocytes and cortical neurons under the treatment of the purified (> 95%) SAL. Moreover, WST-1 assay was also used to evaluate the cytoprotective effects of SAL preventing astrocytes from staurosporine-induced cell death; Thirdly, we examined the spontaneous reactive oxygen species (ROS) and staurosporine-induced ROS generation in astrocytes in the absence or presence of SAL. Results SAL was observed to improve the astrocytes viability but not cortical neurons. In addition, SAL was able to ameliorate staurosporine-induced cell death. Moreover, SAL was able to attenuate the spontaneous ROS and staurosporine-induced ROS generation. Conclusion We here confirm that the anti-oxidative effect of SAL on primary astrocytes might be an important mechanism accounting for the cytoprotective effects from SAL.

Six flavonoids, diosmetin (1), apigenin (2), luteolin (3), chrysoeriol (4), apigenin-7-O-glucuronide (5) and acacetin (6), two caffeic acid sugar esters, verbascoside (7) and isoverbascoside (8), as well as one norsesquiterpene, blumenol A (9) were isolated or detected from the EtOAc and n-BuOH extract of the whole plants of Striga asiatica. Their structures were identified by H-1 & C-13 NMR, HR-LC-TOF-MS and co-injection with the reference standards. Among them, blumenol A (9) was firstly found from the Scrophulariaceae. Diosmetin (1) and isoverbascoside (8) were firstly reported from the genus Striga, while verbascoside (7) was reported from this plant for the first time. Based on our chemotaxonomic studies and the previous phytochemical studies on Scrophulariaceae, Striga could be tentatively treated as a member of the family Scrophulariaceae instead of Orobanchaceae. Radix astragali (Huangqi in Chinese) is a well-known traditional Chinese medicinal herb that has been used clinically in China for centuries to cure various diseases. To profile the antiviral constituents of this herb, a high-performance liquid chromatography-diode array detector-electrospray ionization-tandem mass spectrometry (HPLC-DAD-ESI-MS/MS) analytical method was developed to separate and determinate the active part of the extract of Radix astragali, which showed potent inhibition of several viruses. By comparing their retention time and MS data with those obtained from the authentic compounds and the published data, a total of 18 compounds, comprising 11 flavonoids and 7 saponins, were identified. This study provides an approach to rapidly characterize bioactive constituents in traditional Chinese medicines (TCMs).

The FY-94 and FY-97 National Defense Authorization Acts amended the Act to Prevent Pollution from Ships (APPS) to require fleet-wide installation of Plastics Waste Processors by December 1998, and installation of Pulpers and Shredders by December 2000. These requirements translate to an acquisition program, which could rapidly develop, procure and deploy processing equipment on about 200 surface ships (over 24 ship classes) within three years. To date, this program has successfully completed the ship integration of the Plastic Waste Processor and is in the stages of implementing the Pulpers and Shredders. » A previous paper on this subject broadly described the development of the acquisition strategy, the development of the equipment and the fleet integration plan. This paper focuses on the ownership issues of the program and how they affect program decisions. It reviews how the Navy selected its current compliance strategy, including the consideration of its effects on marine environment through the Environmental Impact Statement (EIS) studies. It details the various processes NAVSEA implemented to ensure the delivery of quality and affordable "turn-key" system to the fleet. These processes included a comprehensive installation design and review; rapid incorporation of lessons learned; timely deployment of Integrated Logistics Support to meet fleet operations to meet fleet introduction which included establishing interim spares; and instituting a comprehensive In-Service Engineering Agent assist and inspection program.

Eight compounds including four caffeoyl phenylpropanoid glycosides, jasnervosides A-D (1-4), one monoterpenoid glycoside, jasnervoside E (5), and three secoiridoid glycosides, jasnervosides F-H (10-12), were isolated from the stems of Jasminum nervosum Lour. (Oleaceae), along with four known compounds, poliumoside (6), verbascoside (7), alpha-L-rhamnopyranosyl-(1 -> 3)-O-(alpha-L-rhamnopyranosyl(1 -> 6)-1-O-E-caffeoyl-beta-D-glucopyranoside (8), and jaspolyanthoside (9). Their structures were elucidated on the basis of their physicochemical and spectroscopic properties. Compounds 1,2,4 and 11 displayed potent antioxidant activities in the DPPH assay, while 2 and 3 displayed good activities against LPS-induced TNF-alpha and IL-1 beta production in BV2 cells. Compounds 1-5 and 10-12 were evaluated for their cytotoxic activities against three human cancer cell lines (A-549, Bel-7402, and HCT-8), but none displayed significant activity. Three new monoterpene lactones, cimicifugolides A-C (1-3), along with a known one (4), were identified from the dried rhizome of Actaea cimicifuga L. that was used as traditional Chinese medicine for thousands of years with the Chinese common name of shengma. The structures of the new isolates were established using spectroscopic methods, including NMR, mass, UV, and IR spectra. The inhibition activity of compounds 1, 2, and 4 against pancreatic lipase was evaluated.

Matrix metalloproteinase 26 (MMP-26) is a novel member of the matrix metalloproteinase (MMP) family and is widely expressed in cancer cells of epithelial origin. MMP-26 has been shown to contribute to tumor development and to the restoration of tissue injury. In this study, in order to identify the functions of MMP-26 that contribute to the biological phenotype and behavior of human lung carcinoma A549 cells, we established an MMP-26 low-expressing tumor cell model using RNA interference (RNAi) transfection. These cells were used to investigate the role of MMP-26 in tumor progression. The MTT, colony forming, adhere-nce and spreading, wound-healing and Transwell chamber invasion assays were performed to analyze the invasion ability of pshRNA-MMP26-transfected A549 cells. Semi-quantitative reverse transcription polymerase chain reaction, Western blotting and double immunofluorescent staining were employed to detect the relationship between MMP-26 and MMP-9. Results showed that the adhesive rate was down-regulated in pshRNA-MMP26-transfected cells, compared to the controls. Silencing of the MMP-26 gene significantly retarded the invasiveness of A549 cells in Transwell insert invasion assays. The cells proliferated in the three-dimensional (3D) culture system. A549 cells transfected with the pshRNA-MMP26-C plasmid mainly developed reticular structures in morphology, and formed few clones with clear and smooth edges as well as tight intercellular junctions. The mRNA and protein expression of MMP-9 in pshRNA-MMP26-transfected cells were significantly lower than those of the controls. Double immunofluorescence labeling and focal laser scanning microscopy showed that MMP-26 was colocalized with MMP-9 in the controls. In conclusion, we successfully established an MMP-26 low-expressing cell model and confirmed that MMP-26 contributed to A549 cell invasion and migration in vitro. We also demonstrated that MMP-26 plays an important role in local invasion at least in part through coordination with MMP-9.

To study the effects of growth factors on the survival and proliferation of human spermatogonial stem cells (SSCs) in vitro.SSCs were treated with the growth factors SCF, LIF and bFGF added to the culture, each at the concentrations of 0, 5, 10 and 20 microg/L and repeated three times. The survival time and proliferation rate of the cells were determined every 8-12 hours and their morphological features observed with the light microscope and electron microscope.The survival time and proliferation rate of the SSCs were significantly increased in the treated groups as compared with the control (P0.05).The growth factors SCF, LIF and bFGF can promote the survival and proliferation of SSCs in vitro.

To investigate whether chronic childhood constipation (CCC) may cause oxidative stress and potential free radical damage to children, and to explore the mechanisms by which CCC may cause oxidative stress and potential free radical damage to chronic constipation patients (CCPs).Sixty CCPs and sixty healthy child volunteers (HCVs) whose ages, gender and others were matched for the CCPs were enrolled in a randomized controlled study, in which levels of vitamin C (VC) and vitamin E (VE) in plasma as well as activities of superoxide dismutase (SOD) and catalase (CAT) in erythrocytes were determined by spectrophotometric analytical methods.Compared with average values of the above biochemical parameters in the HCVs group, the average values of VC and VE in plasma as well as those of SOD and CAT in erythrocytes in the CCPs group were significantly decreased (P0.0001). Linear regression and bivariate correlation analysis showed that with prolonged course of the CCPs, the levels of VC and VE in plasma as well as the activities of SOD and CAT in erythrocytes in the CCPs were decreased gradually (P0.0001).The findings in the present study suggest that chronic childhood constipation causes oxidative stress and potential free radical damage to children with chronic constipation.
