The sub-50 nm pH-responsive nanoparticles have cross-linked acidic cores and galactosylated shells, which endow them with high targeting efficacy to hepatoma cells, high drug loading efficiency at pH 7.4, and rapid squeezable drug release at pH 5.0.

Trichogramma spp. (Hymenoptera: Trichogrammatidae) are generalist egg parasitoids widely used as biological control agents, yet with limited efficacy against Tuta absoluta (Meyrick; Lepidoptera: Gelechiidae), a key pest of tomato. Olfactory cues related to the host-plant complex influence the searching behavior in parasitoids. Studies have shown that female Trichogramma spp. can learn odor cues affecting their preference. We assessed if learning increases the response to tomato odor and whether it improves the parasitism rate of Trichogramma achaeae, Trichogramma evanescens, and Trichogramma dendrolimi. The parasitoids were either naive, reared for one generation on T. absoluta eggs, or conditioned associatively with an oviposition experience on a factitious host in the presence of tomato odor. Subsequently, their arrestment response (searching time) to T. absoluta-infested tomato leaves was observed in a static four-chamber olfactometer. Rearing the parasitoids on T. absoluta increased the arrestment time of T. dendrolimi strongly and that of T. evanescens slightly but not that of T. achaeae. In contrast, oviposition experience on a factitious host with the tomato odor (leaves or extract) increased the searching time of T. achaeae and T. evanescens strongly but did not improve the response of T. dendrolimi. Concerning parasitism improvement, only T. achaeae showed an increased parasitism rate after associative learning. Hence, the learning capacity of T. achaeae should be further explored to improve its biological control efficacy against T. absoluta.

The South American tomato pinworm, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), has invaded most Afro-Eurasian countries and is threatening worldwide tomato production. Various strategies have been developed and implemented to manage this pest. Here, we present a timely review on the up-to-date development and practical implementation of integrated pest management (IPM) programs for tomato crops across different world regions infested by T. absoluta. While insecticide resistance is a growing concern, biological control via releasing or conserving arthropod natural enemies and sex pheromone-based biotechnical control are the most successful management practices. Agronomic control-related research is an emerging area where the soil fertilization and/or irrigation, as well as breeding of resistant cultivars, has the potential to enhance IPM effectiveness. Grower survey responses in the native areas (i.e., South America), early-invaded areas (i.e., first report between 2006 and 2012) and newly invaded areas (i.e., first report after 2012) showed that the control programs evolved along with the areas and time since invasion. Growers in the early-invaded areas shifted more rapidly from chemical control to biological control compared to those from the native area. In all concerned regions, the pest control failure risk following chemical insecticide applications and the high cost associated with either biological or biotechnical control methods have been the greatest concerns for growers. The information gathered from the native and/or early-invaded areas may help achieve a more effective management in newly invaded areas. Lastly, researchers are expected to break the bottlenecks of some key issues that would enable lowering application cost of novel biorational alternative management options.

In this paper, the contact interface between FGH96 superalloy melts and refractory slurry with corundum powder and silica sol at 1600°C with different soaking time in 10-240 min range was investigated. The morphology and composition of the contact interface were studied by optical microscopy (OM), scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS). The results show that the reaction layer formed on the contact interface between the melting superalloy and the refractory slurry, and is mainly consist of Alumina and contains a small amount of other metal elements such as Ti and Cr. The reaction layer was not smooth on the micro level, and there was a peninsula-like structure protruding into the internal part of the melting superalloy on the reaction zone, and even started to fall off at some places to form islands. With the increase of soaking time, the reaction between melt of superalloy and refractory slurry increased gradually and the reaction layer began to combine with the refractory slurry substrate and form obvious interaction layered structure, resulting in the corrosion of refractory slurry substrate. With the soaking time over 120 min, the stable contact interface was destroyed. Thermodynamic calculation shows that the substitution reaction between Al in superalloy and SiO2 in refractory slurry meets the thermodynamic conditions, and the reaction can proceed forward.

M1 microglial activation is crucial for the pathogenesis of early brain injury (EBI) following subarachnoid hemorrhage (SAH), and there is growing evidence that glucose metabolism is frequently involved in microglial activation. However, the molecular mechanism of glycolysis and its role in M1 microglial activation in context of EBI are not yet fully understood. In this study, firstly, the relationship between aerobic glycolysis and M1 microglial activation as well as SAH-induced EBI was researched in vivo. Then, intervention on mammalian target of rapamycin (mTOR) was performed to investigate the effects on glycolysis-dependent M1 microglial activation and EBI, and its relationship with hypoxia-inducible factor-1α (HIF-1α) in vivo. Next, Hif-1α was inhibited to analyze its role in aerobic glycolysis, M1 microglial activation and EBI in vivo. Lastly, both in vivo and in vitro, mTOR inhibition and Hif-1α enhancement were administered simultaneously, and the combined effects were further confirmed again. The results showed that aerobic glycolysis and M1 microglial polarization were increased after SAH, and glycolytic inhibition could attenuate M1 microglial activation and EBI. Inhibition of mTOR reduced glycolysis-dependent M1 microglial polarization and EBI severity by down-regulating HIF-1α expression, while enhancement had the opposite effects. Blockading HIF-1α had the similar effects as suppressing mTOR, while HIF-1α agonist worked against mTOR antagonist when administered simultaneously. In conclusion, the present study showed new evidence that aerobic glycolysis induced by mTOR/HIF-1α might promote EBI after SAH by activating M1 microglia. This finding provided new insights for the treatment of EBI.

To explore the prevalence and related factors influencing diabetes stigma among Chinese young and middle-aged patients with type 2 diabetes.A cross-sectional, quantitative study was conducted through questionnaires.A total of 453 young and middle-aged patients with type 2 diabetes were recruited. Questionnaire survey was conducted with convenience sampling from September 2019 to December 2020. This included demographic questionnaire, Type 2 Diabetes Stigma Assessment Scale (DSAS-2) and Connor-Davidson Resilience Scale (CD-RISC). The STROBE checklist was applied as the reporting guideline for this study.The scores of diabetes stigma and resilience of patients were 48.18 ± 14.52 and 66.02 ± 7.18, respectively. Diabetes stigma was negatively correlated with resilience, the correlation coefficient was -0.581. The results showed that the complications, diabetes year, monthly income and insulin treatment are important factors affecting stigma. In addition, it is resilience that was a protective factor of diabetes stigma, explaining 27.2% of the variance in diabetes stigma.

Stearoyl-CoA-desaturase 1 (SCD1) deficiency mediates apoptosis in colorectal cancer cells by promoting ceramide de novo synthesis. The mechanisms underlying the cross-talk between SCD1 and ceramide synthesis have not been explored. We treated colorectal cancer cells with an SCD1 inhibitor and examined the effects on gene expression, cell growth, and cellular lipid contents. The main effect of SCD1 inhibition on the fatty acid contents of colorectal cancer cells was a decrease in monounsaturated fatty acids (MUFAs). RNA sequencing (RNA-seq) showed that the most intense alteration of gene expression after SCD1 inhibition occurred in the NF-κB signaling pathway. Further experiments revealed that SCD1 inhibition resulted in increased levels of phosphorylated NF-κB p65 and increased nuclear translocation of NF-κB p65. Treatment with an NF-κB inhibitor eliminated several effects of SCD1 inhibition, mainly including overexpression of serine palmitoyltransferase1 (SPT1), elevation of dihydroceramide contents, and suppression of cell growth. Furthermore, treatment with supplemental oleate counteracted the SCD1-induced NF-κB activation and downstream effects. In summary, our data demonstrate that the NF-κB pathway plays a role in SCD1 deficiency-induced ceramide de novo synthesis in colorectal cancer cells, and that reduced MUFA levels contribute to the course.

Oxygen is an essential molecule for animal respiration, growth, and survival. Unlike in terrestrial environments, contamination and climate change have led to the frequent occurrence of hypoxia in aquatic environments, thus impacting aquatic animal survival. However, the adaptative mechanisms underlying fish responses to environmental hypoxia remain largely unknown. Here, we used large yellow croaker ( Larimichthys crocea) and large yellow croaker fry (LYCF) cells to investigate the roles of the Hif-1α/Hsf1/Hsp70 signaling pathway in the regulation of cellular redox homeostasis, and apoptosis. We confirmed that hypoxia induced the expression of Hif-1α, Hsf1, and Hsp70 in vivo and in vitro. Genetic Hsp70 knockdown/overexpression indicated that Hsp70 was required for maintaining redox homeostasis and resisting oxidative stress in LYCF cells under hypoxic stress. Hsp70 inhibited caspase-dependent intrinsic apoptosis by maintaining normal mitochondrial membrane potential, enhancing Bcl-2 mRNA and protein expression, inhibiting Bax and caspase3 mRNA expression, and suppressing caspase-3 and caspase-9 activation. Hsp70 suppressed caspase-independent intrinsic apoptosis by inhibiting nuclear translocation of apoptosis-inducing factor (AIF) and disturbed extrinsic apoptosis by inactivating caspase-8. Genetic knockdown/overexpression of Hif-1α and dual-luciferase reporter assay indicated that Hif-1α activated the Hsf1 DNA promoter and enhanced Hsf1 mRNA transcription. Hsf1 enhanced Hsp70 mRNA transcription in a similar manner. In summary, the Hif-1α/Hsf1/Hsp70 signaling pathway plays an important role in regulating redox homeostasis and anti-apoptosis in L. crocea under hypoxic stress.

The large yellow croaker (Larimichthys crocea), which is an economically important mariculture fish in China, is often exposed to environmental hypoxia. Reactive oxygen species (ROS) homeostasis is essential for the maintenance of normal physiological conditions in an organism. Direct evidence that environmental hypoxia leads to ROS overproduction is scarce in marine fish. Furthermore, the sources of ROS overproduction in marine fish under hypoxic stress are poorly known. In this study, we investigated the effects of hypoxia on redox homeostasis in L. crocea and the impact of impaired redox homeostasis on fish. We first confirmed that hypoxia drove ROS production mainly via the mitochondrial electron transport chain and NADPH oxidase complex pathways in L. crocea and its cell line (large yellow croaker fry (LYCF) cells). We subsequently detected a marked increase in the antioxidant systems of the fish. However, imbalance between the pro-oxidation and antioxidation systems ultimately led to excessive ROS and oxidative stress. Cell viability showed a remarkable decrease while oxidative indicators, such as malondialdehyde, protein carbonylation, and 8-hydroxy-2 deoxyguanosine, showed a significant increase after hypoxia, accompanied by tissue damage. N-acetylcysteine (NAC) reduced ROS levels, alleviated oxidative damage, and improved cell viability in vitro. Appropriate uptake of ROS scavengers (e.g., NAC and elamipretide Szeto-Schiller-31) and inhibitors (e.g., apocynin, diphenylene iodonium, and 5-hydroxydecanoate) may be effective at overcoming hypoxic toxicity. Our findings highlight previously unstudied strategies of hypoxic toxicity resistance in marine fish.

Under dehumidifying conditions, the condensed water will directly affect the heat transfer and resistance characteristics of a fin-and-tube heat exchanger. The geometrical form of condensed water on fin surfaces of three different fin materials (i.e., copper fin, aluminum fin, and aluminum fin with hydrophilic layer) in a fin-and-circular-tube heat exchanger was experimentally studied in this paper. The effect of the three different fin materials on heat transfer and friction performance of the heat exchanger was researched, too. The results show that the condensation state on the surface of copper fin and aluminum fin are dropwise condensation. The condensation state on the surface of the aluminum fin with the hydrophilic layer is film condensation. For the three different material fins, increasing the air velocity (ua,in) and relative humidity (RHin) of the inlet air can enhance the heat transfer of the heat exchanger. Friction factor (f) of the three different material fins decreases with the increase of ua,in, however, increases with the increase of RHin. At the same ua,in or RHin, Nusselt number (Nu) of the copper fin heat exchanger is the largest and Nu of the aluminum fin with hydrophilic layer is the smallest, f of the aluminum fin heat exchanger is the largest and f of the aluminum fin with hydrophilic layer is the smallest. Under the identical pumping power constrain, the comprehensive heat transfer performance of the copper fin heat exchanger is the best for the studied cases.

The large yellow croaker (Larimichthys crocea) is an important marine economic fish in China, however, its intolerance to hypoxia causes widespread mortality. To understand the molecular mechanisms underlying hypoxia tolerance in L. crocea, the transcriptome gene expression profiling of three different tissues (blood, gills, and liver) of L. crocea exposed to hypoxia and reoxygenation stress were performed. In parallel, the gene relationships were investigated based on weighted gene co-expression network analysis (WGCNA). Accordingly, the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis showed that several pathways (e.g., energy metabolism, signal transduction, oxygen transport, and osmotic regulation) may be involved in the response of L. crocea to hypoxia and reoxygenation stress. In addition, also, four key modules (darkorange, magenta, saddlebrown, and darkolivegreen) that were highly relevant to the samples were identified by WGCNA. Furthermore, some hub genes within the association module, including RPS16, EDRF1, KCNK5, SNAT2, PFKL, GSK-3β, and PIK3CD, were found. This is the first study to report the co-expression patterns of a gene network after hypoxia stress in marine fish. The results provide new clues for further research on the molecular mechanisms underlying hypoxia tolerance in L. crocea.

The frequency and duration of heat waves are expected to increase under global warming, so parasitoids could increasingly encounter heat waves at some stage of their life cycle. However, few studies have focused on how host-feeding parasitoids respond to heat waves. We investigated how the frequency of heat waves during different developmental stages (larva, pupa and adult) of a host-feeding parasitoid, Eretmocerus hayati, affects its life history traits and life table parameters. The results showed that when continuous heat waves were experienced during a single stage, the development time of the stage that encountered the heat waves (either the larval or pupal stage) was significantly prolonged, while one heat wave did not have the same effect. Regardless of which stage (larval or pupal stage) of parasitoids encountered continuous heat waves, life history traits (host feeding, fecundity and longevity) and life table parameters (intrinsic rate of increase, r) were significantly decreased. Moreover, heat waves at the pupal stage had greater detrimental effects on adult performance than heat waves at the larval stage. However, when adult parasitoids experienced continuous heat waves, their lifetime fecundity did not differ significantly from that of parasitoids that experienced heat waves only once during a single stage. When two or three stages faced continuous heat waves, adult performance sharply decreased, especially in terms of r. Hence, these findings suggest that the timing and duration of heat waves should be assessed before releasing a parasitoid as a biological control agent. If such an assessment is not performed, the biological control efficiency could be greatly decreased.

Background Clindamycin is a lincosamide antibiotic used to treat staphylococcal and streptococcal infections. Reports of clinical Streptococcus agalactiae isolates with the rare lincosamide resistance/macrolide susceptibility (LR/MS) phenotype are increasing worldwide. In this study, we characterised three clinical S. agalactiae strains with the unusual L phenotype from China. Methods Three clinical S. agalactiae strains, Sag3, Sag27 and Sag4104, with the L phenotype were identified from 186 isolates collected from 2016 to 2018 in Shanghai, China. The MICs of clindamycin, erythromycin, tetracycline, levofloxacin, and penicillin were determined using Etest. PCR for the lnu(B) gene was conducted. Whole genome sequencing and sequence analysis were carried out to investigate the genetic context of lnu(B). Efforts to transfer lincomycin resistance by conjugation and to identify the circular form by inverse PCR were made. Results Sag3, Sag27, and Sag4104 were susceptible to erythromycin (MIC ≤0.25 mg/L) but resistant to clindamycin (MIC ≥1 mg/L). lnu(B) was found to be responsible for the L phenotype. lnu(B) in Sag3 and Sag27 were chromosomally located in an aadE-spw-lsa(E)-lnu(B) resistance gene cluster adjacent to an upstream 7-kb tet(L)-cat resistance gene cluster. Two resistance gene clusters were flanked by the IS6-like element, IS1216. Sag4104 only contained partial genes of aadE-spw-lsa(E)-lnu(B) resistance gene cluster and was also flanked by IS1216. Conclusion These results established the presence of the L phenotype associated with lnu(B) in clinical S. agalactiae isolates in China. The lnu(B)-containing multi-resistance gene cluster possibly acts as a composite transposon flanked by IS1216 and as a vehicle for the dissemination of multidrug resistance among S. agalactiae.

When two parasitoids exploit a common host simultaneously, competitive interactions between them usually occur, and are more complex among host-killing species. Diglyphus isaea (Walker) and Neochrysocharis formosa (Westwood) are larval parasitoids of agromyzid leafminers. They have widespread distributions and share the same host resources when sympatric. In this study, interactions between these two parasitoids and different parasitoid release modes were investigated. Parasitoids were exposed to leaves infested with late second to early third instar Liriomyza sativae at two different host densities in Petri dishes. The total death rate of N. formosa released alone was density-dependent. And release modes also impacted the total death rate of parasitoids at a high host density. However, the oviposition number of D. isaea was significantly greater than that of N. formosa when they coexisted. In addition, host-feeding proportions were greatest among three host-killing behaviors: parasitism, host feeding and host stinging. These results contribute to a further understanding of parasitoids’ host-killing strategies.

Background: Aortic valve sclerosis (AVSc), a common precursor to calcific aortic valve disease, may progress into advanced aortic stenosis with hemodynamic instability. However, plasma biomarkers of such a subclinical condition remain lacking. Since impaired fibrinolysis featuring dysregulated tissue plasminogen activator (t-PA) is involved in several cardiovascular diseases, we investigated whether endogenous t-PA was also associated with AVSc.Methods: Plasma t-PA levels were measured in 295 consecutive patients undergoing standard echocardiography and Doppler flow imaging. Multiple logistic regression analyses were used to assess the association between t-PA and AVSc. Receiver operating characteristic curve analysis was performed for determining the diagnostic value of t-PA for AVSc. The performance of adding t-PA to clinical signatures of AVSc was evaluated. Concentration of t-PA was assessed in human sclerotic and non-sclerotic aortic valves by histology and immunohistochemistry analysis.Results: Plasma t-PA was higher in patients with AVSc than in non-AVSc counterparts (median, 2063.10 vs. 1403.17 pg/mL, p < 0.01). C-statistics of plasma t-PA for discriminating AVSc was 0.698 (95%CI: 0.639–0.758). The performance of t-PA for identifying AVSc was better among male and non-hypertensive patients [C-statistics (95%CI): 0.712 (0.634–0.790) and 0.805 (0.693–0.916), respectively]. Combination of t-PA and clinical factors improved classification of the patients (category-free NRI: 0.452, p < 0.001; IDI: 0.020, p = 0.012). The concentration of t-PA was three times higher in sclerotic compared to non-sclerotic aortic valves.Conclusion: Elevated circulating t-PA level confers an increased risk for AVSc. Further prospective studies with larger sample size are needed to examine if t-PA could serve as a diagnostic clinical marker for AVSc.

As the main exploitation mean of low permeability reservoirs, multi-stage fracturing plays a significant role in horizontal well. However, due to the multi-stage fracturing technology lack of focused research on crack interaction phenomenon, limiting its further development. In this paper, considering the basis of rock and soil mechanics, flow mechanics theory, the fluid-solid coupling and combining with extended finite element method (XFEM) to simulate the fracture propagation of 2–4 vertical cracks for simultaneous fracturing in horizontal well. Based on the fracture morphology, optimizing the number of fractures, cluster spacing and segment spacing by comparing the crack width of deflection point with the crack width of injection point. The simulation results are applied in field for verification. The study results are as follows: Selecting 2–3 cracks per segment can efficiently satisfy the requirement of production; the optimal cluster spacing of simultaneous fracturing for two fractures is 20–30 m; the optimal cluster spacing of simultaneous fracturing for three fractures is 30–40 m; the optimal segment spacing is 30–40 m. The results suggested as above illustrate that the more the number of fractures, the better the effect of multi-stage fracturing is not correct. Meanwhile, the reasonable fracture spacing can effectively reduce the negative impact of crack interaction phenomenon. The research method and optimal parameters proposed in this paper provides a theoretical basis for practical production in horizontal well.

Aquatic vegetation is an important part of lake ecosystems and plays a vital role in improving water quality and maintaining biodiversity. At present, China's lakes are facing eutrophication and the degradation of aquatic vegetation. The monitoring of temporal and spatial variations in aquatic vegetation and elucidating the main influencing factors are of great significance for protecting aquatic vegetation and restoring eutrophic lake ecosystems. Therefore, we introduced the Vegetation Present Frequency (VPF) method to extract data on aquatic vegetation and combined this with meteorological factors and human activities to analyze the temporal and spatial in Lake Hongze based on MODIS data from 2007 to 2017. The VPF of aquatic vegetation in Lake Hongze showed clear seasonal and interannual variations. The VPF was significantly higher in spring and summer than in autumn and winter (

The purpose of this study is to research the effect of high speed stirring by emulsifier blades on the thermal safety of emulsion matrix. In order to analyze thermal safety of emulsion matrix in emulsifying process, the heating effect influenced by rotor speed and emulsify temperature was combined with self-heat properties of emulsion matrix in this study. Computational fluid dynamics (CFD) is used for analysis. The emulsifier model adopts simplified CYJ type emulsifier, which is a kind of vertical emulsifier and has a large stirring area. The self-heat properties of emulsion matrix are described by Arrhenius equation. The simulation results showed that heat could not accumulate in emulsion matrix due to the strong turbulence formed by stirring. But, because of the shear stress and friction, a high temperature region will be formed around the blades of the rotor in a short time. If the stirring speed reaches 10300 rpm, the temperature in this region would be able to reach the critical ignition temperature of emulsion matrix.

Hemiptarsenus varicornis (Girault), a destructive host-feeding wasp, is an important biocontrol agent/larval ectoparasitoid of agromyzid leafminers worldwide. In the present study, the life history and life table of H. varicornis reared with Liriomyza trifolii (Burgess) were studied at a constant 27 °C. The developmental durations of female and male eggs, larvae, prepupae, pupae and total immature wasps were 1.00 and 1.00, 2.57 and 2.62, 0.45 and 0.37, 3.88 and 3.52, and 7.90 and 7.52 days, respectively. This wasp showed three types of host-killing behavior: reproductive parasitization (parasitism), non-reproductive host feeding (host feeding), and host stinging without oviposition or feeding (host stinging), resulting in 133.9, 303.8, and 84.2, respectively, killed host larvae. We confirm that H. varicornis is a strong synovigenic parasitoid, with an ovigeny index of 0.003. The number of host-feeding events was strongly correlated with parasitism, host-stinging events, longevity and total host mortality. The intrinsic rate of increase, the finite rate of increase, the net reproductive rate, the gross reproduction rate, and the mean length of a generation of H. varicornis were 0.3011/day, 1.3624/day, 66.22 offspring/individual, 168.33 offspring/individual, and 13.56 days, respectively. These results could contribute to a better understanding of the biocontrol efficiency of this destructive host feeder.
