Abstract The evolutions of MHD instability behaviors and enhancement of both electrostatic and electromagnetic turbulence towards the plasma disruption have been clearly observed in the HL-2A plasmas. Two types of plasma disruptive discharges have been investigated for similar equilibrium parameters: one with a distinct stage of a small central temperature collapse ( $$\sim$$ ∼ 5–10%) around 1 millisecond before the thermal quench (TQ), while the other without. For both types, the TQ phase is preceded by a rotating 2/1 tearing mode, and it is the development of the cold bubble from the inner region of the 2/1 island O-point along with its inward convection that causes the massive energy loss. In addition, the micro-scale turbulence, including magnetic fluctuations and density fluctuations, increases before the small collapse, and more significantly towards the TQ. Also, temperature fluctuations measured by electron cyclotron emission imaging enhances dramatically at the reconnection site and expand into the island when approaching the small collapse and TQ, and the expansion is more significant close to the TQ. The observed turbulence enhancement near the X-point cannot be fully interpreted by the linear stability analysis by GENE. Evidences suggest that nonlinear effects, such as the reduction of local $$E_r\times B$$ E r × B shear and turbulence spreading, may play an important role in governing turbulence enhancement and expansion. These results imply that the turbulence and its interaction with the island facilitate the stochasticity of the magnetic flux and formation of the cold bubble, and hence, the plasma disruption.

The hyperspectral image (HSI) has poor spatial resolution and is vulnerable to cloud, which limits its application in practical tasks. However, current hyperspectral and multispectral image fusion algorithms do not consider the cloud contamination problem. In view of this situation, we propose a novel dual-branch fusion framework to improve spatial resolution of HSI while restoring the details of areas covered by clouds. The network extracts multiscale and multilevel spatiospectral features through the dual-branch encoder–decoder structure to focus on local information, and acquires the nonlocal similar relationship by transformer to aggregate global information of features for better reconstruction. To produce a more natural image, the spatiospectral gradient loss with L1 loss of reconstructed image is introduced to guide network training. It can be seen from the cloud-free and cloud-contaminated experiment results that the proposed method achieves the best results in both visual and accuracy evaluation, compared with the recent state of the arts.

Heat stress hinders the growth and productivity of sweetpotato plants, predominantly through oxidative damage to cellular membranes. Therefore, the development of efficient approaches for mitigating heat-related impairments is essential for the long-term production of sweetpotatoes. Melatonin has been recognised for its capacity to assist plants in dealing with abiotic stress conditions. This research aimed to investigate how different doses of exogenous melatonin influence heat damage in sweetpotato plants. Heat stress drastically affected shoot and root fresh weight by 31.8 and 44.5%, respectively. This reduction resulted in oxidative stress characterised by increased formation of hydrogen peroxide (H2O2) by 804.4%, superoxide ion (O2•−) by 211.5% and malondialdehyde (MDA) by 234.2%. Heat stress also reduced chlorophyll concentration, photosystem II efficiency (Fv/Fm) by 15.3% and gaseous exchange. However, pre-treatment with 100 μmol L−1 melatonin increased growth and reduced oxidative damage to sweetpotato plants under heat stress. In particular, melatonin decreased H2O2, O2•− and MDA by 64.8%, 42.7% and 38.2%, respectively. Melatonin also mitigated the decline in chlorophyll levels and improved stomatal traits, gaseous exchange and Fv/Fm (13%). Results suggested that the favorable outcomes of melatonin treatment can be associated with elevated antioxidant enzyme activity and an increase in non-enzymatic antioxidants and osmo-protectants. Overall, these findings indicate that exogenous melatonin can improve heat stress tolerance in sweetpotatoes. This study will assist researchers in further investigating how melatonin makes sweetpotatoes more resistant to heat stress.

Abstract Evaluating and predicting how carbon storage (CS) is impacted by land use change can enable optimizing of future spatial layouts and coordinate land use and ecosystem services. This paper explores the changes in and driving factors of Zunyi CS from 2000 to 2020, predicts the changes in CS under different development scenarios, and determines the optimal development scenario. Woodland and farmland are the main land use types in Zunyi. Land use change was reflected mainly in the mutual conversion among woodland, farmland, and grassland and by their conversion to construction land and water. In 2000, 2010, and 2020, the CS in Zunyi was 658.77 × 10^6 t, 661.44 × 10^6 t, and 658.35 × 10^6 t, respectively. Woodland, farmland and grassland conversions to construction land and water were primarily responsible for CS loss. The normalized difference vegetation index (NDVI) is the main factor influencing the pattern of CS (q > 10%). Furthermore, the impacts of the human footprint index and population density are increasing. In 2030, the CS of Zunyi is trending downward. Under the ecological-farmland conservation scenario (ECS), the CS is estimated to be 656.67 × 10^6 t, with the smallest decrease (− 0.26%) among timepoints. The effective control of woodland and farmland weakens the trend of CS reduction.

Abstract Deep phenotyping can enhance the power of genetic analysis, including genome-wide association studies (GWAS), but the occurrence of missing phenotypes compromises the potential of such resources. Although many phenotypic imputation methods have been developed, the accurate imputation of millions of individuals remains challenging. In the present study, we have developed a multi-phenotype imputation method based on mixed fast random forest (PIXANT) by leveraging efficient machine learning (ML)-based algorithms. We demonstrate by extensive simulations that PIXANT is reliable, robust and highly resource-efficient. We then apply PIXANT to the UKB data of 277,301 unrelated White British citizens and 425 traits, and GWAS is subsequently performed on the imputed phenotypes, 18.4% more GWAS loci are identified than before imputation (8710 vs 7355). The increased statistical power of GWAS identified some additional candidate genes affecting heart rate, such as RNF220, SCN10A, and RGS6, suggesting that the use of imputed phenotype data from a large cohort may lead to the discovery of additional candidate genes for complex traits.

Statistical characteristics and the classification of the topside ionospheric mid-latitude trough are systemically analyzed, using observations from the Defense Meteorological Satellite Program F18 (DMSP-F18) satellite. The data was obtained at an altitude of around 860 km in near polar orbit, throughout 2013. Our study identified the auroral boundary based on the in-situ electron density and electron spectrum, allowing us to precisely determine the location of the mid-latitude trough. This differs from most previous works, which only use Total Electron Content (TEC) or in-situ electron density. In our study, the troughs exhibited a higher occurrence rate in local winter than in summer, and extended to lower latitudes with increasing geomagnetic activity. It was found that the ionospheric mid-latitude trough, which is associated with temperature changes or enhanced ion drift, exhibited distinct characteristics. Specifically, the ionospheric mid-latitude troughs related to electron temperature (Te) peak were located more equatorward of auroral oval boundary in winter than in summer. The ionospheric mid-latitude troughs related to Te-maximum were less frequently observed at 60−70°S magnetic latitude and 90−240°E longitude. Furthermore, the troughs related to ion temperature (Ti) maximums were observed at relatively higher latitudes, occurring more frequently in winter. In addition, the troughs related to ion velocity (Vi) maximums could be observed in all seasons. The troughs with the maximum-Ti and maximum-Vi were located closer to the equatorward boundary of the auroral oval at the nightside, and in both hemispheres. This implies that enhanced ion drift velocity contributes to increased collisional frictional heating and enhanced ion temperatures, resulting in a density depletion within the trough region.

Abstract The superconducting diode effect (SDE) is defined by the difference in the magnitude of critical currents applied in opposite directions. It has been observed in various superconducting systems and attracted high research interests. However, the operating temperature of the SDE is typically low and/or the sample structure is rather complex. For the potential applications in non-dissipative electronics, efficient superconducting diodes working in zero magnetic field with high operating temperatures and a simple configuration are highly desired. Here, we report the observation of a SDE under zero magnetic field with operating temperatures up to 72 K and efficiency as high as 22% at 53 K in high-transition-temperature (high-T c) cuprate superconductor Bi2Sr2CaCu2O8+δ (BSCCO) flake devices. The rectification effect persists beyond two hundred sweeping cycles, confirming the stability of the superconducting diode. Our results offer promising developments for potential applications in non-dissipative electronics, and provide insights into the mechanism of field-free SDE and symmetry breakings in high-T c superconductors.

Abstract The prevalence of obesity and osteoporosis (OP) represents a significant public health concern on a global scale. A substantial body of evidence indicates that there is a complex relationship between obesity and OP, with a correlation between the occurrence of OP and obesity. In recent years, sirtuins have emerged as a prominent area of interest in the fields of aging and endocrine metabolism. Among the various research avenues exploring the potential of sirtuins, the effects of these proteins on obesity and OP have garnered significant attention from numerous researchers. Sirtuins regulate energy balance and lipid balance, which in turn inhibit the process of adipogenesis. Additionally, sirtuins regulate the balance between osteogenic and osteoblastic activity, which protects against the development of OP. However, no study has yet provided a comprehensive discussion of the relationship between the three: sirtuins, obesity, and OP. This paper will therefore describe the relationship between sirtuins and obesity, the relationship between sirtuins and OP, and a discussion focusing on the possibility of treating OP caused by obesity by targeting sirtuins. This will be based on the common influences on the occurrence of obesity and OP (such as mesenchymal stem cells, gut microbiota, and insulin). Finally, the potential of SIRT1, an important member of sirtuins, in polyphenolic natural products for the treatment of obesity and OP will be presented. This will contribute to a better understanding of the interactions between sirtuins and obesity and bone, which will facilitate the development of new therapeutic strategies for obesity and OP in the future.

Abstract Repeated closed-head injuries (rCHI) from activities like contact sports, falls, military combat, and traffic accidents pose a serious risk due to their cumulative impact on the brain. Often, rCHI is not diagnosed until symptoms of irreversible brain damage appear, highlighting the need for preventive measures. This study assessed the prophylactic efficacy of remote photobiomodulation (PBM) targeted at the lungs against rCHI-induced brain injury and associated behavioral deficits. Utilizing the “Marmarou” weight-drop model, rCHI was induced in rats on days 0, 5, and 10. Remote PBM, employing an 808 nm continuous wave laser, was administered daily in 2-min sessions per lung side over 20 days. Behavioral deficits were assessed through three-chamber social interaction, forced swim, grip strength, open field, elevated plus maze, and Barnes maze tests. Immunofluorescence staining and 3D reconstruction evaluated neuronal damage, apoptosis, degeneration, and the morphology of microglia and astrocytes, as well as astrocyte and microglia-mediated excessive synapse elimination. Additionally, 16S rDNA amplicon sequencing analyzed changes in the lung microbiome following remote PBM treatment. Results demonstrated that remote PBM significantly improved depressive-like behaviors, motor dysfunction, and social interaction impairment while enhancing grip strength and reducing neuronal damage, apoptosis, and degeneration induced by rCHI. Analysis of lung microbiome changes revealed an enrichment of lipopolysaccharide (LPS) biosynthesis pathways, suggesting a potential link to neuroprotection. Furthermore, remote PBM mitigated hyperactivation of cortical microglia and astrocytes and significantly reduced excessive synaptic phagocytosis by these cells, highlighting its potential as a preventive strategy for rCHI with neuroprotective effects.

In hyperspectral images, the high dimensionality of spectral data often leads to redundant spectral information, making it difficult to extract features. Two-dimensional CNNs fail to effec-tively extract spatial and spectral information, and deploying three-dimensional CNNs on microprocessors is challenging as these net-works consume excessive resources. If graph convolutional networks (GCN) are adopted, most networks employ superpixel segmentation for HSI classification. However, this approach tends to overlook pixel-level features and thus fails to achieve fine classification.To efficiently extract spectral features while reducing resource consumption, we proposed the spectral shift module (SPCSM). This module extracts features by circularly shifting spectral information. It emphasizes the internal correlations within the spectral and offers the advantage of fewer parameters. Based on the SPCSM, we designed the spatial–spectral shift module (S2SM). It models and analyzes correlations between spatial and spectral data, facilitating the extraction of spatial and spectral features. Additionally, it addresses the issues of redundancy in shallow information. To enable deployment on microprocessors and effectively classify hyperspectral images, we optimized the MambaOut network by integrating the S2SM with a multihead MambaOut (LS2SM-MHMambaOut). Experiments demonstrated that our network struck a favorable balance between classification accuracy and model complexity. Therefore, it is promising to be integrated into microprocessors.

Abstract Rapeseed (Brassica napus L.) possesses substantial economic value as an oil, vegetable, and forage crop, while also exhibiting notable ornamental characteristics. Recent advances in flower colour breeding have significantly enhanced the visual appeal of rapeseed, with anthocyanins identified as the primary contributor to the development of red, purple, and pink flowers. However, the mechanisms underlying the synthesis and regulation of anthocyanins during petal coloration in rapeseed are still poorly understood. This research combined miRNA and mRNA expression data from four different color phases, along with degradome analysis, to discover important miRNA-mRNA modules responsible for controlling the accumulation of anthocyanin in purple-flowered rapeseed. In the process of petal development, a grand sum of 247 miRNAs (including 223 known and 24 novel miRNAs) were effectively detected, with 64 of them displaying differential expression patterns. Degradome sequencing was used to conduct a comprehensive analysis of 152 targets for the differential expression miRNAs. Out of these, 108 miRNA-mRNA modules exhibit contrasting expression patterns. Some miRNAs and their corresponding targets have additionally been discovered, potentially playing a role in governing the buildup of anthocyanin in purple-flowered rapeseed. The regulatory modules miR156-SPL9 and miR828-PAP2 composed of miR156b and miR828 and their targets may play a key role in this process. The results offer a thorough analysis of miRNAs linked to the regulation of anthocyanin in B. napus, offering valuable understanding into the regulatory processes that govern miRNA-mediated anthocyanin production in Brassica crops.

Abstract Urolithiasis is a common and recurrent condition in the urological spectrum. Despite various proposed mechanisms, the causal relationship between sleep traits and the risk of urolithiasis remains unclear. We used publicly available genome-wide association study (GWAS) summary data from the UK Biobank and FinnGen to perform a two-sample Mendelian randomization (MR) analysis and genetic correlation analysis, evaluating the causal relationship and genetic correlation between sleep traits (chronotype, getting up in the morning, sleep duration, nap during the day, and insomnia) and urolithiasis (calculus of the kidney and ureter, and calculus of the lower urinary tract). Additionally, multivariable MR (MVMR) analysis adjusted for body mass index (BMI) and other sleep characteristics was conducted to assess the direct impact of sleep traits on the risk of urinary tract stones. The LD score regression (LDSC) analysis indicated a genetic correlation between insomnia and upper urinary tract stones (rg = 0.082, P = 0.017, Adjusted P = 0.085), but no significant genetic correlation was found for other sleep traits. Our results indicated no causal relationship between sleep traits and upper urinary tract stones. However, insomnia was significantly associated with a higher risk of lower urinary tract stones (IVW [inverse variance weighted] OR [odds ratio] = 5.91, 95% CI [confidence interval] 1.52–22.98, P = 0.010, Adjusted P = 0.030), while early rising exhibited a protective effect (IVW OR = 0.29, 95% CI 0.11–0.76, P = 0.012, Adjusted P = 0.030). In the MVMR analysis, insomnia consistently showed a similar trend, whereas daytime napping significantly reduced the risk of lower urinary tract stones (OR = 0.28, 95% CI 0.12–0.65, P = 0.003). This study provides MR-based evidence suggesting that insomnia may increase the risk of lower urinary tract stones, while daytime napping may reduce this risk. No causal relationship was found between sleep characteristics and upper urinary tract stones.

Abstract Background Recurrent observations have indicated the presence of deficits in mismatch negativity (MMN) among schizophrenia. There is evidence suggesting a correlation between increased dopaminergic activity and reduced MMN amplitude, but there is no consensus on whether antipsychotic medications can improve MMN deficit in schizophrenia. Methods We conducted clinical assessments, cognitive function tests, and EEG data collection and analysis on 31 drug-naïve patients with schizophrenia. Comprehensive evaluation tools such as PANSS and MCCB. MMN amplitude was analyzed by event-related potential (ERP) approaches, evoked theta power was analyzed by event-related spectral perturbation (ERSP) approaches. Results Our findings indicate that antipsychotic treatment significantly improved clinical symptoms, as evidenced by reductions in PANSS positive, negative, general symptoms, and total scores (all p 0.05). Conclusion These results suggest that while antipsychotic medications effectively alleviate clinical symptoms, their impact on MMN amplitude and evoke theta power deficit is limited in the short term. Moreover, the amelioration of cognitive impairment in individuals with schizophrenia is not readily discernible, and it cannot be discounted that the enhancement observed in language acquisition and working memory may be attributed to a learning effect. These findings underscore the complexity of the neurobiological mechanisms involved and highlight the need for further research to optimize individualized treatment strategies for schizophrenia. Abstract Introduction Severe acute pancreatitis (SAP) is a crucial gastrointestinal disease characterized by systemic inflammatory responses and persistent multiple organ failure. The role of bile acids (BAs) in diverse inflammatory diseases is increasingly recognized as crucial, but the underlying role of BA conjugation remains elusive. Objectives Our study aim to investigate the potential role of conjugated bile acids in SAP and reveal the molecular mechanisms underlying its regulatory effects. We hypothesized that taurochenodeoxycholic acid (TCDCA) and glycochenodeoxycholic acid (GCDCA) could protect SAP through inhibiting the activation of NLRP3 inflammasomes via the TGR5 pathway in macrophages. Methods To test our hypothesis, we used BA-CoA: amino acid N-acyltransferase knockout (Baat −/− ) mice and established SAP mouse models using caerulein- and sodium taurocholate- induced. We utilized a range of methods, including pathology sections, qRT-PCR, immunofluorescence, Western blotting, and ELISA, to identify the mechanisms of regulation. Results BA-CoA: Amino acid N-acyltransferase knockout (Baat −/− ) mice significantly exacerbated pancreatitis by increasing pancreatic and systemic inflammatory responses and pancreatic damage in SAP mouse models. Moreover, the serum TCDCA levels in Baat −/− mice were lower than those in wild-type (WT) mice with or without SAP, and GCDCA and TCDCA showed stronger anti-inflammatory effects than chenodeoxycholic acid (CDCA) in vitro. TCDCA treatment alleviated SAP in a Takeda G protein-coupled receptor 5 and NOD-like receptor family, pyrin domain containing 3—dependent manner in vivo. Reinforcing our conclusions from the mouse study, clinical SAP patients exhibited decreased serum content of conjugated BAs, especially GCDCA, which was inversely correlated with the severity of systemic inflammatory responses. Conclusion Conjugated bile acids significantly inhibit NLRP3 inflammasome activation by activating TGR5 pathway, thereby alleviating pancreatic immunopathology. The results provide new insights into the variability of clinical outcomes and paves the way for developing more effective therapeutic interventions for AP.

Abstract Background Inhibiting cholesterol metabolism has shown great potential in non-small cell lung cancer (NSCLC). However, the regulatory mechanism of the lipid metabolism key factor Sect. 14-like lipid binding 2 (SEC14L2) in NSCLC remains unclear. This study investigates the effects of differentially expressed genes related to cholesterol metabolism on the development of NSCLC. Methods Cox regression and survival analysis were performed to screen cholesterol metabolism-related genes and predict survival prognosis in NSCLC patients. The proliferation and migration of NSCLC cells were assessed by CCK-8, EdU, colony formation and wound-healing assay. Cholesterol depletion and rescue trials were used to evaluate the effect of SEC14L2 on cholesterol transport in NSCLC cells. IF and Co-IP were used to analyze the targeting relationship between SEC14L2 and scavenger receptor class B member 1 (SCARB1). Results SEC14L2 was a key gene related to prognosis in NSCLC patients and was highly expressed in A549 and Calu-1 cells. Subsequent studies demonstrated that knockdown of SEC14L2 significantly reduced the proliferation and migration of NSCLC cells, resulting in inhibited tumor growth. Furthermore, both in vitro and in vivo experiments indicated that SEC14L2 regulated cholesterol uptake. Silencing SEC14L2 partially counteracted the promotion of cholesterol content by MβCD-chol in A549 and Calu-1 cells. We then verified that there was a protein interaction between SEC14L2 and SCARB1. Conclusion SEC14L2 promoted cholesterol uptake in NSCLC cells by up-regulating SCARB1 expression, thereby promoting NSCLC development.

Abstract Neuronal oscillations refer to rhythmic and periodic fluctuations of electrical activity in the central nervous system that arise from the cellular properties of diverse neuronal populations and their interactions. Specifically, gamma oscillations play a crucial role in governing the connectivity between distinct brain regions, which are essential in perception, motor control, memory, and emotions. In this context, we recapitulate various current stimulation methods to induce gamma entrainment. These methods include sensory stimulation, optogenetic modulation, photobiomodulation, and transcranial electrical or magnetic stimulation. Simultaneously, we explore the association between abnormal gamma oscillations and central nervous system disorders such as Alzheimer’s disease, Parkinson’s disease, stroke, schizophrenia, and autism spectrum disorders. Evidence suggests that gamma entrainment-inducing stimulation methods offer notable neuroprotection, although somewhat controversial. This review comprehensively discusses the functional role of gamma oscillations in higher-order brain activities from both physiological and pathological perspectives, emphasizing gamma entrainment as a potential therapeutic approach for neuropsychiatric disorders. Additionally, we discuss future opportunities and challenges in implementing such strategies.

Abstract Extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue (EMZL) is an indolent B-cell lymphoma that can involve various anatomic sites. EMZL is derived from post-germinal center marginal zone B cells and typically lacks bcl-6 expression. Herein, we report two post-treatment cases of EMZL where unexpected bcl-6 protein expression was observed in specimens obtained following recurrence or progression. This contrasts with the primary specimens, which were negative for the bcl-6. Additionally, we confirm that the altered bcl6 expression observed in relapsed EMZL cases is independent of BCL6 gene rearrangement, as demonstrated by fluorescence in situ hybridization analysis. Relevant literature was reviewed and summarized to enhance the understanding of this phenomenon, particularly for pathologists.

Abstract The ratmouth barbel (Ptychidio jordani) is a critically endangered freshwater fish from the Cyprinidae family, primarily due to overfishing and habitat disruption. To address the challenges of its shrinking wild populations and the difficulties in artificial reproduction, we sequenced, assembled, and annotated a high-quality chromosome-level genome of P. jordani using next-generation short-read sequencing, third-generation long-read sequencing, and Hi-C sequencing. The final genome assembly was 1.14 Gb, consisting of 25 chromosomes with a contig N50 of 25.14 Mb and a scaffold N50 of 42.91 Mb. We identified 25,183 protein-coding genes, 751.75 Mb of repeats, and 19,373 ncRNAs. Methylation loci on most chromosomes ranged from 1,000 to 3,000 per 100 kb window. Gene expression levels across various tissues were analyzed, revealing 12,135 (caudal fin), 11,465 (liver), 14,438 (gill), 12,413 (heart), 8,301 (spleen), and 3,578 (kidney) differentially expressed genes compared to muscle. The comprehensive genomic and transcriptomic resources generated here will aid in understanding the ecology, adaptation, and environmental responses of P. jordani, supporting future research and conservation efforts.

Abstract Background Scutellaria baicalensis Georgi, a traditional Chinese herb, is known for its various biological effects, including antibacterial, anti-inflammatory, antioxidative, and antitumor properties. However, the function and mechanisms of methanol extract of Scutellaria baicalensis Georgi (MESB) in treating hepatic fibrosis remain unclear. Methods This study utilized a CCl4-induced mouse model of hepatic fibrosis to assess the effects of MESB through histopathological analysis and serum tests. The anti-fibrosis mechanism of MESB was investigated using qPCR, Western blotting, RNA interference, proteomics, and metabolomics. Spatial metabolomics identified key components of MESB in liver tissue, while molecular docking determined their targets. Results Treatment with MESB alleviated hepatic pathological changes and reversed hepatic fibrosis in the CCl4-induced models, as evidenced by decreased collagen fibers deposition, reduced expression of hepatic fibrosis markers COL1A1, FN, and PAI-1, and lowered serum levels of AST and ALT. In vitro, MESB inhibited the proliferation of LX-2 cells and the expression of hepatic fibrosis markers. Furthermore, MESB intervention modulated various pathways, particularly those involved in metabolic pathways. Subsequent metabolomics analysis demonstrated that MESB disrupted glycerophospholipid metabolism and suppressed arachidonic acid metabolism. MESB downregulated the expression of cPLA2 in LX-2 cells, leading to decreased production of arachidonic acid and its downstream inflammatory mediators. Meanwhile, MESB inhibited the expression of cPLA2 and its downstream NF-κB pathway in the liver tissues of models induced by CCl4. Additionally, silencing cPLA2 markedly reduced the expressions of COL1A1, FN, and PAI-1. Spatial metabolomics analysis confirmed the penetration of baicalein, wogonin and wogonoside into liver tissue. Further results indicated that baicalein and wogonin inhibited the expression of cPLA2, while baicalin and wogonoside do not exhibit this effect. Moreover, molecular docking suggested that baicalein and wogonin possess the potential to directly interact with cPLA2. Conclusion This study reveals that MESB is crucial in preventing hepatic fibrosis via the cPLA2-mediated arachidonic acid metabolic pathway, highlighting its key active components as potential drugs for fibrosis treatment.

Abstract Sodium butyrate (NaBu), well-known as a histone deacetylase inhibitor and for its capacity to impede cell growth, can enhance the production of a specific protein, such as an antibody, in recombinant Chinese hamster ovary (CHO) cell cultures. In this study, two CHO cell lines, namely K1 and DG44, along with their corresponding mAb-producing lines, K1-Pr and DG44-Pr, were cultivated with or without NaBu. A SWATH-based profiling method was employed to analyze the proteome. Cells cultured in the presence of NaBu exhibited a reduction in mitosis and gene expression, supported by their culture data demonstrating growth inhibition. The presence of NaBu corresponded to upregulation of intracellular trafficking and secretion pathways, aligned with an observed increase in mAb production, and was associated with an elevated glycosylation pathway and a slight alteration in the glycosylation profile of the mAbs. Increased fatty acid oxidation, redox interactions, and lipid biosynthesis were also observed and are likely attributable to the metabolism of NaBu. A comprehensive understanding of the systemic effects of NaBu will facilitate the discovery of strategies to enhance or prolong the productivity of CHO cells. Graphical Abstract

Abstract Background Breastfeeding is the optimal source of nutrition for infants and young children, essential for their healthy growth and development. However, a gap in cohort studies tracking breastfeeding up to six months postpartum may lead caregivers to miss critical intervention opportunities. Methods This study conducted a three-wave prospective cohort analysis to examine maternal breastfeeding trajectories within the first six months postpartum and to develop risk prediction models for each period using advanced machine learning algorithms. Conducted at a leading Maternal and Children's hospital in China from October 2021 to June 2022, data were gathered via self-administered surveys and electronic health records. Results Of the 3307 women recruited, 3175 completed the surveys, yielding a 96% effective response rate. Breastfeeding(BF) rates were observed at 100%, 96%,93% and 83% at discharge, 42 day, 3 month and 6 month postpartum, respectively. Exclusively breastfeeding(EBF) rates were recorded at 91%, 64%,72% and 58% for the same intervals. Among the five machine learning methods employed, Random Forest (RF) demonstrated superior accuracy in predicting breastfeeding patterns, with classification accuracies of 0.629 and an area under the receiver operating characteristic curve (AUC) of 0.8122 at 42 days, 0.925 and an AUC of 0.9800 at 3 months, and 0.836 and an AUC of 0.9463 at 6 months postpartum, respectively. Key predictive factors for breastfeeding at 42 days postpartum included the newborn’s birth weight and the mother’s pre-delivery and prenatal weights. Predictors for feeding type at 3 months and 6 months postpartum included early feeding types and the scores from the Breastfeeding Self-Efficacy Scale-short Form (BSES-SF) at 6 months. The predictive model based on follow-up data showed strong performance. Conclusion Breastfeeding rates slightly declined from discharge to 6 months postpartum. The breastfeeding context in this region is comparatively optimistic both within China and internationally. Factors such as newborn’s birth weight, gestational age, maternal weight management before and during pregnancy, early support and breastfeeding success, breastfeeding knowledge and self-efficacy are intricately linked to long-term breastfeeding outcomes. This study highlights critical, modifiable risk factors for early breastfeeding stages, providing valuable insights for enhancing breastfeeding intervention programs and informed decision-making.

Abstract Background Compassion fatigue impacts nurses’ well-being and work efficiency. Extensive research has explored its prevalence, but evidence regarding related factors is broadly categorized and lacks descriptive data. There’s also a lack of systematic reviews on compassion fatigue among nursing students during internships. Objectives To synthesize evidence on the levels and factors of compassion fatigue among nurses and nursing students during internships. Methods This systematic review is registered with PROSPERO (ID CRD42023444173). Literature searches were conducted in five databases (Cochrane Library, PubMed, EMBASE, CNKI, and WanFang) up to November 30, 2022, with updates planned until January 17, 2024, if necessary. Inclusion criteria covered studies reporting data on the prevalence and related factors of compassion fatigue or its dimensions, burnout and secondary traumatic stress, among registered nurses or nursing students during internships. Independent study selection, quality assessment, and data extraction were performed. The Agency for Healthcare Research and Quality tool was used for critical appraisal of study quality. Random-effects model analyses were conducted using Stata 17.0 to pool data on prevalence rates and mean scores of compassion fatigue. When comprehensive data on compassion fatigue were unavailable, its dimensions were analyzed for both prevalence and mean scores. Results Our review included 196 studies (73,034 nurses and 4,551 nursing students). For nurses, pooled mean scores for burnout and secondary traumatic stress were 26.81 (95% CI 26.28 to 27.35) and 25.88 (95% CI 25.39 to 26.37), respectively. For nursing students during internships, pooled mean scores were 29.16 (95% CI 26.95 to 31.37) and 25.64 (95% CI 20.95 to 30.34), respectively. Subgroup analyses revealed that post-COVID-19 pandemic, nurses exhibited higher compassion fatigue, especially in ICU or emergency departments. Evidence from 93 studies suggested that nurses’ burnout and secondary traumatic stress are both influenced by work environment, social support, job satisfaction, workload, and psychological capital (moderate to low-certainty evidence). For nursing students, psychological capital plays a significant role (moderate to low-certainty evidence). Conclusions Both registered nurses and nursing students experience moderate compassion fatigue. Work environment, social support, job satisfaction, workload, and psychological capital are key factors associated with burnout and secondary traumatic stress in registered nurses. For nursing students, psychological capital plays a similarly significant role. Given that this is the first systematic review to explore these factors among nursing students, further research is essential to deepen understanding and develop effective interventions.

ABSTRACT Background Cancer cachexia can impact prognosis, cause resistance to anticancer treatments and affect the tolerability of treatments. This study aims to identify hepatocellular carcinoma (HCC) with cachexia by characterizing longitudinal body composition (BC) trajectories. Methods This longitudinal, multicentre cohort study included unresectable HCC patients treated with first‐line programmed death‐(ligand)1 inhibitors plus anti‐vascular endothelial growth factor antibody/tyrosine kinase inhibitors between 01/2018–12/2022. BC measurements including skeletal muscle mass (SMM) and total adipose tissue area (TATA) were evaluated by computed tomography at the third lumbar vertebra at baseline and follow‐up imaging. Unsupervised latent class growth mixed models were applied to distinguish potential longitudinal SMM and TATA trajectories for identifying cachexia. The primary study endpoint was overall survival (OS), with secondary endpoints including progression‐free survival (PFS), objective response rate (ORR) and safety. Multiple Cox proportional hazards models were used to calculate adjusted hazard ratios (HRs) for survival. Results A total of 411 patients with 2138 time‐point measurements were included. The median age was 56 years, and 50 (12.2%) patients were female. Two distinct trajectories were identified for SMM and TATA: sharp‐falling and stable. In order to obtain the dynamic stress response of the drum and main shaft of a friction hoist under dynamic loads, a dynamic stress calculation model for the friction hoist was constructed based on the flexible multibody modelling method. This approach integrates modal flexible bodies and finite element flexible bodies to visualize the kinematic and dynamic characteristics of the friction hoist. The fatigue load spectrum of the main shaft and drum of the friction hoist was obtained, addressing the structural strength analysis and dynamic stress response calculation issues under dynamic loads. This method can be used as an alternative to traditional constant amplitude design load applications for fatigue strength assessment.

Rock masses in high-elevation or cold regions are vulnerable to the combined effects of freeze-thaw (F-T) weathering and dynamic mixed-mode loading, posing a serious threaten to the safety and stability of geotechnical engineering. In this study, a series of dynamic fracture tests were conducted on notched semi-circular bend (NSCB) granite specimens subjected to different mixed-mode loading and F-T cycles using a split Hopkinson pressure bar (SHPB) test system. The effects of F-T treatment and dynamic mixed-mode loading on the fracture properties of granite, including effective fracture toughness, progressive fracture process, and macroscopic morphology of fracture surface, were comprehensively revealed. The experimental results suggest that the dynamic effective fracture toughness of NSCB specimens is dependent on the loading rate, particularly when the mode I loading is dominant. Additionally, the fracture toughness decreases as the number of F-T cycles increases, with an inflection point at 30 F-T cycles. All granite specimens subjected to mixed-mode loading exhibit a curved fracture path, with faster crack propagation speed and more fine cracks in specimens exposed to higher F-T cycles. Macroscopic morphology of fracture surface obtained using three-dimensional (3D) scanning indicates that the fractal dimension of the fracture surface increases with increasing F-T cycles, and the increment is more pronounced for specimens subjected to a higher mode II loading component. Moreover, this study compared the fracture resistance of F-T treated granite subjected to dynamic mixed loading using the maximum tangential stress (MTS) criterion and the generalized maximum tangential stress-based semi-analytical (SA-GMTS) criterion. Compared with the MTS criterion, the SA-GMTS criterion shows a more reasonable consistency with the experimental results, with a root mean square error within ±7%.

Background: S100A8 is a member of the S100 protein family and plays a pivotal role in regulating inflammation and tumor progression. This study aimed to comprehensively assess the expression patterns and functional roles of S100A8 in glioma progression. Methods: Glioma tissues were collected from 98 patients who underwent surgical treatment at Fudan University Shanghai Cancer Center. S100A8 expression in glioma tissues was analyzed using immunohistochemistry (IHC) to establish its correlation with clinicopathological features in patients. The expression and prognostic effect of S100A8 in glioma were analyzed using TCGA and CGGA public databases. Then, we investigated the role of S100A8 in glioma through a series of in vivo and in vitro experiments including Transwell, wound healing, CCK8, and intracranial tumor models. Subsequently, bioinformatics analysis, single-cell sequencing and coimmunoprecipitation (Co-IP) were used to explore the underlying mechanism. Results: S100A8 was upregulated in gliomas compared to paracancerous tissues, and this phenotype was significantly correlated with poor prognosis. Subgroup analysis showed that S100A8 expression was higher in the high-grade glioma (HGG) group than that in the low-grade glioma (LGG) group. S100A8 overexpression in glioma cell lines promoted cell proliferation, migration and invasion, while silencing S100A8 reversed these effects. In vivo experiments showed that S100A8 knockdown can significantly reduce the tumor burden of glioma cells. Notably, S100A8 was observed to stimulate microglial M2 polarization by interacting with TLR4, which subsequently induced NF-κB signaling and IL-10 secretion within the tumor microenvironment. Conclusions: S100A8 promotes tumor progression by inducing phenotypic polarization of microglia through the TLR4/IL-10 signaling pathway in glioma. It might represent a therapeutic target for further basic research or clinical management of glioma.

Abstract Background Acute myeloid leukemia (AML) is characterized by abundant immature myeloid cells, relapse and refractory due to leukemia stem cells (LSCs). Bone marrow mesenchymal stem/ stromal cells (BMSCs) supported LSCs survival, meanwhile, chemotherapy improved connexin43 (CX43) expression. CX43, as the most intercellular gap junction, facilitated transmit mitochondria from BMSCs into AML. We hypothesized that increased mitochondria transferred from BMSCs supported metabolic remodeling in LSCs to sustain their stemness. Methods Primary BMSCs from AML patients were isolated. CX43-BMSCs, overexpressing CX43, were cocultured with KG-1a cells. Fluorescence and confocal microscopy observed mitochondrial transfer. Flow cytometry, EdU assay, and clonogenicity evaluated cell cycle, proliferation, and clonogenic potential. Xenograft mouse models were used to evaluate the tumorigenicity of KG-1a in vivo. Seahorse, RNA-seq, and LC-MS assessed mitochondrial function, transcriptomes, and metabolites post-coculture. Results CX43-BMSCs promoted unidirectional mitochondrial transfer, enhancing KG-1a adhesion and proliferation to maintain LSCs stemness in vitro and vivo. RNA-seq revealed coculture with CX43-BMSCs upregulated genes related to adhesion, proliferation, and migration in KG-1a cells. Elevated CX43 expression strengthened BMSCs-KG-1a interaction, facilitating mitochondrial transfer and nucleoside metabolism, fueling KG-1a cells. This enhanced mitochondrial energy metabolism, promoting metabolic reprogramming and clonogenicity. Conclusion CX43-mediated mitochondrial transfer from BMSCs to KG-1a enhances LSCs adhesion, proliferation, clonogenicity, and metabolic reprogramming. CX43 emerges as a potential therapeutic target for AML by sustaining LSCs stemness through metabolic remodeling.

Abstract Background The 12-oxo-phytodienoic acid reductase (OPR) enzyme is crucial for the synthesis of jasmonates (JAs), and is involved in the plant stress response. However, the OPR gene family in sweetpotato, an important horticultural crop, remains unidentified. Results In this study, we employed bioinformatics techniques to identify nine IbOPR genes. Phylogenetic analysis revealed that these genes could be divided into Group I and Group II. Synteny analysis indicated that IbOPR evolution was driven by tandem duplication, whole-genome duplication (WGD), and segmental duplication events. The promoter sequences of IbOPRs were found to be associated with stress and hormonal responses. Additionally, we successfully cloned four IbOPRs from "Haida HD7791" and "Haida HD7798" using homologous cloning technology. These sequences were 1203 bp, 1200 bp, 1134 bp, and 1137 bp in length and encoded 400, 399, 377, and 378 amino acids, respectively. The protein sequence similarity between the salt-tolerant variety "Haida HD7791" and the salt-sensitive variety "Haida HD7798" was determined to be 96.75% for IbOPR2, 99.75% for IbOPR3, 92.06% for IbOPR6, and 98.68% for IbOPR7. Phylogenetic analysis categorized IbOPR2 and IbOPR3 proteins into Group II, while IbOPR6 and IbOPR7 proteins belonged to Group I. Subcellular localization experiments showed IbOPR2 protein present in the peroxisome, while IbOPR3, IbOPR6, and IbOPR7 proteins were found in the cytoplasm and nucleus. Salt stress induction experiments demonstrated that IbOPR2, IbOPR3, and IbOPR7 were significantly upregulated only in 'Haida HD7791' after 6 h. In contrast, IbOPR6 was induced in 'Haida HD7798' at 6 h but inhibited in 'Haida HD7791' at later time points (12, 24, 48, and 72 h), highlighting functional differences in salt stress responses. Conclusions Our findings suggest that IbOPR2 may play a crucial role in sweetpotato's response to salt stress by participating in JAs synthesis. These results provide a foundation for future functional analyses of OPR genes in sweetpotato.

Abstract The meticulous design of advanced electrocatalysts and their integration into gas diffusion electrode (GDE) architectures is emerging as a prominent research paradigm in the H2O2 electrosynthesis community. However, it remains perplexing that electrocatalysts and assembled GDE frequently exhibit substantial discrepancies in H2O2 selectivity during bulk electrolysis. Here, we elucidate the pivotal role of mass transfer behavior of key species (including reactants and products) beyond the intrinsic properties of the electrocatalyst in dictating electrode-scale H2O2 selectivity. This tendency becomes more pronounced in high reaction rate (current density) regimes where transport limitations are intensified. By utilizing diffusion-related parameters (DRP) of GDEs (i.e., wettability and catalyst layer thickness) as probe factors, we employ both short- and long-term electrolysis in conjunction with in-situ electrochemical reflection-absorption imaging and theoretical calculations to thoroughly investigate the impact of DRP and DRP-controlled local microenvironments on O2 and H2O2 mass transfer. The mechanistic origins of diffusion-dependent conversion selectivity at the electrode scale are unveiled accordingly. The fundamental insights gained from this study underscore the necessity of architectural innovations for mainstream hydrophobic GDEs that can synchronously optimize mass transfer of reactants and products, paving the way for next-generation GDEs in gas-consuming electroreduction scenarios.

Evaluating the recognizability of street built environments provides crucial support for urban planning, security monitoring and navigation. Although street view images (SVIs) are widely used in urban studies, it overlooks the interconnection among different locations, which can also affect perceptions about environmental recognizability. To address this issue, this study proposes a deep learning-based model called RB-Node, which comprehensively integrates spatial structural features in a road network view and visual features from SVIs, achieving 82.56% accuracy. It appears that image information from visual features dominates environmental recognizability. Additionally, structural information contributes significantly to the accurate classification of nodes and waterfront promenade areas. Moreover, scene-text information, a subset of visual features, helps classify commercial and historical areas. Furthermore, 1056 samples were collected through an eye-tracking experiment to validate the recognizability evaluation results, as well as compare the decision-making process between humans and RB-Node. According to the results, RB-Node behaviour and human observed behavior follow similar patterns, although human perceptions tend to be more holistic than RB-Node's. This study contributes to a better understanding of environmental recognizability and provides targeted recommendations for urban renewal.

The experiment sought to determine the effects of substituting Flammulina velutipes residue (FVR) for corn on growth performance, serum biochemical parameters, slaughter performance and caecum microbiota of geese. One hundred and ninety-two 35old Hordobagy geese were used in this experiment, and they were divided into four groups and fed with various diets for 28 d. The geese in the control group were fed with a basal diet. The other groups substituted 12%, 24% and 32% FVR for corn in their diets, respectively. Loess has unique physical, hydrodynamic and mechanical properties, which are influenced by both internal and external geological processes, as well as human engineering activities. Consequently, surface disasters are especially prevalent in the Loess region of China. The study area is situated in the middle part of Lüliang Mountain in the middle part of the Loess Plateau, which is characterised by a typical loess landform with a complex system of gullies and hill ridges. According to current theory, the cracking boundary of the goaf profile is a straight line. However, these surface disasters are actually caused by the shear action of the deep rock layer and the original vertical joint structure of the loess. By analysing the cracking process of the ‘Goaf-Overburden-Loess’ section in the study area, it can be found that the boundary of the movement basin section presents a broken line shape, which has important implications for the accurate estimation of the area affected by loess-type surface subsidence.

Background: Following recent research advancements, an increasing level of evidence had been published to indicate that celastrol exerted a therapeutic effect on a range of nervous system diseases. This study therefore aimed to investigate the potential involvement of celastrol on ferroptosis and the blood-brain barrier disruption in intracerebral haemorrhage. Methods: We established a rat intracerebral haemorrhage and adrenal pheochromocytoma cell (PC12) OxyHb models using an ACSL4 overexpression vector. Ferroptosis-related indices were assessed using corresponding assay kits, and immunofluorescence and flow cytometry were used to measure reactive oxygen species (ROS) levels. Additionally, quantitative PCR (qPCR) and western blot analyses were conducted to evaluate the expression of key proteins and elucidate the role of celastrol in intracerebral haemorrhage (ICH). Results: Celastrol significantly improved neurological function scores, blood-brain barrier integrity, and brain water content in rats with ICH. Moreover, subsequent analysis of ferroptosis-related markers, such as Fe2+, ROS, MDA, and SOD, suggested that celastrol exerted a protective effect against the oxidative damage induced by ferroptosis in ICH rats and cells. Furthermore, Western blotting indicated that celastrol attenuated ferroptosis by modulating the expression levels of key proteins, including acyl-CoA synthetase long-chain family member 4 (ACSL4), glutathione peroxidase 4 (GPX4), ferritin heavy chain 1 (FTH1), and anti-transferrin receptor 1 (TFR1) both in vitro and in vivo. ACSL4 overexpression attenuated the neuroprotective effects of celastrol on ICH in vitro. Molecular docking analysis revealed that celastrol interacted with ACSL4 via the GLU107, GLN109, ASN111, and LYS357 binding sites. Conclusions: Celastrol exerted antioxidant properties and aids in neurological recovery after stroke by suppressing ACSL4 expression during ferroptosis. As such, this drug represented a promising pharmaceutical candidate for the treatment of ICH.

The precious metal based multi-principal elements alloy (MPEA) PdAgCuAuPtZn is mainly used in electrical contact applications, as a new type of multi-component alloy. However, there is a lack of detailed information about the precipitation evolution and its effect on the creep properties of the PdAgCuAuPtZn alloys. Combining nanoindentation and detailed electron microscopy analyses, we investigated the microstructural evolution and creep mechanism of the Pd–30Ag–14Cu–10Au–10Pt–1Zn alloy during aging treatment systematically. Microstructural analyses show that discontinuous precipitation (DP) with Cu-rich and Ag-rich phases is observed in aging-treated samples. As the aging temperature increases, DP gradually replaces the matrix and coarsens. Meanwhile, the grain size gradually decreases because of the existence of DP. Spinodal decomposition (SD) and chemical short-range order (CSRO) can be observed in the matrix and CSRO can be observed in the Cu-rich phase. Nanoindentation results show that when the aging temperature is below 500 °C, the matrix and DP hardness gradually increase, from 4.06 GPa to 7.42 GPa and 3.78 GPa–5.29 GPa, respectively, and gradually decreases at temperatures higher than 500 °C. Based on the creep displacement analysis, the increasing temperature contributes to improving the creep resistance and DP has higher creep resistance than matrix. The results of the creep stress exponent (n) and activation volume (V) also indicate that the creep mechanism is dominated by dislocation movement. Both CSRO and SD contribute to improving creep resistance. These results will be beneficial to both deeply understanding the creep behavior of PdAgCuAuPtZn alloys and designing high-performance precious metal multi-component alloys.

Objective and methods In this study, the hydrochemical method and self-organized neural network (SOM-KM) coupling method were employed to identify recharge sources and reveal the water-rich mechanism in the karst groundwater-rich area of the Changxiao karst water system in Jinan City. The contribution ratio of karst groundwater recharge sources in the karst groundwater-rich area was quantitatively calculated using the end-element mixed model. The enrichment mechanism of karst groundwater is explored by combining with topography, geological structure, stratigraphic lithology, and catchment conditions. Results The results showed that the karst groundwater in the catchment drainage area had similar water chemistry to that in the southern recharge area, the karst groundwater in the lateral runoff area, and the Yellow River, indicating a close hydraulic connection. This implies that the karst groundwater in the catchment drainage area is recharged by three sources: The southern mountain area, the karst groundwater in the lateral runoff area, and the Yellow River. The contribution ratios of the three components are 75.09%, 21.02%, and 3.89%, respectively. Carbonate rocks are widely distributed, and fissured karst is well developed in the accumulation and discharge areas, especially in the Maji-Xiaoli-Guide area. Moreover, there are abundant karst groundwater recharge sources in this area. During the runoff process of karst groundwater from southeast to northwest, it is impeded by sandstone and mudstone in the north. As a result, it accumulates in the contact zone between soluble rock and insoluble rock, thus forming impeded-type karst groundwater-rich structures. Conclusion Revealing the enrichment mechanism of karst groundwater in the Changxiao karst water system can provide scientific support for accurate calculations of recoverable resources and the protection of the springs in Jinan.

BackgroundThe threshold limit values (TLVs) established and regularly updated by the American Conference of Governmental Industrial Hygienists (ACGIH) are widely adopted and referenced globally, serving as a crucial reference for China's occupational exposure limits (OELs). It is necessary to track it regularly and compare it with China's OELs. ObjectiveTo compare the OELs stipulated in Occupational exposure limits for hazardous agents in the workplace—Part 1: Chemical hazardous agents (GBZ 2.1—2019) and the ACGIH TLVs (2024) and to provide references for subsequent formulation and revision of OELs in China. MethodsThe OELs specified in GBZ 2.1—2019 and the TLVs issued by ACGIH were used to establish a database using Microsoft Excel 2019 software. Cross verification was conducted through matching Chemical Abstracts Service Registry Numbers (CAS Rn) and both Chinese and English names to ensure accuracy. Then, comparisons and analyses were carried out based on the type of limit values, which were matched as follows: permissible concentration-time weighted average (PC-TWA) with threshold limit value-time weighted average (TLV-TWA), permissible concentration-short term exposure limit (PC-STEL) with threshold limit value-short term exposure limit (TLV-STEL), and maximum allowable concentration (MAC) with threshold limit value-ceiling (TLV-C). Comparisons included types, quantities, and sizes of limits. ResultsThe GBZ 2.1—2019 OELs and the ACGIH TLVs (2024) were generally consistent in terms of types and definitions, but there were differences in the number and size of the limits. In terms of the number of limits, GBZ 2.1—2019 specified 365 OELs for 358 chemical hazardous agents, while ACGIH TLVs (2024) included 316 corresponding limits. Among these, 148 (46.9%) limits were consistent, 38 (12.0%) were basically consistent, and 130 (41.1%) were inconsistent. In terms of the size of the limits, out of the 130 inconsistent limits, 51 OELs were lower than the corresponding TLVs, 67 OELs were higher than the corresponding TLVs, and 12 were under different limit types. For some chemical hazardous agents, their OELs were significantly lower or higher than their TLVs. ConclusionSome of the OELs for chemical hazardous agents specified in GBZ 2.1—2019 are significantly lower or higher than the TLVs. For these chemical hazardous factors, it is recommended to prioritize their inclusion in research projects and to complete the revisions as soon as possible based on the latest scientific evidence.

Austenitization is significant for understanding the microstructure and residual stress evolution in additive manufacturing of non-austenitic steels. However, the accurate modeling of austenite transformation of additively manufactured parts is rarely reported. In this work, a new kinetic model is proposed to describe the diffusional austenitization of additively manufactured 17-4 PH martensitic stainless steel. The proposed kinetic model is based on the classic Johnson-Mehl-Avrami-Kolmogorov (JMAK) theory, and incorporates a new austenite grain growth model that accounts for the maximum austenite grain size and the effective driving force for growth. Experimental results obtained through dilatometry show that the proposed kinetic model is able to fit and predict the austenite transformation curves across a wide range of heating rates. This model accurately predicts the saturated austenite fraction by maximum austenite grain size and facilitates the understanding of the effect of heating rate on diffusional austenite transformation behavior. The findings imply that the drag force restricting the maximum austenite grain size originates from the initial martensitic microstructure. The transition from the diffusional austenitization to massive or displacive phase transformation at the heating rate of 100 °C/s is identified for additively manufactured 17-4 PH martensitic stainless steel.

Abstract Background Klebsiella pneumoniae has emerged as a significant pathogen that causes community and hospital infections due to its high resistance rate and transmissibility. Klebsiella pneumoniae complex is classified into three phylogroups: Klebsiella pneumoniae (KpI), Klebsiella quasipneumoniae (KpII), and Klebsiella variicola (KpIII) in classical taxonomy, but KpII and KpIII are infrequently isolated clinically. Although pathogenic KpII has been reported worldwide, the understanding of this pathogen remains limited. Methods Whole-genome sequencing (WGS) of K. quasipneumoniae subsp. similipneumoniae ACESH00366hy was performed. Plasmid characterization was demonstrated using S1-PFGE, Southern blotting, and conjugation assays. Antimicrobial susceptibility testing was performed and interpreted according to CLSI, EUCAST, and FDA standards. The virulence of the strain was assessed using the Galleria mellonella infection assay, serum-killing assay, and biofilm formation assay. A phylogenetic tree was constructed to explore its biological evolution. Results The K. quasipneumoniae subsp. similipneumoniae isolate ACESH00366hy, belonging to ST2012 and KL139, contains several resistance genes including bla NDM−1, bla SHV−12, bla OKP−B−2, and oqxAB, as well as various virulence genes including iroE, iutA, mrkABCDFHIJ, entABCDEFS, and fepABCDG. The bla NDM−1 and bla SHV−12 genes were present on the 53,624 bp IncX3 plasmid. Virulence assays showed that the virulence of ACESH00366hy was greater than that of the low-toxicity strain ATCC 700,603. Phylogenetic analysis demonstrated the emergence of ST2012 KpII-B in Asia and revealed the spread of K. quasipneumoniae subsp. similipneumoniae in humans, animals, and the environment. Conclusion This study highlights the emergence of clinical ST2012 KpII strains harbouring bla NDM−1 via the IncX3 plasmid in China. We evaluated its resistance and virulence characteristics and performed phylogenetic analysis, thereby enhancing our understanding of its resistance and pathogenicity and contributing to the clinical surveillance of K. quasipneumoniae.

Abstract Colorectal cancer (CRC) patients with mismatch repair (MMR)-deficient (dMMR) but not MMR-proficient (pMMR) tend to benefit from immune checkpoint blockade (ICB) therapy. To profile the tumor microenvironments (TME) underlying these varied therapeutic responses, we integrate spatial enhanced resolution omics-sequencing (Stereo-seq), single-cell RNA sequencing, and multiplexed imaging analysis to create high-definition spatial maps of tumors from treatment-naïve and ICB-treated CRC patients. Our results identify the spatial organization and immune status of the tumor-stroma boundary as a distinctive feature of dMMR and pMMR CRCs, which associates with ICB response. The physical interactions and abundance of LAMP3 +DCs and CXCL13 +T cells may shape the ICB-responsive tumor-stroma boundary, whereas CXCL14 +cancer-associated fibroblasts tend to remodel extracellular matrix to form a structural barrier in non-responders. Our work therefore points out the importance of the molecular and cellular spatial structures of tumors in ICB response, raising the possibility of reprogramming tumor-stroma boundary for sensitizing immunotherapies in the majority of CRCs.

Abstract Background Uncertainty in speech perception and emotional disturbances are intertwined with psychiatric symptoms. How prosody embedded in target speech affects speech-in-noise recognition (SR) and is related to psychiatric symptoms in patients with schizophrenia (SCHs) remains unclear. This study aimed to examine the neural substrates of prosodic SR deficits and their associations with psychiatric symptom dimensions in patients with schizophrenia. Methods Fifty-four SCHs and 59 healthy control participants (HCs) completed the SR task (participants were required to identify the contents of the target pseudo-sentences expressed in neutral, happy, sad, angry, fearful, and disgusted prosody by actors), positive and negative syndrome scale (PANSS) assessment, and magnetic resonance imaging scanning. We examined the deficits of the six prosodic SRs in schizophrenia patients and their associations with brain gray matter volume (GMV) and psychiatric symptoms. Results Negative emotional (sad, angry, fearful, and disgusted) prosodies of the target sentences worsened SR across groups. Both participant groups had equal SR between the neutral and happy prosody conditions. Compared to the anger and disgusted conditions, SCHs and HCs had better SR under sad and fearful conditions. Better prosodic SR was associated with shorter duration and lower local shimmer of target sentences. A partial least squares (PLS) component of GMV (covering 47 brain regions with group differences) was associated with six prosodic SRs. The happy SR was associated with the PANSS total, negative, and general scores, adjusting for covariates. Conclusions Negative emotional prosodies of the target sentences dampened the recognition of the target sentences. The prosodic SR abnormalities in SCHs were associated with not only brain GMV reductions in the regions involved in the processing of sensorimotor, speech, and emotion but also with negative and general psychiatric symptoms. These findings suggest the possibility of improving negative symptoms by improving a happy SR in schizophrenia patients based on neuroplasticity.

Abstract Background Orthodontic treatment carries potential risks and complications, especially if the path of tooth movement extends beyond the alveolar bone or if the patient has a thin bone plate. This amplifies the patient’s risk of complications such as bone fenestration and bone dehiscence. Hence, orthodontists should consider the patient’s periodontal and dental condition during treatment and opt for a multidisciplinary approach when necessary to achieve optimal results. Case presentation This case report describes a successfully treated case of Class III malocclusion using an adjuvant therapy known as periodontally accelerated osteogenic orthodontics (PAOO). A 25-year-old man sought treatment primarily due to an evident anterior negative overjet. The patient was diagnosed with a skeletal Class III relationship and mandibular prognathism. A clinical examination revealed a severe Class III molar relationship and an anterior negative overjet. Moreover, cone-beam computed tomography revealed a thin labial bone plate in relation to the mandibular anterior teeth. The molar relation was corrected through molar distalization using miniscrews, and the mandibular incisors were retracted in conjunction with the PAOO procedure. Post-treatment outcomes demonstrated improved occlusal relationship, thickened labial bone plate of the mandibular anterior teeth, and absence of bone fenestration or dehiscence. Conclusions This clinical case shows that orthodontic treatment requires a multidisciplinary approach. The favorable results indicate that PAOO is effective in maintaining alveolar bone thickness and reducing the risk of periodontal complications.
