Missing data can significantly influence results of epidemiological studies. The National Health and Nutrition Examination Survey (NHANES) is a popular epidemiological dataset. We examined recent practices related to the prevalence and the reporting of the amount of missing data, the underlying mechanisms, and the methods used for handling missing data in recent doctoral dissertations and subsequent publications using NHANES data as a case study. We also explored missing data handling coverage in top-selling applied statistical textbooks. Thirty-seven doctoral dissertations (published from 2007 to 2017) and 17 subsequent journal articles were included in the analysis. Overall, 29 (78.4%) dissertations did not explicitly state whether they had missing data. Five (62.5%) dissertations reporting missing data did not report an assumed mechanism of missingness. Only one subsequent journal article reported the missing data percent for key variables. 28 (75.7%) dissertations and 16 (94.1%) journal articles reported the use of NHANES sample weights. Of the top-selling 20 applied statistics/biostatistics textbooks examined, 14 did not mention imputation. This sample reflects poor rigor in analysis, reporting, and handling of missing data found among recent graduates and poor coverage in textbooks. Simple Summary: With the increasing demand for aquaculture, the resource of high-quality traditional protein (such as fishmeal) becomes limited, and plant ingredients are often used as substitutes for fishmeal because of their low price and wide availability. However, plant proteins are usually deficient in certain essential amino acids (EAAs), which adversely affects the growth performance of aquatic animals. Lysine is generally the first limiting amino acid in plant protein ingredients used to formulate the aquafeed, and it plays an important role in physiological processes including antioxidant, differentiation, growth, immunity, and reduction of nitrogen emission. In this study, the effects of dietary graded L-lysine concentrations on the growth, proximate composition, and AA (amino acid) profile of coho salmon alevins were evaluated by a feeding trial. The results showed that an adequate dietary lysine level significantly improved the specific growth rate (SGR), protein efficiency ratio (PER), body protein deposition (BPD), and feed conversion ratio (FCR). The dietary optimum L-lysine requirements for coho salmon alevins were 3.74%, 3.73%, 3.91%, and 3.77% of the diet or 6.80%, 6.78%, 7.11%, and 6.85% of dietary proteins against the SGR, PER, BPD, and FCR, respectively. The suitable dietary L-lysine concentration for coho salmon (Oncorhynchus kisutch) alevins was assessed by a dose response feeding trial. Six experimental diets were made with graded L-lysine concentrations of 2.29%, 2.81%, 3.32%, 3.80%, 4.27%, and 4.78% of the dry matter, respectively, each of which was fed to triplicate groups of 100 alevins (initial body weight: 0.30 ± 0.01 g) in 18 plastic baskets (water volume 240 L). The alevins were cultured in a flowing freshwater system and fed manually to apparent satiation four times a day for 12 weeks. The survival rate of alevins did not differ significantly among the dietary groups. The specific growth rate (SGR), protein efficiency ratio (PER), and body protein deposition (BPD) increased significantly (p < 0.05) with the increase in dietary lysine concentration up to 3.80% and then reduced as lysine level further increased. The feed conversion ratio (FCR) had an inverse trend to SGR. The whole-body crude protein content of the alevins increased significantly with increasing dietary lysine level, while crude lipid content showed the opposite trend. In comparison, the contents of morphological indices, whole-body moisture, and ash were not affected significantly (p > 0.05) by the different dietary lysine concentrations. The highest contents of lysine, arginine, and total essential amino acids (EAAs) were observed in the group with 4.27% dietary lysine concentration, which did not differ significantly from those in the 3.32%, 3.80%, and 4.78% groups but was significantly higher than those in the 2.29% and 2.81% groups. Similarly, valine had the highest content in the group with 4.78%. The variations in dietary lysine had no significant impacts on other EAA and non-EAA contents except glycine, which increased with increasing dietary lysine level. Simple Summary: As an excellent but most expensive source of protein, fish meal (FM) is commonly used in 30–60% of the aquafeed for salmon fish species including coho salmon. However, the high cost and unstable supply of FM led it no longer to be considered as a sustainable protein source for aquafeeds. Accordingly, alternative protein sources with low cost and high availability have become a research hotspot in aquafeed for carnivorous species. Poultry by-product meal (PBPM), one of the rendered animal protein sources, can be used as protein substitute of FM in aquafeed due to its economical availability and high protein content and growth promotion effects in comparison to plant proteins. Until now, little information has been available on evaluating the effects of replacing FM by animal proteins including PBPM for coho salmon. In this study, the effects of PBPM instead of FM on growth, muscle composition, and tissue biochemical indexes of coho salmon were investigated. The results showed that the growth performance, feed utilization, muscle composition, serum biochemical indices, and liver antioxidant enzyme activities of coho salmon were negatively affected by high dietary inclusion level of PBPMs, and the optimum substitution level was evaluated based on the specific growth rate (SGR). The present study evaluated the effects of partially substituting fish meal (FM) with poultry by-product meal (PBPM) on the growth, muscle composition, and tissue biochemical parameters of coho salmon (Oncorhynchus kisutch) post-smolts. Five isonitrogenous (7.45% nitrogen) and isoenergetic (18.61 MJ/kg gross energy) experimental diets were made by substituting 0%, 10%, 20%, 40%, and 60% FM protein with PBPM protein, which were designated accordingly as PBPM0 (the control), PBPM10, PBPM20, PBPM40, and PBPM60, respectively. Each diet was fed to triplicates of ten post-smolts (initial individual body weight, 180.13 ± 1.32 g) in three floating cages three times daily (6:50, 11:50, and 16:50) to apparent satiation for 84 days. Both specific growth rate (SGR) and protein efficiency ratio did not differ significantly (p > 0.05) among the control, PBPM10, and PBPM20 groups, which were remarkably (p < 0.05) higher than those of the PBPM40 and PBPM60 groups. Feed conversion ratio varied inversely with SGR. The PBPM replacement had no remarkable effects on the morphological indices and proximal muscle components. The control and PBPM10 groups led to significantly higher muscle contents of leucine, lysine, and methionine than groups of higher PBPM inclusion. The groups of PBPM40 and PBPM60 obtained significantly (p < 0.05) higher serum alanine aminotransferase and aspartate aminotransferase activities than the control and low PBPM inclusion groups. The control group had significantly higher albumin and total cholesterol contents than the groups with PBPM inclusion. The control group had significantly higher triglycerides content than the PBPM60 group. The PBPM60 group had significantly lower contents of high-density lipoprotein, low-density lipoprotein, and total protein than the control and PBPM10 groups. The high PBPM replacement level up to 40% and 60% had adverse effects on hepatic malondialdehyde levels. The catalase and superoxide dismutase activities were not affected by low PBPM inclusion, but significantly decreased in high-PBPM-inclusion groups. The present study evaluated the effects of supplemental dietary α-linolenic acid (ALA, 18:3n-3) on growth performance, body composition, hepatic fat metabolizing enzymes, and fatty acid profiles of coho salmon (Oncorhynchus kisutch) alevins cultured in freshwater. Six isonitrogenous and isolipidic experimental diets were formulated to attain different levels of ALA (0.09, 0.41, 0.76, 1.03, 1.32, and 1.68% dry weight) by adding linseed oil in a basal diet, respectively. Each diet was fed to triplicate groups of alevins (50 fish in each tank) with initial mean body weight of 0.352 ± 0.002 g. Fish were reared in a freshwater flow-through rearing system and fed to apparent satiation 4 times daily for 12 weeks. The specific growth rate (SGR) increased with increasing dietary ALA level, which reached the peak at 1.03% ALA group (p < 0.05). The final body weight (FBW) and SGR showed linear, quadratic, and cubic responses with the dietary ALA level. The whole-body crude protein increased with increasing dietary ALA level while lipid content was the opposite trend (p < 0.05). However, the content of crude lipid in the whole-body of fish showed quadratic and cubic responses with the dietary ALA level. In addition, except for fatty acid synthase (FAS), the hepatic enzymes activities of lipoprotein lipase (LPL), hepatic lipase (HL), and malate dehydrogenase (MDH) showed linear, quadratic, and cubic response with the dietary ALA level (p < 0.05) The activities of hepatic LPL and HL in 1.32% ALA group were significantly higher than other groups (p < 0.05). The activities of hepatic MDH and FAS in 1.32% ALA group were the lowest, which were significantly lower than those in ALA groups (form 0.09% to1.03%) (p < 0.05). Except for the content of MUFA, the contents of other fatty acids showed linear, quadratic, and cubic response with the dietary ALA level. The ALA and ∑n-3 PUFA content significantly increased with increasing dietary ALA levels (p < 0.05). These results indicated that dietary ALA actively impacted fish growth performance, hepatic lipid metabolizing enzymes, and muscle fatty acid profile. Simple Summary: In China, coho salmon is becoming an increasing farming species in recent years. Thus, full understanding of the fatty acid requirements of coho salmon is particularly relevant, especially during the larval stage, since they need high-energy demands and rapid lipid metabolism. As a vital essential fatty acid, linoleic acid (18:2n-6) also plays an important role in the larval stage due to the production of longer-chained and more essential highly unsaturated fatty acids (HUFA). However, there is a lack of knowledge about the dietary linoleic acid (18:2n-6) requirement for this fish species. In this study, six isonitrogenous and isolipidic diets were formulated to contain different dietary linoleic acids (18:2n-6) by increasing corn oil at the expense of coconut oil to study the growth performance, fatty acid profile, and hepatic lipid metabolism enzymes of coho salmon alevins. The results showed that the dietary linoleic acid (18:2n-6) affected the growth performance, muscle fatty acid profile, and hepatic lipid metabolism enzyme activities of coho salmon, and determined the optimal demand of linoleic acid based on the specific growth rate (SGR) and feed conversion ratio (FCR). A 12-week feeding trial aimed to evaluate the effects of dietary linoleic acid (LA, 18:2n-6) on the growth performance, fatty acid profile, and lipid metabolism enzyme activities of coho salmon (Oncorhynchus kisutch) alevins. Six experimental diets (47% crude protein and 15% crude lipid) were formulated to contain graded LA levels of 0.11%, 0.74%, 1.37%, 2.00%, 2.63%, and 3.26%. Each diet was fed to triplicate groups of 50 alevins with an initial body weight of 0.364 ± 0.002 g, which were randomly assigned to 18 white plastic tanks (0.8 × 0.6 × 0.6 m, 240 L/tank). Fish were reared in a freshwater flow-through rearing system and fed to apparent satiation four times daily. The survival rate was not significantly different among the treatments (p > 0.05). However, the 1.37% LA group significantly improved the final body weight and specific growth rate (SGR) (p < 0.05) of alevins. The feed conversion ratio (FCR) in the 1.37% LA group was significantly lower than those in other groups (p < 0.05). The whole-body lipid content significantly decreased (p < 0.05) with dietary LA levels increasing from 0.74% to 2.00%. The fatty acid composition of the total lipid in muscle was closely correlated with those in the diets. The dietary LA level of 1.37% led to significantly higher activities of liver lipoprotein lipase (LPL) and hepatic lipase (HL) than those of other groups (p < 0.05). Hepatic malate dehydrogenase (MDH) and fatty acid synthase (FAS) decreased with the increase in the dietary LA levels from 0.11% to 1.37%. The lowest MDH and FAS activities were obtained in the 1.37% LA group (p < 0.05). This study indicated that an appropriate amount of dietary LA was beneficial for the growth and lipid metabolism of coho salmon alevins, and the results of the quadratic regression analysis of the SGR and FCR indicated that the optimal dietary LA requirements were 1.25% and 1.23% for coho salmon alevins, respectively. In this study, the effects of different dietary iron levels on the growth performance, body composition and antioxidant responses were investigated in the coho salmon Oncorhynchus kisutch alevins. Six experimental diets were formulated to contain graded levels (23.7, 46.4, 59.8, 77.3, 101.7 and 127.7 mg/kg) of iron provided as FeSO4∙7H2O. Each diet was fed to triplicate groups of coho salmon alevins with initial mean weight of 0.35 ± 0.02 g in a flow‐through rearing system for 84 days. The results showed that there was no significant difference in survival among dietary treatments. Specific growth rate (SGR) increased significantly (p < 0.05) while feed conversion rate (FCR) decreased significantly (p < 0.05) with increasing dietary iron level from 23.7 up to 101.7 mg/kg. Whole body moisture and ash showed no significant differences among dietary treatments. Fish fed the diet with 127.7 mg/kg had the significantly highest whole body protein and lipid levels (p < 0.05). Whole body and hepatic iron concentrations increased with increasing dietary iron levels (p < 0.05). However, hepatic iron concentrations were decreased when the dietary iron was higher than 77.3 mg/kg treatment. Fish fed the diet with 77.3 mg/kg had the significantly highest activities of catalase (CAT) and total superoxide dismutase (T‐SOD) but the lowest malondialdehyde (MDA) activity in liver (p < 0.05). Based on the SGR, iron concentrations in the whole body and liver and CAT, T‐SOD and MDA activities in liver, the optimal dietary iron requirements for coho salmon alevins were 101.7, 79.3, 75.7, 90.8, 83.6 and 68.9 mg/kg respectively. In this study, the mechanism that the inhibition of glycogen synthase kinase-3β (GSK-3β) promotes the production of reactive oxygen species (ROS) via β-catenin/CCAAT/enhancer binding protein α (C/EBPα) signaling was investigated in the spleen of zebrafish ( Danio rerio ). The results demonstrated that the inhibition of GSK-3β induced the mRNA expression of β-catenin and C/EBPα by lithium (Li) treatments or GSK-3β RNA interference. The levels of hydrogen peroxide (H 2 O 2 ), superoxide anion (O 2 .- ), and hydroxy radical (·OH) as well as the activity of superoxide dismutase (SOD) were increased, while the activities of catalase (CAT) and glutathione peroxidase (GSH-PX) were decreased in the spleen and ZF4 cells of zebrafish by Li + treatments. In addition, GSK-3β RNA interference increased ROS levels and decreased the activities of CAT and GSH-PX in the spleen. The fluorescence intensity of ROS was increased but the mitochondrial membrane potential (MMP) was decreased by Li + treatments in ZF4 cells labeled with 2′,7′-dichlorofluorescein diacetate (DCFH-DA) and Rhodamine-123, respectively. The results of present study indicated that the inhibition of GSK-3β promoted the ROS production via β-catenin/C/EBPα signaling in the spleen of zebrafish, and the balance between ROS and antioxidants could be destroyed by the GSK-3β/β-catenin/C/EBPα signaling. • Microplastic ingestion in fish occurred in urban water relevant scenario. • Accumulation occured mainly via waterborne uptake rather than food chain transfer. • Both water- and foodborne microplastics exposure caused severe fish hyperactivity. • Cholinergic system alteration was mainly responsible for the neurotoxicity after waterborne exposure. • Obvious dopamine and 5-HT neurotransmitter concentration changes were observed after foodborne exposure. Organisms constantly ingest microplastics directly from the environment or indirectly via trophic transfer due to the pervasiveness of microplastic pollution. However, most previous studies have only focused on waterborne exposure at the individual level, while few studies have investigated the contribution of trophic transfer to the exposure in organisms. We comprehensively evaluated the differences in microplastic ingestion and toxic effects in zebrafish exposed to microplastics via two concurrent routes (waterborne and foodborne). The polyethylene microplastics (40–47 μm, 0.1–10 mg/L) concentration used here was set in a range closed to the environmentally relevant microplastic concentrations, especially considering the extreme high concentration scenarios in wastewater. The concentration of microplastics resulting from foodborne exposure (0.01±0.01 μg/mg; 0.1±0.1 particles/mg) was significantly lower than that through waterborne exposure (0.06±0.02 μg/mg; 0.8±0.3 particles/mg), suggesting the ingestion of microplastics in their tissues occurs mainly through direct environmental uptake rather than food chain transfer (though the initial microplastic concentration was 1000 folds lower). However, more sublethal impacts, including the significant abnormal hyperactive swimming behaviour (107±5% induction; p < 0.05), were observed in the foodborne group than waterborne group. Additionally, ingenuity pathway analysis predicted both exposure routes caused obvious nervous system interference but through opposite modes of action. This was further verified by the alteration of neurotransmitter biomarkers that neurotoxicity mechanisms were completely different for the two exposure routes. A 10-week feeding trial was conducted to evaluate the effects of replacing fish meal (FM) with meat and bone meal (MBM) on growth and body composition of juvenile snakehead Ophiocephalus argus. Six approximately isonitrogenous (450 g kg crude protein) and isoenergetic (18.5 MJ kg gross energy) diets were formulated by replacing 0, 200, 400, 600, 800 and 1000 g kg FM protein with MBM protein, and the diets were designated as control, MBM20, MBM40, MBM60, MBM80 and MBM100, respectively. Each diet was fed to triplicate groups of fish with initial mean body weight of 12.11 ± 0.08 g (mean ± SD) in floating cages in an earthen pond, and each cage was initially stocked with 40 fish. Both specific growth rate (SGR) and protein efficiency ratio (PER) in the control and MBM20 group had no significant differences ( p > 0.05), but were significantly higher ( p < 0.05) than the other groups, and MBM100 showed the lowest SGR and PER. Whole-body crude protein decreased significantly with increasing dietary MBM levels ( p < 0.05). A feeding trial was conducted to investigate the effects of dietary administration of probiotic Bacillus subtilis and prebiotic fructooligosaccharide (FOS) on growth performance, immune responses and disease resistance of juvenile ovate pompano, Trachinotus ovatus. One thousand six hundred and twenty individuals (initial body weight: 10.32 ± 0.46 g, mean ± S.E) were fed nine practical diets according to a 3 × 3 factorial design: the basal diet as the control diet supplemented with three levels of B. subtilis (0, 1.05 × 107 or 5.62 × 107 CFU g−1 diet), crossed with 0, 0.2% or 0.4% FOS. After an 8-week feeding experimental period, six fish per cage were sampled for immunity determination. Then 18 fish of each cage left were challenged by Vibrio vulnificus. The results showed that fish fed with 5.62 × 107 CFU B. subtilis g−1 in combination with 0.2% FOS produced the highest specific growth rate, and were significantly higher than the groups fed with 0 and 0.2% FOS without B. subtilis supplementation (P < 0.05). Feed efficiency ratio significantly increased with the increasing doses of dietary FOS without B. subtilis added (P < 0.05). The immune assay showed that fish fed with the control diet produced the lowest respiratory burst activity and was significantly different from the groups fed the diets containing 0.2% FOS at each B. subtilis level and containing 0.4% FOS single (P < 0.05). Phagocytic activity was significantly decreased with the increasing doses of dietary B. subtilis at 0.4% FOS level (P < 0.05). Alternative complement pathway activity of the fish fed with 0.2% FOS single was significantly lower than those fed with 5.62 × 107 CFU B. subtilis g−1 diet supplemented at each FOS level (P < 0.05). Fish fed with the control diet had the lowest lysozyme activity, and were significantly different from those fed with 0.2 or 0.4% FOS at 1.05 and 5.62 × 107 CFU B. subtilis g−1 diet level. Moreover, fish fed with diets supplemented with 0.2% and 0.4% FOS at each B. subtilis level had notably lower cumulative mortality after 10 days following V. vulnificus infection (P < 0.05). This paper investigates the effect of lanthanum (La) on lipid deposition and Wnt10b signaling in the liver of male zebrafish with exposure of 0, 10, 20, and 30 μmol/L La. It suggests that La can be accumulated in liver, and its treatments decrease the activities and gene expression of enzymes related to fatty acid synthesis. The levels of total cholesterol (TC), triglyceride (TG), and nonesterified fatty acids (NEFA) as well as the size of lipid droplets are decreased by La treatments. Moreover, La treatments affect the composition of fatty acids and the content of nutrient elements. Meanwhile, they also induce the gene expression of wnt10b, β-catenin, pparα , and pparγ , but inhibit gsk-3β gene expression in liver. Further study on the result of wnt10b gene interference shows that Wnt10b/β-catenin signaling plays a crucial role in the regulatory process of hepatic lipid deposition. A feeding trial was conducted to investigate the effects of dietary graded protein levels on the growth, survival, amylase and trypsin activities of large yellow croaker ( Pseudosciaena crocea R.) larvae from 12 to 42 days after hatching (DAH). Five approximately isoenergetic microbound diets (16.65 MJ/kg diet) were formulated to contain different protein (47.1%, 52.0%, 57.1%, 62.2% and 67.5%) levels. Frozen copepods, containing 54.5% crude protein (CP), 6.0% crude lipid, 27.2% ash and 6.7% glycogen, were used as a control. Each diet was randomly fed to triplicate groups of larvae with an initial mean body weight of 1.76 ± 0.09 mg (mean ± SD) in 180 L white plastic tanks, and each tank was stocked initially with 3500 larvae. Both the survival and the specific growth rate (SGR) of large yellow croaker larvae significantly increased with increasing dietary protein level up to 57.1%, and decreased thereafter. Frozen copepods resulted in intermediate survival and low SGR compared with the other diets. Whole-body moisture and protein of larvae were not significantly affected by the dietary protein level. In contrast, whole-body lipid of larvae fed diet with 47.1% CP was significantly higher ( P<0.05) than those from fish fed the diets containing more than 57.1% CP. Additionally, fish fed the frozen copepods had the lowest whole-body protein and lipid. The amylase-specific activity increased with increasing dietary carbohydrate level during the period of this experiment. However, trypsin activity was not significantly affected by the dietary protein content before 42 DAH, indicating a later onset of trypsin than amylase in the regulation of enzymatic synthesis induced by a dietary substrate. Background: Spider silk is a proteinaceous fiber with remarkable mechanical properties spun from spider silk proteins (spidroins). Engineering spidroins have been successfully produced in a variety of heterologous hosts and the most widely used expression system is Escherichia coli (E. coli). So far, recombinantly expressed spidroins often form insoluble inclusion bodies (IBs), which will often be dissolved under extremely harsh conditions in a traditional manner, e.g. either 8 mol/L urea or 6 mol/L guanidine hydrochloride, highly risking to poor recovery of bioactive proteins as well as unexpected precipitations during dialysis process. Results: Here, we present a mild solubilization strategy—one-step heating method to solubilize spidroins from IBs, with combining spidroins' high thermal stability with low concentration of urea. A 430-aa recombinant protein (designated as NM) derived from the minor ampullate spidroin of Araneus ventricosus was expressed in E. coli, and the recombinant proteins were mainly present in insoluble fraction as IBs. The isolated IBs were solubilized parallelly by both traditional urea-denatured method and one-step heating method, respectively. The solubilization efficiency of NM IBs in Tris-HCl pH 8.0 containing 4 mol/L urea by one-step heating method was already comparable to that of 7 mol/L urea with using traditional urea-denatured method. The effects of buffer, pH and temperature conditions on NM IBs solubilization of one-step heating method were evaluated, respectively, based on which the recommended conditions are: heating temperature 70–90 °C for 20 min, pH 7.0–10, urea concentration 2–4 mol/L in normal biological buffers. The recombinant NM generated via the one-step heating method held the potential functions with self-assembling into sphere nanoparticles with smooth morphology. Conclusions: The one-step heating method introduced here efficiently solubilizes IBs under relatively mild conditions compared to the traditional ones, which might be important for the downstream applications; however, this protocol should be pursued carefully in terms of urea-induced modification sensitive applications. In this study, we evaluated the effects of fermented soybean meal (FSBM) or/and unfermented SBM replacing a portion of fish meal (FM) on the growth performance, antioxidant capacity, immunity, and mechanistic target of rapamycin (mTOR) signaling pathway of juvenile coho salmon (Oncorhynchus kisutch). Four groups of juvenile coho salmon (initial weight 152.23 ± 3.21 g) in triplicate were fed for 12 weeks on four different iso-nitrogen and iso-lipid experimental diets: G0 diet (28% FM protein, control group), G1 diet (18% FM protein and 10% SBM protein), G2 diet (18% FM protein, 5% SBM protein, and 5% FSBM protein), and G3 diet (18% FM protein and 10% FSBM protein). The main results were compared with the G0 diet; the weight gain rate, specific growth rate, and condition factor of juveniles in G3 were increased significantly (p < 0.05). The content of muscle crude protein, the total protein, glucose, albumin, total cholesterol in serum, and the total antioxidant capacity in the liver of juveniles in G3 was increased significantly (p < 0.05). The activities of pepsin, trypsin, α-amylase, and lipase in the intestine, the superoxide dismutase, catalase, and alkaline phosphatase in the liver of juveniles in G3 were increased significantly (p < 0.05). The expression levels of phosphatidylinositide 3-kinases, serine/threonine kinase, mTOR, and ribosomal protein S6 kinase 1 genes in the liver of juveniles in G3 were upregulated significantly (p < 0.05). The feed coefficient ratio, viscerosomatic index, the contents of muscle moisture, and malondialdehyde in the liver of juveniles in G3 were decreased significantly (p < 0.05). The expression levels of tumor necrosis factor α, interleukin 1β, and interleukin 6 genes in the liver of juveniles in G3 were downregulated significantly (p < 0.05). However, there was no significant effect (p > 0.05) on the survival rate, food intake, and muscle crude lipid and ash of juveniles among the experimental groups. In conclusion, FSBM to replace a portion FM had a positive effect on the growth performance, protein deposition, antioxidant enzyme activity, digestive enzyme activity, protein synthesis, and immune-related genes of juvenile coho salmon. Simple Summary: As one of the essential mineral elements, manganese (Mn) is involved in maintaining the growth and physiological function of fish species. An optimal dietary level of manganese accelerated the growth, lipid metabolism, Mn accumulation, and antioxidant capacity of coho salmon. The dietary Mn requirement for post-larvae coho salmon was 17.35 mg kg−1 and 19.75 mg kg−1 according to the SGR and FCR, respectively. An optimal dietary level of manganese had a positive effect on the antioxidant capacity by scavenging free radicals in fish bodies. In addition, dietary Mn improved lipid metabolism, and the signaling pathway of PI3K/AKT/mTOR may be involved in regulating the activity of lipid metabolism enzymes. In this study, post-larval coho salmon Oncorhynchus kisutch (initial weight 0.37 ± 0.03 g) were fed with 6 experimental diets with increasing manganese (Mn) content (2.4, 8.5, 14.8, 19.8, 24.6, and 33.7 mg kg−1) for 12 weeks. Our results indicated that the feed conversion rate (FCR), specific growth rate (SGR), condition factor (CF), crude protein, moisture, crude lipid, ash, whole-body Mn, and vertebral Mn were affected by the elevation of Mn content in the diet. The activities of hepatic GSH-PX, Mn-SOD, and CAT were enhanced with increasing Mn content in the diet and reached the highest value at 19.8 mg kg−1 Mn. However, the level of hydrogen peroxide (H2O2), superoxide anion (O2·−), and malondialdehyde (MDA) was reduced with increasing Mn content in the diet. In addition, the activity of hepatic lipase (HL) and lipoprotein lipase (LPL) was increased with the elevation of dietary Mn content and reached a peak value at 14.8 mg kg−1 Mn. The activity of fatty acid synthetase (FAS) and the content of nonesterified fatty acid (NEFA) were increased following the elevation of Mn content from 2.4 to 19.8 mg kg−1 in the diet. The results indicated that the appropriate dietary Mn supplementation improved the feeding efficiency, lipid metabolism, and antioxidant capacity of coho salmon. Simple Summary: Fish meal has been the main aquatic feed protein source for aquaculture. However, global fish meal is lacking, and the price of fish meal continues to rise, which has been unable to meet the needs. Soybean meal is currently recognized as the best choice to replace fish meal in aquatic feed, but soybean meal contains anti-nutritional factors which can affect the health of aquatic animals. Microbial fermentation is a commonly used biological method for treating soybean meal antigens and palatability. The optimal dietary protein to energy ( P/ E) ratio for juvenile peanut worm Sipunculus nudus Linnaeus (initial average weight 46.16 ± 0.07 mg) was determined using practical diets in a 3 × 3 factorial experiment. Three dietary protein levels (38, 43, and 48 %) and three lipid levels (6, 9, and 12 %) were tested, yielding P/ E ratios of 19.5-26.4 mg protein kJ. Each diet was fed to juveniles in three plastic tanks (65 × 55 × 45 cm) for 56 days. S. nudus juveniles fed a diet containing 43 % protein and 9 % lipid, yielding a P/ E ratio of 23.1 mg protein kJ, presented the highest specific growth rate among the treatments ( P < 0.05). The highest protease and lipase activities were observed in S. nudus juveniles fed a diet with a P/ E ratio of 23.1 mg protein kJ. Carcass moisture and ash contents were not significantly affected by the dietary P/ E ratio ( P > 0.05). Carcass protein content improved as dietary protein increased at each lipid level, and carcass lipid content improved as dietary lipid increased at each protein level. The structure of alimentary tract has been studied in a cold water fish Glyptosternum maculatum, an endemic teleost species of notable economic importance and with high potential for controlled rearing of the species in Tibet, by light and electron microscope. Glyptosternum maculatum has short oesophagus, large caecal-type stomach and short intestine, and the digestive tract with four layers: mucosa, submucosa, muscularis and serosa. Taste buds were found in the epithelium of lips, buccopharynx and oesophagus. The stratified epithelium of buccopharynx and oesophagus was located with numerous goblet cells. The U-shaped stomach has three parts, corresponding to mammalian cardiac, fundus and pyloric portion, lined with a single-layered columnar epithelium, and tubular gastric glands are present in cardiac and fundic portion, but absent in pyloric portion. No pyloric caeca was detected. The intestine is separated from the stomach by a loop valve. The intestine epithelium is composed of simple columnar cells with a distinct microvillus brush border and many goblet cells. Meanwhile, the intestinal coefficient was 0.898. At the ultrastuctural level, three type cells (mucous, glandular and endocrine cell) were found in the stomach, and glandular cell with a great amount of pepsinogen granules. This study aims to explore the single and joint effects of depression and malnutrition on the incidence of first fall onset in a Chinese community-dwelling elderly population. This cohort study consisted of 739 residents without a history of falls who were aged 60 years and older (mean age: 67.08 ± 5.79 years, female: 58.2%). Depression was defined with the Geriatric Depression Scale (GDS)-30; a score of ≥11 was considered to be depressed, while malnutrition was defined with the Mini Nutritional Assessment where a score <17 was defined as malnourished. Over a 2-year follow-up period, older adults who experienced at least one fall were allocated to the first fall onset group. The prevalence of baseline falls was 21.36%. During the 2-year follow-up, incidence of first fall onset was 13.13%. After adjusting for all confounders, depression alone (adjusted odds ratio [OR] = 3.545, 95% confidence interval [CI] = 1.318-9.535) and malnutrition alone (adjusted OR = 2.204, 95% CI = 1.183-4.108) were observed to be independent risk factors for first fall onset, while comorbidity of depression and malnutrition showed progressively increased risk of promoting first fall (adjusted OR = 8.161, 95% CI = 3.591-18.545) than those with only depression or malnutrition or without both depression and malnutrition. Malnutrition mediated 56% effects in the association between depression and first fall onset, while depression mediated 76% effects in the promoting role of malnutrition in first fall. Depression and malnutrition were found to be independent causes for promoting first fall, while mental health and nutrition should be treated as commonly prior interventions to delay first fall onset. Epidemiological studies report that more than half of the people over the age of 65 years suffer from variable sleep problems. In this study, we conducted a cohort study to investigate the relationship between sleep duration, muscle mass, and function within a community-dwelling, elderly Chinese population. Our study population consisted of residents living in the township central hospital of suburban Tianjin, China. We measured muscle strength and walking speed. We divided sleep duration into the following four groups: <7 hours, 7-8 hours, >8-9 hours, and >9 hours. A total of 902 participants completed the 3-year follow-up. We observed a U-shaped relationship between sleep duration and fall risk. Compared with the 7-8-hour group, fall risk within the <7-hour group was 3.67 (2.59, 5.42) times higher and fall risk within the >9-hour group was 2.35 (1.29, 3.52) times higher. After adjustment, muscle mass declined by -6.82% (-11.27%, -3.83%) in the <7-hour group. In summary, we observed a U-shaped relationship between sleep duration and falls. Abstract Opioid neuropeptides are developed early in the course of a long evolutionary process. As the endogenous messengers of immune system, opioid neuropeptides participate in regulating immune response. In this study, the mechanism that Met-enkephalin (M-ENK) inhibits ROS production through Wnt/β-catenin signaling was investigated in the ZF4 cells of zebrafish. ZF4 cells were exposed to 0, 10, 20, 40, 80, and 160 μM Met-enkephalin (M-ENK) for 24 h, and the cell viability was detected with 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay. The cell viability was significantly increased by 10, 20, 40, 80, and 160 μM M-ENK. After ZF4 cells were exposed to 0, 20, 40, and 80 μM M-ENK for 24 h, the mRNA expression of Wnt10b, β-catenin, and CCAAT/enhancer binding protein α (C/EBPα) was significantly increased by 40 and 80 μM M-ENK. However, the mRNA and protein expression of GSK-3β was significantly decreased by 40 and 80 μM M-ENK. The protein expression of β-catenin was significantly induced by 40 and 80 μM M-ENK, while the protein expression of p-β-catenin was significantly decreased by 20, 40, and 80 μM M-ENK. In addition, the mRNA expression of CAT, SOD, and GSH-PX was significantly increased by 40 and 80 μM M-ENK. The levels of H 2 O 2 , ·OH, and O 2 ·- were significantly decreased, but the activity of CAT, SOD, and GSH-PX was significantly increased by 40 and 80 μM M-ENK. The fluorescence intensity of reactive oxygen species (ROS) was decreased, and that of mitochondrial membrane potential (MMP) was increased with the increase of M-ENK concentration in ZF4 cells. The results showed that M-ENK could induce Wnt/β-catenin signaling, which further inhibited ROS production through the induction of C/EBPα, MMP, and the activities of antioxidant enzymes. Background: The aim of the present study was to examine the risk factors for developing diabetes after 3 years in an elderly Chinese suburban population with impaired fasting glucose (IFG). Methods: The study population comprised residents of the Hangu area of Tianjin, China, with IFG, aged ≥60 years, who joined the national free physical examination program (n = 328; mean [±SD] age 68.0 ±6.1 years; 48.2% men). Diabetes was defined by self‐report of a physician's diagnosis or fasting plasma glucose (FPG) ≥7.0 mmol/L; IFG was defined as FPG ≥5.6 and < 7.0 mmol/L. Risk factors of incident diabetes at the 3‐year follow‐up were analyzed individually using logistic regression analysis. Results: Between baseline and the 3‐year follow‐up, 56 subjects with IFG at baseline had developed diabetes. After multivariate adjustment for demographic and clinical factors, the incidence of diabetes increased with higher FPG (odds ratio [OR] 9.30, 95% confidence interval [CI] 2.84–30.48), but decreased with the grip strength/weight ratio (OR 0.88, 95% CI 0.82–0.94). Moreover, the combination of higher FPG and lower grip strength/weight was associated with a higher incidence of diabetes than higher FPG only or lower grip strength/weight (P < 0.05). Conclusion: The present study indicates that higher FPG and lower muscle strength are associated with glycemic deterioration among subjects with IFG after 3 years. The results suggest that not only glucose levels, but also physical performance may be useful markers of the risk of diabetes in this population. Abstract Objective The objective of this study was to examine the incidence of new onset depressive symptoms and associated factors over a 1-year period in an older Chinese suburban population. Design Prospective cohort study. Setting and Participants The sample comprised 691 Chinese community-dwelling participants (304 men; mean age 67.5 ± 5.7 years) without depressive symptoms at baseline, recruited from Chadian of Tianjin, China. Measures We had documented detailed information regarding sociodemographics, behavioral characteristics, and medical conditions. Sarcopenia was defined according to the Asian Working Group for Sarcopenia (AWGS) criteria. The outcome was new onset depressive symptoms at 1-year follow-up, defined as a score of ≥11 on the 30-item Geriatric Depression Scale. Results We found that 83 (12.0%) of the 691 participants without depressive symptoms at baseline had developed depressive symptoms. After multivariate adjustments, it was found that the incidence of new onset depressive symptoms was associated with sarcopenia, type 2 diabetes mellitus, and cardiovascular disease. People with a higher level of muscle mass and better sleep quality were significantly less likely to develop depressive symptoms than their counterparts. Conclusions/Implications We found that the incidence of depressive symptoms increased with some chronic diseases, such as sarcopenia, type 2 diabetes mellitus, and cardiovascular disease. In addition, muscle mass was the most related protective factor among sarcopenia's 3 basic diagnosis components—muscle mass, muscle strength, and physical performance. The objective of this study was to determine falls risk profiles to derive a falls risk prediction score and establish a simple and practical clinical screening tool for Chinese community-dwelling elderly individuals. This was a prospective cohort study (n = 619) among adults aged 60 years and older. Falls were ascertained at a 1-year follow-up appointment. Sociodemographic information, medical history, and physical performance data were collected. The mean age was 67.4 years; 57.7% were women. Female sex (odds ratios [ORs] 1.82; 95% confidence interval [95% CI] 1.17-2.82), diabetes (OR 2.13; 95% CI 1.13-3.98), a Timed Up and Go Test (TUGT) ≥10.49 seconds (OR 1.51; 95% CI 1.23-1.94), a history of falls (OR 3.15; 95% CI 1.72-5.79), and depression (Geriatric Depression Scale [GDS] ≥11, OR 2.51; 95% CI 1.36-4.63) were the strongest predictors. These predictors were used to establish a risk score. The area under the curve of the score was 0.748. From a clinical point of view, the most appropriate cutoff value was 7 (97.5% specificity, 70.7% positive predictive value, and 83.6% negative predictive value). For this cutoff, the fraction correctly classified was 82.5%. A cutoff score of 7 derived from a risk assessment tool using four risk factors (gender, falls history, diabetes, and depression) and the TUGT may be used in Chinese community-dwelling elderly individuals as an initial step to screen those at low risk for falls. In this study, the mechanism that lithium (Li) promotes the production of reactive oxygen species (ROS) via the glycogen synthase kinase-3β (GSK-3β)/tuberous sclerosis complex 2 (TSC2)/target of rapamycin (TOR) signaling was investigated in the gill of zebrafish ( Danio rerio ). After the zebrafish were treated by 25 and 50 mg/L Li + , the mRNA expression of GSK-3β and TSC2 was inhibited, but the expression of TOR was induced in the gill of zebrafish. The levels of hydrogen peroxide (H 2 O 2 ), superoxide anion (O 2 ·- ), and hydroxy radical (·OH) as well as the activity of superoxide dismutase (SOD) were increased, while the activities of catalase (CAT), glutathione peroxidase (GSH-PX), and peroxidase (POD) were decreased by 25 and 50 mg/L Li + treatments. In the ZF4 cells, the mRNA expression of GSK-3β and TSC2 was inhibited, but TOR expression was induced by 1, 5, and 10 mmol/L Li + treatments. To further confirm that lithium promoted ROS production via GSK-3β inhibition, GSK-3β RNA was interfered. It was found that the interference of GSK-3β RNA induced the TSC2/TOR signaling. The levels of H 2 O 2 , O 2 ·- , and ·OH were increased, but the activities of CAT, GSH-PX, and POD were decreased by GSK-3β RNA interference. In addition, lithium decreased the mitochondrial membrane potential (MMP) with Rhodamine-123 assay, but increased the levels of ROS by 2′,7′-dichlorofluorescein diacetate (DCFH-DA) assay. This paper puts forward the reasonable choice of live transportation strategy to guarantee survival rate and quality of Urechis unicinctus by designing its packaging mode and carrying out real-time monitoring technologies. Ambient data sensing devices are designed and deployed in water and waterless transportation facilities by which the simulation of real live transportation is tested. During the delivery process of live Urechis unicinctus, the key ambient factors – temperature, dissolved oxygen/oxygen, carbon dioxide, PH, salinity–are dynamically sampling by on-line electronic monitoring equipment. Urechis unicinctus transport ambient data and their correlations are statistically calculated and analyzed by transportation monitoring and analysis system for the two transportation strategies. Through five control groups test for water and waterless shipment, it is found that waterless transportation is more suitable for over 30 h live transportation by studying transportation facilities management, surface characteristics and survival rate, which can provide a sound statistical basis of reasonable transportation mode to guarantee consumers to eat healthier and cheaper seafood with more convenient and economical way. This article aims to examine the effect of mobility limitation and obesity on the risk of new incidence of type 2 diabetes mellitus (T2DM) and mortality. The design was a cohort study (n = 1075) among adults aged 60 years and older. Obesity was defined as body-mass index greater than or equal to 28 kg/m2. Mobility limitation was defined as participants scoring in the top 20% on the timed up and go test or in the slowest 20% for the 4-m walking test. The mean age of the study population was 67.4 ± 5.4 years (age range: 60-86 years), and 57.4% were women. Overall, 5.1% of women and 1.9% of men had both obesity and mobility limitations. During 3-year follow-up, the new incidence of T2DM was 2.98% and the adjusted risk of the new incidence of T2DM was progressively greater in obese subjects without mobility limitation, but not greater in the single mobility limitation subjects. The combination of mobility limitation and obesity (odds ratio = 10.3, 95% confidence interval = 2.25-70.13) has a significantly higher risk than obesity only or mobility limitation only. What is more, obesity with mobility limitation could be an independent predictor of 3-year mortality compared with the other subjects. Background: Physical performance is an important healthy factor in elder people. Good living habits, which include sleep, can maintain physical strength and physical performance. The aim of the present study was to conduct a cross-sectional study to determine the association between total sleep duration and physical performance. Methods: Our study population comprised residents of the township central hospital in the suburban of Tianjin, China. We measured muscle strength, walk speed and balance function by grip, 4-m walk test and timed up and go test (TUGT). We divided sleep duration into four groups <7h, 7-8h, >8-9h, >9h. Results: A total 898 participants had completed data (392 men and 506 women, mean age 67.71 years). In man, adjusted sleep duration was associated with lower grip in > 9 h group, the mean value (95% CI) was 0.429 (0.409, 0.448), and longer TUGT time was also associated with long sleep duration, 10.46s (9.97 s, 10.95 s). In women, adjusted slower 4-m walk speed present an inverse U-shaped relation with sleep duration, by 0.93 m/s (0.86 m/s, 0.98 m/s), 0.97 m/s (0.96 m/s, 1.00 m/s), 0.97 m/s (0.95 m/s, 0.99 m/s) and 0.92 m/s (0.89 m/s, 0.96 m/s); longer TUGT time were associated with long sleep duration (> 9 h), by 11.23 s (10.70 s, 11.77 s). Conclusion: In Chinese community-dwelling elderly, lower muscle strength and lower balance function were associated with long sleep duration in men. Abstract: Large yellow croaker is an important marine fish species for aquaculture in China. Larval rearing of this species is still based on live prey feeding. The aim of this study was to describe the onset and development of the main digestive enzymes during larval development of this species, in order to obtain essential data for the formulation of a compound diet adapted to larvae. Enzymatic assays were conducted from day 1 post hatching to day 40 in larvae fed the following live prey sequence: rotifers, Artemia nauplii, copepods, and then frozen copepods. Assays were done on whole larvae until day 19, then on dissected larvae and on purified brush border membranes of intestine. Pancreatic enzymes, amylase, and trypsin, showed high activities from early stages, even before mouth opening. Mechanisms of pancreatic secretion were achieved from day 19. Enzymes of brush border membranes, alkaline phosphatase, and aminopeptidase N, increased abruptly between day 23 and 25, showing maturation of the intestinal digestive process. These data, together with data describing mouth enlargement during development, suggest that large yellow croaker larvae could be fed successfully with a compound diet from early stages, at least from day 23. • Relative handgrip strength predicts MetS superior to absolute handgrip strength. • The overall prevalence of MetS is 38.4 % in this study. • HS/body fat mass appears to be the index best associated with MetS. • The cut-off value of HS/body fat mass to detect MetS is 1.92 in men and 1.25 in women. Currently there is no consensus on the correlation between metabolic syndrome (MetS) and muscle strength. The objective of this study was to examine the associations between MetS and its components and different handgrip strength (HS) indexes among Chinese community-dwelling elderly individuals. In addition, we hoped to find an optimal cutoff point for the index most relevant to MetS. Data were obtained from 909 participants aged ≥ 60 years (385 men, average age, 68.0 ± 5.9 y). We used the International Diabetes Federation metabolic syndrome guidelines to define MetS. General data of all participants were collected through questionnaires and anthropometric data were measured. At the same time, blood samples were collected. The prevalence of MetS was 26.8 % in men and 46.9 % in women. In all HS indexes, HS/body fat mass was most strongly correlated with MetS, and the areas under the receiver-operating characteristic curve were 0.723 (95 % confidence interval [CI] = 0.669-0.776) in men and 0.619 (95 % CI = 0.571-0.667) in women, and the optimal cutoffs were 1.92 in men and 1.25 in women. The adjusted odds ratios (ORs) of MetS for low HS/body fat mass were 5.38 (95 % CI = 3.03–9.56, p < 0.001) in men and 2.39 (95 % CI = 1.56–3.64, p < 0.001) in women. There are 19 Wnt genes in mammals that belong to 12 subfamilies. Wnt signaling pathways participate in regulating numerous homeostatic and developmental processes in animals. However, the function of Wnt10b in fatty acid synthesis remains unclear in fish species. In the present study, we uncovered the role of the Wnt10b signaling pathway in the regulation of fatty acid synthesis in the muscle of zebrafish. The gene of Wnt10b was overexpressed in the muscle of zebrafish using pEGFP-N1-Wnt10b vector injection, which significantly decreased the expression of glycogen synthase kinase 3β (GSK-3β), but increased the expression of β-catenin, peroxisome proliferators-activated receptor γ (PPARγ), and CCAAT/enhancer binding protein α (C/EBPα). Moreover, the activity and mRNA expression of key lipogenic enzymes ATP-citrate lyase (ACL), acetyl-CoA carboxylase (ACC) and fatty acid synthetase (FAS), and the content of non-esterified fatty acids (NEFA), total cholesterol (TC), and triglyceride (TG) were also significantly decreased. Furthermore, interference of the Wnt10b gene significantly inhibited the expression of β-catenin, PPARγ, and C/EBPα, but significantly induced the expression of GSK-3β, FAS, ACC, and ACL. The content of NEFA, TC, and TG as well as the activity of FAS, ACC, and ACL significantly increased. In this study, the mechanism by which GSK-3β regulates protein synthesis and lipid deposition was investigated in zebrafish (Danio rerio). The vector of pEGFP-N1-GSK-3β was constructed and injected into the muscle of zebrafish. It was found that the mRNA and protein expression of tuberous sclerosis complex 2 (TSC2) was significantly increased. However, the mRNA and protein expression of mammalian target of rapamycin (mTOR), p70 ribosomal S6 kinase 1 (S6K1), and 4E-binding protein 1 (4EBP1) was significantly decreased by the pEGFP-N1-GSK-3β vector in the muscle of zebrafish. In addition, the mRNA and protein expression of β-catenin, CCAAT/enhancer binding protein α (C/EBPα), and peroxisome proliferators-activated receptor γ (PPARγ) was significantly decreased, but the mRNA expression of fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), ATP-citrate lyase (ACL), and HMG-CoA reductase (HMGCR) was significantly increased by the pEGFP-N1-GSK-3β vector. The activity of FAS, ACC, ACL, and HMGCR as well as the content of triglyceride (TG), total cholesterol (TC), and nonesterified fatty acids (NEFA) were significantly increased by the pEGFP-N1-GSK-3β vector in the muscle of zebrafish. The content of free amino acids Arg, Lys, His, Phe, Leu, Ile, Val, and Thr was significantly decreased by the pEGFP-N1-GSK-3β vector. The results indicate that GSK-3β may participate in regulating protein synthesis via TSC2/mTOR signaling and regulating lipid deposition via β-catenin in the muscle of zebrafish (Danio rerio).
