Abstract Background The electrophoretic mobility shift assay (EMSA) is a common technology to detect DNA-protein interactions. However, in most cases, the protein used in EMSA is obtained via prokaryotic expression, and rarely from plants. At the same time, the proteins expressed from prokaryotic systems usually cannot fold naturally and have no post translationally modification, which may affect the binding of proteins to DNA. Results Here, we develop a technique to quickly isolate proteins of interest from host plants and then analyze them using fluorescent EMSA. This technology system is called: protein from plants fluorescent EMSA method (PPF-EMSA). In PPF-EMSA, a special transient transformation method is employed to transiently deliver genes into the plant, enabling efficient synthesis the encoded proteins. Then, the target protein is isolated using immunoprecipitation, and the DNA probes were labeled with cyanine 3 (Cy3). Both fluorescent EMSA and super-shift fluorescent EMSA can be performed using the proteins from plants. Three kinds of plants, Betula platyphylla, Populus. davidiana×P. bolleana and Arabidopsis thaliana, are used in this study. The proteins isolated from plants are in a natural state, can fold naturally and are posttranslationally modified, enabling true binding to their cognate DNAs. Conclusion As transient transformation can be performed quickly and not depended on whether stable transformation is available or not, we believe this method will have a wide application, enabling isolation of proteins from host plant conveniently.

Heavy metals and per- and polyfluoroalkyl substances (PFASs) have become particularly important when studying the development of depression, a common illness that severely restricts psychosocial functioning and diminishes quality of life. Therefore, the potential joint effects of heavy metal and PFAS exposure on depression, as well as the underlying mechanisms involved, were investigated by using integrated epidemiological and bioinformatic approaches in the present study. A thorough analysis of 7301 samples from the National Health and Nutrition Examination Survey (NHANES) cycles that occurred between 2005 and 2018 was performed. Single-exposure studies have shown that cadmium exposure is positively associated with depression, whereas perfluorooctanesulfonic acid (PFOS) exposure and perfluorodecanoic acid (PFDE) exposure are negatively associated with depression. Furthermore, the Bayesian kernel machine regression (BKMR) and quantile g-computation (QGcomp) models were employed to investigate the collective impact of exposure to mixed metals on depression. Cadmium emerged as the principal contributor to depression. Moreover, the addition of PFAS to the metal mixture had an antagonistic effect on depression, with PFOS having the most prominent influence. Analysis of the effects of co-exposure to cadmium and PFOS confirmed the presence of an antagonistic effect. The inflection points of cadmium and PFOS were determined to be −1.11 and 2.27, respectively. Additionally, exposure to cadmium and PFOS had the opposite effects on two crucial pathways, namely, the rap1 and calcium signaling pathways, which involve core genes related to depression such as ADORA2A, FGF2, and FGFR1. These findings have significant implications for future studies and provide new strategies for exploring the mechanisms underlying co-exposure effects.

BackgroundDepression poses a major threat to public health with an increasing prevalence in the United States. Mindfulness-based interventions, such as mindfulness-based cognitive therapy (MBCT), are effective methods for managing depression symptoms and may help fortify existing efforts to address the current disease burden. The in-person group format of MBCT, however, incurs barriers to care such as expenses, childcare needs, and transportation issues. Alternate delivery modalities such as MBCT delivered via the web can be investigated for their capacity to overcome these barriers and still reduce symptoms of depression with adequate feasibility and efficacy. ObjectiveThis study protocol aims to examine the feasibility and efficacy of MBCT delivered via the web for the treatment of depression. MethodsTo attain study aims, 2 phases will be implemented using a waitlist control design. A total of 128 eligible participants will be randomized into either an 8-week MBCT intervention group plus treatment as usual (MBCT + TAU; group 1) or an 8-week waitlist control group (group 2). In phase I (8 weeks), group 1 will complete the intervention and group 2 will proceed with TAU. In phase II (8 weeks), group 2 will complete the intervention and group 1 will continue with TAU until reaching an 8-week follow-up. TAU may consist of receiving psychotherapy, pharmacotherapy, or combined treatment. Data collection will be completed at baseline, 8 weeks (postintervention for group 1 and preintervention for group 2), and 16 weeks (follow-up for group 1, postintervention for group 2). The primary outcomes will include (1) current, residual, or chronic depression symptoms and (2) psychiatric distress. Secondary outcomes will include perceived stress and facets of mindfulness. The feasibility will be measured by assessing protocol adherence, retention, attendance, and engagement. Finally, the extent of mindfulness self-practice and executive functioning skills will be assessed as mediators of intervention outcomes. ResultsThis study began screening and recruitment in December 2022. Data collection from the first cohort occurred in January 2023. By November 2023, a total of 30 participants were enrolled out of 224 who received screening. Data analysis began in February 2024, with an approximate publication of results by August 2024. Institutional review board approval took place on September 11, 2019. ConclusionsThis trial will contribute to examining mindfulness-based interventions, delivered via the web, for improving current, residual, or chronic depression symptoms. It will (1) address the feasibility of MBCT delivered via the web; (2) contribute evidence regarding MBCT’s efficacy in reducing depression symptoms and psychiatric distress; and (3) assess the impact of MBCT on several important secondary outcomes. Findings from this study will develop the understanding of the causal pathways between MBCT delivered via the web and depression symptoms further, elucidating the potential for future larger-scale designs. Abstract Aims Circulating biomarkers can provide important information for the diagnosis and prognosis of dilated cardiomyopathy (DCM). We explored novel biomarkers for the diagnosis and prognosis of DCM to improve clinical decision‐making. Methods and results A total of 238 DCM patients and 65 control were consecutively enrolled at Zhongshan Hospital between January 2017 and January 2019. In the screening set, four DCM patients and four controls underwent measurements of serum proteomic analysis. Seventy‐six differentially expressed circulating proteins were screened by data‐independent acquisition proteomics, and three of these proteins (S100A4, S100A8/A9, and S100A12) were validated by multiple‐reaction monitoring‐mass spectrometry. In the validation set, subsequently, a total of 234 DCM patients and 61 control subjects were evaluated by enzyme‐linked immunosorbent assay. Circulating S100A4, S100A8/A9, and S100A12 were significantly increased in DCM patients (P 0.05). Cox proportional hazards regression analysis revealed that S100A4 [≥75th percentile vs.

Abstract Background Clear cell renal cell carcinoma (ccRCC) is characterized as one of the most common types of urological cancer with high degrees of malignancy and mortality. Due to the limited effectiveness of existing traditional therapeutic methods and poor prognosis, the treatment and therapy of advanced ccRCC patients remain challenging. Tryptophan metabolism has been widely investigated because it significantly participates in the malignant traits of multiple cancers. The functions and prognostic values of tryptophan metabolism-related genes (TMR) in ccRCC remain virtually obscure. Methods We employed the expression levels of 40 TMR genes to identify the subtypes of ccRCC and explored the clinical characteristics, prognosis, immune features, and immunotherapy response in the subtypes. Then, a model was constructed for the prediction of prognosis based on the differentially expressed genes (DEGs) in the subtypes from the TCGA database and verified using the ICGC database. The prediction performance of this model was confirmed by the receiver operating characteristic (ROC) curves. The relationship of Risk Score with the infiltration of distinct tumor microenvironment cells, the expression profiles of immune checkpoint genes, and the treatment benefits of immunotherapy and chemotherapy drugs were also investigated. Results The two subtypes revealed dramatic differences in terms of clinical characteristics, prognosis, immune features, and immunotherapy response. The constructed 6-gene-based model showed that the high Risk Score was significantly connected to poor overall survival (OS) and advanced tumor stages. Furthermore, increased expression of CYP1B1, KMO, and TDO2 was observed in ccRCC tissues at the translation levels, and an unfavorable prognosis for these patients was also found. Conclusion We identified 2 molecular subtypes of ccRCC based on the expression of TMR genes and constructed a prognosis-related model that may be used as a powerful tool to guide the prediction of ccRCC prognosis and personalized therapy. In addition, CYP1B1, KMO, and TDO2 can be regarded as the risk prognostic genes for ccRCC.

Abstract Background Clear-cell renal cell carcinoma (ccRCC) is one of prevalent kidney malignancies with an unfavorable prognosis. There is a need for a robust model to predict ccRCC patient survival and guide treatment decisions. Methods RNA-seq data and clinical information of ccRCC were obtained from the TCGA and ICGC databases. Expression profiles of genes related to natural killer (NK) cells were collected from the Immunology Database and Analysis Portal database. Key NK cell-related genes were identified using consensus clustering algorithms to classify patients into distinct clusters. A NK cell-related risk model was then developed using Least Absolute Shrinkage and Selection Operator (LASSO) Cox regression to predict ccRCC patient prognosis. The relationship between the NK cell-related risk score and overall survival, clinical features, tumor immune characteristics, as well as response to commonly used immunotherapies and chemotherapy, was explored. Finally, the NK cell-related risk score was validated using decision tree and nomogram analyses. Results ccRCC patients were stratified into 3 molecular clusters based on expression of NK cell-related genes. Significant differences were observed among the clusters in terms of prognosis, clinical characteristics, immune infiltration, and therapeutic response. Furthermore, six NK cell-related genes (DPYSL3, SLPI, SLC44A4, ZNF521, LIMCH1, and AHR) were identified to construct a prognostic model for ccRCC prediction. The high-risk group exhibited poor survival outcomes, lower immune cell infiltration, and decreased sensitivity to conventional chemotherapies and immunotherapies. Importantly, the quantitative real-time polymerase chain reaction (qRT-PCR) confirmed significantly high DPYSL3 expression and low SLC44A4 expression in ACHN cells. Finally, the decision tree and nomogram consistently show the dramatic prediction performance of the risk score on the survival outcome of the ccRCC patients. Conclusions The six-gene model based on NK cell-related gene expression was validated and found to accurately mirror immune microenvironment and predict clinical outcomes, contributing to enhanced risk stratification and therapy response for ccRCC patients.

Abstract The current reliability criterion (RC) of the film capacitor (FC) is usually a fixed value, which does not consider the staged capacitance loss caused by moisture diffusion under high temperature and/or high humidity. The expected lifetime of capacitors would deviate far from the actual one. The moisture diffusion process of the encapsulated FC is analysed based on Fick's second law. In order to investigate the capacitance loss and moisture diffusion characteristics, 4 types of film capacitors are stressed with AC voltage under different temperatures and humidities. Based on the staged analysis of the capacitance and weight gain curves, the effect of moisture diffusion on capacitance loss is revealed. Results indicate that the linear stage of capacitance loss corresponds to the Fickian moisture diffusion stage of weight gain. The effect of voltage on FC aging is the most significant. The capacitance loss rate increases by 301% when the voltage rises by 30%. An RC calculation method of the FC is further proposed, which could also screen the encapsulation quality and shorten the aging test duration by at least 80%.

Abstract Background The incidence of kidney disease caused by thyroid cancer is rising worldwide. Observational studies cannot recognize whether thyroid cancer is independently associated with kidney disease. We performed the Mendelian randomization (MR) approach to genetically investigate the causality of thyroid cancer on immunoglobulin A nephropathy (IgAN). Methods and results We explored the causal effect of thyroid cancer on IgAN by MR analysis. Fifty-two genetic loci and single nucleotide polymorphisms were related to thyroid cancer. The primary approach in this MR analysis was the inverse variance weighted (IVW) method, and MR‒Egger was the secondary method. Weighted mode and penalized weighted median were used to analyze the sensitivity. Liriomyza trifolii is a significant invasive pest that targets horticultural and vegetable crops, causing large-scale outbreaks characterized by pronounced thermotolerance and insecticide resistance. This study examined the impact of long-term selection for abamectin resistance during the larval stage of L. trifolii on its population dynamics and thermal tolerance. We conducted a comprehensive comparison between the abamectin-resistant strain (AB-R) and the susceptible strain (S), including age-stage, two-sex life table analysis, thermal preference (Tpref), critical thermal maximum (CTmax), heat knockdown times (HKDTs), eclosion and survival rates, and LtHsp expression under heat stress. Our results showed that while selection for abamectin resistance was detrimental to survival and reproduction, it activated self-defense mechanisms and rapid adaptive adjustments and conferred modest thermal tolerance, which suggests a dual nature of insecticide effects. The AB-R strain exhibited significantly higher thermal preference and CTmax values, along with a longer HKDT and improved survival. Additionally, there was a significant upregulation of LtHsp expression in the AB-R strain compared to the S strain. These findings indicate that the evolution of thermal adaptation was accompanied by abamectin resistance development, emphasizing the necessity of considering temperature effects when applying chemical control. Our study provides valuable insights into how physiological acclimation may help mitigate the toxic effects of insecticides and illustrate how insects respond to multiple environmental pressures.

Hatchery rearing significantly influences the phenotypic development of fish, with potential adverse effects for the post-release performance of hatchery-reared individuals in natural environments, especially when targeted for stock enhancement. To assess the suitability of releasing hatchery-reared fish, a comprehensive understanding of the phenotypic effects of captive rearing, through comparisons with their wild conspecifics, is essential. In this study, we investigated the divergence in body coloration between wild and hatchery-reared marbled rockfish Sebastiscus marmoratus. We examined the selection preferences for different light colors and assessed the impact of different ambient light colors on the morphological color-changing ability of juvenile marbled rockfish. Our findings revealed significant differences in body color between wild and hatchery-reared marbled rockfish. The hue and saturation values of wild marbled rockfish were significantly higher than those of their hatchery-reared counterparts, indicative of deeper and more vibrant body coloration in the wild population. Following a ten-day rearing period under various light color environments, the color of wild marbled rockfish remained relatively unchanged. In contrast, hatchery-reared marbled rockfish tended to change their color, albeit not reaching wild-like coloration. Light color preference tests demonstrated that wild juvenile marbled rockfish exhibited a preference for a red-light environment, while hatchery-reared individuals showed a similar but weaker response. Both wild and hatchery-reared marbled rockfish displayed notable negative phototaxis in the presence of yellow and blue ambient light. These results highlight the impact of hatchery rearing conditions on the body color and morphological color-changing ability, and provide insight into light color selection preferences of marbled rockfish. To mitigate the divergence in phenotypic development and produce more wild-like fish for stocking purposes, modifications to the hatchery environment, such as the regulation of ambient light color, should be considered.

BackgroundMyocardial infarction (MI) caused by severe coronary artery disease has high incidence and mortality rates, making its prevention and treatment a central and challenging aspect of clinical work for cardiovascular practitioners. Recently, researchers have turned their attention to a novel mechanism of cell death caused by Cu2+, cuproptosis.MethodsThis study integrated data from three MI-related bulk datasets downloaded from the Gene Expression Omnibus (GEO) database, and identified 16 differentially expressed genes (DEGs) related to cuproptosis by taking intersection of the 6378 DEGs obtained by differential analysis with 49 cuproptosis-related genes. Four hub genes, Dbt, Dlat, Ube2d1 and Ube2d3, were screened out through random forest analysis and Lasso analysis. In the disease group, Dbt, Dlat, and Ube2d1 showed low expression, while Ube2d3 exhibited high expression.ResultsFocusing on Ube2d3 for subsequent functional studies, we confirmed its high expression in the MI group through qRT-PCR and Western Blot detection after successful construction of a MI mouse model by left anterior descending (LAD) coronary artery ligation, and further clarified the correlation of cuproptosis with MI development by detecting the levels of cuproptosis-related proteins. Moreover, through in vitro experiments, Ube2d3 was confirmed to be highly expressed in oxygen-glucose deprivation (OGD)-treated cardiomyocytes AC16. In order to further clarify the role of Ube2d3, we knocked down Ube2d3 expression in OGD-treated AC16 cells, and confirmed Ube2d3’s promoting role in the hypoxia damage of AC16 cells by inducing cuproptosis, as evidenced by the detection of MTT, TUNEL, LDH release and cuproptosis-related proteins.ConclusionIn summary, our findings indicate that Ube2d3 regulates cuproptosis to affect the progression of MI.

Export production, which is defined as the export of organic matter fixed by photosynthesis, is crucial for sustaining oceanic carbon uptake. The export route in the open ocean is the sinking of biogenic particles through the bottom of the euphotic layer. In contrast, the export routes in the shelf seas are the sinking of biogenic particles to the sediment and the horizontal transport of biogenic particles across the boundary of the shelf seas to the open ocean. The biogenic particles in the shelf seas are supported by multisource nutrients including riverine and oceanic ones. Their exports depend on the hydrodynamic conditions and biogeochemical processes responsible for different sources of nutrients. Here, a unique physical-biological coupled model with a tracking approach is applied to evaluate the export production supported by multisource dissolved inorganic nitrogen (DIN) over the East China Sea. The total export production is 6.83 kmol N s-1 (=17.16 Tg C yr-1), which is slightly lower than the reported atmospheric CO2 absorption. Approximately 80% of particulate organic nitrogen (PON) is exported via off-shelf transport, and the remaining 20% is buried in the sediment. The PON supported by DIN from rivers accounts for 8% of export production, with an e-ratio (export production/primary production) of 0.09. In comparison, that from the Kuroshio accounts for 64%, with an e-ratio of 0.22. This suggests that offshore areas here are more efficient in exporting local production than nearshore ones, largely supported by oceanic nutrients.

ObjectivesThe heat transfer characteristics of steam condensation in a passive residual heat removal system heat exchanger (PRHR HX) are among the most important factors affecting the operational capability of marine nuclear power platform PRHR systems. This study aims to analyze the heat transfer characteristics of steam condensation in a PRHR HX and obtain the accurate heat transfer coefficient relationship.MethodsThe heat transfer characteristics of steam condensation in a PRHR HX are analyzed by building an experimental device with a power ratio of 1∶50. ResultsThe experimental results show that the heat transfer coefficient increases with the increase in steam pressure. The internal flow pattern of the PRHR HX is a stratified flow or wave flow-annular flow-wave flow. The maximum flow rate of HX vapor is 6.72 m/s when the system pressure is 0.52 MPa, and when the pressure exceeds 0.52 MPa, the steam velocity decreases with the increase in heating power. The proposed new correlation shows good agreement, with an error of ±8% between the calculated and experimental results.ConclusionsThis study can provide useful references for the design of reactor safety systems for marine nuclear power platforms.

To evaluate the protective effects of sodium butyrate (SB) on intestinal morphology, anti-oxidant status, inflammatory response and barrier integrity in male broilers under heat stress. A total of 180 21-d-old male Arbour Acres broilers with similar body weight were used for this 21-d experiment. Broilers were randomly allocated to three groups: CON group, basal diet and raised under 24 °C; HS group, basal diet and raised under heat stress condition (34 °C from 10:00 to 18:00 and 24 °C for the rest time); HS-SB group, basal diet with 1200 mg/kg SB and raised under heat stress condition. Compared to the HS group, SB supplementation increased average daily gain (ADG), villus height: crypt depth ratio (VH: CD), VH, glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activity, and interleukin-10 (IL-10) level, decreased feed conversion ratio (FCR), CD, reactive oxygen species (ROS), IL-1β and tumour necrosis factor-alpha (TNF-α) level, up-regulated the mRNA expression of nuclear factor erythroid 2-related factor 2, haem oxygenase 1, GSH 1 (GPX1), SOD and CAT, down-regulated the mRNA expression of toll-like receptor 4 (TLR4), nuclear transcription factors, IL-1β, and TNF-α in jejunum of heat-stressed broilers. Furthermore, SB supplementation up-regulated the mRNA expression of occluding, claudin-1 and zona occludens-1 (ZO-1) in jejunum of heat-stressed broilers, accompanied with decreasing serum endotoxin and D-lactic acid concentration, and diamine oxidase (DAO) activity. There results suggested that SB supplementation could alleviate heat stress-induced jejunal tight junction structure dysfunction by up-regulated the expression of occluding, claudin-1 and ZO-1, presumably through alleviating oxidative stress and inflammatory response.HIGHLIGHTS Sodium butyrate alleviated heat stress-induced jejunal morphology damage. Bemisia tabaci (Gennadius) is one of the most important invasive species in China, with strong insecticide resistance and thermotolerance. In this study, we investigated the effects of elevated temperature on the tolerance of B. tabaci MEMA1 to abamectin (AB) and thianethixam (TH) insecticides. We firstly cloned two new CYP450 genes from B. tabaci MEAM1, including one CYP6 family gene (BtCYP6k1) and one CYP305 family gene (BtCYP305a1). The expression patterns of the two BtCYP450 genes were compared in response to high-temperature stress and insecticide exposure, and RNAi was then used to demonstrate the role that these two genes play in insecticide tolerance. The results showed that expression of the two BtCYP450 genes could be induced by exposure to elevated temperature or insecticide, but this gene expression could be inhibited to a certain extent when insects were exposed to the combined effects of high temperature and insecticide treatment. For AB treatment, the expression of the two BtCYP450 genes reached the lowest level when insects were exposed to a temperature of 41 °C and treated with AB (combined effects of temperature and insecticide). In contrast, TH treatment showed a general decrease in the expression of the two BtCYP450 genes with exposure to elevated temperatures. These findings suggest that insecticide tolerance in B. tabaci MEAM1 could be mediated by high temperatures. This study provides a prospective method for the more effective application of insecticides for the control of B. tabaci in the field.

The resolving power of metalens telescopes rely on their aperture size. Flat telescopes are advancing with the research on super-resolution confocal metalenses with large aperture. However, the aperture sizes of metalenses are usually bound within hundreds of micrometers due to computational and fabrication challenges, limiting their usage on practical optical devices like telescopes. In this work, we demonstrated a two-step designing method for the design of dual-band far-field super-resolution metalens with aperture sizes from the micro-scale to macro-scale. By utilizing two types of inserted unit cells, the phase profile of a dual-wavelength metalens with a small aperture of 100 μm was constructed. Through numerical simulation, the measured FWHM values of the focal spots of 5.81 μm and 6.81 μm at working wavelengths of 632.8 nm and 1265.6 nm were found to all be slightly smaller than the values of 0.61 λ/NA, demonstrating the super-resolution imaging of the designed metalens. By measuring the optical power ratio of the focal plane and the incident plane, the focusing efficiencies were 76% at 632.8 nm and 64% at 1265.6 nm. Based on the design method for small-aperture metalens, far-field imaging properties through the macro metalens with an aperture of 40 mm were simulated by using the Huygens–Fresnel principle. The simulation results demonstrate confocal far-field imaging behavior at the target wavelengths of 632.8 nm and 1265.6 nm, with a focal length of 200 mm. The design method for dual-band far-field super-resolution metalens with a large aperture opens a door towards the practical applications in the dual-band space telescope system.

Composite materials are widely used in many fields due to their excellent properties. Quality defects in composite materials can lead to lower quality components, creating potential risk of accidents. Experimental and simulation methods are commonly used to predict the quality of composite materials. However, it is difficult to predict the quality of composite materials accurately due to the uncertain curing environment and incomplete feature space. To address this problem, a digital twin (DT) visual model of a composite material is first constructed. Then, a static autoclave DT virtual model is coupled with a variable composite material DT virtual model to construct a model of the curing process. Features are added to the proposed model by generating simulated data to enhance the quality prediction. An extreme learning machine (ELM) for quality prediction is trained with the generated data. Finally, the effectiveness of the proposed method is verified through result analysis.

Vibrations profoundly influence the liquid film formed by spray droplets in spray cooling applications. However, understanding heat transfer mechanisms in the non-boiling region under vibrating conditions is limited in the existing literature. This study aims to bridge this gap by numerically investigating the heat transfer performance of spray cooling in non-boiling regions under vibration using the Lagrange-Euler method. To validate the computational fluid dynamics (CFD) model, experimental measurements of spray heat transfer were conducted on a heated vibrating surface test rig. The results demonstrate that the average film thickness (AFT) and average film velocity (AFV) increase at lower frequencies, leading to a decrease in the heat transfer coefficient. Conversely, the AFT decreases at higher frequencies while the AFV remains relatively constant. This reduction in AFT enhances the heat transfer coefficient, resulting in an up to 12% improvement in heat transfer performance. When a larger vibration amplitude is introduced, a higher inertial force acts on the liquid film, causing it to converge towards the centre. Generally, the heat transfer coefficient of spray cooling in the non-boiling region decreases as the vibration amplitude increases.

Effective vibration control has potential to enhance convective heat transfer. However, the precise heat transfer mechanism of single-phase spray cooling under vibration remains elusive. This study aims to elucidate this mechanism by establishing a closed-loop system for vibrating surface spray cooling. Various vibration parameters, including vibration Reynolds number (Rev), dimensionless acceleration number (Ac), amplitude, and frequency, were investigated experimentally to determine their impact on heat transfer. The research findings demonstrate that the heat flux, heat transfer coefficient, cooling efficiency, and vibration enhancement factor all decrease with an increase in Rev and amplitude. At a heating power of 200 W and Rev of 4947, the spray cooling performance on the vibrating surface is inhibited by 22%. However, as the heating power increases, the inhibitory effects of Rev and amplitude on the spray cooling heat transfer performance are weakened. Conversely, the amplitude and frequency enhance the heat flux, heat transfer coefficient, cooling efficiency, and vibration enhancement factor. At a heating power of 200 W and an Ac of 3.6, the spray cooling heat transfer performance on the vibrating surface is enhanced by 17%. Finally, a dimensionless correlation equation and a Nusselt number (Nu) prediction model for single-phase spray cooling under vibrating conditions are derived and fitted with an error of ±13.2%.

Siglecs are important lectins found in different types of immune cells and function as regulatory molecules by recognizing self-associated glycans and converting extracellular interactions into signals for inhibiting immune cell functions. Although many Siglecs have been found to show broad specificities and recognize different types of sulfated oligosaccharides, Siglec-8 and Siglec-9 displayed a high degree of specificity for sialyl N-acetyllactosamine (sLacNAc) with sulfations at O6-positions of the galactose (6’-sulfation) and N-acetylglucosamine (6-sulfation), respectively. Siglec-3 was recently discovered to bind sLacNAc both sulfations. In addition to a conserved arginine residue for binding to sialic acid residue, the sequence variety in the CC’ loop may provide binding specificities to sulfated oligosaccharides in Siglecs. Thus, the present study employed molecular models to study the impact of different residues in the CC’ loops of Siglec-8/9/3 to the recognitions of 6-sulfations in Gal and/or GlcNAc of sLacNAc. The negatively charged residues in the CC’ loop of Siglec-9 formed unfavorable electrostatic repulsions with the 6-sulfate in Gal and resulted no recognitions, in contrast to the favorable interactions formed between the positively charged residues in the CC’ loop of Siglec-8 and the 6-sulfate in Gal resulting strong specificity. A two-state binding model was proposed for Siglec-3 recognizing 6-sulfations in Gal and GlcNAc of sLacNAc, as the neutral residues in the CC’ loop of Siglec-3 could not form strong favorable interactions to lock the 6-sulfate in Gal within a single binding pose or strong unfavorable interactions to repel the 6-sulfate in Gal. The oligosaccharide adopted two distinctive binding poses and oriented the sulfate groups to form interactions with residues in the CC’ loop and G-strand. The present study provided a structural mechanism for the sequence variety in the CC’ loop of Siglec-8/9/3 determining the recognitions to the sulfated oligosaccharides and offered insights into the binding specificities for Siglecs.

Human respiratory syncytial virus (RSV) is a leading cause of lower respiratory tract infection worldwide. Until now, there are no licenced vaccines or effective antiviral drugs against RSV infections. In our previous work, we found 2-((1H-indol-3-yl)thio/sulfinyl)-N-pheny acetamide derivatives (4-49 C and 1-HB-63) being a novel inhibitor against RSV in vitro. Here, we explored the underlying mechanism of 2-((1H-indol-3-yl)thio/sulfinyl)-N-pheny acetamide derivatives to inhibit RSV replication in vitro and disclosed that 4–49 C worked as the inhibitor of membrane fusion and 1-HB-63 functioned at the stage of RSV genome replication/transcription. Yet, both of them could not inhibit RSV infection of BALB/c mice by using RSV-Luc, in vivo imaging and RT-qPCR analyses, for which it may be due to the fast metabolism in vivo. Our work suggests that further structural modification and optimisation of 2-((1H-indol-3-yl) thio/sulfinyl)-N-pheny acetamide derivative are needed to obtain drug candidates with effective anti-RSV activities in vivo.

Titanium has a low density and high corrosion resistance. In order to achieve the goal of a lightweight material, and to extend the normal working hour of proton-exchange membrane fuel cells (PEMFCs), ultra-thin titanium plates were chosen to manufacture the key components—bipolar plates (BPs). For the purpose of overcoming the challenges of manufacturing with a large depth to width ratio, a multi-stage formation process was established with characteristics such as high efficiency and a lower price. In this study, the process parameters were examined through an experimental approach. The outcomes show that the channel formed by multistage forming is deeper than that formed by single-stage forming under the same displacement conditions. To achieve greater flow depths, it is recommended to increase the displacements as much as possible during both the first- and second-stage forming processes. The implementation of three-stage forming can effectively reduce the maximum thinning rates within flow channels while improving the overall deformation uniformity. This method deviates from traditional one-stage loading processes by adopting multi-stage loading instead. By employing appropriate mold designs, material deformation and flow can be enhanced throughout gradual loading processes, thereby preventing strain concentration and enhancing the ultimate formation height accuracy within micro-flow channels. Consequently, the proposed three-stage forming process proves highly appropriate for the mass production of BPs utilizing titanium plates.

IntroductionCognitive decline in the elderly population is a growing concern, and vascular factors, such as hypertension, diabetes, cerebrovascular disease, and coronary heart disease, have been associated with cognitive impairments. This study aims to provide deeper insights into the structure of cognitive function networks under these different vascular factors and explore their potential associations with specific cognitive domains.MethodsCognitive function was assessed using a modified Chinese version of the mini-mental state examination (MMSE) scale, and intensity centrality and side weights were estimated by network modeling. The network structure of cognitive function was compared across subgroups by including vascular factors as subgroup variables while controlling for comorbidities and confounders.ResultsThe results revealed that cerebrovascular disease and coronary heart disease had a more significant impact on cognitive function. Cerebrovascular disease was associated with weaker centrality in memory and spatial orientation, and a sparser cognitive network structure. Coronary heart disease was associated with weaker centrality in memory, repetition, executive function, recall, attention, and calculation, as well as a sparser cognitive network structure. The NCT analyses further highlighted significant differences between the cerebrovascular disease and coronary heart disease groups compared to controls in terms of overall network structure and connection strength.ConclusionOur findings suggest that specific cognitive domains may be more vulnerable to impairments in patients with cerebrovascular disease and coronary heart disease. These insights could be used to improve the accuracy and sensitivity of cognitive screening in these patient populations, inform personalized cognitive intervention strategies, and provide a better understanding of the potential mechanisms underlying cognitive decline in patients with vascular diseases.

IntroductionCurrently, deep-learning-based prediction of Significant Wave Height (SWH) is mostly performed for a single location in the ocean or simply relies on a single factor (SF). Such approaches have the disadvantage of lacking spatial correlations or dynamic complexity, leading to an inevitable growth of the prediction error with time.MethodsHere, attempting a solution, we develop a Multi-Factor (MF) data-driven 2D SWH prediction model for the Bohai, Yellow, and East China Seas (BYECS). Our model is developed based on a multi-channel PredRNN algorithm that is an improved deep-learning calculation of the ConvLSTM.ResultsIn our model, the MF of historical SWH, 10 m surface winds, ocean surface currents, bathymetries, and open boundaries are used to predict 2D SWH in the next 1-72h. Our modeled SWHs show the correlation coefficients as 0.98, 0.90, and 0.87 for the next 6h, 24h, and 72h, respectively.DiscussionAccording to the ablation experiments, winds are the dominant factor in the MF model and the memory-decoupling module is the key improvement of the PredRNN compared to the ConvLSTM. Furthermore, when the historical SWH is excluded from the input, the correlation coefficients remain around 0.95 in the 1-72h prediction due to the elimination of the error accumulation. It was worse than the MF-PredRNN with the historical SWH before 10h but better than it after 10h. Overall, for the prediction of SWH in the BYECS, our MF-PredRNN-based 2D SWH prediction model significantly improves the accuracy and extends the effective prediction time length.

PurposeNephrolithiasis is thought to be a risk factor for osteoporosis, but data assessing if osteoporosis predisposes to the risk of nephrolithiasis are lacking. The present study aims to investigate whether patients with nephrolithiasis have a prominently higher prevalence of osteoporosis than the controls and vice versa via a cumulative analysis.MethodsFour databases were used to detect the eligible studies. We calculated the relative risk (RR) with a 95% confidence interval (CI) to assess the combined effect. The methodologies for conducting this study followed the PRISMA guidelines and were registered in the PROSPERO (ID: CRD42023395875),ResultsNine case-control or cohort studies with a total of 454,464 participants were finally included. Combined results indicated that there was a significantly higher prevalence of osteoporosis in patients with nephrolithiasis as compared to the general population without nephrolithiasis (overall RR from six studies= 1.204, 95%CI: 1.133 to 1.28, P< 0.001; heterogeneity: I2 = 34.8%, P= 0.162). Conversely, osteoporosis was significantly correlated to an increased risk of nephrolithiasis as compared to the controls without osteoporosis (overall RR from four studies= 1.505, 95%CI: 1.309 to 1.731, P< 0.001; I2 = 89.8%, P< 0.001). Sensitivity analysis on the two categories validated the above findings. No significant publication bias was identified in this study.ConclusionsThe present study highlighted a significantly high prevalence of osteoporosis in patients with nephrolithiasis and vice versa. Ordered intermetallic Pt-M alloys are promising as next-generation low-Pt catalysts for oxygen reduction reaction (ORR) in proton exchange membrane fuel cells (PEMFCs) due to the high ORR activity and excellent long-term stability. However, the mechanism behind the enhancement of ORR activity by ordering mostly comes from theoretical calculations, and experimental evidence is very lacking. Herein, we prepared a surface Pt-rich PtCo ordered alloy of about 3 nm in size, which exhibited high ORR catalytic activity, achieving impressive performance of 1.4 W cm−2 @ 0.6 V in H2-air fuel cell and mass activity of 0.62 A mgPt-1 @ 0.9 ViR-free in H2-O2 fuel cell. The key ORR intermediate, adsorbed *O2, was successfully detected by electrochemical in situ infrared spectroscopy. Compared with Pt and disordered PtCo, the ordered PtCo alloy has optimal adsorption interaction with *O2, accelerating the ORR kinetics.

In this paper, the fault detection and isolate problem is investigated for the pitch system of floating wind turbine. In the addressed system model, the system noises and measurement noises are correlated, and the measurement is affected by the missing phenomena. A Kalman filter is designed to handle the correlated noises and estimate the pitch angle, and a residual of the measurement of the pitch system is constructed to detection the faults. Then the fault isolation algorithm is presented based on a multi-attention mechanism one-dimensional convolutional neural network, which is employed to accurately isolate the faults. The simulation results show that the proposed method can significantly improve the accuracy of fault detection and isolation, which the fault isolation accuracy of the simulation results reaches 99.15%.

Virtual marine scene authoring plays an important role in generating large-scale 3D scenes and it has a wide range of applications in computer animation and simulation. Existing marine scene authoring methods either produce periodic patterns or generate unnatural group distributions when tiling marine entities such as schools of fish and groups of reefs. To this end, we propose a new large-scale marine scene authoring method based on real examples in order to create more natural and realistic results. Our method first extracts the distribution of multiple marine entities from real images to create Octahedral Blocks, and then we use a modified Wang Cubes algorithm to quickly tile the 3D marine scene. As a result, our method is able to generate aperiodic tiling results with diverse distributions of density and orientation of entities. We validate the effectiveness of our method through intensive comparative experiments. User study results show that our method can generate satisfactory results which are in accord with human preferences.

Abstract Aims Right ventricular pacing (RVP) dependence could impair left ventricular ejection fraction (LVEF). This study aimed to illuminate the relationship between RVP proportion and LVEF, as well as disclosing independent predictors of RVP dependence. Methods and results Patients indicated for permanent pacemaker implantation were included (2016–2020). The ventricular pacing lead was placed in right ventricular apex or septum. Pacing mode programming followed universal standard. Electrocardiographic, echocardiographic, and serological parameters were collected. RVP dependence was defined according to its influence on LVEF. This study was of case–control design. Included patients were matched by potentially confounding factors through propensity score matching. A total of 1183 patients were included, and the mean duration of follow‐up was 24 months. Percentage of RVP 120 ms (OR = 2.46, 95% CI: 1.42–4.27), QTc interval > 470 ms (OR = 2.01, 95% CI: 1.33–3.05), and pulmonary artery systolic pressure (PASP) > 40 mmHg (OR = 1.93, 95% CI: 1.46–2.56) were proved to predict RVP dependence. Conclusions High RVP percentage (>80%) indicating RVP dependence significantly correlates with poor prognosis of cardiac function. High degree/complete AVB, AF, ischaemic aetiology, PCI history, HRmax 120 ms, QTc interval > 470 ms, and PASP > 40 mmHg were verified as independent risk factors of RVP dependence.

Suxiao Jiuxin Pill (SJP) is a well-known traditional Chinese medicine drug used to manage heart diseases. This study aimed at determining the pharmacological effects of SJP in acute myocardial infarction (AMI), and the molecular pathways its active compounds target to induce coronary artery vasorelaxation. Using the AMI rat model, SJP improved cardiac function and elevated ST segment. LC-MS and GC-MS detected twenty-eight non-volatile compounds and eleven volatile compounds in sera from SJP-treated rats. Network pharmacology analysis revealed eNOS and PTGS2 as the key drug targets. Indeed, SJP induced coronary artery relaxation via activation of the eNOS-NO pathway. Several of SJP’s main compounds, like senkyunolide A, scopoletin, and borneol, caused concentration-dependent coronary artery relaxation. Senkyunolide A and scopoletin increased eNOS and Akt phosphorylation in human umbilical vein endothelial cells (HUVECs). Molecular docking and surface plasmon resonance (SPR) revealed an interaction between senkynolide A/scopoletin and Akt. Vasodilation caused by senkyunolide A and scopoletin was inhibited by uprosertib (Akt inhibitor) and eNOS/sGC/PKG axis inhibitors. This suggests that senkyunolide A and scopoletin relax coronary arteries through the Akt-eNOS-NO pathway. In addition, borneol induced endothelium-independent vasorelaxation of the coronary artery. The Kv channel inhibitor 4-AP, KCa2+ inhibitor TEA, and Kir inhibitor BaCl2 significantly inhibited the vasorelaxant effect of borneol in the coronary artery. In conclusion, the results show that Suxiao Jiuxin Pill protects the heart against acute myocardial infarction.

Synchronizing a few-level quantum system is of fundamental importance to the understanding of synchronization in the deep quantum regime. Whether a two-level system, the smallest quantum system, can be synchronized has been theoretically debated for the past several years. Here, for the first time, we demonstrate that a qubit can indeed be synchronized to an external driving signal by using a trapped-ion system. By engineering fully controllable gain and damping processes, an ion qubit is locked to the driving signal and oscillates in phase. Moreover, upon tuning the parameters of the driving signal, we observe characteristic features of the Arnold tongue as well. Our measurements agree remarkably well with numerical simulations based on recent theory on qubit synchronization. By synchronizing the basic unit of quantum information, our study opens up the possibility of exploring the application of quantum synchronization to quantum information processing in the near future.

Particle counting serves as a pivotal constituent in diverse analytical domains, encompassing a broad spectrum of entities, ranging from blood cells and bacteria to viruses, droplets, bubbles, wear debris, and magnetic beads. Recent epochs have witnessed remarkable progressions in microfluidic chip technology, culminating in the proliferation and maturation of microfluidic chip-based particle counting methodologies. This paper undertakes a taxonomical elucidation of microfluidic chip-based particle counters based on the physical parameters they detect. These particle counters are classified into three categories: optical-based counters, electrical-based particle counters, and other counters. Within each category, subcategories are established to consider structural differences. Each type of counter is described not only in terms of its working principle but also the methods employed to enhance sensitivity and throughput. Additionally, an analysis of future trends related to each counter type is provided.

Podocyte injury is an important cause of proteinuria. Angiopoietin-like protein 4 (Angptl4) is a secreted glycoprotein and has a role in proteinuria. However, the exact role of Angptl4 in podocyte injury and its upstream regulators has not been clarified. In this study, we used lipopolysaccharide (LPS)-induced mice and cultured podocytes as podocyte injury models. Our results indicated that LPS increased the expression of podocyte Angptl4 in vivo and in vitro. Furthermore, we showed that Angptl4 overexpression deteriorated LPS-induced podocyte injury by inducing podocyte cytoskeleton rearrangement, reducing the expression of synaptopodin while Angptl4 knockdown alleviated LPS-induced podocyte injury. In addition, we found that inhibitors and siRNA targeting TLR4/MyD88/NF-κB signaling inhibited the upregulation of Angptl4 in LPS-induced podocytes. Moreover, inhibitors and siRNA targeting calcineurin/NFAT signaling also relieved LPS-induced Angptl4 expression and podocyte injury in vivo and in vitro. Taken together, our study has elucidated that both of the TLR4/MyD88/NF-κB and calcineurin/NFAT signaling mediate the upregulation of Angptl4 in LPS-induced podocytes, which has important implications for further understanding the molecular mechanism of LPS-induced podocyte injury.

BackgroundAcute rejection is a determinant of prognosis following kidney transplantation. It is essential to search for novel noninvasive biomarkers for early diagnosis and prompt treatment.MethodsGene microarray data was downloaded from the Gene Expression Omnibus (GEO) expression profile database and the intersected differentially expressed genes (DEGs) was calculated. We conducted the DEGs with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. Distribution of immune cell infiltration was calculated by CIBERSORT. A hub gene marker was identified by intersecting the rejection-related genes from WGCNA and a selected KEGG pathway—T cell receptor signaling pathway (hsa04660), and building a protein-protein interaction network using the STRING database and Cytoscape software. We performed flow-cytometry analysis to validate the hub gene.ResultsA total of 1450 integrated DEGs were obtained from five datasets (GSE1563, GSE174020, GSE98320, GSE36059, GSE25902). The GO, KEGG and immune infiltration analysis results showed that AR was mainly associated with T cell activation and various T-cell related pathways. Other immune cells, such as B cells, Macrophage and Dendritic cells were also associated with the progress. After utilizing the WGCNA and PPI network, PDCD1 was identified as the hub gene. The flow-cytometry analysis demonstrated that both in CD4+ and CD8+ T cells, PD1+CD57-, an exhausted T cell phenotype, were downregulated in the acute rejection whole blood samples.ConclusionsOur study illustrated that PDCD1 may be a candidate diagnostic biomarker for acute kidney transplant rejection via integrative bioinformatic analysis.

Summary: Searching for an efficient, durable, and low cost catalyst toward oxygen reduction reaction (ORR) is of paramount importance for the application of fuel cell technology. Herein, PtFeCoNiCu high-entropy alloy nanoparticles (PFCNC-HEA) is reported as electrocatalyst toward ORR. It shows remarkable ORR catalytic mass activity of 1.738 A mg−1Pt at 0.90 V, which is 15.8 times higher than that of the state-of-art commercial Pt/C catalyst. It also exhibits outstanding stability with negligible voltage decay (3 mV) after 10k cycles accelerated durability test. High ORR activity is ascribed to the ligand effect caused by polymetallic elements, the optimization of the surface electronic structure, and the formation of multiple active sites on the surface. In the proton exchange membrane fuel cell setup, this cell delivers a power density of up to 1.380 W cm−2 with a cathodic Pt loading of 0.03 mgPt cm−2, demonstrating a promising catalyst design direction for highly efficient ORR.

Background Nonsuicidal self-injury (NSSI) behaviors—an important factor that profoundly affects the physical and mental health of young people—are induced by complex and diverse factors, while showing significant differences at the gender level. We examined mediating behaviors among parenting styles, students’ coping styles, and endogenous and exogenous influencing variables of adolescents’ NSSI behaviors. Methods In this cross-sectional study, Secondary school students in Ningbo, Zhejiang Province, China (n = 2,689; F/M:1532/1157) were surveyed for basic attributes, parenting styles, coping styles, and NSSI behaviors. After the initial screening of the sample data, several external derivatives were screened based on the single factor analysis method. On this basis, the construction of path analysis models under multivariate multiple elicitations was carried out. Results The overall prevalence of NSSI was 15.16%, and the incidence of NSSI in boys was lower than that in girls (OR = 0.334, 95% CI [0.235–0.474]). The path analysis model data fit well; the indicators of female and male part are: CFI = 0.913/0.923, GFI = 0.964/0.977, SRMR = 0.055/0.047, RMSEA = 0.097/0.069 with 90% confidence interval (CI) [0.084–0.111]/[0.054–0.084]. For female, when negative coping style and extreme education affect NSSI respectively, the standardized path coefficient values are 0.478 (z = 20.636, P = 0.000 < 0.01) and 0.151 (z = 6.524, P = 0.000 < 0.01) respectively, while for male, the corresponding values become 0.225 (z = 7.057, P < 0.001) and 0.104 (z = 3.262, P < 0.001). Conclusion In particular, we investigated the mediating effects of gender-specific NSSI influences and found that NSSI behaviors were strongly associated with environmental variables and individual factors, especially family parenting style and adolescent coping style, which influenced NSSI in a gender-specific manner. The results showed that males were the target of both positive and negative parenting styles, whereas females were more likely to choose negative coping styles directed towards emotions in response to external stimuli, and instead showed a more significant predisposition towards NSSI behaviors. This phenomenon seems to be influenced by multilevel factors such as sociocultural, individual value identity, and physiological structure differences. In the path analysis model with the introduction of mediating effects, the influence of gender differences on NSSI behavior becomes more pronounced under the interaction of multiple factors: women seem to be more significantly influenced by the external derivatives in the internal derivatives than male subjects, and are more likely to trigger NSSI behavior under the interaction of multiple factors. These findings effectively reveal the significant role of different end-influencing factors in NSSI behaviors at the level of gender differences, which can provide effective theoretical support to prevent and treat NSSI behaviors in adolescents.

BackgroundAcute allograft rejection (AR) following renal transplantation contributes to chronic rejection and allograft dysfunction. The current diagnosis of AR remains dependent on renal allograft biopsy which cannot immediately detect renal allograft injury in the presence of AR. In this study, sensitive biomarkers for AR diagnosis were investigated and developed to protect renal function.MethodsWe analyzed pre- and postoperative data from five databases combined with our own data to identify the key differently expressed genes (DEGs). Furthermore, we performed a bioinformatics analysis to determine the immune characteristics of DEGs. The expression of key DEGs was further confirmed using the real-time quantitative PCR (RT-qPCR), enzyme-linked immunosorbent assay (ELISA), and immunohistochemical (IHC) staining in patients with AR. ROC curves analysis was used to estimate the performance of key DEGs in the early diagnosis of AR.ResultsWe identified glutamic-oxaloacetic transaminase 2 (GOT2) and syntaxin binding protein 3 (STXBP3) as key DEGs. The higher expression of STXBP3 and GOT2 in patients with AR was confirmed using RT-qPCR, ELISA, and IHC staining. ROC curve analysis also showed favorable values of STXBP3 and GOT2 for the diagnosis of early stage AR.ConclusionsSTXBP3 and GOT2 could reflect the immunological status of patients with AR and have strong potential for the diagnosis of early-stage AR.

Equilibrium Optimizer (EO) is a newly developed intelligent optimization algorithm inspired by control volume mass balance models. EO has been proven to have an excellent solution effect on some optimization problems, with the advantages of ease of implementation and strong adaptability. However, the original EO has some disadvantages when solving complex multimodal problems, including an immature balance between exploration and exploitation, the high probability of falling into local optima entrapment, and the slow rate of convergence. In order to address these shortcomings, a modified equilibrium optimizer (OTLEO) is proposed using teaching-learning-based optimization (TLBO) and opposition-based learning (OBL). These modifications aim to maintain the diversity of the solutions, expand the algorithm’s search range, improve exploration and exploitation capabilities, and avoid local optima. To verify and analyze the modified equilibrium optimizer algorithm’s performance, the OTLEO was tested for 32 classical benchmark functions. All algorithms are independently run 30 times in the same environment. Thereafter, the comparative evaluation against the OTLEO and other six representative algorithms is conducted. Four real-world engineering application problems, including multiple sequence alignment and so on, are used for additional validation. The experimental results, statistical tests, qualitative analysis, and stability analysis demonstrate that the proposed OTLEO outperforms the original EO and other algorithms.

Background: Leukocyte telomere length shortening is a characteristic of premature senescence, a process that can be accelerated by oxidative stress. In general, patients with end-stage renal disease undergoing regular hemodialysis (HD) are repeatedly exposed to oxidative stress. Patients undergoing HD tend to have cardiovascular diseases associated with oxidative stress and inflammation. Therefore, we assumed that telomere length is associated with HD vintage and the degree of vascular calcification. Methods: A total of 144 patients undergoing regular HD before kidney transplantation and 62 patients on hemodialysis, but not undergoing kidney transplantation, were enrolled. We measured common laboratory values, such as calcium, phosphate, and hemoglobin levels, and assessed the degree of vascular calcification in the patients. The leukocyte telomere length was measured using reverse transcription polymerase chain reaction, and Spearman correlation was used for correlation analysis. Results: The leukocyte telomere length was negatively associated with age (rho = −0.306, P＜0.01); it was shorter in middle-aged patients than in young patients (13.48 ± 4.80 vs. 15.86 ± 4.51, P

Biofilm-associated infections exert more severe and harmful attacks on human health since they can accelerate the generation and development of the antibiotic resistance of the embedded bacteria. Anti-biofilm materials and techniques that can eliminate biofilms effectively are in urgent demand. Therefore, we designed a type I photosensitizer (TTTDM) with an aggregation-induced emission (AIE) property and used F-127 to encapsulate the TTTDM into nanoparticles (F-127 AIE NPs). The NPs exhibit highly efficient ROS generation by enhancing intramolecular D–A interaction and confining molecular non-radiative transitions. Furthermore, the NPs can sufficiently penetrate the biofilm matrix and then detect and eliminate mature bacterial biofilms upon white light irradiation. This strategy holds great promise for the rapid detection and eradication of bacterial biofilms.

Leak monitoring is essential for the intelligent operation and maintenance of marine systems, and can effectively prevent catastrophic accidents on ships. In response to this challenge, a machine vision-based leak model is proposed in this study and applied to leak detection in different types of marine system in complex engine room environments. Firstly, an image-based leak database is established, and image enhancement and expansion methods are applied to the images. Then, Standard Convolution and Fast Spatial Pyramid Pooling modules are added to the YOLOv5 backbone network to reduce the floating-point operations involved in the leak feature channel fusion process, thereby improving the detection speed. Additionally, Bottleneck Transformer and Shuffle Attention modules are introduced to the backbone and neck networks, respectively, to enhance the feature representation performance, select critical information for the leak detection task, and suppress non-critical information to improve detection accuracy. Finally, the proposed model’s effectiveness is verified using leak images collected by the ship’s video system. The test results demonstrate that the proposed model exhibits excellent recognition performance for various types of leak, especially for drop-type leaks (for which the accuracy reaches 0.97).

The pH of saline–alkali soil is high because of carbonate salts, and the deleterious effects of saline–alkali soil on the growth of plants are greater than those of saline soil. Few studies have examined the saline–alkali tolerance of Betula platyphylla at the molecular level. To clarify the regulatory mechanism underlying saline–alkali tolerance in B. platyphylla, RNA sequencing analysis of B. platyphylla seedlings treated with NaHCO3 was conducted. Differences in gene expression in the roots of B. platyphylla seedlings under saline–alkali stress (induced via NaHCO3) for 3 h and 6 h were characterized, and a total of 595 and 607 alkali stress-responsive genes were identified, respectively. Most differentially expressed genes were involved in stress, signal transduction, secondary metabolic process, regulation of jasmonic acid, and the abiotic stimulus signaling pathway. The single nucleotide polymorphism loci in the differentially expressed genes were associated with the alkaline-salt tolerance in birch germplasm. In addition, birch plants overexpressing WRKY70 and NAC9 were obtained using the A. tumefaciens-mediated transient transformation method, and these two genes were found to play key roles in saline–alkali tolerance. Additional study revealed that WRKY70 and NAC9 can increase resistance to saline–alkali stress by enhancing reactive oxygen species scavenging and inhibiting cell death in birch plants. The results of this study enhance our understanding of the saline–alkali stress tolerance of B. platyphylla at the molecular level, and provide several key genes that could be used in the breeding of birch plants in the future.

The composition of marine aerosol is quite complex, and its sources are diverse. Across the East China Sea (ECS) and the Yellow Sea (YS), multi-dimensional analysis of marine aerosols was conducted. The characteristics of carbonaceous aerosols and gaseous pollutants were explored through in situ ship-based observation, MERRA-2 reanalysis datasets and TROPOMI data from Sentinel-5P satellite. Black carbon (BC)’s average concentration is 1.35 ± 0.78 μg/m3, with high-value BC observed during the cruise. Through HYSPLIT trajectory analysis, sources of BC were from the northern Eurasian continent, the Shandong Peninsula, the ECS and Northwest Pacific Ocean (NWPO). The transport of marine sources like ship emissions cannot be ignored. According to the absorption Angstrom exponent (AAE), BC originates from biomass burning (BB) in the shortwave band (~370 nm) and from fossil fuel combustion in the longwave band (~660 nm). Organic carbon (OC), sulfate (SO42−) and BC report higher Angstrom exponent (AE) while dust and sea salt reveal lower AE, which can be utilized to classify the aerosols as being fine- or coarse-mode, respectively. OC has the highest AE (ECS: 1.98, YS: 2.01), indicating that anthropogenic activities could be a significant source. The process of biomass burning aerosol (BBA) mixed with sea salt could contribute to the decline in BBA’s AE. Ship emissions may affect the distribution of tropospheric nitrogen dioxide (NO2) in the ECS, especially during the COVID-19 pandemic. Tropospheric NO2 over the YS has the highest value (up to 12 × 1015 molec/cm2). Stratospheric NO2 has a ladder-like distribution from north to south, and the variation gradient was lower than that in the troposphere. Carbon monoxide (CO) accumulates in the south and east of the ECS and the east of the YS, while the variation over the eastern YS is relatively frequent. Seas near the Korean Peninsula have extremely high CO concentration (up to 1.35 × 1017 molec/cm2).

This paper employs regression discontinuity design to estimate the impact of heating modes on residents’ energy consumption, energy efficiency gap, and energy usage behavior, using the Chinese Residential Energy Consumption Survey (CRECS). We find that district heating increases total energy consumption and total heating expenditure. However, energy consumption and expenditure per unit of district heating are lower than self-heating, which means a positive energy efficiency gap between district heating and self-heating. With the estimates, we construct a model of household heating expenditure. The counterfactual simulation shows that district heating expenditure would be reduced by more than 35% if the heating fee is charged based on actual usage. We suggest government provide a quantity-based district heating service by implementing a two-part tariff pricing strategy.

Previously, we have shown that the transcription factor BplMYB46 in Betula platyphylla can enhance tolerance to salt and osmotic stress and promote secondary cell wall deposition, and we characterized its downstream regulatory mechanism. However, its upstream regulatory mechanism remains unclear. Here, the promoter activity and upstream regulatory factors of BplMYB46 were studied. Analyses of β-glucuronidase (GUS) staining and activity indicated that BplMYB46 promoter was specific temporal and spatial expression, and its expression can be induced by salt and osmotic stress. We identified three upstream regulatory factors of BplMYB46: BpDof1, BpWRKY3, and BpbZIP3. Yeast-one hybrid assays, GUS activity, chromatin immunoprecipitation, and quantitative real-time polymerase chain reaction revealed that BpDof1, BpWRKY3, and BpbZIP3 can directly regulate the expression of BplMYB46 by specifically binding to Dof, W-box, and ABRE elements in the BplMYB46 promoter, respectively. BpDof1, BpWRKY3, and BpbZIP3 were all localized to the nucleus, and their expressions can be induced by stress. Overexpression of BpDof1, BpWRKY3, and BpbZIP3 conferred the resistance of transgenic birch plants to salt and osmotic stress. Our findings provide new insights into the upstream regulatory mechanism of BplMYB46 and reveal new upstream regulatory genes that mediate resistance to adverse environments. The genes identified in our study provide novel targets for the breeding of forest tree species.

Objective: Parental and peer support are both associated with mental distress and unhealthy behaviour indices in adolescents.Methods: We used the Global School-Based Student Health Survey data (n = 192,633) from 53 countries and calculated the weighted prevalence of individual and combined parental and peer support. Multiple logistic regression analysis was used to estimate the adjusted associations between combined parental and peer support with mental distress and unhealthy behaviours.Results: The prevalence figures for having all four categories of parental support and two peer-support were 9.7% and 38.4%, respectively. Compared with no parental support, adolescents with all four parental support negatively associated with all five mental distress and eight unhealthy behaviours factors, and the ORs ranged from 0.19 to 0.75. Additionally, adolescents with two peer support were negative association with all mental distress and four health risk behaviours, and positively associated with a sedentary lifestyle.Conclusion: Parental and peer support were lacking in some countries, while greater parental and peer support were negative associated with mental distress and most unhealthy behaviours in adolescents, and the relationships were independent.

The GRAS gene family is a plant-specific family of transcription factors and play a vital role in many plant growth processes and abiotic stress responses. Nevertheless, the functions of the GRAS gene family in woody plants, especially in Betula platyphylla (birch), are hardly known. In this study, we performed a genome-wide analysis of 40 BpGRAS genes (BpGRASs) and identified typical GRAS domains of most BpGRASs. The BpGRASs were unevenly distributed on 14 chromosomes of birch and the phylogenetic analysis of six species facilitated the clustering of 265 GRAS proteins into 17 subfamilies. We observed that closely related GRAS homologs had similar conserved motifs according to motif analysis. Besides, an analysis of the expression patterns of 26 BpGRASs showed that most BpGRASs were highly expressed in the leaves and responded to salt stress. Six BpGRASs were selected for cis-acting element analysis because of their significant upregulation under salt treatment, indicating that many elements were involved in the response to abiotic stress. This result further confirmed that these BpGRASs might participate in response to abiotic stress. Transiently transfected birch plants with transiently overexpressed 6 BpGRASs and RNAi-silenced 6 BpGRASs were generated for gain- and loss-of-function analysis, respectively. In addition, overexpression of BpGRAS34 showed phenotype resistant to salt stress, decreased the cell death and enhanced the reactive oxygen species (ROS) scavenging capabilities and proline content under salt treatment, consistent with the results in transiently transformed birch plants. This study is a systematic analysis of the GRAS gene family in birch plants, and the results provide insight into the molecular mechanism of the GRAS gene family responding to abiotic stress in birch plants.

Liver fibrosis results from the chronic liver injury and no specific medical therapy is approved so far. Recently, new compound, N-(3,4,5-trichlorophenyl) − 2 (3-nitrobenzenesulfonamido) benzamide, referred to as IMB16–4, was developed to resist liver fibrosis. However, IMB16–4 displays poor aqueous solubility and poor oral bioavailability. To increase the dissolution rate, improve the oral bioavailability and enhance the anti-hepatic fibrosis action of IMB16–4, IMB16–4 self-emulsifying drug delivery systems (SEDDS) with negative charge or positive charge were prepared using simple stirring, respectively. Their stability, oral bioavailability and anti-liver fibrosis effect were evaluated. The results showed IMB16–4 SEDDS in simulated gastric juice were nearly spherical with the diameter of 100～200 nm and possessed good stability in 30 days. The oral bioavailability of IMB16–4 SEDDS with negative charge and positive charge were increased to 33 folds and 58 folds compared with that of pure IMB16–4, respectively. In bile duct ligation (BDL) rats, IMB16–4 SEDDS attenuated the degree of liver damage and decreased collagen accumulation. In addition, IMB16–4 SEDDS with negative charge easily accumulated in the liver and alleviated hepatic fibrosis by TGF-β/Smad signaling. These findings indicate that IMB16- 4 SEDDS may be a potential therapy for the treatment of liver fibrosis.
