7. New G-Triplex DNA Dramatically Activates the Fluorescence of Thioflavin T and Acts as a Turn-On Fluorescent Sensor for Uracil-DNA Glycosylase Activity Detection

Abstract

G-triplexes are G-rich oligonucleotides composed of three G-tracts and have absorbed much attention due to their potential biological functions and attractive performance in biosensing. Through the optimization of loop compositions, DNA lengths, and 5′-flanking bases of G-rich sequences, a new stable G-triplex sequence with 14 bases (G3-F15) was discovered to dramatically activate the fluorescence of Thioflavin T (ThT), a water-soluble fluorogenic dye. The fluorescence enhancement of ThT after binding with G3-F15 reached 3200 times, which was the strongest one by far among all of the G-rich sequences. The conformations of G3-F15 and G3-F15/ThT were studied by circular dichroism. The thermal stability measurements indicated that G3-F15 was a highly stable G-triplex structure. The conformations of G3-F15 and G3-F15/ThT in the presence of different metal cations were studied thoroughly by fluorescent spectroscopy, circular dichroism, and nuclear magnetic resonance. Furthermore, using the G3-F15/ThT complex as a fluorescent probe, a robust and simple turn-on fluorescent sensor for uracil-DNA glycosylase activity was developed. This study proposes a new systematic strategy to explore new functional G-rich sequences and their ligands, which will promote their applications in diagnosis, therapy, and biosensing.

Abstract

An advance Li‐sphere possessing a definitely regular morphology in Li deposition enables a well‐defined more robust structure and superior solid‐electrolyte interphase (SEI) to achieve high‐efficiency long‐term cycles in Li metal anode. Here, a new sight of high Li+ cluster‐like solvation sheaths coordinated in a localized high‐concentration NO3− (LH‐LiNO3) electrolyte fully clarifies for depositing advanced Li spheres. Moreover, we elucidate a critical amorphous‐crystalline phase transition in the nanostructure evolution of Li‐sphere deposits during the nucleation and growth. Li‐sphere anode exhibits ultra‐stable structural engineering for suppressing Li dendrite growths and rendering ultralong life of 4000 cycles in symmetrical cells at 2 mA cm−2. The as‐constructed Li spheres/3DCM|LiFePO4 (LFP) full cell delivers a high capacity retention of 90.5% at 1 C after 1000 cycles, and a robust dendrite‐free structure also stably exists in Li‐sphere anode. Combined with high‐loading LFP cathodes (6.6 and 10.9 mg cm−2), superb capacity retentions are up to 96.5% and 92.5% after 800 cycles at 1 C, respectively. Cluster‐like solvation sheaths with high Li+ coordination exert significant influence on depositing a high‐quality Li‐sphere anode. [ABSTRACT FROM AUTHOR]

Abstract

A magnetic beads based immunoaffinity capillary electrophoresis method for total Immunoglobulin E quantification in serum has been developed. The method combines speed, automation ability, and minimal sample consumption. Only 1 [micro]L of serum is required while the whole immunoaffinity capillary electrophoresis method is performed in less than 50 min. The concomitant use of online immunocapture, transient isotachophoresis, and laser-induced fluorescence detection provides a sensitivity in the low picomolar range and a highly linear fluorescence response over 4 orders of magnitude (IgE concentration ranging from 2.4 to 2400 ng/mL). After validation with a reference material, the method has been successfully applied to the quantification of total IgEs in patient sera. The results compared well with classical ImmunoCap data.

Abstract

A competitive immunoassay for estradiol (E2) based on capillary electrophoresis was established. This method was based on the competitive reaction of complete antigen and E2 with a limited amount of monoclonal antibody, with fluorescein isothiocyanate (FITC)-labeled secondary antibody as a fluorescence probe. The addition of the thermally reversible hydrogel, poly-N-isopropylacrylamide (pNIPA) in the buffer as a replaceable packing material improved reproducibility and resolution of the method. This capillary electrophoresis immunoassay with laser-induced fluorescence can be applied to determine E2 with good precision at concentrations as low as 9 pg/mL. Details of typical separations of immunological complex and free reactants are presented.
