This study evaluated the effectiveness and safety of anlotinib with or without programmed death-1 (PD-1) blockades for patients with advanced primary HCC in a real-world setting in China.Patients and Methods: Between July 2019 and May 2021, 27 patients with advanced primary HCC who received at least 2 cycles of anlotinib were included in this retrospective study. Primary endpoint was objective response rate (ORR). Secondary endpoints were disease control rate (DCR), progression-free survival (PFS), overall survival (OS), and safety.Results: Of the 27 patients, ORR and DCR were 25.93% and 74.07%, respectively. The median follow-up time was 6.27 months (range: 1.30– 17.40) with a median PFS and OS of 3.29 months (95% CI: 1.31– 15.47) and 6.21 months (95% CI: 2.23– 15.87), respectively. A total of 14 patients received anlotinib and PD-1 blockade combination therapy, and 13 received anlotinib monotherapy. No significant differences were observed in ORR (28.57 vs 23.08%), DCR (71.43 vs 76.92%), PFS (3.38 [95% CI: 2.66– 13.14] vs 11.86 months [95% CI: 4.27– 15.93]) and OS (4.90 [95% CI: 2.56– 13.60] vs 11.04 months [95% CI: 1.31– 17.18]) between the two groups (all p> 0.05). Treatment-related AEs were reported in 88.89% of patients. Grade 3 AE was bleeding, which occurred in 3 patients (11.11%).Conclusion: Anlotinib yielded a promising efficacy and manageable safety in patients with advanced primary HCC irrespective of whether patients received PD-1 blockades, indicating that anlotinib might be a promising treatment option for this patient population.Keywords: anlotinib, objective response rate, PD-1 blockades, primary hepatocellular carcinoma

In the US FDA database, there are 95 approved pharmaceuticals that stem from pyridine or dihydropyridine, including isoniazid and ethionamide (tuberculosis), delavirdine (HIV/AIDS), abiraterone acetate (prostate cancer), tacrine (Alzheimer’s), ciclopirox (ringworm and athlete’s foot), crizotinib (cancer), nifedipine (Raynaud’s syndrome and premature birth), piroxicam (NSAID for arthritis), nilvadipine (hypertension), roflumilast (COPD), pyridostigmine (myasthenia gravis), and many more. Their remarkable therapeutic applications have encouraged researchers to prepare a larger number of biologically active compounds decorated with pyridine or dihydropyridine, expandeing the scope of finding a cure for other ailments. It is thus anticipated that myriad new pharmaceuticals containing the two heterocycles will be available in the forthcoming decade. This review examines the prospects of highly potent bioactive molecules to emphasize the advantages of using pyridine and dihydropyridine in drug design. CB helps to eliminate heat and toxic components, and prevents the accumulation of phlegm and blood stasis in the liver. In Asian countries, CB Sativus (CBS), an ideal substitute for natural CB, is presently extensively used for long-term treatment of chronic liver diseases. The present study aimed to evaluate the effects and potential mechanism(s) of action of CBS on mice with fructose-induced nonalcoholic fatty liver disease (NAFLD). The NAFLD model was established in C57BL/6 mice by exclusively feeding fluids containing 30% fructose for 8 consecutive weeks. After these 8 weeks, mice were given CBS (50 mg/kg/day or 100 mg/kg/day) for 2 consecutive weeks. Treatment with CBS reversed the fructose-induced impaired glucose tolerance. Compared with the model group, in which mice received 8 weeks of high-fructose diet and 2 weeks of 0.5% sodium carboxymethyl cellulose, CBS treatment significantly decreased the levels of fasting serum glucose, fasting insulin, triglyceride, and total cholesterol, and increased levels of high-density lipoprotein-cholesterol. CBS treatment also significantly decreased the levels of triglyceride, total cholesterol, and free fatty acid in the liver. The activity of superoxide dismutase in the liver was increased after treatment with CBS, however, levels of malondialdehyde and reactive oxygen species decreased. Histopathological examination showed that liver steatosis and injury were significantly reduced in CBS-treated mice. The expression of fatty acid synthase, nuclear factor kappa-light-chain-enhancer of activated B cells, Cysteinyl aspartate-specific proteinase-3, and synonyms B-cell leukemia/lymphoma-2 gene-associated X protein were downregulated after treatment with CBS, whereas the expression of nuclear factor erythroid-2-related factor 2 was upregulated. In conclusion, CBS treatment exerted therapeutic effects in the liver of mice with NAFLD, which may be associated with amelioration of metabolic disorders, enhanced antioxidant effects, and alleviation of apoptosis. Sophia Rangwala, Madeleine Duvic, Chunlei ZhangDepartment of Dermatology, The University of Texas MD Anderson Cancer Center, Houston, Texas, USAAbstract: Epigenetic modification with small molecule histone deacetylase inhibitors has been a promising new anti-neoplastic approach for various solid and hematological malignancies, particularly cutaneous T-cell lymphoma (CTCL). Oral vorinostat was the first histone deacetylase inhibitor approved to enter the clinical oncology market for treating CTCL patients who have progressive, persistent, or recurrent disease after failing two systemic therapies. In two phase II clinical trials, oral vorinostat was found to be safe and effective at a dose of 400 mg/day, with an overall response rate of 24%–30% in heavily pretreated patients with advanced CTCL, including those with large-cell transformed mycosis fungoides and Sézary syndrome. About half of CTCL patients receiving vorinostat also experienced substantial relief in pruritus and thus a marked improvement in quality of life. A subsequent follow-up study reported long-term safety and clinical benefits of vorinostat in patients with refractory CTCL, regardless of previous treatment failures. The most frequent side effects of vorinostat include gastrointestinal symptoms, fatigue, and thrombocytopenia. These adverse reactions are dose-related and reversible upon cessation of therapy. Preclinical studies have supported the therapeutic potential of vorinostat by demonstrating in vitro and in vivo anti-tumor activities against CTCL, including selective induction of apoptosis in malignant T cells, inhibition of angiogenesis, suppression of signal transducer and activator of transcription proteins, and up-regulation of pro-apoptotic proteins. Transmission electron microscope and scanning electron microscope methods were used to observe bacterial adhesion and the bacterial cell membrane. Fibroblast cell viability was determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay.Results: The best FSCP/COS mass ratio for electrospinning was 2:1, and the nanofibers had small dimensions ranging from 50 to 100 nm. The NFM showed good antibacterial activities against Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli. The antibacterial activity against S. aureus was higher than against E. coli. The pili and adhesive fimbriae of E. coli promoted bacterial adhesion to the NFM surfaces, and S. aureus biofilms aided S. aureus adhesion on the surface of NFMs. Damage to the bacterial cell membrane indicates that the NFMs could lead to the release of intracellular materials, particularly with S. aureus. In addition, FSCP/COS NFM rapidly increased the permeability of the outer membranes of E. coli. The electrospun NFM with FSCP and COS had good biocompatibility in vitro and supported proliferation of human skin fibroblasts.Conclusion: FSCP are superior to mammalian collagen, and have feasibility and potency for wound dressings. Abstract Background Mammalian spermatozoa become fully motile and fertile during transit through the luminal fluid of the epididymis. At least 200 proteins are present in the epididymal lumen, but the potential roles of these luminal proteins in male fertility are unknown. Investigation of the function of these proteins will elucidate the mechanism of sperm maturation, and also provide new drug targets for male contraception. We cloned RNase9 from a human epididymis cDNA library for characterization and analysis of its functions. Findings It was predicted that human RNase9 gene was located on chromosome 14q11.2 and encoded a 205 amino acids protein with a signal peptide of 26 amino acids at the N-terminus. The protein had eight conserved cysteine residues characteristic of the RNase A family members and several potential post-translational modification sites. At the transcriptional level, RNase9 was expressed in a wide variety of tissues, and the expression was higher in men than in boys. RNase9 was localized to the post-equatorial region of the sperms' head. Immunofluorescence staining showed that RNase9 protein was present mostly in the epithelium of the epididymal tubule. Recombinant RNase9 had no ribonuclease activity. In addition, RNase9 had no detectable effect on sperm motility and fertilization as demonstrated by blocking spermatozoa with anti-RNase9 polyclonal serum. Conclusion RNase9 is expressed in a wide variety of tissues. It is located on the post-equatorial region of the sperm head and the epithelium of epididymal tubule. Although RNase9 belongs to the RNase A family, it has no ribonuclease activity.

Abstract Almost one in ten Australians has received acupuncture treatment by acupuncturists and/or medical doctors in private clinics. The majority of Australian health insurance funds offer rebates for acupuncture. Statutory regulations for acupuncture have been implemented in the State of Victoria, Australia. Six acupuncture degree courses have been approved by the Chinese Medicine Registration Board of Victoria and/or accredited by the Australian Acupuncture and Chinese Medicine Association. Furthermore, a number of clinical trials of acupuncture on allergic rhinitis, pain and women's health were carried out in Australia. Recent developments of acupuncture in Australia indicate that through adequate and appropriate evaluation, acupuncture begins to integrate into mainstream health care in Australia.

Objective: Black wool can effectively prevent sheep from DNA damage as well as fungal infection, and can improve reproductive performance. In order to explore the candidate genes related to black wool formation in Qira sheep., Methods: We selected 123 adult healthy ewes with different coat colors in Qira sheep groups (black (B), brown (Y) and grey (G)) and extracted DNA from their venous blood to obtain Illumina Ovine SNP 50K chip data. Subsequently, our PCA, NJ-tree, and Admixture population structure analyses of the 3 wool color Qira sheep populations showed that the 3 middle wool color populations exhibited the same genetic traits. Fst, xp-EHH, iHS, and π were detected for selection signals, and the 5% SNPs loci positively selected from the analyses were annotated based on SheepOar_v4.0. The region of exon 1 of the TYRP1 gene was further screened, amplified and sequenced through the DNA of the Qira sheep and associated with goodness-of-fit using the chi-square test., Results: The results showed that 71 SNPs associated with black wool traits, among which TYRP1, PARD3 and CDH2 genes were strongly associated with black wool production. Three mutations were detected in the exon 1 region of the TYRP1 gene that were significantly associated with coat color variation inQira sheep (2:81,183,168, 2:81,183,281 and 2:81,183,284)., Conclusion: In this paper, Qira sheep could not differentiate the genetic structure of this population by wool color, and obtained 71 SNPs related to black wool. Detection of mutation sites on the TYRP1 gene affecting hair color change provides a basis for black sheep line selection as well as breed conservation., Competing Interests: Declarations. Defining the active sites and further optimizing their activity are of great significance for enhancing the hydrogen evolution reaction (HER) performances, especially for inexpensive Ni-based catalysts doped with metals and nonmetal elements. This work reports the role of the incorporated molybdenum and sulfur in enhancing the HER activity of nickel. The prepared molybdenum and sulfur coincorporated Ni (NMS) electrocatalysts exhibit excellent HER performance, with an overpotential and Tafel slope of 77.0 mV (10 mA·cm -2 ) and 64.4 mV·dec -1 , respectively, because of the large electrochemically active surface area, quick reaction kinetics, and charge-transfer capability. The theoretical calculation results reveal that the incorporated Mo atoms play the role of reaction sites, and the introduced S atoms can further enhance the activity of Mo atom. The high HER activity of the Mo atom can be attributed to the regulated d-band center by optimizing the composition of NMS, which tunes the interaction between Mo and intermediate species. The elucidated mechanism can make it possible to design Ni-based electrocatalysts doped with metals and nonmetals for efficient hydrogen evolution.

The majority of genetic Primary progressive aphasia (PPA) patients harbor mutations in the granulin ( GRN ) gene. The present case showed impaired performances in single-word retrieval in spontaneous speech and naming, and repetition. Head MRI revealed marked lateral atrophy in the left parietal cortex. A diagnosis of logopenic variant PPA (lvPPA) was established. Genetic analysis showed a heterozygous 10-bp frameshift deletion in exon 4 of the GRN gene (NM_002087.4), leading to transformation of cysteine into alanine at amino acid 92 and creation of a premature stop codon at position 161. This patient represented a rare case of GRN -associated lvPPA. A new mutation site was detected in exon 4 of GRN gene.

Eleven previously undescribed coumarin derivatives, bonenncarpas A-K (1-11), along with three known analogues (12-14), were isolated from the whole plant of Boenninghausenia sessilicarpa. The structures of these compounds were elucidated through comprehensive spectroscopic and spectrometric analyses. Their absolute configurations were determined by comparing experimental and calculated electronic circular dichroism data, as well as by X-ray crystallography. Notably, compound 1 features an open-ring coumarin structure with a five-membered lactone ring, while compounds 9-11 are characterized by a dihydrofuran ring at C-3 and C-4. All compounds were evaluated for their inhibitory effects on lipopolysaccharide-induced nitric oxide production in RAW 264.7 macrophage cells. Objective: This study aimed to develop a predictive model for the risk of no usable blastocyst formation in patients with normal ovarian reserve undergoing IVF., Methods: The model was derived from 7,901 patients who underwent their first oocyte retrieval and subsequent blastocyst culture, of which 446 cases have no usable blastocysts formed. Univariate regression analyses, least absolute shrinkage and selection operator regression analysis were used to identify the association of patient and cycle characteristics with the presence of no available blastocyst and to create a nomogram. The performance of the nomogram was assessed using the receiver operating characteristic (ROC) curve and calibration curve, the net benefit threshold of prediction was determined using decision curve analysis (DCA)., Results: Multivariate analysis identified three independent predictors: the number of day 3 (D3) embryos, the number of high-quality D3 embryos, and the number of embryos used for blastocyst culture. A nomogram model was developed and internally validated using bootstrapping, demonstrating good discriminative ability with an area under the receiver operating characteristic curve (AUC) of 0.879(95%CI: 0.861-0.890)., Conclusions: The cycle-based nomogram can anticipate the probability of no available blastocyst formation in IVF/ICSI treatment. The catalytic conversion of CO 2 into valuable chemicals using metalized covalent organic frameworks (COFs) as catalysts is a promising method for reducing atmospheric CO 2 levels. Herein, a aldehyde-amine COF (TAPT-Tp) at room temperature and pressure and their metallized results is synthesized, Ni-TAPT-Tp and Ti-TAPT-Tp. The photocatalytic results indicate that the CO 2 to CO reduction rate is 6182.5 µmol g -1  h -1 for Ni-TAPT-Tp, but only 1615.4 µmol g -1  h -1 for Ti-TAPT-Tp. Density functional theory (DFT) simulations further demonstrate that for intermediates * CO 2 , * COOH, and * CO, the energy of Ni-TAPT-Tp is consistently lower than that of Ti-TAPT-Tp, indicating that Ni-TAPT-Tp exhibits superior photocatalytic performance for CO 2 RR. In China, the red swamp crayfish (Procambarus clarkii), a notorious invasive species, has become an important economic freshwater species. In order to compare the genetic diversity and population structure of crayfish from northern and southern China, we collected 60 crayfish individuals from 4 crayfish populations in northern China and 2 populations in southern China for sequencing using the 2b-RAD technique. Additionally, the whole genome sequence information obtained by 2b-RAD of 90 individuals from 2 populations in northern China and 7 populations in southern China were downloaded from NCBI. After quality control, a total of 25,371 SNPs were detected from approximately 54.22 billion raw reads. Based on these SNPs, high genetic diversity was observed in the 15 crayfish populations in China. The pairwise F ST values indicated that there was a large genetic differentiation of crayfish populations in northern and southern China. Despite common genetic backgrounds, due to geographical barriers, genetic divergence has been observed in northern and southern China crayfishes. The principal component analysis in combination with Admixture and Neighbor-Joining tree analysis showed that the crayfish fell into two clusters corresponding to geographical regions. The integrated analysis of whole genome and transcriptome data showed that two genes (CETN4 and CPEB2) might play important roles during crayfish resistance to a cold environment. This study reveals the genetic differentiation of crayfish populations in northern and southern China and provides clues to the genetic mechanism related to cold adaptation., Competing Interests: Declarations. Gestational diabetes mellitus (GDM), a common pregnancy disease, has long-term negative effects on offspring health. Epigenetic changes may have important contributions to that, but the underlying mechanisms are not well understood. Here, we report the influence of GDM on DNA methylation of offspring (GDF1) oocytes and the possible mechanisms. Our results show that GDM induces genomic hyper-methylation of offspring oocytes, and at least a part of the altered methylation is inherited by F2 oocytes, which may be a reason for the inheritance of metabolic disorders. We further find that GDM exposure increases the expression of Ezh2 in oocytes. Ezh2 regulates DNA methylation via DNMT1, and Ezh2 knockdown reduces the genomic methylation level of GDF1 oocytes. These results suggest that GDM may induce oocyte genomic hyper-methylation of offspring via enhancing the Ezh2 expression recruiting more DNMT1 into nucleus., Competing Interests: Competing interests: The authors declare no competing interests., (© 2024. For the efficient detection of Hg 2+ and ClO - , a double-analyte-responsive fluorescent probe PTB was successfully synthesized by combining N-butyl-3-formyl phenothiazine with hydrazine benzothiazole, and designing a specific reaction site for recognizing two analytes (Hg 2+ and ClO - ) in a compound. It was shown that probe PTB successfully formed a stable complex with Hg 2+ in the coordination ratio of 2:1 by using the strong sulfur affinity of Hg 2+ , which resulted in a remarkable "turn-off" effect, with a quenching efficiency of 92.5% and four reversible cycles of Hg 2+ fluorescence detection. For the fluorescence detection of Hg 2+ , the response time is fast (≤ 2 min) and the detection limit is low (7.8 nM), showing extremely high sensitivity, and the performance is obviously better than that of the reported fluorescent probes for detecting Hg 2+ . In particular, probe PTB has low toxicity and good biocompatibility, and has been successfully used for imaging of Hg 2+ in living cells. Herein, we develop a dual catalytic platform for the 1,2- or 1,4-alkylacylation reaction of 1,3-dienes with readily available alkyl halides and aldehydes by merging N -heterocyclic carbene catalysis with photoinduced palladium catalysis. A series of β,γ-unsaturated ketones are obtained in good to high yields. Mechanistic studies suggest that this reaction involves a radical process. The direct synthesis of flavanone from salicylaldehyde exemplified the potential capability of this dual catalytic platform.

Dithiocarbamate is a key structural sequence in pharmaceuticals and agrochemicals, and its synthesis is crucial in organic chemistry. Although significant progress has been made in related synthesis research, developing a practical and universal synthesis method remains fascinating. Herein, we report a new visible-light-induced decarboxylation coupling reaction between N-hydroxyphthalimide esters and tetraalkylthiuram disulfides, which uses Ir(ppy) 3 as a photocatalyst to promote the generation of corresponding decarboxylation thioacylation product-dithiocarbamates in high yields. This redox-neutral protocol uses inexpensive and readily available starting material under mild reaction conditions, exhibiting broad substrate scope and wide functional group compatibility. As an invasive plant, Solanum elaeagnifolium has posed a serious threat to agriculture and natural ecosystems worldwide. In order to better manage and limit its spread, we established niche models by combining distribution information and climate data from the native and invasive ranges of S. elaeagnifolium to analyze its niche changes during its colonization. Additionally, we evaluated its global invasion risk. Our results showed that the distribution of S. elaeagnifolium is affected by temperature, precipitation, altitude, and human activities. Solanum elaeagnifolium exhibits different degrees of niche conservatism and niche shift in different invasion ranges. During the global invasion of S. elaeagnifolium, both the niche shift and conservatism were observed, however, niche shift was particularly significant due to the presence of unoccupied niches (niche unfilling). Solanum elaeagnifolium generally occupied a relatively stable niche. However, a notable expansion was observed primarily in Europe and China. In Australia and Africa, its niche largely remains a subset of its native niche. Compared to the niche observed in its native range, its realized niche in China and Europe has shifted toward lower temperature and higher precipitation levels. Conversely, in Africa, the niche has shifted toward lower precipitation levels, while in Australia, it has shifted toward higher temperature. Our model predicted that S. elaeagnifolium has high invasion potential in many countries and regions. The populations of S. elaeagnifolium in China and Africa have reached the adapted stage, while the populations in Australia and Europe are currently in the stabilization stage. In addition, our research suggests that the potential distribution of S. elaeagnifolium will expand further in the future as the climate warms. All in all, our study suggests that S. elaeagnifolium has high potential to invade globally. Objective: To construct and internally validate a nomogram that predicts the likelihood of postoperative delirium in a cohort of elderly individuals undergoing hip arthroplasty., Methods: Data for a total of 681 elderly patients underwent hip arthroplasty were retrospectively collected and divided into a model ( n  = 477) and a validation cohort ( n  = 204) according to the principle of 7:3 distribution temporally. The assessment of postoperative cognitive function was conducted through the utilization of The Confusion Assessment Method (CAM). The nomogram model for postoperative cognitive impairments was established by a combination of Lasso regression and logistic regression. The receiver operating characteristic (ROC) curve, calibration plot, and decision curve analysis (DCA) were used to evaluate the performance., Results: The nomogram utilized various predictors, including age, body mass index (BMI), education, preoperative Barthel Index, preoperative hemoglobin level, history of diabetes, and history of cerebrovascular disease, to forecast the likelihood of postoperative delirium in patients. The area under the ROC curves (AUC) for the nomogram, incorporating the aforementioned predictors, was 0.836 (95% CI: 0.797-0.875) for the training set and 0.817 (95% CI: 0.755-0.880) for the validation set. The calibration curves for both sets indicated a good agreement between the nomogram's predictions and the actual probabilities., Conclusion: The use of this novel nomogram can help clinicians predict the likelihood of delirium after hip arthroplasty in elderly patients and help prevent and manage it in advance.

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a crucial treatment option for children with M2 subtype acute myeloid leukemia (AML). Human herpesvirus 6 (HHV-6) encephalitis following transplantation is a rare postoperative complication, with a poor prognosis and a high fatality rate in allo-HSCT recipients. In this report, a juvenile patient with AMLwas successfully treated after developing visual impairment as a result of HHV-6B encephalitis during allo-HSCT therapy.
