Objective To evaluate the clinical efficacy and safety of hysteroscopy endo-operative system （HEOS） cold knife hysteroscopic micro-shear separation in the clinical treatment of patients with intrauterine adhesion. Methods 200 patients with intrauterine adhesion were enrolled and divided into the observation （n = 100） and control groups （n = 100） according to different treatment methods. In the control group， patients were treated with traditional hysteroscopic electrosurgical separation， and those in the observation group received HEOS cold knife hysteroscopic micro-shear separation. The surgical indexes， clinical efficacy and postoperative complications were statistically compared between two groups. Results In the observation group， the operation time was significantly shorter， and the amount of intraoperative bleeding and absorption of uterine distention fluid were significantly less than those in the control group （all P < 0.05）. There was a significant difference in clinical efficacy between two groups （P < 0.05）. In the observation group， the cure rate was 61%， higher than 45% in the control group （P < 0.05）. In the observation group， postoperative bleeding occurred in 1 case， 2 cases of infection and 1 case of pelvic adhesion. The incidence of postoperative complications was 4%. In the control group， postoperative bleeding occurred in 5 cases， 2 cases of infection， 3 cases of ectopic pregnancy and 5 cases of pelvic adhesion. The incidence of postoperative complications was 15%， which was significantly higher than 4% in the observation group （P

Objective Histological morphology, immunophenotype and molecular diagnosis of gliomas were reported and classified according to fifth edition of the 2021 World Health Organization (WHO) Classification of Tumors of the Central Nervous System (WHO CNS5). Methods Total 60 glioma patients with a clear diagnosis through genetic testing were collected in General Hospital of Southern Theatre Command of Chinese PLA from July 2019 to July 2021. All of them were diagnosed based on histopathology and molecular alterations by HE staining, immunohistochemical staining, and genetic screening and were classified according to the WHO CNS5. Results A total of 60 gliomas, according to WHO CNS5, were classified into one with pilocytic astrocytoma (1.67%), one with ganglioglioma (1.67%), 15 with astrocytomas, IDH⁃mutant (25%), 11 with oligodendrogliomas, IDH⁃mutant and 1p/19q co⁃deleted (18.33%), one with pleomorphic xanthoastrocytoma (1.67%), 21 with glioblastomas, IDH⁃wildtype (35%), 3 with diffuse midline glioma, H3 K27⁃altered (5%) and 7 with glioma, NEC (11.67%). The 7 patients with glioma, NEC were respectively diagnosed based on histopathology as anaplastic astrocytoma in one case, oligodendrogliomas in 2 cases, diffuse astrocytomas in 2 cases, ganglioglioma in one case, and anaplastic oligodendroglioma in one case, with only one case with methylguanine⁃DNA methyltransferase (MGMT) promoter methylation and one case with BRAF V600E mutation by genetic screening. The 21 patients with glioblastoma, IDH⁃wildtype were respectively diagnosed based on histopathology as glioblastoma in 15 cases, oligodendroglioma in one case, anaplastic oligodendroglioma in 2 cases, diffuse astrocytoma in 2 cases and anaplastic astrocytoma in one case, while TERT promoter mutation in 15 cases (71.43%), EGFR amplification in 7 cases (33.33%), MGMT promoter methylation in 5 patients (23.81%) and amplification of chromosome 7 in one case (4.76%). The 15 patients with astrocytoma, IDH⁃mutant, CNS WHO grade 2-4 were diagnosed based on histopathology as glioblastoma in 3 cases, oligodendroglioma in 2 cases, anaplastic oligodendroglioma in one case, diffuse astrocytoma in 8 cases and anaplastic astrocytoma in one case, with MGMT promoter methylation in 11 cases, homozygous deletion of cyclin⁃dependent kinase inhibitor 2A/B (CDKN2A/B) in 2 cases. Conclusions This article is the first pathological diagnosis report of single⁃center glioma since the publication of WHO CNS5, suggesting the WHO CNS5 could more accurately classify gliomas. However, the proportion of glioma, NEC diagnosed is still relatively high. Even well⁃classified gliomas have many differences in genetic signature. The molecular features of gliomas still require further deeply exploration.

Cardiac fibrosis is one of the major pathological characteristics of diabetic cardiomyopathy (DCM). MicroRNAs (miRNAs, miRs) have been identified as key regulators in the progression of cardiac fibrosis. This study aimed to investigate the role of miR-30a-5p in DCM and the underlying mechanism. The rat model of diabetes mellitus (DM) was established by streptozotocin injection, and the rat primary cardiac fibroblasts (CFs) were isolated from cardiac tissue and then treated with high glucose (HG). MTT assay was performed to assess the viability of CFs. Dual-luciferase reporter gene assay was conducted to verify the interaction between miR-30a-5p and Smad2. The expression of miR- 30a-5p was downregulated in the myocardial tissues of DM rats and HG-stimulated CFs. Overexpression of miR-30a-5p reduced Smad2 levels and inhibited collagen formation in HG-stimulated CFs and DM rats, as well as decreased the proliferation of CFs induced by HG. Smad2 was a target of miR-30a-5p and its expression was inhibited by miR-30a-5p. Furthermore, the simultaneous overexpression of Smad2 and miR-30a-5p reversed the effect of miR-30a-5p overexpression alone in CFs. Our results indicated that miR-30a-5p reduced Smad2 expression and also induced a decrease in proliferation and collagen formation in DCM. La fibrose cardiaque est l'une des principales caracteristiques pathologiques de la cardiomyopathie diabetique (CMD). On a etabli que les microARN (miARN) sont des regulateurs cles dans l'evolution de la fibrose cardiaque. Cette etude portait sur le role du miR-30a-5p dans la CMD et le mode d'action sous-jacent. Nous avons realise le modele de diabete sucre (DS) chez le rat par l'injection de streptozotocine, et nous avons isole des fibroblastes cardiaques (FC) de rat a partir de tissu cardiaque, puis administre des quantites de glucose elevees (HG). Nous avons evalue la vitalite des FC a l'aide de l'epreuve MTT. Nous avons mene l'epreuve de gene rapporteur par la double luciferase afin de verifier la presence d'une interaction entre le miR30a-5p et Smad2. Les resultats ont montre que l'expression du miR-30a-5p etait regulee a la baisse dans le tissu myocardique de rats atteints de DS et dans des FC stimules par l'HG. La subexpression du miR-30a-5p entrainait une diminution des taux de Smad2 avec une inhibition de la formation de collagene dans les FC stimules par l'HG et chez les rats atteints de DS, de meme qu'une diminution de la proliferation des FC induite par l'HG. Smad2 constituait une cible du miR- 30a-5p et son expression etait inhibee par le miR-30a-5p. En outre, la surexpression simultanee de Smad2 et du miR-30a-5p entrainait une inversion de l'effet de la subexpression du miR-30a-5p seul dans les FC. Nos resultats ont montre que le miR-30a-5p permettait de faire diminuer l'expression de Smad2, de meme que la proliferation des cellules et la formation de collagene dans la CMD.
