Abstract Active surveillance (AS) is the primary strategy for managing patients with low or favorable-intermediate risk prostate cancer (PCa). Identifying patients who may benefit from AS relies on unpleasant prostate biopsies, which entail the risk of bleeding and infection. In the current study, we aimed to develop a radiomics model based on prostate magnetic resonance images to identify AS candidates non-invasively. A total of 956 PCa patients with complete biopsy reports from six hospitals were included in the current multicenter retrospective study. The National Comprehensive Cancer Network (NCCN) guidelines were used as reference standards to determine the AS candidacy. To discriminate between AS and non-AS candidates, five radiomics models (i.e., eXtreme Gradient Boosting (XGBoost) AS classifier (XGB-AS), logistic regression (LR) AS classifier, random forest (RF) AS classifier, adaptive boosting (AdaBoost) AS classifier, and decision tree (DT) AS classifier) were developed and externally validated using a three-fold cross-center validation based on five classifiers: XGBoost, LR, RF, AdaBoost, and DT. Area under the receiver operating characteristic curve (AUC), accuracy (ACC), sensitivity (SEN), and specificity (SPE) were calculated to evaluate the performance of these models. XGB-AS exhibited an average of AUC of 0.803, ACC of 0.693, SEN of 0.668, and SPE of 0.841, showing a better comprehensive performance than those of the other included radiomic models. Additionally, the XGB-AS model also presented a promising performance for identifying AS candidates from the intermediate-risk cases and the ambiguous cases with diagnostic discordance between the NCCN guidelines and the Prostate Imaging-Reporting and Data System assessment. These results suggest that the XGB-AS model has the potential to help identify patients who are suitable for AS and allow non-invasive monitoring of patients on AS, thereby reducing the number of annual biopsies and the associated risks of bleeding and infection.

Bacteria exhibit a rich repertoire of RNA molecules that intricately regulate gene expression at multiple hierarchical levels, including small RNAs (sRNAs), riboswitches, and antisense RNAs. Notably, the majority of these regulatory RNAs lack or have limited protein-coding capacity but play pivotal roles in orchestrating gene expression by modulating transcription, post-transcription or translation processes. Leveraging and redesigning these regulatory RNA elements have emerged as pivotal strategies in the domains of metabolic engineering and synthetic biology. While previous investigations predominantly focused on delineating the roles of regulatory RNA in Gram-negative bacterial models such as Escherichia coli and Salmonella enterica, this review aims to summarize the mechanisms and functionalities of endogenous regulatory RNAs inherent to typical Gram-positive bacteria, notably Bacillus subtilis. Furthermore, we explore the engineering and practical applications of these regulatory RNA elements in the arena of synthetic biology, employing B. subtilis as a foundational chassis.

Defect detection is critical to industrial quality control in leather production engineering. The various sizes and locations of defects in leather, as well as significant differences within the same class and indistinctive variations between different classes of defects, contribute to the complexity of the problem. To address this challenge, we propose a Multi-Layer Residual Convolutional Attention (MLRCA) approach. MLRCA enhances its ability to capture both intra-class and inter-class differences by enhancing the semantic feature representation in the backbone network. To improve multiscale fusion effects, we also incorporate the MLRCA module into the feature pyramid network (FPN) and propose a new multi-layer residual convolution attention feature pyramid network (ML-FPN). This approach enables more accurate identification of leather defects at a more detailed level by selectively capturing contextual information from different domains. We then implement the Side-Aware Boundary Localization (SABL) detection head, which accurately locates defects and helps the network distinguish between similar defect categories for more precise positioning. To validate the effectiveness of our approach, we conducted ablation experiments on the created leather dataset. Comparative experiments demonstrate the excellent capability of our model to detect minor defects. The model achieved 83.4, 89.7, and 85.6 for the AP, AP50, and AP75 evaluation metrics. In addition, the model achieves 71.3, 89.9, and 88.9 for APS, APM, and APL. Our approach has been confirmed feasible through experimentation and provides new insights for automated leather defect detection methods.

The probiotic bacterium Escherichia coli Nissle 1917 (EcN) holds significant promise for use in clinical and biological industries. However, the reliance on antibiotics to maintain plasmid-borne genes has overshadowed its benefits. In this study, we addressed this issue by engineering the endogenous cryptic plasmids pMUT1 and pMUT2. The non-essential elements were removed to create more stable derivatives pMUT1NR△ and pMUT2HBC△. Synthetic promoters by integrating binding motifs on sigma factors were further constructed and applied for expression of Bacteroides thetaiotaomicron heparinase III and the biosynthesis of ectoine. Compared to traditional antibiotic-dependent expression systems, our newly constructed antibiotic-free expression systems offer considerable advantages for clinical and synthetic biology applications.

Abstract Background Although studies have indicated that extreme temperature is strongly associated with respiratory diseases, there is a dearth of studies focused on children, especially in China. We aimed to explore the association between extreme temperature and children’s outpatient visits for respiratory diseases and seasonal modification effects in Harbin, China. Methods A distributed lag nonlinear model (DLNM) was used to explore the effect of extreme temperature on daily outpatient visits for respiratory diseases among children, as well as lag effects and seasonal modification effects. Results Extremely low temperatures were defined as the 1st percentile and 2.5th percentile of temperature. Extremely high temperatures were defined as the 97.5th percentile and 99th percentile of temperature. At extremely high temperatures, both 26 °C (97.5th) and 27 °C (99th) showed adverse effects at lag 0–6 days, with relative risks (RRs) of 1.34 [95% confidence interval (CI): 1.21–1.48] and 1.38 (95% CI: 1.24–1.53), respectively. However, at extremely low temperatures, both − 26 °C (1st) and − 23 °C (2.5th) showed protective effects on children’s outpatient visits for respiratory diseases at lag 0–10 days, with RRs of 0.86 (95% CI: 0.76–0.97) and 0.85 (95% CI: 0.75–0.95), respectively. We also found seasonal modification effects, with the association being stronger in the warm season than in the cold season at extremely high temperatures. Conclusions Our study indicated that extremely hot temperatures increase the risk of children’s outpatient visits for respiratory diseases. Efforts to reduce the exposure of children to extremely high temperatures could potentially alleviate the burden of pediatric respiratory diseases, especially during the warm season.

BackgroundThe main functional gastrointestinal disorders (FGIDs) include functional dyspepsia (FD) and irritable bowel syndrome (IBS), which often present overlapping symptoms with gastroesophageal reflux disease (GERD), posing a challenge for clinical diagnosis and treatment. The gut microbiota is closely associated with FGIDs and GERD, although the causal relationship has not been fully elucidated. Therefore, we aimed to investigate the potential causal relationship using bidirectional two-sample Mendelian randomization (MR) analysis.Materials and methodsThe genetic data of the 211 gut microbiota were obtained from the MiBioGen consortium (N = 14,306, from phylum to genus level) and species level of gut microbiota were acquired from the Dutch Microbiome Project (N = 7,738). For FD and IBS, we utilized the FinnGen consortium, whereas, for GERD data analysis, we obtained the IEU OpenGWAS project. The inverse-variance weighted (IVW) method was used as the primary method to calculate causal effect values. Sensitivity analyses were also performed to confirm the robustness of the primary findings of the MR analyses. Moreover, a reverse MR analysis was conducted to assess the likelihood of reverse causality.ResultsCombining the results of the preliminary and sensitivity analyses, we identified that 8 gut microbial taxa were associated with FD. Genus Lachnospiraceae NK4A136 group (p = 3.63 × 10−3) and genus Terrisporobacter (p = 1.13 × 10−3) were strongly associated with FD. At the same time, we found that 8 gut microbial taxa were associated with IBS. Family Prevotellaceae (p = 2.44 × 10−3) and species Clostridium leptum (p = 7.68 × 10−3) display a robust correlation with IBS. In addition, 5 gut microbial taxa were associated with GERD using the IVW approach. In the reverse MR analysis, 2 gut microbial taxa were found to be associated with FD, 5 gut microbial taxa were found to be associated with IBS, and 21 gut microbial taxa were found to be associated with GERD.ConclusionThe study reveals the potential causal effects of specific microbial taxa on FD, IBS, and GERD and may offer novel insights into the diagnosis and treatment of these conditions.

This paper presented the development of the thermo-mechanical coupling model and simulation analysis of a composite refractory layer ladle. The thermomechanical modeling of the ladle was carried out using the Finite Element Method (FEM), taking into account the joints between the slag line bricks as well as the contact between the composite refractory layers. The temperature and thermal stress characteristics of the overall ladle structure under different transit conditions were investigated using the finite element analysis software ANSYS. We also performed a comparative analysis on the improvement effect of thermal stresses by expansion joints of different sizes. The simulation results were validated with previous studies. The results showed that the installation of expansion joints within a suitable range did not significantly change the thermal expansion characteristics of the refractory lining, but it also reduced the thermal stress level of the lining to a large extent. A larger temperature gradient produced higher thermal stresses, and in the direction of the ladle height, the more downward the position of its maximum compressive stress was greater. Under different working conditions, compared with the axial and hoop stress, the radial stress was small in magnitude. Axial stress exhibited a greater degree of sensitivity to pressure variations than hoop stress. The developed thermodynamic model considering the full contact between the bricks and the refractory layer gave us a more complete view of the physical field laws inside the ladle.

Abstract Sulfonation as one of the most important modification reactions in nature is essential for many biological macromolecules to function. Development of green sulfonate group donor regeneration systems to efficiently sulfonate compounds of interest is always attractive. Here, we design and engineer two different sulfonate group donor regeneration systems to boost the biosynthesis of sulfated compounds. First, we assemble three modules to construct a 3'-phosphoadenosine-5'-phosphosulfate (PAPS) regeneration system and demonstrate its applicability for living cells. After discovering adenosine 5’-phosphosulfate (APS) as another active sulfonate group donor, we engineer a more simplified APS regeneration system that couples specific sulfotransferase. Next, we develop a rapid indicating system for characterizing the activity of APS-mediated sulfotransferase to rapidly screen sulfotransferase variants with increased activity towards APS. Eventually, the active sulfonate group equivalent values of the APS regeneration systems towards trehalose and p-coumaric acid reach 3.26 and 4.03, respectively. The present PAPS and APS regeneration systems are environmentally friendly and applicable for scaling up the biomanufacturing of sulfated products.

In high-power photovoltaic systems, the inverter with an LCL filter is widely used to reduce the value of output inductance at which a lower switching frequency is required. However, the effect on the stability of the system caused by an LCL filter due to its resonance characteristic cannot be ignored. This paper studies the stability of a single-phase voltage source full-bridge inverter with an LCL filter through the bifurcation theory as it is a nonlinear system. The simulation results show that low-frequency oscillation appears when the proportional coefficient of the system controller increases or the damping resistance decreases to a certain extent. The average model is derived to analyze the low-frequency oscillation; the theoretical analysis demonstrates that low-frequency oscillation is essentially a period in which doubling bifurcation occurs, which indicates the intrinsic mechanism of the instability of the full-bridge inverter with an LCL filter. Additionally, the limitation of the existing damping resistor design standards, which only considers the main circuit parameters but ignores the influence of the controller on system stability, is identified. To solve this problem, the analytical expression of the system stability boundary is provided, which can not only provide convenience for engineering design to protect the system from low-frequency oscillation but also expand the selection range of damping resistance in practice. The experiments are performed to verify the results of the simulation and theoretical analysis, demonstrating that the analysis method can facilitate the design of the inverter with an LCL filter.

High-speed interior permanent magnet (IPM) motors require highly reliable rotors. Some measures must be adopted to improve rotor safety, but its electromagnetic performance is seriously affected. It is a challenge to achieve excellent electromagnetic characteristics while satisfying mechanical strength. This paper presents an electromagnetic optimization design of high-speed IPM motors considering rotor stress. Firstly, the permanent magnet (PM) is segmented by adding stiffeners to improve stress distribution. The effects of the bridge and stiffener thickness on the rotor stress and electromagnetic performance are analyzed. Secondly, an electromagnetic optimization model is built based on a three-segment PM rotor structure, aiming for maximum efficiency and minimum rotor core losses. Then, the initial design and optimized scheme are compared, the results show that the efficiency, safety and temperature performance of the motor are improved. Finally, a 140 kW, 18,000 rpm prototype is manufactured and tested. The above analysis provides a valuable reference for the design and widespread application of high-speed IPM motors.

Unmanned surface vehicles (USVs) are crucial in ensuring maritime safety and observation, attracting widespread attention and research. However, a single USV exhibits limited performance and cannot effectively observe complex marine environments. In contrast, clusters of USVs can collaborate to execute complex maritime tasks, thereby enhancing the overall operational efficiency. USVs typically form heterogeneous clusters by combining vehicles with varying maneuverabilities and communication network capabilities. This has sparked an increased interest in cooperative communication research within heterogeneous USV clusters. The heterogeneous USVs discussed in this paper share the same dynamic model; however, they differ in dynamic parameters and communication capabilities. First, this study establishes a three-degree-of-freedom motion mathematical model for an underdriven USV considering environmental interference. It estimates the dynamic parameters of four USVs and evaluates their communication abilities, laying the foundation for researching the cooperative control of heterogeneous USV clusters and their application in Ocean Observation. Next, the communication capability of the USVs is assessed by studying the communication mode and signal transmission loss within the USV clusters. This study investigates the problem of cooperative communication in USV cluster formation, starting with the communication delay of USV clusters under a directed switching topology. Finally, a coherent formation controller is designed under a switching communication topology to address the dynamic transformation of communication topologies within heterogeneous USV clusters. This verifies that heterogeneous USV clusters can seamlessly form and maintain formation shapes during communication topology transformations through formation simulation experiments involving four heterogeneous USVs was 22% higher than that of dispersed control topology structures. This study provides a solid foundation for future investigations into the cooperative control of heterogeneous USV clusters and their applications in marine observations.

As a fundamental tool in synthetic biology, promoters are pivotal in regulating gene expression, enabling precise genetic control and spurring innovation across diverse biotechnological applications. However, most advances in engineered genetic systems rely on host-specific regulation of the genetic portion. With the burgeoning diversity of synthetic biology chassis cells, there emerges a pressing necessity to broaden the universal promoter toolkit spectrum, ensuring adaptability across various microbial chassis cells for enhanced applicability and customization in the evolving landscape of synthetic biology. In this study, we analyzed and validated the primary structures of natural endogenous promoters from Escherichia coli, Bacillus subtilis, Corynebacterium glutamicum, Saccharomyces cerevisiae, and Pichia pastoris, and through strategic integration and rational modification of promoter motifs, we developed a series of cross-species promoters (Psh) with transcriptional activity in five strains (prokaryotic and eukaryotic). This series of cross species promoters can significantly expand the synthetic biology promoter toolkit while providing a foundation and inspiration for standardized development of universal components The combinatorial use of key elements from prokaryotic and eukaryotic promoters presented in this study represents a novel strategy that may offer new insights and methods for future advancements in promoter engineering.

Abstract Background Coccidiosis, a prominent intestinal protozoan disease, carries significant economic implications for the poultry industry. The aim of this study was to evaluate the effects of Fengqiang Shengtai (BLES), a probiotics product, and coccidiosis vaccine in modulating the intestinal microbiome and providing insight into mitigating the occurrence and management of avian coccidiosis. Methods Broilers included in the study were divided into four pre-treatment groups: the Pre-Con group (commercial diet), Pre-BLES group (BLES supplement), Pre-Vac group (coccidiosis vaccination) and Pre-Vac-BLES group (combined vaccination and BLES). Body weight gain, feed consumption and feed conversion ratio were monitored from age 25 to 55 days. Cecum contents were collected at 8 and 15 days of age for comparative analysis of intestinal microbiomes. In the Pre-BLES and Pre-Vac-BLES groups, probiotics were administered at a dose of 0.01 g per chicken between ages 3 to 6 days and 10–13 days. At 3 days of age, chickens in the Pre-Vac and Pre-Vac-BLES groups were vaccinated with 1700 sporulated oocysts of the live coccidiosis vaccine per chicken. At the age of 25 days, Eimeria spp. challenge experiments were performed based on the aforementioned immunization strategy, and the oocysts per gram (OPG) in the feces, intestinal lesion score and intestinal pathological characteristics were evaluated. Specifically, 30 chickens were randomly selected from each group and orally administered 34,000 sporulated oocysts of Eimeria spp. per chicken, re-defined as Eimeria group, BLES-Eimeria group, Vac-Eimeria group and Vac-BLES-Eimeria group, respectively. Additionally, 30 chickens were randomly selected from the Pre-Con group and included as negative control without Eimeria spp. challenge. Intestinal microbiota was sequenced and analyzed when the broilers were 32 days old. Results A significant improvement was observed in body weight gain of the broilers in the Pre-BLES and Pre-Vac-BLES group at 45 days of age. Analysis of the intestinal microbiota revealed a positive correlation between the experimental groups receiving BLES and coccidiosis vaccines at 8 and 15 days of age with the Enterococcus genus and Lachnospiraceae NK4A136 group, respectively. In addition to the reduced lesion score and OPG values, the combination of coccidiosis vaccine and BLES also reduced the intestinal epithelial abscission induced by coccidiosis vaccines. The results of intestinal microbial function prediction demonstrated that N-glycan biosynthesis and ferroptosis were the prominent signal pathways in the Vac-BLES-Eimeria group. Conclusions Taken together, the results of the present study suggest that supplementation of BLES with coccidiosis vaccine represents a promising strategy for improving growth performance, alleviating clinical manifestations and inducing favorable alterations to the intestinal microbiota in broiler chickens affected by coccidiosis.

UAV swarm passive positioning technology only requires the reception of electromagnetic signals to achieve the positioning and tracking of radiation sources. It avoids the active positioning strategy that requires active emission of signals and has the advantages of good concealment, long acting distance, and strong anti-interference ability, which has received more and more attention. In this paper, we propose a new UAV swarm formation flight method based on pure azimuth passive positioning. Specifically, we propose a two-circle positioning model, which describes the positional deviation of the receiving UAV using trigonometric functions relative to the target in polar coordinates. Furthermore, we design a two-step adjustment strategy that enables the receiving UAV to reach the target position efficiently. Based on the above design, we constructed an optimized UAV swarm formation scheme. In experiments with UAV numbers of 8 and 20, compared to the representative comparison strategy, the proposed UAV formation scheme reduces the total length of the UAV formation by 34.76% and 55.34%, respectively. It demonstrates the effectiveness of the proposed method in the application of assigning target positions to UAV swarms.

Ectoine is a natural macromolecule protector and synthesized by some extremophiles. It provides protections against radiation-mediated oxidative damages and is widely used as a bioactive ingredient in pharmaceutics and cosmetics. To meet its growing commercial demands, we engineered Escherichia coli strains for the high-yield production of ectoine. The ectABC gene cluster from the native ectoine producer Halomonas elongata was introduced into different Escherichia coli (E. Coil) strains via plasmids and 0.8 g L-1 of ectoine was produced in flask cultures by engineered E. coli BL21 (DE3). Subsequently, we designed the ribosome-binding sites of the gene cluster to fine-tune the expressions of genes ectA, ectB, and ectC, which increased the ectoine yield to 1.6 g L-1. After further combinatorial overexpression of Corynebacterium glutamicum aspartate kinase mutant (G1A, C932T) and the H. elongate aspartate-semialdehyde dehydrogenase to increase the supply of the precursor, the titer of ectoine reached to 5.5 g L-1 in flask cultures. Finally, the engineered strain produced 60.7 g L-1 ectoine in fed-batch cultures with a conversion rate of 0.25 g/g glucose.

Abstract Purpose To demonstrate the practicability of a portable instrument in assessing tear film breakup time (TFBUT): a lens attachment for smartphones (LAS). Methods By applying LAS in combination with the iPhone 12 pro, and a recordable slit-lamp microscope, we obtained TFBUT videos from 58 volunteers. The comparison between the conventional slit-lamp microscope and LAS by an experienced ophthalmologist. Moreover, we also invited an ophthalmic postgraduate student and an undergraduate student with no clinical experience to assess TFBUT in those videos. The inter-observer reliability was assessed using intraclass correlation coefficients (ICC). Results The TFBUT of 116 eyes in total was recorded. Reliability indexes were adequate. Phthalides are a class of unique compounds such as ligustilide, butylphthalide and butyldenephthalide, which have shown to possess multiple bioactivities in new drug research and development. Phthalides are naturally distributed in different plants that have been utilized as herbal treatments for various ailments with a long history in Asia, Europe and North America. Their extensive biological activity has led to a dramatic increase in the study of phthalide compounds in recent years. This review summarizes the latest research progress of plant-derived phthalides, and a total of 133 phthalide compounds are described based on the characteristics of chemical structures. Pharmacological properties of plant-derived phthalides are associated with hemorheological improvement, vascular function modulation and central nervous system protection. Potential treatments for a variety of diseases mainly including cardio-cerebrovascular disorders and neurological complications such as Alzheimer's disease are also concluded. In addition, key metabolic pathways have been clearly elucidated. Further investigations on the molecular mechanisms involved in biological activity are recommended for offering new insights into profound comprehension of phthalide applications.

Leech hyaluronidase (LHyal) is a hyperactive hyaluronic acid (HA) hydrolase that belongs to the hyaluronoglucuronidase family. Traditionally, LHyal is extracted from the heads of leeches, but the recent development of the Pichia pastoris recombinant LHyal expression method permitted the industrial production of size-specific HA oligosaccharides. However, at present LHyal expressed by recombinant yeast strains requires laborious protein purification steps. Moreover, the enzyme is deactivated and removed after single use. To solve this problem, we developed a recyclable LHyal biocatalyst using a yeast surface display (YSD) system. After screening and characterization, we found that the cell wall protein Sed1p displayed stronger anchoring to the P. pastoris cell wall than other cell wall proteins. By optimizing the type and length of the linkers between LHyal and Sed1p, we increased the activity of enzymes displayed on the P. pastoris cell wall by 50.34% in flask cultures. LHyal-(GGGS)6-Sed1p activity further increased to 3.58 × 105 U mL−1 in fed-batch cultivation in a 5 L bioreactor. Enzymatic property analysis results revealed that the displayed LHyal-(GGGS)6-Sed1p generated the same oligosaccharides but exhibited higher thermal stability than free LHyal enzyme. Moreover, displayed LHyal-(GGGS)6-Sed1p could be recovered easily from HA hydrolysis solutions via low-speed centrifugation and could be reused at least 5 times. YSD of LHyal not only increased the utilization efficiency of the enzyme but also simplified the purification process for HA oligosaccharides. Thus, this study provides an alternative approach for the industrial preparation of LHyal and HA oligosaccharides.

Gene regulation by trans-acting small RNAs (sRNAs) has considerable advantages over other gene regulation strategies. However, synthetic sRNAs mainly take natural sRNAs (MicC or SgrS) as backbones and comprise three functional elements folding into two or more stem-loop structures: an mRNA base pairing region, an Hfq-binding structure, and a rho-independent terminator. Due to limited numbers of natural sRNAs and complicated backbone structures, synthetic sRNAs suffer from low activity programmability and poor structural modularity. Moreover, natural sRNA backbone sequences may increase the possibility of unwanted recombination. Here, we present a bottom-up approach for creating structure defined single-stem loop small non-coding RNAs (ssl-sRNAs), which contain a standardized scaffold of a 7 bp-stem-4 nt-loop-polyU-tail and a 24 nt basing pairing region covering the first eight codons. Particularly, ssl-sRNA requires no independent Hfq-binding structure, as the polyU tail fulfills the roles of binding Hfq. A thermodynamic-based scoring model and a web server sslRNAD (http://www.kangzlab.cn/) were developed for automated design of ssl-sRNAs with well-defined structures and programmable activities. ssl-sRNAs displayed weak polar effects when regulating polycistronic mRNAs. The ssl-sRNA designed by sslRNAD showed regulatory activities in both Escherichia coli and Bacillus subtilis. A streamlined workflow was developed for the construction of customized ssl-sRNA and ssl-sRNA libraries. As examples, the E. coli cell morphology was easily modified and new target genes of ergothioneine biosynthesis were quickly identified with ssl-sRNAs. ssl-sRNA and its designer sslRNAD enable researchers to rapidly design sRNAs for knocking down target genes.

Bacillus subtilis as the Gram-positive model bacterium has been widely used in synthetic biology and biotechnology while the regulatory RNA tools for B. subtilis are still not fully explored. Here, a bottom-up approach is proposed for designing artificial trans-acting sRNAs. By engineering the intrinsic sRNA SR6, a minimized core scaffold structure consisting of an 8 bp stem, a 4 nt loop, and a 9 nt polyU tail was generated and proven to be sufficient for constructing sRNAs with strong repression activity (83%). Moreover, we demonstrate this artificial sRNA system functions well in an hfq-independent manner and also achieves strong repression efficiency in Escherichia coli (above 80%). A structure-based sRNA design principle was further developed for the automatic generation of custom sRNAs with this core scaffold but various sequences, which facilitates the manipulation and avoids structure disruption when fusing any base-pairing sequence. By applying these auto-designed sRNAs, we rapidly modified the cell morphology and biofilm formation, and regulated metabolic flux toward acetoin biosynthesis. This sRNA system with cross-species regulatory activities not only enriched the gene regulation toolkit in synthetic biology for B. subtilis and E. coli but also enhanced our understanding of trans-acting sRNAs.

BackgroundImmunogenic cell death (ICD) plays a vital role in tumor progression and immune response. However, the integrative role of ICD-related genes and subtypes in the tumor microenvironment (TME) in prostate cancer (PCa) remains unknown.Materials and methodsThe sample data were obtained from The Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), and Memorial Sloan Kettering Cancer Center (MSKCC) prostate cancer-related databases. We first divided the subtypes based on ICD genes from 901 PCa patients and then identified the prognosis- related genes (PRGs) between different ICD subtypes. Subsequently, all the patients were randomly split into the training and test groups. We developed a risk signature in the training set by least absolute shrinkage and selection operator (LASSO)–Cox regression. Following this, we verified this prognostic signature in both the training test and external test sets. The relationships between the different subgroups and clinical pathological characteristics, immune infiltration characteristics, and mutation status of the TME were examined. Finally, the artificial neural network (ANN) and fundamental experiment study were constructed to verify the accuracy of the prognostic signature.ResultsWe identified two ICD clusters with immunological features and three gene clusters composed of PRGs. Additionally, we demonstrated that the risk signature can be used to evaluate tumor immune cell infiltration, prognostic status, and an immune checkpoint inhibitor. The low-risk group, which has a high overlap with group C of the gene cluster, is characterized by high ICD levels, immunocompetence, and favorable survival probability. Furthermore, the tumor progression genes selected by the ANN also exhibit potential associations with risk signature genes.ConclusionThis study identified individuals with high ICD levels in prostate cancer who may have more abundant immune infiltration and revealed the potential effects of risk signature on the TME, immune checkpoint inhibitor, and prognosis of PCa.

This paper proposed an eco-speed harmonization method at intersections. It is able to reduce carbon emissions by controlling partially connected and automated traffic and signal timing. It has the following features: (i) traffic emission reduction enhancement at various demand levels; (ii) traffic emission achievement while improving the mobility of entire traffic at intersections; (iii) enhanced traffic emission reduction with the help of a small portion of connected and automated vehicles; and (iv) potential implementations in the near feature. To validate the effectiveness, the proposed method is evaluated against a state-of-the-art strategy. Sensitivity analysis is conducted under various demand levels and market penetration rates (MPRs) of connected and automated vehicles (CAVs). The result shows that the proposed method outperforms and has the benefits of traffic emission reduction, throughput improvement, and stop frequency reduction. The proposed method demonstrates consistent performance across all demand levels and CAVs’ MPR. The proposed approach can achieve a reduction in emissions ranging from 4% to 61%, an average increase in throughput of around 14.91%, and a decrease in the stop frequency of at least 26%. This provides the foundation for future CAVs-based eco-approaching strategies.

Avian coccidiosis is the most serious parasitic disease in the poultry industry. Therefore, the aim of the current study was to explore the effects of B. subtilis and live coccidiosis vaccine alone or in combination on the production performance and anticoccidiosis, as well as the dynamic changes of intestinal microbial community. Nine hundred ninty Mahuang chickens were randomized into 4 preimmune groups including control group, coccidiosis vaccine immunization group; B. subtilis administration group and a group that was administrated a combination of live coccidiosis vaccine and B. subtilis group. Intestinal mucosal scraps collected from all these experimental groups at the age of 8 d and 15 d for microbial community 16S rRNA gene sequencing. At the age of 25 d, 30 broilers from each preimmune group were randomly assigned to a subgroup infected with Eimeria spp. and renamed as CI, V-CI BS-CI, and VBS-CI group. The production performance was monitored at the age of 25 d, 35 d, 45 d, and 55 d for the rest broilers from each pre-immune group. Otherwise, in the Eimeria spp. challenge stage, intestinal mucosal scraps collected for microbial community sequencing, while duodenum, jejunum, and cecum collected for pathological examination after sacrifice at the age of 32 d. In addition, the oocysts per gram of feces (OPG) and intestinal lesion score of broilers after Eimeria spp. challenge were also counted. Overall, the probiotics and coccidiosis vaccine resulted in the significantly improvement of the production performance. Otherwise, the intestinal lesion score and OPG after Eimeria spp. infection was significantly decreased in the VBS-CI group (P < 0.05). Moreover, these protective effects may also be closely related to genus such as Romboutsia, Blautia, and Butyricococcus, as well as microbiota functions like the quorum sensing pathway. According to these results, a combination of B. subtilis and coccidiosis vaccines can improve performance and provide additional protection against Eimeria spp. infection.

Food availability represents a major worldwide concern due to climate change and population growth. Low-temperature stress (LTS) severely restricts the growth of tomato seedlings. Exogenous 5-aminolevulinic acid (ALA) can alleviate the harm of abiotic stress including LTS; however, data on its protective mechanism on tomato seedling roots, the effects of organelle structure, and the regulation of metabolic pathways under LTS are lacking. In this study, we hope to fill the above gaps by exploring the effects of exogenous ALA on morphology, mitochondrial ultrastructure, reactive oxygen species (ROS) enrichment, physiological indicators, related gene expression, and metabolic pathway in tomato seedlings root under LTS. Results showed that ALA pretreatment could increase the activity of antioxidant enzymes and the content of antioxidant substances in tomato seedlings roots under LTS to scavenge the massively accumulated ROS, thereby protecting the mitochondrial structure of roots and promoting root development under LTS. Combined transcriptomic and metabolomic analysis showed that exogenous ALA pretreatment activated the glutathione metabolism and β-alanine metabolism of tomato seedling roots under LTS, further enhanced the scavenging ability of tomato seedling roots to ROS, and improved the low-temperature tolerance of tomato seedlings. The findings provide a new insight into the regulation of the low-temperature tolerance of tomato by exogenous ALA.

Chondroitin sulfate (CS) has a wide range of physiological functions and clinical applications. However, the biosynthesis of chondroitin oligosaccharides (o-CHs) and sulfate derivatives with specific length is always challenging. Herein, we report enzymatic strategies for producing homogeneous o-CHs and its sulfate derivatives from microbial sourced chondroitin. Chondroitin disaccharides, tetrasaccharides, hexasaccharides, octasaccharides, and decasaccharides with defined structure were produced by controllably depolymerizing microbial sourced chondroitin with an engineered chondroitinase ABC I. The highest conversion rates of the above corresponding o-CHs were 65.5%, 32.1%, 12.7%, 7.2%, and 16.3%, respectively. A new efficient enzymatic sulfation system that directly initiates from adenosine 5′-triphosphate (ATP) and sulfate was developed and improved the sulfation of chondroitin from 8.3% to 85.8% by optimizing the temperature, sulfate and ATP concentration. o-CHs decasaccharide, octasaccharide, hexasaccharide, tetrasaccharide and disaccharide were modified and the corresponding sulfate derivatives with one sulfate group were prepared. The enzymatic approaches constructed here for preparing o-CHs and its sulfate derivatives pave the way for the study of structure-activity relationship and applications.

Previous studies found that 5-aminolevulinic acid (ALA) and abscisic acid (ABA) can mitigate damage from adversity by enhancing photosynthesis. However, it is not clear whether they have positive effects on iron utilization and chlorophyll synthesis of tomato seedlings under low-temperature stress. To investigate the possible functional relationship between ABA and ALA and elucidate the possible mechanisms of action of ALA to alleviate low-temperature stress in tomato seedlings, this experiment analyzed the effects of ALA and ABA on chlorophyll synthesis in tomato seedling leaves sprayed with exogenous of ALA (25 mg·L−1) or ABA (100 µM) under low-temperature stress (8–18 °C/8–12 °C, day/night). The results show that exogenous ALA increased the Fv/Fm of tomato leaves by 5.31% and increased the accumulation of iron and chlorophyll by 101.15% and 15.18%, respectively, compared to the low-temperature treatment alone, and tomato resistance of low-temperature stress was enhanced. Meanwhile, exogenous application of ALA increased the ABA content by 39.43%, and subsequent application of exogenous ABA revealed that tomato seedlings showed similar effects to exogenous ALA under low-temperature stress, with increased accumulation of iron and chlorophyll in tomato seedlings, which eventually increased the maximum photochemical efficiency of PS II. Under low-temperature stress, application of exogenous ABA significantly reduced ALA content, but the expression of key enzyme genes (PPGD, HEMB1, HEME1, and HEMF1), precursors of chlorophyll synthesis by ALA, was significantly elevated, presumably because the increased activity of these enzymes after external application of ABA accelerated ALA consumption. In conclusion, ABA may crosstalk with ALA to improve the photochemical efficiency and low temperature resistance of tomatoes by regulating chlorophyll synthesis and iron accumulation.

Hyaluronidases are a group of glycosidases catalyzing the degradation of hyaluronic acid (HA). Because of the advantages of effectively hydrolyzing the HA-rich matrix and low immunogenicity, human hyaluronidase PH20 (hPH20) is widely used in the medical field. Here, we realized the active expression of recombinant hPH20 by Pichia pastoris under a methanol-induced promoter PAOX1. By optimizing the composition of the C-terminal domain and fusing protein tags, we constructed a fusion mutant AP2-△491C with the extracellular hyaluronidase activity of 258.1 U·L−1 in a 3-L bioreactor, the highest expression level of recombinant hPH20 produced by microbes. Furthermore, we found recombinant hPH20 hydrolyzed the β-1,4 glycosidic bonds sequentially from the reducing end of o-HAs, with HA6NA as the smallest substrate. The result will provide important theoretical guidance for the directed evolution of the enzyme to prepare multifunctional o-HAs with specific molecular weights.

At present, marine autonomous surface ships (MASSs) play a huge role in marine shipping, surveying and mapping, safeguarding the rights and interests of marine space, and other maritime tasks. The cooperative operation of a swarm of MASSs can extend the scope of operation of the MASSs, and thus allow them to carry out more complex tasks. Path tracking is an important problem for the control of a swarm of MASSs. In this paper, the control of underactuated MASSs is decoupled, to control the heading and speed, respectively. First of all, in the path tracking, the improved arc LOS guidance law is introduced, and the heading torque controller is designed, so that the MASS can track the reference path efficiently and accurately. Then the single-path guided path tracking without formation of the swarm of MASSs is studied, the reference path of the swarm center tracking is defined, and the heave thrust controller of the swarm of MASSs is designed based on consensus theory, so that the surge velocity of the MASS can tend towards consistentcy, and finally converge to the desired speed. Finally, the effectiveness of the proposed control strategy is verified by two groups of simulation experiments.

The problem of small and multi-object polished rice image segmentation has always been one of importance and difficulty in the field of image segmentation. In the appearance quality detection of polished rice, image segmentation is a crucial part, directly affecting the results of follow-up physicochemical indicators. To avoid leak detection and inaccuracy in image segmentation qualifying polished rice, this paper proposes a new image segmentation method (YO-LACTS), combining YOLOv5 with YOLACT. We tested the YOLOv5-based object detection network, to extract Regions of Interest (RoI) from the whole image of the polished rice, in order to reduce the image complexity and maximize the target feature difference. We refined the segmentation of the RoI image by establishing the instance segmentation network YOLACT, and we eventually procured the outcome by merging the RoI. Compared to other algorithms based on polished rice datasets, this constructed method was shown to present the image segmentation, enabling researchers to evaluate polished rice satisfactorily.

Escherichia coli is the most well-studied model prokaryote and has become an indispensable host for the biotechnological production of proteins and biochemicals. In particular, the probiotic status of one E. coli strain, E. coli Nissle 1917 (EcN) has helped it become a new favorite amongst synthetic biologists. To broaden its potential applications, here we assemble a comparative study on the genomes, transcriptomes, and metabolic properties of E. coli strains EcN, BL21(DE3), and MG1655. Comparative genomics data suggests that EcN possesses 1404 unique CDSs. In particular, EcN has additional iron transport systems which endow EcN with a higher tolerance to iron scarcity when compared to two other E. coli strains. EcN transcriptome data demonstrates that E. coli strains EcN, BL21(DE3), and MG1655 all have comparable activities of the central metabolic pathway, however only EcN inherits the arginine deiminase pathway. Additionally, we found that EcN displayed a lower expression of ribosomal proteins compared to BL21(DE3) and MG1655. This comparative study on E. coli strains EcN, BL21(DE3), and MG1655 aims to provide a reference for further engineering EcN as a biotechnological tool.

Here, the author increases hyaluronan production and diversifies the molecular weights through engineering the hyaluronan biosynthesis pathway and disruption of Corynebacterium glutamicum encapsulation caused by secreted hyaluronan.

Abstract Background Trypsin has many applications in food and pharmaceutical manufacturing. Although commercial trypsin is usually extracted from porcine pancreas, this source carries the risks of infectivity and immunogenicity. Microbial Streptomyces griseus trypsin (SGT) is a prime alternative because it possesses efficient hydrolysis activity without such risks. However, the remarkable hydrolysis efficiency of SGT causes autolysis, and five autolysis sites, R21, R32, K122, R153, and R201, were identified from its autolysate. Results The tbcf (K101A, R201V) mutant was screened by a directed selection approach for improved activity in flask culture (60.85 ± 3.42 U mL−1, increased 1.5-fold). From the molecular dynamics simulation, in the K101A/R201V mutant the distance between the catalytical residues D102 and H57 was shortened to 6.5 Å vs 7.0 Å in the wild type, which afforded the improved specific activity of 1527.96 ± 62.81 U mg−1. Furthermore, the production of trypsin was increased by 302.8% (689.47 ± 6.78 U mL−1) in a 3-L bioreactor, with co-overexpression of chaperones SSO2 and UBC1 in Pichia pastoris. Conclusions SGT protein could be a good source of trypsin for insulin production. As a result of the hydrolysates analysis and direct selection, the activity of the tbcf (K101A, R201V) mutant increased 1.5-fold. Furthermore, the production of trypsin was improved threefold by overexpressing chaperone protein in Pichia pastoris. Future studies should investigate the application of SGT to insulin and pharmaceutical manufacturing.

The electromagnetic wave propagation through plasma medium is one of the most important research fields in computational electromagnetics. A numerical formulation based on both the auxiliary differential equation (ADE) and the precise-integration time-domain (PITD) method for solving the plasma problems is proposed to break through the Courant-Friedrich-Levy (CFL) limit on the time-step size in a finite-difference time-domain (FDTD) simulation. In this new method, the current density J is introduced as the auxiliary variable to deal with the complex permittivity of the plasma which is dependent on the frequency, and the precise integration (PI) technique makes the selectable maximum time-step size become much larger and removes the impact of the time-step size to the numerical dispersion error. Numerical experimentations of the typical plasma problems verify and validate the reliability of the proposed formulation. Through the numerical results, it can be found that the maximum allowable time-step size of the new method is much larger than that of the CFL limit of the FDTD method, and the calculation error of the new method is nearly independent of the time-step size. As a consequence, the execution time is significantly reduced by using a larger time-step size.

Prokaryotic gene expression is largely regulated on transcriptional levels with the involvement of promoters, RNA polymerase and sigma factors. Developing new promoters to customize gene transcriptional regulation becomes increasingly demanded in synthetic biology and biotechnology. In this study, we designed synthetic promoters in the Gram-positive model bacterium Bacillus subtilis by interlocking the binding motifs of σA for house-keeping gene expression and that of two alternative sigma factors σH and σB which are involved in responding post-exponential growth and general stress, respectively. The developed promoters are recognized by multiple sigma factors and hence generate strong transcriptional strength when host cells grow under normal or stressed conditions. With green fluorescent protein as the reporter, a set of strong promoters were identified, in which the transcription activities of PHA-1, PHAB-4, PHAB-7 were 18.6, 4.1, 3.3 fold of that of the commonly used promoter P43, respectively. Moreover, some of the promoters such as PHA-1, PHAB-4, PHAB-7, PBA-2 displayed increased transcriptional activities in response to high salinity or low pH. The promoters developed in this study should enrich the biotechnological toolboxes of B. subtilis. Background:To identify the immune-related genes of bladder cancer (BLCA) based on immunological characteristics and explore their correlation with the prognosis. Methods:We downloaded the gene and clinical data of BLCA from the Cancer Genome Atlas (TCGA) as the training group, and obtained immune-related genes from the Immport database. We downloaded GSE31684 and GSE39281 from the Gene Expression Omnibus (GEO) as the external validation group. R (version 4.0.5) and Perl were used to analyze all data. Result:Univariate Cox regression analysis and Lasso regression analysis revealed that 9 prognosis-related immunity genes (PIMGs) of differentially expressed immune genes (DEIGs) were significantly associated with the survival of BLCA patients (p < 0.01), of which 5 genes, including NPR2, PDGFRA, VIM, RBP1, RBP1 and TNC, increased the risk of the prognosis, while the rest, including CD3D, GNLY, LCK, and ZAP70, decreased the risk of the prognosis. Then, we used these genes to establish a prognostic model. We drew receiver operator characteristic (ROC) curves in the training group, and estimated the area under the curve (AUC) of 1-, 3- and 5-year survival for this model, which were 0.688, 0.719, and 0.706, respectively. The accuracy of the prognostic model was verified by the calibration chart. Combining clinical factors, we established a nomogram. The ROC curve in the external validation group showed that the nomogram had a good predictive ability for the survival rate, with a high accuracy, and the AUC values of 1-, 3-, and 5-year survival were 0.744, 0.770, and 0.782, respectively. The calibration chart indicated that the nomogram performed similarly with the ideal model. Conclusion:We had identified nine genes, including PDGFRA, VIM, RBP1, RBP1, TNC, CD3D, GNLY, LCK, and ZAP70, which played important roles in the occurrence and development of BLCA. The prognostic model based on these genes had good accuracy in predicting the OS of patients and might be promising candidates of therapeutic targets. This study may provide a new insight for the diagnosis, treatment and prognosis of BLCA from the perspective of immunology. However, further experimental studies are necessary to reveal the underlying mechanisms by which these genes mediate the progression of BLCA.

It is essential to promote the intelligence and autonomy of Maritime Autonomous Surface Ships (MASSs). This study proposed an automatic collision-avoidance method based on an improved Artificial Potential Field (APF) with the formation of MASSs (F-MASSs). Firstly, the navigation environment model was constructed by the S-57 Electronic Navigation Chart (ENC) data in Tianjin Port. The Formation Ship State Parameter (FSSP) definition was proposed for the port environment under multiple constraints that considered the navigation conditions of the MASSs. The formation pattern transformation was settled by changing the formation ship state parameter. Considering the constraints of an ‘unmanned–manned’ encounter situation, the static obstacles, and the design of the channel area improved artificial potential method for the formation. Finally, the simulation experiment was carried out in the sea near Tianjin Port to verify the effectiveness of the algorithm under multiple constraints. The results indicate that the method can satisfy the integrated operation of collision avoidance and path following in a constrained environment, and it can support the application of merchant F-MASS autonomous navigation in the future.

With the continuous development of hyperspectral image technology and deep learning methods in recent years, an increasing number of hyperspectral image classification models have been proposed. However, due to the numerous spectral dimensions of hyperspectral images, most classification models suffer from issues such as breaking spectral continuity and poor learning of spectral information. In this paper, we propose a new classification model called the enhanced spectral fusion network (ESFNet), which contains two parts: an optimized multi-scale fused spectral attention module (FsSE) and a 3D convolutional neural network (3D CNN) based on the fusion of different spectral strides (SSFCNN). Specifically, after sampling the hyperspectral images, our model first implements the weighting of the spectral information through the FsSE module to obtain spectral data with a higher degree of information richness. Then, the weighted spectral data are fed into the SSFCNN to realize the effective learning of spectral features. The new model can maximize the retention of spectral continuity and enhance the spectral information while being able to better utilize the enhanced information to improve the model’s ability to learn hyperspectral image features, thus improving the classification accuracy of the model. Experiment results on the Indian Pines and Pavia University datasets demonstrated that our method outperforms other relevant baselines in terms of classification accuracy and generalization performance.

Hyperspectral imaging can simultaneously acquire spectral and spatial information of the samples and is, therefore, widely applied in the non-destructive detection of grain quality. Supervised learning is the mainstream method of hyperspectral imaging for pixel-level detection of mildew in corn kernels, which requires a large number of training samples to establish the prediction or classification models. This paper presents an unsupervised redundant co-clustering algorithm (FCM-SC) based on multi-center fuzzy c-means (FCM) clustering and spectral clustering (SC), which can effectively detect non-uniformly distributed mildew in corn kernels. This algorithm first carries out fuzzy c-means clustering of sample features, extracts redundant cluster centers, merges the cluster centers by spectral clustering, and finally finds the category of corresponding cluster centers for each sample. It effectively solves the problems of the poor ability of the traditional fuzzy c-means clustering algorithm to classify the data with complex structure distribution and the complex calculation of the traditional spectral clustering algorithm. The experimental results demonstrated that the proposed algorithm could describe the complex structure of mildew distribution in corn kernels and exhibits higher stability, better anti-interference ability, generalization ability, and accuracy than the supervised classification model.

Purpose: To assess the correlation between Liver Imaging Reporting and Data System (LIRADS) and digital substract angiography (DSA) and lipiodol deposits in cirrhotic nodules of LIRADS category ≥3 receiving interventional treatment. Methods: From June 2014 to June 2016, patients with cirrhotic nodules were identified retrospectively and MR images were reviewed by sub-specialty radiologists according to modified LIRADS v2014. Correlation between nodules of LIRADS category ≥3 and DSA findings and lipiodol deposits were analyzed. Results: 71 cirrhotic nodules were evaluated in 33 patients. 39/71 nodules were classified as LR-3, 9/71 nodules were categorized as LR-4, 23/71 nodules were grouped into LR-5. 43 nodules presented positive DSA, 37 nodules showed presence of lipiodol deposits during follow up. With the upgrade of LIRADS category of cirrhotic nodules, DSA and lipiodol deposits became more conspicuous. Spearman analysis demonstrated positive correlations between LIRADS and DSA (r = 0.567, P = 0.000) as well as LIRADS and lipiodol deposits (r = 0.616, P = 0.000). ROC analysis revealed a cut-off value of LR ≥ 4 resulted in a sensitivity of 67.4% and specificity of 89.3% in predicting positive DSA (RUC = 0.799, P ＜ 0.0001), and a sensitivity of 75.7% and specificity of 88.2% in predicting lipiodol deposits (RUC = 0.818, P ＜ 0.0001). Of 39 lesions of LR-3, 64.1% (25/39) showed negative DSA, and 76.9% (30/39) showed absence of lipiodol deposits during follow up. Logistic regression analysis identified arterial enhancement (OR = 26.837, P = 0.002) and lesion size (OR = 1.325, P = 0.022) were independently associated with positive DSA in nodule of LIRADS category ≥3, while no factors were associated with lipiodol deposits. Conclusion: The LIRADS can be used to predict DSA findings and lipiodol deposits in nodules with LIRADS score 3 and above. LIRADS 3 nodules tend to be DSA-negative and have less lipiodol deposits. Since the e-Navigation strategy was put forward, various countries and regions in the world have researched e-Navigation test platforms. However, the sources of navigation data are multi-source, and there are still difficulties in the unified acquisition, processing, analysis and application of multi-source data. Users often find it difficult to obtain the required comprehensive navigation information. The purpose of this paper is to use e-Navigation architecture to design and develop maritime data security management platform, strengthen navigation safety guarantee, strengthen Marine environment monitoring, share navigation and safety information, improve the ability of shipping transportation organizations in ports, and protect the marine environment. Therefore, this paper proposes a four-layer system architecture based on Java 2 Platform Enterprise Edition (J2EE) technology, and designs a unified maritime data storage, analysis and management platform, which realizes the intelligent, visualized and modular management of maritime data at shipside and the shore. This platform can provide comprehensive data resource services for ship navigation and support the analysis and mining of maritime big data. This paper expounds on the design, development scheme and demonstration operation scheme of the maritime data security management platform from the system structure and data exchange mode.

An optical Kerr gate (OKG) is an ultrafast optical switch based on the optical Kerr effect. The performance of a conventional OKG depends mainly on Kerr materials. Traditional Kerr materials do not demonstrate both large optical nonlinearity and an ultrafast response time. Therefore, the performance of a conventional OKG is limited by an inherent trade-off between high signal transmittance and fast switching time, which limits its application in many fields. We propose an improved femtosecond OKG with double gate pulses, based on the use of a birefringent crystal to realize an ultrashort switching time, even with a slow-response optical Kerr medium. We assessed the dependence of the double gate pulsed OKG (D-OKG)’s performance on the intensity ratio of the double gate pulses. A transmittance of 50% and a switching time of 142 fs were achieved. The D-OKG is convenient to construct, and its integrated performance is superior to that of a conventional OKG.

Background: One of the main aims of the updated Prostate Imaging Reporting and Data System Version 2 (PI-RADS v2) is to diminish variation in the interpretation and reporting of prostate imaging, especially among readers with varied experience levels. This study aimed to retrospectively analyze diagnostic consistency and accuracy for prostate disease among six radiologists with different experience levels from a single center and to evaluate the diagnostic performance of PI-RADS v2 scores in the detection of clinically significant prostate cancer (PCa). Methods: From December 2014 to March 2016, 84 PCa patients and 99 benign prostatic shyperplasia patients who underwent 3.0T multiparametric magnetic resonance imaging before biopsy were included in our study. All patients received evaluation according to the PI-RADS v2 scale (1–5 scores) from six blinded readers (with 6 months and 2, 3, 4, 5, or 17 years of experience, respectively, the last reader was a reviewer/contributor for the PI-RADS v2). The correlation among the readers' scores and the Gleason score (GS) was determined with the Kendall test. Intra-/inter-observer agreement was evaluated using κ statistics, while receiver operating characteristic curve and area under the curve analyses were performed to evaluate the diagnostic performance of the scores. Results: Based on the PI-RADS v2, the median κ score and standard error among all possible pairs of readers were 0.506 and 0.043, respectively; the average correlation between the six readers' scores and the GS was positive, exhibiting weak-to-moderate strength (r = 0.391, P = 0.006). The AUC values of the six radiologists were 0.883, 0.924, 0.927, 0.932, 0.929, and 0.947, respectively. Conclusion: The inter-reader agreement for the PI-RADS v2 among the six readers with different experience is weak to moderate. Different experience levels affect the interpretation of MRI images.

Effective use of port waterways is conducive to enhancing port competitiveness. To minimize the waiting time of ships, improve traffic efficiency, and enhance the applicability of the model to the presence of uncertain factors, a fuzzy scheduling optimization method for ships suitable for one-way waterways is proposed based on fuzzy theory. Considering the ambiguity of the speed of ships entering and exiting the port or the time it takes to cross the channel, the previous research on vessel scheduling on one-way waterways has been extended by introducing a triangular fuzzy number and a method for determining the feasible navigable time window of a ship subject to the tide height constraint was proposed. In this study, the genetic algorithm is used to construct the mathematical model for solving fuzzy vessel scheduling problems based on time optimization, and the minimum delay strategy is used to determine the service sequence. Then, the parameters setting are discussed in detail to find the optimal settings. Finally, an experimental comparative analysis of the randomly generated cases was conducted based on the simulated data. The results show that the designed fuzzy vessel scheduling algorithm reduces the dependence on the port environment, is versatile, and can effectively improve the efficiency of ship schedules and traffic safety compared to other methods. Moreover, it can avoid the problem of the illegal solution occurring in the manual scheduling method.

Supercontinuum (SC) generation using multiple thin plates is demonstrated with a femtosecond laser pulse. We propose an improved technique to obtain larger spectrum broadening and higher spectral intensity by employing mixed multiple thin plates with different thicknesses and materials. Furthermore, the spectrum has good stability, which is superior to that of the spectrum induced by the traditional single filament in bulk material. Our approach offers a route towards simple and stable SC generation for potential applications.

Local fat infiltration of the thoracic cavity is a rare initial presentation of Bochdalek hernia. We report a case of Bochdalek hernia in a child with leukemia that demonstrated initial local fat infiltration of the thoracic cavity on computed tomography scan and progressed to an obvious diaphragmatic hernia on subsequent follow-up. We suggest that initial local fat infiltration of the thoracic cavity on computed tomography scan may indicate a potential diaphragmatic hernia.

Interleaved LLC resonant converters are widely used in various fields. However, interleaved LLC converters under Pulse Frequency Modulation (PFM) will lose the regulation of individual phases, causing a load sharing problem. Existing load sharing solutions have limitations; for example, phase shedding and current sharing cannot be realized at the same time. This paper proposed a novel current sharing method for interleaved full-bridge LLC resonant converters. Based on Zero-Vector-Injection, the voltage applied to the resonant tank is controlled to compensate for the difference in gain caused by component tolerance. The modulation strategy is proposed to maintain soft switching after Zero-Vector-Injection, and the phase shedding technique is also used to improve the efficiency at a light load. The detailed theoretical analysis and implementation method are proposed and validated using simulations. Experiments are also carried out to verify the feasibility of the proposed strategy based on a 2-phase 1.8 kW prototype.

Background. Abernethy malformation is a rare splanchnic vascular abnormality characterizing extrahepatic abnormal shunts that is classified into types I and II. Abernethy malformation type I has a female predilection and is associated with a variety of concurrent hepatic benign or malignant tumours while type II with concurrent tumours is very rare in females. Case Report. We report a rare female case of Abernethy malformation type II with concurrent occupying lesion in the right liver, which was successfully transplanted; the occupying lesion was pathologically proven to be nodular hyperplasia. Conclusion. This case might provide further knowledge regarding Abernethy malformation. On imaging, the anatomy of portal vein should be carefully investigated to categorize Abernethy malformation, and a wide variety of differential diagnosis of concurrent occupying lesions should be taken into account.
