A good fishery water environment is an important guarantee for the formation and development of aquatic economic animals and plants, and the protection of the fishery water environment is the basic premise to maintain the sustainable development of fisheries. In order to firmly establish the concept of "Green water and green mountains are mountains of gold and silver", fully establish and maintain the ecological environment of fishery waters, and achieve a harmonious coexistence between man and nature, it′s necessary to strengthen the protection of the of law. The concept of ecological environment destruction in fishery waters has been clearly expressed in the Fisheries Law of the People′s Republic of China and other relevant laws, but there are many legal problems on how to determine it. Therefore, it is of great theoretical and practical significance to identify the ecological environment destruction in fishery waters. Improving the ecological environment destruction identification system helps to curb ecological environment destruction to a greater extent and protect ecological environment interests.

Nickel and chromium are both common heavy metals that pose serious environmental and health hazards. However, the exact mechanism by which nickel and/or chromium cause renal injury is unclear. Therefore, we explored the molecular mechanisms of renal injury caused by nickel and/or chromium poisoning from the perspective of mitochondrial dynamics and the Nrf2 antioxidant pathway. In this study, eighty 6-week-old C57BL/6J mice were randomly divided into four groups: control (Con, untreated), nickel (Ni, 110 mg/L Ni2+), chromium (Cr, 50 mg/L Cr6+), and combined nickel–chromium (Ni + Cr, 110 mg/L Ni2+, 50 mg/L Cr6+). The results showed that chronic nickel and/or chromium exposure inhibited body weight gain and impaired kidney function and structure in mice. Chronic nickel and/or chromium exposure led to the disruption of mitochondrial dynamics and thus induced oxidative stress. On the other hand, the Nrf2 antioxidant pathway may play an important regulatory role in mitigating oxidative stress-induced oxidative damage in kidney. The present study partially elucidated the molecular mechanism of renal injury induced by nickel and/or chromium exposure in mice and the regulatory role of the Nrf2 pathway in inducing oxidative injury from the perspective of mitochondrial dynamics. This provides a theoretical basis for the development of prevention and control strategies, and environmental protection measures.

In the traditional method, as the loss product and gas are considered to be saturated, the heat leak of a cryogenic vessel is the latent heat of loss product. However, the temperature of loss product and gas exceeds the saturated temperature as measured in tests. Therefore, a portion of the heat leak is absorbed by the loss product when it diffuses into the gas phase of a cryogenic vessel in the storage process. In this study, the energy equations of the gas and liquid are established in cryogenic vessels during storage to investigate the actual heat leak of a cryogenic vessel in a storage process by considering the temperature distribution of the gas. Then, an experiment was conducted to demonstrate that the actual heat leak of a cryogenic vessel contains not only the latent heat but also the enthalpy change of the loss product, which is produced as it diffused into the gas phase. The results showed that the enthalpy change increased as the liquid level decreased and was critical for determining the actual heat leak in cryogenic vessels in the storage process, with a ratio of approximately 12% to the latent heat when the liquid level ratio is 85%.

The thermal insulation performance of cryogenic vessels is evaluated by the boil-off gas (BOG) rate and heat leak as a reference standard; however, it consumes considerable testing time and working medium. In this study, we proposed a method of evaluating the thermal insulation performance of cryogenic vessels from the heat leak in the self-pressurization process, which can minimize the testing time and preserve the working medium. To improve the accuracy of heat leak calculation, the vapor temperature distribution of cryogenic vessels is analyzed and considered in the calculation of heat leak. However, it is ignored in the current standard test method. In this study, the energy equations of cryogenic vessels in the self-pressurization process were established by considering the vapor temperature distribution. Then, based on the experimental data, the heat leak was calculated for demonstrating the thermal insulation performance of cryogenic vessels depending on the new method. The results indicate that the vapor temperature should be seriously considered when the liquid level ratio is less than 50 %.

Article highlights 1. Two new rotor profiles are proposed and designed to improve the performance for Roots vacuum pump. 2. For the new rotor profile formed by a large rolling circle, its volume utilization rate is greater than 60%, which can pump more gas in a cycle and significantly better than that of traditional profile, such as arc profile. 3. For the new rotor profile formed by a small rolling circle, its pressure ratio is greater than traditional rotor profiles because its length of minimum gap between rotor profile and chamber is critical increased to decrease the back flow.

Background: Allergic rhinitis (AR) is a chronic respiratory disease. Hereditary factors played a key role in the pathogenesis of the AR. This study investigated the association between CLEC16A variants and AR risk in the Chinese population.Methods: We applied Agena MassARRAY technology platform to genotype five single nucleotide polymorphisms (SNPs) located in CLEC16A in 1004 controls and 995 cases. The association between CLEC16A SNPs (rs2286973, rs887864, rs12935657, rs11645657 and rs36045143) and AR risk were calculated by logistic regression analysis, with odds ratio (OR) and 95% confidence interval (CI). False-positive report probability (FPRP) was also used to assess the significant results to reduce false positives. Multifactor dimensionality reduction (MDR) was completed to assess the interaction between CLEC16A variants to predict AR risk.Results: Totally, CLEC16A (rs887864, rs12935657, rs2286973, rs11645657 and rs36045143) were significantly associated with AR risk. Therein, rs2286973, rs11645657 and rs36045143 were related to a decreased risk of AR in the people ≤ 43 years old, females and the people with BMI≤24, respectively. And rs887864 and rs12935657 were also associated with a decreased susceptibility of AR in the people >43 years old. Meanwhile, in the results of region stratification, rs887864 conferred a reduced risk to AR in the people from loess hilly area.Conclusion:CLEC16A variants conferred a decreased risk to AR in the Chinese population.

This study aims to investigate the effect of heat stress on the physiological metabolism of young laying hens and whether N-acetyl-l-cysteine (NAC) can effectively alleviate heat stress. 120 Hy-Line Brown laying hens aged 12 weeks were randomly divided into four groups: the control group (fed on basal diet under thermal neutral condition), HS group (fed on basal diet under heat stress condition), CN group (fed on the basic meal supplemented with 1,000 mg NAC per kg under thermal neutral condition), and HS+N group (fed on the basic meal was supplemented with 1000 mg NAC per kg under heat stress condition). The HS and HS+N groups were exposed to 36 ± 1°C for 10 h/day. The effects of NAC on the changes of serum concentrations of T3, T4, and CORT and hypothalamic gene and protein expressions induced by heat stress were measured. Results showed that heat stress upregulated the contents of T3, T4, and CORT, while NAC reduced the contents of T3, T4, and CORT. In addition, NAC downregulated AgRP expression, while upregulated the expression of POMC. Moreover, the expressions of AMPKα1, LKB1, and CPT1 were inhibited by NAC, while the expressions of AKT1, ACC, GPAT, and PPARα were increased after NAC treatment, and HMGR did not change significantly. Western blot and comprehensive immunofluorescence section of AMPK in the hypothalamus showed that NAC attenuated the activity of AMPK. In conclusion, NAC can enhance the resistance of laying hens to heat stress by alleviating the metabolic disorders of serum T3, T4, and CORT induced by heat stress, inhibiting the activation of the AMPK pathway and regulating the expression of appetite-related genes in the hypothalamus.

Nephropathogenic infectious bronchitis virus (NIBV) is one of the most important viral pathogens in the world poultry industry. Here, we used RT–qPCR, WB and immunofluorescence to explore the interaction between NIBV and the host innate immune system of the kidney. Multiple virions were found in the kidney tissues of the disease group under electron microscopy, and pathological changes such as structural damage of renal tubules and bleeding were observed by HE staining. In addition, we found that the mRNA levels of TLR7, TRAF6, and IKKβ were upregulated after NIBV infection. IRF7 mRNA levels decreased significantly at 5 dpi and increased significantly at 11 to 18 dpi. The NF-κB P65 mRNA level increased significantly at 5 to 18 dpi and decreased at 28 dpi. However, NIBV infection-induced NF-κB P65 protein levels were downregulated at multiple time points. Moreover, we demonstrated that the cytokine (IFN-γ, IL-8, and IL-6) mRNA and protein expression levels were increased significantly at multiple time points after NIBV infection. Furthermore, immunofluorescence analysis showed that NF-κB P65 and IFN-γ were mainly located in the nuclear or perinuclear region. The positive signal intensity of NF-κB P65 was significantly lower than that of the normal group at 1 to 5 dpi, and there was no significant change in the subsequent time period. The positive signal intensity of IFN-γ decreased significantly at 5 dpi, and increased significantly at 11 to 28 dpi. In conclusion, we found that NIBV promoted cytokine release through the TLR7/NF-κB signaling axis, thus causing kidney injury.

The heat leak in a cryogenic vessel is currently analyzed using a standard test based on measuring loss product. The test requires 72 h to complete; in addition, it requires that gas is discharged from the vessel to equalize the vessel pressure with the atmospheric pressure before the test can be performed. Therefore, the test is ineffective and wastes gas. In this study, new methods using constant pressure, constant flow rate, and self-pressurization are proposed to resolve the limitations posed by the standard test. Based on the heat leak transfer process and energy equations for cryogenic vessels, all factors, including temperature, pressure, latent heat, that influence the heat leak in cryogenic vessels are determined and analyzed. For a given cryogenic vessel, according to the equation between heat leak, temperature, and pressure, new test methods are proposed. Tests were conducted applying the new methods. The test results of each method showed that the heat leak in the cryogenic vessel was calculated accurately. We concluded that the new methods are effective and save significant amount of gas compared with the standard method.

The lack of cell retention clearly represents a potentially serious limitation for therapeutic efficacy of stem cells. To enhance the efficacy, we developed a novel hydrogel that is thermosensitive and biodegradable and possesses desirable stiffness in a solid form. Immediately after induction of myocardial infarction of male rat, cardiac outgrowth cells embedded in hydrogel (HG) or saline (CO) were injected directly into the peri-infarct area. Left ventricular ejection fraction, cell retention rate, and a spectrum of biochemical markers were measured to evaluate the effect of the treatment. Left ventricular ejection fraction was significantly higher in the cell-injected groups (HG and CO) than in the control group at 1 week after treatment. This functional benefit was continued only in the HG group, accompanied with more retained cells. Furthermore, the expression of insulin-like growth factor-1 was significantly higher in the HG group with less progression of cell apoptosis.

Sambucus williamsii Hance has been used in fractures for thousands of years, but research on its active components, such as morroniside, until now had not been carried out. In this study, morroniside was taken as the leading compound, and fourteen derivatives were synthesized. The promotion of osteoblast proliferation effect of the derivatives was evaluated on MC3T3-E1 cells. Five derivatives (2, 3, 4, 5, and 14) showed a good proliferation effect on MC3T3-E1 cells, and their promoted expression effects on OC (Osteocalcin) and ALP (Alkaline phosphatase) in MC3T3-E1 cells were measured. Compound 3 was shown to have the strongest proliferation effect (EC50 = 14.78 ± 1.17 μg/mL) and to significantly promote the expression of OC and ALP.

Background Complex interconnections are evident among gut microbiota, circulating metabolites, inflammatory cytokines, and the pathogenesis of abdominal aortic aneurysms (AAA), with the causal dynamics yet to be comprehensively elucidated. The primary objective of this study was to elucidate the potential causal relationships involving gut microbiota-mediated plasma metabolites, inflammatory cytokines, and AAA.Methods We utilized data from genome-wide association studies predominantly comprising individuals of European ancestry, encompassing four major gut microbiota signatures, 233 plasma metabolite signatures (N = 136,016), 91 inflammatory cytokine signatures (N = 14,824), and AAA signatures (N = 1,458,875). Mendelian randomization (MR), employed in a two-sample format, was utilized as a tool to investigate the potential causal pathways from gut microbiota to the development of AAA. Additionally, a two-step MR approach was employed to dissect the impact of plasma metabolites and inflammatory cytokines on the relationship between gut microbiota and AAA and to ascertain the mediated fractions.Results Our findings indicate that five phylum or family-identical bacteria, 175 plasma metabolites, and seven inflammatory factors are causally associated with AAA. Among them, five bacterial species from the same phylum or family, identified from different GWAS data, were strongly associated with AAA. Of these, two exhibited negative causality and three exhibited positive causality. We found that the phylum Firmicutes and the families Oscillospiraceae might reduce the risk of AAA, whereas the families Prevotellaceae, Sutterellaceae, and Aminobacteriaceae might increase the risk of AAA. Further screening indicated that phylum Firmicutes id.1672 (GCST90017114) may confer a protective effect against AAA by reducing triglyceride levels in medium/small high-density lipoprotein (HDL).Conclusion MR analysis has delineated a causal pathway from gut microbiota, through plasma circulating metabolites and inflammatory cytokines, to the pathogenesis of AAA. The role of intestinal flora and certain biomarkers may provide a reference for the diagnosis of AAA, and contribute to the prevention, diagnosis, and treatment of AAA disease.

Firstly, the antioxidant capacity of total flavonoids that was extracted from Sorbus pohuashanensis (Hance) Hedl was determined by ABTS+·radical-scavenging method, and MTT assay was used to mensurate the anti-tumor activity of total flavonoids In Vitro. Subsequently, the extraction conditions of flavonoids were optimized by the response surface methodology, the antioxidant activity was investigated simultaneously. Furthermore, the anti-tumor activity mechanism of flavonoids was simulated by molecular docking software. The antioxidant activity of flavonoids was correlated with the content in the process of optimizing the extraction condition. Moreover, The MTT assay revealed that SPF inhibited the activity of four sorts of cancer cells and positively correlated with the concentration of total flavonoids. And the experimental data demonstrated its anticancer activity was better at high concentrations. Finally, its anti-tumor activity was found that may be related to small molecules from flavonoids bind the apoptotic proteins Caspase-3, Caspase-8, and Caspase-9. The study could provide a theoretical basis for further pharmacodynamic research of Sorbus pohuashanensis (Hance) Hedl flavonoids, and improved a valuable attribute to Sorbus pohuashanensis (Hance) Hedl.
