Objective: This study aimed to explore andrographolide’s mechanism of action and its protective effect on noise-induced hearing loss (NIHL). Materials and Methods: A mice animal model for NIHL was established through exposure to broadband noise at 120 dB sound pressure level for 4 h. Transcriptomics analysis and pharmacodynamic experiments were carried out. Results: Andrographolide enters the inner ear and effectively prevents hearing damage following noise exposure in the mice model for permanent hearing loss. Moreover, treatment with andrographolide inhibited the excessive activation of inflammatory factors in the cochleae of noise-exposed mice. Conclusion: Andrographolide might be a promising candidate for auditory protective drug investigation.

Various excitation-induced loss mechanisms have been identified during the development of direct-gap semiconductor lasers. Recently, indirect-gap laser sources, particularly germanium (Ge) or GeSn based, have emerged due to silicon industry compatibility. Tensile strain is crucial for optical gain or low-threshold room-temperature operation in such media. This study investigates an excitation-induced optical loss mechanism of mechanical origin in Ge-based micro-cavities with all-around stressor layers, a popular platform for strain-engineered laser sources. Using Raman spectroscopy, photoluminescence, and simulations, we find that excitation lowers the optical gain by altering the strain profile. Heating causes Ge micro-cavities to expand within a constraining stressor layer, inducing compressive strain, which is explained by the mismatch in thermal expansion coefficients.

The recent advancement in wireless power transmission (WPT) has led to the development of wireless rechargeable sensor networks (WRSNs), since this technology provides a means to replenish sensor nodes wirelessly, offering a solution to the energy challenges faced by WSNs. Most of the recent previous work has focused on charging sensor nodes using wireless charging vehicles (WCVs) equipped with high-capacity batteries and WPT devices. In these schemes, a vehicle can move close to a sensor node and wirelessly charge it without physical contact. While these schemes can mitigate the energy problem to some extent, they overlook two primary challenges of applied WCVs: off-road navigation and vehicle speed limitations. To overcome these challenges, previous work proposed a new WRSN model equipped with one drone coupled with several pads deployed to charge the drone when it cannot reach the subsequent stop. This wireless charging pad deployment aims to deploy the minimum number of pads so that at least one feasible routing path from the base station can be established for the drone to reach every SN in a given WRSN. The major weakness of previous studies is that they only consider deploying a wireless charging pad at the locations of the wireless sensor nodes. Their schemes are limited and constrained because usually every point in the deployed area can be considered to deploy a pad. Moreover, the deployed pads suggested by these schemes may not be able to meet the connected requirements due to sparse environments. In this work, we introduce a new scheme that utilizes the Quad-Tree concept to address the wireless charging pad deployment problem and reduce the number of deployed pads at the same time. Extensive simulations were conducted to illustrate the merits of the proposed schemes by comparing them with different previous schemes on maps of varying sizes. In the case of large maps, the proposed schemes surpassed all previous works, indicating that our approach is more suitable for large-scale network environments.

BackgroundElectrical impedance tomography (EIT) has been shown to be useful in guiding individual positive end-expiratory pressure titration for patients with mechanical ventilation. However, the appropriate positive end-expiratory pressure (PEEP) level and whether the individualized PEEP needs to be adjusted during long-term surgery (>6 h) were unknown. Meanwhile, the effect of individualized PEEP on the distribution of pulmonary ventilation in patients who receive abdominal thermoperfusion chemotherapy is unknown. The primary aim of this study was to observe the effect of EIT-guided PEEP on the distribution of pulmonary ventilation in patients undergoing cytoreductive surgery (CRS) combined with hot intraperitoneal chemotherapy (HIPEC). The secondary aim was to analyze their effect on postoperative pulmonary complications.MethodsA total of 48 patients were recruited and randomly divided into two groups, with 24 patients in each group. For the control group (group A), PEEP was set at 5 cm H2O, while in the EIT group (group B), individual PEEP was titrated and adjusted every 2 h with EIT guidance. Ventilation distribution, respiratory/circulation parameters, and PPC incidence were compared between the two groups.ResultsThe average individualized PEEP was 10.3 ± 1.5 cm H2O, 10.2 ± 1.6 cm H2O, 10.1 ± 1.8 cm H2O, and 9.7 ± 2.1 cm H2O at 5 min, 2 h, 4 h, and 6 h after tracheal intubation during CRS + HIPEC. Individualized PEEP was correlated with ventilation distribution in the regions of interest (ROI) 1 and ROI 3 at 4 h mechanical ventilation and ROI 1 at 6 h mechanical ventilation. The ventilation distribution under individualized PEEP was back-shifted for 6 h but moved to the control group’s ventral side under PEEP 5 cm H2O. The respiratory and circulatory function indicators were both acceptable either under individualized PEEP or PEEP 5 cm H2O. The incidence of total PPCs was significantly lower under individualized PEEP (66.7%) than PEEP 5 cm H2O (37.5%) for patients with CRS + HIPEC.ConclusionThe appropriate individualized PEEP was stable at approximately 10 cm H2O during 6 h for patients with CRS + HIPEC, along with better ventilation distribution and a lower total PPC incidence than the fixed PEEP of 5 cm H2O.Clinical trial registration: identifier ChiCTR1900023897.

Objective: Exposure to bisphenol A (BPA) has been shown to increase the prevalence of obesity and its related insulin resistance (IR). Ceramide is a sphingolipid known to facilitate the production of proinflammatory cytokines and subsequently exacerbate inflammation and IR during the progression of obesity. Here, we investigated the effects of BPA exposure on ceramide de novo synthesis and whether increased ceramides aggravate adipose tissue (AT) inflammation and obesity-related IR. Methods: A population-based case–control study was conducted to explore the relationship between BPA exposure and IR and the potential role of ceramide in AT in obesity. Next, we used mice reared on a normal chow diet (NCD) or a high-fat diet (HFD) to verify the results from the population study and then investigated the role of ceramides in low-level BPA exposure with HFD-induced IR and AT inflammation in mice treated with or without myriocin (an inhibitor of the rate-limiting enzyme in de novo ceramide synthesis). Results: BPA levels are higher in obese individuals and are significantly associated with AT inflammation and IR. Specific subtypes of ceramides mediated the associations between BPA and obesity, obesity-related IR and AT inflammation in the obesity group. In animal experiments, BPA exposure facilitated ceramide accumulation in AT, activated PKCζ, promoted AT inflammation, increased the expression and secretion of proinflammatory cytokines via the JNK/NF-κB pathway, and lowered insulin sensitivity by disrupting IRS1-PI3K-AKT signaling in mice fed a HFD. Myriocin suppressed BPA-induced AT inflammation and IR. Conclusion: These findings indicate that BPA aggravates obesity-induced IR, which is partly via increased de novo synthesis of ceramides and subsequent promotion of AT inflammation. Ceramide synthesis could be a potential target for the prevention of environmental BPA exposure-related metabolic diseases.
