Landscape architecture education in the 21st century witnessed a transition from merely art and engineering to a future that integrates knowledge in science, psychology, environmental studies, ecology, and more. Using planting and environmental design practice as an example, the education focus needs to shift from purely demonstrating aesthetics to a comprehensive framework that integrates aesthetics, ecological services, and social and spatial functionality. This paper explored the gaps in planting and environmental design between pedagogy and practice. The result indicates that college-level education in planting and environmental design is majorly focused on plant identification, ornamental properties, and construction drawing communications, whereas the practical field indicates an expectation of more horticultural and ecological knowledge in design practice from college-level education. To bridge the gap, the article identified the proper trajectories through qualitative research including a questionnaire survey, semi-structured interview, content analysis and cross analysis to explore a sustainable development education framework of planting and environmental design based on the barriers of knowledge dissemination between academia and practice.

Abstract The combination between macroscopic structure designs and microscopic material designs offers tremendous possibilities for the development of advanced electromagnetic wave (EMW) absorbers. Herein, we propose a metamaterial design to address persistent challenges in this field, including narrow bandwidth, low–frequency bottlenecks, and, particularly, the urgent issue of robustness (i.e., oblique, and polarized incidence). Our absorber features a semiconductive metal-organic framework/iron 2D/2D assembly (CuHT–FCIP) with abundant crystal/crystal heterojunctions and strong magneto-electric coupling networks. This design achieves remarkable EMW absorption across a broad range (2 to 40 GHz) at a thickness of just 9.3 mm. Notably, it maintains stable performance against oblique incidence (within 75°) and polarizations (both transverse electric and transverse magnetic). Furthermore, the absorber demonstrates high specific compressive strength (201.01 MPa·cm3·g−1) and low density (0.89 g·cm−3). This advancement holds promise for developing robust EMW absorbers with superior performance.

With global warming, crop plants are challenged by heat stress during reproductive growth, leading to male sterility and yield reduction. The balance between reactive oxygen species (ROS) generation and scavenging is disrupted by stress, resulting in oxidative stress which is harmful to crop growth. However, recent advances have shown that ROS signaling via proteins that sense the redox state is positive for plant performance under stress. ROS signaling is also involved in regulating anther development, such as timely tapetum degeneration. We summarize recent progress in uncovering the mechanism of heat stress effects on ROS homeostasis and discuss the relationship between oxidative stress and metabolic disorder, which is often observed during anther development under heat stress. We propose experimental and breeding strategies to improve field crop tolerance to heat stress.

Abstract Background Misconceptions about male nurses not only exacerbate the gender imbalance in the nursing profession but also negatively impact male nurses embarking on their careers. Currently, no tool exists to measure the gender biases toward males in nursing among nursing students in China. Consequently, the primary objective of this study is to assess the validity and reliability of the Chinese translation of the Gender Misconceptions of Men in Nursing (GEMINI) scale among nursing students. Methods This cross-sectional study involved 1,102 nursing students from China who participated online. We utilized the Brislin translation technique with a forward-backward approach. To determine the factor structure within the Men in Nursing Gender Misconceptions Scale’s Chinese version, both exploratory (EFA) and confirmatory factor analysis (CFA) were applied. The scale’s internal consistency was measured through the Cronbach’s alpha coefficient, corrected item-total correlation, and a retest reliability assessment. Results The scale showed a content validity index of 0.938 and a retest reliability of 0.844. EFA indicated a two-factor structure for the translated instrument. CFA revealed a chi-square/degree of freedom of 3.837, an incremental fit index (IFI) of 0.952, a goodness-of-fit index (GFI) of 0.910, a comparative fit index (CFI) of 0.952, and an RMSEA of 0.073, all of which were within acceptable limits. The scale’s Cronbach’s α was 0.953, and the corrected item-total correlations ranged between 0.539 and 0.838. Gender-based misconceptions about men in nursing among students appeared to be influenced by their gender and whether they considered a nursing program as their first choice when applying for a major. Misconceptions about male nurses are greater among men and those who do not consider nursing programs as their first choice when applying for a major. Conclusions The Chinese adaptation of the GEMINI scale showcased high reliability and validity. It stands as a potential instrument to gauge gender misconceptions concerning male nurses among Chinese nursing students.

Abstract Fusarium wilt (FW) is widespread in global cotton production, but the mechanism underlying FW resistance in superior‐fiber‐quality Sea Island cotton is unclear. This study reveals that FW resistance has been the target of genetic improvement of Sea Island cotton in China since the 2010s. The key nonsynonymous single nucleotide polymorphism (SNP, T/C) of gene Gbar_D03G001670 encoding protein phosphatase 2C 80 (PP2C80) results in an amino acid shift (L/S), which is significantly associated with FW resistance of Sea Island cotton. Silencing GbPP2C80 increases FW resistance in Sea Island cotton, whereas overexpressing GbPP2C80 reduces FW resistance in Arabidopsis. GbPP2C80 and GbWAKL14 exist synergistically in Sea Island cotton accessions with haplotype forms “susceptible–susceptible” (TA) and “resistant–resistant” (CC), and interact with each other. CRISPR/Cas9‐mediated knockout of GbWAKL14 enhances FW and Verticillium wilt (VW) resistance in upland cotton and overexpression of GbWAKL14 and GbPP2C80 weakens FW and VW resistance in Arabidopsis. GbPP2C80 and GbWAKL14 respond to FW and VW by modulating reactive oxygen species (ROS) content via affecting MPK3 expression. In summary, two tandem genes on chromosome D03, GbPP2C80, and GbWAKL14, functions as cooperative negative regulators in cotton wilt disease defense, providing novel genetic resources and molecular markers for the development of resistant cotton cultivars.

ABSTRACT: Exosome-mediated horizontal and vertical transmission of subgroup J avian leukosis virus (ALV-J) in poultry flocks can lead to growth inhibition and severe immunosuppression. However, there are few reports on the early infection of chicken embryonic stem cells (cESCs) with ALV-J. In this study, we confirmed that early infection with ALV-J can accelerate the differentiation of cESCs and promote the secretion of exosomes. To investigate the modulation strategy of ALV-J in cESCs, circRNA sequencing was performed for further analysis. A total of 305 differentially expressed circRNAs (DECs) were obtained, including 71 upregulated DECs. Circ-CCDC7 was found to be the most upregulated DEC and was assessed by qRT-PCR, with the result consistent with the result of circRNA-seq. Based on qRT-PCR, gga-miR-6568-3p was found to be the target of the top 3 DECs, including circ-CCDC7, and the stem cell marker gene Pax7 was identified as the target gene of gga-miR-6568-3p. This study demonstrated that exosomal circ-CCDC7/gga-miR-6568-3p/Pax7 accelerates the differentiation of cESCs after early infection with ALV-J.

Increasing utilization of titanium dioxide nanoparticles (TiO2-NPs), with a wide range of applications, has resulted in the increased release of the material into the environment. In this study, we combined Illumina short-line RNA sequencing (RNA-Seq) and PacBio Iso-sequencing methods to obtain the transcriptome of Acipenser schrenckii livers treated with 5.27 (TH), 0.527 (TM), and 0.0527 mg/L (TL) TiO2-NPs for 14 days to elucidate the mechanisms underlying the effects of TiO2-NPs on sturgeons. In total, 39.67G subread bases and 31092 de-redundant transcripts (8929 unigenes) were obtained from A. schrenckii livers, with a mean length of 2289 bp. Moreover, 293 differentially expressed genes (DEGs) related to TiO2-NP stress were identified using DESeq; 175, 174, and 62 DEGs were identified in the TH vs. TC, TM vs. TC, and TL vs. TC groups. The Kyoto Encyclopedia of Genes and Genomes pathways of DEGs related to immunity (neuroactive ligand-receptor interaction, rheumatoid arthritis, NF-kappa B signaling pathway, and graft-versus-host disease) were significantly altered by TiO2-NP exposure, and the expression of interleukin-8, a possible biomarker for environmental pollution, was significantly upregulated. The pathways of alpha-linolenic acid metabolism, glycerolipid metabolism, and arachidonic acid metabolism associated with the defense against oxidative stress were also influenced by TiO2-NP exposure. The activity of carboxyl ester lipase involved in glycerolipid metabolism was rapidly downregulated in the TH and TM groups which may play a major role in glycerolipid metabolism regulation. The expression patterns of eight selected genes were generally consistent with the quantitative real-time PCR analysis results.

Abstract The potential of three‐dimensional (3D) printing technology in the fabrication of advanced polymer composites is becoming increasingly evident. This review discusses the latest research developments and applications of 3D printing in polymer composites. First, it focuses on the optimization of 3D printing technology, that is, by upgrading the equipment or components or adjusting the printing parameters, to make them more adaptable to the processing characteristics of polymer composites and to improve the comprehensive performance of the products. Second, it focuses on the 3D printable novel consumables for polymer composites, which mainly include the new printing filaments, printing inks, photosensitive resins, and printing powders, introducing the unique properties of the new consumables and different ways to apply them to 3D printing. Finally, the applications of 3D printing technology in the preparation of functional polymer composites (such as thermal conductivity, electromagnetic interference shielding, biomedicine, self‐healing, and environmental responsiveness) are explored, with a focus on the distribution of the functional fillers and the influence of the topological shapes on the properties and functional characteristics of the 3D printed products. The aim of this review is to deepen the understanding of the convergence between 3D printing technology and polymer composites and to anticipate future trends and applications.

PurposeThis study aimed to evaluate the feasibility of establishing an arterial acute mesenteric ischemia (AMI) model in canines using transcatheter autologous thrombus administration.Materials and methodsTen canines were divided into the experimental group (Group A, n = 5) and the sham group (Group B, n = 5). The canines in Group A received thrombus administration to the superior mesenteric artery (SMA) through a guiding catheter, while the canines in Group B received normal saline administration. Blood samples were collected and tested at baseline and 2 h after modelling. Canines in Group A underwent manual thromboaspiration after blood and intestine samples were collected. Ischaemic grades of intestinal mucosa were evaluated under light microscopes.ResultsThe AMI models were successfully conducted in all canines without procedure-related vessel injury or death. At the 2-h follow-up, the high-sensitivity C-reactive protein and D-dimer in Group A were significantly higher than in Group B (5.72 ± 1.8 mg/L vs. 2.82 ± 1.5 mg/L, p = 0.024; 2.25 ± 0.8 μg/mL vs. 0.27 ± 0.10 μg/mL, p = 0.005; respectively). Objectives: To develop a novel and accurate nomogram to predict survival without amputation in patients with acute lower limb ischemia (ALLI) during the first year following endovascular therapy. Methods: Patients with ALLI who underwent endovascular therapy in our department between January 2012 and September 2020 were screened and included in the research. The included patients were randomly divided into a training and validation cohorts, respectively. Univariate and multivariate analyses were used in the training cohort to identify independent risk factors for amputation-free survival (AFS). A nomogram was then developed according to the identified independent risk factors. The nomogram was then validated in the validation cohort. Results: 415 Chinese patients with 417 affected limbs were included in this study. Among these patients, 311 patients were classified into the training cohort and 104 patients were assigned to the validation cohort. Litopenaeus vannamei, also known as the Pacific white shrimp or white foot shrimp, is a member of the Crustacea, Decapoda, Palaemonidae, and Litopenaeus families. This species is mainly distributed on the Mexican and Pacific coasts of South America. Owing to its excellent immune characteristics, L. vannamei is currently one of the most important economic shrimp species in China.In the aquaculture process and under natural environmental conditions, low dissolved oxygen (DO) condition or even hypoxia frequently occurs. Under high-density culture operations, acute hypoxia is one of the major factors affecting the survival rate of shrimp and the quality of aquaculture water, causing not only a large number of shrimp deaths in the short term but also water decay and fermentation, increased turbidity, and other phenomena. The genetic improvement of L. vannamei, as well as the screening and breeding of exceptionally tolerant species under low DO conditions, are crucial to the sustainable development of the shrimp industry. Global selective breeding efforts based on quantitative genetics to improve genetic progress in L. vannamei have been extensive, and the main targeted traits were growth, survival, disease resistance, and stress tolerance. Few studies have been conducted on the evaluation of genetic parameters and screening of strains for hypoxic tolerance traits in L. vannamei; in particular, the genetic parameters and cytological characteristics of key tissues of L. vannamei under short-term highly lethal DO levels (0.3–0.5 mg/L) have not been reported.In this study, we used two strains of L. vannamei, GK (a strain with high disease resistance) and K (a strain with fast growth characteristics), each with a total of 6 560 shrimp from 41 families, to count survival traits at highly lethal DO levels, analyze differences in hypoxia tolerance traits between strains and within families of the same strain, and evaluate genetic parameters. Individual gill, muscle, and hepatopancreas tissues from families with significantly different tolerance levels were histologically investigated and compared at the cellular level. The results showed that there was a significant difference (P < 0.05) in semi-lethal survival (SS50) between families within both the GK and K strains when overall semi-lethality was reached under hypoxic environmental conditions of 0.3 mg/L, with SS50 values of 49.30% for GK and 42.52% for K. The coefficients of variation for survival times of families within each strain were 60% and 45% for GK and K, respectively. Using the threshold trait animal model, the genetic parameters were estimated to be between 0.345±0.031 and 0.378±0.029 using the survival status of the individual at semi-lethal levels as the observed value (1 for survival and 0 for death), and the heritability was between 0.219±0.031 and 0.237±0.029 following transformation on a continuously varying scale of observed values, indicating a moderate level of heritability. Moreover, the gill, muscle, and hepatopancreas tissues of the hypoxia-tolerant and hypoxia-sensitive families in the GK strain exhibited varying degrees of damage and different degrees of variations in the physiological characteristics of resistance among different families. This may be related to the resistance tolerance strength of different GK families. In contrast to the gill, muscle, and hepatopancreas tissues of the control shrimp, the equivalent tissues of shrimp from tolerant and sensitive families showed different degrees of changes following stress. The gill lumen of the filaments of the experimental group from sensitive families increased, the number of blood cells increased, the structure of the epithelial layer was gradually destroyed or even disintegrated, the vacuolation of the hepatopancreas was severe, the lumen of the ducts became irregularly deformed, and the muscle tissue muscle bundle interval widened with a certain degree of deformation. These results suggested that there was abundant genetic variation in the tolerance traits of L. vannamei at highly lethal DO levels and that this species was amendable to selective breeding practices. This study provides a reference and basis for the selection and breeding of hypoxia-tolerant traits in L. vannamei.

Aquaculture in large water bodies has become an important culture mode of Litopenaeus vannamei in coastal waters. Fractionated chlorophyll-a (Chl-a) and environmental factors of the large water ponds with high salinity (54, n=3) and the control ponds (32, n=3) were investigated from May to July 2020 to explore the variations in Chl-a, phytoplankton particle size, and the response to environmental factors during the aquaculture season. Pearson correlation analysis was performed to analyze the relationship between the environmental factors and the size-fractionated Chl-a concentration. Partial redundancy analysis (RDA) was applied to assess the effects of environmental factors (including silicate, active phosphate, ammonia salt, nitrite, nitrate, water temperature, salinity, dissolved organic nitrogen, and dissolved organophosphorus) on total Chl-a, Chl-a of micro phytoplankton (micro Chl-a), Chl-a of nano phytoplankton (nano Chl-a), and Chl-a of pico phytoplankton (pico Chl-a). The following results were obtained:1) Diurnal variation of Chl-a: Total Chl-a of the high-salinity group showed no significant diurnal variation (P > 0.05). Total Chl-a of the control group showed significant diurnal change in May and June (P < 0.05). The highest value of total Chl-a in May occurred at 15:00, while the highest value of that in June was at 08:00. For size-fractionated Chl-a, pico Chl-a in the high-salinity group showed significant diurnal variation in July (P < 0.05), with the highest value appearing at 12:00. Micro Chl-a in the control group showed significant diurnal changes in May, June, and July (P < 0.05), and nano Chl-a in the control group showed significant diurnal changes in June (P < 0.05).2) Monthly changes of Chl-a: The lowest and highest values of total Chl-a occurred in June and July, respectively. Total Chl-a in July was significantly higher than that in May and June (P < 0.05). For size-fractionated Chl-a, the pico Chl-a and nano Chl-a of the high-salinity group in July were significantly higher than those in May and June (P < 0.05) and showed no significant difference between May and June (P > 0.05). Pico Chl-a, nano Chl-a, and micro Chl-a of the control group in July were significantly higher than those in May and June (P < 0.05) and showed no significant difference between May and June (P > 0.05).3) Contribution of size-fractionated phytoplankton in high-salinity and control groups: The contribution of micro Chl-a, nano Chl-a, and pico Chl-a to total Chl-a in the high-salinity group were (15.64±0.16)%, (73.81±0.13)%, and (10.55±0.06)%, respectively. Nano Chl-a was dominant in May, June, and July. The contribution of pico Chl-a increased from 6.43% in May to 16.81% in July, and exceeded that of micro Chl-a. The contributions of micro Chl-a, nano Chl-a, and pico Chl-a to total Chl-a in the control group were (52.29±0.10)%, (41.82±0.10)%, and (5.59±0.01)%, respectively. Micro Chl-a concentration had a major advantage in May and June, accounting for 59.64% and 57.49%, respectively. Nano Chl-a concentration accounted for 35.46% and 36.90%, respectively. By July, nano Chl-a had a major advantage, contributing to 53.09%.4) Pearson correlation analysis showed no significant correlation between the diurnal variation of Chl-a and the environmental factors of the high-salinity group in May and June (P < 0.05). Yet, the concentrations of nano Chl-a and total Chl-a were negatively correlated with the concentration of nitrate in July (P < 0.05). The concentrations of micro Chl-a and total Chl-a were positively correlated with those of silicate (P < 0.05). For the control group, Pearson correlation analysis showed a significant positive correlation between nano Chl-a and water temperature (P < 0.05). Total Chl-a and phosphate were negatively correlated in May (P < 0.05). There was a significant negative correlation between pico Chl-a and nitrate in July (P < 0.05).5) For the high-salinity group, RDA revealed a significant positive correlation between Chl-a and water temperature, and the contribution of nano Chl-a increased with the increase in temperature. Total Chl-a was positively correlated with silicate and negatively correlated with phosphate, dissolved organic nitrogen, and dissolved organophosphorus in the high-salinity group. For the control group, RDA showed that total Chl-a was positively correlated with dissolved organic nitrogen and negatively correlated with silicate and nitrite. In general, Chl-a in high-salinity ponds has a small diurnal variation, and the phytoplankton particle size gradually decreased with cultivation, which may be caused by the increasing temperature and high organic nitrogen concentration.

Abstract Background Currently, there are few scales used to assess the stressors experienced by undergraduate nursing interns during clinical practice, and the assessment of stressors during clinical practice is not comprehensive; the scale includes some unique stressors during training that is not available in the existing instruments used to assess nursing student practice stress in China. Aim The study aimed to explore the structure of the Chinese revision of the Student Nurse Stressor-14 Scale(SNS-14-CHI)and investigate the psychometric properties it among Chinese undergraduate nursing interns. Methods The original scale was culturally adjusted and revised after expert correspondence on the entries, and 414 undergraduate nursing interns were recruited from three cities in China to administer the questionnaire. Reliability was measured by internal consistency, fold-half reliability, and stability. Content validity was evaluated using exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) to assess the validity of the SNS-14-CHI. Results The SNS-14-CHI retained 14 items, the EFA supported a 2-factor structure, and the items’ factor attribution differed from the original scale. The CFA results showed a good model fit. The Cronbach coefficient of the scale was 0.934, and the coefficient values of the two factors were 0.890 and 0.898. The content validity index of the scale was 0.964.The cumulative variance contribution of the 2-factor structure was 60.445%. The split-half reliability and stability were 0.869,0.762, respectively. Conclusion The SNS-14-CHI has excellent reliability and validity among undergraduate nursing trainees. The evaluation results of the scale can provide a reference for nursing managers to develop educational programs and interventions to quantify nursing student stress.

In this study, the Ni/NiO catalyst was demonstrated to enhance the hydrogen storage performance of MgH2. The dehydrogenation of MgH2+10 wt% Ni/NiO started at approximately 180 °C, achieving 5.83 wt% of dehydrogenation within 10 min at 300 °C. Completely dehydrogenated, MgH2 began to rehydrogenate at about 50 °C, absorbing about 4.56 wt% of hydrogen in 10 min at 150 °C. In addition, the activation energies of dehydrogenation and rehydrogenation of MgH2+10 wt% Ni/NiO were 87.21 and 34.84 kJ/mol. During the dehydrogenation/rehydrogenation cycle, Mg2Ni/Mg2NiH4 could promote hydrogen diffusion, thus enhancing the hydrogen storage performance of Mg/MgH2.

ObjectiveThis study used latent profile analysis to explore the level of depression among US adults with obstructive sleep apnea hypopnea syndrome (OSAHS) symptoms and to identify different latent categories of depression to gain insight into the characteristic differences between these categories.MethodsThe data of this study were obtained from the National Health and Nutrition Examination Survey (NHANES) database, and the subjects with OSAHS symptoms were aged 18 years and older. The latent profile analysis (LPA) method was used to fit the latent depression categories in subjects with OSAHS symptoms. The chi-square test, rank sum test, and binary logistic regression were used to analyze the influencing factors of depression subgroups in subjects with OSAHS symptoms.ResultsThree latent profiles were identified: low-level (83.7%), moderate-level (14.5%) and high-level (1.8%) depression. The scores of 9 items in the high-level depression group were higher than those in the other two groups. Among them, item 4 “feeling tired or lack of energy” had the highest score in all categories.ConclusionDepression in subjects with OSAHS symptoms can be divided into low-level, moderate-level and high-level depression. There are significant differences among different levels of depression in gender, marital status, PIR, BMI, smoking, general health condition, sleep duration and OSAHS symptom severity.

Summary: 3D or 4D printing of metal structures requires extreme conditions or a multistage process. Here, we present a protocol for the preparation of highly conductive metallic composites using liquid metal gels at ambient conditions. We describe the steps to prepare ternary gels composed of copper particles, liquid metal, and water. We then detail procedures for 3D or 4D printing gels into highly conductive structures after adding a small amount of rheological modifier (methyl cellulose) using direct ink writing techniques.For complete details on the use and execution of this protocol, please refer to Xing et al. Avian primordial germ cells (PGCs) are essential in avian transgenic research, germplasm conservation, and disease resistance breeding. However, cultured PGCs are prone to fragmentation and apoptosis, regulated at transcriptional and translational levels, with N6-methyladenosine (m6A) being the most common mRNA modification. Resveratrol (RSV) is known for its antioxidant and anti-apoptotic properties, but its effects on PGCs and the underlying mechanisms are not well understood. This study shows that RSV supplementation in cultured PGCs improves cell morphology, significantly enhances total antioxidant capacity (p < 0.01), reduces malondialdehyde levels (p < 0.05), increases anti-apoptotic BCL2 expression, and decreases Caspase-9 expression (p < 0.05). Additionally, RSV upregulates the expression of m6A reader proteins YTHDF1 and YTHDF3 (p < 0.05). m6A methylation sequencing revealed changes in mRNA m6A levels after RSV treatment, identifying 6245 methylation sites, with 1223 unique to the control group and 798 unique to the RSV group. Combined analysis of m6A peaks and mRNA expression identified 65 mRNAs with significantly altered methylation and expression levels. Sixteen candidate genes were selected, and four were randomly chosen for RT-qPCR validation, showing results consistent with the transcriptome data. Notably, FAM129A and SFRP1 are closely related to apoptosis, indicating potential research value. Overall, our study reveals the protective effects and potential mechanisms of RSV on chicken PGCs, providing new insight into its use as a supplement in reproductive stem cell culture.

Subgroup J avian leukosis virus (ALV-J) is a major pathogen in poultry, causing substantial economic losses to the poultry industry worldwide. Exosomal small RNAs derived from virus-infected cells or biological fluids can serve as viral transmission vectors. However, the role and mechanism of exosomal miRNA in ALV-J infection are unclear. In this study, we demonstrated that exosomal microRNA-7-25207 (miR-7-25207) could increase the titers of ALV-J. Exosomes isolated from ALV-J-infected DF-1 cells (Exo-ALV-J) contained partial viral proteins from ALV-J and could transmit the infection to uninfected DF-1 cells, leading to productive infection. Additionally, the RNA expression profile of exosomes was altered following ALV-J infection. miRNA analysis revealed that the expression of exosomal miR-7-25207 increased. Overexpression of miR-7-25207 significantly increased the titers of ALV-J in transfected cells. Furthermore, miR-7-25207 directly suppressed the expression of Akt and PRC1. Akt, in turn, directly inhibited CyclinQ1 expression, while PRC1 directly interfered with YAF2 expression. In conclusion, ALV-J infection activates the expression of miR-7-25207, which is subsequently delivered via exosomes to uninfected cells, increasing ALV-J titers by targeting Akt-CyclinQ1 and PRC1-YAF2 dual pathways. These findings suggest that exosomal miR-7-25207 may serve as a potential biomarker for clinical parameters in ALV-J infection.

The current study aimed to provide a precise assessment of the genetic parameters associated with growth and white spot syndrome virus (WSSV) resistance traits in Pacific white shrimp (Litopenaeus vannamei). This was achieved through a controlled WSSV challenge assay and the analysis of phenotypic values of five traits: body weight (BW), overall length (OL), body length (BL), tail length (TL), and survival hour post-infection (HPI). The analysis included test data from a total of 1017 individuals belonging to 20 families, of which 293 individuals underwent whole-genome resequencing, resulting in 18,137,179 high-quality SNP loci being obtained. Three methods, including pedigree-based best linear unbiased prediction (pBLUP), genomic best linear unbiased prediction (GBLUP), and single-step genomic BLUP (ssGBLUP) were utilized. Compared to the pBLUP model, the heritability of growth-related traits obtained from GBLUP and ssGBLUP was lower, whereas the heritability of WSSV resistance was higher. Both the GBLUP and ssGBLUP models significantly enhanced prediction accuracy. Specifically, the GBLUP model improved the prediction accuracy of BW, OL, BL, TL, and HPI by 4.77%, 21.93%, 19.73%, 19.34%, and 63.44%, respectively. Similarly, the ssGBLUP model improved prediction accuracy by 10.07%, 25.44%, 25.72%, 19.34%, and 122.58%, respectively. The WSSV resistance trait demonstrated the most substantial enhancement using both genomic prediction models, followed by body size traits (e.g., OL, BL, and TL), with BW showing the least improvement. Furthermore, the choice of models minimally impacted the assessment of genetic and phenotypic correlations. Genetic correlations among growth traits ranged from 0.767 to 0.999 across models, indicating high levels of positive correlations. Genetic correlations between growth and WSSV resistance traits ranged from (−0.198) to (−0.019), indicating low levels of negative correlations. This study assured significant advantages of the GBLUP and ssGBLUP models over the pBLUP model in the genetic parameter estimation of growth and WSSV resistance in L. vannamei, providing a foundation for further breeding programs.

ObjectivesThis study aimed to create and validate a radiomics nomogram for non-invasive preoperative Ki-67 expression level prediction in patients with bladder cancer (BCa) using contrast-enhanced CT radiomics features.MethodsA retrospective analysis of 135 patients was conducted, 79 of whom had high levels of Ki-67 expression and 56 of whom had low levels. For the dimensionality reduction analysis, the best features were chosen using the least absolute shrinkage selection operator and one-way analysis of variance. Then, a radiomics nomogram was created using multiple logistic regression analysis based on radiomics features and clinical independent risk factors. The performance of the model was assessed using the Akaike information criterion (AIC) value, the area under the curve (AUC) value, accuracy, sensitivity, and specificity. The clinical usefulness of the model was assessed using decision curve analysis (DCA).ResultsFinally, to establish a radiomics nomogram, the best 5 features were chosen and integrated with the independent clinical risk factors (T stage) and Rad-score. This radiomics nomogram demonstrated significant correction and discriminating performance in both the training and validation sets, with an AUC of 0.836 and 0.887, respectively. This radiomics nomogram had the lowest AIC value (AIC = 103.16), which was considered to be the best model. When compared to clinical factor model and radiomics signature, DCA demonstrated the more value of the radiomics nomogram.ConclusionEnhanced CT-based radiomics nomogram can better predict Ki-67 expression in BCa patients and can be used for prognosis assessment and clinical decision making.

Abstract Understanding the host‐guest interactions for thermally activated delayed fluorescence (TADF) emitters is critical because the interactions between the host matrices and TADF emitters enable precise control on the optoelectronic performance, whereas technologically manipulating the singlet and triplet excitons by using different kinds of host‐guest interactions remains elusive. Here, we report a comprehensive picture that rationalizes host‐guest interaction‐modulated exciton recombination by using time‐resolved spectroscopy. We found that the early‐time relaxation is accelerated in polar polymer because dipole‐dipole interaction facilitates the stabilization of the 1CT state. However, an opposite trend is observed in longer delay time, and faster decay in the less polar polymer is ascribed to the π‐π interaction that plays the dominant role in the later stage of the excited state. Our findings highlight the technological engineering singlet and triplet excitons using different kinds of host‐guest interactions based on their electronic characteristics.

AbstractCurrently, the inversion of remote sensing satellite images of water environment indicators mostly stays in the indicators with active optical characteristics, while there is less research on the inversion of most water quality indicators with non-optical activity properties, weak scattering and absorption of optical radiation, the size of their concentration has little effect on the spectral characteristics of the water body, such as TOC(Total Organic Carbon). In this paper, based on Pingzhai Reservoir, a dammed river in the karst mountainous area, the inversion model of TOC concentration was established based on BP neural network (BPNN) and sentinel-2 satellite remote sensing images. The results showed that the single bands with high correlation with the measured TOC concentration data were two vegetation red-edge bands B6 (740 nm) and B7 (783 nm) and one NIR band B8 (842 nm), and finally b7, b6 + b7, b7 + b8, b7 × b8 were selected as the input layers of BPNN for modeling through the combination of the bands, and their Pearson’s coefficients were −0.667, −0.656, −0.655, − 0.675.The inverse model established could reach a minimum RMSE of 0.235 mg/L and a maximum R2 of 0.889, which was superior to that of the conventional empirical model. Demonstrate the feasibility of a TOC inversion method based on Sentinel-2 data and BPNN to monitor TOC concentrations in Pingzhai Reservoir. The study successfully established a BP neural network inversion model of TOC concentration in Pingzhai Reservoir with low error, meanwhile, we analyzed the correlation between common water quality indicators and TOC in the reservoir, in which TOC showed significant positive correlation with WT and significant negative correlation with TN and EC, with Pearson’s coefficients of 0.655, −0.666, and −0.393, respectively. The article provides scientific theoretical foundation and technical support for water quality protection of water sources.

Abstract Plant anthers are composed of different specialized cell types with distinct roles in plant reproduction. High temperature (HT) stress causes male sterility, resulting in crop yield reduction. However, the spatial expression atlas and regulatory dynamics during anther development and in response to HT remain largely unknown. Here, the first single‐cell transcriptome atlas and chromatin accessibility survey in cotton anther are established, depicting the specific expression and epigenetic landscape of each type of cell in anthers. The reconstruction of meiotic cells, tapetal cells, and middle layer cell developmental trajectories not only identifies novel expressed genes, but also elucidates the precise degradation period of middle layer and reveals a rapid function transition of tapetal cells during the tetrad stage. By applying HT, heterogeneity in HT response is shown among cells of anthers, with tapetal cells responsible for pollen wall synthesis are most sensitive to HT. Specifically, HT shuts down the chromatin accessibility of genes specifically expressed in the tapetal cells responsible for pollen wall synthesis, such as QUARTET 3 (QRT3) and CYTOCHROME P450 703A2 (CYP703A2), resulting in a silent expression of these genes, ultimately leading to abnormal pollen wall and male sterility. Collectively, this study provides substantial information on anthers and provides clues for heat‐tolerant crop creation.

Abstract Flavivirus remodels the host endoplasmic reticulum (ER) to generate replication compartments (RCs) as the fundamental structures to accommodate viral replication. Here, a centralized replication mode of flavivirus is reported, i.e., flavivirus concentrates host ER in perinuclear main replication compartments (MRCs) for efficient replication. Superresolution live‐cell imaging demonstrated that flavivirus MRCs formed via a series of events, including multisite ER clustering, growth and merging of ER clusters, directional movement, and convergence in the perinuclear region. The dynamic activities of viral RCs are driven by nonstructural (NS) proteins and are independent of microtubules and actin. Moreover, disrupting MRCs formation by small molecule compounds inhibited flavivirus replication. Overall, the findings reveal unprecedented insight into dynamic ER reorganization by flavivirus and identify a new inhibition strategy.

ABSTRACT Translucent post-larvae disease (TPD), caused by Vibrio parahaemolyticus (Vp TPD), has become an emerging shrimp disease, affecting more than 70%–80% of coastal shrimp nurseries in China in spring 2020. Here, we investigated the key virulence factors of Vp TPD by analyzing protein fragments, related genomic information, as well as experimental challenge tests. After investigating the toxic effects of purified protein fragments with different molecular weights (MWs) from Vp TPD, we found that only the protein fragments with MW >100 kDa showed similar lethality to live Vp TPD in the experimental challenge test using post-larvae shrimp. Meanwhile, similar histopathological changes exhibiting in the hepatopancreas and midgut of the diseased individuals were observed in the post-larvae shrimp challenged with either bacterial protein fragments (MW >100 kDa) or live Vp TPD. Based on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and mass spectrometry analyses, two novel proteins, Vibrio high virulent protein (VHVP)-1 and VHVP-2, were identified as the candidates of key virulence factors to cause TPD. Moreover, VHVP-1 and VHVP-2 were found to be encoded by two genes (vhvp-1 and vhvp-2) tandemly located on a 187,791-bp plasmid and were predicted to depend on the same promoter following a comparative genomic analysis. Further epidemiological investigation and challenge test indicated that the V. parahaemolyticus isolate carrying only the vhvp-1 gene and lacking vhvp-2 gene could not cause mortality of experimental Penaeus vannamei post-larvae. The mutant (Δvhvp-2) by deleting vhvp-2 gene could only cause 4.92% of accumulative mortality of post-larvae that is similar to the non-Vp TPD Vibrio strain. Additionally, the complemented strains, Δvhvp-2/pBT3-vhvp-2 and Δvhvp-2/pwtCas9-vhvp-2, showed similar virulence to the wild-type Vp TPD. The results demonstrated that V. parahaemolyticus becomes lethal to post-larval shrimp by acquiring the VHVP-2 virulence factor. This study sheds light on further investigations of the pathogenic mechanism of Vp TPD and the development of strategies for early diagnosis of TPD in shrimp hatcheries. IMPORTANCE As a severe emerging shrimp disease, TPD has heavily impacted the shrimp aquaculture industry and resulted in serious economic losses in China since spring 2020. This study aimed to identify the key virulent factors and related genes of the Vp TPD, for a better understanding of its pathogenicity of the novel highly lethal infectious pathogen, as well as its molecular epidemiological characteristics in China. The present study revealed that a novel protein, Vibrio high virulent protein-2 (MW >100 kDa), is responsible to the lethal virulence of V. parahaemolyticus to shrimp post-larvae. The results are essential for effectively diagnosing and monitoring novel pathogenic bacteria, like Vp TPD, in aquaculture shrimps and would be beneficial to the fisheries department in early warning of Vp TPD emergence and developing prevention strategies to reduce economic losses due to severe outbreaks of TPD. Elucidation of the key virulence genes and genomics of Vp TPD could also provide valuable information on the evolution and ecology of this emerging pathogen in aquaculture environments.

Steel slag composite grout (SSCG) was prepared by a large amount of steel slag (SS) to reinforce expressway photovoltaic (PV) slopes and reduce carbon emission. Laboratory tests were conducted to investigate SSCG properties, including flowability, effective water-solid ratio (W/S), stone rate, initial and final setting time, as well as flexural strength (FS), unconfined compressive strength (UCS), FS/UCS ratio and microstructure of SSCG stone body. Ordinary Portland cement (OPC, 42.5) and sulphoaluminate cement (SAC, 42.5) grouts were selected to make contrasts. Based on the performance results, the recommended SSCG composition for engineering application is 40 % SS + 25 % blast furnace slag (BFS) + 20 % silicate cement clinker (CC) + 10 % fly ash (FA) + 5 % flue gas desulfurisation gypsum (FGDG). and the recommended admixture is: 6 % AA + 2 % bentonite + 0.4 % FND-C. With the above SSCG and related grouting techniques for slope soils, the soil cohesion was increased by 7.3–12.2 % and the angle of internal friction was increased by 10.2–19.7 %. To further validate the beneficial effects of SSCG on slope stability and the safe application of PV systems, numerical modelling calculations were conducted based on the soil sample test results. The simulations demonstrated the positive effect of SSCG in enhancing slope stability and facilitating the secure installation of PV systems on expressway slopes. The successful operation of grid-connected PV systems provided compelling evidence for the technical applicability of SSCG and it contributed to the application feasibility of PV systems to expressway slopes. The innovative use of SSCG in this study not only ensures the safety of construction projects but also contributes to environmental sustainability through the application of industrial slags.

ABSTRACT: Adenovirus serves as an excellent viral vector and is employed in vector vaccine research. Duck hepatitis A virus type 1 (DHAV1) and duck adenovirus type 3 (DAdV3) cause significant economic losses in the Chinese duck industry. In this study, we found an excellent exogenous gene insertion site in DAdV3 genome of CH-GD-12-2014 strain, within 3 intergenic regions (IGR). Subsequently, we generated a recombinant duck adenovirus named rDAdV3-VP1-188, which exhibits excellent replication characteristics and immunogenicity of DAdV3 and DHAV1. Animal experiments showed that rDAdV3-VP1-188 can provide 100% protection against the DAdV3 and 80% protection against DHAV1. These results showed that rDAdV3-VP1-188 could induce protection against DAdV3 and DHAV1 in ducks, thus indicating the feasibility of DAdV3 as a vector for the development of avian vector vaccines. These insights contribute to the further development of DAdV3 vectors and other adenovirus vectors.

ABSTRACT: In the early stages of embryonic development, a precise and strictly controlled hierarchy of gene expression is essential to ensure proper development of all cell types and organs. To better understand this gene control process, we constructed a small RNA library from 1- to 5-day-old chick embryos, and identified 2,459 miRNAs including 827 existing, 695 known, and 937 novel miRNAs with bioinformatic analysis. There was absolute high expression of a number of miRNAs in each stage, including gga-miR-363-3p (Em1d), gga-miR-26a-5p (Em2d and Em3d), gga-miR-10a-5p (Em4d), and gga-miR-199-5p (Em5d). We evaluated enriched miRNA profiles, identifying VEGF, Insulin, ErbB, MAPK, Hedgehog, TLR and Hippo signaling pathways as primary regulatory mechanisms enabling complex morphogenetic transformations within tight temporal constraints. Pathway analysis revealed miRNAs as pivotal nodes of interaction, coordinating cascades of gene expression critical for cell fate determination, proliferation, migration, and differentiation across germ layers and developing organ systems. Weighted Gene Co-Expression Network Analysis (WGCNA) generated hub miRNAs whose modular connections spanned regulatory networks, including: gga-miR-181a-3p (blue module), coordinating immunegenesis and myogenesis; gga-miR-126-3p (brown module), regulating vasculogenesis and angiogenesis; gga-miR-302c-5p (turquoise module), enabling pluripotency and self-renew; and gga-miR-429-3p (yellow module), modulating neurogenesis and osteogenesis. The findings of this study extend the knowledge of miRNA expression in early embryonic development of chickens, providing insights into the intricate gene control process that helps ensure proper development.

Abstract 3D orthogonal woven composites are receiving increasing attention with the ever-growing market of composites. A current challenge for these materials’ development is how to improve their damage tolerance in orthogonal and layer-to-layer structures under extreme loads. In this paper, a damage reduction strategy is proposed by combining structural and electromagnetic properties. An integrated experimental platform is designed combining a power system, a drop-testing machine, and data acquisition devices to investigate the effects of pulse current and impact force on woven composites. Experimental results demonstrate that pulse current can effectively reduce delamination damage and residual deformation. A multi-field coupled damage model is developed to analyze the evolutions of temperature, current and damage. Parallel current-carrying carbon fibers that cause yarns to be transversely compressed enhance the mechanical properties. Moreover, the microcrack formation and extrusion deformation in yarns cause the redistribution of local current among carbon fibers, and its interaction with the self-field produces an obvious anti-impact effect. The obtained results reveal the mechanism of damage reduction and provide a potential approach for improving damage tolerance of these composites.

Abstract The Multi-purpose Time Projection Chamber (TPC) for nuclear AsTrophysical and Exotic beam experiments (MATE) is being upgraded for the decay and active target experiments at the Heavy Ion Research Facility in Lanzhou (HIRFL). We have developed a gating grid driver to control the transitions between the closed and open states of the gating grid of the MATE-TPC to detect interesting rare decay events from a large amount of implanted ions. The gating grid driver is mainly composed of a digital control unit and a high-voltage switch unit. The digital control unit responds to the external trigger and generates control signals for the operation of the high-voltage control part based on the presetting instruction. The high-voltage switch unit is connected to two negative high voltages with different values and changes the voltages of neighboring wires of the gating grid based on the request for closing or opening the gate. A 500 ns switching time of the gating grid driver has been achieved from the closed to open state. The duration of the open state can be adjusted from 1 µs to 99 ms based on the experimental requirements. This gating grid driver can be used in a particle detector with a high voltage bias of up to ± 3000 V.

The Pacific white shrimp, Penaeus vannamei, is highly susceptible to white spot syndrome virus (WSSV). Our study explored the transcriptomic responses of P. vannamei from resistant and susceptible families, uncovering distinct expression patterns after WSSV infection. The analysis revealed a higher number of differentially expressed genes (DEGs) in the susceptible family following WSSV infection compared to the resistant family, when both were evaluated against their respective control groups, indicating that the host resistance of the family line influences the transcriptome. The results also showed that subsequent to an identical duration following WSSV infection, there were more DEGs in P. vannamei with a high viral load than in those with a low viral load. To identify common transcriptomic responses, we profiled DEGs across families at 96 and 228 h post-infection (hpi). The analysis yielded 64 up-regulated and 37 down-regulated DEGs at 96 hpi, with 33 up-regulated and 34 down-regulated DEGs at 228 hpi, showcasing the dynamics of the transcriptomic response over time. Real-time RT-PCR assays confirmed significant DEG expression changes post-infection. Our results offer new insights into shrimp’s molecular defense mechanisms against WSSV.

Sea Island cotton is renowned for its superior fiber quality. Although mechanical harvesting has the potential to significantly increase efficiency and reduce the production cost of Sea Island cotton, there is still little research in this area. In this study, we analyzed 240 Sea Island cotton germplasm resources and evaluated 19 traits related to mechanical harvesting. The coefficient of variation ranged from 5.42% to 66.96%, and the genetic diversity index spanned from 1.57 to 2.07. In most traits studied, there was a strong correlation between the height of the first fruiting branch and the defoliation rate. The 19 traits were categorized into 6 factorial groups by principal component analysis, in which the defoliation factor contributed the most (30.89%). The cluster analysis divided the 240 cotton accessions into four main groups, with the second group exhibiting favorable mechanical harvesting characteristics such as higher defoliation rate and first fruit branch height. Using stepwise regression, a model was constructed with the joint evaluation score F-value as the response variable and eight traits (X1: PH, X2: SNB, X3: SBN, X4: MBL, X5: AFBM, X7: MLIA, X8: NB, and X13: 15 d DR) as predictors: Y = −7.2 + 0.01X1 + 0.23X2 + 0.192X3 + 0.038X4 + 0.007X5 + 0.014X7 + 0.025X8 + 2.952X13. Selected materials suitable for machine harvesting, such as MoShi729, were identified. This study provides valuable theoretical insights into the mechanical harvesting of Sea Island cotton germplasm resources and identifies promising materials for targeted breeding and improvement programs.

White spot disease (WSD) outbreaks pose a significant threat to the Pacific white shrimp (Litopenaeus vannamei) farming industry. The causative agent is the white spot syndrome virus (WSSV). There are no effective treatments for WSD so far. Therefore, understanding the resistance mechanisms of L. vannamei against the WSSV is crucial. C-type lectins (CTLs) are important pattern recognition receptors (PRRs) that promote agglutination, phagocytosis, encapsulation, bacteriostasis, and antiviral infections. This study cloned the C-type lectin domain family 4 member F (LvCLEC4F) from L. vannamei. LvCLEC4F contains a 492 bp open reading frame (ORF) encoding a protein of 163 amino acids, including a carbohydrate recognition domain (CRD). Following a challenge with the WSSV, the expression profile of LvCLEC4F was significantly altered. Using RNA interference (RNAi) technology, it was found that LvCLEC4F promotes WSSV replication and affects the expression levels of genes related to the regulation of apoptosis, signaling and cellular stress response, and immune defense. Meanwhile, the hemolymph agglutination phenomenon in vivo was weakened when LvCLEC4F was knocked down. These results indicated that LvCLEC4F may play an important role in the interaction between L. vannamei and WSSV.

Marine animals possess genomes of considerable complexity and heterozygosity. Their unique reproductive system, characterized by high fecundity and substantial early mortality rates, increases the risk of inbreeding, potentially leading to severe inbreeding depression during various larval developmental stages. In this study, we established a set of inbred families of Fenneropenaeus chinensis, with an inbreeding coefficient of 0.25, and investigated elimination patterns and the manifestations of inbreeding depression during major larval developmental stages. Reduced-representation genome sequencing was utilized to explore the genotype frequency characteristics across two typical elimination stages. The results revealed notable mortality in hatching and metamorphosis into mysis and post-larvae stages. Inbreeding depression was also evident during these developmental stages, with depression rates of 24.36%, 29.23%, and 45.28%. Segregation analysis of SNPs indicated an important role of gametic selection before hatching, accounting for 45.95% of deviation in the zoea stage. During the zygotic selection phase of larval development, homozygote deficiency and heterozygote excess were the main selection types. Summation of the two types explained 82.31% and 89.91% of zygotic selection in the mysis and post-larvae stage, respectively. The overall distortion ratio decreased from 22.37% to 12.86% in the late developmental stage. A total of 783 loci were identified through selective sweep analysis. We also found the types of distortion at the same locus could change after the post-larvae stage. The predominant shifts included a transition of gametic selection toward normal segregation and other forms of distortion to heterozygous excess. This may be attributed to high-intensity selection on deleterious alleles and genetic hitchhiking effects. Following larval elimination, a greater proportion of heterozygous individuals were preserved. We detected an increase in genetic diversity parameters such as expected heterozygosity, observed heterozygosity, and polymorphic information content in the post-larvae stage. These findings suggest the presence of numerous recessive deleterious alleles and their linkage and suggest a major role of the partial dominance hypothesis. The results provide valuable insights into the mechanisms of inbreeding depression in marine animals and offer guidance for formulating breeding strategies in shrimp populations.

The Pacific whiteleg shrimp (Penaeus vannamei) is a highly significant species in shrimp aquaculture. In the production of shrimp larvae, noticeable variations in the reproductive capacity among female individuals have been observed. Some females experience slow gonadal development, resulting in the inability to spawn, while others undergo multiple maturations and contribute to the majority of larval supply. Despite numerous studies that have been conducted on the regulatory mechanisms of ovarian development in shrimp, the factors contributing to the differences in reproductive capacity among females remain unclear. To elucidate the underlying mechanisms, this study examined the differences in the ovarian characteristics between high and low reproductive bulks at different maturity stages, focusing on the cellular and molecular levels. Transmission electron microscopy analysis revealed that the abundance of the endoplasmic reticulum, ribosomes, mitochondria, and mitochondrial cristae in oocytes of high reproductive bulk was significantly higher than that of the low reproductive bulk in the early stages of ovarian maturation (stages I and II). As the ovaries progressed to late-stage maturation (stages III and IV), differences in the internal structures of oocytes between females with different reproductive capacities gradually diminished. Transcriptome analysis identified differentially expressed genes (DEGs) related to the mitochondria between two groups, suggesting that energy production processes might play a crucial role in the observed variations in ovary development. The expression levels of the ETS homology factor (EHF) and PRDI-BF1 and RIZ homology domain containing 9 (PRDM9), which were significantly different between the two groups, were compared using qRT-PCR in individuals at different stages of ovarian maturation. The results showed a significantly higher expression of the EHF gene in the ovaries of high reproductive bulk at the II and IV maturity stages compared to the low reproductive bulk, while almost no expression was detected in the eyestalk tissue of the high reproductive bulk. The PRDM9 gene was exclusively expressed in ovarian tissue, with significantly higher expression in the ovaries of the high reproductive bulk at the four maturity stages compared to the low reproductive bulk. Fluorescence in situ hybridization further compared the expression patterns of EHF and PRDM9 in the ovaries of individuals with different fertility levels, with both genes showing stronger positive signals in the high reproductive bulk at the four ovarian stages. These findings not only contribute to our understanding of the regulatory mechanisms involved in shrimp ovarian development, but also provide valuable insights for the cultivation of new varieties aimed at improving shrimp fecundity.

Pathogenic Aeromonas spp. are the etiological agents of Motile Aeromonas Septicemia (MAS). This study aimed to identify the pathogen of diseased tadpoles (Quasipaa spinosa) and the antibiotic-resistance characteristics of this bacterium. A Gram-negative bacterium, named strain QST31, was isolated from the ascites of diseased tadpoles and was identified as Aeromonas media based on physiological and biochemical tests, as well as molecular identification. Artificial infection experiments showed that strain QST31 was highly virulent to tadpoles, with an LC50 of 2.56 × 107 CFU/mL. The antimicrobial susceptibility of strain QST31 was evaluated using the disk diffusion method, and the results indicated that strain QST31 was resistant to 28 antibacterial agents. In addition, the whole genome of strain QST31 was sequenced, and the presence of antimicrobial resistance genes, integron, and transposon was investigated. Genes involved in adherence, hemolysis, type II secretion system (T2SS), T6SS, iron uptake system, and quorum sensing were identified in the genome of strain QST31. More than 12 antimicrobial resistance genes were predicted in the genome of strain QST31. Interestingly, a novel Tn7709 transposon harboring sul1, aadA16, catB3, blaOXA-21, aac(6′)-IIa, and tet(A) genes was identified. In conclusion, this is the first report on the isolation and identification of pathogenic A. media with multidrug resistance genes from diseased tadpoles. The results revealed that preventing and controlling aquatic animal diseases caused by multidrug resistance A. media will be a huge challenge in the future.

Abstract Background Although vaccination is one of the critical interventions to address global health issues, inadequate vaccination rates has become an international challenge. Vaccine hesitancy is the key to affecting inadequate vaccination rates. According to the WHO SAGE working group’s definition, vaccine hesitancy refers to delaying or refusing vaccination and has been ranked as one of the top 10 health threats. There has yet to be a scale that evaluates vaccination attitudes among Chinese adults. However, an attitude quantity, the adult vaccination attitude scale, has been developed to assess adult vaccination attitudes and reasons for vaccine hesitancy. Objective The Adult Attitudes to Vaccination Scale (ATAVAC) was initially developed by Professor Zoi Tsimtsiou et al. This study aimed to analyze the structure of the Chinese version of the ATAVAC and explore the relationship between adult vaccination attitudes, e-health literacy, and medical distrust. Methods After obtaining author permission for the initial scales, the study was translated using the Brislin back-translation method. 693 adults were enrolled to the study. To validate this hypothesis, participants finished the socio-demographic questionnaire, the Chinese version of the ATAVAC, the electronic Health Literacy Scale (e-HEALS) and the Medical Mistrust Index (MMI). The exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) were used to examine the underlying structure of the factors of the Chinese version of the Adult Vaccination Attitude Scale and to measure its reliability and validity. Results The Cronbach’s alpha coefficient for the Chinese version of the ATAVAC was 0.885, with Cronbach’s alpha coefficients ranging from 0.850 to 0.958 for each dimension. The content validity index was 0.90, and the retest reliability was 0.943. The exploratory factor analysis (EFA) supported the 3-factor structure of the translation instrument, and the scale had good discriminant validity. The confirmatory factor analysis (CFA) revealed a degree of freedom of 1.219, a model fit index (GFI) of 0.979, a normative fit index (NFI) of 0.991, a Tucker-Lewis index (TLI) of 0.998, a comparability index (CFI) of 0.998 and a root mean square error of approximation (RMSEA) of 0.026. Conclusion The results show that the Chinese version of the ATAVAC has demonstrated good reliability and validity. Hence, it can be used as an effective tool to assess vaccination attitudes among Chinese adults.

Temperature is an important environmental factor that affects the growth, development, and metabolism of shrimp. High temperatures tend to cause a decrease in the immunity and production performance of shrimp, resulting in a significant increase in shrimp disease and mortality. Based on transcriptome sequencing data, we screened for the involvement of DNA damage-inducible transcript 4 like (ddit4l) in the temperature stress process of Litopenaeus vannamei. Ddit4l (also known as Redd2 and Rtp801L) is involved in regulating cell survival, proliferation, and apoptosis by affecting DNA damage repair factors, hypoxia-inducible factors, and other signaling pathways when stimulated by DNA damage, hypoxia, ischemia, and oxidative stress, among others. Ddit4l may be an important transducer of pathological stress in autophagy through mammalian target of rapamycin (mTOR) signaling. Autophagy is a process in which cells use lysosomes to degrade their own damaged organelles and macromolecules, and the process plays an important role in maintaining the balance of protein metabolism and the stability of the intracellular environment, promoting cell growth and development. An important regulator of autophagy is the mTOR, which responds to various stimuli, such as growth factors, cellular energy status, oxygen concentrations, and stress, to regulate cell metabolism and growth. Ddit4l and its homolog ddit4 are upstream mTOR inhibitors in several tissues and cell models. We set up sudden temperature change and gradual temperature change experiments to induce a stress response in L. vannamei. In the gradual change experiment, the water temperature was increased from 26℃ to 38℃ at a rate of 4℃ per day, and cooled back to 26℃ at the same rate. Two individuals were randomly selected from each of the three parallel experiments in the experimental group at each time point. The eyestalk, gill, hepatopancreas, nerve, and muscle were used for subsequent RNA extraction and gene expression quantification. RNA quality and concentration were also examined. The partial ddit4l cDNA (complete ORF and partial UTR) of L. vannamei was obtained directly by PCR amplification to evaluate the structure and function of the Lv-ddit4l gene. The Lv-ddit4l sequence contained a 495 bp open reading frame, encoding 164 amino acids, with a molecular weight of 18.51 kDa, and an isoelectric point of 5.35. The instability index was 65.83, which classifies the protein as unstable. Lv-ddit4l contains a highly conserved RTP801-C domain. BLAST alignment and phylogenetic tree analysis showed that the Lv-ddit4l gene had 59.76% similarity with the Chionoecetes opilio ddit4l gene, and first clustered with C. opilio. A real-time RT-PCR confirmatory experiment considered three parallel groups for each sample with 18S rRNA as the reference gene. Relative gene expression was analyzed using the 2–ΔΔCt method. Quantitative analysis of gene expression levels showed that Lv-ddit4l was widely expressed in all of the examined tissues, with similar expression levels at 26℃. When the temperature was increased gradually from 26℃ to 32℃, the Lv-ddit4l gene in the eyestalk was significantly increased (P < 0.05), and was 10.37 times higher than that in the control (A-26℃). Therefore, this gene is hypothesized to respond to temperature changes in the eyestalk of L. vannamei. The expression of Lv-ddit4l in the eyestalk, gills, hepatopancreas, nerves, and muscles did not change significantly when the water temperature was gradually increased to the highest temperature of 38℃; it is speculated that this gene may play a role in pre-heat stress. When the temperature was gradually cooled from 38℃ to 32℃, the expression of Lv-ddit4l increased significantly in the gill and hepatopancreas (P < 0.05), which were respectively 3.37 and 2.03 times higher than that of the control group (A-26℃). There was no significant difference in gene expression under different levels of gradual changes in temperature stress in the nerve and muscle. The expression in the gill, hepatopancreas, and nerve was significantly up-regulated (P < 0.05) at a sudden change in water temperature to 36℃ for 10 min, and the change in expression in the hepatopancreas was 24.06 times higher than that in the control group (A-26℃). Gene expression in the gills and nerves was 1.22-fold and 3.67-fold higher than that in the control (A-26℃), respectively. When the water temperature was increased to 36℃ suddenly for 24 h, Lv-ddit4l gene expression increased significantly in the muscle (P < 0.05). Lv-ddit4l showed significant up-regulation of expression in the gill, hepatopancreas, nerve, and muscle with sudden changes in temperature, suggesting that this gene may play a role in the stress induced by sudden increases in temperature. When the temperature was restored from 38℃ and 36℃ to 26℃, the expression of the Lv-ddit4l gene was downregulated in L. vannamei. The above results indicate that Lv-ddit4l is associated with the response to high-temperature stress.

Abstract Purpose The present study aimed to investigate the preliminary safety and efficacy of rheolytic thrombectomy (RT) using an AngioJet Zelante DVT catheter or a Solent Omni catheter for acute proximal deep vein thrombosis (DVT). Material and methods We conducted a retrospective review of 40 patients who were treated with an AngioJet RT between January 2019 and January 2021, and then the patients were divided into the ZelanteDVT group (n = 17) and the Solent group (n = 23). Data on demographics, clinical characteristics, technical success, clinical success, complications, and early follow-up were analysed. Results No significant differences regarding demographics were detected (all p > .05). The technical success rates were both 100%. The ZelanteDVT group had a shorter duration of RT and a higher primary RT success than the Solent group (all p .05). Apart from transient macroscopic haemoglobinuria occurring in all the patients during the first 24 hours post-RT, none of the patients in either group suffered other procedure-related adverse events or major complications. Minor complications included bleeding events in 21.7% (5/23) of the patients in the Solent group and one (5.9%) patient in the ZelanteDVT group (p > .05). At 6 months, the frequency of PTS was 5.9% (1/17) in the ZelanteDVT group and 17.4% (4/23) in the Solent group (p > .05). Conclusion Both catheters are safe and effective in managing patients with proximal DVT, thus leading to improved clinical outcomes with few complications. The ZelanteDVT catheter was more effective than the Solent catheter in thrombectomy, thus allowing for faster extraction of the DVT with a shorter run time and lower proportions of patients with adjunctive CDT.

Cold seeps create diverse habitats in the deep sea and play an important role in the global carbon cycling. Anaerobic oxidation of methane (AOM) and biogenic mineralization are essential carbon pathways of methane and carbon transformation in cold seeps, however, the effects of habitat heterogeneity on the processes are still poorly understood. In this study, we investigated the microbial communities and mineral assemblages at distinct habitats in the Haima cold seep and their relationships with environmental factors. These habitats were classified as methane seep site (MS), seep-free faunal habitat (FH), and control site (CS). Bacterial communities were significantly different among the three habitats. ANME-3 archaea, Sulfurovum bacteria, and mineralization-associated microbes (e.g., Campylobacterales) were detected in high relative abundances at ROV2. Mineralogical analysis revealed abundant calcite minerals at the seep site, indicating that authigenic carbonate minerals were formed at highly active seep. Multivariate statistical analysis demonstrated that the concentrations of SO42–, Ca2+, and Mg2+ were significantly correlated with the presence of calcite minerals and bacterial communities. These results suggested that AOM-accompanied authigenic carbonate formation is an important factor influencing the mineral assemblages in seep habitats. This finding improves our understanding of marine microbial carbon cycling.

Summary: Electromagnetic waves have caused great harm to military safety, high-frequency electronic components, and precision instruments, and so forth, which urgently requires the development of lightweight, high-efficiency, broadband electromagnetic waves (EMW) absorbing materials for protection. As the basic fibrous materials, carbon fibers (CFs) and SiC fibers (SiCf) have been widely applied in EMW absorption due to their intrinsic characteristics of low density, high mechanical properties, high conductivity, and dielectric loss mechanism. Nevertheless, it has remained a great challenge to develop lightweight EMW-absorbing fibrous materials with strong absorption capability and broad frequency range. In this review, the fundamental electromagnetic attenuation mechanisms are firstly introduced. Furthermore, the preparation, structure, morphology, and absorbing performance of CFs and SiCf-based EMW absorbing composites are summarized. In addition, prospective research opportunities are highlighted toward the development of fibrous absorbing materials with the excellent absorption performance.

Background: Inferior vena cava (IVC) filters are commonly used intravascular devices designed to prevent fatal pulmonary embolism (PE), maintaining the IVC filter as centered as possible is fundamental for achieving its filtration function. Objective: This study aimed to characterize the tilt angles of IVC filter between the vascular access of internal jugular vein (IJV) and femoral vein (FV), as well as to identify factors associated with increased or decreased tilt angles between placement and retrieval. Design: This is a multicenter retrospective study. Methods: A multicenter retrospective study was conducted from October 2017 to March 2019. The primary outcome was the change in filter tilt between placement and retrieval. The secondary outcome was the identifications of factors associated with increased or decreased tilt angle. Relevant variables were analyzed using t- tests, Chi-square tests, Fisher’s exact tests, while multivariate logistic regression analysis was used to determine risk factors. Results: A total of 184 eligible patients were included in this study. The IJV group had a lower likelihood of tilt angle over 10° at the time of placement compared to the FVs group (0% versus 12.5%, p = 0.040). Among the 171 patients with a mean dwell time of 22.1 days, the IJV group had a higher likelihood of tilt angle over 10° than the FVs group (10.3% versus 2.3%, p = 0.080). [Objective] The variation rule of water transparency and the driving factors of its spatial differentiation were analyzed in order to provide a scientific basis for the management of lake reservoirs and the restoration of lake reservoir ecosystems. [Methods] A remote sensing inversion model of water transparency in Pingzhai reservoir was constructed based on sentinel-2 MSI satellite images and quasi-synchronous measured transparency data collected on May 18, August 26, and November 14, 2020. We quantitatively analyzed the drivers affecting spatial differentiation of water transparency using the GeoDetector package in R. [Results] ① The water transparency of Pingzhai reservoir was most sensitive to the B3 band of Sentinel-2 MSI, and the transparency inversion model constructed by using the band combination B3×B4 as the most sensitive factor had high accuracy (R2=0.81, RMSE=0.11 m, MRE=16.91%). ② The water transparency of Pingzhai reservoir showed a spatial distribution trend of high in the central reservoir area, low in the upstream region of each tributary, and low on both sides of the near water body. Water transparency was greatest in November followed by August and then May. [Conclusion] The contents of total suspended solids, chlorophyll a, and total organic carbon were the main factors affecting the spatial differentiation of water transparency in Pingzhai reservoir. Total phosphorus, total nitrogen, water temperature, and wind speed affected the spatial distribution of water transparency by affecting the content of total suspended solids, chlorophyll a, and total organic carbon in the water.

The severity of Verticillium wilt on cotton caused by defoliating strains of Verticillium dahliae has gradually increased and threatens production worldwide. Identification of the molecular components of leaf defoliation may increase cotton tolerance to V. dahliae. Ethylene, a major player in plant physiological processes, is often associated with senescence and defoliation of plants. We investigated the cotton–V. dahliae interaction with a focus on the role of ethylene in defoliation and defense against V. dahliae. Cotton plants inoculated with V. dahliae isolate V991, a defoliating strain, accumulated more ethylene and showed increased disease symptoms than those inoculated with a non-defoliating strain. In cotton with a transiently silenced ethylene synthesis gene (GhACOs) and signaling gene (GhEINs) during cotton–V. dahliae interaction, ethylene produced was derived from cotton and more ethylene increased cotton susceptibility and defoliation rate. Overexpression of AtCTR1, a negative regulator in ethylene signaling, in cotton reduced sensitivity to ethylene and increased plant resistance to V. dahliae. Collectively, the results indicated precise regulation of ethylene synthesis or signaling pathways improve cotton resistant to Verticillium wilt.

Abstract Background The transition from fertilized egg to embryo in chicken requires activation of hundreds of genes that were mostly inactivated before fertilization, which is accompanied with various biological processes. Undoubtedly, transcription factors (TFs) play important roles in regulating the changes in gene expression pattern observed at early development. However, the contribution of TFs during early embryo development of chicken still remains largely unknown that need to be investigated. Therefore, an understanding of the development of vertebrates would be greatly facilitated by study of the dynamic changes in transcription factors during early chicken embryo. Cold seeps are highly productive chemosynthetic ecosystems in the deep-sea environment. Although microbial communities affected by methane seepage have been extensively studied in sediments and seawater, there is a lack of investigation of prokaryotic communities at the surface sediments and bottom seawater. We revealed the effect of methane seepage on co-occurrence networks and ecological niches of prokaryotic communities at the surface sediments and bottom seawater in the Haima cold seep. The results showed that methane seepage could cause the migration of Mn and Ba from the surface sediments to the overlying seawater, altering the elemental distribution at seepage sites (IS) compared with non-seepage sites (NS). Principal component analysis (PCA) showed that methane seepage led to closer distances of bacterial communities between surface sediments and bottom seawater. Co-occurrence networks indicated that methane seepage led to more complex interconnections at the surface sediments and bottom seawater. In summary, methane seepage caused bacterial communities in the surface sediments and bottom seawater to become more abundant and structurally complex. This study provides a comprehensive comparison of microbial profiles at the surface sediments and bottom seawater of cold seeps in the South China Sea (SCS), illustrating the impact of seepage on bacterial community dynamics.

ABSTRACT: Novel Duck Reovirus (NDRV) that has been found throughout the world in waterfowl, and it has been extensively described. Here, we report the complete genome sequence of a NDRV strain isolated in China called NDRV YF10. This strain was collected from 87 samples with infected ducks in South Coastal Area. The NDRV genome consists of 23,419 bp. With the assistance of computer analysis, the promoter and terminator of each gene segment and 10 viral genes segments were identified, which encode polypeptides ranging from 98 to 1,294 amino acids. All gene fragments of this virus strain were determined and compared to previously reported strains, revealing genetic variation with similarity rates ranging from 96 to 99% for each gene segment. Each gene segment formed 2 host-associated groups, the waterfowl-derived reovirus and the avian-derived reovirus, except for the S1 gene segment, which was closely related to ARV evolution and formed a host-independent subcluster. This difference may be due to Avian Reovirus (ARV) evolving in a host-dependent manner. In order to evaluate the pathogenicity of YF10, a novel isolated strain of NDRV was tested in 2 types of ducks. It was observed that the YF10 isolated strain exhibits varying degrees of virulence, highlighting the potential risk posed to different types of ducks. In conclusion, our findings emphasize the importance of epidemiology studies, molecular characterization, and prevention of NDRV in waterfowl.

ABSTRACT Benefiting from the rapid developments and wide applications of high-throughput sequencing, great advancements have been made in investigating microbiota, which are highly diverse and play key roles in both element cycling and the energy flow of ecosystems. There have been inherent limitations of amplicon sequencing that could introduce uncertainty and raise concerns about the accuracy and reproducibility of this technology. However, studies focusing on the reproducibility of amplicon sequencing are limited, especially in characterizing microbial communities in deep-sea sediments. To evaluate reproducibility, 118 deep-sea sediment samples were used for 16S rRNA gene sequencing in technical replicates (repeated measurements of the same sample) that demonstrate the variability of amplicon sequencing. The average occurrence-based overlaps were 35.98% and 27.02% between two and among three technical replicates, respectively, whereas their abundance-based overlaps reached 84.88% and 83.16%, respectively. Although variations of alpha and beta diversity indices were found between/among technical replicates, alpha diversity indices were similar across samples, and the average beta diversity indices were much smaller for technical replicates than among samples. Moreover, clustering methods (i.e., operational taxonomic units [OTUs] and amplicon sequence variants [ASVs]) were shown to have little impact on the alpha and beta diversity patterns of microbial communities. Taken together, although there are variations between/among technical replicates, amplicon sequencing is still a powerful tool with which to reveal diversity patterns of microbiota in deep-sea sediments. IMPORTANCE The reproducibility of amplicon sequencing is vital for whether the diversities of microbial communities could be accurately estimated. Thus, reproducibility influences the drawing of sound ecological conclusions. Nevertheless, few studies have focused on the reproducibility of microbial communities that are characterized by amplicon sequencing, and studies focusing on microbiota in deep-sea sediments have been especially lacking. In this study, we evaluated the reproducibility of amplicon sequencing targeting microbiota in deep-sea sediments of cold seep. Our results revealed that there were variations between/among technical replicates and that amplicon sequencing was still a powerful tool with which to characterize the diversities of microbial communities in deep-sea sediments. This study provides valuable guidelines for the reproducibility evaluation of future work in experimental design and interpretation.

Background: Guidewire fracture and dislodgment are very rare but can cause serious complications.Method: A case was reported where part of the guidewire was blocked to the posterior tibial. We attempted to remove it using a Solitaire AB Neurovascular Remodeling Device.Result: A Solitaire AB Neurovascular Remodeling Device was successfully used to remove the fracture and dislodgment of a guidewire. There were no complications during the removal of the guidewire.Conclusion: A Solitaire AB can be used to remove a fractured guidewire, which is a simple and effective method to reduce complications. The Solitaire stent gradually crimped and tightened to form a dense inverted conical meshwork, similar to the “trans embolic protection device.” We can use minimally invasive methods to remove a broken and displaced guidewire within the vascular lumen.

To explore the performance differences among offspring obtained by the intra- and inter-strain crosses, strain A with outstanding WSSV resistance and strain B with outstanding growth of Litopenaeus vannamei were used to construct four experimental groups in a complete diallel cross. 27 growth-related phenotypic traits and WSSV resistance represented by the surviving hours post-infection were obtained via the growth and the virus challenge test. Mixed linear model along with physical pedigree were utilized to calculate the least squares means, correlations, heritability and heterosis. The results showed that crossbreeding can realize character complementation of advantage strain. The performance of WSSV resistance was ranked AA> BA> AB> BB, and the reciprocal crosses performed significantly better than intra-strain group BB. Most growth-related traits in two reciprocal cross groups were also significantly better than those in AA group. Comparisons between the two reciprocal crosses showed that BA cross was better than AB in terms of WSSV resistance and growth. Heterosis of WSSV resistance in BA cross was 5.74∼10.18% greater than that in AB, meanwhile the absolute weight gain rate of BA cross was 4.83% higher than that of AB. The heritability estimates of body length and body weight were 0.0278 and 0.339, both of which reached medium level. The genetic and phenotypic correlations among growth-related traits were from 0.36 to 0.99 and 0.32 to 0.99, respectively. In term of WSSV resistance, the heritability estimate was 0.135 ± 0.082. The genetic and phenotypic correlations between WSSV resistance and growth-related traits were from − 0.034 to − 0.573 and − 0.001 to − 0.202, respectively. The results showed that hybrid offspring obtained by crossing female parents with outstanding WSSV resistance and male parents with outstanding growth culminated in a superior heterosis in both traits. This study provides basic reference data for L. vannamei breeding of new strains that are disease resistant and fast-growing.
