Epidemiological studies indicate that the consumption of caffeine, the most commonly ingested psychoactive substance found in coffee, tea or soft drinks, reduces the risk of developing Alzheimer’s disease (AD). Previous treatment studies with transgenic AD mouse models reported a reduced amyloid plaque load and an amelioration of behavioral deficits. It has been further shown that moderate doses of caffeine have the potential to attenuate the health burden in preclinical mouse models of a variety of brain disorders (reviewed in Cunha in J Neurochem 139:1019–1055, 2016). In the current study, we assessed whether long-term caffeine consumption affected hippocampal neuron loss and associated behavioral deficits in the Tg4-42 mouse model of AD. Treatment over a 4-month period reduced hippocampal neuron loss, rescued learning and memory deficits, and ameliorated impaired neurogenesis. Neuron-specific RNA sequencing analysis in the hippocampus revealed an altered expression profile distinguished by the up-regulation of genes linked to synaptic function and processes, and to neural progenitor proliferation. Treatment of 5xFAD mice, which develop prominent amyloid pathology, with the same paradigm also rescued behavioral deficits but did not affect extracellular amyloid-β (Aβ) levels or amyloid precursor protein (APP) processing. These findings challenge previous assumptions that caffeine is anti-amyloidogenic and indicate that the promotion of neurogenesis might play a role in its beneficial effects. Background Caffeine is a widely used stimulant with potentially beneficial effects on cognition as well as vasoconstrictive properties. In functional magnetic imaging research, caffeine has gained attention as a potential enhancer of the blood oxygenation level-dependent (BOLD) response. In order to clarify changes of oxy- and deoxyhemoglobin (HbO and HbR) induced by caffeine during a cognitive task, we investigated a working memory (WM) paradigm (visual 2-back) using near-infrared spectroscopy (NIRS). Results Behaviorally, caffeine had no effect on the WM performance but influenced reaction times in the 0-back condition. NIRS data demonstrate caffeine-dependent alterations of the course of the hemodynamic response. The intake of 200 mg caffeine caused a significant decrease of the HbO response between 20 and 40 s after the onset of a 2-back task in the bilateral inferior frontal cortex (IFC). In parallel, the HbR response of the left IFC was significantly increased due to caffeine intake. Conclusions In line with previous results, we did not detect an effect of caffeine on most aspects of behavior. Effects of caffeine on brain vasculature were detected as general reduction of HbO. Neuronal effects of caffeine are reflected in an increased concentration of HbR in the left hemisphere when performing a verbal memory task and suggest influences on metabolism.

Spontaneous calcium oscillations in mushroom bodies of late stage pupal and adult Drosophila brains have been implicated in memory consolidation during olfactory associative learning. This study explores the cellular mechanisms regulating calcium dynamics in Kenyon cells, principal neurons in mushroom bodies. Fura-2 imaging shows that Kenyon cells cultured from late stage Drosophila pupae generate spontaneous calcium transients in a cell autonomous fashion, at a frequency similar to calcium oscillations in vivo (10-20/h). The expression of calcium transients is up regulated during pupal development. Although the ability to generate transients is a property intrinsic to Kenyon cells, transients can be modulated by bath application of nicotine and GABA. Calcium transients are blocked, and baseline calcium levels reduced, by removal of external calcium, addition of cobalt, or addition of Plectreurys toxin (PLTX), an insect-specific calcium channel antagonist. Transients do not require calcium release from intracellular stores. Whole cell recordings reveal that the majority of voltage-gated calcium channels in Kenyon cells are PLTX-sensitive. Together these data show that influx of calcium through PLTX-sensitive voltage-gated calcium channels mediates spontaneous calcium transients and regulates basal calcium levels in cultured Kenyon cells. The data also suggest that these calcium transients represent cellular events underlying calcium oscillations in the intact mushroom bodies. However, spontaneous calcium transients are not unique to Kenyon cells as they are present in approximately 60% of all cultured central brain neurons. This suggests the calcium transients play a more general role in maturation or function of adult brain neurons.
