Abstract Background Proteases catalyze the hydrolysis of peptide bonds of proteins, thereby improving dietary protein digestibility, nutrient availability, as well as flavor and texture of fermented food and feed products. The lactobacilli Lactiplantibacillus plantarum (formerly Lactobacillus plantarum) and Pediococcus acidilactici are widely used in food and feed fermentations due to their broad metabolic capabilities and safe use. However, extracellular protease activity in these two species is low. Here, we optimized protease expression and secretion in L. plantarum and P. acidilactici via a genetic engineering strategy. Results To this end, we first developed a versatile and stable plasmid, pUC256E, which can propagate in both L. plantarum and P. acidilactici. We then confirmed expression and secretion of protease PepG1 as a functional enzyme in both strains with the aid of the previously described L. plantarum-derived signal peptide LP_0373. To further increase secretion of PepG1, we carried out a genome-wide experimental screening of signal peptide functionality. A total of 155 predicted signal peptides originating from L. plantarum and 110 predicted signal peptides from P. acidilactici were expressed and screened for extracellular proteolytic activity in the two different strains, respectively. We identified 12 L. plantarum signal peptides and eight P. acidilactici signal peptides that resulted in improved yield of secreted PepG1. No significant correlation was found between signal peptide sequence properties and its performance with PepG1. Conclusion The vector developed here provides a powerful tool for rapid experimental screening of signal peptides in both L. plantarum and P. acidilactici. Moreover, the set of novel signal peptides identified was widely distributed across strains of the same species and even across some closely related species. This indicates their potential applicability also for the secretion of other proteins of interest in other L. plantarum or P. acidilactici host strains. Our findings demonstrate that screening a library of homologous signal peptides is an attractive strategy to identify the optimal signal peptide for the target protein, resulting in improved protein export.

The development of new food preservatives is essential to prevent foodborne outbreaks or food spoilage due to microbial growth, enzymatic activity or oxidation. Furthermore, new compounds that substitute the commonly used synthetic food preservatives are needed to stifle the rising problem of microbial resistance. In this scenario, we report herein, as far as we know, for the first time the use of the zein protein as a gating moiety and its application for the controlled release of essential oil components (EOCs). The design of microdevices consist of mesoporous silica particles loaded with essential oils components (thymol, carvacrol and cinnamaldehyde) and functionalized with the zein (prolamin) protein found in corn as a molecular gate. The zein protein grafted on the synthesized microdevices is degraded by the proteolytic action of bacterial enzymatic secretions with the consequent release of the loaded essential oil components efficiently inhibiting bacterial growth. The results allow us to conclude that the new microdevice presented here loaded with the essential oil component cinnamaldehyde improved the antimicrobial properties of the free compound by decreasing volatility and increasing local concentration.

Aspergillus fumigatus is the most common mold species to cause disease in immunocompromised patients. Infection usually begins when its spores (conidia) are inhaled into the airways, where they germinate, forming hyphae that penetrate and destroy the lungs and disseminate to other organs, leading to high mortality. The ability of hyphae to penetrate the pulmonary epithelium is a key step in the infectious process. A. fumigatus produces extracellular proteases that are thought to enhance penetration by degrading host structural barriers. This study explores the role of the A. fumigatus transcription factor XprG in controlling secreted proteolytic activity and fungal virulence. We deleted xprG, alone and in combination with prtT, a transcription factor previously shown to regulate extracellular proteolysis. xprG deletion resulted in abnormal conidiogenesis and formation of lighter colored, more fragile conidia and a moderate reduction in the ability of culture filtrates (CFs) to degrade substrate proteins. Deletion of both xprG and prtT resulted in an additive reduction, generating a mutant strain producing CF with almost no ability to degrade substrate proteins. Detailed proteomic analysis identified numerous secreted proteases regulated by XprG and PrtT, alone and in combination. Interestingly, proteomics also identified reduced levels of secreted cell wall modifying enzymes (glucanases, chitinases) and allergens following deletion of these genes, suggesting they target additional cellular processes. Surprisingly, despite the major alteration in the secretome of the xprG/prtT null mutant, including two to fivefold reductions in the level of 24 proteases, 18 glucanases, 6 chitinases, and 19 allergens, it retained wild-type virulence in murine systemic and pulmonary models of infection. This study highlights the extreme adaptability of A. fumigatus during infection based on extensive gene redundancy.

Protease secretion by Giardia duodenalis trophozoites upon interaction with epithelial cells and its association with the parasite adhesion was studied in co-cultures of parasites with IEC6 epithelial cell monolayers in the presence or absence of protease inhibitors. Proteolytic activity in supernatants from trophozoites was enhanced when they were co-cultured with IEC6 cells. This activity was strongly inhibited by pre-incubation of live trophozoites with E-64 and TPCK and a concomitant inhibition of parasite adhesion to IEC6 cells was observed. These data suggest that trophozoites secrete cysteine-type proteases that play a role in the adhesion of G. duodenalis to epithelial cells.

The aim of this study was to test the ovicidal effect ofMetarhizium brunneumonAscaridia gallieggs and the protease activity ofM. brunneumgrown on standard media enriched withA. gallieggs.Ascaridia gallieggs were transferred to water agar plates and the fungus,M. brunneum, was added as a spore suspension. The viability of the eggs was assessed at 7, 14, 21 and 28 days post-inoculation.M. brunneumwas grown on three types of growth media: SDA, SDA + chitin and SDA +A. gallieggs. Crude extract of the fungus was produced by mincing and centrifuging biomass taken from the growth plate. The supernatant was added to Azurine cross-linked-casein plates to test for protease activity.M. brunneumshowed a significant impact on the viability ofA. gallieggs, decreasing the percentage of viable eggs to 5.1%, indicating that it is a potential candidate for biological control of this nematode. Crude extract ofM. brunneumgrown on SDA +A. gallieggs showed a significantly higher protease activity than the crude extract ofM. brunneumgrown on SDA alone andM. brunneumgrown on SDA + chitin, indicating a possible ovicidal effect of these proteases. Some characteristics of root-secreted proteases were studied. We measured their molecular weights and mechanism of BSA digestion in comparison to endogenous root proteases. We related these studies to culture medium N composition. The seedlings of Allium porrum L. (cv. Bartek) were cultivated on MS medium, MS without inorganic nitrogen (MS-IN), and MS without IN, but with 0.1% casein (MS-IN + 0.1% casein). Electrophoretic study showed that root-secreted proteases had one isoform with a mol wt of 45 kD, regardless of medium N composition. Difference in molecular weights of root-secreted proteases and endogenous root proteases active under used conditions (>66 kD) provide us another strong evidence that root-secreted proteases were not just leaking from the roots, but they were secreted. Proteases exuded by roots degraded BSA in a similar way as endogenous proteases, with only one SDS-PAGE-detectable product of degradation. Abstract: In the pathogenic yeast Candida albicans, nitrogen availability regulates phenotypes that contribute to the virulence of the fungus, including filamentous growth and protease secretion. Under limiting nitrogen conditions, the ammonium permease Mep2 induces the switch from yeast to filamentous growth. Mep2 is a cytoplasmic membrane protein that mediates uptake of the preferred nitrogen source ammonium. It contains a signaling domain in its C-terminal cytoplasmic tail that induces morphogenesis in response to ammonium availability, presumably by activating the cAMP-PKA pathway and the Cph1-dependent MAP kinase pathway. MEP2 expression itself is regulated by the GATA transcription factors Gat1 and Gln3. These central regulators also control expression of the secreted aspartic protease Sap2, which is induced when proteins are the only available nitrogen source. Under these conditions, Gat1 and Gln3 upregulate the expression of STP1, which encodes a proteolytically activated transcription factor that in turn mediates the expression of SAP2 and several oligopeptide transporters required for growth on proteins. In this way, C. albicans integrates the expression of different virulence-associated phenotypes into the regulatory network controlling nitrogen metabolism. [Copyright &y& Elsevier]

Eight strains of Cryptococcus neoformans var. neoformans isolated from AIDS patients in the Infectious Disease Institute, University of Turin, Italy, were examined for growth and extracellular proteolytic activity in culture with solid and liquid media. All of the strains grew well on Yeast Carbon Base (YCB) agar medium supplemented with both 0.1% (w/v) bovine serum albumin (BSA) and 0.01% (w/v) polypeptone (Pp), and produced a clear proteolytic zone around their colonies, whereas they exhibited less growth and proteolytic activity on YCB medium supplemented with BSA alone. Strain #8 with a strong proteolytic activity was cultured in three different liquid media. Its growth was limited in YCB medium supplemented with 0.1% BSA, but was moderate in that with 0.01% Pp. Enhanced growth was supported by the addition of both BSA and Pp to the YCB medium. The relative value of the final cellular yields obtained with the above YCB-0.1% BSA, YCB-0.01% Pp and YCB-0.1% BSA-0.01% Pp media was approximately 1:10:20. In the culture with YCB medium containing both BSA and Pp, a rapid decrease in the amount of BSA was demonstrated by a spectrophotometric assay and gel electrophoresis of the culture supernatant after the log-to-stationary phase. The proteolytic activity in the culture supernatant became detectable after the log phase when tested with skim milk agarose plates. These results allowed us to conclude that Cr. neoformans var. neoformans is able to secrete protease and to utilize protein as a source of nitrogen. [ABSTRACT FROM AUTHOR]
