Background. Premenstrual syndrome (PMS) characterized by cyclic symptoms during the luteal phase of the menstrual cycle, presents an uncertain etiology in adolescents involving hormonal fluctuations and serotonin-related neurotransmitters with a limited existing literature on the impact of oxidative stress. This study aimed to explore the potential association between PMS and oxidative stress in adolescents. Methods. In a cross-sectional study conducted at a university hospital, involving 45 adolescent girls aged 12 to 18, participants were categorized based on the presence or absence of PMS using the cut-off point of 110 on the PMS Scale developed by Gençdoğan. Oxidative stress was assessed through dynamic thiol-disulfide homeostasis. The shift from the balance towards disulfide form is associated with oxidative stress, whereas towards thiol it shows a greater antioxidant capacity. Results. Thirty out of the forty-five participants were found to have PMS with a mean age of 15.5 years. The PMS group demonstrated a significant increase in antioxidant markers, specifically elevated native (631.6±57.55 vs 598.2±41.08, p=0.048) and total thiol levels (675.15±3.4 vs 639.3±44.9, p=0.031). Despite a significant increase in thiol, thiol to disulfide ratio was not found to be significant (p=0.849). Conclusion. Contradictory to other studies in adults, we have demonstrated an increase in the antioxidant markers in adolescents with PMS. Elevated antioxidant status in adolescents with PMS may be an adaptive response to acute cyclic inflammation in the adolescent period, which might decrease with the progression of age. Hashimoto thyroiditis is a common cause of goiter and acquired hypothyroidism in individuals residing in areas of no iodine deficiency. The fact that the structure of serum albumin exhibits changes in ischemic conditions has paved the way for the discovery of a new serum cardiac ischemia marker, Ischemia Modified Albumin. The other one, thiol/disulphide homeostasis, plays an important part in antioxidative protection, detoxification, cell growth, and apoptosis. In this study, we aimed to investigate both the relationship between Thiol/Disulphide homeostasis and Ischemia Modified Albumin in patients diagnosed with Hashimoto’s Thyroiditis. A total of 70 Hashimoto’’s thyroiditis patients and 50 healthy ones were included in this study. Age, gender, thyroid-stimulating hormone (TSH), anti-thyroid peroxidase (TPO), anti-thyroglobulin (TG) levels were recorded. Ischemia Modified Albumin and thiol-disulphid homeostasis parameters were measured through automated spectrophotometric methods. The ages of individuals included in the study ranged from 35 to 58 years. The native thiol/total thiol were found to be significantly lower in Hashimoto patients when compared to those enrolled in the control group (P < 0.05), whereas the Ischemia Modified Albumin, disulphide, native thiol, total thiol, disulphide/native thiol, and disulphide/total thiol were found to be significantly higher in Hashimoto patients when compared to those in the control (P < 0.05). Increased Ischemia Modified Albumin, native and total thiol, and disulphide levels are related to increased oxidative stress. Although Ischemia Modified Albumin and Thiol-disulphide defense are important oxidative indicators in Hashimoto’s Thyroiditis, many determinants are known to be involved in this process.

METHODS: A total of 146 participants who were admitted to the emergency department and who consisted of 69 patients and 77 healthy volunteers were included in the study. Detailed blood samples were obtained from the patients at the time of admission. Thiol/disulphide levels were examined using a brand-new method developed by Erel and Neselioglu on the patients diagnosed with obstructive jaundice during the admission. Data were evaluated in the computer system. RESULTS: The levels of total thiol (p =0.025) and native thiol (p =0.023) were statistically significantly lower in the patient with obstructive jaundice than in the control group. Background: Alzheimer's disease (AD) is a steadily advancing neurodegenerative condition, the occurrence and prevalence of which are on the rise in various populations. Suspected factors contributing to its development encompass the buildup of amyloid β (Aβ) plaques, the formation of neurofibrillary tangles induced by tau proteins, and heightened oxidative stress. In this study, we aimed to evaluate intra-cellular glutathione status and extracellular thiol-disulphide status in patients with AD., Methods: Adult patients (>60 years old) diagnosed with AD based on DSM-IV diagnostic criteria were included in the study. Patients were divided into 3 groups as mild, moderate and severe according to Mini Mental Status Examination (MMSE) and clinical findings. Extracellular thiol-disulfide and intracellular oxidized-reduced glutathione status parameters for patient and control groups were analyzed before and after reduction procedures by using reaction of thiol groups with DTNB., Results: The reduced forms of both balances (native thiol (NT) and reduced glutathione (GSH)) were significantly lower in the patient group than the control group (p = 0.031 and <0.001, respectively), while oxidized forms (disulphide (SS) and oxidized glutathione (GSSG)) and SS/NT and GSSG/GSH percent ratios were significantly higher (p < 0.05 for all). The disease duration and oxidative stress were significantly higher in the severe group of AD. Background: Urticaria is an unknown, sudden, and itchy skin disease that is recognized with redness, swelling, and is sometimes seen with angioedema. It is classified as acute or chronic, depending on the duration of symptoms. Thiols in plasma are powerful antioxidants that physiologically eliminate free radicals. The mostly and rapidly affected proteins are thiols that contain the sulfhydryl group. In the present study, the thiol/disulfide homeostasis was investigated as a brand new indicator of oxidative stress in patients who had acute urticaria and presented to the emergency department. Objective: In the present study, the thiol/disulfide homeostasis, ischemia-modified albumin (IMA), and and neutrophil lymphocyte ratio (N/L ratio) were investigated in the etiopathogenesis of acute urticaria. Material and Method: A total of 37 patients and 40 healthy volunteers were included in the study. Background: Thiol/disulphide homeostasis (TDH) has a critical role in many cellular activities such as antioxidant protection. Alterations of oxidative stress in the transition period play an important role in development of some diseases and disorders in dairy cows., Objectives: The purpose of this study is to assess the total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), total thiol, native thiol, disulphide and lipid profile in Simmental and Montofon dairy cows (aged 2-3) before and after calving., Methods: Blood samples were collected 233-280 days of pregnancy and the 30 days of post-partum. Serum total thiol, native thiol and disulphide levels were determined as well as TAS, TOS and paraoxonase-1 (PON-1) levels were measured using colorimetric assays. Triglycerides (TG), total cholesterol, HDL cholesterol and LDL cholesterol levels were measured with an automatic analyser., Results: Total thiol (p = 0.038) and disulphide (p = 0.015) levels were higher after calving compare to pregnancy in Montofon. TAS was found lower (p < 0.001), and OSI was higher in both breeds (Montofon p = 0.012, Simmental p = 0.028) after calving than in pregnancy. When compared between pregnancy and after calving levels in the same breed, HDL was found to be higher after calving (p < 0.001) and TG was lower after calving (p = 0.020) in Montofon. PON (p = 0.090), HDL (p < 0.001) and cholesterol levels were found higher (p < 0.001) and TG level was lower (p < 0.001) after calving in Simmental., Conclusions: According to our results, we observed different responses between two breeds before and after calving. Thiol-disulphide homeostasis (TDH) is a new parameter indicating oxidative stress that plays a role in the pathogenesis of various clinical disorders. Our study planned to investigate TDH in COVID-19 patients. Age and gender-matched healthy subjects (n = 70) and COVID-19 patients (n = 144) were included in the study. In addition to the routine laboratory parameters of the groups, their native thiol (NT), total thiol (TT) and disulphide levels were measured. Primarily, we compared COVID-19 patients to the healthy control group for inflammatory parameters, NT, TT and disulphide levels. Then, COVID-19 patients were divided into two groups according to the severity of the disease as mild to moderate and severe COVID-19, and the three groups were compared with each other. Predictive value of thiol parameters in the diagnosis of COVID-19 and in the determining its severity, and its correlation with presence and duration of symptoms were investigated. Severe COVID-19 patients had lower NT and TT levels compared with healthy controls and mild to moderate patients (P < 0.001 for both). The results of ROC analysis show that the greatest AUC was IL-6 and NT (AUC = 0.97, AUC = 0.96, respectively) between control and COVID-19 patients, while it was CRP and NT (AUC = 0.85, AUC = 0.83) between mild to moderate and severe patients. A negative correlation was found between duration of symptoms of dyspnoea, cough, fever, and sore throat and NT (r = −0.45, P = 0.017, r = −0.418, P < 0.001, r = −0.131, P = 0.084, r = −0.452, P = 0.040, respectively). NT and TT levels have a strong predictive value in the diagnosis of COVID-19 and in determining disease severity. Our results support that changing TDH parameters appears to have an important role in disease pathogenesis and it can be used in clinical management of patients. Objectives: In infectious diseases, various inflammatory cells are active and reactive oxygen species are produced to fight against intra cellular and extra cellular microorganisms. This leads to an increase in potential antioxidant capacity and free radical production. Thiol is an important antioxidant. Thiols enter oxidation reactions with oxidative molecules and it form disulphide bonds. The aim of this study was to evaluate the dynamic thiol/disulphide homeostasis in patients with surgical site infection (SSI). Methods: Sixty-eight patients with SSI and 66 healthy persons (control group) were included in this study. Complete blood count, biochemistry, erythrocyte sedimentation rate, C-reactive protein values and thiol levels were studied in blood of the patients at 0th day and 10th day. The blood thiol disulphide homeostasis was analysed using a new automated method developed by Erel and Neselioglu. Results: Native thiol, total thiol, albumin levels and native thiol/total thiol ratio were found significantly lower in the blood samples at day 0th compared to 10th day. There was a statistically significant difference between the patient group and the control group in IMA, native thiol, total thiol, albumin, disulphide levels, disulphide/native thiol, disulphide/total thiol and native thiol/total thiol ratios Conclusions: The thiol-disulphide balance is impaired in SSI. The elevated disulphide/total thiol ratio and disulphide/native thiol ratio, and reduced native thiol/total thiol ratio indicate increased oxidation in SSI. The pathogenesis chronic urticaria (CU) hasn't been fully understood. In recent years, it has been shown that thiol-disulphide homoeostasis, as an antioxidant system, plays important roles in both healthy individuals and various diseases. In different ischaemia-reperfusion states, high oxidative stress causes ischaemia-modified albumin (IMA) generation. To investigate thiol/disulphide balance and IMA level in children with CU and their association with disease severity. Thirty children with CU and 20 healthy children as controls, aged 1–18 years, were included in this cross-sectional study. In all subjects, total thiol, native thiol, disulphide levels and IMA levels were measured in plasma by spectrophotometry. Disulphide/native thiol, disulphide/total thiol and native thiol/total thiol ratios were calculated. The disease severity was rated by Urticaria Activity Score (UAS). In the children with CU, the levels of native thiol (375.56 ± 56.22 μmol/L) and total thiol (415.69 ± 54.75 μmol/L) were significantly lower than the control group (475.20 ± 71.87 and 511.20 ± 73.73 μmol/L, respectively) (p = 0.000, p = 0.000). The ratio of native/total thiol * 100 was lower in patients than the control group (p = 0.002). IMA was significantly higher in the patient group than control group (p = 0.000). No significant correlation was found between UAS and thiol/disulphide homoeostasis (p > 0.05). The disulphide levels, disulphide/native thiol and disulphide/total thiol levels were found to be higher in patients with positive family history for autoimmune disorders than those without (p < 0.05). One of the most important causes of varicocele‐related infertility is oxidative stress (OS). One of the markers considered as an indicator of OS is thiol–disulphide homeostasis (TDH). Based on the hypothesis that OS should decrease after varicocelectomy in the light of this information, in our current study, we investigated the relationship between TDH levels and sperm parameters. The data of 56 infertile varicocele men were prospectively analysed. The post‐operative total and native thiol levels were significantly higher than those pre‐operative total and native thiol levels (477.7 & 436.7 nmol/L, 417.6 & 372.1 nmol/L). Positive correlation was found between total thiol change and change in semen volume (ρ:.277, p:.039), ratio of spermatozoa with normal morphology (ρ:.342, p:.01), progressive (ρ:.334, p:.012) and nonprogressive motility (ρ:.385, p:.003). Positive correlation was also found between native thiol change and semen volume (ρ:.349, p:.008), ratio of spermatozoa with normal morphology (ρ:.362, p:.006), progressive (ρ:.297, p:.026) and nonprogressive motility (ρ:.368, p:.005). Change in the level of TDH was found as positively correlated with progressive and nonprogressive motility change. Objective: The aim of this study was to evaluate the dynamic thiol-disulphide homeostasis as a novel oxidative stress parameter in newly diagnosed and medication-naive general anxiety disorder (GAD) and panic disorder (PD) patients. Method: Medication-naive patients with GAD (n=21) or PD (n=23) and healthy individuals (n=23) were enrolled. Patients were administered a sociodemographic form, Beck Depression Inventory, and Beck Anxiety Inventory. Routine biochemical parameters, native thiol, total thiol, and disulphide parameters were measured in all patients and healthy individuals. Results: The native thiol levels of the GAD and PD groups were found significantly lower than those of the control group (p<0.001 and p=0.03, respectively), whereas the disulphide levels and disulphide/native thiol ratios of both patient groups were significantly higher than that of the control group (p<0.001 and p=0.011, respectively). There was no statistically significant difference between the GAD and PD groups with regard to thiol and disulphide levels. Conclusion: Oxidative stress is considered to be one of the molecular changes underlying the pathogenesis of GAD and PD. This study showed that dynamic thiol-disulphide homeostasis was disturbed due to an increase in oxidants and a decrease in antioxidants in GAD and PD patients, suggesting that antioxidant replacement interventions might be an option in the treatment of these disorders. Objective: To investigate oxidative stress parameters [total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI), paraoxonase (PON), arylesterase (ARE) and thiol-disulphide homeostasis] in patients who were diagnosed as having somatic symptom disorder in accordance with Diagnostic and Statistical Manual of Mental Disorders-5. Methods: The study included 41 medication-free patients with somatic symptom disorder and 47 age, sex, and sociodemographic-matched healthy individuals. The patients were administered the Patient Health Questionnaire-15, Somatic Symptom Amplification Scale, Beck Depression Inventory, and Beck Anxiety Inventory. TOS, TAS, OSI, PON, ARE thiol, disulphide levels, and routine biochemical parameters were compared between the two groups. Results: TOS, OSI, disulphide levels, disulphide/native thiol, and disulphide/total thiol ratios were found significantly higher in the patient group compared with the control group (p < 0.001). There was no significant difference in PON, ARE, and TAS parameters between the two groups (p > 0.05). Conclusion: This study showed that the level of oxidants increased and oxidative balance was impaired in somatic symptom disorder. Oxidative stress may play a role in the aetiopathogenesis of this disorder. This is the first study to report an association between oxidative stress and somatic symptom disorder. Purpose: To examine the thiol-disulphide homeostasis during an optic neuritis episode in patients with multiple sclerosis and the relationship between this homeostasis and P100 wave latency.Materials and Method: Visual evoked potential reviews of multiple sclerosis patients who presented with an optic neuritis episode were conducted and P100 latencies were measured. Peripheral blood samples were collected from all patients. Native thiol and total thiol concentrations were measured with the automated method that was recently developed. Their amount of disulphide bonds, disulphide/native thiol, disulphide/total thiol and native thiol/total thiol ratios were calculated. The relationship between P100 latency and thiol-disulphide homeostasis was investigated.Results: A significant positive correlation was determined between the disulphide/native thiol ratio and both mean P100 latency and maximum P100 latency (p = 0.021, r = 0.136; p = 0.030, r = 0.177, respectively).Discussion: As the balance of the plasma dominated by antioxidants moves towards the oxidant side, in other words as a higher rate of thiol is oxidised from the thiol pool, P100 latency is extended. N-acetylcysteine and alpha lipoic acid as well as thiol supplements can improve the thiol-disulphide balance, reinforce antioxidant defence and it can help in slowing down the demyelinating damage. Background: Gunshot injuries result in serious traumatic tissue damage due to high velocity of the bullet, deep penetration, and ballistic effect. Trauma is known to be related with oxidative stress. Serum thiol levels and disulphide/thiol homeostasis are novel oxidative stress biomarkers. In this study, we aimed to investigate serum thiol levels and disulphide/thiol homeostasis in injury patterns of patients admitted to the emergency department with a gunshot injury.Method: A total of 128 participants were included in the study. The participants were divided into two groups: the patient group (Group 1; n = 73) and healthy controls (Group 2; n = 55). Native thiol, total thiol, disulphide levels, disulphide/native thiol, disulphide/total thiol, and neutrophil-to-lymphocyte ratio (NLR) were measured. The Revised Trauma Scale (RTS) and Glasgow Coma Scale (GCS) scores were calculated.Results: Native thiol, total thiol, and disulphide levels were significantly lower in Group 1 (p < 0.001). Disulphide/native thiol ratio, disulphide/total thiol ratio, and NLR were significantly higher in Group 1, compared to Group 2 (p < 0.05). There was a positive correlation between thiol levels and RTS and GCS scores and NLR. Stepwise linear regression analysis showed that native thiol was an independent indicator of RTS and GCS scores. The receiver operating characteristic curve (ROC) analysis revealed that serum native thiol levels of ≤ 342.9 could predict gunshot injury with a sensitivity of 82% and a specificity of 77% (area under the curve = 0.853; 95% confidence interval 0.783-0.924).Conclusion: Our study results suggest that thiol-disulphide homeostasis is disrupted in patients sustaining gunshot injuries, and thiol levels decrease in correlation with the severity of trauma with a high sensitivity and specificity. As the level of native thiol is an independent predictor of the severity of trauma, reduced thiol levels may be of prognostic value in the early assessment of patients in the emergency room. Aim: This study was conducted to investigate thiol-disulphide homeostasis, a novel oxidative stress marker in primary restless legs syndrome (RLS) patients. Material and Method: Thirty-eight patients and 43 healthy subjects were included in the study. Serum native thiol, total thiol, disulphide and disulphide / native thiol percent ratio levels were measured by using a novel automated method. Diagnosis of RLS was based on the criteria proposed by the International Restless Legs Syndrome Study Group (IRLSSG), and RLS severity was assessed using the IRLSSG Severity Scale. Results: Serum native thiol and total thiol levels were significantly lower (p< 0.001; p= 0.007, respectively) while disulphide and disulphide/native thiol percent ratio levels significantly higher (p= 0.017; p< 0.001, respectively), in patients with RLS compared with control group. Significant correlations were found between the IRLSSG Severity Scale and thiol-disulphide homeostasis parameters (r= -0.56, p< 0.001 for native thiol; r= -0.51, p< 0.001 for total thiol; r= 0.37, p= 0.020 for disulphide and r= 0.60, p< 0.001 for disulphide/native thiol percent ratio). Conclusions: Decreased native thiol, increased disulphide levels and increased disulphide/native thiol percent ratio show that thiol-disulphide balance shifted to the oxidation side. Strong correlations between the IRLSSG Severity Scale and thiol-disulphide homeostasis parameters indicate that homeostasis may have a role in the pathogenesis of the Primary RLS. Objective:: The objective of this study is to examine thiol-disulphide homeostasis in patients with polyneuropathy dominated by diabetic or non-diabetic axonal degeneration.Materials-methods:: Fifty-four patients diagnosed with polyneuropathy dominated by axonal damage and 41 healthy subjects were included in the study. The patients were grouped into two groups according to whether or not they had diabetes. The native thiol and total thiol concentrations were measured with the newly developed automated method.Results:: While there was no significant difference between the patients with diabetic and non-diabetic polyneuropathy in terms of native thiol and total thiol levels (p > 0.05), the native thiol and total thiol levels of the groups with both diabetic polyneuropathy and non-diabetic polyneuropathy were significantly low compared to the control group (p < 0.01). The level of disulphides in the patients with diabetic polyneuropathy was significantly higher than that of the patients with non-diabetic polyneuropathy and the healthy individuals (p < 0.05). The loss in the sural nerve sensory neural action potential amplitude was positively correlated with the decrease in the levels of both native thiol and total thiol (p < 0.05).Discussion:: In our study, we observed that the thiol-disulphide balance was also impaired in patients with non-diabetic polyneuropathy similar to patients with diabetic polyneuropathy, and we therefore considered that impaired the thiol-disulphide homeostasis could be the last common path in patients with polyneuropathy with axonal damage, regardless of the aetiology. Therefore, fortification of thiol deficiency with N-acetyl cysteine or alpha-lipoic acid can fix the thiol-disulphide balance and help decelerate the axonal damage. [ABSTRACT FROM AUTHOR]

Purpose: The purpose of the study was to calculate serum total oxidant status (TOS), total antioxidant status (TAS), and dynamic thiol-disulphide (T-D) homeostasis in patients with age-related macular degeneration (AMD), and compare the results with healthy individuals. Methods: Thirty-three exudative AMD patients and 33 healthy controls were included in this case-control study. Participants' serum TAS and TOS levels were measured. In addition, total thiol (TT), native thiol (NT), and disulphide (DS) concentrations were assessed using a novel automated method of measurement. Results: In comparison with the control group, serum TAS, TT, and NT levels were found to be significantly lower ( p < 0.0001, p = 0.004, p = 0.003, respectively) and TOS levels were detected higher ( p = 0.032) in AMD patients. Serum DS levels were elevated in the AMD patient group, but the difference was not statistically significant ( p = 0.219). DS/TT and DS/NT ratios were significantly higher ( p = 0.012, p = 0.013, respectively) in AMD patients. A positive correlation was found between TT and NT ( p < 0.0001) in AMD group. Conclusions: Serum TOS levels are higher, TAS levels are lower, and the T-D balance is shifted to the DS bond side in AMD patients. These results suggest that increased oxidative stress and decreased antioxidant levels may play a role in AMD progression. Objective: Thiol-disulphide homeostasis (TDH) has a critical role in various clinical disorders. We aimed to assess the association of TDH with acute tonsillopharyngitis (AT) in children. Methods: This study included 94 (73 viral and 21 bacterial) tonsillopharyngitis patients and 88 control children. Their native thiol, total thiol, and disulphide levels were measured. Results: Viral and bacterial tonsillopharyngitis patients had lower native thiol levels compared with healthy children (P < 0.001 andP = 0.008, respectively). Both groups had lower total thiol levels compared with control children (P = 0.002 for viral,P = 0.011 for bacterial). The disulphide levels were lower in bacterial than in viral tonsillopharyngitis patients (P = 0.04), and there was a significant difference between viral tonsillopharyngitis patients and the control group (P < 0.001). The native/total thiol ratio in each patient group was lower than in the control group (P < 0.001 for viral,P = 0.017 for bacterial). The disulphide/native thiol and disulphide/total thiol ratios were significantly higher in viral (P < 0.001 for both) and bacterial tonsillopharyngitis patients (P = 0.017 for both) than in healthy children. In all patients, a correlation was found between the levels of C-reactive protein (CRP) and native thiol (r = −0.211,P = 0.04), CRP and total thiol (r = −0.217,P = 0.036), white blood cell (WBC) and native thiol (r = −0.228,P = 0.002), WBC and total thiol (r = −0.191,P = 0.01), and WBC and disulphide (r = 0.160,P = 0.03). Oxidative stress plays an important role in the development of infertility secondary to varicocele. We aimed to investigate the dynamic thiol–disulphide homeostasis as an oxidative stress marker in the spermatic vein of infertility secondary to varicocele. Sixty‐one patients with varicocele were included in the study. Blood was drawn from the median cubital vein and the testicular venous return side before the spermatic vein was separated during surgery. Total thiol, native thiol, disulphide, ischaemia modified albumin (IMA) and albumin values were measured from both the dilated spermatic vein and the median cubital vein. The disulphide/native thiol, disulphide/total thiol and native thiol/total thiol ratios were determined. The mean age of the patients was 27.0 ± 6.68 (15–50) years. While the albumin, native thiol and total thiol values and the native thiol/total thiol ratio were significantly lower (p = 0.004, p < 0.001, p < 0.001, p < 0.001 respectively), the IMA value and the disulphide/native thiol and disulphide/total thiol ratios were significantly higher (p < 0.001, p < 0.001, p < 0.001 respectively) in the samples taken from spermatic venous blood. Thiol–disulphide balance had deteriorated towards disulphide formation in the spermatic vein compared with the peripheral vein. Introduction: The aim of this study was to investigate the relationship among serum uric acid concentrations and thiol-disulphide homeostasis tests in different groups of the patients. Material and Methods: Patients' uric acid and thiol-disulphide homeostasis data were screened retrospectively between November 2016 and February 2017 using the laboratory information system of Atatürk Training and Research Hospital of Ankara Yildirim Beyazit University Medical Faculty. The 389 patients included in the study were divided into three groups, the uric acid concentration being below the reference limits (45), within the reference limits (256) and above the reference limits (88). The relationship between serum uric acid levels and thiol‐disulphide homeostasis was evaluated in three groups. All data were analyzed using IBM SPSS Statistics 23.0. Results: There was a significant difference between the native, total thiol and disulphide averages of the groups (p<0.001). The disulphide values of the groups were 15.4, 15.8 and 20.7 and the native thiol values of the groups were 353.9, 432.3 and 383.4, respectively. There was a poor positive correlation between uric acid and disulphide values (r=0.281, p<0.05). Discussion: Several studies have reported that uric acid has an antioxidant function at normal concentrations and has a prooxidant function at high concentrations. In this study, it was observed that oxidative stress was increased and thiol-disulphide homeostasis was impaired in the third group. Objective: The aim of this study was to investigate dynamic thiol-disulphide homeostasis in patients with idiopathic Parkinson's disease and to determine its relationship with the clinical stage as assessed by the modified Hoehn and Yahr scale., Design and Methods: Fifty-two patients with Parkinson's disease (PD), diagnosed according to the United Kingdom Brain Bank Criteria for idiopathic PD, and 41 healthy individuals were included in the study. Clinical staging of patients was performed according to the Hoehn and Yahr scale. Peripheral blood samples were taken from all participants, and their native thiol and total thiol concentrations were measured using the newly developed automated method. In addition, their amount of disulphide bonds, disulphide/native thiol, disulphide/total thiol and native thiol/total thiol ratios were calculated., Results: Considering the data obtained from Parkinson's patients and the control group, both native thiol (-SH) and total thiol (-SH+-S-S) levels were found significantly lower in patients with Parkinson's disease. A negative and statistically significant relationship was found between both disease duration and disease stage and native thiol (-SH), total thiol (-SH+-S-S) levels and -SH/(-SH+-S-S-) ratio. A positive and statistically significant relationship was found between both disease duration and stage and -S-S-/-SH and -S-S-/(-SH+-S-S-) ratios., Conclusion: In patients with Parkinson's disease, dynamic thiol-disulphide homeostasis is disrupted, according to disease stage and duration. This balance, easily measured by using the newly developed automated method, can be used in monitoring disease progression. Background: The balance of oxidant and antioxidant status plays a key role in the coronary artery diseases (CAD). Thiol is one of the most important antioxidant barriers in humans, and thiol/disulphide homeostasis is a novel oxidative stress marker., Aim: We aimed to investigate the relation of serum thiol levels and thiol/disulphide homeostasis with the presence and severity of CAD., Methods: A total of 161 patients who underwent coronary angiography owing to stable angina pectoris were consecutively enrolled. They were divided into three groups. Group I - 47 age- and gender-matched subjects with normal coronary angiography (control); group II - 71 newly diagnosed CAD patients with noncritical stenosis; and group III - 43 newly diagnosed CAD patients with critical stenosis. Serum native thiol, total thiol, and disulphide levels were measured, and disulphide/thiol ratios were calculated. Gensini scores were calculated in CAD patients., Results: While the highest thiol levels were found in group I, the lowest one was observed in group III (p < 0.001). Total and native thiol levels were significantly lower in group II than in group I (p < 0.001 for each), but they increased considerably in group II compared with group III (p = 0.031 and p = 0.028, respectively). Disulphide levels decreased in group II and III compared with group I (p < 0.001 for each). No statistically significant changes were observed in disulphide/thiol ratios (p > 0.05). Gensini scores were negatively correlated with total and native thiols, and positively with age and dyslipidaemia. Stepwise linear regression analyses showed that native thiol was an independent predictor in the final model for Gensini score. Receiver operating characteristic curve analysis demonstrated that thiol values of 310.7 or below could predict CAD with 89% sensitivity and 85% specificity (AUC = 0.918; 95% CI 0.870-0.965)., Conclusions: While the disulphide/thiol ratio did not change significantly, decreased native thiol levels were associated with the presence and severity of CAD. This result indicates that the reduction of thiols may be an important factor in the development of CAD.
