Item does not contain fulltext The literature on sustainable management has seen a stark increase in publications in the last few years. A considerable number of papers were written on models or specific human resource practices that aim to provide clarity on what sustainable human resources entail. However, many publications seem to show that sustainable management focuses primarily on employer-driven outcomes and external stakeholders. In this chapter we therefore focus on employees and high-quality employment relationships through the lens of the psychological contract. We propose that by focusing on developing a high-quality psychological contract, companies will select the right contract terms in the form of sustainable human resource practices so as to realize the desired outcomes such as employee commitment and retention. After a short introduction on sustainability, we discuss psychological contracts and zoom in on three specific areas in which companies can make a real difference in generating high-quality relationships, thereby putting in place an employee-centered sustainable human resource management.

6. Case report: leg ulcers resolving after discontinuation of clozapine for schizophrenia

- Abstract
There is a high prevalence of skin conditions and metabolic disturbances in patients with schizophrenia. Skin ulcers, in particular those of the lower extremities, mostly arise from arterial and/or venous pathology. Diabetes is considered a prominent risk factor. Atypical antipsychotics carry an increased risk for cardiometabolic side effects, clozapine being among those with the highest risk. We explore the case of a 57-year-old female patient with refractory schizophrenia who received a combination of clozapine and haloperidol. She soon developed type 2 diabetes mellitus and persistent malleolar ulcerations shortly thereafter. We discontinued clozapine and the ulcerations resolved definitively. We discuss underlying ulcer pathophysiology and review the literature for metabolic and cutaneous side effects of antipsychotics. To our knowledge, this is the first report of a patient developing leg ulcers upon treatment with clozapine., Psicosomàtica y Psiquiatría, Núm. 15 (2020)

Lethal yellowing is a disease that causes severe damages to coconut palms and it has devastated the coconut canopy of several Caribbean islands, and areas of southern Florida. The Fiji dwarf coconut accession has demonstrated tolerance to this disease. Pollen storage and germination provides useful information for conservation strategy and genetic improvement programs. The objective of this study was to evaluate pollen viability of Fiji dwarf coconut by in vitro germination under different nutritional and abiotic conditions. For the first experiment, two liquid culture media were evaluated for in vitro germination of Fiji dwarf coconut pollen; the Lora culture medium and the Armendariz culture medium combined with four temperature levels. In the second experiment, different sucrose concentrations in the Lora culture medium (4, 8, 12 and 16%) were evaluated combined with two incubation times (6 and 24 h) in the presence or absence of light. The highest in vitro germination of pollen grain was obtained in Lora culture medium by 24 h incubation period at 28 ºC. The light condition does not influence the germination process. In vitro germination percentage improved as the sucrose concentration increased according to a linear regression. O estudo foi realizado em duas propriedades situadas nos municípios de Paracatu (MG) e Silvânia (GO), manejadas sob sistema orgânico, que fazem parte do projeto ?Corredores agroecológicos como estratégias para a produção de alimentos e sementes, focados no manejo da agrobiodiversidade e sustentabilidade de pequenas propriedades familiares? entre os anos de 2016 a 2019. Seu objetivo foi fazer o levantamento fitossociológico da composição da vegetação espontânea nos corredores agroecológicos, obtendo dados de frequência, densidade e índice de valor de cobertura das espécies ocorrentes naquela formação, além de realizar estudo das semelhanças e diferenças de comunidades espontâneas entre as áreas avaliadas, através de coeficiente de similaridade. No total, foram identificadas 29 espécies, sendo 10 presentes em ambas as regiões. Asteraceae e Poaceae foram as duas principais famílias, entre as 16 encontradas. O coeficiente de similaridade entre as duas regiões foi de 57%, mostrando similaridade mediana entre a flora nos dois anos estudados. SE hace, además, una descripción de las características morfométricas y fisicoquímicas de los embalses en donde fue encontrada.

Este artigo apresenta alguns aspectos da obra do pesquisador inglês Tony Bennett (1988, 1998, 2008, 2011) sobre as políticas culturais e os circuitos artísticos. O autor adota o conceito de governamentalidade, proposto por Foucault (2016), e sugere que o encontro da estética pós-kantiana com o pensamento liberal, no final do século XVIII, criou condições para que as artes e a cultura passassem a ser vistas como um campo de governo. Essa tendência é visível no desenvolvimento das políticas culturais, especialmente a partir de meados do século XX e, mais recentemente, na difusão do conceito de economia criativa. Em seguida, a proposta é avançar sobre o pensamento de Foucault (2008), especialmente sobre as noções de empreendedorismo e capital humano. A hipótese é que um dos fatores que faz dos circuitos da arte um foco de interesse na sociedade atual é a maneira como os artistas investem a própria vida na criação de suas obras. Com isso, multiplicam-se grupos de interesse, que lutam por melhores condições de trabalho e políticas públicas. Por outro lado, corre-se o risco de que as redes se fechem em seus próprios circuitos, fragmentando o campo cultural. O desafio é fazer com que as artes e a cultura se transformem, de fato, em um bem comum, que faça parte do cotidiano da população.

WOS: 000434263000007, PubMed: 29766772, BACKGROUND Embolic strokes of undetermined source represent 20% of ischemic strokes and are associated with a high rate of recurrence. Anticoagulant treatment with rivaroxaban, an oral factor Xa inhibitor, may result in a lower risk of recurrent stroke than aspirin. METHODS We compared the efficacy and safety of rivaroxaban (at a daily dose of 15 mg) with aspirin (at a daily dose of 100 mg) for the prevention of recurrent stroke in patients with recent ischemic stroke that was presumed to be from cerebral embolism but without arterial stenosis, lacune, or an identified cardioembolic source. The primary efficacy outcome was the first recurrence of ischemic or hemorrhagic stroke or systemic embolism in a time-to-event analysis; the primary safety outcome was the rate of major bleeding. RESULTS A total of 7213 participants were enrolled at 459 sites; 3609 patients were randomly assigned to receive rivaroxaban and 3604 to receive aspirin. Patients had been followed for a median of 11 months when the trial was terminated early because of a lack of benefit with regard to stroke risk and because of bleeding associated with rivaroxaban. The primary efficacy outcome occurred in 172 patients in the rivaroxaban group (annualized rate, 5.1%) and in 160 in the aspirin group (annualized rate, 4.8%) (hazard ratio, 1.07; 95% confidence interval [CI], 0.87 to 1.33; P=0.52). Recurrent ischemic stroke occurred in 158 patients in the rivaroxaban group (annualized rate, 4.7%) and in 156 in the aspirin group (annualized rate, 4.7%). 17. Pyogenic Liver Abscesses Derived From Dental Disease in an Immunocompetent Patient

Introduction: Pyogenic liver abscess of odontogenic origin is extremely rare. The authors report a case of pyogenic liver abscesses with oral origin caused by Prevotella denticola and Streptococcus anginosus, in an immunocompetent adult. Case Report: A 54-year-old healthy female was presented to our hospital with fever, pain in the right hypochondriac region, constipation, anorexia and history of toothache with gingival swelling. Abdominal X-ray presented showed signs of intestinal obstruction and computed tomography suggested two liver abscesses. The larger one, with 113 mm, was drained under percutaneous ultrasound guidance. Abscess culture isolated Prevotella denticola and Streptococcus anginosus. Under antibiotic therapy, the most common causes for liver abscess were excluded. Dental evaluation found multiple septic foci. Root extractions, dental restorations and periodontal treatments were performed. In this case, abscess culture result is highly suggestive of a dental origin, as species isolated are common oral pathogens. Along with the clinical findings, we are reassured about the original source of infection. Conclusion: This case illustrates the underestimated risk associated with untreated dental diseases. We believe oral infectious foci removal is imperative as primary care. Due to the increase of the cryptogenic liver abscess, we find a systematic dental evaluation is required for the etiological diagnosis of liver abscess. info:eu-repo/semantics/publishedVersion

Drought is the most limiting environmental factor to crop productivity and presents a great variability in the degree of tolerance among and within species, among varieties. The aim of this study was to characterize sugarcane accessions regarding tolerance to water stress during in vitro cultivation based on changes in biometric, physiological and biochemical characteristics, within species and among species, to support future breeding programs. Adventitious shoots of five sugarcane accessions: Saccharum robustum, Saccharum spontaneum and Saccharum officinarum species, cultivated in Murashige and Skoog medium supplemented with 2% sucrose and 4 g/l Phytagel were used in five water potentials, 0, -0.3, -0.6, -0.9, -1.2 MPa, induced by mannitol. Survival, length of shoots and roots, number of shoots and roots, biomass, proline content in leaves and activity of antioxidant enzymes were analyzed. There is difference among species, and also, within the same sugarcane species when submitted to in vitro drought stress, and S. officinarum was shown to be the most tolerant. Proline can be used as a biochemical indicator of response to drought in sugarcane accessions and its accumulation was intensified in S. robustum and S. spontaneum accessions. BACKGROUND: Patients with systolic heart failure and anemia have worse symptoms, functional capacity, and outcomes than those without anemia. We evaluated the effects of darbepoetin alfa on clinical outcomes in patients with systolic heart failure and anemia. METHODS: In this randomized, double-blind trial, we assigned 2278 patients with systolic heart failure and mild-to-moderate anemia (hemoglobin level, 9.0 to 12.0 g per deciliter) to receive either darbepoetin alfa (to achieve a hemoglobin target of 13 g per deciliter) or placebo. The primary outcome was a composite of death from any cause or hospitalization for worsening heart failure. RESULTS: The primary outcome occurred in 576 of 1136 patients (50.7%) in the darbepoetin alfa group and 565 of 1142 patients (49.5%) in the placebo group (hazard ratio in the darbepoetin alfa group, 1.01; 95% confidence interval, 0.90 to 1.13; P=0.87). There was no significant between-group difference in any of the secondary outcomes. The neutral effect of darbepoetin alfa was consistent across all prespecified subgroups. Fatal or nonfatal stroke occurred in 42 patients (3.7%) in the darbepoetin alfa group and 31 patients (2.7%) in the placebo group (P=0.23). Thromboembolic adverse events were reported in 153 patients (13.5%) in the darbepoetin alfa group and 114 patients (10.0%) in the placebo group (P=0.01). Cancer-related adverse events were similar in the two study groups. CONCLUSIONS: Treatment with darbepoetin alfa did not improve clinical outcomes in patients with systolic heart failure and mild-to-moderate anemia. A Estratégia Nacional para a Qualidade na Saúde 2015 -2020 visa assegurar que as funções de governação, coordenação e ação local, centradas no doente, estão devidamente alinhadas pelo mesmo diapasão, com respeito pelas dimensões da efetividade, eficiência, acesso, segurança, equidade, adequação, oportunidade, continuidade e respeito pelo cidadão. A pesquisa da qualidade nos serviços de saúde deixou de ser um ato isolado e tornou-se hoje um dado adquirido e categórico. A Sociedade está cada vez mais exigente, no que se refere à qualidade dos serviços prestados, fundamentalmente nas instituições públicas. Esta exigência desenvolveu uma necessidade de criar normas e mecanismos de avaliação e controlo da qualidade nos serviços de saúde. Na última década os meios complementares de diagnóstico e terapêutica transformaram-se em ferramentas essenciais para todos os ramos e especialidades da medicina. O recurso a procedimentos radiológicos para diagnóstico e terapêutica tem determinado um aumento da dose de radiação nos pacientes, tornando-se numa preocupação constante dos utilizadores. Contudo, a sua quantificação e monitorização permite assegurar a segurança dos pacientes, assim como contribuir para a manutenção de serviços de qualidade no SNS. Os meios complementares de diagnóstico e terapêutica transformaram-se em ferramentas essenciais para todos os ramos e especialidades da Medicina. Devido às suas propriedades únicas, a radiação ionizante apresenta múltiplas aplicações terapêuticas. Contudo, pode também originar potenciais danos para os utilizadores e para os pacientes. A quantificação da dose de radiação nos pacientes e o tempo de exposição dos procedimentos são uma preocupação crescente dos utilizadores. Este trabalho tem como objetivo apresentar a importância da Comissão de Proteção contra Radiações (CPCR) no contexto da qualidade do Centro Hospitalar do Porto (CHP). A CPCR detém múltiplas atividades, estando, atualmente, as suas sinergias direcionadas para a normalização do registo de dose no processo clínico dos pacientes e para a notificação, pelos Técnicos de Radiologia, dos eventos radiológicos contemplados nas normas pré--estabelecidas pela Comissão. A atribuição à CPCR da competência de identificação, acompanhamento e avaliação das exposições dos doentes a procedimentos específicos com radiação tem contribuído para reforçar a sensibilização dos profissionais para os valores de dose e os seus efeitos. A implementação dos procedimentos da Comissão permite assegurar um melhor acompanhamento dos doentes em risco, uma maior acuidade na monitorização das doses e uma otimização de protocolos. info:eu-repo/semantics/publishedVersion

Resumo. A geoinformação é essencial para o planejamento e monitoramento das atividades agropecuárias, justificando o emprego de esforços para reuni-la e padronizá-la de acordo com as diretrizes governamentais e possibiltando sua disponibilização à sociedade em geral. O objetivo deste trabalho é apresentar a experiência da Embrapa em construir sua Infraestrutura de Dados Espaciais, denominada GeoInfo, uma iniciativa para organizar, preservar, documentar e ofertar dados geoespaciais abertos produzidos nas pesquisas da empresa, ampliando o potencial de aplicação dessa informação na produção e difusão de conhecimento e inovação. O GeoInfo implementa os padrões da Infraestrutura Nacional de Dados Espaciais e viabiliza a interoperabilidade de dados geoespaciais provenientes de diferentes fontes, inclusive sob o aspecto semântico. Essa iniciativa possibilita a integração das informações geoespaciais produzidas na Embrapa com diversas informações disponíveis em nosso país e permite que a redundância de esforços na obtenção e produção de dados geoespaciais sejam evitados. El objetivo del trabajo fue desarrollar un modelo de simulación de calidad foliar de pasturas, de base morfogenética,para integrar el efecto de la edad y el largo foliar sobre la digestibilidad en relación con el manejode la defoliación. El modelo se construyó y calibró con datos de morfogénesis foliar, fibra detergente neutro(FDN), digestibilidad de la FDN (DFDN) y digestibilidad in vitro de láminas de festuca alta (Festuca arundinaceaSchreb.). Luego se lo validó con datos observados en pasturas manejadas a dos alturas de forrajeremanente con similar producción de biomasa. El modelo predijo adecuadamente la calidad nutritiva de laspasturas de diferente altura de remanente. Las variaciones en calidad durante el rebrote de pasturas vegetativasse debieron principalmente a los cambios de DFDN. Estos resultados indican que la consideración de ladinámica de DFDN en relación con la morfogénesis foliar permitiría mejorar la predicción de la calidad nutritivaen los modelos de pasturas. The study takes an interdisciplinary approach, in that it seeks to demonstrate the close relationship between law and economics to identify the economic and social impacts of environmental crimes and for their valuation degraded property. To support the work, the environment was characterized as well legal, listing up the penalties charged to agents of crimes against him; described is the social function of the sentence and its value to the law; They were listed basic principles of Traditional Theory of Criminal Law and Economic Theory of Crimes and Punishments and analyzed briefly fines charged against environmental crimes agents, with regard to the said correlation. The research was guided by questions such as: how are valued monetized degraded natural resources for penalty purposes for environmental crimes? Environmental crimes, the penalty has maintained its sense of "sensitive and powerful means to quell the despotic spirit that returned to plunge society into chaos"? "As the rules (or feathers) can affect behavior and how they can be modified to generate more beneficial behavior for society?" The conclusion was that there is a positive correlation between the monetary penalty attributed to environmental crimes agents and their damage done. O objetivo deste trabalho ?? verificar a correla????o entre a pena pecuni??ria imputada a agentes de crimes ambientais e o respectivo dano causado, com base na an??lise econ??mica do Direito. This study was developed to evaluate the effect of seasonality on the yield and chemical composition of the essential oil (EO) of Hesperozygis ringens (Benth.) Epling, a native species from the Brazilian Pampa. Leaves were collected from four specimens of a single population in each of the four seasons for a year and were extracted in triplicate by hydro-distillation for 2 hours. The yield of EO (% w/w) was calculated on fresh weight basis (FWB), and the 16 oil samples were analyzed by gas chromatography coupled to mass spectrometry (GC-MS) and gas chromatography with flame ionization detector (GC-FID). Hierarchical Cluster Analysis (HCA) and Principal Component Analysis (PCA) were used as statistical tools to evaluate differences in chemical composition. The highest yields were obtained in autumn, spring and summer (2.32-4.38%), while the lowest yields were detected in winter, ranging from 1.15 to 1.91%. Oxygenated monoterpenoids were the predominant class of chemical constituents in the EO obtained in all seasons, showing the highest contents in autumn and summer, and pulegone was identified as a major compound, whose contents varied between 54.13 and 81.17%. The EO samples were divided into three chemical groups by HCA and PCA and were assigned to the same group, except for the three samples gathered in winter. The results showed a seasonal influence on the yield and chemical composition of the EO. Resumo Este estudo foi desenvolvido a fim de avaliar o efeito da sazonalidade no rendimento e composição química do óleo essencial (OE) de Hesperozygis ringens (Benth.) Epling., uma espécie nativa do Pampa brasileiro. Folhas foram coletadas de quatro indivíduos de uma mesma população, em cada uma das quatro estações de um ano, e foram extraídas em triplicada por hidrodestilação durante 2 horas. O rendimento do OE (% m/m) foi calculado considerando a base fresca (BF) e as 16 amostras de óleo foram analisadas por cromatografia gasosa acoplada à espectrometria de massas (CG-EM) e cromatografia gasosa com detector de ionização de chamas (CG-DIC). Análise Hierárquica de Cluster (AHC) e Análise de Componentes Principais (ACP) foram utilizadas como ferramentas estatísticas para avaliar as diferenças na composição química. Os maiores rendimentos foram obtidos no outono, primavera e verão (2,32-4,38%), enquanto que os menores foram detectados no inverno, variando de 1,15 até 1,91%. Os monoterpenoides oxigenados foram a classe predominante dos constituintes do OE obtido em todas as estações, apresentando os maiores teores no outono e no verão, e a pulegona foi identificada como o constituinte majoritário, cujos teores variaram entre 54,13 e 81,17%. As amostras de OE foram divididas em três grupos químicos por AHC e ACP e foram classificadas no mesmo grupo, com exceção de três amostras coletadas no inverno. Os resultados demonstraram influência sazonal no rendimento e na composição química dos OE.

The productivity of herds may be negatively affected by inbreeding depression, and it is important to know how intense is this effect on the livestock performance. We performed a comprehensive analysis involving five Zebu breeds reared in Brazil to estimate inbreeding depression in productive and reproductive traits. Inbreeding depression was estimated for 13 traits by including the individual inbreeding rate as a linear covariate in the standard genetic evaluation models. For all breeds and for almost all traits (no effect was observed on gestation length), the performance of the animals was compromised by an increase in inbreeding. The average inbreeding depression was B0.222% and B0.859% per 1% of inbreeding for linear regression coefficients scaled on the percentage of mean (bm) and standard deviation (br), respectively. The means for bm (and B) were B0.269% (B1.202%) for weight/growth traits and B0.174% (B0.546%) for reproductive traits. Hence, inbreeding depression is more pronounced in weight/growth traits than in reproductive traits. These findings highlight the need for the management of inbreeding in the respective breeding programmes of the breeds studied here.

- Abstract
In the eastern Pacific, lithogenic input to the ocean responds to variations in the atmospheric and oceanic system and their teleconnections over different timescales. Atmospheric (e.g., wind fields), hydrological (e.g., fresh water plumes) and oceanic (e.g., currents) conditions determine the transport mode and the amount of lithogenic material transported from the continent to the continental shelf. Here, we present the grain size distribution of a composite record of two laminated sediment cores retrieved from the Peruvian continental shelf that record the last 1000 years at a sub-decadal to centennial time-series resolution. We propose novel grain size indicators of wind intensity and fluvial input that allow reconstructing the oceanic atmospheric variability modulated by sub-decadal to centennial changes in climatic conditions. Four grain size modes were identified. Two are linked to aeolian inputs (M3: similar to 54; M4: similar to 91 mu m on average), the third is interpreted as a marker of sediment discharge (M2: similar to 10 mu m on average), and the last is without an associated origin (M1: similar to 3 mu m). The coarsest components (M3 and M4) dominated during the Medieval Climate Anomaly (MCA) and the Current Warm Period (CWP) periods, suggesting that aeolian transport increased as a consequence of surface wind stress intensification. In contrast, M2 displays an opposite behavior, exhibiting an increase in fluvial terrigenous input during the Little Ice Age (LIA) in response to more humid conditions associated with El Nino-like conditions. Comparison with other South American paleoclimate records indicates that the observed changes are driven by interactions between meridional displacement of the Intertropical Convergence Zone (ITCZ), the South Pacific Subtropical High (SPSH) and Walker circulation at decadal and centennial timescales.

Resumo: A suspeição está na base de uma investigação clÃnica completa, sendo de extrema importância a revisão dos casos clÃnicos nos quais não consigamos estabelecer um diagnóstico definitivo. Desta revisão surgem hipóteses diagnósticas que contemplam doenças raras, como a macroglobulinemia de Waldenstrom, doença linfoproliferativa de células tipo B, caracterizada por infiltração medular e produção de IgM monoclonal. Em 1% destas gamapatias são diagnosticadas formas biclonais. Apresentamos o caso clÃnico de um doente com 74 anos, internado em Outubro/04 por colecistite aguda, tendo os exames pré-operatórios revelado derrame pleural bilateral. Durante o internamento foi feita minuciosa investigação etiológica, que foi contudo inconclusiva. Em Maio/05 foi internado, tendo sido submetido a talcagem pleural direita e a investigação clÃnica, também esta inconclusiva. Em Julho/05 foi reinternado num serviço de medicina interna, por anasarca e dor nos pés. Fez-se nova investigação e o diagnóstico final foi de macroglobulinemia de Waldenstrom.Rev Port Pneumol 2008; XIV (6): 869-874 Abstract: Clinical suspicion is the bedrock of a thorough clinical work-up, and a review of the clinical files with no definitive diagnosis is a must. Such a review can uncover rare diagnoses, such as Waldenstrom's macroglobulinaemia, a proliferative B-cell disease characterised by medular infiltration and monoclonocal IgM production. 1% of these are biclonal. We present a case of a 74 year old patient diagnosed in October 2004 with acute cholecystitis. Surgical evaluation revealed bilateral pleural effusion with an inconclusive aetiology. Patient underwent a right pleurodesis in May 2005 and the aetiology remained inconclusive. The Grid Computing paradigm aims to create a ‘virtual’ and powerful single computer with many distributed resources to solve resource intensive problems. The term ‘gridification’ involves the process of transforming a conventional application to run in a Grid environment. In that sense, the more automatic this process is, the easier is for developers with low expertise in parallel and distributed computing to take advantage of these resources. To date, many semiautomatic gridifiers were built to support different gridification approaches and application code structures or anatomies. Furthermore, agricultural simulation applications have a particular common anatomy based on biophysical entities, such as animals, crops, and pastures, which are updated by actions, such as growing animals, growing crops, and growing pastures, along simulation execution. However, this anatomy is not fully supported by any of the existing gridifiers. Thus, this paper presents Agricultural Simulation Applications Gridifier (ASAG), a method for easy gridification of agricultural simulation applications, and its Java implementation, named Java ASAG (JASAG). The main design drivers of JASAG are middleware independence, separation of business logic and Grid behavior, and performance increase. An experimental evaluation showing the feasibility of the gridification method and its implementation is also reported, which resulted in speedups of up to 25 by using a real agricultural simulation application. A total of 7,256 records of animals participating in weight gain trials were used. The (co) variance components were estimated by restricted maximum likelihood, under an animal model. Contemporary group, age and weight as covariate linears were included as fixed effects. Additive genetic effects were the only random effect. The heritability estimates for structure, precocity and muscling were 0.30; 0.37 and 0.32 respectively. The genetic correlations between visual score were 0.76 to 0.95, indicating that the visual scores are controlled by the same set of genes, a fact making individual selection more complex. The visual scores showed similar genetic correlations with daily weight gain, weight at 18 months and scrotal circumference, ranging from 0.45 to 0.50, from 0.80 to 0.83 and from -0.05 to -0.03, respectively, indicating that individual selection for visual scores will result in genetic changes in the same direction in weight at 18 months and, to a lesser extent, in daily weight gain. 48. Transcriptional vs post-transcriptional regulation of the central carbon metabolism of E. coli

- Abstract
Transcriptomics data are currently one of the most available types of large-scale biological data. A large number of methods have been developed to improve constraint-based simulations using these data. We recently performed a systematic comparison of these methods and observed that, at least for central carbon metabolism, there is no significant improvement in the prediction of flux distributions when gene expression data is used. These results are consistent with recent studies, in different organisms, showing that central carbon metabolism is predominantly regulated at post-transcriptional levels. Central carbon metabolism provides the precursors for the production of multiple compounds used in industrial biotechnology. Hence, it is the main target for intervention in most rational strain design strategies. However, its complexity is still not completely understood. In this work, we analyze the role of allosteric regulation, one of the main mechanisms of post-transcriptional regulation, for the control of central carbon metabolism. We extend a model of central carbon metabolism of E. coli with allosteric interactions, revealing a hidden topology in metabolic networks. We use this model to integrate a multi-omic dataset containing transcript, protein, flux and metabolite levels to further dissect the contribution of different types of regulation for metabolic flux control in these central pathways. Situations of predominant allosteric control could be identified, highlighting the importance of this kind of regulation in central carbon metabolism.

Introduction The use of anti-retroviral therapy (ART) has dramatically reduced HIV-1 associated morbidity and mortality. However, HIV-1 infected individuals have increased rates of morbidity and mortality compared to the non-HIV-1 infected population and this appears to be related to end-organ diseases collectively referred to as Serious Non-AIDS Events (SNAEs). Circulating miRNAs are reported as promising biomarkers for a number of human disease conditions including those that constitute SNAEs. Our study sought to investigate the potential of selected miRNAs in predicting mortality in HIV-1 infected ART treated individuals. Materials and Methods A set of miRNAs was chosen based on published associations with human disease conditions that constitute SNAEs. This case: control study compared 126 cases (individuals who died whilst on therapy), and 247 matched controls (individuals who remained alive). Cases and controls were ART treated participants of two pivotal HIV-1 trials. The relative abundance of each miRNA in serum was measured, by RTqPCR. Associations with mortality (all-cause, cardiovascular and malignancy) were assessed by logistic regression analysis. Correlations between miRNAs and CD4+ T cell count, hs-CRP, IL-6 and D-dimer were also assessed. Results None of the selected miRNAs was associated with all-cause, cardiovascular or malignancy mortality. The levels of three miRNAs (miRs -21, -122 and -200a) correlated with IL-6 while miR-21 also correlated with D-dimer. Additionally, the abundance of miRs -31, -150 and -223, correlated with baseline CD4+ T cell count while the same three miRNAs plus miR-145 correlated with nadir CD4+ T cell count. Discussion No associations with mortality were found with any circulating miRNA studied. These results cast doubt onto the effectiveness of circulating miRNA as early predictors of mortality or the major underlying diseases that contribute to mortality in participants treated for HIV-1 infection.

Posteriormente as amostras foram submetidas a tratamento térmico nas temperaturas de 475°C, 600°C e 750°C por 8 horas, seguidas de resfriamento ao ar. As tensões residuais médias foram avaliadas e diferentes perfis de tensões foram construídos. A tensão residual das fases foi determinada pela técnica sen2 Ψ, utilizando um difratômetro com geometria Bragg-Brentano, empregando fonte de radiação CuKα ( = 0,15419 nm) para a fase austenita e radiação CrKα ( = 0,2291 nm) para a fase ferrita. Avaliou-se ainda o teor de ferrita, microdureza e microestrutura do material, os quais foram relacionados aos perfis obtidos. Verificou-se que tanto a temperatura quanto a variação do gás de proteção influenciam no perfil de tensões para as diferentes fases deste aço. Palavras chave: Aço inoxidável duplex UNS S32304.
