Emerging studies demonstrates that mitochondrial G-quadruplex (G4) may be a prospective diagnostic and therapeutic target for cancer. Fluorescent probes targeting mitochondrial G4 are crucial for in-depth research on G4 function and mechanism and developing new diagnostic techniques. Here, we report a benzothiazole derivative named ThT-E, which can be prepared with high yield through only one step of reaction. ThT-E exhibits superior detection sensitivity compared to existing probes, and also demonstrates reliable performance for in situ characterization of mitochondrial G4 in living cells. Interestingly, ThT-E has been used to differentiate tumor cells from normal cells by quantifying the formation of mitochondrial G4s, implicating that ThT-E may have wide applications in tumor diagnosis. • A benzothiazole derivative for monitoring mitochondrial G-quadruplexes. • More pronounced fluorescence enhancement in tumor cells than normal cells. • More G-quadruplex formation in tumor mitochondria. • Mitochondrial G-quadruplex as a potential cancer biomarker. • New probe tool for tumor diagnosis.
