Objective To investigate the integration characteristics of hepatitis B virus (HBV) viral DNA in the host genome of hepatocellular carcinoma HepG2.2.15 cells and HepG2-derived cell line HepAD38 by probe-based capture and high-throughput sequencing technology. Methods After the genomic DNA of HepG2.2.15 and HepAD38 cells were extracted, probe-based capture technique was used to capture the integrated DNA fragments of HBV DNA in the obtained genomic sequences, and the intergrated fragments were further studied with high-throughput sequencing technology. The bioinformatics software such as Trimmatic, BBAP, and BWA (Burrows-Wheeler Aligner) were empolyed for data analysis. Finally, the characteristics of integrated DNA were validated through PCR and cloning and sequencing. Results The probe-based capture and high-throughput sequencing technology detected 12 and 7 integrated DNA fragments in HepG2.2.15 and HepAD38 cells, respectively. These integration events were randomly distributed on chromosomes chr1, chr2, chr7, chr9, chr16, chr17, chr19, and chrX. DPP7, TRIM56 and GPC3 appeared with higher frequencies at the upstream and downstream of the integration sites. Conclusion We successfully establish a method to detect and analyze the integrated DNA fragments of HBV DNA in host genome based on probe-based capture and high-throughput sequencing technology. HBV integration events occur randomly on the chromosomes in HepG2.2.15 and HepAD38 cells, and most of integration sites are in the X gene region of the HBV genome.

The effect of baking degree on the change of seven chlorogenic acid compounds in 10 different origins (origin 1: Yunnan, origin 2: Africa and America) of Cardim coffee. The content of chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, isochlorogenic acid A, isochlorogenic acid B, isochlorogenic acid C and 5-ferulicquinic acid in green coffee and baked coffee were determined by high performance liquid chromategraphy (HPLC). The optimum extraction conditions of seven chlorogenic acid compounds were optimized by response surface methodology. 100 mL 0.1% phosphoric acid solution was extracted in water bath for 30 min, and the calibration curves of the seven chlorogenic acid compounds showed good linearity with correlation coefficients of above 0.999 4 in the range of 2~150 mg/L. Limit of the detection (LOD) and the limit of the quantitative (LOQ) were 0.005~0.5 mg/g and 0.02~2 mg/g respectively. Objective To investigate the molecular mechanism of hepatitis B virus X (HBx) regulating the degradation of SAMHD1 and promoting the proliferation of hepatocellular carcinoma (HCC) cells. Methods The SAMHD1 expression in hepatoma and para-cancerous tissues collected from 15 HCC patients was detected by Western blotting. A stable hepatocellular carcinoma cell line of HepAD38 with SAMHD1 knockout was constructed using CRISP9/Cas9 technology, and the effects of SAMHD1 knockout on the change of proliferation and cell cycle were measured by MTT assay and flow cytometry, respectively, and its effects on related cyclin expression and Akt/p27 pathway were also detected by Western blotting. The impacts of HBx protein overexpression on the mRNA and protein expression of SAMHD1 were subsequently examined. In addition, the molecular mechanism of HBx regulating the degradation of SAMHD1 was further investigated through ubiquitin-proteasome pathway using cycloheximide (CHX) and MG132 treatment along with immunoprecipitation (IP) analysis. Results ① The expression of SAMHD1 was significantly lower in liver cancer tissues than the para-cancerous tissues (P < 0.01).②A stable SAMHD1 knockout cell line HepAD38 (KO-SAMHD1) was successfully constructed, and MTT assay showed that the KO-SAMHD1 group had obviously faster cell proliferation (P < 0.01), and flow cytometry indicated the group had larger amounts of cells arrested in G2/M phase when compared with the control group. ③Western blot analysis indicated that the expression of related cyclins was changed in the KO-SAMHD1 group when compared with the control group, including the expression of p27 protein decreased and p-Akt protein increased. ④In vitro cell experiments showed that HBx overexpression induced decreased expression of SAMHD1 in Huh7.0 cells. ⑤The results of immunoprecipitation suggested that HBx were combined and interacted with SAMHD1. CHX and ubiquitination IP assay indicated an earlier decay time of SAMHD1 protein and enhanced ubiquitination of SAMHD1 induced by HBx. Conclusion HBx interacts with SAMHD1 and degrades it by regulating its ubiquitination, and thereby affects the Akt/p27 pathway to modulate the cell cycle and promote the proliferation of HCC cells.

Alclad and unclad 2E12 aerospace aluminum alloy were treated by sulfuric acid anodic oxidation. The effects of alclad layer and anodizing time on the anodization behaviour and corrosion resistance of anodic oxide layer on 2E12 aluminum alloy were studied. Surface and cross-section morphology of anodic oxide films were observed by scanning electron microscopy. The electrochemical properties of anodic oxide films were analyzed by potentiodynamic polarization curve and electrochemical impedance spectroscopy. The results show that the protective anodic oxide layers are formed on alclad and unclad 2E12 aluminum alloy. The film thickness increases with anodizing time extending. The copper rich second phase particles lead to more cavity defects and even micro cracks on anodic oxide films of unclad 2E12 aluminum alloy. The anodic oxide films on alclad 2E12 aluminum alloy are thicker and have fewer cavity defects, resulting in better corrosion resistance. The films obtained after 30min and 45min anodic oxidation treatment exhibit lower corrosion current and higher impedance of the porous layer than other anodizing time.

The magnetorheological fluid transmission device power transmission mechanism is analyzed according to the Bingham model,and a disk- type magnetorheological fluid transmission device is designed. By using finite element analysis software Ansoft Maxwell,the magnetic circuit design and magnetic field simulation are carried out and the magnetic field distribution nephogram is obtained. A magnetic field tester is built,on which the magnetic field is tested. This transmission device is able to meet the working requirements,and the foundation for the next experiment is laid.

With the combination of airborne Lidar and panchromatic images in 1981 and 2021, we investigated the canopy height structure of tropical forests in Menglun sub-reserve in the Xishuangbanna National Nature Reserve of Yunnan Province, and analyzed its relationship with environmental factors by using multiple regression tree (MRT) method. The results showed that forests in the Menglun sub-reserve could be clustered into seven types based on canopy height structures, with tropical rainforest, monsoon evergreen broad-leaved forest, secondary forest, and flood plain forest as the main types. The potential solar radiation, altitude, terrain profile curvature, slope and the brightness value of imageries in 1981 and 2021 were main factors that drove the classification. The tropical seasonal rainforest dominated by Pometia pinnata occupied the largest area in valley and low-land. The monsoon evergreen broad-leaved forest dominated by Castanopsis echinocarpa mainly distributed in the ridge and disturbed areas. The secondary forests had homogeneous canopy surface, which was significantly different from the primary forests. LANDIS Pro 7.0 model was used to simulate the dynamics of aboveground biomass of ten broadleaved tree species in the Xiao Xing' an Mountains area under current and various climate change scenarios from 2000 to 2200, and carbon content coefficients (CCCs) were coupled to cal- culate the aboveground carbon sequestration rates (ACSRs) of these species. The results showed that in the initial year of simulation, the biomasses and their proportions of Fraxinus mandshurica, Phellodendron amurense, Quercus mongolica, Ulmus propinqua, and Acer mono were relatively low, while those of Betula costata, Betula platyphylla, and Populus davidiana were higher. A trend of rise after decline occurred in ACSR for pioneer species in the mid and late periods of simulation years, but ACSRs for the other broadleaved tree species were considerably complex. The ACSRs of Q. mongolica and Tilla amurensis fluctuated in the ranges of -0.05-0.25 t . hm-2 . 10 a-1 and 0.16-1.29 t. hm-2. 10 a-1 in simulation years, respectively. The ACSRs of F. mandshurica, U. propinqua, A. mono, and B. costata showed a trend of decline after rise in late simulation years. There were significant differences in ACSR for P. amurense and B. davurica among various climate change scenarios in the periods of 2050-2100 and 2150-2200, while no significant difference in ACSR for the other species would be detected. Difference of sensitivity of various species in ACSR for future climate scenarios in the Small Khingan Mountains area existed. However, the uncertainty of future climates would not yield significant difference in ACSR for most broadleaved tree species. Urban expansion was simulated by SLEUTH model based on the data of Fuxin City, Northeast China in 1997-2013. The optimal parameters of urban expansion were obtained from SLEUTH model calibration, with the diffusion coefficient as 6, breed coefficient as 64, spread coefficient as 44, slope resistance as 52 and road gravity as 90. Urban growth types in Fuxin mainly belonged to new center growth and edge growth, i.e., the further expansion of new and old urban centers. Urban expansion was greatly influenced by roads. Fuxin, as a resource-exhausted city, suffered from the natural disasters, such as landslides, subsidence, and so on. The slope resistance of urban expansion was large in the development of urban land. From the perspective of urban scale, road gravity in smaller city was greater than in larger city. The urban expansion in smaller city was more inclined to the new center growth. The locations of enterprises and new development zones were more interested in the area of good transport facilities. Meanwhile, they were inclined to new center growth. Urban expansions were simulated based on optimal parameters of SLEUTH model. The simulated result of edge growth was better than the simulated result of new spreading center growth, because new spreading center growth was susceptible to policymaking, and cellular influence was little. Objective: To observe the clinical efficacy of intercondylar fossa plasty in preventing intercondylar fossa impingement syndrome after high tibial osteotomy., Methods: From August 2018 to August 2020, 84 patients with inverted knee osteoarthritis were treated by arthroscopy combined with high tibial osteotomy, and were divided into two groups with 42 cases in each group according to different surgical methods. In the intercondylar fossa plasty group, there were 13 males and 29 females, age ranged from 52 to 67 years old with an average of(58.27±4.32) years old, and arthroscopic intercondylar fossa plasty was performed first, and then high tibial osteotomy. In the arthroscopic cleansing group, 16 males and 26 females, age ranged from 50 to 71 years old with an average of (59.02±5.14) years old, underwent arthroscopic cleansing and then high tibial osteotomy. Postoperative treatment was evaluated using visual analogue scale(VAS), hospital for special surgery (HSS) score for the knee, and the occurrence of intercondylar percussa impingement., Results: All 84 patients were followed up, the duration ranged from 12 to 18 months with an average of (14.1±1.6) months. The VAS and HSS score of knee joint at 6, 12 and 18 months after surgery were significantly improved compared with preoperative period, and there was no significant difference between the two groups ( P >0.05), but the incidence of intercondylar fossa index and intercondylar fossa impact between the two groups was significantly compared 18 months after surgery ( P <0.05)., Conclusion: Intercondylar fossa plasty can effectively prevent the incidence of intercondylar fossa impact after high tibial osteotomy, and has a more significant effect on postoperative knee pain and function improvement.

Purpose: To investigate the difference in the retention effect of 3D printing resin splint and Hawley retainer combined with lingual retention wire in periodontal disease patients after orthodontic treatment., Methods: Forty patients who finished orthodontic treatment for periodontal disease from January 2021 to February 2022 were selected and divided into experimental group and control group according to the random number table method, with 20 patients in each group. Patients in the experimental group used 3D printing resin splint for retention, while patients in the control group used Hawley retainer combined with a lingual retention wire for retention. The chair-side operation time of the retainers was recorded for both groups. The maxillary and mandibular cuspid width, molar width, overjet, overbite and irregularity index were measured at 1 month and 6 months after orthodontic treatment in both groups. Statistical analysis was performed with SPSS 12.0 software package., Results: The mean time for chair-side manipulation of experimental group and control group was 8.23, 11.17 min, respectively. The difference between the two groups was statistically significant (P＜0.05) when comparing the width of the maxillary and mandibular cusps, the width of the molars, overjet, overbite and the irregularity index after 1 month and 6 months of wear of the retainers, which were significantly greater in the experimental group than in the control group at 6 months(P＜0.05)., Conclusions: 3D printing resin splint, with short chair-side manipulation time, was more effective than Hawley retainer combined with a lingual retention wire, and the efficacy of both groups was otherwise generally consistent.

To study the effects of different drying methods on the quality of male flowers of Eucommia ulmoides(MFOEU), we treated fresh MFOEU samples with drying in the shade(DS), vacuum freeze drying(VFD), high-or low-temperature hot air drying(HTHAD, LTHAD), microwave drying(MD), and vacuum drying(VD), respectively. The color, total flavonoid content, total polysaccharide content, and main active components such as geniposide, geniposidic acid, rutin, chlorogenic acid, galuteolin, pinoresinol diglucoside, and aucubin in MFOEU were taken as the evaluation indicators. The quality of MFOEU was comprehensively evaluated by entropy weight method combined with color index method, partial least squares discriminant analysis and content clustering heat map. The experimental results showed that VFD and DS basically kept the original color of MFOEU. The MFOEU treated with MD had higher content of total polysaccharides, phenylpropanoids, lignans, and iridoids. The MFOEU treated with LTHAD had higher content of total flavonoids and that treated with VD had lower content of active components. According to the results of comprehensive evaluation, the quality of MFOEU dried with different methods followed the order of MD&gt;HTHAD&gt;VFD&gt;LTHAD&gt;DS&gt;VD. Considering the color of MFOEU, the suitable drying methods were DS and VFD. Considering the color, active components, and economic benefits of MFOEU, MD was the suitable drying method. The results of this study are of a reference value for the determination of suitable methods for MFOEU processing in the producing areas.

This study aims to develop the pre-column derivatization high performance liquid chromatography(HPLC) method for the determination of 16 kinds of amino acids in Eucommia ulmoides leaves, and compare the content of amino acids in the leaves harvested at different time and under leaf-oriented cultivation mode(LCM) and arbor forest mode(AFM). The HPLC conditions are as below: phenyl isothiocyanate(PITC) as pre-column derivatization agent, Agilent ZORBAX C&#95;(18 )column(4.6 mm×250 mm, 5 μm), mobile phase A of acetonitrile-water(80∶20), mobile phase B of 0.1 mol·L~(-1) sodium acetate solution-acetonitrile(94∶6), gradient elution, flow rate of 1.0 mL·min~(-1), injection volume of 5 μL, column temperature of 40 ℃, and detection wavelength of 254 nm. The HPLC profile indicated well separation of 16 kinds of amino acids and the amino acid content in E. ulmoides leaves was up to 16.26%. In addition, the amino acid content in leaves of E. ulmoides under LCM was higher than under AFM. The amino acid content varied with the harvesting time. Through orthogonal partial least squares discriminant analysis, the amino acids of E. ulmoides under LCM and AFM were compared, which can distinguish the leaves under LCM from those under AFM. Principal component analysis was applied to comprehensively score the amino acids of E. ulmoides leaves. The results showed that the score of leaves under LCM was higher than that under AFM. Nutritional evaluation results indicated that the proteins in E. ulmoides leaves belonged to high-quality vegetable proteins. The established method for the determination of amino acid content is reliable. With the amino acid content as index, the leaf quality of E. ulmoides under LCM is better than that under AFM. This study lays a theoretical basis for the promotion of LCM for E. ulmoides and the development of medicinal and edible products from E. ulmoides leaves.

The main objective of this study was to explore the changes in bacterial communities and antibiotic resistance genes (ARGs) in an integrated fixed-film activated sludge (IFAS)+magnetic coagulation process wastewater treatment plant (WWTP) in Xinjiang. The bacterial communities and ARGs in the influent, suspended activated sludge, attached biofilm, and effluent were studied using 16S rRNA gene sequencing and metagenomic sequencing. The results showed that the average relative abundances of Chloroflexi and Nitrospirae in activated sludge were 3.50% and 0.03%, respectively, and their relative abundances in biofilm reached 10.02% and 2.12%, respectively. The average removal rates of NH 4 + -N and TN increased from 91.89% and 66.76% to 97.71% and 91.90% after the reformation of this wastewater treatment plant, respectively, indicating that IFAS enhanced the biological nitrogen removal capacity of wastewater treatment plants in cold regions. The average relative abundances of Ferruginibacter and Rhodoferax related to iron redox in the biological treatment section were 5.24% and 3.72%, respectively, and the relative abundance of Rhodoferax in effluent reached 9.48%, indicating that the magnetic powder had an impact on the bacterial community. The IFAS wastewater treatment plant had an obvious removal effect on ARGs, and the relative abundance of ARGs decreased from 191.08×10 -3 ‰ in the influent to 32.58×10 -3 ‰ in the effluent. The relative abundance of ARGs in activated sludge was 63.25×10 -3 ‰-72.38×10 -3 ‰, which was significantly higher than 41.31×10 -3 ‰ in biofilm. However, the relative abundances of dominant subtypes of ARGs such as sul2 , floR , and rpoB2 in biofilm were 5.77×10 -3 ‰, 2.52×10 -3 ‰, and 2.03×10 -3 ‰, respectively, which were higher than the 3.15×10 -3 ‰-3.57×10 -3 ‰, 1.73×10 -3 ‰-2.24×10 -3 ‰, and 1.28×10 -3 ‰-1.76×10 -3 ‰ in activated sludge. The network analysis indicated that Caldilineaceae&#95;norank and Trichococcus were respectively positively correlated with sul2 and floR . These results can provide theoretical reference for the optimal operation and ARGs control of WWTPs in cold regions.

This study established the EA.hy926 cell myocardial ischemia model to compare the effects of two Kaixin Powder prescriptions, Buxin Decoction(BXD) and Dingzhi Pills(DZP), at three dosages(500, 200, and 100 μg·mL~(-1)) on the cell viability. Further, the public databases(TCMSP, TCMID, SYMMAP, and STRING) and the network pharmacology methods such as KEGG pathway enrichment were employed to decipher the possible molecular mechanism of BXD in exerting the cardioprotective effect. The pharmacological effect of BXD was evaluated with the rat model of isoprenaline(ISO)-induced myocardial ischemia. The expression levels of proteins involved in the phosphatidylinositol-3-kinase/protein kinase B(PI3 K/AKT) signaling pathway were measured by Western blot. BXD significantly increased the viability of EA.hy926 cells, showing the performance superior to DZP. The network pharmacology analysis predicted that BXD might exert cardiac protection through the PI3 K/AKT signaling pathway. The in vivo experiment on rats showed that BXD treatment significantly increased the cardiac ejection fraction(EF), fractional shortening(FS), diastolic left ventricular anterior wall(LVAWd), systolic left ventricular anterior wall(LVAWs), and diastolic left ventricular posterior wall(LVPWd), significantly decreased the beat per minute(BPM) and diastolic left ventricular internal diameter(LVIDd), and significantly improved the ST segment in the electrocardiogram. The pathological results(Masson staining) showed that BXD restored the myocardial thickness, decreased the collagen fiber, increased the muscle fiber, and reduced the infarct area to alleviate myocardial ischemia. Furthermore, BXD lowered the serum levels of inflammatory cytokines [tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6)] and myocardial enzymes [creatine kinase(CK) and lactate dehydrogenase(LDH)], increased the p-AKT/AKT ratio, up-regulated the protein levels of PI3 K, NF-κB, IKK-α, and Bcl-xl, and down-regulated that of the apoptotic protein Bax. In conclusion, BXD may exert cardiac protection effect by regulating the PI3 K/AKT signaling pathway.

Objective: To compare the effectiveness of percutaneous coaxial large-channel endoscopic lumbar interbody fusion (PE-LIF) and minimal invasive transforaminal lumbar interbody fusion (MIS-TLIF) in the treatment of degenerative lumbar spinal stenosis., Methods: The clinical data of 134 patients with single-segment degenerative lumbar spinal stenosis who met the selection criteria between January 2019 and January 2021 were retrospectively analyzed, including 52 cases in PE-LIF group and 82 cases in MIS-TLIF group. There was no significant difference in general data such as gender, age, disease duration, surgical segment, and preoperative visual analogue scale (VAS) scores of low back pain and lower extremity pain, and Oswestry disability index (ODI) between the two groups ( P >0.05). The operation time, intraoperative blood loss, postoperative drainage, hospitalization stay, and complications were recorded and compared between the two groups. The level of serum creatine kinase (CK) was recorded at 1 day before operation and at 1 and 3 days after operation to evaluate intraoperative muscle damage. The Brantigan criteria was used to evaluate the interbody fusion in the two groups. The VAS scores of low back pain and lower extremity pain at 1 day before operation and at 3 days, 3 months, and 1 year after operation, and the ODI scores at 1 day before operation and at 3 months and 1 year after operation were recorded and compared between the two groups., Results: There was no significant difference in operation time and hospitalization stay between the two groups ( P >0.05). The intraoperative blood loss and postoperative drainage in the PE-LIF group were significantly lower than those in the MIS-TLIF group ( P <0.05). There was no significant difference in serum CK between the two groups before operation ( P >0.05), and the serum CK in the PE-LIF group at 1 and 3 days after operation were significantly lower than those in the MIS-TLIF group ( P <0.05). All patients were followed up regularly for 1 year. The postoperative VAS scores of low back pain and lower extremity pain and ODI score in both groups were significantly lower than those before operation ( P <0.05); there was no significant difference between the two groups ( P >0.05). At 1 year after operation, 48 patients in PE-LIF group had successful interbody fusion, and 77 patients in MIS-TLIF group had successful interbody fusion. There was no significant difference in the interbody fusion distribution between the two groups at 3 months and 1 year after operation ( P >0.05). There were 2 and 3 cases of lower limb numbness, 1 and 3 cases of neuroedema pain, 1 and 1 case of Cage displacement, 1 and 1 case of pedicle screw loosening in the PE-LIF group and MIS-TLIF group, respectively. No infection or dural sac tearing occurred in the two groups. There was no significant difference in the incidence of complications between the two groups (9.6% vs. 9.8%) ( χ 2 =0.001, P =0.979)., Conclusion: In the treatment of single-segment degenerative lumbar spinal stenosis, PE-LIF can achieve similar effectiveness as MIS-TLIF, and PE-LIF has less intraoperative blood loss and less muscle damage.

Objective To study the influence of recombinant bovine basic fibroblast growth factor as an adjuvant therapy on scar alleviation and inflammatory cytokines in patients with atrophic acne scar. Methods The random number table was employed to randomly assign 120 patients with atrophic acne scar into a test group and a control group.Both groups of patients were treated with CO 2 lattice laser.After the operation,the control group was routinely smeared with erythromycin ointment and the test group was coated with recombinant bovine basic fibroblast growth factor gel.The clinical efficacy,clinical indicators,scar alleviation,and inflammatory cytokine levels before and after treatment were compared,and adverse reactions were counted. Results The test group had higher total effective rate( P =0.040) and lower total incidence of adverse reactions( P =0.028) than the control group.Compared with the control group,the test group showcased short erythema duration after treatment( P =0.025),early scab forming( P =0.002),and early edema regression( P <0.001).After treatment,the proportion of grade 1 scars graded by Goodman and Baron's acne scar grading system in the test group and control group increased( P =0.001, P =0.027),and the proportion of grade 4 scars decreased( P <0.001, P =0.034).Moreover,the proportion of grade 1 scars in the test group was higher than that in the control group( P =0.031) after treatment,and the proportion of grade 4 scars presented an opposite trend( P =0.031).After treatment,the levels of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) in both groups declined(all P <0.001),and the test group had lower TNF-α and IL-1β levels than the control group(all P <0.001). Conclusion The recombinant bovine basic fibroblast growth factor gel as an adjuvant therapy of CO 2 lattice laser can effectively alleviate the atrophic acne scar,relieve local inflammatory reaction,and has good curative effect and less adverse reactions.

Purpose: To compare the effect of different root canal preparation methods on the incidence of interappointment emergencies (IAE) and root canal filling., Methods: A total of 96 teeth requiring root canal therapy due to pulpitis or periapical periodontitis from August 2018 to August 2021 were selected. They were randomly divided into 2 groups: MT group was treated with Mtwo root canal preparation method modified by Mtwo machine nickel-titanium file, while synchronous group was treated with modified Mtwo preparation method and synchronous root canal length measurement. After root canal preparation, the trial point film was taken, calcium hydroxide was used to seal the root canal, and routine thermoplasticizied gutta-percha root canal filling was performed during the follow-up visit. SPSS 22.0 software package was used to analyze the incidence of IAE and filling effect after root canal therapy., Results: There was no significant difference in the incidence of IAE between the two groups immediately after operation, three days and 1 week after operation(P＞0.05); the incidence of IAE in synchronous group was significantly lower than that in MT group at 1 and 2 days after operation(P＜0.05). The qualified rate of root canal filling in synchronous group was significantly higher than that in MT group (P＜0.05)., Conclusions: Synchronous method can reduce mechanical stimulation of apical area during root canal preparation, strictly control the working length of root canal and maintain apical barrier, thus reducing the incidence of IAE and effectively improving the qualification rate of root canal filling.

Objectives: To systematically evaluate the effect of prophylactic use of hydrolyzed protein formula on gastrointestinal diseases and physical development in preterm infants., Methods: A computerized search was performed in the databases including China National Knowledge Infrastructure, Wanfang Data, Weipu, PubMed, Embase, and the Cochrane Library to identify randomized controlled trials of the effect of prophylactic use of hydrolyzed protein formula on gastrointestinal diseases and physical growth in preterm infants. RevMan 5.3 software was used to perform a Meta analysis for the included studies., Results: A total of 7 randomized controlled studies were included. The results of Meta analysis showed that compared with the whole protein formula, the prophylactic use of hydrolyzed protein formula could reduce the risk of neonatal necrotizing enterocolitis ( RR =0.40, P =0.04) and feeding intolerance ( RR =0.40, P =0.005), and had no significant effect on the growth of weight, length and head circumference ( P >0.05)., Conclusions: Compared with the whole protein formula, the prophylactic use of hydrolyzed protein formula in preterm infants may reduce the occurrence of necrotizing enterocolitis and feeding intolerance, and can meet the nutrient requirement of physical development. However, the evidence is limited, and the results of this study cannot support the routine prophylactic use of hydrolyzed protein formula in preterm infants.

Traditional Chinese medicine(TCM) has unique advantages in the prevention and treatment of diseases owing to its holistic view and more than 2 000 years of experience in the clinical use of natural medicine. The &quot;holistic&quot; characteristic of TCM gives birth to a new generation of research paradigm featuring &quot;network&quot; and &quot;system&quot;, which has been developing rapidly in the era of biomedical big data and artificial intelligence. Network pharmacology, a representative research field, provides new ideas and methods for the research of the interdiscipline of artificial intelligence and medicine, the analysis of massive biomedical data, and the transformation from data to knowledge. TCM plays an important role in proposing the core theory of &quot;network target&quot; and promoting the establishment and development of network pharmacology, and has taken the lead in formulating the first international standard of network pharmacology--Network Pharmacology Evaluation Method Guidance. In terms of theory, network target can systematically link drugs and diseases and quantitatively interpret the overall regulatory mechanism of drugs. In the aspect of method, network pharmacology is developing towards a research model that combines computational, experimental, and clinical approaches. This review introduces the resent important progress of TCM network pharmacology in predicting drug targets, understanding the biological basis of drugs and diseases, and searching for disease and syndrome biomarkers. Under the guidance of Network Pharmacology Evaluation Method Guidance, the development of network pharmacology is expected to become more and more standardized and healthy. Network target will help produce more high-quality research outcomes in TCM and effectively boost the modernization and internationalization of TCM.

Urban street canyon is one of the most important characteristics and spatial forms of cities. It is one of the most frequently used public spaces in cities, with the most serious automobile exhaust pollution and the largest population density. The unreasonable space configuration and internal composition might decrease self-purification of urban ventilation but increase local air pollutant concentration. Here, we reviewed the impacts of street canyon morphology, street trees, vehicle flow and meteorological factors on the distribution of air pollutants in street canyons. We scrutinized the relevant methods of numerical simulation, wind tunnel experiments, and field monitoring on the distribution and diffusion of air pollutants in street canyons. We recommended that future research should concentrate on the impacts of various parameters on the distribution and diffusion of air pollutants based on the field monitoring data. Meanwhile, further research should develop optimization strategies for street canyon design which is conducive to the dispersion of air pollutants, and put forward scientific support and optimization scheme for the controlling of air pollutants from the perspective of urban planning and pattern optimization.

Objective: To compare clinical effects of different postoperative rehabilitation modes on lumbar degenerative diseases, and explore influence of rehabilitation mode and other factors on postoperative effect., Methods: From June 2013 to July 2016, totally 900 patients were admitted from nine tertiary hospitals in Beijing to perform single segment bone grafting and internal fixation due to lumbar degenerative diseases were prospectively analyzed. There were 428 males and 472 females, the age of patient over 18 years old, with an average of (51.42±12.41) years old;according to patients' subjective wishes and actual residence conditions, all patients were divided into three groups, named as observation group 1 (performed integrated rehabilitation approach and orthopedic treatment model intervention), observation group 2 (performed integrated rehabilitation approach and orthopedic treatment, classified rehabilitation model intervention), and control group(performed routine rehabilitation model intervention). Visual analogue scale(VAS), Oswestry Disability Index(ODI) and Japanese Orthopaedic Association (JOA) were used to evaluate postoperative efficacy among three groups at 24 weeks. Possible factors affecting the postoperative efficacy including age, age grouping, gender, body mass index (BMI), BMI grouping, education level, visiting hospital, payment method of medical expenses, preoperative complications, preoperative JOA score, clinical diagnosis, surgery section, operative method, intraoperative bleeding volume, postoperative complications and rehabilitation mode were listed as independent variables, and postoperative ODI score at 24 weeks as dependent variables. Univariate analysis was used to analyze relationship between influencing factors and postoperative efficacy. Multiple linear regression was used to analyze relationship between influencing factors, rehabilitation mode and postoperative ODI score at 24 weeks, in further to find out the main reasons which affect postoperative efficacy, and to analyze impact of rehabilitation mode on postoperative efficacy., Results: All patients were followed up for 24 weeks after operation. All incisions healed at stage I with stable internal fixation. (1)Evaluation of postoperative efficacy:① There were no statistical differences in preoperative VAS and ODI among three groups( P >0.05), the degree of pain and dysfunction decreased among three groups after operation, and had differences in postoperative VAS and ODI among three groups ( P <0.05). There were no significant differences between observation group 1 and observation group 2( P >0.05); while compared with observation group 1 and control group, observation group 2 and control group, there were significant differences ( P <0.05). ②The function among three groups were improved in varying degrees after operation. There was difference in JOA score among three groups before operation and 24 weeks after operation ( P <0.05). There were no difference in JOA score among three groups between observation group 1 and observation group 2 ( P >0.05);while compared with observation group 1 and control group, observation group 2 and control group, there were significant differences ( P <0.05). (2)Influencing factors at 24 weeks after operation:①Univariate analysis showed gender, age, age grouping, education level, preoperative complications, clinical diagnosis, operative section, operative method, preoperative JOA score and rehabilitation mode had statistical significance with postoperative ODI score at 24 weeks ( P <0.05). BMI, BMI grouping, payment method of medical expenses, visiting hospital, intraoperative bleeding volume, postoperative complications had no statistical significance with postoperative ODI score at 24 weeks ( P <0.05).②Multivariate analysis results showed gender, rehabilitation mode, age, preoperative JOA score entered the equation eventually, stepwise multiple linear equation obtained had statistical significance ( F =12.294, P = 0.000). Among rehabilitation mode, standardized regression coefficient of the integrated rehabilitation approach and orthopedic treatment with classified rehabilitation model was absolute value of the largest (0.176), which had the greatest influence on postoperative curative effect. The degree of dysfunction in control group was higher than that in observation group 1 and observation group 2. Postoperative dysfunction was more severe in males than that of in females. Older age has higher degree of dysfunction after operation. Lower preoperative JOA score has higher degree of dysfunction after operation., Conclusion: Preoperative JOA score, gender, age could predict postoperative clinical effects of lumbar degenerative diseases in varying degrees treated with single level bone graft fusion and internal fixation. Different rehabilitation modes could improve clinical effects. Intergrated rehabilitation orthopedic treatment model and integrated rehabilitation approach and orthopedic treatment with classifiedrehabilitation model are superior to conventional rehabilitation model in improving patients' postoperative function and relieving pain, which is worthy of promoting in clinical.

Atmospheric environment in urban built-up area is severely influenced by the surrounding landscape pattern. Understanding the relationship between air pollution and surrounding landscape pattern at small scale has great significance for mitigating air pollution from the perspective of urban construction. The annual average concentrations of NO 2 , SO 2 , PM 2.5 and PM 10 from 266 air pollution monitoring stations in 30 provincial capitals of China in 2017 were chosen as dependent variables. Ten two-dimensional and three-dimensional landscape pattern indices (number of buildings, building aggregation, building density, impervious water ratio, quantitative density of catering, building footprint area, high building ratio, floor area ratio, total building area and building type Shannon diversity index) within the 3 km area around the monitoring stations were used as independent variables. The effects of landscape pattern on the concentration of four air pollutants were analyzed using the boosted regression trees model. The results showed that the concentration of four air pollutants in the central and northern cities were significantly higher than that in the southeast coastal cities and southwest cities. The most important factor affecting the concentrations of NO 2 , SO 2 , PM 2.5 and PM 10 was the impervious ratio, with relative contribution rates of 40.7%, 36.3%, 51.0% and 51.8% respectively. The results of sub-region analysis showed that the most important influencing factor differed in different regions, including the impervious ratio in the East and Central China; the number and density of buildings in South China; the impervious ratio and diversity of building types in North China; the impervious ratio and the number of buildings in Northeast China, the density of buildings in Northwest China. Such differences were mainly caused by climate, topography, urban planning, and other factors.

Objective: To explore the value of non-invasive prenatal testing (NIPT) for the detection of fetal chromosome copy number variations (CNVs)., Methods: Clinical data of 18 661 pregnant women who underwent NIPT were collected. For fetuses suspected for carrying CNVs, amniotic fluid samples were collected for chromosomal karyotyping and/or chromosomal microarray analysis (CMA)., Results: Among all samples, NIPT suggested that 58 fetuses carried trisomy 21, 18 carried trisomy 18, 19 carried trisomy 13, 1 carried trisomies 18 and 21. Eighty eight women accepted invasive prenatal diagnosis. The results of CMA in 59 cases were consistent with those of NIPT, which yielded a consistency rate of 67.05%. In addition, 37 cases of fetal CNVs were detected by NIPT, of which 19 (15 microdeletions and 4 microduplications) have accepted invasive prenatal diagnosis. In 14 cases, the results were consistency with those of NIPT, with a consistent rate of 73.68%., Conclusion: NIPT features high sensitivity and accuracy. Invasive prenatal diagnosis should be considered for CNVs detected by NIPT, and by tracing its parental origin, it can provide guidance for clinical practice.

Objective: To investigate the clinical effect of high tibial osteotomy combined with arthroscopic lateral retinacular release in the treatment of knee varus osteoarthritis., Methods: From October 2017 to April 2019, a retrospective analysis was performed on 43 patients with knee varus osteoarthritis and lateral patellar compression syndrome treated by high tibial osteotomy combined with arthroscopic lateral retinacular release. There were 15 males and 28 females, aged 53 to 72(62.05±5.17) years. The visual analogue scale(VAS), Lysholm, and the knee range of motion were used to evaluate knee pain and functional recovery before operation, 2 weeks, 3 months and 12 months after operation. And the congruence angle (CA), patellar tilt angle (PTA), and femala-tibial angle (FTA) were measured respectively before and 12 months after operation to evaluate the congruence of patellar joint, and the improvement of line of gravity of lower limb., Results: All 43 patients were followed up for more than 12 months, with a follow-up time of 14 to 28 (19.60±4.50) months. The VAS scores decreased from 6.65±0.65 before operation to 2.16±0.95, 0.51±0.77 and 0.33±0.64 at 2 weeks, 3 months and 12 months after operation, and the difference was statistically significant ( P <0.001). Lysholm score increased from 43.02±8.54 before operation to 46.84±2.81, 72.42±5.30, and 93.40±5.44 at 2 weeks, 3 months and 12 months after operation, and the difference was statistically significant ( P <0.001). The knee range of motion increased from (86.97±5.02)° before operation to (99.38±3.27)°, (110.13±4.13)°, and (113.03±4.85)° at 2 weeks, 3 months and 12 months after operation, the difference was statistically significant ( P <0.001). The CA decreased from (7.81±1.21)° before operation to (5.82±4.10)° at 12 months after operation, the PTA decreased from (15.87±2.89)° before operation to (13.79±4.26)° at 12 months after operation, and the FTA decreased from(182.61±2.07)° before operation to(170.89±0.89)° at 12 months after operation, and the differences were statistically significant ( P <0.05). One case received proper braking and anticoagulation after operation, and was improved after 1 week. The swelling was observed in 14 patients after operation, and subsided about 2 weeks later., Conclusion: High tibial osteotomy combined with arthroscopic lateral retinacular release can relieve weight-bearing pain in frontal axis and improve the function of knee in sagittal axis.

Objective: To study the expression and function of long non-coding RNA linc00467 in childhood acute myeloid leukemia (AML)., Methods: Bone marrow samples were collected from 5 children with AML who were diagnosed from May 2016 to June 2018. Normal bone marrow samples based on bone marrow examination were collected from 3 children as controls. Quantitative real-time PCR was used to measure the expression of linc00467 in the two groups. A lentivirus system was used to achieve overexpression of linc00467 in AML cells (HL-60) (linc00467 overexpression group), and empty vector expressing green fluorescent protein (GFP) was transfected into AML cells to establish a GFP control group. A lentivirus system was used to insert an interfering sequence into AML cells (sh-linc00467 interfering group), and a random sequence was inserted to establish an sh-NC control group. Cell proliferation and resistance to doxorubicin were observed for all groups., Results: Compared with the normal control group, the children with AML had a significant increase in linc00467 (P=0.018). Overexpression and interference with linc00467 expression had no significant effect on cell proliferation. Compared with the GFP control group, the linc00467 overexpression group had a significant increase in the viability of HL-60 cells at the adriamycin concentrations of 0.1, 0.2, 0.3, 0.4, and 0.5 μg/mL (P<0.05). Compared with the sh-NC control group, the sh-linc00467 interfering group had a significant reduction in the viability of HL-60 cells at the adriamycin concentrations of 0.1, 0.2, 0.3, 0.4, and 0.5 μg/mL (P<0.05). Compared with the untreated group, the adriamycin treatment group had a significant increase in the expression of linc00467 in HL-60 cells (P<0.05)., Conclusions: This study reveals the biological function of linc00467 to promote the resistance to adriamycin in AML, which provides a basis for developing new therapeutic drugs for AML.

Objective: To evaluate the cartilage regeneration in the knee joint by arthroscopy after high tibial osteotomy., Methods: Eleven patients were included in the study who were treated with high tibial osteotomy and underwent microscopy when the internal fixation was unloaded from September 2017 to September 2019. Among them, there were 2 males and 9 females, aged from 55 to 64 years old. The internal and external compartment pictures of the knee were taken before and after surgery of removing the internal fixation and the International Cartilage Repair Society (ICRS) grading systerm was used to evaluate the degree of cartilage damage on the medial and lateral femoralcondyles and tibial plateau. The Westrn Ontarioand Mcmaster Universities osteoarthritis index (WOMAC) and the weight bearing line (WBL) were used to evaluate the function of the knee and the alignment of the lower limb., Results: All 8 patients were followed up for more than 12 months, ranging from 12 to 22 months. The degenerated cartilage of the medial femoral condyle and medial tibial plateau was covered by newly regenerated cartilage. WOMAC score decreased from 102-127 to 41-52 and WBL was improved from 17%-34% to 58%-64%. All incisions healed in stageⅠ, and no complications such as internal fixation rupture and infection occurred during and after the operation., Conclusion: High tibial osteotomy can relieve the pain of the knee and the dysfunction by adjusting lower limb alignment, and the degenerated cartilage could be regenerated in the medial femoral condyle and medial tibial plateau.

The close interactions between abiotic and biotic components create a variety of three-dimensional (3-D) landscape patterns. Landscape ecology, as a discipline of studying patterns and ecological processes, has made rapid progress in the exploration of 3-D space with the improvement of data acquirement ability, such as lidar technology. The real surface landscape can be described considering 3-D data, which improves the consistency between landscape indices and ecological process, and has overcome the shortage of ecological meanings of traditional researches. However, the lack of universality of methods and conclusions still exist due to different study backgrounds. The sensitivities of research results will increase with the expansion from two-dimensional to 3-D scale because of the increasing data quantity and accuracy. How to select and process the suitable scaled data to get more scientific conclusions need to be discussed in the future. The integration of multi-scale, multi-source and a long time series data will be the study trend with data acquisition becoming more convenient. In addition to the dynamic monitoring and prediction studies, the sustainable and ecological restoration application combined with landscape planning and design will be an important research direction.

Genetic analysis is an important part of undergraduate genetics teaching and tetrad analysis is unique and integral for genetic analysis of fungi. The ordered tetrad in Neurospora is an important material for genetic analysis, which can not only be used to study recombination between genes and centromeres, but also between genes themselves, as well as study the fine cross patterns between non-sister chromatids of homologous chromosomes. However, in textbooks and related professional journals, there is a lack of specific introduction to the induction methods of the seven basic class asci used in two genes analysis. In the present paper, we designed a table presenting the correlation between the three tetrad types (PD, NPD, T) and the four segregation pattern groups (Ⅰ Ⅰ, Ⅱ Ⅱ, Ⅰ Ⅱ, Ⅱ Ⅰ) to visually show the 12 possible combinations (3×4=12). Then five of them were excluded through the "×" symbol and in addition with three comments attached with the table, thus finally we obtained seven basic ascus types. We hope that this analytical method can assist the teaching of ordered tetrad analysis in Neurospora.

The bioweathering of stone cultural relics is a ubiquitous problem. Weathering prevention is an escalating challenge under the increasing global climate and environmental changes. Here, the mechanisms of lichen-microorganism mediated weathering of stone materials and their relationships with climatic and environmental factors were reviewed. The biological protection of lichens and the evaluation of the efficacy of biocides in lichen-control were discussed. The potential research directions in this field were proposed. Research on lichen-rock interfaces suggested that biological weathe-ring could be mainly attributed to physical and chemical weathering which represented by mycelium penetration and calcium oxalate formation. Bioweathering of outdoor stone cultural relics is closely related with the whole ecosystem encompassing factors, such as stone matrix, surrounding environment, and climate factors. Lichens have both biological weathering and protection effects on stone heritage. For the restoration of bioweathered stone cultural relics, environmental conditions for pre-servation of stone cultural relics should be improved step by step. The related industry regulations and national standards for evaluating biological weathering and control efficiency should be established to promote the efficient development of scientific protection.

Maxent niche model was used to project the potential distribution of alien plant species Spartina alterniflora in China, with 96 S. alterniflora records and marine and climatic data along China coastal area. The results showed that suitable area for the distribution of S. alterniflora occupied 85% of coastal areas, with the highly suitable area, the medium, and low suitable area accounting for 18%, 34% and 33%, respectively. Only 15% of the coastal areas were regions with distribution possibility of less than 5%. Its distribution suitability was mainly related to annual lowest seawater temperature, annual mean seawater temperature, annual mean air temperature, and annual lowest temperature in January. However, the importance of annual mean precipitation, annual mean daily diurnal range, seawater salinity, annual highest seawater temperature, annual highest tempe-rature in June and current velocity was relatively small. At the highly suitable areas for S. alterni-flora, the annual lowest seawater temperature ranged from 0.62 to 24.81 ℃, the annual mean seawater temperature ranged from 10.46 to 27.29 ℃, the annual mean air temperature was between 9 to 25 ℃, and the annual lowest temperature in January was between -13.5 and 16.7 ℃. The distribution possibility of S. alterniflora in the northern coastal area was over 20%, indicating it has a great potential to invade further north in China, especially in Bohai Bay areas. According to the current distribution records and climatic data in China, the invasion risk of S. alterniflora in central and southern coasts of Hainan and most regions of Taiwan Province was low, but the possibility of future invasion can not be excluded.

Objective: To validate the expressions of G protein-coupled receptor 81 (GPR81), monocarboxylate transporter (MCT) 1 and MCT4 in cervical squamous carcinoma and to explore their role in the onset of cervical squamous carcinoma.
 Methods: Immunohistochemical method was used to detect the expressions of GPR81, MCT1 and MCT4 in 16 normal cervical tissue and 44 cervical squamous carcinoma tissue. The associations of these proteins expression with cervical squamous carcinoma or clinicopathological factors were analyzed.
 Results: The expressions of GPR81, MCT1 and MCT4 in cervical squamous carcinoma tissue were higher than those in normal cervical tissue (P<0.01). In cervical squamous carcinoma group, the expressions of GPR81, MCT1 and MCT4 in clinical stage I-II group were lower than those in clinical stage III-IV group, with significant difference (P<0.05). The expressions of GPR81, MCT1 and MCT4 in middle-differentiated tumor group were lower than those in low-differentiated tumor group, but there was no significant difference between the two groups (P>0.05). No difference of the expressions of GPR81, MCT1 and MCT4 were found between cases with or without lymphatic metastasis (P>0.05). No correlation was found among GPR81, MCT1 and MCT4 in cervical squamous carcinoma (P>0.05).
 Conclusion: GPR81, MCT1 and MCT4 may be associated with the onset of cervical squamous carcinoma, and GPR81 may be associated with the progression of cervical squamous carcinoma.

The SanJiang Plain is one of the most concentrated and contiGuous area of marshes, which plays an irreplaceable role in maintaining regional ecological security. Based on the 3S technology, we examined the changes in land use and landscape pattern of the SanJiang Plain from 1980 to 2010. The results showed that marshland area lost 7135 km 2 , with a loss rate of 59.1%. The paddy area increased 18010 km 2 , with a growth rate of 610.1%. The results of landscape indices analysis showed that the number of patches increased, the landscape fragmentation became stronger, the landscape heterogeneity increased, and the different landscape types became homogenized. The CLUE-S model was validated based on the five different periods of land use maps during 1980-2010. The Kappa index between the simulation and actual measurement at the time scale of 30 years was 0.71, indicating that the model was suitable for 30 years simulation in the study area. The future wetland changes in the SanJiang Plain from 2010 to 2030 was simulated with validated CLUE-S models, including historical development scenario, planning scenario, and ecological restoration scenario. The simulation results showed that the marsh land would decrease 2515.44 km 2 and the paddy area would increase 19656.24 km 2 in the historical development scenario. The marsh land would decrease 303.28 km 2 , but the paddy area would increase 1392.08 km 2 in the planning scenario. The marsh land would increase 3585.61 km 2 and the paddy area would increase 289.72 km 2 in the ecological restoration scenario. The landscape patterns of the three scenarios were estimated using landscape indices. The results showed that the landscape pattern fragmentation would become more and more serious in the historical development scenario. The landscape pattern would have no signifi-cant changes in the planning scenario. The wetland area and connectivity would increase, the different landscape types would become balanced, and the landscape pattern would be gradually optimized in the ecological restoration scenario.

Objective: To investigate the current status of vitamin A deficiency in preschool children in Dongguan, China, as well as the effect of vitamin A on serum ferritin, red blood cell, and reticulocyte parameters., Methods: Cluster sampling was performed from April 2015 to December 2016 to select 2 085 preschool children (3-6 years old) without any disease in Dongguan. Routine blood test, reticulocyte count, serum ferritin measurement, hemoglobin electrophoresis, and vitamin A measurement were performed for all children. The associations of age and sex with vitamin A and serum ferritin concentrations were analyzed. The effect of vitamin A concentration on serum ferritin, red blood cell, and reticulocyte parameters and the effect of reduced iron storage caused by vitamin A deficiency on red blood cell parameters were evaluated., Results: Of the 2 085 children, 140 (6.71%) had reduced iron storage, and 678 (32.52%) had vitamin A deficiency. Among the 678 children with vitamin A deficiency, 647 (95.4%) had subclinical deficiency and 31 (4.6%) had clinical deficiency. There was no significant difference in vitamin A concentration between boys and girls, however girls had a significantly higher serum ferritin concentration than boys (P<0.05). The clinical vitamin A deficiency group had a significantly higher serum ferritin concentration than the subclinical vitamin A deficiency group and the normal group (P<0.05). In cases of vitamin A deficiency, the reduced iron storage group had significant reductions in mean corpuscular volume and mean corpuscular hemoglobin than the normal iron storage group (P<0.05). Compared with the normal vitamin A group, the vitamin A deficiency group had significantly lower hemoglobin concentration, mean corpuscular hemoglobin, red blood cell count, hematocrit, absolute reticulocyte count, reticulocyte percentage, and reticulocyte hemoglobin content, as well as a significantly higher mean corpuscular volume (P<0.05)., Conclusions: Vitamin A deficiency is prevalent in preschool children in Dongguan, China, and it may adversely affect serum ferritin, red blood cell, and reticulocyte parameters.

Objective: To investigate the regulation of Bruton's tyrosine kinase (Btk) and to explore its possible mechanism., Methods: After treatment with the proteasome inhibitors and/or phorbol esters (PMA), the mRNA and protein expression level of Btk was detected by RT-PCR and Western blot, respectively. The ubiquitination level of Btk in B lymphoblastoid A20 cells was estimated after stimulation via the crosslinking of BCR with anti-IgM antibody. The cotransfection of COS-7 cell with Btk, ubiquitin and Cbl was performed, then the ubiquitination level of Btk was measured. The Btk ubiquitination level was detected after ectopic expression of ubiquitin transfected with the wild type or triple mutant of Ub (K29R, K48R, K63R) . Mono-ubiquitination of Btk was detected with antibodies preferentially against monovalent ubiquitin; in addition, the protein expression levels of chloroquine-treated stably transfected cells expressing Btk-GFP were detected by Western blot, and quantified with the strength of GFP fluorescence., Results: In the presence of proteasome-specific inhibitors and/or PMA, steady-state levels of Btk protein were reduced due to decrease of transcription. Posttranslational modification of Btk by ubiquitination was observed, which was related with the level of Btk expression and activation. The E3 ubiquitin ligase Cbl, which binds to Btk, was also found to ubiquitinate this kinase. Altogether, the data of this study strongly suggest that Btk is regulated by poly- and/or mono-ubiquitination events., Conclusion: The Btk protein is dictated by its expression level through the ubiquitination pathway.

Objective: To explore whether neurotrophin receptor-interacting MAGE homolog (NRAGE) is involved in the intestinal ischemia-reperfusion (I/R) and its effect on the apoptosis of intestinal epithelial cells and the expression of occludin protein., Methods: The level of NRAGE protein after the rat small intestine I/R was detected by immunohistochemical (IHC) In vivo . The level of NRAGE protein and mRNA in IEC-6 cells after hypoxia and reoxygenation were tested by Western blot and RT-PCR respectively in vitro . The IEC-6 cells were divided into four groups, including NRAGE overexpression by lentivirus infection (Lv- NRAGE group), interference (sh- NRAGE group), lentivirus control (Lv-control group), and normal control group without lentivirus infection (NC group). The apoptosis of IEC-6 cells after infection was analyzed by flow cytometry. The level of the tight junction protein occludin was detected by Western blot., Results: The expression of NRAGE were highly increased in intestinal mucosa epithelial cells after I/R ( P <0.01). The proteins and mRNA levels of NRAGE were increased after 6 h of hypoxia in IEC-6 cells in vitro . Compared with the Lv-control group, the early apoptosis rate was raised ( P <0.01) and the level of occludin was reduced ( P <0.01) in Lv- NRAGE group; while the early apoptosis rate was reduced ( P <0.01) and the level of occludin was raised in sh- NRAGE group( P <0.001)., Conclusion: NRAGE may be involved in intestinal I/R and promote the apoptosis and decrease occludin expression of intestinal epithelial cells.

Objective: To explore the effect of Ibrutinib on the chemoresistance mediated by SDF-1α/CXCR4 axis in ALL cells., Methods: Flow cytometry was used to detect the apoptosis of cell line and expression of surface membrane CXCR4, Western blot was used to determine the expression level of CXCR4, ERK and Bcl-xL proteins, qPCR was used to assay the mRNA level of CXCR4., Results: Ibrutinib enhanced the apoptosis induced by adriamycin(ADR) (17.100±4.3% to 28.133±3.16%); Ibrutinib inhibited the phosphorylation of CXCR4 induced by SDF-1α and with concentration- and time- dependent manner (r 24h =-0.99659, r 48h =-0.99764, r=-0.99980). Ibrutinib inhibited the expression and activity of CXCR4 downstream signaling molecules pERK and BCL-xL., Conclusion: Ibrutinib can enhance the sensitivity of SUP-B15 to ADR, reverse SDF-1α/CXCR4-mediated chemoresistance in Ph + acute lymphoblastic leukemia cells. This mechanism of ibrutinib may be assosiated with inhibiting CXCR4/ERK/BCL-xL.

Proinsulin (Pins) is the precursor of insulin. The expression of proinsulin in Escherichia coli forms inclusion body, so that the recombinant protein should be processed with multiple steps to form active insulin. With the development in biotechnology, cell-free protein synthesis (CFPS) system is becoming a valuable tool in protein expression by decoupling the cell growth with protein production, which allows it to express proteins that would interfere with cell physiology. In this study, we synthesized soluble proinsulin in CFPS system in order to establish a new approach for both insulin expression and delivery. The soluble proinsulin was successfully expressed in CFPS system by fusing proinsulin with two types of fluorescent protein. The expression of Pins-mCherry was confirmed by Western blotting analysis, and the Pins-eGFP titer was (12.28±3.45) μg/mL in CFPS system. These results implicated that the proinsulin was expressed partially in soluble form. Here, for the first time, we successfully expressed soluble proinsulin in CFPS system by fluorescent protein fusion. These results provide useful information in developing new insulin expression and delivery method.

With the expansion of urban area, many cities are facing urban water environment issues, i.e., water resources shortage, lack of groundwater reserves, water pollution, urban waterlogging. For resolving these urban issues, 'sponge city' was proposed in 2015 in China. Liaodong Bay area of Panjin City in Liaoning Province of China was chosen as case study. Based on 'Sponge City Construction Technology Guide: Low Impact Development Rainwater System Building (Trial)', the underlying surface and types of land use in the typical area were analyzed. Sponge city plan of the study area was designed through combining topography, hydrology, rainfall intensity and other factors, and selecting LID measures. The results showed that when the study area reached the ove-rall target control rate (the control rate of the total annual runoff was ＞75%), the subsidence greenbelt rate was 1%-31%, with a total area of 13.73 km 2 ; the pervious pavement rate was 1%-13%, with a total area of 2.29 km 2 . This study could provide a case study for planning and designing of 'sponge city', proposing new ideas and methods for the research on landscape pattern and process.

Objectives: To improve the method of culturing and obtain purified alveolar epithelial type1 (AT1) cells of SD rats., Methods: 1 d newborn SD rats were applied for cell culture and brains were decapitated for lung tissues obtaining after respiratory cessation. Collagenase1and DNase1 were used to digest and isolate cells. Then, cells were put into the plate coated with type1 rat tail collagen and different adherence was used to purify cells. Meanwhile, culture medium replacement was conducted after 48 h. The growth status was observed under an inverted microscope. Immunofluorescence including specific marker AQP5, SPC, BSI, Vimentin were used to identify cells., Results: 2 d after incubation, the cells began to adhere to plate. At day 4 and 6, cells began to proliferate and exhibited a shape of spindle, cube and polygon. 8 d after incubation, the character of proliferation reached the highest and the cell viability was (87±8)% and purification was (87±5)%., Conclusion: By modifying the methods of tissues harvest, isolation and culture, we can obtain AT 1 cells with high yield, good growth state and super purity.

A case of primary yolk sac tumor of endometrium was retrospectively analyzed and relevant literature was systematically reviewed. We found that the primary yolk sac tumor of endometrium showed low incidence rate and was clinically characterized by abnormal vaginal bleeding, intrauterine lesions and significantly elevated alpha-fetoprotein (AFP) level. The final diagnosis should be confirmed by pathological examination. There is no guideline for the treatment of primary yolk sac tumor of endometrium due to its rareness. For now, surgery and chemotherapy are the major therapies and the curative effect is satisfactory in some cases.

Objective: To examine the IgG and IgM antibodies for parasites Cysticercus cellulosae and Toxoplasma gondii in 122 patients with meningitis encephalitis syndrome, and provide basis for clinical diagnosis of the meningitis encephalitis syndrome., Methods: The sera were collected from patients with meningitis encephalitis syndrome in Shanghai Jiaotong University Affiliated Sixth People’s Hospital, People’s Hospital of Danyang City, and Jiangsu University Affiliated Hospital from August, 2014 to December, 2015. Serum IgG and IgM antibodies for cysticercus and T. gondii were examined using antibody test kits. The antibody positive rate was calculated and its distribution was analyzed by gender, season, age and occupation., Results: A total of 122 patients with meningitis encephalitis syndrome were included. Seventeen and 22 patients of them were positive for IgG （13.9%, 17/122） and IgM（18.0%, 22/122） against cysticercus, respectively, while 29 and 8 cases were positive for IgG （23.8%, 29/122） and IgM （6.6%, 8/122） against T. gondii. The positive rate of cysticercus and T. gondii in males was 30.6%（22/72） and 31.9%（23/72） respectively, while that in females was 26.0%（13/50） and 24.0% （12/50）. The positive rate of IgM against cysticercus was 12.0%（3/25）, 27.0%（17/63）, 6.9% （2/29）, and 0（0/5） from spring to winter, highest within 13-25 years（45.0%, 9/20） among age groups, and highest in workers（7/14） among various occupations. The positive rate of IgM against T. gondii was 4.0%（1/25）, 11.1% （7/63）, 0（0/29）, and 0（0/5） from spring to winter, highest in ages >65 years（44.0%, 11/25）, and highest in patients with other occupations（4/10）. There was no statistically significant difference in the positive rate between males and females, and among different seasons, ages and occupations., Conclusion: The positive rate of antibodies against cysticercus and T. gondii is high in the patients included, suggesting that a serological test for parasite infection might be performed during clinical diagnosis of meningitis encephalitis syndrome.
