1. Developing an initial set of quality indicators for chiropractic care: a scoping review

Abstract: Abstract Background Quality indicators are standardized, evidence-based measures of health care quality. Currently, there is no basic set of quality indicators for chiropractic care published in peer-reviewed literature. The goal of this research is to develop a preliminary set of quality indicators, measurable with administrative data. Methods We conducted a scoping review searching PubMed/MEDLINE, CINAHL, and Index to Chiropractic Literature databases. Eligible articles were published after 2011, in English, developing/reporting best practices and clinical guidelines specifically developed for, or directly applicable to, chiropractic care. Eligible non-peer-reviewed sources such as quality measures published by the Centers for Medicare and Medicaid Services and the Royal College of Chiropractors quality standards were also included. Following a stepwise eligibility determination process, data abstraction identified specific statements from included sources that can conceivably be measured with administrative data. Once identified, statements were transformed into potential indicators by: 1) Generating a brief title and description; 2) Documenting a source; 3) Developing a metric; and 4) Assigning a Donabedian category (structure, process, outcome). Draft indicators then traversed a 5-step assessment: 1) Describes a narrowly defined structure, process, or outcome; 2) Quantitative data can conceivably be available; 3) Performance is achievable; 4) Metric is relevant; 5) Data are obtainable within reasonable time limits. Indicators meeting all criteria were included in the final set. Results Literature searching revealed 2562 articles. After removing duplicates and conducting eligibility determination, 18 remained. Most were clinical guidelines (n = 10) and best practice recommendations (n = 6), with 1 consensus and 1 clinical standards development study. Data abstraction and transformation produced 204 draft quality indicators. Of those, 57 did not meet 1 or more assessment criteria. After removing duplicates, 70 distinct indicators remained. Most indicators matched the Donabedian category of process (n = 35), with 31 structure and 4 outcome indicators. No sources were identified to support indicator development from patient perspectives. Conclusions This article proposes a preliminary set of 70 quality indicators for chiropractic care, theoretically measurable with administrative data and largely obtained from electronic health records. Future research should assess feasibility, achieve stakeholder consensus, develop additional indicators including those considering patient perspectives, and study relationships with clinical outcomes. 2. Deriving a year 2000 glacier inventory for New Zealand from the existing 2016 inventory

Abstract: Due to adverse snow and cloud conditions, only a few inventories are available for the maritime glaciers in New Zealand. These are difficult to compare as different approaches and baseline data have been used to create them. In consequence, glacier fluctuations in New Zealand over the past two decades are only known for a few glaciers based on field observations. Here we present the results of a new inventory for the ‘year 2000’ (some scenes are from 2001 and 2002) that is based on glacier outlines from a recently published inventory for the year 2016 and allowed consistent change assessment for nearly 3000 glaciers over this period. The year 2000 inventory was created by manual on-screen digitizing using Landsat ETM+ satellite imagery (15 m panchromatic band) in the background and the year 2016 outlines as a starting point. Major challenges faced were late and early seasonal snow, clouds and shadow, the geo-location mismatch between Landsat and Sentinel-2 as well as the correct interpretation of ice patches and ice under debris cover. In total, we re-mapped 2967 glaciers covering an area of 885.5 km2 in 2000, which is 91.7 km2 (or 10.4%) more than the 793.8 km2 mapped in 2016. Area change rates (mean rate −0.65% a−1) increase towards smaller glaciers. Strongest area loss from 2000 to 2016 occurred at elevations ~1900 m but the highest relative loss was found below 800 m a.s.l. In total, 109 glaciers split into two or more entities and 264 had wasted away by 2016.

3. Illumination of understudied ciliary kinases

Abstract: Cilia are cellular signaling hubs. Given that human kinases are central regulators of signaling, it is not surprising that kinases are key players in cilia biology. In fact, many kinases modulate ciliogenesis, which is the generation of cilia, and distinct ciliary pathways. Several of these kinases are understudied with few publications dedicated to the interrogation of their function. Recent efforts to develop chemical probes for members of the cyclin-dependent kinase like (CDKL), never in mitosis gene A (NIMA) related kinase (NEK), and tau tubulin kinase (TTBK) families either have delivered or are working toward delivery of high-quality chemical tools to characterize the roles that specific kinases play in ciliary processes. A better understanding of ciliary kinases may shed light on whether modulation of these targets will slow or halt disease onset or progression. For example, both understudied human kinases and some that are more well-studied play important ciliary roles in neurons and have been implicated in neurodevelopmental, neurodegenerative, and other neurological diseases. Similarly, subsets of human ciliary kinases are associated with cancer and oncological pathways. Finally, a group of genetic disorders characterized by defects in cilia called ciliopathies have associated gene mutations that impact kinase activity and function. This review highlights both progress related to the understanding of ciliary kinases as well as in chemical inhibitor development for a subset of these kinases. We emphasize known roles of ciliary kinases in diseases of the brain and malignancies and focus on a subset of poorly characterized kinases that regulate ciliary biology.

4. Southern Alps equilibrium line altitudes: four decades of observations show coherent glacier–climate responses and a rising snowline trend

Abstract: An end of summer snowline (EOSS) photographic dataset for Aotearoa New Zealand contains over four decades of equilibrium line altitude (ELA) observations for more than 50 index glaciers. This dataset provides an opportunity to create a climatological ELA reference series that has several applications. Our work screened out EOSS sites that had low temporal coverage and also removed limited observations when the official survey did not take place. Snowline data from 41 of 50 glaciers in the EOSS dataset were retained and included in a normalised master snowline series that spans 1977–2020. Application of the regionally representative normalised master snowline series in monthly and seasonally resolved climate response function analyses showed consistently strong relationships with austral warm-season temperatures for land-based stations west of the Southern Alps and the central Tasman Sea. There is a trend towards higher regional snowlines since the 1990s that has been steepening in recent decades. If contemporary decadal normalised master snowline series trends are maintained, the average Southern Alps snowline elevation will be displaced at least 200 m higher than normal by the 2025–2034 decade. More frequent extremely high snowlines are expected to drive more extreme cumulative mass-balance losses that will reduce the glacierised area of Aotearoa New Zealand.

5. Comparison of Machine-Learning and Deep-Learning Methods for the Prediction of Osteoradionecrosis Resulting From Head and Neck Cancer Radiation Therapy

Abstract: Purpose: Deep-learning (DL) techniques have been successful in disease-prediction tasks and could improve the prediction of mandible osteoradionecrosis (ORN) resulting from head and neck cancer (HNC) radiation therapy. In this study, we retrospectively compared the performance of DL algorithms and traditional machine-learning (ML) techniques to predict mandible ORN binary outcome in an extensive cohort of patients with HNC. Methods and Materials: Patients who received HNC radiation therapy at the University of Texas MD Anderson Cancer Center from 2005 to 2015 were identified for the ML (n = 1259) and DL (n = 1236) studies. The subjects were followed for ORN development for at least 12 months, with 173 developing ORN and 1086 having no evidence of ORN. The ML models used dose-volume histogram parameters to predict ORN development. These models included logistic regression, random forest, support vector machine, and a random classifier reference. The DL models were based on ResNet, DenseNet, and autoencoder-based architectures. The DL models used each participant's dose cropped to the mandible. The effect of increasing the amount of available training data on the DL models’ prediction performance was evaluated by training the DL models using increasing ratios of the original training data. Results: The F1 score for the logistic regression model, the best-performing ML model, was 0.3. The best-performing ResNet, DenseNet, and autoencoder-based models had F1 scores of 0.07, 0.14, and 0.23, respectively, whereas the random classifier's F1 score was 0.17. No performance increase was apparent when we increased the amount of training data available for DL model training. Conclusions: The ML models had superior performance to their DL counterparts. The lack of improvement in DL performance with increased training data suggests that either more data are needed for appropriate DL model construction or that the image features used in DL models are not suitable for this task.

Abstract: Abstract The World Magnetic Model (WMM) is a geomagnetic main field model that is widely used for navigation by governments, industry and the general public. In recent years, the model has been derived using high accuracy magnetometer data from the Swarm mission. This study explores the possibility of developing future WMMs in the post-Swarm era using data from the Iridium satellite constellation. Iridium magnetometers are primarily used for attitude control, so they are not designed to produce the same level of accuracy as magnetic data from scientific missions. Iridium magnetometer errors range from 30 nT quantization to hundreds of nT errors due to spacecraft contamination and calibration uncertainty, whereas Swarm measurements are accurate to about 1 nT. The calibration uncertainty in the Iridium measurements is identified as a major error source, and a method is developed to calibrate the spacecraft measurements using data from a subset of the INTERMAGNET observatory network producing quasi-definitive data on a regular basis. After calibration, the Iridium data produced main field models with approximately 20 nT average error and 40 nT maximum error as compared to the CHAOS-7.2 model. For many scientific and precision navigation applications, highly accurate Swarm-like measurements are still necessary, however, the Iridium-based models were shown to meet the WMM error tolerances, indicating that Iridium is a viable data source for future WMMs. Graphical Abstract

7. Updated inventory of glacier ice in New Zealand based on 2016 satellite imagery

Abstract: The only complete inventory of New Zealand glaciers was based on aerial photography starting in 1978. While there have been partial updates using 2002 and 2009 satellite data, most glaciers are still represented by the 1978 outlines in contemporary global glacier databases. The objective of this project is to establish an updated glacier inventory for New Zealand. We have used Landsat 8 OLI satellite imagery from February and March 2016 for delineating clean glaciers using a semi-automatic band ratio method and debris-covered glaciers using a maximum likelihood classification. The outlines have been checked against Sentinel-2 MSI data, which have a higher resolution. Manual post processing was necessary due to misclassifications (e.g. lakes, clouds), mapping in shadowed areas, and combining the clean and debris-covered parts into single glaciers. New Zealand glaciers cover an area of 794 ± 34 km2 in 2016 with a debris-covered area of 10%. Of the 2918 glaciers, seven glaciers are >10 km2 while 71% is 40%. Only 15 glaciers are located on the North Island. For a selection of glaciers, we were able to calculate the area reduction between the 1978 and 2016 inventories.

8. Reconceptualizing the U.S. Military’s Transition Assistance Program: The Success in Transition Model

Abstract: This article describes and proposes the Success in Transition Model as a guiding framework for a reconceptualized Transition Assistance Program (TAP) to support U.S. military members as they leave the service. Key groups of veterans have ongoing difficulties adjusting to civilian settings after their time in the service. Even with recent changes, the current TAP has a narrow focus of primarily aiding members to prepare for finding post-military employment, education, and benefits. Little attention is given to other key areas of their functioning such as adjusting to their new work/educational/cultural settings, meeting family transition needs, financial management issues, procuring housing, dealing with trauma responses, or assuring that veterans truly obtain the benefits and support they need. This new model addresses key flaws with the current TAP and is grounded in human and military transition theories and research into optimal ways to foster improved outcomes for transitioning military members.

Abstract: Abstract Glacier mass loss is recognized as a major contributor to current sea level rise. However, large uncertainties remain in projections of glacier mass loss on global and regional scales. We present an ensemble of 288 glacier mass and area change projections for the 21st century based on 11 glacier models using up to 10 general circulation models and four Representative Concentration Pathways (RCPs) as boundary conditions. We partition the total uncertainty into the individual contributions caused by glacier models, general circulation models, RCPs, and natural variability. We find that emission scenario uncertainty is growing throughout the 21st century and is the largest source of uncertainty by 2100. The relative importance of glacier model uncertainty decreases over time, but it is the greatest source of uncertainty until the middle of this century. The projection uncertainty associated with natural variability is small on the global scale but can be large on regional scales. The projected global mass loss by 2100 relative to 2015 (79 ± 56 mm sea level equivalent for RCP2.6, 159 ± 86 mm sea level equivalent for RCP8.5) is lower than, but well within, the uncertainty range of previous projections.

Abstract: The Darwin–Hatherton Glacial system (DHGS) connects the East Antarctic Ice Sheet (EAIS) with the Ross Ice Shelf and is a key area for understanding past variations in ice thickness of surrounding ice masses. Here we present the first detailed measurements of ice thickness and grounding zone characteristics of the DHGS as well as new measurements of ice velocity. The results illustrate the changes that occur in glacier geometry and ice flux as ice flows from the polar plateau and into the Ross Ice Shelf. The ice discharge and the mean basal ice shelf melt for the first 8.5 km downstream of the grounding line amount to 0.24 ± 0.05 km3 a−1 and 0.3 ± 0.1 m a−1, respectively. As the ice begins to float, ice thickness decreases rapidly and basal terraces develop. Abstract: Abstract We report on the large-scale evolution of dipolarization in the near-Earth plasma sheet during an intense (AL ~ −1000 nT) substorm on August 10, 2016, when multiple spacecraft at radial distances between 4 and 15 R E were present in the night-side magnetosphere. This global dipolarization consisted of multiple short-timescale (a couple of minutes) B z disturbances detected by spacecraft distributed over 9 MLT, consistent with the large-scale substorm current wedge observed by ground-based magnetometers. The four spacecraft of the Magnetospheric Multiscale were located in the southern hemisphere plasma sheet and observed fast flow disturbances associated with this dipolarization. The high-time-resolution measurements from MMS enable us to detect the rapid motion of the field structures and flow disturbances separately. A distinct pattern of the flow and field disturbance near the plasma boundaries was found. We suggest that a vortex motion created around the localized flows resulted in another field-aligned current system at the off-equatorial side of the BBF-associated R1/R2 systems, as was predicted by the MHD simulation of a localized reconnection jet. The observations by GOES and Geotail, which were located in the opposite hemisphere and local time, support this view. We demonstrate that the processes of both Earthward flow braking and of accumulated magnetic flux evolving tailward also control the dynamics in the boundary region of the near-Earth plasma sheet. Abstract: Recognising the scarcity of glacier mass-balance data in the Southern Hemisphere, a mass-balance measurement programme was started at Brewster Glacier in the Southern Alps of New Zealand in 2004. Evolution of the measurement regime over the 11 years of data recorded means there are differences in the spatial density of data obtained. To ensure the temporal integrity of the dataset a new geostatistical approach is developed to calculate mass balance. Spatial co-variance between elevation and snow depth allows a digital elevation model to be used in a co-kriging approach to develop a snow depth index (SDI). By capturing the observed spatial variability in snow depth, the SDI is a more reliable predictor than elevation and is used to adjust each year of measurements consistently despite variability in sampling spatial density. The SDI also resolves the spatial structure of summer balance better than elevation. Co-kriging is used again to spatially interpolate a derived mean summer balance index using SDI as a co-variate, which yields a spatial predictor for summer balance. The average glacier-wide surface winter, summer and annual balances over the period 2005–15 are 2484, −2586 and −102 mm w.e., respectively, with changes in summer balance explaining most of the variability in annual balance.

13. Peptide ELISA and FRET-qPCR Identified a Significantly Higher Prevalence of Chlamydia suis in Domestic Pigs Than in Feral Swine from the State of Alabama, USA

Abstract: Chlamydia suis is an important, highly prevalent, and diverse obligate intracellular pathogen infecting pigs. In order to investigate the prevalence and diversity of C. suis in the U.S., 276 whole blood samples from feral swine were collected as well as 109 fecal swabs and 60 whole blood samples from domestic pigs. C. suis-specific peptide ELISA identified anti-C. suis antibodies in 13.0% of the blood of feral swine (26/276) and 80.0% of the domestic pigs (48/60). FRET-qPCR and DNA sequencing found C. suis DNA in 99.1% of the fecal swabs (108/109) and 21.7% of the whole blood (13/60) of the domestic pigs, but not in any of the assayed blood samples (0/267) in feral swine. Phylogenetic comparison of partial C. suis ompA gene sequences and C. suis-specific multilocus sequencing typing (MLST) revealed significant genetic diversity of the C. suis identified in this study. Highly genetically diverse C. suis strains are prevalent in domestic pigs in the USA. As crowding strongly enhances the frequency and intensity of highly prevalent Chlamydia infections in animals, less population density in feral swine than in domestic pigs may explain the significantly lower C. suis prevalence in feral swine. A future study is warranted to obtain C. suis DNA from feral swine to perform genetic diversity of C. suis between commercial and feral pigs.

Abstract: The model single-stranded DNA binding protein of bacteriophage T4, gene 32 protein (gp32) has well-established roles in DNA replication, recombination, and repair. gp32 is a single-chain polypeptide consisting of three domains. Based on thermodynamics and kinetics measurements, we have proposed that gp32 can undergo a conformational change where the acidic C-terminal domain binds internally to or near the single-stranded (ss) DNA binding surface in the core (central) domain, blocking ssDNA interaction. To test this model, we have employed a variety of experimental approaches and gp32 variants to characterize this conformational change. Utilizing stopped-flow methods, the association kinetics of wild type and truncated forms of gp32 with ssDNA were measured. When the C-domain is present, the log-log plot of k vs. [NaCl] shows a positive slope, whereas when it is absent (*I protein), there is little rate change with salt concentration, as expected for this model.A gp32 variant lacking residues 292-296 within the C-domain, ΔPR201, displays kinetic properties intermediate between gp32 and *I. The single molecule force-induced DNA helix-destabilizing activitiesas well as the single- and double-stranded DNA affinities of ΔPR201 and gp32 truncated at residue 295 also fall between full-length protein and *I. Finally, chemical cross-linking of recombinant C-domain and gp32 lacking both N- and C-terminal domains is inhibited by increasing concentrations of a short single-stranded oligonucleotide, and the salt dependence of cross-linking mirrors that expected for the model. Taken together, these results provide the first evidence in support of this model that have been obtained through structural probes.

16. Accelerated Resolution Therapy for treatment of pain secondary to symptoms of combat-related posttraumatic stress disorder

Abstract: Background: As many as 70% of veterans with chronic pain treated within the US Veterans Administration (VA) system may have posttraumatic stress disorder (PTSD), and conversely, up to 80% of those with PTSD may have pain. We describe pain experienced by US service members and veterans with symptoms of PTSD, and report on the effect of Accelerated Resolution Therapy (ART), a new, brief exposure-based therapy, on acute pain reduction secondary to treatment of symptoms of PTSD. Methods: A randomized controlled trial of ART versus an attention control (AC) regimen was conducted among 45 US service members/veterans with symptoms of combat-related PTSD. Participants received a mean of 3.7 sessions of ART. Results: Mean age was 41.0 + 12.4 years and 20% were female. Most veterans (93%) reported pain. The majority (78%) used descriptive terms indicative of neuropathic pain, with 29% reporting symptoms of a concussion or feeling dazed. Mean pre-/post-change on the Pain Outcomes Questionnaire (POQ) was −16.9±16.6 in the ART group versus −0.7±14.2 in the AC group (p=0.0006). Among POQ subscales, treatment effects with ART were reported for pain intensity (effect size = 1.81, p=0.006), pain-related impairment in mobility (effect size = 0.69, p=0.01), and negative affect (effect size = 1.01, p=0.001). Conclusions: Veterans with symptoms of combat-related PTSD have a high prevalence of significant pain, including neuropathic pain. Brief treatment of symptoms of combat-related PTSD among veterans by use of ART appears to acutely reduce concomitant pain.

17. 'My Education, Not Only for Me and My Family but My People': Storied Experiences of Native American Students' College Choice at Four-Year Institutions in North Carolina

Abstract: While increased scholarship has begun to explore the stories of Native American students in higher education, less research has explored Native American student college choice. In this qualitative, narrative study, the experiences of seven Native American students' college choice at four-year institutions in North Carolina were explored. Perna's (2006) conceptual model of college choice provided a framework for the analysis. One research question guided this study: What were the storied experiences of Native American students from North Carolina when they chose to attend college? Findings from this study indicate that Native American students described their college choice process through four themes: family, Native identity and community, external factors, and growth and opportunities. Native American students in this study were inspired by their families and Native communities to pursue higher education. The participants pursued medical and human services disciplines to return and support their Native communities after completing their degrees. To be successful and have a "home feeling" on campus, they looked for universities with thriving Native communities through student organizations and supportive Native faculty and staff. Factors such as distance from home, academic preparation, and federal recognition impacted the participants choice. Through the four themes of Native college choice, higher education researchers, policymakers, and administrators have a better understanding of how to connect and serve Native American students. With increased attention, Native American student participation rates within higher education may increase as they have a home feeling on campus and perceive higher education as a gateway for success for their families and communities. Abstract: The land ice contribution to global mean sea level rise has not yet been predicted1 using ice sheet and glacier models for the latest set of socio-economic scenarios, nor using coordinated exploration of uncertainties arising from the various computer models involved. Two recent international projects generated a large suite of projections using multiple models2,3,4,5,6,7,8, but primarily used previous-generation scenarios9 and climate models10, and could not fully explore known uncertainties. Here we estimate probability distributions for these projections under the new scenarios11,12 using statistical emulation of the ice sheet and glacier models. We find that limiting global warming to 1.5 degrees Celsius would halve the land ice contribution to twenty-first-century sea level rise, relative to current emissions pledges. The median decreases from 25 to 13 centimetres sea level equivalent (SLE) by 2100, with glaciers responsible for half the sea level contribution. The projected Antarctic contribution does not show a clear response to the emissions scenario, owing to uncertainties in the competing processes of increasing ice loss and snowfall accumulation in a warming climate. However, under risk-averse (pessimistic) assumptions, Antarctic ice loss could be five times higher, increasing the median land ice contribution to 42 centimetres SLE under current policies and pledges, with the 95th percentile projection exceeding half a metre even under 1.5 degrees Celsius warming. This would severely limit the possibility of mitigating future coastal flooding. Given this large range (between 13 centimetres SLE using the main projections under 1.5 degrees Celsius warming and 42 centimetres SLE using risk-averse projections under current pledges), adaptation planning for twenty-first-century sea level rise must account for a factor-of-three uncertainty in the land ice contribution until climate policies and the Antarctic response are further constrained.

Abstract: Slope failures in high plasticity clay deposits are common occurrences in many parts of the world. In western and central Alabama, expansive Prairie clays are commonly found, and shallow slope failures have occurred in both fill and cut slopes containing these high plasticity clays. The objective of this study was to examine the effects of suction and the use of nonlinear strength envelopes on the embankment stability of a section of highway AL-5. The testing program consisted of fifteen ring shear tests performed using a Bromhead Ring Shear Device. The results of the tests were used to develop both linear and nonlinear fully softened and residual strength envelopes. The saturated strength envelopes are then used in a limit equilibrium slope stability analysis with and without the effects of suction. The results show stability (factor of safety &gt, 1) for all cases except the residual friction angle without suction. Given these results, large slope failures are unlikely to occur in this area, but surficial failures and deformations due to creep may be possible. These results demonstrate the importance of considering the effects of suction and nonlinear strength envelopes when examining the potential for shallow slope failures in high plasticity clays.

Abstract: In 1995, a geotechnical experimentation site was established in rural Lee County, Alabama near the area often referred to as Spring Villa. The Auburn University National Geotechnical Experimentation Site (AUNGES) site is situated near the southern terminus of the Appalachian Piedmont Physiographic province. The site consists of two sub sites, one that is a thick profile of residual soil and the other is an outcrop of fractured weathered quartzite. Extensive site investigation has been conducted at the site by many agencies and researchers using a wide array of in-situ soil tests. Foundations, excavations, and retaining structures have been constructed and load tested at the AUNGES. This paper summarizes all the work conducted at the site since its inception. Maps that detail the location of all in-situ tests and structures tested were generated. Representative profiles are included that relate early site investigation, between 1995 and 2005, and contemporary work done after 2010 are generated based on the results of piezocone, dilatometer, standard penetration, and shear wave velocity tests.

Abstract: The land ice contribution to global mean sea level rise has not yet been predicted 1 using ice sheet and glacier models for the latest set of socio-economic scenarios, nor using coordinated exploration of uncertainties arising from the various computer models involved. Two recent international projects generated a large suite of projections using multiple models 2–8, but primarily used previous-generation scenarios 9 and climate models 10, and could not fully explore known uncertainties. Here we estimate probability distributions for these projections under the new scenarios 11,12 using statistical emulation of the ice sheet and glacier models. We find that limiting global warming to 1.5 degrees Celsius would halve the land ice contribution to twenty-first-century sea level rise, relative to current emissions pledges. The median decreases from 25 to 13 centimetres sea level equivalent (SLE) by 2100, with glaciers responsible for half the sea level contribution. The projected Antarctic contribution does not show a clear response to the emissions scenario, owing to uncertainties in the competing processes of increasing ice loss and snowfall accumulation in a warming climate. However, under risk-averse (pessimistic) assumptions, Antarctic ice loss could be five times higher, increasing the median land ice contribution to 42 centimetres SLE under current policies and pledges, with the 95th percentile projection exceeding half a metre even under 1.5 degrees Celsius warming. This would severely limit the possibility of mitigating future coastal flooding. Given this large range (between 13 centimetres SLE using the main projections under 1.5 degrees Celsius warming and 42 centimetres SLE using risk-averse projections under current pledges), adaptation planning for twenty-first-century sea level rise must account for a factor-of-three uncertainty in the land ice contribution until climate policies and the Antarctic response are further constrained.

Abstract: Chlamydia suis is an important, highly prevalent, and diverse obligate intracellular pathogen infecting pigs. In order to investigate the prevalence and diversity of C. suis in the U.S., 276 whole blood samples from feral swine were collected as well as 109 fecal swabs and 60 whole blood samples from domestic pigs. C. suis-specific peptide ELISA identified anti-C. suis antibodies in 13.0% of the blood of feral swine (26/276) and 80.0% of the domestic pigs (48/60). FRET-qPCR and DNA sequencing found C. suis DNA in 99.1% of the fecal swabs (108/109) and 21.7% of the whole blood (13/60) of the domestic pigs, but not in any of the assayed blood samples (0/267) in feral swine. Phylogenetic comparison of partial C. suis ompA gene sequences and C. suis-specific multilocus sequencing typing (MLST) revealed significant genetic diversity of the C. suis identified in this study. Highly genetically diverse C. suis strains are prevalent in domestic pigs in the USA. As crowding strongly enhances the frequency and intensity of highly prevalent Chlamydia infections in animals, less population density in feral swine than in domestic pigs may explain the significantly lower C. suis prevalence in feral swine. A future study is warranted to obtain C. suis DNA from feral swine to perform genetic diversity of C. suis between commercial and feral pigs.

Abstract: Acute bacterial endocarditis is a rapid, difficult to manage, and frequently lethal disease. Potent antibiotics often cannot efficiently kill Staphylococcus aureus that colonizes the heart’s valves. S. aureus relies on virulence factors to evade therapeutics and the host’s immune response, usurping the host’s clotting system by activating circulating prothrombin with staphylocoagulase and von Willebrand factor–binding protein. An insoluble fibrin barrier then forms around the bacterial colony, shielding the pathogen from immune cell clearance. Targeting virulence factors may provide previously unidentified avenues to better diagnose and treat endocarditis. To tap into this unused therapeutic opportunity, we codeveloped therapeutics and multimodal molecular imaging to probe the host-pathogen interface. We introduced and validated a family of small-molecule optical and positron emission tomography (PET) reporters targeting active thrombin in the fibrin-rich environment of bacterial colonies. The imaging agents, based on the clinical thrombin inhibitor dabigatran, are bound to heart valve vegetations in mice. Using optical imaging, we monitored therapy with antibodies neutralizing staphylocoagulase and von Willebrand factor–binding protein in mice with S. aureus endocarditis. This treatment deactivated bacterial defenses against innate immune cells, decreased in vivo imaging signal, and improved survival. Aortic or tricuspid S. aureus endocarditis in piglets was also successfully imaged with clinical PET/magnetic resonance imaging. Our data map a route toward adjuvant immunotherapy for endocarditis and provide efficient tools to monitor this drug class for infectious diseases.

Abstract: After deep venous thrombosis (DVT), many patients have impaired quality of life (QOL). We aimed to assess whether pharmacomechanical catheter-directed thrombolysis (PCDT) improves short-term or long-term QOL in patients with proximal DVT and whether QOL is related to extent of DVT.The Acute Venous Thrombosis: Thrombus Removal with Adjunctive Catheter-Directed Thrombolysis (ATTRACT) trial was an assessor-blinded randomized trial that compared PCDT with no PCDT in patients with DVT of the femoral, common femoral, or iliac veins. QOL was assessed at baseline and 1 month, 6 months, 12 months, 18 months, and 24 months using the Venous Insufficiency Epidemiological and Economic Study on Quality of Life/Symptoms (VEINES-QOL/Sym) disease-specific QOL measure and the 36-Item Short Form Health Survey (SF-36) physical component summary (PCS) and mental component summary general QOL measures. Change in QOL scores from baseline to assessment time were compared in the PCDT and no PCDT treatment groups overall and in the iliofemoral DVT and femoral-popliteal DVT subgroups.Of 692 ATTRACT patients, 691 were analyzed (mean age, 53 years; 62% male; 57% iliofemoral DVT). VEINES-QOL change scores were greater (ie, better) in PCDT vs no PCDT from baseline to 1 month (difference, 5.7; P = .0006) and from baseline to 6 months (5.1; P = .0029) but not for other intervals. SF-36 PCS change scores were greater in PCDT vs no PCDT from baseline to 1 month (difference, 2.4; P = .01) but not for other intervals. Among iliofemoral DVT patients, VEINES-QOL change scores from baseline to all assessments were greater in the PCDT vs no PCDT group; this was statistically significant in the intention-to-treat analysis at 1 month (difference, 10.0; P .0001) and 6 months (8.8; P .0001) and in the per-protocol analysis at 18 months (difference, 5.8; P = .0086) and 24 months (difference, 6.6; P = .0067). SF-36 PCS change scores were greater in PCDT vs no PCDT from baseline to 1 month (difference, 3.2; P = .0010) but not for other intervals. In contrast, in femoral-popliteal DVT patients, change scores from baseline to all assessments were similar in the PCDT and no PCDT groups.Among patients with proximal DVT, PCDT leads to greater improvement in disease-specific QOL than no PCDT at 1 month and 6 months but not later. In patients with iliofemoral DVT, PCDT led to greater improvement in disease-specific QOL during 24 months.

Abstract: Glaciers experienced worldwide retreat during the twentieth and early twenty first centuries, and the negative trend in global glacier mass balance since the early 1990s is predominantly a response to anthropogenic climate warming. The exceptional terminus advance of some glaciers during recent global warming is thought to relate to locally specific climate conditions, such as increased precipitation. In New Zealand, at least 58 glaciers advanced between 1983 and 2008, and Franz Josef and Fox glaciers advanced nearly continuously during this time. Here we show that the glacier advance phase resulted predominantly from discrete periods of reduced air temperature, rather than increased precipitation. The lower temperatures were associated with anomalous southerly winds and low sea surface temperature in the Tasman Sea region. These conditions result from variability in the structure of the extratropical atmospheric circulation over the South Pacific. While this sequence of climate variability and its effect on New Zealand glaciers is unusual on a global scale, it remains consistent with a climate system that is being modified by humans., Many New Zealand glaciers advanced during recent global warming, bucking a worldwide trend of glacier retreat. Here, the authors show that these glacier advances were forced by a sequence of unusually cool years in the New Zealand region, rather than a period of increased precipitation.

Abstract: Glucose dysregulation and disruption of insulin signaling in the brain have been suggested as a possible etiology of Alzheimer’s Disease. In the present study, we show that oral delivery of MA-[D-Leu-4]-OB3 to young male Swiss Webster (SW) mice rendered hyperglycemic by intraperitoneal injection of streptozotocin (STZ), prevented insulin-induced increase in body weight gain and enhanced the effects of insulin alone on fasting blood glucose to levels approaching those of normal SW mice of the same age and sex. Novel object testing indicated that insulin treatment improved working memory in STZ-induced hyperglycemic mice, and that MA-[D-Leu-4]-OB3 given in combination with insulin significantly accelerated the time course of this improvement. Although positive results in an animal model may not be recapitulated in human pathology, our data suggest that MA-[D-Leu-4]-OB3, when added to an insulin regimen, may have application to the treatment of Alzheimer’s Disease by improving insulin sensitivity and glycemic control, thus slowing or preventing the progression of memory disruption associated with this disease.

Abstract: Estimating sediment deposition in a stream, a standard procedure for dealing with aggradation problem is complicated in an ungauged catchment due to the absence of necessary flow data. A serious aggradation problem within an ungauged catchment in Alabama, USA, blocked the conveyance of a bridge, reducing the clearance under the bridge from several feet to a couple of inches. A study of historical aerial imageries showed deforestation in the catchment by a significant amount over a period consistent with the first identification of the problem. To further diagnose the aggradation problem, due to the lack of any gauging stations, local rainfall, flow, and sediment measurements were attempted. However, due to the difficulty of installing an area-velocity sensor in an actively aggrading stream, the parameter transfer process for a hydrologic model was adopted to understand/estimate streamflow. Simulated discharge combined with erosion parameters of MUSLE (modified universal soil loss equation) helped in the estimation of sediment yield of the catchment. Sediment yield for the catchment showed a significant increase in recent years. A two-dimensional hydraulic model was developed at the bridge site to examine potential engineering strategies to wash sediments off and mitigate further aggradation. This study is to quantify the increase of sediment yield in an ungauged catchment due to land cover changes and other contributing factors and develop strategies and recommendations for preventing future aggradation in the vicinity of the bridge.

Abstract: Fish in nature have several distinct advantages over traditional propeller driven underwater vehicles including maneuverability and flow sensing capabilities. Taking inspiration from biology, this work seeks to answer three questions related to bioinspired pursuit and apply the knowledge gained therein to the control of a novel, reaction-wheel driven autonomous fish robot. Which factors are most important to a successful pursuit? How might we guarantee capture with underwater pursuit? How might we track the wake of a flapping fish or vehicle? A technique called probabilistic analytical modeling (PAM) is developed and illustrated by the interactions between predator and prey fish in two case studies that draw on recent experiments. The technique provides a method for investigators to analyze kinematics time series of pursuit to determine which parameters (e.g. speed, flush distance, and escape angles) have the greatest impact on metrics such as probability of survival. Providing theoretical guarantees of capture become complicated in the case of a swimming fish or bioinspired fish robot because of the oscillatory nature fish motion. A feedback control law is shown to result in forward swimming motion in a desired direction. Analysis of this law in a pursuit scenario yields a condition stating whether capture is guaranteed provided some basic information about the motion of the prey. To address wake tracking inspiration is taken from the lateral line sensing organ in fish, which is sensitive to hydrodynamic forces in the local flow field. In experiment, an array of pressure sensors on a Joukowski foil estimates and controls flow-relative position in a Karman vortex street using potential flow theory, recursive Bayesian filtering, and trajectory-tracking, feedback control. The work in this dissertation pushes the state of the art in bioinspired underwater vehicles closer to what can be found in nature. A modeling technique provides a means to determine what is most important to pursuit when designing a vehicle, analysis of a control law shows that a robotic fish is capable of pursuit engagements with capture guarantees, and an estimation framework demonstrates how the wake of a swimming fish or obstacle in the flow can be tracked.

Abstract: Glacier behaviour during the mid-Holocene (MH, 6000 years BP) in the Southern Hemisphere provides observational data to constrain our understanding of the origin and propagation of palaeoclimate signals. In this study we examine the climatic forcing of glacier response in the MH by evaluating modelled glacier equilibrium line altitudes (ELAs) and climatic conditions during the MH compared with pre-industrial time (PI, year 1750). We focus on the middle latitudes of the Southern Hemisphere, specifically Patagonia and the South Island of New Zealand. Climate conditions for the MH were obtained from PMIP2 model simulations, which in turn were used to force a simple glacier mass balance model to simulate changes in ELA. In Patagonia, the models simulate colder conditions during the MH in austral summer (−0.2 °C), autumn (−0.5 °C), and winter (−0.4), and warmer temperatures (0.2 °C) during spring. In the Southern Alps the models show colder MH conditions in autumn (−0.7 °C) and winter (−0.4 °C), warmer conditions in spring (0.3 °C), and no significant change in summer temperature. Precipitation does not show significant changes but exhibits a seasonal shift, with less precipitation from April to September and more precipitation from October to April during the MH in both regions. The mass balance model simulates a climatic ELA that is 15–33 m lower during the MH compared with PI conditions. We suggest that the main causes of this difference are driven mainly by colder temperatures associated with the MH simulation. Differences in temperature have a dual effect on glacier mass balance: (i) less energy is available for ablation during summer and early autumn and (ii) lower temperatures cause more precipitation to fall as snow rather than rain in late autumn and winter, resulting in more accumulation and higher surface albedo. For these reasons, we postulate that the modelled ELA changes, although small, may help to explain larger glacier extents observed by 6000 years BP in South America and New Zealand.

Abstract: Knowledge of the ice thickness distribution of glaciers and ice caps is an important prerequisite for many glaciological and hydrological investigations. A wealth of approaches has recently been presented for inferring ice thickness from characteristics of the surface. With the Ice Thickness Models Intercomparison eXperiment (ITMIX) we performed the first coordinated assessment quantifying individual model performance. A set of 17 different models showed that individual ice thickness estimates can differ considerably – locally by a spread comparable to the observed thickness. Averaging the results of multiple models, however, significantly improved the results: On average over the 21 considered test cases, comparison against direct ice thickness measurements revealed deviations in the order of 10 &pm; 24% of the mean ice thickness (1-sigma estimate). For models relying on multiple data sets -- such as surface ice velocity fields, surface mass balance, or rates of ice thickness change – the results highlighted the sensitivity to input data consistency. Together with the requirement of being able to handle large regions in an automated fashion, the capacity of better accounting for uncertainties in the input data will be a key for an improved next generation of ice thickness estimation approaches.

Abstract: The Darwin–Hatherton Glacial system (DHGS) connects the East Antarctic Ice Sheet (EAIS) with the Ross Ice Shelf and is a key area for understanding past variations in ice thickness of surrounding ice masses. Here we present the first detailed measurements of ice thickness and grounding zone characteristics of the DHGS as well as new measurements of ice velocity. The results illustrate the changes that occur in glacier geometry and ice flux as ice flows from the polar plateau and into the Ross Ice Shelf. The ice discharge and the mean basal ice shelf melt for the first 8.5 km downstream of the grounding line amount to 0.24 ± 0.05 km3 a−1 and 0.3 ± 0.1 m a−1, respectively. Abstract: Observations show that glaciers around the world are in retreat and losing mass. Internationally coordinated for over a century, glacier monitoring activities provide an unprecedented dataset of glacier observations from ground, air and space. Glacier studies generally select specific parts of these datasets to obtain optimal assessments of the mass-balance data relating to the impact that glaciers exercise on global sea-level fluctuations or on regional runoff. In this study we provide an overview and analysis of the main observational datasets compiled by the World Glacier Monitoring Service (WGMS). The dataset on glacier front variations (∼42 000 since 1600) delivers clear evidence that centennial glacier retreat is a global phenomenon. Intermittent readvance periods at regional and decadal scale are normally restricted to a subsample of glaciers and have not come close to achieving the maximum positions of the Little Ice Age (or Holocene). Glaciological and geodetic observations (∼5200 since 1850) show that the rates of early 21st-century mass loss are without precedent on a global scale, at least for the time period observed and probably also for recorded history, as indicated also in reconstructions from written and illustrated documents. This strong imbalance implies that glaciers in many regions will very likely suffer further ice loss, even if climate remains stable.

Abstract: Titanic scholars contend that the demise of'the unsinkable ship'left more behind than a memory of April 15, 1912, as an important point in history. Through books, films, stories, and songs, the archetypal shipwreck has endured as a metaphor for the perils of mankind's hubris and the fallibility of technology. In 1985, the discovery of the long-missing wreckage two miles below the surface of the Atlantic revitalized interest in the Titanic and spawned a new generation of books, films, and, for the first time, websites, and computer games. James Cameron's blockbuster Titanic became the biggest movie of all time and engendered still greater popular interest in the tragic event. This bibliography is a survey of the immense volume of literary, dramatic, and commercial endeavors that came out of history's most compelling shipwreck. Organized by genre in accessible categories and short entries, the book includes Titanic-inspired documentaries, narrative films, children's books, histories, short stories, novels, plays, articles, essays, software, websites, poems, and songs. 50. Recent developments in the pharmacological management of pain in children.

Abstract: PURPOSE OF REVIEW: This review explores progress in developmental pharmacokinetics, pharmacogenomics and formulations of analgesic agents, and discusses potential implications for pain therapy.RECENT FINDINGS: Characterization of the developmental pharmacokinetics of morphine, tramadol, paracetamol and nonsteroidal anti-inflammatory drugs has improved dosing in children. Oral sugar solutions have replaced the brandy/sugar pacifier and are effective for single painful events in neonates. Intravenous paracetamol offers increased dosing accuracy, and avoids absorption and bioavailability variability. New nitric-oxide-releasing versions of paracetamol and nonsteroidal anti-inflammatory drugs offer safer alternatives to their parent drugs with enhanced potency. Ketamine has come under a cloud for its possible effects on the neonatal developing brain, but it is being used increasingly in children to supplement opioids for pain after major surgery. Hopes that morphine analgesia may improve neurological outcome in premature babies have not materialized. Reports concerning chronic pain are generally case series and controlled trials are rare and nearly nonexistent in children.SUMMARY: Unlicensed drug use in the very young will increase as familiarity increases. Pharmacogenomic studies have the potential to tailor drug therapy to the individual and decrease between-patient variability. Unfortunately, the pharmacodynamic knowledge in children of analgesic agents remains neglected and is usually extrapolated from adult data. [ABSTRACT FROM AUTHOR]
