Fysisk aktivitet har visat sig förebygga allvarliga hälsorelaterade sjukdomar som till exempel diabetes och fetma, men trots den vetskapen blir vi allt mer stillasittande i vår vardag. För att upprätthålla en hälsosam livsstil är motivation en viktig aspekt att ta hänsyn till. För att skapa förståelse för de psykologiska behov som stödjer motivation används i den här studien makroteorin Self-determination theory (SDT). SDT konkretiserar motivation utifrån inre och yttre motivation, där yttre motivation traditionellt ansetts vara en svagare form av motivation. Allt fler använder sig av mobila hälsoapplikationer som till exempel RunKeeper för att stödja sin motivation för fysisk aktivitet. Mobila hälsoapplikationer använder sig ofta av spelifiering. Spelifiering innebär att spelelement implementeras i en annan kontext än den generella spelkontexten. Spelifiering som är implementerad i dagens mobila hälsoapplikationer stödjer främst yttre motivation för fysisk aktivitet, vilket kan orsaka att den inre motivationen minskar och att den fysiska aktiviteten upphör. Med tanke på att fysisk aktivitet är grundläggande för en hälsosam livsstil och att dagens mobila hälsoapplikationer främst använder sig av spelifiering som stödjer yttre motivation, ämnar studien att undersöka hur spelifiering kan designas för att stödja inre motivation för fysisk aktivitet. Studien utfördes med en designorienterad forskningsansats där spelelement implementerades i en prototyp. Prototypen användes för att utvärdera om spelelementen kunde stödja en känsla av inre motivation för fysisk aktivitet. Spelelementen analyserades med hjälp av SDT för att se hur de kunde stödja de tre psykologiska behoven. Utvärderingen av prototypen resulterade i tio designförslag för hur spelifiering kan designas för att stödja inre motivation för fysisk aktivitet.

Det overordnede målet med prosjektet var å bidra til økt kunnskap om hvordan genetisk bakgrunn, miljø (ulike lengde i ferskvann før utsett i sjø) og ulik fettsyreprofil i fôr kan påvirke laksens medfødte evne til å produsere og deponere EPA og DHA. Vi benyttet 4 Høy- og 3 Lav-desaturase familier som gjennomgikk to ulike smoltifiseringsregimer (normalt og utsatt) og ble gitt to ulike fôr med lav (25 %RO) eller høy (90 %RO) innblanding av rapsolje (RO) i fôret. Det ble foretatt fire ulike prøveuttak (før, under og etter smoltifisering). Resultatene viste at regulering av EPA og DHA syntese er kompleks og styres blant annet av faktorer som miljø (lysstyring, overføring til saltvann), genetikk og fôr. Laks som gjennomgikk et utsatt smoltifiseringsregime viste bedre kapasitet til syntese av EPA og DHA, enn laks som fulgte normalt smoltifiseringsregime. Laks som ble seinere smoltifisert viste også høyere DHA nivå i filet enn laks som ble smoltifisert ved et tidligere tidspunkt. Genuttrykk av alle desaturasene ble nedregulert av lysstyring, mens overføring til saltvann oppregulerte 6fad_b og 6fad_c. Lysstyring kan se ut til å hemme enzymaktiviteten av 5 desaturase og stimulere aktiviteten av 6 desaturase. Laks fra de fleste Høy-desaturase gruppene viste i februar-uttaket høyere kapasitet til EPA+DHA syntese enn Lav-desaturase gruppen. Dette viser at familiene som er selektert for høyt genuttrykk av en 6 desaturase (6fad_b), også har bedre kapasitet til EPA+DHA syntese. Høy innblanding av RO i fôret ga større økning av DHA i filet, enn lav innblanding av RO. Gruppen med 90 %RO i fôret hadde også høyere genuttrykk av alle desaturasene, enn gruppen som fikk 25 %RO i fôret. Dette kan skyldes en inhiberende effekt av DHA i fôret på genuttrykk av desaturasene. The objective of the project was to increased knowledge on how genetic background, smoltification and feed composition affect salmon’s innate capacity to produce and dispose of EPA and DHA. Salmon that followed a postponed smoltification regime showed better capacity to synthesize EPA and DHA, than salmon going through a normal smoltification regime. Salmon smoltified at a later stage also showed higher level of DHA in the fillet than salmon smoltified at a normal time point. Gene expression of all desaturases were down-regulated by light treatment, while transfer to saltwater upregulated 6fad_b and 6fad_c. Light seems to inhibit the enzyme activity of 5 desaturase and stimulate activity of 6 desaturase. Salmon from most of the High-desaturase groups showed in February higher capacity to synthesize EPA + DHA than Low-desaturase groups. These results show that families selected for high gene expression of 6 desaturase (6fad_b), also have better capacity to synthesize EPA + DHA. Groups fed 90 % RO showed higher gene expression level of all desaturases, than the group fed 25 % RO. This may be caused by an inhibitory effect of dietary DHA on gene expression of desaturases.

Et viktig mål i dette prosjektet var å studere potensialet for å påvirke EPA og DHA nivået i muskel i slaktefisk ved fôring med ulike nivåer av EPA og DHA i tidlige livsfaser; henholdsvis før smoltifisering og rett etter smoltifisering. Hypotesen er at ernæringsmessig programmering i tidlige livsfaser kan påvirke fiskens lipidmetabolisme i seinere livsfaser. Startfôringsperioden (fram til 40 gram fiskestørrelse) gav ikke signifikant utslag på fettsyresammensetningen av laksefilet ved 4 kg. Fôring i perioden fra smoltifisering og opp til 400 gram derimot, hadde signifikant betydning for innhold av EPA og DHA i filet etter 13 måneder i sjø og 3,5 kg økning i kroppsvekt. To andre viktige mål i dette prosjektet var å identifisere eventuelle langtidseffekter mellom Høy og Lav desaturase familier på fettdeponering, EPA og DHA nivå i filet, kvalitet og utvalgte helsemarkører. Våre data viser at genetisk bakgrunn hadde betydning for fettfordelingen i fiskekroppen, med spesielt stor betydning for utvikling av fettlever og fordeling mellom innvollsfett og muskelfett. Høy desaturase gruppen viste høyere kapasitet til EPA og DHA syntese i tidlige livsfaser, lavere forekomst av fettlever og mer fett fordelt til muskel sammenlignet med Lav desaturasegruppen. Den genetiske pre-disponeringen til høyere kapasitet til EPA og DHA syntese i Høy desaturase familien sammenlignet med Lavdesaturasefamilien i tidlig livsfase, synes å forsvinne når dietter med høyt nivå av planteolje ble benyttet i vekstfasen i sjø. Dette resulterte i ingen forskjell i prosent EPA + DHA i filet til fisk på 4 Kg mellom de genetiske gruppene, men på grunn av det høyere fettnivået i muskel i Høydesaturase gruppen var nivået av EPA + DHA i gram per 100 gram muskel signifikant høyere i denne gruppen. I tillegg til betydelige effekter på hele kroppens lipid metabolisme, fant vi også helseforskjeller mellom Høy og Lavdesaturasegruppene som muligens kan være relatert til forskjeller i fettmetabolisme. Høydesaturasegruppen hadde bl.a. signifikant lavere dødelighet etter sjøvannsoverføring. Det var ingen forskjeller i muskelkvalitet mellom de genetiske gruppene i slaktefisk. An important goal of this project was to study the potential to influence EPA and DHA levels in muscle in harvest size fish by feeding different levels of EPA and DHA in early life stages; before smoltification and after smoltification respectively. The hypothesis is that nutritional programming in early life stages may affect lipid metabolism in later stages of life. Our study showed that the start feeding period (up to 40 grams) did not give significant effect on fatty acid composition of salmon fillets when the fish reached 4 kg. Feeding during the period from smoltification to 400 grams, however, had a significant effect on the levels of EPA and DHA in fillets after 13 months at sea and 3.5 kg increase in fish body weight. Two other goals was to identify any long-term effects between High and Low desaturase families on fat deposition, EPA and DHA levels in fillets, quality and selected health markers. Our data shows that genetic background of the fish had significant effect on fat distribution in fish body, with particular importance for the development of fatty liver and distribution of fat between internal organs and muscle. High desaturase group showed higher capacity to EPA and DHA synthesis in early life stages, lower incidence of fatty liver and more fat distributed to muscle compared with Low desaturase group. The genetic pre-disposition to higher capacity to EPA and DHA synthesis in High desaturase family compared with Low desaturase family, seems to disappear when diets with high levels of plant oil was used in the growth phase in the sea. Due to the higher fat level in muscle in High desaturase group, the level of EPA + DHA in grams per 100 grams of muscle was significantly higher in this group than in Low desaturase group. The High desaturase group had significantly lower mortality after seawater transfer. There were no differences in muscle quality between the genetic groups in harvest size fish.

13 Pág., Knowledge of population structure and genetic diversity within and between wild and farmed populations of gilthead sea bream (Sparus aurata) and European seabass (Dicentrarchus labrax) is important to achieve sustainable aquaculture production of these species and to assess the risk of genetic impacts of fish escaped from farms. Previous population genetic studies on these species have been based on a limited number of genetic markers and samples. In this study, these features were assessed using samples from 24 seabream and 25 seabass populations distributed throughout the Mediterranean Sea, and 3 wild seabream Atlantic populations. Samples were genotyped with a newly developed combined species SNP array that includes ~60K SNPs. Data from sequencing pools of individual DNA from the same populations were also used. Different approaches were employed for identifying the extent of population stratification within species. The effective population size (a parameter inversely related to the rate at which genetic variability is lost) was estimated for each population based on linkage disequilibrium. Population structure results revealed a clear differentiation between wild and farmed populations in both species. Wild populations showed a low degree of differentiation, particularly in seabream. Despite this, a slight differentiation was observed between Atlantic and Mediterranean seabream populations and between western and eastern Mediterranean seabass populations. However, farmed populations were quite heterogeneous and showed a high degree of differentiation. Some farmed populations of both species showed a genetic makeup similar to that found in wild populations. In general, the effective population size was large (> 1000) for wild and small (< 100) for farmed populations of both species. Decreasing contents of the omega-3 fatty acids EPA and DHA in farmed Atlantic salmon reduces the nutritional value for consumers and influences physiological processes in the fish. Most studies of physiological omega-3 effects in Atlantic salmon have focused on responses to changed dietary levels. However, there are inherent differences in contents of EPA and DHA between Atlantic salmon fed the same diet. Revealing the metabolic differences between these individuals may contribute towards the goal of producing healthy fish of high nutritional value. Previous studies have showed the potential of selective breeding to increase the total n-3 LC-PUFA levels in salmon tissues, but knowledge on the genetic parameters for individual muscle fatty acids and their relationships with other traits were lacking. The overall aim of this thesis was to identify the genetic basis and underlying biological mechanisms associated with omega-3 content in Atlantic salmon fillet. For this purpose, the Atlantic salmon from a SalmoBreed AS broodstock population studied were reared together under the same conditions and fed the same diet throughout their life, in order to reduce environmental factors to a minimum.
