Abstract Background The updated status of hypertension and its risk factors are poorly evaluated in Tibetan highland areas. We initiated a large-scale cross-sectional survey to provide updated status of hypertension and its risk factors (especially salt intake) in the Ganzi Tibetan Plateau, China. Methods Stratified multistage random sampling was performed to obtain a representative sample of 4,036 adult residents from 4 counties in the Ganzi Tibetan area. The whole survey population was used to present the epidemiology and risk factors of hypertension. The participants with blood and urine biochemistry data were used to analyze the relationship between salt intake parameters and hypertension. Results Stratified multistage random sampling was performed to obtain a representative sample of 4,036 adult residents. The overall prevalence rate of hypertension was 33.5% (the age-adjusted prevalence rate was 28.9%). A total of 50.9% of the hypertensive patients knew their conditions; 30.1% of them received antihypertensive treatment; and 11.2% of them had their blood pressure controlled. Age, male sex, living altitude ≥ 3500 m, overweight and abdominal obesity were positively correlated with hypertension. In addition, the adjusted odds ratio (OR) for hypertension was 1.33 (95% CI: 1.01–1.74) for drinking tea with salt, and 1.51 (95% CI: 1.32–1.72) for per SD increase in the estimation of 24-hour urinary sodium excretion (e24hUSE). Furthermore, per 100mmol/day increase in e24hUSE was associated with elevation of blood pressure (+ 10.16, 95% CI: 8.45–11.87 mmHg for SBP; +3.83, 95% CI: 2.74–4.93 mmHg for DBP) in this population. Conclusions Our survey suggests a heavy disease burden of hypertension in the Ganzi Tibetan Plateau. Age, male sex, altitude of residence ≥ 3500 m, overweight, abdominal obesity, and excessive salt intake (shown as drinking tea with adding salt and a higher level of e24hUSE) all increased the risk of hypertension in this highland area.

Abstract Background Pelvic organ-preserving radical cystectomy (POPRC) has been reported to result in a better postoperative quality of life in female with bladder cancer compared to standard radical cystectomy (SRC). However, its oncological outcomes remain a concern. Patients and methods Female patients with bladder cancer who underwent POPRC or SRC were identified from the Surveillance, Epidemiology, and End Results (SEER) database. Logistic regression was used to identify predictors of POPRC usage. To avoid the potential impact of baseline differences between groups on survival, a 1:2 propensity score matching (PSM) was implemented. After that, Kaplan-Meier curves and Log-rank tests were used to determine the significance of overall survival (OS) differences between patients in the SRC group and POPRC group. Finally, subgroup analysis based on predetermined indicators was performed. Results A total of 2193 patients were included with a median follow-up of 53 months, of whom 233 (10.6%) received POPRC and 1960 (89.4%) received SRC. No definitive predictors of POPRC were identified. Before PSM, POPRC resulted in comparable OS to SRC (HR = 1.09, p = 0.309), while after PSM, POPRC was associated with significantly worse OS (HR = 1.23, p = 0.038). In subgroup analyses, POPRC led to non-inferior OS (HR = 1.18, 95%CI 0.71–1.95, p = 0.531) in patients with non-muscle invasive bladder cancer (NMIBC) and T2 patients (HR = 1.07, p = 0.669), but significantly worse OS in T3 patients (HR = 1.41, p = 0.02). Conclusion Currently, patients undergoing POPRC have not undergone strict screening, and candidates for POPRC should have more stringent criteria in the future to achieve satisfactory oncological outcomes. However, flaws in the study make more evidence needed to support our findings.

Abstract Under the target of ‘emission peak and carbon neutrality’, electricity distribution networks will massively access low‐carbon technologies, which will result in problems such as insufficient hosting capacity, unbalanced electricity loads, degraded power quality etc. The low‐voltage flexible distribution network (LVFDN), which interconnects its low‐voltage service transformers using flexible power electronic devices (flexible interconnected devices [FIDs]) is considered an effective means to deal with the challenges above. The total supply capability (TSC) of LVFDN is proposed. Firstly, the typical structures of LVFDN and their operation modes are proposed. Then, the TSC model of LVFDN, which formulates flexible power flow control and multi‐level (medium‐voltage feeder and low‐voltage flexible interconnection) load transfer is proposed. Due to the non‐linear non‐convex characteristics of the proposed TSC model, a new algorithm based on the ‘branch and bound algorithm’ is also provided. In the case study, the TSC of an actual electricity distribution network is calculated and tested by the N‐1 verification method. Finally, the variations of TSC with different capacities of the low‐voltage FID are analysed. Suggestions for the planning and operation of LVFDN are also given. A theoretical basis for the application of flexible interconnection technology in low‐voltage electricity distribution networks is provided.

Abstract The role of cybersecurity in cyber‐physical power systems (CPPS) is reviewed, focusing on the applications of dynamic state estimation (DSE) techniques. These DSE techniques are particularly relevant with the integration of phasor measurement units (PMUs) and advanced communication infrastructure. A comprehensive review on DSE techniques and applications to efficiently protect CPPS against cyberattacks is classified into three cyber resilience phases including prevention, detection, and mitigation. The DSE techniques in the prevention phase are surveyed to improve the observability of the CPPS by the robust design of the Kalman filter and optimal protection of PMUs. The DSE techniques in the detection phase are surveyed to improve the adaptability of CPPS in various attack detection scenerios and optimise the detection accuracy. The DSE techniques in the mitigation phase are surveyed to enhance the flexibility of CPPS resource utilisation with compensation‐based, isolation‐based, and scheduling‐based strategies. Finally, the benefits and limitations of each DSE technique are summarised with potential suggestions on research directions for enhancing the cyber resilience of CPPS.

Abstract Background Patient safety (PS) is a core competency for registered nurses. However, there is a gap between the PS competence of nursing students and their clinical experience in PS. This study explored the effect of PS competence levels on the occurrence of adverse events (AEs) among nursing master’s students in China. Methods A sequential mixed methods design was used, with a purposive sample across seven colleges. A total of 327 graduate nursing students, aged 22 to 38, participated in the survey, and 15 participated in qualitative interviews. The Health Professional Education in Patient Safety Survey (H-PEPSS) assessed the students’ competence levels in PS. The respondents also reported any AEs that they had been involved in over the past year. Results A total of 78 AEs occurred in the past year, with 17.7% of the participants involved 1 to 3 AEs. The most common AEs were medication administration errors (30.77%) and improper use of medical equipment/supplies (28.20%). Students acquired more competencies from the clinical setting than from the classroom setting. Three competencies learned from classroom settings were associated with clinical AEs: low clinical safety skills [OR = 0.61], inappropriate identify, response to and disclosing AE and close calls [OR = 0.454], and low confidence in working in teams with other health professionals [OR = 2.168]. Qualitative data analysis revealed five themes: recognizing AEs, reducing harm by addressing immediate risks to patients and others involved, promoting safe medication and clinical practice, managing members’ authority and team dynamics, and dealing with inter-professional conflict. Conclusions The quantitative and qualitative data align, supporting the enhancement of students’ PS competence.

Abstract Glioblastoma (GBM) is associated with a median survival rate of less than 15 months, necessitating innovative treatment approaches. This study investigates the safety and efficacy of the low-frequency magnetic field (LFMF) OM-100 instrument in GBM therapy. In vitro experiments utilized normal astrocyte and GBM cell lines, determining that OM-100 at 100 kHz for 72 h selectively targeted GBM cells without harming normal cells. Subsequent analyses revealed OM-100’s impact on cell viability, apoptosis, migration, invasion, reactive oxide species levels, and PD-L1 expression. In vivo studies on mice with U87-induced GBM demonstrated OM-100's synergy with anti-PD-1 therapy, leading to significant tumor volume reduction and increased apoptosis. Notably, OM-100 exhibited safety in healthy mice. Overall, OM-100 could enhance anti-PD-1 immunotherapy effectiveness probably by directly inhibiting tumor proliferation and migration as well as promoting PD-L1 expression, offering a promising therapeutic strategy for GBM treatment.

Abstract Background Pancreatic cancer (PaCa) is one of the most intractable and fatal malignancies and is associated with the dysregulation of long noncoding RNAs (lncRNAs), which are a large class of noncoding RNAs larger than 200 nt that act as competing endogenous RNAs or sponges for miRNAs to induce tumour biological behaviours. However, their clinical value in treating pancreatic cancer has been poorly explained, but they are essential for improving the prognosis of PaCa patients. Methods We analysed the plasma-derived exosomal lncRNA profiles of PaCa patients by using whole-transcriptome sequencing analysis and identified significantly differentially expressed lncRNAs, including LINC01268, LINC02802, AC124854.1, and AL132657.1. In the current study, the expression levels of four plasma-derived exosomal lncRNAs in PaCa plasma were validated via quantitative real-time polymerase chain reaction (qRT‒PCR). The relationship between the expression of the four lncRNAs and the clinicopathological features of patients with PaCa was also evaluated. Results We demonstrated that exosomal LINC01268, LINC02802, AC124854.1 and AL132657.1 were highly expressed in PaCa plasma compared with those in normal controls; moreover, they were positively correlated with the serum expression of carbohydrate antigen 19–9 (CA19-9). The receiver operating characteristic curves (AUCs) of the four lncRNAs were 0.8421, 0.6544, 0.7190, and 0.6321, and the AUC value of the combination of the four exosomal lncRNAs increased to 0.8476, with a sensitivity of 0.72 and specificity of 0.89. These results suggested that the plasma-derived exosomal genes LINC01268, LINC02802, AC124854.1, and AL132657.1 may be novel diagnostic markers for PaCa. Conclusions Our research demonstrated that the plasma-derived exosomal lncRNAs of PaCa patients are novel blood-based biomarkers of disease.

Abstract Polymeric-based dielectric materials hold great potential as energy storage media in electrostatic capacitors. However, the inferior thermal resistance of polymers leads to severely degraded dielectric energy storage capabilities at elevated temperatures, limiting their applications in harsh environments. Here we present a flexible laminated polymer nanocomposite where the polymer component is confined at the nanoscale, achieving improved thermal-mechanical-electrical stability within the resulting nanocomposite. The nanolaminate, consisting of nanoconfined polyetherimide (PEI) polymer sandwiched between solid Al2O3 layers, exhibits a high energy density of 18.9 J/cm3 with a high energy efficiency of ~ 91% at elevated temperature of 200°C. Our work demonstrates that nanoconfinement of PEI polymer results in reduced diffusion coefficient and constrained thermal dynamics, leading to a remarkable increase of 37°C in glass-transition temperature compared to bulk PEI polymer. The combined effects of nanoconfinement and interfacial trapping within the nanolaminates synergistically contribute to improved electrical breakdown strength and enhanced energy storage performance across temperature range up to 250°C. By utilizing the flexible ultrathin nanolaminate on curved surfaces such as thin metal wires, we introduce an innovative concept that enables the creation of a highly efficient and compact metal-wired capacitor, achieving substantial capacitance despite the minimal device volume.

Abstract Multicolor tuning of persistent luminescence has been extensively studied by deliberately integrating various luminescent units, known as activators or chromophores, into certain host compounds. However, it remains a formidable challenge to fine-tune the persistent luminescence spectra either in organic materials, such as small molecules, polymers, metal-organic complexes and carbon dots, or in doped inorganic crystals. Herein, we present a strategy to delicately control the persistent luminescence wavelength by engineering sub-bandgap donor-acceptor states in a series of single-phase Ca(Sr)ZnOS crystals. The persistent luminescence emission peak can be quasi-linearly tuned across a broad wavelength range (500–630 nm) as a function of Sr/Ca ratio, achieving a precision down to ~5 nm. Theoretical calculations reveal that the persistent luminescence wavelength fine-tuning stems from constantly lowered donor levels accompanying the modified band structure by Sr alloying. Besides, our experimental results show that these crystals exhibit a high initial luminance of 5.36 cd m− 2 at 5 sec after charging and a maximum persistent luminescence duration of 6 h. The superior, color-tunable persistent luminescence enables a rapid, programable patterning technique for high-throughput optical encryption.

Abstract Modern human society is burdened with the pandemic of cardiovascular and metabolic diseases. Metrnl is a widely distributed secreted protein in the body, involved in regulating glucose and lipid metabolism and maintaining cardiovascular system homeostasis. In this review, we present the predictive and therapeutic roles of Metrnl in various cardiovascular and metabolic diseases, including atherosclerosis, ischemic heart disease, cardiac remodeling, heart failure, hypertension, chemotherapy-induced myocardial injury, diabetes mellitus, and obesity.

Abstract Subarachnoid hemorrhage (SAH) significantly compromises the blood-brain barrier (BBB) and impairs patient recovery. This study elucidates the critical role of astrocytic Neogenin-1 (NEO1) in BBB integrity post-SAH and examines the regulatory effects of hepcidin on endothelial cell (EC) function amid NEO1-mediated disruptions in iron homeostasis. Proteomic analyses of cerebrospinal fluid (CSF) from SAH patients revealed a substantial decrease in NEO1 expression, identifying it as a key factor in BBB integrity. 111 CSF proteins were significantly reduced in early SAH stages (days 1–3), with NEO1 among the most significantly altered. Detecting and removing pesticide and veterinary drug residues in agricultural products are important for ensuring food safety. Current methods for detecting pesticide residues are mainly relied on sophisticated instruments, which require long detection time and skilled operators, limiting their accessibility. Metal-Organic Frameworks (MOFs) are a hybrid material composed of metal ions/clusters and organic ligands. MOFs possess multiple functions such as adsorption enrichment, catalytic degradation, and fluorescent and electrochemical signal generation. Therefore, MOFs have great potential in rapidly detecting and removing pesticide and veterinary drug residues. This review introduces functionalities of MOFs: Enrichment and removal, catalytic degradation, fluorescence sensing, and electrochemical sensing, and then summarizes their applications for rapid detecting and removing pesticide and veterinary drug residues. Additionally, this article comprehensively analyzes the advantages and disadvantages about combining RGB analysis and portable monitoring devices with MOFs. Also, the challenges of applying MOFs to detecting and removing pesticide and veterinary drug residue are discussed. Overall, this paper provides a reference for developing techniques to rapidly detect and remove containments in agricultural products.

To improve the solubility and stability of lutein (LUT), this study utilized the electrostatic self-assembly method to encapsulate LUT within food-grade protein nanoparticles and composite nanoparticles derived from soybean protein isolates (SPI) and fucoidin (FUD). The impact of the SPI/FUD mass ratio on the formation of ternary composite nanoparticles, known as lutein-soybean protein isolate-fucoidin nanoparticles (LUT-SPI-FUD NPs), was examined. The preparation process underwent optimization through single-factor experiments and the Box-Behnken response surface methodology. Various characterization techniques, including ultraviolet spectroscopy, Fourier-transform infrared spectroscopy, differential scanning calorimetry and X-ray diffraction were employed to delve into the mechanisms governing the formation of these two types of nanoparticles to compare with their solubility, stability, and antioxidant properties. The results showed that LUT-SPI-FUD NPs with SPI/FUD mass ratio of 4:1 had an absolute value of zeta potential greater than 30 mV, a polydispersity index, and a smaller particle size. Under these conditions, the most favorable formulation and process parameters were as follows: A LUT/SPI mass ratio of 1:7.587, SPI mass concentration of 0.83 mg/mL, and a stirring time of 1.493 hours. Characterization analysis verified the ideal particle size and more concentrated distribution of both LUT-SPI NPs and LUT-SPI-FUD NPs, and the binding forces were the interplay of hydrogen bonds, hydrophobic interactions, and electrostatic forces. Solubility, stability, and antioxidant assessments indicated that the conversion of LUT into LUT-SPI NPs and LUT-SPI-FUD NPs substantially increased its solubility in purified water by 350.74 and 432.42 times, respectively. Abstract A body area network involving wearable sensors distributed around the human body can continuously monitor physiological signals, finding applications in personal healthcare and athletic evaluation. Existing solutions for near-field body area networks, while facilitating reliable and secure interconnection among battery-free sensors, face challenges including limited spectral stability against external interference. Here we demonstrate a textile metamaterial featuring a coaxially-shielded internal structure designed to mitigate interference from extraneous loadings. The metamaterial can be patterned onto clothing to form a scalable, customizable network, enabling communication between near-field reading devices and battery-free sensing nodes placed within the network. Proof of concept demonstration shows the metamaterial’s robustness against mechanical deformation and exposure to lossy, conductive saline solutions, underscoring its potential applications in wet environments, particularly in athletic activities involving water or significant perspiration, offering insights for the future development of radio frequency components for a robust body area network at a system level.

Excessive consumption of refined grains harms human health and ecosystem viability. Whole grains, as a healthy and sustainable alternative to refined grains, can benefit individual health by providing dietary fiber, B vitamins, and bioactive substances. Additionally, they aid in improving the environment due to their higher extraction rate and lower carbon emission during the processing stage. However, few studies have attempted to evaluate the economic and social benefits of increasing the amount of whole grain in grain intake. This paper estimates the potential savings in healthcare costs and reduced food carbon footprints (CFs) that could result from a shift toward whole grain consumption following the Chinese Dietary Guidelines (CDG). We investigate hypothetical scenarios where a certain proportion (5–100%) of Chinese adults could increase their whole grain intakes as proposed by CDG to meet the average shortfall of 30.2 g. In that case, the healthcare costs for associated diseases (e.g., type 2 diabetes mellitus (T2DM), cardiovascular disease (CVD), and colorectal cancer (CRC)) are expected to reduce by a substantial amount, from USD 2.82 to 56.37 billion; the carbon emission levels are also projected to decrease by 0.24–5.72 million tons. This study provides compelling evidence that advocating for the transition towards greater consumption of whole grain products could benefit individual health, the environment, and society, by reducing both healthcare costs and carbon emissions.

Abstract Background Tumor heterogeneity presents a formidable challenge in understanding the mechanisms driving tumor progression and metastasis. The heterogeneity of hepatocellular carcinoma (HCC) in cellular level is not clear. Methods Integration analysis of single-cell RNA sequencing data and spatial transcriptomics data was performed. Multiple methods were applied to investigate the subtype of HCC tumor cells. The functional characteristics, translation factors, clinical implications and microenvironment associations of different subtypes of tumor cells were analyzed. The interaction of subtype and fibroblasts were analyzed. Results We established a heterogeneity landscape of HCC malignant cells by integrated 52 single-cell RNA sequencing data and 5 spatial transcriptomics data. We identified three subtypes in tumor cells, including ARG1+ metabolism subtype (Metab-subtype), TOP2A+ proliferation phenotype (Prol-phenotype), and S100A6+ pro-metastatic subtype (EMT-subtype). Enrichment analysis found that the three subtypes harbored different features, that is metabolism, proliferating, and epithelial-mesenchymal transition. Trajectory analysis revealed that both Metab-subtype and EMT-subtype originated from the Prol-phenotype. Translation factor analysis found that EMT-subtype showed exclusive activation of SMAD3 and TGF-β signaling pathway. HCC dominated by EMT-subtype cells harbored an unfavorable prognosis and a deserted microenvironment. We uncovered a positive loop between tumor cells and fibroblasts mediated by SPP1-CD44 and CCN2/TGF-β-TGFBR1 interaction pairs. Inhibiting CCN2 disrupted the loop, mitigated the transformation to EMT-subtype, and suppressed metastasis. Conclusion By establishing a heterogeneity landscape of malignant cells, we identified a three-subtype classification in HCC. Among them, S100A6+ tumor cells play a crucial role in metastasis. Targeting the feedback loop between tumor cells and fibroblasts is a promising anti-metastatic strategy.

AIM: To investigate the effects of fibrillin-1 (FBN1) deletion on the integrity of retina-blood barrier function and the apoptosis of vascular endothelial cells under diabetic conditions. METHODS: Streptozotocin (STZ)-induced diabetic mice were used to simulate the diabetic conditions of diabetic retinopathy (DR) patients, and FBN1 expression was detected in retinas from STZ-diabetic mice and controls. In the Gene Expression Omnibus (GEO) database, the GSE60436 dataset was selected to analyze FBN1 expressions in fibrovascular membranes from DR patients. Using lentivirus to knock down FBN1 levels, vascular leakage and endothelial barrier integrity were detected by Evans blue vascular permeability assay, fluorescein fundus angiography (FFA) and immunofluorescence labeled with tight junction marker in vivo. High glucose-induced monkey retinal vascular endothelial cells (RF/6A) were used to investigate effects of FBN1 on the cells in vitro. The vascular endothelial barrier integrity and apoptosis were detected by trans-endothelial electrical resistance (TEER) assay and flow cytometry, respectively. RESULTS: FBN1 mRNA expression was increased in retinas of STZ-induced diabetic mice and fibrovascular membranes of DR patients (GSE60436 datasets) using RNA-seq approach. Besides, knocking down of FBN1 by lentivirus intravitreal injection significantly inhibited the vascular leakage compared to STZ-DR group by Evans blue vascular permeability assay and FFA detection. Expressions of tight junction markers in STZ-DR mouse retinas were lower than those in the control group, and knocking down of FBN1 increased the tight junction levels. In vitro, 30 mmol/L glucose could significantly inhibit viability of RF/6A cells, and FBN1 mRNA expression was increased under 30 mmol/L glucose stimulation. Down-regulation of FBN1 reduced high glucose (HG)-stimulated retinal microvascular endothelial cell permeability, increased TEER, and inhibited RF/6A cell apoptosis in vitro. CONCLUSION: The expression level of FBN1 increases in retinas and vascular endothelial cells under diabetic conditions. Down-regulation of FBN1 protects the retina of early diabetic rats from retina-blood barrier damage, reduce vascular leakage, cell apoptosis, and maintain vascular endothelial cell barrier function.

Non-small cell lung cancer (NSCLC) is a prevalent and aggressive global malignancy. Conventional surgical treatments, radiotherapy, chemotherapy, and targeted therapies often fall short in halting disease progression due to inherent limitations, resulting in suboptimal prognosis. Despite the advent of immunotherapy drugs offering new hope for NSCLC treatment, current efficacy remains insufficient to meet all patient needs. Therefore, actively exploring novel immunotherapeutic approaches to further reduce mortality rates in NSCLC patients has become a crucial focus of NSCLC research. This article aims to systematically review the anti-tumor effects of interleukin-21 and follicular helper T cells in NSCLC immunotherapy by summarizing and analyzing relevant literatures from both domestic and international sources, as well as exploring the potential for enhancing NSCLC treatment prospects through immune checkpoint regulation via immunotherapeutic means.

Abstract The early diagnosis of esophageal cancer (EC) is extremely challenging due to a lack of effective diagnostic methods. The study presented herein aims to assess whether serum volatile organic compounds (VOCs) could be utilised as emerging diagnostic biomarkers for EC. Gas chromatography-ion mobility spectrometry (GC-IMS) was used to detect VOCs in the serum samples of 55 patients with EC, with samples from 84 healthy controls (HCs) patients analysed as a comparison. All machine learning analyses were based on data from serum VOCs obtained by GC-IMS. A total of 33 substance peak heights were detected in all patient serum samples. The ROC analysis revealed that four machine learning models were effective in facilitating the diagnosis of EC. In addition, the random forests model for 5 VOCs had an AUC of 0.951, with sensitivities and specificities of 94.1 and 96.0%, respectively.

Abstract The evolution and mechanism of ground collapse caused by underground water pipeline leakage have become increasingly significant as more urban areas experience collapses. Based on the principle of similarity, and considering the engineering context of road collapses in Anqing City, Anhui Province, this study designed a 3 m × 2 m × 2 m rupture-collapse model test device. Digital Image Correlation (DIC) technology was employed to investigate the erosion process and collapse mechanisms caused by underground pipeline leakage. The results indicate that groundwater seepage provides the driving force for collapses, combined with the migration space provided by defects, collectively triggering the collapses. When groundwater seepage is minimal, the cohesive forces between soil particles maintain soil stability. As groundwater seepage increases, the soil particle framework is eroded, leading to soil structure destabilization and collapse initiation. The depth of collapse significantly influences stress evolution: stress evolution intensity beneath and above the collapse pit is positively correlated with the distance from the collapse pit bottom, but negatively correlated with the distance from the defect. The research provides insights for the early warning and management of ground collapse.

To explore the variances between single-reinforced and hybrid-reinforced Al–Mg–Si alloys with TiC and B4C, 10 wt.%-(TiC + B4C)/6061Al, 10 wt.%-B4C/6061Al, 10 wt.%-TiC/6061Al and 6061Al were prepared via wet mixing for 8 h followed by vacuum hot-press sintering at 580 °C and 30 MPa for 2 h. Microstructure evolution, mechanical properties, and wear behavior were investigated in this study. The results show that unlike single reinforcement of TiC, single reinforcement of B4C and hybrid reinforcement of TiC and B4C altered the distribution of Si and facilitated the in-situ formation of SiC. Both single reinforcement and hybrid reinforcement resulted in mixed fracture characteristics of ductile fracture and cleavage fracture, while hybrid reinforcement of TiC and B4C demonstrated superior strength and plasticity matching. At equivalent particle mass fractions, single reinforcement of TiC was more conducive to inducing recrystallization during sintering, followed by single reinforcement of B4C, and hybrid reinforcement of TiC and B4C had the least effect. At equivalent particle mass fractions, single reinforcement of B4C had the most significant effect on improving the wear resistance, followed by hybrid reinforcement of TiC and B4C, and single reinforcement of TiC had the least effect. All particle addition methods demonstrated a mixed wear mechanism involving abrasive wear, adhesive wear, and spalling wear. This study provides valuable insights into the development of aluminum matrix composites.

Abstract The utilization of drag-reducing polymers has long been hindered by their irritancy, corrosiveness, and toxicity across various domains. In this investigation, we explored sodium alginate, a natural drag reducer, for its efficacy in reducing drag and its resilience to shear in millimeter-scale pipelines. Initially, an experimental setup was devised to assess the drag reduction capabilities of sodium alginate at varying concentrations and flow rates using Response Surface Methodology (RSM). The relationship between drag reduction (DR), concentration (C), and flow rate (Q) was established by analyzing the experimental data. Subsequently, variance analysis was employed to validate the data accuracy, with a comparison between predicted and experimental DR values revealing an error margin within ± 20%. Analysis of cyclic shear testing of sodium alginate solution in tubes demonstrated its effectiveness as a shear flow drag reducer. Furthermore, results from laser particle size analysis indicated minimal molecular breakage of sodium alginate during cyclic shear.

Abstract To investigate the damage mechanisms in granite’s physical and mechanical properties after high-temperature water quenching, this study employed MTS815.04 for uniaxial compression tests on thermally treated specimens, with concurrent acoustic emission monitoring, and utilized nanoindentation for micromechanical analysis. The results show that with increasing temperature, granite's peak strength and elastic modulus decrease, with a sharp decline after 400–500 °C, corresponding to a significant increase in the internal damage, which can be detected by acoustic emission monitoring. Below 500 °C, macroscopic mechanical degradation is due to mineral thermophysical property differences, while above 500 °C, microcrack development is the main deterioration factor. The failure mode shifts from tensile to tensile-shear complex to shear failure, with transition points at 400 °C and 800 °C. The results of this study are of certain reference value for improving the efficiency of extracting thermal energy from dry-hot rocks and providing security guidance for the tunnel restoration process following fire damage.

Abstract α particles must be monitored to be managed as radioactive diagnostic agents or nuclear activity indicators. The new generation of perovskite detectors suffer from limited energy resolution, which affects spectroscopy and imaging applications. Here, we report that the solution-grown CsPbBr3 crystal exhibits a low and stable dark current (34.6 nA·cm−2 at 200 V) by thinning the as-grown crystal to decrease the high concentration CsPb2Br5 phase near the surface. The introduction of the Schottky electrode for the CsPbBr3 detector further reduces the dark current and improves the high-temperature stability. An energy resolution of 6.9% is achieved with the commercial electronic system, while the effects of air scattering and absorption are investigated. Moreover, 1.1% energy resolution is recognized by a full-customized readout application-specific integrated circuit without any additional signal processing, which matches well with the given parameters of the CsPbBr3 detector by reducing the parasitic capacitance and electronic noise.

Abstract The behavior of quark matter with both external electric field and chiral chemical potential is theoretically and experimentally interesting to consider. In this paper, the case of simultaneous presence of imaginary electric field and chiral chemical potential is investigated using the lattice QCD approach with $$N_f=1+1$$ N f = 1 + 1 dynamical staggered fermions. We find that overall both the imaginary electric field and the chiral chemical potential can exacerbate chiral symmetry breaking, which is consistent with theoretical predictions. However we also find a non-monotonic behavior of chiral condensation at specific electric field strengths and chiral chemical potentials. The behavior of Polyakov loop in the complex plane is not significantly affected by chiral chemical potential in the region of the parameters considered.

Abstract Recently, N6-methyladenosine (m6A) has aroused widespread discussion in the scientific community as a mode of RNA modification. m6A comprises writers, erasers, and readers, which regulates RNA production, nuclear export, and translation and is very important for human health. A large number of studies have found that the regulation of m6A is closely related to the occurrence and invasion of tumors, while the homeostasis and function of the tumor microenvironment (TME) determine the occurrence and development of tumors to some extent. TME is composed of a variety of immune cells (T cells, B cells, etc.) and nonimmune cells (tumor-associated mesenchymal stem cells (TA-MSCs), cancer-associated fibroblasts (CAFs), etc.). Current studies suggest that m6A is involved in regulating the function of various cells in the TME, thereby affecting tumor progression. In this manuscript, we present the composition of m6A and TME, the relationship between m6A methylation and characteristic changes in TME, the role of m6A methylation in TME, and potential therapeutic strategies to provide new perspectives for better treatment of tumors in clinical work.

Abstract The Marine Isotope Stage (MIS) 11c interglacial and its preceding glacial termination represent an enigmatically intense climate response to relatively weak insolation forcing. So far, a lack of radiometric age control has confounded a detailed assessment of the insolation-climate relationship during this period. Here, we present 230Th-dated speleothem proxy data from northern Italy and compare them with palaeoclimate records from the North Atlantic region. We find that interglacial conditions started in subtropical to middle latitudes at 423.1 ± 1.3 thousand years (kyr) before present, during a first weak insolation maximum, whereas northern high latitudes remained glaciated (sea level ~ 40 m below present). Some 14.5 ± 2.8 kyr after this early subtropical onset, peak interglacial conditions were reached globally, with sea level 6–13 m above present, despite weak insolation forcing. We attribute this remarkably intense climate response to an exceptionally long (~15 kyr) episode of intense poleward heat flux transport prior to the MIS 11c optimum.

Abstract The current strategies for building 2D organic-inorganic heterojunctions involve mostly wet-chemistry processes or exfoliation and transfer, leading to interface contaminations, poor crystallizing, or limited size. Here we show a bottom-up procedure to fabricate 2D large-scale heterostructure with clean interface and highly-crystalline sheets. As a prototypical example, a well-ordered hydrogen-bonded organic framework is self-assembled on the highly-oriented-pyrolytic-graphite substrate. The organic framework adopts a honeycomb lattice with faulted/unfaulted halves in a unit cell, resemble to molecular “graphene”. Interestingly, the topmost layer of substrate is self-lifted by organic framework via strong interlayer coupling, to form effectively a floating organic framework/graphene heterostructure. The individual layer of heterostructure inherits its intrinsic property, exhibiting distinct Dirac bands of graphene and narrow bands of organic framework. Our results demonstrate a promising approach to fabricate 2D organic-inorganic heterostructure with large-scale uniformity and highly-crystalline via the self-lifting effect, which is generally applicable to most of van der Waals materials.

Abstract Intangible cultural heritage is the carrier of national memory and a historical witness. Exploring the spatial distribution and chronological evolution of intangible cultural heritage is of great significance to understand the cultural integrity and historical continuity. Taking the intangible cultural heritage along the Grand Canal as an example, this study carries out spatial deconstruction based on the extent of the spread of intangible cultural heritage and reveales the aggregation pattern through spatial autocorrelation analysis. Then, the historical process and the spatial center of gravity are shown through an evolution tree model. The influencing factors are illustrated via the methods of Geodetector and qualitative analysis. The results are as follows: (1) The two ends of the canal exhibit obvious advantages in the number of intangible cultural heritage items; however, the existence of a spatial mismatch at the city–county level makes the southern endpoint occupy a dominant position. (2) Although there is a high–high aggregation pattern along the whole canal, 29.82% of the counties are distributed widely with low–low aggregation. (3) The spatial center of intangible cultural heritage items from different historical periods generally presents a changing path from north to south and returning north. (4) Intangible cultural heritage items are mainly distributed in areas with a prosperous culture, a high level of economic development, and strong policy and financial support from government. Changes in the natural environment, population migration, economic development, war disasters, and canal shipping have profound impacts over time. This study provides a new idea for the methodological advancement of spatial distribution of intangible cultural heritage, as well as studies on local cultural identity and regional consistency.

Two taxonomically controversial polypore genera with reddish brown to orange basidiomata that stain reddish with KOH solution, Aurantiporus and Hapalopilus, are revised based on additional sampling, morphological examination, and phylogenetic analysis of a combined dataset of ITS1-5.8S-ITS2-nLSU sequences. Hapalopilus is a monophyletic genus belonging to Phanerochaetaceae, whereas Aurantiporus is a polyphyletic genus belonging to Meruliaceae. Hapalopilus and Aurantiporus s. str. are circumscribed, and two new species – Aurantiporus orientalis and Hapalopilus tabuliformis – are described and illustrated from temperate China. In addition, four new combinations, viz. Aurantiporus alboaurantius, A. mutans, A. tropicus and Luteoporia albocitrina, are proposed based on morphology and phylogenetic analysis. The relationships between Aurantiporus and Hapalopilus are discussed.

Abstract Measurement of the cervical range of motion (CROM) is significant for early diagnosis of cervical spondylosis and determination of the severity of the disease. Aiming at the problem of convenient and continuous measurement of CROM and the reduction of the impact of data fluctuations, this paper proposes a low‐cost method for testing and analyzing CROM. This paper analyzes the correspondence between smartphone orientation sensor angle and CROM, obtains smartphone orientation sensing data, and uses the extreme‐point symmetric mode decomposition method by calculating the energy of the difference value to extract the adaptive global mean curve of the CROM data, and calculates the cervical ROM. The statistical analysis of the CROM test results is carried out. Results show that the method proposed can obtain the CROM including flexion and extension, lateral flexion, and rotation in a single measurement, which has the advantages of continuous measurement and low cost.

The underwater 10CrNi3MoV high-strength low-alloy steel thin-walled part with forming quality up to in-air laser deposition quality was successfully manufactured for the first time. The influencing mechanisms of complicated thermal history and water environment on microstructure evolution and mechanical properties of thin-walled part were researched. Due to austenitization and interlocking effect, the microstructure in the bottom, middle and top (below the 56th layer) areas of underwater sample and the middle area of in-air sample were composed of fine grain ferrite and granular bainite. With the increase of building height, the heat accumulation effect of underwater sample enhanced, resulting in small grain size (GS), high dislocation density, strong solution strengthening effect and low maximum texture strength at the bottom area, while the high proportion of high-angle grain boundaries (HAGBs) at the top area. These resulted in the greatest tensile strength (714 MPa) and microhardness (232 HV) at the bottom area and the highest elongation (11%) at the top area. Due to the cooling effect of water environment, the proportion of HAGBs reduced and there were oxides in fracture. As a result, the horizontal elongation of underwater sample was about 90.9% of in-air sample. Additionally, the GS reduced, and the dislocation density and solution strengthening effect increased, so the microhardness of underwater sample was slightly higher than that of in-air sample. However, due to some microscopic defects, the horizontal and vertical tensile strength of underwater sample were about 97.6% and 90.7% of in-air sample.

Abstract The Mg–9Gd–3Y–2Zn–0.5Zr alloy was studied for its long-term corrosion behavior in a simulated coastal storage environment. The results show that the Mg12 (Y, Gd) Zn phase in the Mg–9Gd–3Y–2Zn–0.5Zr alloy forms a galvanic couple with α-Mg, and the Mg12 (Y, Gd) Zn phase acts as a cathode to accelerate α-Mg during the corrosion initiation period. The corrosion of the anode is subsequently transformed into corrosive dissolution of the anode. With the dissolution of the Mg12 (Y, Gd) Zn phase, elements such as Gd and Y are gradually distributed into the entire corrosion product layer, improving the protective performance of the corrosion product layer by forming dense Gd2O3 and Y2O3.

Abstract Background Excessive backfat deposition lowering carcass grade is a major concern in the pig industry, especially in most breeds of obese type pigs. The mechanisms involved in adipogenesis and fat accumulation in pigs remain unclear. Lysine 2-hydroxyisobutyrylation (Khib), is a novel protein post-translational modification (PTM), which play an important role in transcription, energy metabolism and metastasis of cancer cells, but its role in adipogenesis and fat accumulation has not been shown. Results In this study, we first analyzed the modification levels of acetylation (Kac), Khib, crotonylation (Kcr) and succinylation (Ksu) of fibro-adipogenic progenitors (FAPs), myogenic precursors (Myo) and mesenchymal stem cells (MSCs) with varied differentiation potential, and found that only Khib modification in FAPs was significantly higher than that in MSCs. Abstract Helicobacter pylori (H. pylori) is currently recognized as the primary carcinogenic pathogen associated with gastric tumorigenesis, and its high prevalence and resistance make it difficult to tackle. A graph neural network-based deep learning model, employing different training sets of 13,638 molecules for pre-training and fine-tuning, was aided in predicting and exploring novel molecules against H. pylori. A positively predicted novel berberine derivative 8 with 3,13-disubstituted alkene exhibited a potency against all tested drug-susceptible and resistant H. pylori strains with minimum inhibitory concentrations (MICs) of 0.25–0.5 μg/mL. Pharmacokinetic studies demonstrated an ideal gastric retention of 8, with the stomach concentration significantly higher than its MIC at 24 h post dose. Oral administration of 8 and omeprazole (OPZ) showed a comparable gastric bacterial reduction (2.2-log reduction) to the triple-therapy, namely OPZ + amoxicillin (AMX) + clarithromycin (CLA) without obvious disturbance on the intestinal flora. A combination of OPZ, AMX, CLA, and 8 could further decrease the bacteria load (2.8-log reduction). More importantly, the mono-therapy of 8 exhibited comparable eradication to both triple-therapy (OPZ + AMX + CLA) and quadruple-therapy (OPZ + AMX + CLA + bismuth citrate) groups. SecA and BamD, playing a major role in outer membrane protein (OMP) transport and assembling, were identified and verified as the direct targets of 8 by employing the chemoproteomics technique. In summary, by targeting the relatively conserved OMPs transport and assembling system, 8 has the potential to be developed as a novel anti-H. pylori candidate, especially for the eradication of drug-resistant strains.

Abstract Background The intrusion of maxillary anterior teeth is often required and there are various intrusion modes with mini-implants in clear aligner treatment. The objective of this study was to evaluate the effectiveness of maxillary anterior teeth intrusion with different intrusion modes, aiming to provide references for precise and safe intrusion movements in clinical practice. Methods Cone-beam computed tomography and intraoral optical scanning data of a patient were collected. Finite element models of the maxilla, maxillary dentition, periodontal ligaments (PDLs), clear aligner (CA), attachments, and mini-implants were established. Different intrusion modes of the maxillary anterior teeth were simulated by changing the mini-implant site (between central incisors, between central and lateral incisor, between lateral incisor and canine), loading site (between central incisors, on central incisor, between central and lateral incisor, between lateral incisor and canine), and loading mode (labial loading and labiolingual loading). Ten conditions were generated and intrusive forces of 100 g were applied totally. Then displacement tendency of the maxillary anterior teeth and CA, and stress of the PDLs were analyzed. Results For the central incisor under condition L14 and for the canine under conditions L11, L13, L23, and L33, the intrusion amount was negative. Under other conditions, the intrusion amount was positive. The labiolingual angulation of maxillary anterior teeth exhibited positive changes under all conditions, with greater changes under linguoincisal loading. The mesiodistal angulation of canine exhibited positive changes under labial loading, while negative changes under linguoincisal loading except for condition L14. Conclusions The intrusion amount, labiolingual and mesiodistal angulations of the maxillary anterior teeth were affected by the mini-implant site, loading site, and loading mode. Labial and linguoincisal loading may have opposite effects on the intrusion amount of maxillary anterior teeth and the mesiodistal angulation of canine. The labiolingual angulation of the maxillary incisors would increase under all intrusion modes, with greater increases under linguoincisal loading.

Ultra-intense, ultra-fast X-ray free-electron lasers (XFELs) enable the imaging of single protein molecules under ambient temperature and pressure. A crucial aspect of structure reconstruction involves determining the relative orientations of each diffraction pattern and recovering the missing phase information. In this paper, we introduce a predicted model-aided algorithm for orientation determination and phase retrieval, which has been tested on various simulated datasets and has shown significant improvements in the success rate, accuracy and efficiency of XFEL data reconstruction.

Aiming at the problem that the crack extension law of deep high-gas coal seams is not clear during blasting and permeability enhancement under the action of ground stress, firstly, according to the propagation and superposition of blasting stress wave, the stress distribution law around the blast hole under the coupling effect of ground stress and blasting was theoretically analyzed. Then, the crack extension characteristics of single-hole blasting under different ground stress conditions were investigated through laboratory tests, on the basis, the numerical simulation was carried out to investigate the crack extension mechanism and the penetration process of double-hole blasting under different lateral stress coefficients, and combined with the ImageJ image recognition software and LS-PrePost software, the effect of ground stress on the degree of crack development was characterized using crack density and extension length as a quantitative index. Finally, based on the results of laboratory test and numerical simulation, the layout scheme of coal seam blasting holes for cracking and permeability enhancement considering the influence of ground stress was proposed. The results of single-hole blasting laboratory test shown that, the ground stress can reduce the tensile stress induced by the blasting load and thus inhibit crack extension. Under the condition of non-hydrostatic ground stress, the tensile stresses orthogonal to the direction of the larger principal stresses were weakened, resulting in the inhibition of crack extension in this direction, causing the cracks to preferentially extend in the direction of higher stresses. The numerical simulation results of double-hole blasting shown that, when the lateral stress coefficient was not 1, the blasting crack expansion was directionality, and the main crack tended to expand in the direction of higher ground stress, resulting in an elliptical shape of the blast crack area, and the direction of the long axis and the larger in-situ stress were consistent. According to the results of laboratory tests and numerical calculations, it was advisable to drill holes along the direction of large ground stress to improve the effect of fracturing and permeability when blasting in deep coal seams. The research results are of great significance for understanding the crack expansion mechanism and optimizing the blast hole layout scheme when blasting for permeability enhancement in deep high-gas coal seams.

Study Design A retrospective cohort study. Purpose This study aimed to assess the reliability of quantitative computed tomography (QCT) in measuring bone mineral density (BMD) of instrumented vertebrae and investigate the effect of less paraspinal muscle damage on BMD changes after lumbar interbody fusion. Overview of Literature Patients always experience a decrease in vertebral BMD after lumbar interbody fusion. However, to the best of our knowledge, no study has analyzed the effect of paraspinal muscles on BMD changes. Methods This retrospective analysis included a total of 155 patients who underwent single-level lumbar fusion, with 81 patients in the traditional group and 74 patients in the Wiltse group (less paraspinal muscle damage). QCT was used to measure the volumetric BMD (vBMD), Hounsfield unit value, and cross-sectional area of the paraspinal muscles at the upper instrumented vertebrae (UIV), vertebrae one segment above the UIV (UIV+1), and the vertebrae one segment above the UIV+1 (UIV+2). Statistical analyses were performed. Results No significant differences in general data were observed between the two groups (p>0.05). Strong correlations were noted between the preoperative and 1-week postoperative vBMD of each segment (p0.05). Vertebral BMD loss was significantly higher in UIV+1 and UIV+2 in the traditional group than in the Wiltse group (−13.6%±19.1% vs. −4.2%±16.5%, −10.8%±20.3% vs. −0.9%±37.0%; p0.05). Conclusions QCT can reliably determine BMD in the instrumented spine after lumbar interbody fusion. With QCT, we found that reducing paraspinal muscle destruction through the Wiltse approach during surgery can help preserve the adjacent vertebral BMD; however, it does not help increase the BMD in the instrumented vertebrae.

Binocular image stereo matching is a key technology to realize autonomous obstacle avoidance and visual reconnaissance of unmanned auxiliary transport vehicles in coal mines. However, factors such as high dust and unstable lighting conditions in coal mines can lead to Salt-and-pepper noise in the images collected by the visual sensor, resulting in a high stereo matching error rate. Therefore, a Census stereo matching algorithm based on the combination of threshold and weight is proposed to reduce the impact of Salt-and-pepper noise on stereo matching. The main contributions include: ① threshold processing is carried out on the gray values of all pixels in the support window to remove the pixels with maximum and minimum gray values in the support window and solve the impact of outlier on the weighted fusion; ② the four diagonal pixels corresponding to the center point are weighted and fused to replace the center point pixel. Select pixel points along the four diagonal lines intersecting at the center pixel, with step sizes ranging from 1 to 3. According to the corresponding steps, weights of 0.7, 0.2, and 0.1 are assigned. Multiply the valid pixel points among these 12 points by their respective weights, then divide by the sum of the valid weights. This process yields the reference value of the center pixel point after weighted processing, addressing the issue of traditional algorithms' dependency on the center pixel of the Census transform window. Consequently, this approach enhances matching precision. The experimental results show that the average error rate calculated by the proposed algorithm is reduced by 5.64% compared to traditional Census algorithms, and reduced by 1.71% compared to the mean-based Census algorithm. What's more, the average error rate under different noise levels calculated by the proposed algorithm is reduced by 15.93% compared to the traditional Census algorithm, and reduced by 16.62% compared to the mean-based one. In non-occluded areas, the error matching rate of our algorithm is reduced by 17.19% compared to the traditional Census algorithm and 18.11% compared to the mean-based Census algorithm. The proposed Census stereo matching algorithm, which combines threshold and weight, effectively enhances the robustness against noise, reduces the error rate, and improves matching accuracy.

Abstract Retinoblastoma is one of the most common ocular malignancies in children. Bmi-1, a member of the Polycomb group family of transcriptional repressors, is expressed in a variety of tumors. The purpose of our study was to explore the role of Bmi-1 in retinoblastoma. RT-qPCR and western blot were used for calculating the mRNA and protein levels of Bmi-1 and RKIP. MTT, Wound healing and Transwell assays were performed to measure the proliferation, migration and invasion in retinoblastoma cells. Cell apoptosis was detected by flow cytometry. The volume and mass of transplanted tumors were detected in nude mice. Bmi-1 was over expressed, and RKIP was low expressed in retinoblastoma cells. Bmi-1 promoted cell proliferation, migration and invasion and suppressed cell apoptosis of Y79 and SO-RB50 cells. Downregulation of Bmi-1 and overexpression of RKIP inhibited cell proliferation, migration and invasion, and increased cell apoptosis. The functions of Bmi-1 knockdown on retinoblastoma cells were blocked by RKIP knockdown, but promoted by RKIP. Down-regulated Bmi-1 inhibited xenograft tumor growth, and RKIP exacerbated this inhibitory effect. Bmi-1 served as a potential therapeutic target for improving the efficacy of clinical treatment in retinoblastoma. All the findings revealed the functions of Bmi-1/RKIP axis in retinoblastoma tumorigenesis.

Abstract Metallic structures produced with laser powder bed fusion (LPBF) additive manufacturing method (AM) frequently contain microscopic porosity defects, with typical approximate size distribution from one to 100 microns. Presence of such defects could lead to premature failure of the structure. In principle, structural integrity assessment of LPBF metals can be accomplished with nondestructive evaluation (NDE). Pulsed infrared thermography (PIT) is a non-contact, one-sided NDE method that allows for imaging of internal defects in arbitrary size and shape metallic structures using heat transfer. PIT imaging is performed using compact instrumentation consisting of a flash lamp for deposition of a heat pulse, and a fast frame infrared (IR) camera for measuring surface temperature transients. However, limitations of imaging resolution with PIT include blurring due to heat diffusion, sensitivity limit of the IR camera. We demonstrate enhancement of PIT imaging capability with unsupervised learning (UL), which enables PIT microscopy of subsurface defects in high strength corrosion resistant stainless steel 316 alloy. PIT images were processed with UL spatial–temporal separation-based clustering segmentation (STSCS) algorithm, refined by morphology image processing methods to enhance visibility of defects. The STSCS algorithm starts with wavelet decomposition to spatially de-noise thermograms, followed by UL principal component analysis (PCA), fine-tuning optimization, and neural learning-based independent component analysis (ICA) algorithms to temporally compress de-noised thermograms. The compressed thermograms were further processed with UL-based graph thresholding K-means clustering algorithm for defects segmentation. The STSCS algorithm also includes online learning feature for efficient re-training of the model with new data. For this study, metallic specimens with calibrated microscopic flat bottom hole defects, with diameters in the range from 203 to 76 µm, were produced using electro discharge machining (EDM) drilling. While the raw thermograms do not show any material defects, using STSCS algorithm to process PIT images reveals defects as small as 101 µm in diameter. To the best of our knowledge, this is the smallest reported size of a sub-surface defect in a metal imaged with PIT, which demonstrates the PIT capability of detecting defects in the size range relevant to quality control requirements of LPBF-printed high-strength metals.

Abstract Dilated cardiomyopathy (DCM) is a common cause of heart failure, thromboembolism, arrhythmias, and sudden cardiac death. The quality of life and long-term survival rates of patients with dilated DCM have greatly improved in recent decades. Nevertheless, the clinical prognosis for DCM patients remains unfavorable. The primary driving factors underlying the pathogenesis of DCM remain incompletely understood. The present study aimed to identify driving factors underlying the pathogenesis of DCM from the perspective of gene regulatory networks. Single-cell RNA sequencing data and bulk RNA data were obtained from the Gene Expression Omnibus (GEO) database. Differential gene analysis, single-cell genomics analysis, and functional enrichment analysis were conducted using R software. The construction of Gene Regulatory Networks was performed using Python. We used the pySCENIC method to analyze the single-cell data and identified 401 regulons. Through variance decomposition, we selected 19 regulons that showed significant responsiveness to DCM. Next, we employed the ssGSEA method to assess regulons in two bulk RNA datasets. Significant statistical differences were observed in 9 and 13 regulons in each dataset. By intersecting these differentiated regulons and identifying shared targets that appeared at least twice, we successfully pinpointed three differentially expressed targets across both datasets. In this study, we assessed and identified 19 gene regulatory networks that were responsive to the disease. Furthermore, we validated these networks using two bulk RNA datasets of DCM. The elucidation of dysregulated regulons and targets (CDKN1A, SAT1, ZFP36) enhances the molecular understanding of DCM, aiding in the development of tailored therapies for patients.

Abstract Purpose Symptom assessment is central to appropriate adenomyosis management. Using a WeChat mini-program-based portal, we aimed to establish a valid symptom assessment scale of adenomyosis (AM-SAS) to precisely and timely identify needs of symptom management and ultimately, to alert disease recurrence. Methods A combination of intensive interviews of patients with adenomyosis and natural language processing on WeChat clinician-patient group communication was used to generate a pool of symptom items-related to adenomyosis. An expert panel shortened the list to form the provisional AM-SAS. The AM-SAS was built in a Wechat mini-programmer and sent to patients to exam the psychotically validity and clinical applicability through classic test theory and item response theory. Results Total 338 patients with adenomyosis (29 for interview, 179 for development, and 130 for external validation) and 86 gynecologists were included. The over 90% compliance to the WeChat-based symptom evaluate. The AM-SAS demonstrated the uni-dimensionality through Rasch analysis, good internal consistency (all Cronbach’s alphas above 0.8), and test-retest reliability (intraclass correlation coefficients ranging from 0.65 to 0.84).
