In this study, the temporal and spatial variation of mechanical effects (ME) and sonochemical effects (SE), the degradation behavior of polyphenols as well as the correlation coefficients were comprehensively analyzed. The ME and SE showed inverse dependence along with the axial direction of transducer. Higher irradiation distance (50 mm) enhanced the ME but inhibited the generation of hydroxyl radicals (8.29 μmol/L). Rutin and cyanidin-3-glucoside, compared with thermal degradation (70°C, 6.41%, and 11.45%), were more susceptible to free radical degradation due to the boost of SE at low temperature (10°C, 20.23%, and 34.41%). The concentration of hydroxyl radicals at 70°C (18.38 μmol/L) was about 5.77 times lower than that at 10°C (3.22 μmol/L). No significant differences in the ME were observed for a wide range of temperatures (10–70°C). The impacts of increasing duty cycle on further promoting the ME and SE were limited. The concentration of hydroxyl radicals (0–23.37 μmol/L) increased linearly with sonication time (0–45 min) and the degradation of caffeic acid only occurred on a time scale (25 min). The free radical scavenger can minimize the degradation of polyphenols by inhibiting the hydroxyl radicals (23.37 μmol/L without t-BuOH; 12.17 μmol/L with t-BuOH). The above results showed that the degradation of polyphenols attacked by hydroxyl radicals presented a dose-response relationship. The correlation between the hydroxyl radicals and polyphenols as well as the mechanical effects and extraction yield indicated that the stability of polyphenols exposed to ultrasonic field depended on the structure-radical scavenging activity relationship and the extraction may be accompanied by the degradation.

Self-aggregating polymeric micelles are novel promising vehicles to solubilize and targeting delivery hydrophobic food ingredients. Solubilization of β -carotene with octenylsuccinate oat β -glucan (OSG) micelles, in terms of the apparent solubility of β -carotene ([ S ] β -car ), was investigated in this study. Its dependence on the degree of substitution (DS) and molecular weight (M w ) of OSG and stirrer input power (IP) was explored with the aid of response surface methodology (RSM). More importantly, the freeze-thaw, thermal and storage stability of β- carotene-loaded OSG micelles were determined. Results from RSM revealed that the maximum [ S ] β -car (12.84 μg/mL) encountered with the intermediate DS (0.0183), high M w (17.1 × 10 4 g/mol) and IP (4.3 W). TEM images evidenced a minor deformation of OSG micelles upon β -glucan solubilization. In terms of size and polydispersity index, β -carotene-loaded OSG micelles exhibited good thermal stability, intermediate freeze-thaw stability and inferior storage stability. Furthermore, regarding to the retention of β -carotene, β -carotene-loaded OSG micelles were susceptible to thermal treatment and storage but kept stable against freeze-thaw treatment, but presented nice freeze-thaw stability. The loss of [ S ] β -car upon thermal treatment and storage was of temperature and time dependence. These results cast new light on the possible fortification of insoluble ingredients by OSG micelles in aqueous food systems.

Summary Barley (Hordeum vulgare L.) possesses a large and highly repetitive genome of 5.1 Gb that has hindered the development of a complete sequence. In 2012, the International Barley Sequencing Consortium released a resource integrating whole‐genome shotgun sequences with a physical and genetic framework. However, because only 6278 bacterial artificial chromosome (BACs) in the physical map were sequenced, fine structure was limited. To gain access to the gene‐containing portion of the barley genome at high resolution, we identified and sequenced 15 622 BACs representing the minimal tiling path of 72 052 physical‐mapped gene‐bearing BACs. This generated ~1.7 Gb of genomic sequence containing an estimated 2/3 of all Morex barley genes. Exploration of these sequenced BACs revealed that although distal ends of chromosomes contain most of the gene‐enriched BACs and are characterized by high recombination rates, there are also gene‐dense regions with suppressed recombination. We made use of published map‐anchored sequence data from Aegilops tauschii to develop a synteny viewer between barley and the ancestor of the wheat D‐genome. Except for some notable inversions, there is a high level of collinearity between the two species. The software HarvEST:Barley provides facile access to BAC sequences and their annotations, along with the barley–Ae. tauschii synteny viewer. These BAC sequences constitute a resource to improve the efficiency of marker development, map‐based cloning, and comparative genomics in barley and related crops. Additional knowledge about regions of the barley genome that are gene‐dense but low recombination is particularly relevant., Significance Statement We provide new genome sequence resources for barley and related species, to improve the efficiency of marker development, map‐based cloning, comparative genomics studies, and to provide insights into genome architecture. Notably, some of the gene‐rich islands in the barley genome deviate markedly from assumption that gene density and high recombination frequency are correlated.

Summary Cowpea (Vigna unguiculata L. Walp.) is a legume crop that is resilient to hot and drought-prone climates, and a primary source of protein in sub-Saharan Africa and other parts of the developing world. However, genome resources for cowpea have lagged behind most other major crops. Here we describe foundational genome resources and their application to analysis of germplasm currently in use in West African breeding programs. Resources developed from the African cultivar IT97K-499-35 include a whole-genome shotgun (WGS) assembly, a bacterial artificial chromosome (BAC) physical map, and assembled sequences from 4,355 BACs. These resources and WGS sequences of an additional 36 diverse cowpea accessions supported the development of a genotyping assay for 51,128 SNPs, which was then applied to five biparental RIL populations to produce a consensus genetic map containing 37,372 SNPs. This genetic map enabled the anchoring of 100 Mb of WGS and 420 Mb of BAC sequences, an exploration of genetic diversity along each linkage group, and clarification of macrosynteny between cowpea and common bean. The SNP assay enabled a diversity analysis of materials from West African breeding programs. Two major subpopulations exist within those materials, one of which has significant parentage from South and East Africa and more diversity. There are genomic regions of high differentiation between subpopulations, one of which coincides with a cluster of nodulin genes. The new resources and knowledge help to define goals and accelerate the breeding of improved varieties to address food security issues related to limited-input small-holder farming and climate stress. The genomes of West African breeding lines and landraces have regions of marked depletion of diversity, some of which coincide with QTL that may be the result of artificial selection or environmental adaptation. The new publicly available resources and knowledge help to define goals and accelerate the breeding of improved varieties to address food security issues related to limited-input small-holder farming and climate stress.

Knowledge of these deviant regions is relevant to trait introgression, genome-wide association studies, genomic selection model development and map-based cloning strategies. The dependence of the curcumin loading capacity (CLC) of octenylsuccinate oat β-glucan (OSG) micelles on the structural parameters (degree of substitution, DS; molecular weight, Mw) of OSG was unknown and explored in this study. Meanwhile, the curcumin-loaded OSG micelle (COM) was first characterized. The results from response surface methodology revealed that the linear effects of Mw and stirrer input power, as well as the quadratic effect of DS, were significant (p < 0.05). The maximum CLC value of the OSG micelle was obtained as 4.21 μg/mg. Dynamic light scattering showed that the average size and ζ potential of the COM were 308 nm and -10.8 mV, respectively. Transmission electron microscopy and atomic force microscopy evidenced that the COM was elliptical in shape. Fourier transform infrared spectroscopy, differential scanning calorimeltry, and X-ray diffraction revealed that curcumin was loaded in OSG micelles in an amorphous form by interacting with OSG molecules.
